Die erhaltenen Basen können in ihre Säureadditions- oder quaternären Salze überführt werden.
Die Herstellung der für das erfindungsgemässe Verfahren benötigten Ausgangsmaterialien kann nach an sich bekannten Methoden geschehen.
Gegebenenfalls substituierte Thioxanthone können z. B. durch Kondensation von gegebenenfalls substitu- ierter Thiosalicylsäure mit gegebenenfalls substituiertem Benzol gewonnen werden.
Die Herstellung der aliphatischen, eine Ätherfunktion aufweisenden metallorganischen Verbindungen lässt sich ebenfalls nach bereits vorbekannten Methoden durchführen. Die für eine bevorzugte Ausführungsform benötigten Magnesiumverbindungen von Alkoxy-alkylhalogeniden lassen sich sich z. B. durch direkten Umsatz von Magnesium mit einer ätherischen Lösung des Alkoxyalkylhalogenides gewinnen, wobei die Umsetzung zweckmässigerweise durch Zusatz von Methyljodid oder Äthylbromid angeregt wird. Beispiele für Alkylhalogenide, welche eine Alkoxygruppe tragen können, sind Propylhalogenide, Butylhalogenide, Amylhalogenide, Hexylhalogenide, Heptylhalogenide.
Die Alkylreste können gerade oder verzweigt sein. Die Alkoxygruppe ist zweckmässigerweise eine niedere Alkoxygruppe wie Methoxy-, Äthoxy-, Propoxy, Butoxy, welche sich z. B. in Endstellung des Alkylhalogenides befindet. co-Methoxy-alkylmagnesiumverbindun- gen sind bevorzugte Ausgangsmaterialien. Die Ätherfunktion kann neben Alkoxy z. B. auch Aralkoxy bedeuten, wie Benzyloxy oder Phenäthyloxy, oder auch Tetrahydropyranyloxy.
Für die Durchführung des erfindungsgemässen Verfahrens lassen sich insbesondere magnesium-, lithium- und zinkorganische Metallverbindungen verwenden. In einer bevorzugten Ausführungsform werden die Ausgangsketone der allgemeinen Formel I entweder in fester, feingepulverter Form oder in einem indifferenten organischen Lösungsmittel, wie z. B. absolutem Äther, Benzol, Tetrahydrofuran, zu der in einem derartigen organischen indifferenten Lösungsmittel befindlichen Magnesiumverbindung eines eine Ätherfunktion aufweisenden Alkylhalogenides zugegeben. Nach beendeter Umsetzung wird das Reaktionsprodukt der Hydrolyse unterworfen. Besonders vorteilhaft gestaltet sich die Zerlegung der entstandenen organischen Metallverbindungen unter praktisch neutralen Bedingungen, z. B. durch Hydrolyse in wässriger Ammoniumchloridlösung.
Auf diese Weise werden die tertiären Carbinole der untenstehenden allgemeinen Formel II gebildet, welche durch Behandlung mit geeigneten, mit Wasser nicht mischbaren organischen Lösungsmitteln, wie z. B. Äther, Essigsäureäthylester, Chloroform, Methylenchlorid, von den Nebenprodukten der Reaktion abgetrennt und isoliert werden können. Die tertiären Carbinole stellen feste, meist gut kristallisierbare Verbindungen dar und besitzen folgende Formel

worin A einen mehr als zwei Kohlenstoffatome aufweisenden aliphatischen Rest, der als Substituent eine Äthergruppe trägt, bedeutet. Diese Verbindungen können wie gesagt, in den aromatischen Kernen gegebenenfalls durch Alkyl-, Trifluormethyl-, Aralkyl-, Aryl-, Alkoxy-, Aralkoxy-, Aryloxy-, Alkylthio-, Aralkylthio-, AryIthio-, Halogen-, Acyl-, Amino-, Hydroxy- und Carboxygruppen substituiert sein.
Allenfalls vorhandene Acyl-, Amino-, Hydroxylund Carboxylgruppen müssen vor dem Umsatz mit der metallorganischen Verbindung geschützt werden, um eine Reaktion dieser Gruppen mit dem metallorganischen Reagens zu verhüten. Solche Schutzgruppen können eingeführt werden, z. B. durch Ketalisierung der Acylgruppe (z. B. Bildung des Acetyl-monoketals), durch Acylierung der Aminogruppe (z. B. Bildung des Acet- oder Benzamides) oder durch Veresterung der Hydroxyl- oder Carboxylgruppen (z. B. Bildung der Acetoxyverbindung einerseits oder der Carbomethoxyoder Carbäthoxyverbindung andererseits). Die Abspaltung der Schutzgruppen erfolgt meist gleichzeitig mit der hydrolytischen Zerlegung der gebildeten organischen Metallverbindungen.
In einer weiteren Reaktionsfolge wird die Ätherfunktion des oben erhaltenen Hydrolyseproduktes der Formel II, unter gleichzeitiger Dehydratisierung des Carbinols, durch Halogen ersetzt, wobei Halogen-Derivate der untenstehenden allgemeinen Formel II erhalten werden. Diese Reaktionsfolge wird mit Vorteil gemäss zwei bevorzugten Ausführungsformen durchgeführt: a) Das Hydrolyseprodukt wird mit Bromwasserstoff behandelt, zweckmässig mit einer konzentrierten wässrigen Lösung von Bromwasserstoff bei Siedehitze, wobei die entsprechende Bromverbindung entsteht.
b) Das Hydrolyseprodukt wird mit Phosphoroxychlorid behandelt, was mit Vorteil bei Rückflusstemperatur geschieht. Dabei entsteht die entsprechende Chlorverbindung. Die Halogenverbindungen der Formel III sind meist viscose, z. T. kristallisierbare Substanzen, welche im Hochvakuum unzersetzt destilliert werden können und besitzen folgende Formel

Diese Verbindungen können in den aromatischen Kernen gegebenenfalls durch Alkyl-, Trifluormethyl-, Aralkyl-, Aryl-, Alkoxy-, Aralkoxy-, Aryloxy-, Alkylthio-, Aralkylthio-, Arylthio-, Halogen-, Acyl-, Amino-, Hydroxy- und Carboxygruppen substituiert sein.
In einer weiteren Reaktionsstufe werden die oben erhaltenen Halogenverbindungen der Formel III mit gegebenenfalls durch Hydroxy- oder weitere Aminogruppen substituierten primären oder sekundären Aminen umgesetzt, wobei die in 9-Stellung den zweiwertigen Rest D tragenden Thioxanthenverbindungen entstehen. Geeignete Amine sind z. B. Monoalkylamine oder Dialkylamine, wie z. B. Methylamin, Äthylamin, Dimethylamin, Diäthylamin. Bei Verwendung von Monoalkylaminen gelangt man zu therapeutisch besonders interessanten neuen Verbindungen. Der Alkylrest kann auch durch einen Cycloalkylrest ersetzt sein, wie im Falle des Methylcyclopentylamins. Die sekundäre Aminogruppe kann auch Glied eines gesättigten heterocyclischen Ringsystems sein. Beispiele solcher cyclischen Amine sind Piperidin, Pyrrolidin, Morpholin.
Gegebenenfalls können wie gesagt, die zur Umsetzung verwendeten primären oder sekundären Amine noch Hydroxylgruppen oder zusätzliche Aminogruppen aufweisen. Beispiele solcher Amine sind 2-Amino-5diäthylamino-pentan, Diäthanolamin, 4-Hydroxy-piperidin, Piperazin, 1-Hydroxyäthyl-piperazin. Die Reaktion wird zweckmässigerweise in einem inerten organischen Lösungsmittel durchgeführt, wie Benzol, Toluol, usw. Man arbeitet am besten bei erhöhter Temperatur, z. B. bei Siedetmperatur des Gemisches, oder im Falle von leicht flüchtigen Aminen, im Einschlussrohr, z. B.
bei einer Temperatur von etwa 1000. Zudem ist es angebracht, die Reaktion in Gegenwart eines anorganischen oder organischen säurebindenden Mittels durchzuführen. Als anorganisches säurebindendes Mittel kann z. B. wasserfreies Kaliumcarbonat angewendet werden. Als organisches säurebindendes Mittel dient vorteilhafterweise das gleiche Amin, welches zur Reaktion mit der Halogenverbindung eingesetzt ist. Eine zweckmässige Ausführungsform besteht darin, dass man auf 1 Mol Halogenverbindung 2 Mol Amin verwendet. Bei Verendung eines in den aromatischen Rin gen asymmetrisch substituierten Carbinols gelangt man bei der oben geschilderten Dehydratisierung zu einer Halogenalkylidenverbindung, welche aus einem Isomerengemisch besteht.
Dieses Isomerengemisch kann vor oder nach der Reaktion mit der Aminkomponente aufgetrennt werden, was nach an sich bekannten Methoden, z. B. durch fraktionierte Kristallisation, geschehen kann.
Die Erfindung umfasst auch die Herstellung der Säureadditionssalze, z. B. die Salze der vorgenannten in 9-Stellung basisch substituierten Thioxanthene mit anorganischen Säuren, wie Chlorwasserstoffsäure, Bromwasserstoffsäure, Schwefelsäure; mit organischen Säuren, wie Oxalsäure, Citronensäure, Essigsäure, Milchsäure, Weinsäure; sowie die Verwendung der Verfahrensprodukte zur Herstellung von quaternären Salzen durch Umsetzung der Verfahrensstoffe mit quaternisierenden Mitteln, z. B. Alkylhalogeniden, wie Methylbromid, ithyljodid; Dialkylsulfaten, wie Dimetthylsulfat und Aralkylhalogeniden, wie Benzylbromid.
Die erfindungsgemäss erhältlichen Verbindungen zeichnen sich durch vielfältige Wirkungen auf das Nervensystem aus. So sind narkosepotenzierende, adrenolytische, sedative, antihistaminartige, antiemetische, antipyretische, lokal-anästhetische und hypotherme Wir- kungen festgestellt worden. Die Verbindungen können als Heilmittel in Form pharmazeutischer Präparate Verwendung finden, welche sie oder ihre Salze in Mischung mit einem für die enterale oder parenterale Applikation geeigneten pharmazeutischen organischen oder anorganischen inerten Trägermaterial enthalten.
Die pharmazeutischen Präparate können in fester Form oder in flüssiger Form vorliegen. Gegebenenfalls enthalten sie Hilfsstoffe, wie Konservierungs-, Stabilisierungs-, Netz- oder Emulgiermittel, Salze zur Veränderung des osmotischen Druckes oder Puffer. Sie können auch noch andere therapeutisch wertvolle Stoffe enthalten.
Beispiel 1
In einem 2 Liter Dreihalskolben, der mit Rührer, Tropftrichter und Kühler versehen ist, werden 15 g Magnesiumspäne mit 50 ml trockenem Äther überdeckt und mit 0,5 ml Methyljodid versetzt.
Nachdem die lebhafte Reaktion etwas nachgelassen hat, wird eine Lösung von 54,6 g 1-Chlor-3-methoxypropan in 300 ml trockenem Äther derart eingetropft, dass das Reaktionsgemisch im Sieden gehalten wird.
Es wird noch 4 Stunden bei 400 unter Rückfluss gekocht.
Dann wird die Reaktionsmischung mit Eiswasser gekühlt, eine Suspension von 51,4g Thioxanthon in 600 ml trockenem Äther im Laufe einer Stunde zugegeben und das Ganze noch 15 Stunden bei 40 unter Rückfluss gekühlt. Am folgenden Tag wird nochmals mit Eiswasser gekühlt und die Reaktionsmischung mit einer kalt gesättigten Ammoniumchloridlösung versetzt. Die organische Schicht wird abgetrennt, die wässrige Phase zweimal mit je 150 ml Ather ausgeschüttelt und die vereinigten Ätherportionen über Natriumsulfat getrocknet und eingedampft. Der Rückstand liefert nach Umkristallisation aus hochsiedendem Petroläther gelbe, bei 94-950 schmelzende Kristalle von 9-Hydroxy-9-(3-methoxy-propyl)-thioxanthen.
Ausbeute 90 0/0.
20 g des erhaltenen 9-Hydroxy-9-(3'-methoxy-propyl)-thioxanthen und 50 ml einer 48 0/o-igen wässrigen Lösung von Bromwasserstoff werden 12 Stunden unter starkem Rühren unter Rückfluss gekocht. Nach dem Abkühlen wird das Reaktionsgemisch zweimal mit je 100 ml Äther geschüttelt, die vereinigten Ätherportionen mit Wasser, mit Natriumbicarbonatlösung und nochmals mit Wasser neutral gewaschen und eingedampft. Der Rückstand liefert nach Destillation im Hochvakuum 9-(3'-Brom-propyliden)-thioxanthen vom Siedepunkt 170-180 /0,05 mm Hg.
5 g 9-(3'-Brom-propyliden)ithioxanthen und 5 mol 100 O/o-iges Dimethylamin werden in 25 ml trockenem Toluol 8 Stunden im Einschlussrohr bei 1000 erwärmt.
Nach dem Erkalten wird mit Äther verdünnt, die ätherische Lösung mit Wasser gewaschen, getrocknet und eingedampft. Der Rückstand wird im Hochvakuum destilliert und besteht aus 9-(3'-Dimethylamino-propyliden)-thioxanthen (Siedepunkt 1400/0,04 mmHg). Das Hydrochlorid bildet farblose, wasserlösliche Kristalle vom Schmelzpunkt 186-188 .
Beispiel 2
7,2 g 9-(3'-Brom-propyliden)-thioxanthen (erhalten gemäss Beispiel 1) und 3,9 g Piperidin werden in 25 ml trockenem Toluol gelöst und 5 Stunden unter Rückfluss gekocht. Nach dem Erkalten wird mit 150 ml Äther verdünnt, mit Wasser gewaschen, über Natriumsulfat getrocknet und eingedampft. Aus dem Rückstand erhält man durch Destillation im Hochvakuum 9-(3'-Piperidino-propyliden)-thioxanthen vom Siedepunkt 170-175"/0,lmm Hg als sehr dickes Ö1. Das Hydrochlorid, aus Athanol-Sither umkristallisiert, schmilzt bei 220-2220. Ausbeute75 O/o.
Beispiel 3
In einem 2 Liter Dreihalskolben, der mit Rührer, Tropftrichter und Kühler versehen ist, werden 15 g Magnesiumspäne mit 60 ml trockenem Äther überdeckt und mit 5 Tropfen Methyljodid versetzt.
Nachdem die lebhafte Reaktion etwas nachgelassen hat, wird eine Lösung von 54,6 g 1-Chlor-3-methoxypropan in 300 ml trockenem Äther im Laufe einer Stunde eingetropft. Es wird noch 4 Stunden bei 40, unter Rückfluss gekocht.
Dann wird die Suspension von 59,5 g 2-Chlor-thioxanthon in 600 ml absolutem Äther im Laufe einer Stunde zu dem mit Eiswasser gekühlten Reaktionsgemisch zugetropft und das Ganze noch 18 Stunden unter Rückfluss gekocht. Am folgenden Tag wird nochmals mit Eiswasser gekühlt und die Reaktionsmischung mit einer kalt gesättigten Ammoniumchloridlösung versetzt. Die organische Schicht wird abgetrennt, die wässrige Phase zweimal mit je 200 ml Äther ausgeschüttelt und die vereinigten Ätherportionen über Natriumsulfat getrocknet und eingedampft. Der Rückstand liefert nach Umkristallisation aus hochsiedendem Petroläther gelbliche, bei 91-92 C schmelzende Kristalle von 2-Chlor-9-hydroxy-9-(3'-methoxypropyl)-thioxanthen. Ausbeute: 85 0/0.
20 g 2-Chlor-9-hydroxy-9 (3'-methoxy-propyl)-thioxanthen und 50 ml einer 48 0/o-igen, wässrigen Lösung von Bromwasserstoff werden 17 Stunden unter starkem Rühren unter Rückfluss gekocht. Nach dem Abkühlen wird das Reaktionsgemisch zweimal mit je 150 ml Äther geschüttelt, dievereinigten Ätherportionen mit Wasser, mit Natriumbicarbonatlösung und nochmals mit Wasser neutral gewaschen und eingedampft. Der Rückstand liefert nach Destillation im Hochvakuum 2-Chlor-9-(3 '-brom-propyliden) -thioxanthen vom Siedepunkt 185 /0,005 mm Hg.
8,5 g 2-Chlor-9 (3'-brom-propyliden)-thioxanthen und 6,3 g 1-Hydroxyäthyl-piperazin werden in 25 ml trokkendem Toluol gelöst und 4 Stunden unter Rückiluss erwärmt. Nach dem Erkalten wird mit 200 ml Äther verdünnt und zweimal mit je 150 ml 1 n Salzsäure geschüttelt. Die wässrige Phase wird alsdann durch Zugabe von festen Kaliumcarbonat alkalisch gestellt, die ausgefallene, ölige Base durch Schütteln mit Äther extrahiert, die ätherische Lösung mit Wasser gewaschen, über Natriumsulfat getrocknet und eingeengt. Der Rückstand stellt 2-Chlor-9- [3'-(4"-hydroxyäthyl- piperazin-1")-propyliden] -thioxan then dar. Das Dihydrochlorid, aus Methanol-Äther umkristallisiert, schmilzt bei 257-2580.
Beispiel 4
10,9 g 2-Chlor-9 (3-brom-propyliden)-thioxanthen (erhalten gemäss Beispiel 3) und 10 g 100 0/o-iges Dimethylamin werden in 50 ml trockenem Toluol gelöst und 8 Stunden bei 1000 im Einschlussrohr erwärmt. Nach dem Erkalten wird mit 300 ml Äther verdünnt, mit Wasser gewaschen, über Natriumsulfat getrocknet und die Lösungsmittel eingedampft. Aus dem Rückstand erhält man durch Destillation im Hochvakuum 2-Chlor-9-(3'-dimethylamino propyliden)-thioxanthen vom Siedepunkt 1600/0,04 mm Hg in Form eines Stereoisomerengemisches. Ausbeute 65 0/6.
Zur Gewinnung des höher schmelzenden Isomeren, das als Trans-Isomeres bezeichnet wird, werden 4g des obigen Isomerengemisches mit 4 ml Petroläther verdünnt und im Kühlschrank stehen gelassen. Im Laufe einiger Tage, rascher nach Animpfen, scheiden sich Kristalle der Trans-Form aus, die abgetrennt und aus Methanol-Wasser umkristallisiert werden; Schmelzpunkt 96-98 . Das Hydrochlorid des trans-2-Chlor-9-(3'dimethylamino-propyliden)-thioxanthen wird aus Alkohol-Äther umkristallisiert und schmilzt bei 221-223 .

PATENTANSPRUCH II
Verwendung der nach dem Patentanspruch I erhaltenen tricyclischen Verbindungen zur Herstellung von quaternären Salzen dieser tricyclischen Verbindungen durch Umsetzen der erhaltenen Verfahrensprodukte mit Quaternisierungsmitteln.
UNTERANSPRÜCHE
1. Verfahren nach Patentanspruch I, dadurch gekennzeichnet, dass man die erhaltenen Basen in ihre Säureadditionssalze überführt.
2. Verfahren nach Patentanspruch I, dadurch gekennzeichnet, dass als aliphatische, eine Äthergruppe aufweisende metallorganische Verbindung eine Alkoxyalkylmetallverbindung insbesondere eine Methoxyalkylmetallverbindung, verwendet wird.
3. Verfahren nach Unteranspruch 2, dadurch gekennzeichnet, dass als metallorganische Verbindung eine magueslumorganische Verbindung verwendet wird.
4. Verfahren nach Patentanspruch I, dadurch gekennzeichnet, dass das erhaltene Carbinol zwecks Dehydratisierung mit Bromwasserstoff behandelt wird, wobei zugleich die Äthergruppe durch Brom ersetzt wird.
5. Verfahren nach Patentanspruch I, dadurch gekennzeichnet, dass das erhaltene Carbinol zwecks Dehydratisierung mit Phosphoroxychlorid behandelt wird,

The bases obtained can be converted into their acid addition or quaternary salts.
The starting materials required for the process according to the invention can be prepared by methods known per se.
Optionally substituted thioxanthones can, for. B. obtained by condensation of optionally substituted thiosalicylic acid with optionally substituted benzene.
The preparation of the aliphatic, organometallic compounds having an ether function can also be carried out according to previously known methods. The magnesium compounds of alkoxy-alkyl halides required for a preferred embodiment can be, for. B. win by direct conversion of magnesium with an ethereal solution of the alkoxyalkyl halide, the reaction is conveniently stimulated by the addition of methyl iodide or ethyl bromide. Examples of alkyl halides which can carry an alkoxy group are propyl halides, butyl halides, amyl halides, hexyl halides and heptyl halides.
The alkyl radicals can be straight or branched. The alkoxy group is conveniently a lower alkoxy group such as methoxy, ethoxy, propoxy, butoxy, which z. B. is in the end position of the alkyl halide. co-methoxy-alkylmagnesium compounds are preferred starting materials. The ether function can in addition to alkoxy z. B. also mean aralkoxy, such as benzyloxy or phenäthyloxy, or tetrahydropyranyloxy.
In particular, magnesium, lithium and organozinc metal compounds can be used to carry out the process according to the invention. In a preferred embodiment, the starting ketones of the general formula I are either in solid, finely powdered form or in an inert organic solvent, such as. B. absolute ether, benzene, tetrahydrofuran, is added to the magnesium compound of an alkyl halide having an ether function, which is in such an organic inert solvent. After the reaction has ended, the reaction product is subjected to hydrolysis. The decomposition of the organic metal compounds formed is particularly advantageous under practically neutral conditions, e.g. B. by hydrolysis in aqueous ammonium chloride solution.
In this way, the tertiary carbinols of the general formula II below are formed, which by treatment with suitable, water-immiscible organic solvents, such as. B. ether, ethyl acetate, chloroform, methylene chloride, separated from the by-products of the reaction and isolated. As already mentioned, these compounds can be optionally substituted in the aromatic nuclei by alkyl, trifluoromethyl, aralkyl, aryl, alkoxy, aralkoxy, aryloxy, alkylthio, aralkylthio, arythio, halogen, acyl, amino , Hydroxy and carboxy groups.
Any acyl, amino, hydroxyl and carboxyl groups that may be present must be protected from reaction with the organometallic compound in order to prevent these groups from reacting with the organometallic reagent. This reaction sequence is advantageously carried out in two preferred embodiments: a) The hydrolysis product is treated with hydrogen bromide, expediently with a concentrated aqueous solution of hydrogen bromide at boiling point, the corresponding bromine compound being formed.
b) The hydrolysis product is treated with phosphorus oxychloride, which is advantageously done at reflux temperature. This creates the corresponding chlorine compound. The halogen compounds of the formula III are usually viscous, e.g. T. crystallizable substances which can be distilled without decomposition in a high vacuum and have the following formula

These compounds can optionally be substituted in the aromatic nuclei by alkyl, trifluoromethyl, aralkyl, aryl, alkoxy, aralkoxy, aryloxy, alkylthio, aralkylthio, arylthio, halogen, acyl, amino, hydroxy and carboxy groups.
In a further reaction stage, the halogen compounds of the formula III obtained above are reacted with primary or secondary amines optionally substituted by hydroxyl or further amino groups, the thioxanthene compounds bearing the divalent radical D in the 9-position being formed. Suitable amines are e.g. B. monoalkylamines or dialkylamines, such as. B. methylamine, ethylamine, dimethylamine, diethylamine. The use of monoalkylamines leads to new compounds of particular therapeutic interest. The alkyl radical can also be replaced by a cycloalkyl radical, as in the case of methylcyclopentylamine. The secondary amino group can also be a member of a saturated heterocyclic ring system. Examples of such cyclic amines are piperidine, pyrrolidine and morpholine.
If appropriate, as stated, the primary or secondary amines used for the reaction can also contain hydroxyl groups or additional amino groups. Examples of such amines are 2-amino-5-diethylamino-pentane, diethanolamine, 4-hydroxy-piperidine, piperazine and 1-hydroxyethyl-piperazine. The reaction is conveniently carried out in an inert organic solvent, such as benzene, toluene, etc. It is best to work at an elevated temperature, e.g. B. at boiling temperature of the mixture, or in the case of volatile amines, in the containment tube, for. B.
at a temperature of about 1000. It is also appropriate to carry out the reaction in the presence of an inorganic or organic acid-binding agent. As an inorganic acid-binding agent, for. B. anhydrous potassium carbonate can be used. The same amine which is used for the reaction with the halogen compound is advantageously used as the organic acid-binding agent. An expedient embodiment consists in using 2 moles of amine for 1 mole of halogen compound. If a carbinol which is asymmetrically substituted in the aromatic rings is used, the dehydration described above leads to a haloalkylidene compound which consists of a mixture of isomers.
This mixture of isomers can be separated before or after the reaction with the amine component, which can be achieved by methods known per se, e.g. B. by fractional crystallization can happen.
The invention also includes the preparation of the acid addition salts, e.g. B. the salts of the above-mentioned thioxanthenes which are basic substituted in the 9-position with inorganic acids, such as hydrochloric acid, hydrobromic acid, sulfuric acid; with organic acids such as oxalic acid, citric acid, acetic acid, lactic acid, tartaric acid; and the use of the process products for the production of quaternary salts by reacting the process materials with quaternizing agents, e.g. B. alkyl halides, such as methyl bromide, ethyl iodide; Dialkyl sulfates such as dimethyl sulfate and aralkyl halides such as benzyl bromide.
The compounds obtainable according to the invention are distinguished by diverse effects on the nervous system. For example, narcosis-potentiating, adrenolytic, sedative, antihistamine-like, antiemetic, antipyretic, local anesthetic and hypothermic effects have been found. The compounds can be used as medicaments in the form of pharmaceutical preparations which contain them or their salts in a mixture with a pharmaceutical, organic or inorganic inert carrier material suitable for enteral or parenteral administration.
The pharmaceutical preparations can be in solid form or in liquid form. They may contain auxiliaries, such as preservatives, stabilizers, wetting agents or emulsifiers, salts to change the osmotic pressure or buffers. They can also contain other therapeutically valuable substances.
example 1
In a 2 liter three-necked flask equipped with a stirrer, dropping funnel and cooler, 15 g of magnesium shavings are covered with 50 ml of dry ether and 0.5 ml of methyl iodide is added.
After the lively reaction has subsided somewhat, a solution of 54.6 g of 1-chloro-3-methoxypropane in 300 ml of dry ether is added dropwise in such a way that the reaction mixture is kept boiling.
It is refluxed at 400 for a further 4 hours.
The reaction mixture is then cooled with ice water, a suspension of 51.4 g of thioxanthone in 600 ml of dry ether is added over the course of an hour and the whole is refluxed at 40 for a further 15 hours. The following day, the mixture is cooled again with ice water and a cold saturated ammonium chloride solution is added to the reaction mixture. The organic layer is separated off, the aqueous phase is extracted twice with 150 ml of ether each time and the combined ether portions are dried over sodium sulfate and evaporated. After recrystallization from high-boiling petroleum ether, the residue gives yellow crystals of 9-hydroxy-9- (3-methoxy-propyl) -thioxanthene which melt at 94-950.
Yield 90%.
20 g of the 9-hydroxy-9- (3'-methoxy-propyl) -thioxanthene obtained and 50 ml of a 48% strength aqueous solution of hydrogen bromide are refluxed for 12 hours with vigorous stirring. After cooling, the reaction mixture is shaken twice with 100 ml of ether each time, the combined portions of ether are washed neutral with water, with sodium bicarbonate solution and again with water and evaporated. After distillation in a high vacuum, the residue gives 9- (3'-bromopropylidene) thioxanthene with a boiling point of 170-180 / 0.05 mm Hg.
5 g of 9- (3'-bromopropylidene) ithioxanthene and 5 mol of 100% strength dimethylamine are heated in 25 ml of dry toluene for 8 hours at 1000 in a containment tube.
After cooling, it is diluted with ether, the ethereal solution washed with water, dried and evaporated. The residue is distilled in a high vacuum and consists of 9- (3'-dimethylamino-propylidene) -thioxanthene (boiling point 1400 / 0.04 mmHg). The hydrochloride forms colorless, water-soluble crystals with a melting point of 186-188.
Example 2
7.2 g of 9- (3'-bromo-propylidene) -thioxanthene (obtained according to Example 1) and 3.9 g of piperidine are dissolved in 25 ml of dry toluene and refluxed for 5 hours. After cooling, it is diluted with 150 ml of ether, washed with water, dried over sodium sulfate and evaporated. 9- (3'-Piperidino-propylidene) -thioxanthene with a boiling point of 170-175 "/ 0.1 mm Hg is obtained as a very thick oil from the residue by distillation under high vacuum. The hydrochloride, recrystallized from ethanol-sither, melts at 220- 2220. Yield 75 O / o.
Example 3
In a 2 liter three-necked flask equipped with a stirrer, dropping funnel and cooler, 15 g of magnesium shavings are covered with 60 ml of dry ether and 5 drops of methyl iodide are added.
After the lively reaction has subsided somewhat, a solution of 54.6 g of 1-chloro-3-methoxypropane in 300 ml of dry ether is added dropwise over the course of one hour. It is refluxed for another 4 hours at 40.
The suspension of 59.5 g of 2-chloro-thioxanthone in 600 ml of absolute ether is then added dropwise over the course of one hour to the reaction mixture, which has been cooled with ice water, and the whole is refluxed for a further 18 hours. The following day, the mixture is cooled again with ice water and a cold saturated ammonium chloride solution is added to the reaction mixture. The organic layer is separated off, the aqueous phase is shaken out twice with 200 ml of ether each time and the combined ether portions are dried over sodium sulfate and evaporated. After recrystallization from high-boiling petroleum ether, the residue gives yellowish crystals of 2-chloro-9-hydroxy-9- (3'-methoxypropyl) -thioxanthene melting at 91-92 ° C. Yield: 85%.
20 g of 2-chloro-9-hydroxy-9 (3'-methoxy-propyl) -thioxanthene and 50 ml of a 48% strength aqueous solution of hydrogen bromide are refluxed for 17 hours with vigorous stirring. After cooling, the reaction mixture is shaken twice with 150 ml of ether each time, the combined portions of ether are washed neutral with water, with sodium bicarbonate solution and again with water and evaporated. After distillation in a high vacuum, the residue gives 2-chloro-9- (3'-bromopropylidene) thioxanthene with a boiling point of 185 / 0.005 mm Hg.
8.5 g of 2-chloro-9 (3'-bromopropylidene) thioxanthene and 6.3 g of 1-hydroxyethyl piperazine are dissolved in 25 ml of drying toluene and heated under reflux for 4 hours. After cooling, it is diluted with 200 ml of ether and shaken twice with 150 ml of 1N hydrochloric acid each time. The aqueous phase is then made alkaline by adding solid potassium carbonate, the oily base which has precipitated is extracted by shaking with ether, the ethereal solution is washed with water, dried over sodium sulphate and concentrated. The residue is 2-chloro-9- [3 '- (4 "-hydroxyäthyl-piperazine-1") -propylidene] thioxane. The dihydrochloride, recrystallized from methanol ether, melts at 257-2580.
Example 4
10.9 g of 2-chloro-9 (3-bromopropylidene) thioxanthene (obtained according to Example 3) and 10 g of 100% dimethylamine are dissolved in 50 ml of dry toluene and heated at 1000 for 8 hours in the containment tube. After cooling, it is diluted with 300 ml of ether, washed with water, dried over sodium sulfate and the solvents evaporated. From the residue, 2-chloro-9- (3'-dimethylamino propylidene) thioxanthene with a boiling point of 1600 / 0.04 mm Hg is obtained in the form of a stereoisomer mixture by distillation in a high vacuum. Yield 65%.
To obtain the higher melting isomer, which is referred to as the trans isomer, 4 g of the above isomer mixture are diluted with 4 ml of petroleum ether and left to stand in the refrigerator.
