Verfahren zur Herstellung von Benzodiazepin-Derivaten
Die vorliegende Erfindung betrifft ein neues Verfahren zur Herstellung von Benzodiazepin-Derivaten der Formel

worin R1 Wasserstoff oder Methyl und Rt Wasserstoff, Chlor oder Fluor bedeuten bzw. von Säureadditionssalzen dieser Verbindungen.
Das erfindungsgemässe Verfahren ist dadurch gekennzeichnet, dass man eine Verbindung der Formel

worin R1 und R2 obige Bedeutung haben und R2 Was- serstoff oder Acyl bedeutet, mit einer starken Base oder, falls R3 Wasserstoff ist, mit einer starken Base oder einem Isocyanat oder Carbodiimid behandelt. Das erhaltene Produkt kann erwünschtenfalls in ein Säureadditionssalz übergeführt werden.
Eine bevorzugte Ausführungsform umfasst die Dehydratisierung von 7-Chlor-2,3,4,5-tetrahydro-4-hydroxy- - 1 -methyl-5-phenyl- 111-1 4-benzodiazepin, oder die Säureabspaltung des entsprechenden O-Acetyl-Derivates.
Für die Durchführung des erfindungsgemässen Verfahrens geeignete Basen sind anorganische und organische Basen, wie Alkalihydroxyde (z.B. Natriumhydroxyd, Kaliumhydroxyd u. dgl.), Erdalkalihydroxyde (wie Calciumhydroxyd u. dgl.), Alkoxyde (wie Natriumäthylat, Natrium-tert.-Butoxyd u. dgl.), Alkalihydride (wie Lithiumhydrid, Natriumhydrid u. dgl.), Erdalkalihydride, Alkaliamide (wie Natriumamid), quaternäre Basen (wie Tetramethylammoniumhydroxyd u. dgl.) und stark basische Ionenaustauscher.
Geeignete Carbodiimide sind z.B. Dicyclohexylcarbodiimid u. dgl. Unter den für die Dehydratisierung geeigneten Isocyanaten sind z.B. Phenylisocyanat u. dgl. zu nennen.
Die Dehydratisierung bzw. Säureabspaltung wird zweckmässigerweise in einem organischen Lösungsmittel, wie Alkohole (z.B. niedere Alkanole, wie Methanol, Äthanol, tert.Butanol), Kohlenwasserstoffe (wie Benzol, Toluol), Äther (wie Dioxan), niedere Dialkylsulfoxyde (z. B. Dimethylsulfoxyd) u. dgl. durchgeführt. Die Wahl eines bestimmten Lösungsmittels hängt selbstverständlich weitgehend von der Natur des eingesetzten Dehydrati sierungs- bzw. Deacylierungsmittels ab.
Für diese Reaktion geeignete Temperaturen sind Temperaturen zwischen OO und 1500C. Nach einer ersten Ausführungsart des erfindungsgemässen Verfahrens wird eine Verbindung der Formel II, worin R Wasserstoff bedeutet, durch Behandlung mit einer starken Base dehydratisiert.
Hierbei verwendet man als Ausgangsprodukt beispielsweise 7-Chlor-2,3,4,5-tetrahydro-4-hydroxy -5-phenyl- lH-1,4-benzodiazepin, 7-Chlor-5-(2'-chlorphe nyl)-2,3,4,5-tetrahydro-4-hydroxy- 1 H - 1,4 - benzodiazepin, 7-Chlor-2,3,4,5-tetrahydro-4-hydroxy- 1 - methyl -5-phenyl -IH-1,4-benzodiazepin, 7Chlor 5-(2'-chlorphenyl)-2,3,4, 5- tetrahydro - 4- hydroxy-1 -methyl-lH-1 ,4-benzodiazepin usw. und erhitzt dieses in einem geeigneten organischen Lösungsmittel in Gegenwart einer starken Base während ca. 5 bis 30 Stunden am Rückfluss zum Sieden. Die Wahl des Lösungsmittels hängt von der eingesetzten Base ab; man verwendet beispielsweise Alkalihydride, wie Natriumhydrid, in Mineralöl, Dimethylsulfoxyd oder tert.
Butanol, Alkalihydroxyde, wie Natrium- oder Kaliumhydroxyd, in Äthanol u. dgl., Alkoxyde im entsprechenden Alkanol (z.B. Natriumäthylat in Äthanol), Tetramethylammoniumhydroxyd oder andere quartäre Ammoniumbasen in einem niederen Alkanol, wie Äthanol, stark basische Ionenaustauscher in Äthanol usw. In vielen Fällen erweist es sich als vorteilhaft, die Reaktion in einer Inertgasatmosphäre durchzuführen, beispielsweise unter Stickstoff.
Nach einer zweiten Ausführungsart des erfindungsgemässen Verfahrens verwendet man als Ausgangsprodukt ebenfalls eine Verbindung der Formel II, worin R, Wasserstoff bedeutet, und dehydratisiert diese durch Behandlung mit einem Carbodiimid, wie Dicyclohexylcarbodiimid, oder einem Isocyanat, z.B. Phenylisocyanat.
Die Reaktion wird beispielsweise so durchgeführt, dass man das Ausgangsprodukt und das Dehydratisierungsmittel in einem Kohlenwasserstoff, wie Toluol u. dgl., oder in einem anderen geeigneten organischen Lösungsmittel während ca. 2-20 Stunden am Rückfluss zum Sieden erhitzt.
Eine weitere Ausführungsart des erfindungsgemässen Verfahrens besteht darin, dass man eine Verbindung der Formel II, worin R3 eine Acylgruppe bedeutet, durch Behandlung mit einer starken Base einer Säureabspaltung unterzieht. So gelangt man beispielsweise ausgehend von 4-Acetoxy-7-chlor-2,3,4,5-tetrahydro- 1 -methyl-5-phe- nyl-lH-1,4-benzodiazepin zu 7-Chlor-2,3 -dihydro- 1 -me- thyl-5-phenyl-lH-1,4-benzodiazepin, indem man dieses mit einem Alkalihydrid, wie Natriumhydrid, in Dimethylsulfoxyd während ca. 1z; Stunde auf etwa 600 erwärmt oder mit einem Alkalihydroxyd, wie Natriumhydroxyd, in Äthanol oder einem anderen geeigneten niederen Alkanol während ca. 20-30 Stunden am Rückfluss zum Sieden erhitzt.
Die erfindungsgemäss hergestellten Verbindungen der Formel I, unter denen insbesondere dass 7-Chlor-2,3-dihydro-l -methyl-5-phenyl-l H-1 ,4-benzodiazepin hervorzuheben ist, sind wertvolle Sedativa, Muskelrelaxantien und Anticonvulsiva.
In den folgenden Beispielen sind alle Temperaturen in Grad C angegeben.
Beispiel I
1 g 7- Chlor-2,3,4,5-tetrahydro-4-hydroxy-5-phenyl- -lH-1,4-benzodiazepin wird in 10 ml 50%iger Natriumhydrid-Suspension in Mineralöl unter Stickstoff während 6 Stunden gekocht. Nach Zugabe von Wasser wird mit Äther ausgeschüttelt, die Ätherphase mit Wasser gewaschen, über Nariumsulfat getrocknet und eingedampft. Chromatographie des Rückstandes an 40 g Kieselgel mit 10% Alkohol in Essigester liefert 7-Chlor-2,3 -dihydro-5-phenyl- 1 H- 1 ,4-benzodiazepin vom Schmelz punkt 171.1730.
Beispiel 2
1 g 7-Chlor-2,3,4,5-tetrahydro-4-hydroxy-5-phenyl- -1H-1,4-benzodiazepin und 0,2 g Natriumhydridsuspen sion werden in 10 ml tert. Butanol unter Stickstoff wäh rend 24 Stunden gekocht. Das Reaktionsgemisch wird in Wasser und Äther aufgenommen, die Ätherphase mit
Wasser gewaschen, über Natriumsulfat getrocknet und eingedampft. Der Rückstand wird aus Äther kristalli siert und liefert 7-Chlor-2,3-dihydro-5-phenyl-lH-1,4- -benzodiazepin vom Schmelzpunkt 171 - 1730.
Beispiel 3
1 g 7-Chlor-2,3,4,5-tetrahydro-4-hydroxy-5-phenyl- -lH-1,4-benzodiazepin wird in einer Lösung von 0,1 g
Natrium in 15 ml Alkohol während 18 Stunden unter
Stickstoff gekocht. Nach dem Einengen im Vakuum wird zwischen Wasser und Methylenchlorid verteilt, die Me thylenchloridphase über Natriumsulfat getrocknet und eingedampft. Der Rückstand liefert nach Kristallisation aus Äther 7-Chlor-2,3-dihydro-5-phenyl- 1 H- 1 ,4-benzo- -diazepin vom Schmelzpunkt 171 - 1730.
Beispiel 4
1 g 7-Chlor-2,3,4,5-tetrahydro-4-hydroxy-5-phenyl- -lH-1,4-benzodiazepin wird in einer Lösung von 0,5 g
Kaliumhydroxyd in 15 ml Alkohol während 18 Stunden unter Stickstoffatmosphäre gekocht. Nach der in Bei spiel 3 beschriebenen Aufarbeitung erhält man 7-Chlor -2,3-dihydro-5-phenyl-lH-1,4-benzodiazepin vom Smp.
171-1730.
Beispiel 5
1 g 7-Chlor-2,3,4,5-tetrahydro-4-hydroxy-5-phenyl- -lH-1,4-benzodiazepin wird in 20 ml Alkohol in Gegen wart von 1 g < sAmberlite JR 401 in der Hydroxylform während 16 Stunden gekocht. Nach Abtrennung des
Ionentauschers wird eingedampft und der Rückstand an
30 g Kieselgel mit 10% Alkohol in Äthylacetat chroma tographiert. Man erhält 7-Chlor-2,3-dihydro-5-phenyl -1H-1,4-benzodiazepin vom Schmelzpunkt 171 - 1730.
Beispiel 6
1 g 7-Chlor-2,3,4,5-tetrahydro-4-hydroxy-5-phenyl- -lH-1,4-benzodiazepin wird in 20 ml Alkohol und 2 ml -1 0%iger wässriger Tetramethylammoniumhydroxydlö.
sung während 24 Stunden unter Rückfluss gekocht. Nach dem Eindampfen wird zwischen Wasser und Methylen chlorid verteilt, die Methylenchloridphase abgetrennt, ge trocknet und eingedampft. Kristallisation des Rückstandes liefert 7-Chlor-2,3-dihydro-5-phenyl- 1 H- 1 ,4-benzo- diazepin vom Schmelzpunkt 170-1720.
Beispiel 7
2 g 7-Chlor-2,3,4,5-tetrahydro-4-hydroxy- 1 -methyl-5- -phenyl-1H-1,4-benzodiazepin werden in einer Lösung von 0,4 g Natriumhydridsuspension in 20 ml tert.-Buta nol während 16 Stunden unter Rückfluss und Stickstoff atmosphäre gekocht. Nach Versetzen mit Wasser wird mit Äther extrahiert. Die Ätherphase wird über Natrium sulfat getrocknet und eingedampft. Der Rückstand liefert nach Kristallisation aus Äther/Hexan 7-Chlor-2,3 -dihydro- 1 -methyl-5-phenyl- 1 H -1 1,4-benzodiazepin vom
Schmelzpunkt 98-1000.
Das Ausgangsmaterial kann wie folgt hergestellt werden:
Zu einer Suspension von 6,5 g Lithiumaluminium hydrid in 250 ml Äther werden 20 g 7-Chlor-1,3-dihydro - 1 -methyl-5-phenyl-2H- 1 ,4-benzodiazepin-2-on-4-oxyd in kleinen Portoinen zugefügt. Nach beendeter Zugabe wird während 4 Stunden unter Rückfluss gekocht. Durch vorsichtiges Eintropfen von 35 ml Wasser wird hydrolysiert und das anorganische Material abfiltriert. Der nach dem Eindampfen des Filtrates erhaltene Rückstand wird an 300 g Kieselgel mit 10% Äthylacetat in Methylenchlorid chromatographiert. Man erhält kristallines 7-Chlor-2,3, 4,5-tetrahydro-4-hydroxy- 1 -methyl-5-phenyl - lH- 1 ,4-ben- zodiazepin vom Schmelzpunkt 138-1400. Nach Umkristallisation aus Methanol schmilzt das reine Produkt bei 141.1430.
Beispiel 8
1,45 g (5 mMol) 7-Chlor-2,3,4,5-tetrahydro-4-hy droxy-l -methyl-5-phenyl-1H- 1A-benzodiazepin u. 1,25 g Dicyclohexylcarbodiimid werden während 3 Stunden in 20 ml Toluol zum Rückfluss erhitzt. Nach dem Abkühlen wird der ausgeschiedene Dicyclohexylharnstoff abgesaugt und das Filtrat dreimal mit 20 ml 2n Salzsäure extrahiert. Die rohe, salzsaure Lösung wird mit konz.
Ammoniak alkalisch gestellt und mit Äther extrahiert.
Die Ätherextrakte werden getrocknet und eingedampft.
Nach Krisallisation des Rückstandes aus Äther/Hexan erhält man 7-Chlor-2,3-dihydro- 1 -methyl-5-phenyl- 1H- -1,4-benzodiazepin vom Schmelzpunkt 96-980.
Beispiel 9
Eine Lösung von 1,45 g (5 mMol) 7-Chlor-2,3,4,5- -tetrahydro-4-hydroxy- 1 -methyl-5-phenyl- iH- 1,4- benzo- diazepin in 20 ml abs. Toluol wird mit 0,75 g (6,3 mMol) Phenylisocyanat versetzt und während 18 Stunden zum Rückfluss erhitzt. Das beim Abkühlen auskristallisierte Nebenprodukt wird abgesaugt und das Filtrat zweimal mit je 25 ml 2n Salzsäure extrahiert. Aus der salzsäuren Lösung werden die Basen wie in Beispiel 8 angegeben isoliert und an 20 g Kieselgel mit Äthylacetat chromatographiert. Kristallisation aus Hexan liefert 7-Chlor-2,3 -dihydro- 1 -methyl-5-phenyl - 1H - 1,4-benzodiazepin vom Schmelzpunkt 97-990.
Beispiel 10
2 g 4-Acetoxy-7-chlor-2,3,4,5-tetrahydro- 1 -methyl-5- -phenyl-lH-1,4-benzodiazepin werden in 20 ml abs. Dimethylsulfoxyd gelöst, mit 0,5 g Natriumhydridsuspension (50%ig in Mineralöl) versetzt und während 30 Minuten auf 600 erwärmt. Das Reaktionsgemisch wird auf Wasser gegossen und mit Äther extrahiert. Der aus den Ätherextrakten erhaltene Rückstand liefert nach Chromatographie an 40 g Kieselgel mit Äthylacetat 7-Chlor-2,3 -dihydro- 1 -methyl-5-phenyl - 1H- 1,4-benzodiazepin vom Schmelzpunkt 95-980.
Das Ausgangsprodukt kann wie folgt hergestellt werden:
Eine Lösung von 2 g 7-Chlor-2,3,4,5-tetrahydro-4 -hydroxy - 1 - methyl - 5 - phenyl-lH.l,4-benzodiazepin in
10 ml Pyridin wird mit 2 ml Acetanhydrid versetzt und während 3 Tagen bei Raumtemperatur stehengelassen.
Nach dem Eindampfen im Vakuum wird der Rückstand in Äther aufgenommen und mit wässriger Bicarbonatlösung ausgewaschen. Die über Natriumsulfat getrocknete Ätherphase wird eingedampft und der Rückstand durch Chromatographie an 50 g Kieselgel mit 10% Äther in Benzol gereinigt.
Beispiel 11
1 g 7-Chlor-5-(2'-chlorphenyl)-2,3,4,5-tetrahydro-4- -hydroxy-1H-1,4-benzodiazepin wird in 15 ml Alkohol in Gegenwart von 0,5 g Kaliumhydroxyd während 24 Stunden unter Rückfluss gekocht. Die heisse Lösung wird mit Wasser verdünnt und zur Kristallisation gestellt. Das auskristallisierte, hellgelbe Produkt wird abgesaugt, mit Wasser gewaschen und im Vakuum getrocknet. Man er hält 7-Chlor-5- (L'-chlorphenyl) -2,3-dihydro- 1H- 1 4-ben- zodiazepin vom Schmelzpunkt 176-1780.
Das Ausgangsmaterial kann wie folgt hergestellt werden:
9 g 7-Chlor-5-(2'-chlorphenyl)-1,3-dihydro-2H-1,4- -benzodiazepin-2-on-4-oxyd werden in kleinen Portionen in eine Suspension von 4 g Lithiumaluminiumhydrid in 200 ml Äther eingetragen. Nach beendeter Zugabe wird noch während 2 Stunden zum Sieden erhitzt. Das Reaktionsgemisch wird durch Zutropfen von 20 ml Wasser hydrolysiert. Das anorganische Material wird abgesaugt und das Filtrat eingedampft. Chromatographie des Rohproduktes an 200 gKieselgel mit 20% Äthylacetat in Methylenchlorid liefert nach Kristallisation aus Äther/ Hexan 7- Chlor - 5-(2'-chlorphenyl)-2,3,4,5-tetrahydro-4- -hydroxy-1H-1,4-benzodiazepin vom Schmelzpunkt 1531540.
Beispiel 12
1 g 7-Chlor-5-(2'-chlorphenyl)-2,3,4,5-tetrahydro-4- .hydroxy.1-methyl-lH.l,4.benzodiazepin und 0,5 g Kaliumhydroxyd werden in 15 ml Alkohol während 24 Stunden unter Rückfluss gekocht. Nach dem Einengen im Vakuum wird zwischen Wasser und Äther verteilt, die Ätherphase getrocknet und eingedampft. Kristallisation des Rückstandes aus Äther/Hexan liefert 7-Chlor-5-(2'- -chlorphenyl)-2,3-dihydro- 1 -methyl- 1 H- 1,4-benzodiazepin vom Schmelzpunkt 92-950.
Das Ausgangsmaterial kann wie folgt hergestellt werden:
In eine Suspension von 2 g Lithiumaluminiumhydrid in 100 ml Äther werden 3 g 7-Chlor-5-(2'-chlorphenyl) - 1,3-dihydro- 1 -methyl-2H- 1,4 - benzodiazepin-2-on-4-oxyd in kleinen Portionen eingetragen. Nach der Zugabe wird während 2 Stunden unter Rückfluss gekocht. Das Reaktionsgemisch wird hierauf mit 10 ml Wasser hydrolysiert und filtriert. Das nach dem Eindampfen des Filtrates erhaltene Harz wird an 50 g Kieselgel mit Methylenchlorid chromatographiert. Man erhält 7-Chlor-5-(2' .chlorphenyl).2,3 ,4,5.tetrahydro-4-hydroxy- 1 -methyl- lH- - 1,4-benzodiazepin vom Schmelzpunkt 158-1590.
Beispiel 13
Eine Lösung von 2 g 4-Acetoxy-7-chlor-2,3,4,5-tetra hydro-l -methyl-5-phenyl-1H-1 A-benzodiazepin in 20 ml Äthanol wird mit 1 g Natriumhydroxid versetzt und während 24 Stunden unter Rückfluss gekocht. Nach dem Einengen im Vakuum wird zwischen Wasser und Äther verteilt.
Die ätherische Phase wird mit Wasser gewaschen, über Natriumsulfat getrocknet und eingedampft. Kristallisation des Rückstandes aus Äther-Hexan liefert 7-Chlor -2,3 - dihydro - 1 - methyl - 5 - phenyl.lH-1,4.henzodiazepin vom Smp. 96-980C.

Example I.
1 g of 7-chloro-2,3,4,5-tetrahydro-4-hydroxy-5-phenyl-1H-1,4-benzodiazepine is boiled in 10 ml of 50% sodium hydride suspension in mineral oil under nitrogen for 6 hours . 20 g of 7-chloro-1,3-dihydro-1-methyl-5-phenyl-2H-1,4-benzodiazepin-2-one-4-oxide are added to a suspension of 6.5 g of lithium aluminum hydride in 250 ml of ether added in small postage. 2 g of 4-acetoxy-7-chloro-2,3,4,5-tetrahydro-1-methyl-5-phenyl-1H-1,4-benzodiazepine are dissolved in 20 ml of abs. A solution of 2 g of 7-chloro-2,3,4,5-tetrahydro-4-hydroxy - 1 - methyl - 5 - phenyl-lH.l, 4-benzodiazepine in
10 ml of pyridine are mixed with 2 ml of acetic anhydride and left to stand for 3 days at room temperature.
