Verfahren zur Herstellung basisch substituierter Heterocyclen
Die Erfindung betrifft ein Verfahren zur Herstellung von 11-basisch substituierten Dibenzo[b,f]- 1,4-thiazepinen der allgemeinen Formel:

sowie von Säure-Additionssalzen davon. In Formel I bedeutet Rt eine höchstens 3 C-Atome enthaltende Alkyl- oder Hydroxyalkylgruppe oder eine höchstens 6 C-Atome enthaltende Alkoxyalkyl- oder Alkoyloxyalkylgruppe. R2 stellt ein Wasserstoffatom oder ein Halogenatom, insbesondere ein Chlor-, Brom- oder Fluoratom, eine höchstens 3 C-Atome enthaltende Alkyl- oder Alkoxygruppe, die Dimethylaminosulfonylgruppe (-SO2N(CHs)2), die Methylsulfonylgruppe (-SO2CHS) oder die Nitrogruppe dar.
Die gewünschten Produkte werden erhalten, wenn man 11-basisch substituiertes Dibenzo[b,fi-1 ,4-thiaze- pin der allgemeinen Formel:

worin Rj und R2 die genannte Bedeutung haben, in einem nicht sauren Medium mit nicht wesentlich mehr als einem äquivalent eines Oxydationsmittels behang delt, welches weder selbst sauer ist noch unter den Reaktionsbedingungen Säure freisetzen kann.
Eine bevorzugte Ausführungsweise des beschriebenen Verfahrens besteht darin, dass man auf die Verbindungen der allgemeinen Formel II während einigen Stunden bis zu 3 Tagen bei Temperaturen zwischen Raumtemperatur und Rückflusstemperatur des Lösungsmittels das Oxydationsmittel einwirken lässt.
Besonders günstig ist die Oxydation mit Wasserstoffperoxyd, vorzugsweise einer 300/obigen wässrigen Lösung, in Gegenwart eines Alkohols, insbesondere Methanol und Äthanol, oder von Aceton. Die OxydsZ tion kann aber ebensogut auch erfolgen mit Calciumhypochlorit in Dioxan/Wasser oder mit organischen Hydroperoxyden, insbesondere tert. Butylhydroperoxyd, in Benzol und in Gegenwart eines Schwermetallkatalysators, vorzugsweise Vanadiumpentoxyd.
Die Isolation der erhaltenen Verbindungen der allgemeinen Formel I erfolgt auf an sich bekannte Weise, z. B. durch Extraktion, Ausfällung, Salzbildung usw.; ebenfalls auf an sich bekannte Weise, z. B. durch Umkristallisation, werden die Produkte gereinigt.
Die im erwähnten Verfahren als Ausgangsmaterialien benötigten Verbindungen der allgemeinen Formel II sind, soweit R2 ein Wasserstoff- oder Halogenatom oder eine höchstens 3 C-Atome enthaltende Alkyloder Alkoxygruppe darstellt, z. B. aus der britischen Patentschrift Nr. 980 853 bekannt und können nach den dort beschriebenen Verfahren hergestellt werden.
Die Herstellung der Verbindungen der allgemeinen Formel II, worin R2 die Dimethylaminosulfonyl-, Methylsulfonyl- oder Nitrogruppe bedeutet, kann in analoger Weise erfolgen.
Die erhaltenen Verbindungen der allgemeinen Formel I sind bei Zimmertemperatur feste, gegebenenfalls kristalline basische Verbindungen, die durch Umsetzen mit geeigneten anorganischen oder organischen Säuren in ihre Säure-Additionssalze übergeführt werden können. Hierfür haben sich als organische Säuren die Toluolsulfonsäure, Malonsäure, Bernsteinsäure, Sipfel- säure, Maleinsäure, Weinsäure etc. und als anorganische Säuren Halogenwasserstoffsäuren, Schwefelsäure, Salpetersäure, Phosphorsäure etc., als geeignet erwiesen.
Die 11-basisch substituierten Dibenzo [b,f]-1 ,4-thia- zepine der allgemeinen Formel I sowie ihre Säure-Additionssalze sind neue Verbindungen, die als Wirkstoffe in Arzneimitteln Verwendung finden sollen. Sie üben eine günstige Wirkung auf das Zentralnervensystem aus und fallen insbesondere als Analgetika, Sedativa und Antiemetika in Betracht.
Insbesondere diejenigen Verbindungen gemäss Formel I, worin R2 ein Wasserstoff- oder Halogenatom, eine höchstens 3 C-Atome aufweisende Alkyl- oder Alkoxygruppe oder die Nitrogruppe bedeutet, zeigen das für Neuroleptika typische Wirkungsbild. Dieses äussert sich pharmakologisch z. B. in einer Motilitäts; dämpfung, welche von einer kataleptischen und/oder apomorphinantagonistischen Wirkung begleitet sein kann, bei Mäusen bzw. Ratten. In dieser Wirkungsrichtung zeichnen sich insbesondere das 2-Chlor-1 1-(4-methyl-1- piperazinyl)-dibenzo [b,f] - 1,4-thiazepin-(4-oxyd) und 2-Methyl-1 1-(4-Methyl-1- piperazinyl)-dibenzo [b,f]1 ,4-thiazepin-(4-oxyd) sowie ihre Säure-Additionssalze aus.
Bei Verbindungen gemäss Formel I, worin R2 die Dimethylaminosulfonyl oder Methylsulfonylgruppe bedeutet, steht die antiemetische Wirkung im Vordergrund. Diese zeigt sich pharmakologisch z. B. in einer starken apomorphinantagonistischen Wirkung bei Hunden und Ratten sowie einer im Vergleich zur apomorphinantagonistischen Wirkung geringen kataleptischen und motilitätsdämpfenden Wirkung.
Die Wirkungsstärke der erfindungsgemässen Verbindungen ist vergleichbar mit derjenigen der entsprechenden tertiären Amine, doch hat sich überraschend gezeigt, dass die N-Oxyde gemäss allgemeiner Formel I erheblich weniger toxisch sind als die tertiären Amine und demnach eine weitaus grössere therapeutische Breite besitzen.
In den nachfolgenden Beispielen, welche die Ausführung des Verfahrens erläutern, die Erfindung aber in keiner Weise einschränken sollen, erfolgen alle Temperaturabgaben in Celsiusgraden und sind korrigiert.
Beispiel 1
6,88 2-Chlor-ll-(4-methyl-i- piperazinyl)-dibenzo[b,f]1,4-thiazepin (Smp. 117-120 C) werden mit 2,2 ml 30 O/oiger wässriger Wasserstoffperoxyd-Lösung in 30 ml 95 0/oigem Äthanol während 3 1/2 Stunden auf Rückfluss; erhitzt.
Nach dem Erkalten wird das Reaktionsgemisch mit 0,3 g 50/oigem Palladium-Kohlepulver während 15 Minuten gerührt und anschliessend während 15 Minuten gekocht. Nach Abtrennung des Katalysators durch Filtrieren und Nachwaschen mit Methanol wird das Filtrat mit 50 ml Wasser versetzt und im Vakuum bis auf einen zähflüssigen Sirup eingeengt. Dieser wird zwischen Äther und Wasser verteilt, wobei massive Kristallisation eintritt. Die Kristalle werden abgetrennt und mit Äther und Wasser gewaschen. Man erhält 2-Chlor-1 1-(4-methyl-1piperazinyl)-dibenzo[b,f] 1,3 -thiazepin-(4-oxyd) in Form von farblosen Prismen, welche sich ab 2350 C alngsam und bei 245-2480 C rasch zersetzen.
Beispiel 2
5,7 g 2-Methyl-11 (4-methyl-1- piperazinyl)-dibenzo [b,f]-1,4-thiazepin (Smp. 99-1070 C) werden mit 1,95 ml 30 0/obiger wässriger Wasserstoffperoxydlösung in 27 ml 95 0/oigem Äthanol während 3 1/2 Stunden auf Rückfluss erhitzt.
Nach dem Erkalten wird das Reaktionsgemisch mit 0,3 g 5 0/oigem Palladium-Kohlepulver während 15 Minuten gerührt und anschliessend während 15 Minuten gekocht. Nach Abtrennung des Katalysators durch Filtrieren und Nachwaschen mit Methanol wird das Filtrat mit 50 ml Wasser versetzt und im Vakuum eingedampft. Der Eindampfrückstand wird durch systematisches Ausschütteln zwischen Äther und Wasser verteilt und die wässrigen Phasen werden zur Trockne eingedampft. Der Rückstand wird aus Aceton/Äther/Petrol äther kristallisiert, wobei man 2-Methyl-1 1-(4-methyl-1- piperazinyl)-dibenzo [b,f] -1,4- thiazepin-(4-oxyd)-Dihydrat in Form von feinen Nadeln vom Schmelzpunkt 183-186 C erhält.

R2 represents a hydrogen atom or a halogen atom, in particular a chlorine, bromine or fluorine atom, an alkyl or alkoxy group containing at most 3 carbon atoms, the dimethylaminosulfonyl group (-SO2N (CHs) 2), the methylsulfonyl group (-SO2CHS) or the nitro group represent.
The desired products are obtained if 11-basic substituted dibenzo [b, fi-1, 4-thiazepine of the general formula:

where Rj and R2 have the meaning given, in a non-acidic medium with not significantly more than one equivalent of an oxidizing agent which is neither acidic itself nor can release acid under the reaction conditions.
A preferred embodiment of the process described consists in allowing the oxidizing agent to act on the compounds of the general formula II for a few hours up to 3 days at temperatures between room temperature and the reflux temperature of the solvent.
Oxidation with hydrogen peroxide, preferably a 300% above aqueous solution, in the presence of an alcohol, in particular methanol and ethanol, or of acetone, is particularly favorable. The OxydsZ tion can also take place just as well with calcium hypochlorite in dioxane / water or with organic hydroperoxides, especially tert. Butyl hydroperoxide, in benzene and in the presence of a heavy metal catalyst, preferably vanadium pentoxide.
The compounds of general formula I obtained are isolated in a manner known per se, for. B. by extraction, precipitation, salt formation, etc .; also in a manner known per se, e.g. B. by recrystallization, the products are purified.
The compounds of the general formula II required as starting materials in the process mentioned are, if R2 represents a hydrogen or halogen atom or an alkyl or alkoxy group containing at most 3 carbon atoms, e.g. B. from British Patent No. 980 853 and can be prepared by the methods described there.
The compounds of the general formula II in which R2 is the dimethylaminosulfonyl, methylsulfonyl or nitro group can be prepared in an analogous manner.
The compounds of general formula I obtained are solid, optionally crystalline, basic compounds at room temperature which can be converted into their acid addition salts by reaction with suitable inorganic or organic acids. For this purpose, toluenesulphonic acid, malonic acid, succinic acid, sipulic acid, maleic acid, tartaric acid etc. have proven to be suitable as organic acids and hydrohalic acids, sulfuric acid, nitric acid, phosphoric acid etc. as inorganic acids.
The 11-base substituted dibenzo [b, f] -1, 4-thiazepines of the general formula I and their acid addition salts are new compounds which are to be used as active ingredients in medicaments. They have a beneficial effect on the central nervous system and are particularly suitable as analgesics, sedatives and antiemetics.
In particular, those compounds according to formula I in which R2 is a hydrogen or halogen atom, an alkyl or alkoxy group containing at most 3 carbon atoms or the nitro group show the pattern of action typical of neuroleptics. This is expressed pharmacologically z. B. in a motility; attenuation, which can be accompanied by a cataleptic and / or apomorphine-antagonistic effect, in mice or rats. The 2-chloro-1 1- (4-methyl-1-piperazinyl) -dibenzo [b, f] -1,4-thiazepine (4-oxide) and 2-methyl-1 1- (4-Methyl-1-piperazinyl) -dibenzo [b, f] 1,4-thiazepine- (4-oxide) and their acid addition salts from.
In the case of compounds according to formula I in which R2 is the dimethylaminosulfonyl or methylsulfonyl group, the antiemetic effect is in the foreground. This is shown pharmacologically z. B. in a strong apomorphine antagonistic effect in dogs and rats and in comparison to the apomorphine antagonistic effect, low cataleptic and motility-suppressing effect.
The potency of the compounds according to the invention is comparable to that of the corresponding tertiary amines, but it has surprisingly been found that the N-oxides according to general formula I are considerably less toxic than the tertiary amines and therefore have a much greater therapeutic range.
In the following examples, which explain how the process is carried out but are not intended to restrict the invention in any way, all temperatures are given in degrees Celsius and are corrected.
