Die Erfindung betrifft ein Verfahren zur Herstellung von neuen Aminoketonen der Formel I

sowie von ihren physiologisch verträglichen Säureaddi tionssalzen mit anorganischen oder organischen Säuren.
In der obigen Formel bedeuten
Hal ein Chlor- oder Bromatom,
R1 ein Wasserstoff- oder Chlor- oder Bromatom, Ro und Rs, die gleich oder verschieden sein können,
Wasserstoffatome, geradkettige oder verzweigte
Alkylreste, Alkenyl-, Alkinyl-, Hydroxyalkyl-, Alk oxyalkyl-, Dialkylaminoalkyl-, Cycloalkyl-, Phenyl-,
Benzyl- oder Adamantylreste oder zusammen mit dem Stickstoffatom einen gegebenenfalls durch nie dere Alkylreste oder durch Phenylreste substituierten
Pyrrolidino-, Piperidino-, Hexamethylenimino-, Pi perazino-, Morpholino- oder Camphidinorest, R4 und R5, die gleich oder verschieden sein können,
Wasserstoffatome, Alkylreste mit 1 bis 3 Kohlen stoffatomen oder Phenylgruppen und n 0, 1 oder 2.
Die neuen Verbindungen werden hergestellt, indem man Mono- oder Diacylamino-phenylaminoalkylketone der Formel II

worin mindestens einer der Reste Rg und R7 eine beliebige Acylgruppe und der andere Rest gegebenen falls ein Wasserstoffatom bedeutet, entacyliert.
Die Entacylierung erfolgt beispielsweise durch Erhit zen mit verdünnten Mineralsäuren.
Die Ausgangsverbindungen der Formel II lassen sich beispielsweise durch Umsetzung der entsprechend substituierten Phenyl-halogenalkyl-ketone mit Aminen gewinnen.
Nach der genannten Methode wurden beispielsweise folgende Ausgangsstoffe hergestellt: 2-Acetamino-3 ,5-dibrom-o-dipropylamino-aceto- phenon-hydrochlorid, Fp.: 1870 C (Zers.);

Die erhaltenen Verbindungen der Formel I können gegebenenfalls über ihre Basen, falls sie in Form ihrer Säureadditionssalze anfallen, mit beliebigen anorganischen oder organischen Säuren in ihre physiologisch verträglichen Säureadditionssalze überführt werden, beispielsweise durch Umsetzung mit einer alkoholischen Lösung der betreffenden Säure. Als Säuren haben sich beispielsweise Salzsäure, Bromwasserstoffsäure, Schwe felsäure, Phosphorsäure, Milchsäure, Zitronensäure, Weinsäure, Maleinsäure oder Fumarsäure als geeignet erwiesen. Es lassen sich Salze mit einem, zwei oder falls im Molekül drei basische Reste vorhanden sind, auch mit drei Äquivalenten der betreffenden Säure herstellen. Die erhaltenen Salze sind wasserlöslich.
Die erfindungsgemäss hergestellten Verbindungen sowie ihre Salze besitzen wertvolle pharmakologische Eigenschaften. So weisen sie je nach den vorhandenen Substituenten eine antiphlogistische und Kreislauf-Wirkung sowie zentralnervöse Eigenschaften auf, die in einer analgetischen, antipyretischen, sedativen und antikonvulsiven oder auch in einer zentral erregenden Wirkung bestehen können.
Die pharmakologische Wirksamkeit von
A = 4-Amino-3,5-dibrom-ss-propylamino isobutyrophenon-hydrochlorid wurde im Vergleich zu
B = N-(3 -Dimethylamino-propyl)-3 -chlorpheno- thiazin-hydrochlorid (Chlorpromazin) und
C = 4-Amino-3 ,5-dibrom-ss-propylamino propiophenon-hydrochlorid im Vergleich zu
D = 2-Methyl-3-(o-tolyl)-4(3H)-chinazolon (Methaqualon) und
E = 4-Amino-3-brom-a-tert.-butylamino butyrophenon-hydrochlorid im Vergleich zu F 1 ,2-Diphenyl-3 ,5-dioxo-4-(n-butyl)- pyrazolidin (Phenylbutazon) geprüft.
1. Sedierende Wirkung:
Die sedierende Wirkung der Substanzen A, B, C und D an der Maus wurde mit Hilfe lichtelektrischer Schranken als eine die Spontanmotilität hemmende Wirkung bestimmt.
Die Prüfung erfolgte in Anlehnung an methodische Angaben von Dews [Brit. J. Pharmacol. 8, 46 (1953)] mit Hilfe eines besonderen trommelförmigen Zählkäfigs, in dessen Boden 10 Photozellen eingebaut waren, deren Abdeckung durch die über den Boden hinweglaufenden Mäuse fortlaufend registriert wurde.
Für diese Untersuchungen wurden verschiedene Substanzmengen 15 Minuten (Substanz A und B) bzw.
60 Minuten (Substanz C und D) vor Beginn der Registrierung an Kollektiven von je n weissen männlichen Mäusen mit einem durchschnittlichen Körpergewicht von 20 g per Schlundsonde verabfolgt und die Motilität der Mäuse über einen Zeitraum von 30 Minuten hinweg gemessen.
Die Tabelle 1 gibt die Wirkung der Substanz A und B, die Tabelle 2 die Wirkung der Substanz C und D auf die Spontanmotilität der Mäuse wieder.

Aus der prozentualen Abnahme der Motilität nach den verschiedenen Dosen der zu untersuchenden Substanzen gegenüber den Kontrollen wurde durch graphische Extrapolation für jede Substanz die Dosis ermittelt, die eine ziege Hemmung der Spontanmotilität bewirkt, das Ergebnis zeigt folgende Tabelle:

Die antiphlogistische Wirkung der Substanz E und F wurde als antiexsudative Wirkung gegenüber dem Kaolinödem der Rattenhinterpfote geprüft.
Zur Auslösung des Ödems diente entsprechend den Angaben von Hillebrecht [Arzneimittel-Forschung 4, 607 (1954)] eine subplantare Injektion von 0,05 ml einer lOSigen Suspension von Kaolin in 0,85 %oder wässriger NaCl-Lösung. Die Messung der Pfotendicke wurde mit Hilfe einer von Doepfner und Cerletti [In.
Arch. Allergy Immunol. 12, 89 (1958)] angegebenen Technik vorgenommen.
Für die Untersuchungen wurden je n männliche weisse Ratten mit einem durchschnittlichen Körpergewicht von 145 g verwendet. Die auf ihre antiexsudative Wirkung zu prüfenden Substanzen E und F wurden 30 Minuten vor Auslösung des Ödemes als Verreibung in Tylose in verschiedenen Dosen per Schlundsonde beigebracht. 5 Stunden nach Auslösung des Ödemes wurde die zur Bewertung einer antiexsudativen Wirkung als Grundlage dienende Messung vorgenommen. Die gemittelten Schwellungswerte der mit Substanz behandelten Tiere wurden mit denen der scheinbehandelter Kontrolltiere verglichen.
Für die einzelnen Dosen der zu prüfenden Substanzen E und F ergab sich folgende Hemmung der Schwellung:

Durch graphische Extrapolation wurde aus den mit den verschiedenen Dosen der Substanzen E und F erzielten prozentualen Hemmwerten die Dosis ermittelt, die zu einer 35 %igen Abschwächung (ED35) der Schwellung führte:

Die akute Toxizität der Substanzen A bis F wurde in Gruppen zu je 10 weissen männlichen Mäusen mit einem durchschnittlichen Körpergewicht von 20 g bestimmt.
Es wurde die LDjo, die Dosis bei deren peroralen Verabreichung 50 so der Tiere innerhalb von 7 Tagen verstarben, nach der Methode von Litchfield und Wilcoxon berechnet, dabei wurden folgende Werte erhalten:

Die nachstehenden Beispiele sollen die Erfindung näher erläutern:
Beispiel I 4-Amino-2-chlor-a-diäthylamino-acetophenon
1 g 4-Acetamino.2-chlor-a-diäthylamino-acetophenon wird mit 10 ml 6n Salzsäure 5 Minuten gekocht; man stellt die Lösung mit Natronlauge alkalisch und extrahiert mit Chloroform. Die Chloroformlösung wird mit Natriumsulfat getrocknet und über wenig Aktivkohle filtriert.
Mit isopropanolischer Salzsäure fällt man das Hydrochlorid des 4 - Amino - 2 - chlor - a - diäthylamino- acetophenons aus und kristallisiert aus Isopropanol um; Fp.: 1500 C (Zers.).

Beispiel 2
3-Amino-4,6-dibrom-ss-propylamino-propiophenon
7,0 g 3-Acetamino-4,6-dibrom-ss-propylamino-propiophenon werden in 70 ml 3n-isopropanolischer Salzsäure und 20 ml Wasser gelöst. Man kocht 3 Stunden unter Rückfluss, bringt dann im Vakuum zur Trockene und verreibt den Rückstand mit Isopropanol. Das abgesaugte Hydrochlorid der gewünschten Verbindung schmilzt nach Umkristallisieren aus Methanol bei 139 bis 1410 C (Zers.).
Beispiel 3 2-Amino-5-brom-α-piperidino-propiophenon
26,1 g 2-Acetylamino-5 -brom-a-piperidino-propio- phenon werden mit 250 ml 2n Salzsäure 1 Stunde auf dem Dampfbad erhitzt. Man engt im Vakuum ein und kristallisiert den schaumigen Rückstand durch Verreiben mit Aceton. Nach Umkristallisieren aus Isopropanol zersetzt sich das Hydrochlorid des 2-Amino-5-brom-apiperidino-propiophenons bei 202-203 C (Dunkelfärbung ab etwa 180 C).
Analog wird erhalten:
2-Aminoa-diäthylamino-3 ,5-dibrom-butyro phenon, Fp. des Hydrochlorids: 138-142 C, hergestellt aus 2-Acetylamino-a-diäthylamino 3,5-dibrom-butyrophenon.
Beispiel 4
4-Amino-3 -brom-5-chlor-a-methyl- ss-morpholino-propiophenon
5,0 g 4-Acetylamino-3-brom-5-chlor-α-methyl-ss- morpholino-propiophenon werden mit 50 ml 6n Salzsäure 5 Minuten lang auf 600 C erhitzt. Nach Abkühlen stellt man die Lösung mit Natronlauge alkalisch, extrahiert mit Chloroform, trocknet die Chloroformlösung, fällt mit äthanolischer Salzsäure das 4-Amino-3-brom 5-chlor-α-methyl-ss-morpholino-propiophenon-hydro- chlorid aus und kristallisiert aus Methanol/Isopropanol um. Fp.: 204-206 C.

The resulting compounds of the formula I can, if appropriate, via their bases, if they are obtained in the form of their acid addition salts, be converted into their physiologically acceptable acid addition salts with any inorganic or organic acids, for example by reaction with an alcoholic solution of the acid in question. For example, hydrochloric acid, hydrobromic acid, sulfuric acid, phosphoric acid, lactic acid, citric acid, tartaric acid, maleic acid or fumaric acid have proven suitable as acids. 1. Sedative effect:
The sedative effect of substances A, B, C and D on the mouse was determined with the aid of photoelectric barriers as an effect which inhibits spontaneous motility.
The test was carried out on the basis of methodological information from Dews [Brit. J. Pharmacol. 8, 46 (1953)] with the help of a special drum-shaped counting cage, in the bottom of which 10 photocells were installed, the coverage of which was continuously registered by the mice running over the floor.
For these investigations, different amounts of substance were required for 15 minutes (substance A and B) or
60 minutes (substance C and D) before the start of registration, it was administered to collectives of n white male mice each with an average body weight of 20 g by gavage and the motility of the mice was measured over a period of 30 minutes.
Table 1 shows the effect of substances A and B, and Table 2 shows the effect of substances C and D on the spontaneous motility of the mice.

The anti-inflammatory effects of substances E and F were tested as anti-exudative effects against kaolin edema of the rat hind paw.
To trigger the edema, according to the information provided by Hillebrecht [Arzneimittel-Forschung 4, 607 (1954)], a subplantar injection of 0.05 ml of a loose suspension of kaolin in 0.85% or aqueous NaCl solution was used. The measurement of the paw thickness was carried out using a method developed by Doepfner and Cerletti [In.
Arch. Allergy Immunol. 12, 89 (1958)].
For the studies, n male white rats with an average body weight of 145 g were used. Substances E and F to be tested for their antiexudative effect were introduced as trituration in Tylose in various doses by gavage 30 minutes before the edema was triggered. 5 hours after the edema was triggered, the measurement serving as the basis for evaluating an anti-exudative effect was carried out. The mean swelling values of the substance-treated animals were compared with those of the sham-treated control animals.

Example 2
3-amino-4,6-dibromo-ss-propylamino-propiophenone
7.0 g of 3-acetamino-4,6-dibromo-ss-propylamino-propiophenone are dissolved in 70 ml of 3N-isopropanolic hydrochloric acid and 20 ml of water. It is refluxed for 3 hours, then brought to dryness in vacuo and the residue is triturated with isopropanol. The extracted hydrochloride of the desired compound melts after recrystallization from methanol at 139 to 1410 C (decomp.).
Example 3 2-Amino-5-bromo-α-piperidino-propiophenone
26.1 g of 2-acetylamino-5-bromo-a-piperidino-propiophenone are heated with 250 ml of 2N hydrochloric acid on the steam bath for 1 hour. It is concentrated in vacuo and the foamy residue is crystallized by trituration with acetone. After recrystallization from isopropanol, the hydrochloride of 2-amino-5-bromo-apiperidino-propiophenone decomposes at 202-203 ° C. (darkening from about 180 ° C.).
