In den Verbindungen der allgemeinen Formel I und den zugehörigen, weiter unten genannten Ausgangsstoffen ist R1 beispielsweise durch Alkylgruppen, wie die Methyl-, Äthyl-, n-Propyl-, Isopropyl-, n-Butyl-, Isobutyl-, sek. Butyl-, n-Pentyl-, Isopentyl-, n-Hexyl-, n-Heptyl-, n-Octyl-, n-Nonyl-, oder n-Decylgruppe; durch Alkenylgruppen, wie die Allyl-, Crotyl-, l-Methylallyl- oder 2 -Methylallylgruppe;
Process for the preparation of new piperidine derivatives
The present invention relates to a process for the preparation of new piperidine derivatives with valuable pharmacological properties.
It has surprisingly been found that piperidine derivatives of the general formula I,
EMI1.1
in which R1 is an aliphatic hydrocarbon radical with a maximum of 10 carbon atoms, the cinnamyl group, an unsubstituted or on the benzene nucleus by halogen up to atomic number 35 or lower alkyl or
Alkoxy-substituted phenyl (lower alkyl) group with a maximum of 12 carbon atoms, in which the phenyl group can be connected to the lower alkyl group instead of directly via oxygen, the carbonyl group, alkylimino group or alkenylimino group, and R2 and R3 denote an alkyl group with 2-4 Carbon atoms and their addition salts with inorganic and organic acids mean valuable pharmacological properties,
in particular have analgesic as well as antitussive and partly also centrally damping effectiveness with a favorable therapeutic index.
The analgesic activity of the compounds of general formula I and their salts, e.g. the fumarates of 1- [1- (3-phenylpropyl) -4-propyl-4-piperidyl] -1-butanone and of 1 - [- cinnamyl-4-propyl-4-piperidyl] -1-butanone eg by measuring the prolongation of the reaction time caused by their intraperitoneal or oral administration to mice when the tail is irritated by thermal radiation according to the experimental arrangement by H. Friebel and Cl. Reichle, Arch. Exp.
Path. and Pharmakol. 226, 551 (19i5). According to the test results, the compounds of general formula I with the n-propyl group as R5 and their salts are strong analgesics, those with other groups R3, such as the ethyl and n-butyl groups, are mild to moderate analgesics.
The antitussive effectiveness of the abovementioned and other compounds of general formula I is evident, for example, when administered intravenously to cats using the R. Domenjoz method. Arch. Exp. Path.
and Pharmakol. 215: 19-24 (1952).
Another method to prove the antitussive effectiveness is the determination of the inhibition of the irritable cough caused by sulfur dioxide caused by the subcutaneous or oral administration of the test substances in guinea pigs: in a preliminary experiment for the selection of the test animals, male guinea pigs are given an SO2-CO2 gas flowing through at atmospheric pressure in a plexiglass chamber. Air mixture with a constant mixing ratio of 20 ml: 1.5 liters: 10.5 liters per minute until the onset of the cough or a maximum of 120 seconds. The assessment of the onset of a cough is carried out by inspection. The guinea pigs that react with coughs (approx. 2/3 of all animals) are combined in groups of 6 animals each.
Such test animal groups receive the test substance in various doses suitable for determining the ED 50 in mg / kg administered subcutaneously or orally about 24 hours after the preliminary test. The irritant gas exposure takes place in the same way as in the preliminary test after 30 u. after 90 minutes after application of the test substances. The onset of a cough is assessed again by inspection. From the percentages of animals no longer reacting to SO3 determined at different doses, the dose preventing coughing in 50% of the animals = ED30 is determined by graphical interpolation with the probability network Schleicher and Schüll 29872.
The new piperidine derivatives of the general formula I and their pharmaceutically acceptable acid addition salts are suitable as active ingredients for pharmaceutical preparations for the relief and elimination of pain of various origins and of coughing stimuli. Administration can take place orally, rectally or parenterally.
In the compounds of general formula I and the associated starting materials mentioned below, R1 is, for example, represented by alkyl groups such as methyl, ethyl, n-propyl, isopropyl, n-butyl, isobutyl, sec. Butyl, n-pentyl, isopentyl, n-hexyl, n-heptyl, n-octyl, n-nonyl, or n-decyl group; by alkenyl groups, such as the allyl, crotyl, 1-methylallyl or 2-methylallyl group;
by the benzyl group, the p-fluorine, o-, m- or p-chlorine, p-bromine, 3,4-dichloro, p-methyl, p-isopropyl, p-methoxy, p- Ethoxy, p-isopropoxy, 3,4-dimethoxy, 3,4,5-trimethoxy, benzyl, the unsubstituted 2-phenylethyl, 3-phenylpropyl, 2-phenylpropyl, 4-phenylbutyl, 2-phenoxyethyl, 3-phenoxypropyl, 4-phenoxybutyl, 2-benzoylethyl, 3-benzolpropyl group or by the corresponding groups substituted in the benzene nucleus analogously to the aforementioned benzyl groups; by methyl, ethyl, n-propyl, which in their position by the anilino, o-toluidino, m-toluidino, p-toluidino, 3,4-xylidino or 2,6-xylidino Group are substituted.
New piperidine derivatives of the general formula I and their acid addition salts are prepared by relying on a compound of the general formula II,
EMI2.1
in which R1 and R have the meaning given under formula I, R3 is the methyl, ethyl or propyl group, Y is a lower alkyl group and Z is the group -COO-Y or the cyano group, allows an acid to act and if desired from an acid addition salt obtained a compound of general formula I is released and this or the salt initially obtained is converted into another addition salt with an inorganic or organic acid.
For example, you can concentrate on a compound of general formula II. Mineral acids, e.g. conc.
Hydrochloric acid or conc. Hydrobromic acid or sulfuric acid act at temperatures of 600 to 1500C, the required reaction temperature and duration can easily be determined from the evolution of carbon dioxide.
For the preparation of the starting materials of the general formula II one uses, for example, an acid halide of the general formula III,
EMI2.2
in which Hal is chlorine or bromine and R1 and R2 have the meaning given under formula I, with an alkali metal or an alkoxymagnesium compound of a malonic acid diester or cyanoacetic acid ester of the general formula formula IV
EMI2.3
in which Y, Z and R5, have the meaning given under formula II, where the solvent e.g.
Ether, ethanol, methanol or benzene are used.
If desired, the piperidine derivatives of the general formula I obtained by the process according to the invention are then converted in the customary manner into their addition salts with inorganic and organic acids. For example, a solution of a piperidine derivative of the general formula 1 in an organic solvent, such as diethyl ether, methanol or ethanol, is mixed with the acid or a solution thereof desired as the salt component and this is separated immediately or after adding a second organic liquid, such as e.g. Diethyl ether to methanol, precipitated salt.
For use as active ingredients for drugs, pharmaceutically acceptable acid addition salts can be used instead of free bases, i. Salts with acids whose anions show either no pharmacological effect or a desired one at the dosages in question. It is also advantageous if the salts to be used as active ingredients can be readily crystallized and are not or only slightly hygroscopic.
For salt formation with piperidine derivatives of the general formula I, e.g. Hydrochloric acid, hydrobromic acid, sulfuric acid, phosphoric acid, methanesulphonic acid, ethanesulphonic acid, p-hydroxyethanesulphonic acid, acetic acid, malic acid, tartaric acid, citric acid, lactic acid, succinic acid, fumaric acid, maleic acid, benzoic acid, salicylic acid, phenylacetic acid, mandelic acid, phenylacetic acid, mandelic acid, emaphamic acid, 1,5-naphthalenesulphonic acid, phenylacetic acid, emaphamic acid, emaphamic acid, 1,5-naphthosulphonic acid, phenylacetic acid will.
The following example explains the preparation of the new compounds of general formula I.
The temperatures are given in degrees Celsius.
example
In a four-necked flask, 28.2 g of ethyl malonic acid diethyl ester with 3.45 g of finely divided sodium are dissolved in 900 ml of abs for 15 hours. Ether stirred. 17.3 g of 1 - (2-phenylethyl-4-propyl-isonipecotinoyl chloride hydrochloride are added to the thick suspension of sodium ethyl malonic acid diethyl ester and the mixture is heated first at room temperature and then under reflux for a further hour 5N hydrochloric acid is decomposed, the ether phase is separated off, extracted once more with ether and then the aqueous phase is made alkaline with concentrated ammonia. It is extracted with ether, and after evaporation the (1-phenylethyl-4-propyl-4-piperidyl) 8.9 g of the crude product mentioned above are mixed with 20 ml of concentrated hydrochloric acid and refluxed for 2 hours.
Then it is evaporated in vacuo, the residue taken up in a little water, with conc.
Ammonia made alkaline and extracted with ether.
It is dried, evaporated and distilled in an air bath at 0.02 torr and 135-145. A silica gel is optionally chromatographed in chloroform-methanol 97: 3. The base is then converted into the fumarate in the usual way. The 1- [1- (2-phenylethyl) -4-propyl-4-piperidyl] -1-butanone fumarate melts at 1760.
The following were prepared in the same way: 1 - [1 - (3-phenyl-propyl) -4-propyl-4-piperidyl] -1-butanone-fumarate, m.p. 172-173, 1 - (1-cinnamyl-4-propyl -4-piperidyl] -1-butanone fumarate, m.p. 178-179 and 1- [1- (2-phenoxyethyl) -4-propyl-1-piperidyl] -1-butanone fumarate, m.p. 159 -160.
