Verfahren zur Herstellung von tricyclischen Aminen
5H-Dibenzo[a,b]cycloheptane und 10,11-Dihydro-5H-dibenzo[a,d]cycloheptane mit einer basischen Seitenkette in 5-Stellung sind bereits als lantidepressiv wirkende Psychopharmaka bekannt, z. B.
10,11-Dihydro-5-(3'-dimethylaminopropyliden)
5H-dibenzo[a,d]cyclohepten und
10,11-Dihydro-5-(3'-methylaminopropyliden)
5H-dibenzo[a,d]cyclohepten.
Es wurde nun überraschend gefunden, dass tricyclische Amine der Formel

in der X eine Äthylengruppe oder eine gegebenenfalls durch ein Halogenatom substituierte Vinylengruppe und Hal Chlor oder Fluor bedeutet, oder die entsprechenden 5-Methylaminopropylidenverbindungen sowie die in den aromatischen Ringen weitere Chlor- bzw. Fluoratome tragenden Derivate davon, sowie Säureadditionssalze dieser Verbindungen, die sich von den bekannten Vertretern Ider betreffenden Substanzgruppen durch die Anwesenheit eines Chlor- oder Fluoratoms in 1-Stellung unterscheiden, durch eine beträchtlich erhöhte antide- pressive Wirkung und stark verminderte Toxizität auszeichnen.
Als besonders vorteilhaft ist die fehlende oder nur geringfügige anticholinergische Wirkung zu erwäh- nen. Diese Verbindungen zeichnen sich ausserdem durch vielfältige Wirkungen auf das Nervensystem aus. So sind narkosepotenzierende, adrenolytische, sedative, antihist aminaftige und lokalanästhetische Wirkungen festge- stellt worden.
Verbindungen gemäss obiger Definition mit einer exocyclischen Doppelbindung in 5-Stellunlg bilden geometrische Isomere. Auch diese Isomere zeigen die vorstehend erwähnten überragenden Eigenschaften. Sie werden im folgenden als a- bzw. ss-Isomere bezeichnet.
Von den Chlor- bzw. Fluor-Substituenten im aromatischen Teil ist Chlor bevorzugt. X ist vorzugsweise eine unsubstituierte Äthylen- oder Vinylengruppe, kann aber auch eine durch ein Halogenatom, insbesondere ein Chlor- oder Bromatom, substituierte Vinylengruppe darstellen. Besonders interessant sind z. B.: l-Cihlor-l 0,1 1-dihydro-5-(3'-methylaminopropyl)-
5H-dibenzo[a,b]cyclohepten und 1 -Chlor-5 -(3 -methylaminopropyliden)-5 H- dibenzo[a,b]cyclohepten (insbesondere Dessen ss-Isomeres).
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von tricyclischen Aminen der Formel I sowie von den oben genannten Derivaten dieser Verbindungen, das dadurch gekennzeichnet ist, dass man eine Verbindung der Formel

die im aromatischen Teil in gleicher Weise wie die Verbindungen der Formel I substituiert sein kann, oder eine entsprechende N-Methyl-N-benzyl-aminopropylidenverbin dung debenzyliert. Erwünschtenfalls werden Isomere aus einem Isomerengemisch isoliert, und erwünschten falls wird eine erhaltene Base in ein Salz überführt.
Die erfindungsgemässe Debenzylierung wird zweckmässig durch Behandeln mit freiem Wasserstoff in Anwesenheit eines Katalysators, wie Palladium, Palladium auf Aluminiumoxid, Raney-Nickel oder Platindioxid, durchgeführt. Die Temperatur liegt zweckmässig zwischen etwa Raumtemperatur und etwa 800 C.
Die erfindungsgemäss eingesetzten Ausgangsverbindungen können auf vielen Wegen erhalten werden. Eine Methode besteht in Umsetzen des entsprechenden tricyclischen 5-Ketons mit geeigneten Grignard-Verbin- dungen. Die tricyclischen 5-Ketone sind nach an sich bekannten Methoden erhältlich.
Man kann z. B. ein entsprechendes tricyciisches 5 Keton mit
Methyl-benzylaminopropyl-magnesiumchlorid umsetzen und anschliessend das Reaktionsprodukt hydrolysieren.

worin X und Hai die oben gegebene Bedeutung haben, sowie die im aromatischen Teil in gleicher Weise wie die Verbindungen der Formel I substituierten Derivate davon können durch Reduktion in die exocyclisch gesättigten Verbindungen der Formel II übergeführt werden. Die Reduktion lässt sich zweckmässig mit nascierendem Wasserstoff, z.B. Zink in Eisessig, durch führen. Ein anderes, sehr geeignetes Reduktionsmittel ist Jodwasserstoffsäure.
Durch Behandeln einer Verbindung der Formel III mit Jodwasserstoffsäure, insbesondere in Anwesenheit von rotem Phosphor, gelangt man besonders glatt zu solchen Verbindungen der Formel II, welche in 10,11-Stellung gesättigt sind. Bei der Reduktion arbeitet man bevorzugt in Anwesenheit eines Lösungsmittels wie Essigsäure oder Essigsäureanhydrid und bei einer Temperatur zwischen Raumtemperatur und dem Siedepunkt des Reaktionsgemisches.
Die Dehydratisierung der Verbindungen der Formel III führt zu den entsprechenden Verbindunlgen mit einer exocyclischen Doppilbindung in 5-StelZung, welche, wie gesagt, ebenfalls im erfindungsgemässen Verfahren als Ausgangsstoffe eingesetzt werden können. Diese Dehydratisierung wird zweckmässig unter Verwendung von Mineralsäuren, wie Chlor- oder Bromwasserstoffsäure, durchgeführt, wobei in wasserfreiem oder wässrigem Medium gearbeitet werden kann. Vorzugsweise wird die Dehydratisierung in äthanolischer Salzsäure bei einer Temperatur zwischen Raumtemperatur und dem Siedepunkt des Reaktionsgemisches ausgeführt.
Sie gelingt jedoch auch ,durch Erwärmen, z.B. bei 500 bis Rückflusstemperatur, vorzugsweise bei Rückflusstempe- ratur, mit einem hochsiedenden wasserfreien Lösungsmittel, wie Dimethylsulfoxid. Andere übliche Wasserabspaltungsmittel können ebenfalls eingesetzt werden, z. B.
Acetylchlorid, Essigsäureanhydrid,
Trichloressigsäurenhydrid, Schwefelsäure oder
Phosphoroxichlorid, p-Tolualsulfochlorid,
Zinkchlorid oder Kaliumbisulfat, z. B. in einem inerten organischen Lösungsmittel, beispielsweise in Chloroform oder Methylenchlorid, und bei einer Temperatur zwischen Raumtemperatur und dem Siedepunkt des Reaktionsgemisches.
Erhaltene, unsymmetrisch substituierte Verfahrensprodukte, die eine exocyclische Doppelbindung tragen, und deren Salze können in ihre geometrischen Isomeren, d. h. a- bzw. ss-Isomerens getrennt werden.
Die Trennungsmethoden sind an sich bekannt. Bevorzugt trennt man die geometrischen Isomeren durch fraktionierte Kristallisation der Säureadditionssalze aus einem Lösungsmittel, z. B. Aceton, oder aus einem Lösungsmittelgemisch, z. B. Methanol/Diäthyläther.
Die Erfindung erfasst, wie gesagt, auch die Herstellung der Säureadditionssalze der vorliegenden Verfahrensprodukte. Solche Salze sind beispielsweise die wenigen mit organischen Säuren, wie Oxalsäure, Zitronensäure, Essigsäure, Milchsäure, Maleinsäure und Weinsäure, oder mit anorganischen Säuren, wie Chlorwasserstoffsäure, Bromwasserstoffsäure oder Schwefelsäure.
Die Verfahrensprodukte können als Heilmittel in Form pharmazeutischer Präparate Verwendung finden, welche sie oder ihre Salze in Mischung mit einem für die enterale oder parenterale Applikation geeigneten pharmazeutischen, organischen oder anorganischen inerten Trägermaterial enthalten. Die pharmazeutischen Präparate können in fester Form oder in flüssiger Form vorliegen. Gegebenenfalls enthalten sie Hilfsstoffe, wie Konservierungs-, Stabilisierungs-, Netz- oder Emulgiermittel, Salze zur Veränderung des osmotischen Druckes oder Puffer. Sie können auch noch andere thelrapeu- tisch wertvolle Stoffe enthalten.
Beispiel I
2 g 1-Chlor-10,11-dihydro-5-(3'-N-benzyl) Nmethyl-aminopropyi)-5 H-dibenzo [a,d]cyciohepten werden in 20 mi Methanol und 25 ml konz. Salzsäure gelöst. Nach Zusatz von 400 mg Aktivokohle und 5 ml 5% Palladiumchloridlösung wird das Gemisch bei Zimmertemperatur und 2 Atm. Überdruck etwa 20 Stunden hydriert. Das Gemisch wird anschliessend filtriert und das Filtrat unter vermindertem Druck zur Trockne eingedampft. Der Rückstand wird in Wasser gelöst, mit verdünnter Natronlauge alkalisch gestellt und mit Äther extrahiert. Die ätherische Lösung wird mit Wasser gewaschen, über Natriumsulfat getrocknet und eingedampft.
Das rohe, ölige Produkt, welches das
1-Chlor-10,11-dihydro-5-(3'-methylaminopropyl)
5H-dibenzo[a,b]cyclohepten enthält, enthält gemäss Dünnschichtchromatogramm noch etwas Ausgangsmaterial. Nach Acetylierung des Produktes mit Acetanhydrid inl Pyridin kann das Acetamid von
1-Chlor-10,11-dihydro-5-(3'-methylaminopropyl) SH-dibenzo[a,d]cyclohepten erhalten werden, welches gemäss Gaschromatogramm 20 % deschlorierte Verbindung enthält.
Das als Ausgangsverbindung verwendete
1-Chlor-10,11-dihydro-5-(3'-N-benzyl-N-methyl aminopropyl)-5H-dibenzo[a,b]cyclohepten kann wie folgt hergestellt werden:
20,6 g Gilman-Legierung in 100 ml Äther werden nach Zusatz einer Spur Methyljodid mit 146 g N-Benzyl-N-methyl-aminopropylchlorid in 250 ml absolutem Äther versetzt. Das Gemisch wird 12 Stunden unter Rühren erhitzt. Anschliessend werden 112 g l-Chlor-10,1 l-dlihydro-5H-dibenzo[a,d]cyclo- hepten-5-on in 250 ml Benzol derart zugetropft, dass die Tem peratur zwischen 15 und 20 C erhalten bleibt.
Das so erhaltene Gemisch wird 1 Stunde unter Rücluss- bedingungen gekocht, dann bei 150 C mit 120 ml gesättigter Ammoniumchloridlösung hydrolysiert und schliesslich filtriert. Das Filtrat wird unter vermindertem Druck eingeengt. Der erhaltene Rückstand kristallisiert aus hochsiedenden Petroläther beim Abkühlen auf -10 C. Man erhält
1-Chlor- 10,11 -dihydro-5 -83 '-N-benzyl-N-methyl aminopropyl)-5-hydroxy-5H-dibenzo[a,d]cy olohepten, das bei 112-114 C schmilzt.
20 g 1-Chlor-10,11-dihydro-5-(3'-N-benzyl-N-me thyl-aminopropyl)-5 -hydroxy-SH-dibenzo- [a,ld]cyolohepten in 130 ml Eisessig werden nach Zugabe von 72 ml Jodwasserstoffsäure (Dichte 1,7), 0,65 ml 50 % iger unterphosphoriger Säure und 14,2 g rotem Phosphor 12 Stunden unter Rückflussbedingnngen erhitzt. Nach dem Erkalten und Zugabe von etwas Aceton wird das Gemisch auf Eis gegossen, mit Äther versetzt und mit verdünnter Natronlauge alkaLisch gestellt. Das Gemisch wird filtriert und die ätherische Phase mit Natriumthiosulfatlösung und Wasser gewaschen, über Natriunm- sulfat getrocknet und unter vermindertem Druck eingedampft.
Man erhält öliges
1-Chlor-10,11-dihydro-5-(3'-N-benzyl-N-methyl aminopropyl)-5H-dibenzo[a,d]cyclohepten.
Dieses kann durch Zugabe von methanolischer Salzsäure, Einengen, Trocknen unter vermindertem Druck und Kristallisieren aus Aceton/tiefs. Petroläther in das entsprechende Hydrochlorid vom Schmelzpunkt 146 bis 148 C übergeführt werden.
Beispiel 2
1,7 g 1-Flüor-10,11-dihydro-5-(3'-N-benzyl-N-me thyl-aminopropyl)-5H-dibenzo[a,d]cyclo hepten werden 70 Stunden, analog wie in Beispiel 1 beschrieben, hydriert. Nach Aufarbeitung wie in Beispiel 1 beschrieben, erhält man öliges
1-Fluor-10,11-dihydro-5-(3'-methylaminopropyl)
5H-dibenzo[a,d]cyclohepten, das durch Zugabe von methanolischer Salzsäure in das reine kristalline Hydrochlorid vom Schmelzpunkt 167 bis 167,5 C überge;Sührt werden kann.
Das tals Ausgangsverbindung verwendete
1-Fluor-10,11-dihydro-5-(3'-N-benzyl-N-methyl aminopropyl)-5 Hdiberzo[a,d] cyclohepten kann wie folgt hergestellt werden.
14,8 g pulverisiertes Phthalsäureanhydrid, 18,5 g o-Fluorphenylessigsäure und 0,5 g frisch geschmolzenes Natriumacetat werden 3 Stunden auf 235-240 C geheizt und das entstehende Wasser abdestiliert. Das heisse Reaktionsgemisch wird in eine Reibschale geleert und pulverisiert. Das rohe Produkt wird aus Aceton/ Petroläther umkristallisiert. Man erhält o-Fluorbenzyl;i- denphthalid, das bei 148-151 C schmiltzt.
72,6 g o-Fluorbenzylidenphthalid, 55,0 g roter Phosphor und 450 ml Jodwasserstoffsäure (d= 1,75) werden in einer Argonatmosphäre 24 Stunden unter Rückllussbedingnngen erhitzt. Das Reaktionsgemisch wird abgekühlt, mit 500 ml Wasser versetzt und fil tariert. Der Rückstand wird mit 2n Natronlauge auf 80 C erwärmt und wieder filtriert. Die alkalische Lösung wird sauer gestellt, mit Methylenchlorid extrahiert, mit Wasser gewaschen, getrocknet und eingeengt. Der zurückbleibende rohe Feststoff wird aus Aceton/tiefsiedendem Petroläther umkristallisiert, Man erhält weisse Kristalle von 2-(o-Fluorphenyläthyl)-benzoesäure, die bei 93-94 C schmelzen.
280 g Polyphosphorsäure wird auf 1200 C erhitzt.
Es werden nun 70,0 g 2-(o-Fluorphenyläthyl)-benzov säure in einer Argonatmosphäre auf einmal hinzugefügt, und das Reaktionsgemisch wird anschliessend 3 Stunden bei 1500 C gehalten. Das heisse Reaktionsgemisch wird auf Eiswasser gegossen und mit Äther extrahiert. Die Ätherauszüge werden mit Wasser, 2n Na- triumhydroxyd und wiederum mit Wasser bis zu neutraler Reaktion gewaschen, über Natriumsulfat getrocknet, filtriert und eingeengt. Der Rückstand wird unter stark vermindertem Druck destilliert.
Man erhält ein gelbes Öl, das beim Stehen allmählich kristallisiert. Es wird
1-Fluor-10,11-dihydro-5H-dibenzo[a,d]cyclo hepten-5-on erhalten, das bei 64-65 C schmitzt.
Aus 1-Fluor-10,11-dihydro-5H-dibenzo[a,d]cyclo hepten-5-on erhält man in Analogie zu Beispiel 1 über
1-Fluor-10,11-dihydro-5-(3'-N-benzyl-N-methyl aminopropyl)-5-hydroxy-5H-dibenzo[a,d] cyclohepten (Schmelzpunkt 111-111,5 C) das
1-Fluor-10,11-dihydro-5-(3'-N-benzyl-N-methyl aminopropyl)-5 H-dibenzo .
Diese Verbindung kann in das entsprechende Hydrochlorid übergeführt werden, das bei 184-190 C schmilzt.

in which X is an ethylene group or a vinylene group optionally substituted by a halogen atom and Hal is chlorine or fluorine, or the corresponding 5-methylaminopropylidene compounds and the derivatives thereof bearing further chlorine or fluorine atoms in the aromatic rings, as well as acid addition salts of these compounds which are derived from distinguish the known representatives of the substance groups in question by the presence of a chlorine or fluorine atom in the 1-position, are distinguished by a considerably increased antidepressant effect and greatly reduced toxicity.
The lack of or only a slight anticholinergic effect should be mentioned as being particularly advantageous. These compounds are also characterized by diverse effects on the nervous system. Of the chlorine or fluorine substituents in the aromatic part, chlorine is preferred. The debenzylation according to the invention is expediently carried out by treatment with free hydrogen in the presence of a catalyst such as palladium, palladium on aluminum oxide, Raney nickel or platinum dioxide. The temperature is conveniently between about room temperature and about 800 C.

The reduction can conveniently be carried out with nasal hydrogen, e.g. Zinc in glacial acetic acid, run through. Another very suitable reducing agent is hydriodic acid.
By treating a compound of the formula III with hydriodic acid, in particular in the presence of red phosphorus, compounds of the formula II which are saturated in the 10, 11-position are obtained particularly smoothly. The reduction is preferably carried out in the presence of a solvent such as acetic acid or acetic anhydride and at a temperature between room temperature and the boiling point of the reaction mixture.
The dehydration of the compounds of the formula III leads to the corresponding compounds with an exocyclic double bond in 5-position, which, as stated, can also be used as starting materials in the process according to the invention. This dehydration is expediently carried out using mineral acids, such as hydrochloric or hydrobromic acid, it being possible to work in an anhydrous or aqueous medium. The dehydration is preferably carried out in ethanolic hydrochloric acid at a temperature between room temperature and the boiling point of the reaction mixture.

The separation methods are known per se. The geometric isomers are preferably separated by fractional crystallization of the acid addition salts from a solvent, e.g. B. acetone, or from a mixed solvent, e.g. B. methanol / diethyl ether.
As stated, the invention also covers the preparation of the acid addition salts of the present process products. Such salts are, for example, the few with organic acids such as oxalic acid, citric acid, acetic acid, lactic acid, maleic acid and tartaric acid, or with inorganic acids such as hydrochloric acid, hydrobromic acid or sulfuric acid.
The products of the process can be used as medicaments in the form of pharmaceutical preparations which contain them or their salts in a mixture with a pharmaceutical, organic or inorganic inert carrier material suitable for enteral or parenteral administration. The pharmaceutical preparations can be in solid form or in liquid form. They may contain auxiliaries, such as preservatives, stabilizers, wetting agents or emulsifiers, salts to change the osmotic pressure or buffers. They can also contain other thelapeutically valuable substances.
Example I.
2 g of 1-chloro-10,11-dihydro-5- (3'-N-benzyl) Nmethyl-aminopropyi) -5 H-dibenzo [a, d] cycloheptene are dissolved in 20 ml of methanol and 25 ml of conc. Dissolved hydrochloric acid. After adding 400 mg of activated charcoal and 5 ml of 5% palladium chloride solution, the mixture is at room temperature and 2 atm. Hydrogenated overpressure for about 20 hours. The mixture is then filtered and the filtrate is evaporated to dryness under reduced pressure. The residue is dissolved in water, made alkaline with dilute sodium hydroxide solution and extracted with ether. The ethereal solution is washed with water, dried over sodium sulfate and evaporated.
