Verfahren zur Herstellung von Sonnenlicht filternden Stoffen
Die Erfindung betrifft ein Verfahren zur Herstellung neuer und geeigneter, das Sonnenlicht filternder Stoffe sowie die Verwendung der Verfahrensprodukte als Komponente in Kompositionen zum Schutz der menschlichen Haut gegen verbrennende, gerbende und abbauende Wirkungen des Sonnenlichts. Insbesondere werden die erfindungsgemäss hergestellten Verbindungen in kosmetische Kompositionen eingearbeitet, die ausser- ordentlich gut geeignet sind zum Schutz der Haut gegen das Sonnenlicht.
Es sind im allgemeinen Sonnenlicht filternde Stoffe entwickelt worden, die ganz befriedigenden Schutz gegen die verbrennenden Strahlen gewähren, die aber die gerbend wirkenden Strahlen durchlassen. Die am schädlich- sten verbrennend wirkenden Wellenlängen des Sonnenlichts befinden sich im Bereich von ungefähr 2900 bis 3100 A. Die Wirkung dieser Wellenlängen auf die menschliche Haut ruft eine ernsthafte Verbrennung, Rötung, Blasenbildung und Ödeme hervor, die für viele Verbrennungen charakteristisch sind. Dies ist vom asthe- tischen Standpunkt aus unerwiinscht und bedeutet gleichzeitig eine ernste Gefahr für die menschliche Gesundheit. Um die Wirkung der verbrennenden Strahlen des Sonnenlichts zu überwinden, sind verschiedene Verbindungen angewendet worden, die die verbrennend wirkenden Strahlen aufheben.
Die Öffentlichkeit nimmt an und im allgemeinen hin, dass das Gerben ein wtinschens- wertes Phänomen ist und irgend etwas mit guter Gesundheit zu tun hat. Die Öffentlichkeit ist dazu erzogen worden, die zahlreichen im Handel erhältlichen Produkte, welche die verbrennend wirkenden Strahlen aussieben und die gerbend wirkenden Strahlen durchlassen, zu akzeptieren. Innerhalb der letzten Jahre hat man im allgemeinen angenommen, dass das Gerben, obgleich es nlltzlich ist zum Schutze der Haut gegen fortgesetzte Sonnenbestrahlung, es jedoch verbunden ist mit einem allgemeinen Abbau der Haut durch die chemisch wirkende Strahlung.
Obgleich die grösste $Zahl der Menschen nicht schwer durch gerbend wirkende Strahlen angegriffen werden, gibt es eine grove Zahl, die sogar gegen die gerbend wirkenden Strahlen der Sonne empfindlich sind, so dass die Bestrahlung in dieser Wellenlänge bei ihnen grosse gesundheitliche Gefahren hervorruft. Daher besteht für die, die sowohl Schutz gegen Verbrennen und Gerben der Sonnenstrahlen wtinschen und für diejenigen, die nicht gegerbt werden wollen, ein echtes Bedürfnis nach einem in einem breiten Spektrum, das Sonnenlicht filterndem Mittel, welches der Haut einen angemessenen Schutz verleiht, genügend stabil ist unter den Bedingungen des Gebrauchs und seine Wirkungen mehrere Stunden lang behält, leicht auf die 1Haut aufgebracht werden kann und nicht leicht entfernt werden kann, zum Beispiel durch einfaches Waschen mit Wasser.
Diese letztere Eigenschaft muB gewährleistet sein, weil es unerwtinscht ist, ein beispielsweise schon beim Baden leicht entfernbares Material zu verwenden.
Es ist ein besonderer Vorteil der vorliegenden Erfindung, dass durch sic Verbindungen hergestelit werden können, die überragende Fil\tereigenschaften haben für Sonnenstrahlen mit Wellenlängen von 2900 bis 3500 A; der Bereich der Wellenlänge von 3200 bis 3500 A umschliesst den Bereich des Sonnenlichtes, der gerbend wirkt.
Es wurden nun Verbindungen gefunden, die iiberragende Filtcreigenschaften haben für die verbrennend und gerbend wirkenden Sonnenstrahlen und insbeson dcre der Strahlen mit Wellenlängen im ultravioletten Bereich von 2900 A bis ungefähr 3500 A. Diese Verbindungen haben hervorragende K-Werte sowie unerwartete Lichtbeständigkeit und Stabilität gegen ultraviolette Strahlen.
Der K-Wert ist ein Mass für die Wirksamkeit der Verbindung, spezifische Wellenlänge des Lichtes herauszufiltern. Eine Verbindung mit einem K-Wert von 1 reduziert das durchgehende Licht in einer Konzentration von 0,1 S und einer Schichtdicke von 1 cm auf 10% des eingefallenen Lichtes. So reduziert eine Ver bindung mit einem K = 10 bei einer gegebenen Wellenlänge in einer Konzentration von 0,1% die durchgehende Strahlung auf 10% der eingefallenen Strahlung bereits bei einer Schichtdicke von nur 1 mm.
Die Verfahrensprodukte der vorliegenden Erfindung haben die folgende allgemeine Formel:

in der As und Aa Benzolringe bedeuten, die mindestens eine Sulfonsäure- oder Carbonsäuregruppe sowie gegebenenfalls weitere Substituenten tragen, und R ein ali phatischer oder cycloaliphatischer Rest, vorzugsweise ein niedermolekularer Alkylrest mit 1 bis 6 Kohlenstoffatomen ist.
Die bevorzugt herstellbaren Verbindungen der vorliegenden Erfindung haben die folgende allgemeime Formel:

in der (Ar)l und (Ar)2 Benzolringe, Z die Reste - SO3M oder -COOM sind, n und m den Wert 1 bis 3 besitzen, R ein niedermolekularer Alkylrest mit 1 bis 6 Kohlenstoffatomen und M Wasserstoff oder ein salzbildendes Kation ist, vorteilhaft ein Alkalimetallion, wie Natrium, Kalium, Lithium oder Ammonium bzw. Der Benzolring kann zusätzlich zu der Salz- oder salzbildenden Gruppe andere Substituenten enthalten, die nicht chromophor sind. Diese Substituenten können folgende Reste sein : Hydroxyl, Halogen, Alkyl, substituiertes Alkyl, Alkenyl, substituiertes Alkenyl, Cyan, Alkylsulfonyl (z. B. -SO2CH3), Carboxamido oder Sulfonamido.
Chromophore Gruppen, wie beispielsweise Nitro-, Azo-, Azoxy- oder andere Gruppen sollten vermieden werden.
Wenn ein Oxyrest (z. B. ein Hydroxy-, Alkoxy- oder Alkenoxy-Rest) auxochrom oder bathochrom wirksam ist, sollte nicht mehr als ein solcher Rest vorhanden sein, weil das Absorptionsmaximum der Verbindungen mit 2 oder mehr Oxyresten in den meisten Fallen zu nahe, wenn nicht gar in sichtbarem Bereich des Spek drums liegt, wodurch sie dazu neigen, die Wirksamkeit dieser Verbindungen als Sonnenlicht filLernde Stoffe zu vermindern.
Es ist auch vorteilhaft, wenn die Erfindung gemäss verwandten und hergestellten Verbindungen keine Aminoreste, msbesondere direkt an den Benzolring gebundene, enthalten, weil diese Verbindungen im allgemeinen niedrigere Lichtbeständigkeit besitzen, cbgleich ihre Absorptionseigenschaften befrie digend sein können.
Das erfindungsgemässe VeRahren zur Herstellung der Verbindungen der Formel I ist dadurch gekennzeichnet, dass eine Säure der Formel A-CHO oder ein Salz davon mit einer Säure der Formel

bzw. einem Salz davon kondensiert wird.
Die Erfindung bezieht sich somit auf die Kondensation eines Sulfo- oder Carboxybenzolaldehyds, ge wünschtenfalls in Form seines Natriumsalzes, mit einem Sulfo- oder Carboxyphenly-(R-substituierten)-hydrazin.
Die Kondensation kann in einem wässrigen Medium, besonders wenn die Ausgangsstoffe löslich in Wasser sind, oder in jedem ausgewählten Lösungsmittelmedium durchgeführt werden. Das Verfahrensprodukt kann in üblicher Weise durch Destillation, Extraktion, Kristallisation oder Fällung isoliert werden. Im allgemeinen ergibt das ein- bis zweistündige Erhitzen der Ausgangsstoffe mit dem Aldehyd, der in Form des Natriumsalzes vorliegt, unter Rückfluss hohe Ausbeuten.

Eine der Methoden zur Herstellung der Hydrazinkomponenten besteht darin, N-Alkylanilin zu nitrosieren, zu dem entsprechenden Hydrazin mit Zink in Essigsäure zu reduzieren und schliesslich mit Oleum zu sulfonieren. In alternativer Weise können die N-Alkylaniline zuerst sulfoniert und dann nitrosiert und reduziert wer- den. Wenn die N-Alkylaniline in para-Stellung einen Substituenten, wie Halogen oder einen Alkyl-, Cyan-, Sulfonyl-, Carbalkoxy- oder einen ähnlichen Rest enthalten, findet die Sulfonierung in ortho-Stellung statt.
Wenn diese Substituenten in ortho- oder meta-Stellung stehen, findet die Sulfonierung in para-Stellung statt.

Die Carboxyphenylhydrazine können in gleicher Weise durch Nitrosieren und Reduzieren der entsprechenden Carboxyaniline hergestellt werden.
In den folgenden Beispielen sind die Teile Gewichtsteile, wenn nichts anderes angegeben wurde.
Beispiel I
In einen Drei-Liter-Kolben, der mit Rührer, Thermometer, Rückflusskühler und Heizmantel ausgestattet war, wurden folgende Ingredientien gegeben: 42,3 g 1-Methyl-1-(p-sulfophenyl)-hydrazin (Molekulargewicht 188, Kmax = 69,2 bei 253 m,), 52,5 g Natriumsalz des 2-Sulfobenzaldehyds (88,2 % rein; Molekulargewicht 186),
1200 ml Äthanol und
30,6 g Natriumacetatkristalle.
Die Mischung wurde zweieinhalb Stunden lang unter Rückflusskühlen gerührt und dann in einem Eisbad auf 10 C ab,,ekühlt. Die erhaltenen Kristalle wurden abfiltriert und der Filterkuchen mit 240 ml kaltem Ethanol gewaschen. Der abgepresste Filterkuchen wurde dann in 750 ml Wasser aufgeschlämmt, mit einem Teelöffel voll Aktivkohle (entfärbend wirkender Kohlenstoff) versetzt und filtriert. Zu dem Filtrat wurde dann bei Zimmertemperatur so viel Isopropanol gegeben, bis die Bildung von Kristallen beobachtet wurde.
Die erhaltene Masse wurde dann auf 10 C abgekühlt, filtriert und bei 40 C im Vakuumofen getrocknet. Es wurden 77 g 1-Methyl-1-(p-sulfophenyl)-2-sulfophenyl- hydrazon-Natriumsalz erhalten, mit einem Kmax gleich 70 bei 360 m,te.
Beispiel 2
Das Beispiel 1 wurde mit der abändernden Mass- nahme wiederholt, dass anstelle des Natriumsalzes des 2-Sulfobenzaldehyds die gleiche Gewichtsmenge (das heist 4e,3 g 100Yriges Produkt) 3-Sulfobenzaldehyd verwendet wurden. Das Produkt war l-Methyl-l-(psulfophenyl)-2-sulfopenylhydrazon-Natriu
Beispiel 3
Das Beispiel 1 wurde mit der abändernden Mass- nahme wiederholt, dass eine äquivalente Menge, bezogen auf 100 Siges Produkt, 2,4-Disulfobenzaldehyd- Natriumsalz anstelle des Sulfobenzaldehyds des Beispiels 1 verwendet wurden. Das erhaltene Produkt war 1-methyl-1-(2,4-disulfophenyl)-2-sulfophenylhydrazon Natriumsalz.
Beispiel 4
Das Beispiel 2 wurde mit der abändernden Mass- nahme wiederholt, dass anstelle des p-sulfonierten Hydrazins äquimolare Mengen des o-sulfonierten Produkts verwendet wurden. Das erhaltene Produkt war das Natriumsalz des 1-Methyl-1-(0-sulfophenly)-2-sulfo- phenylhydrazons .
Beispiele 6 bis 25
In den folgenden Beispielen wurde das Beispiel 1 mit der abändernden Massnahme wiederholt, dass anstelle des 2-Sulfobenzaldehyds die folgenden Benzaldehyde in äquivalenten Mengen angewendet wurden:

Beispiel A
Das Produkt des Beispiels 1 wurde in eine neutrale Detergent-Komposition in Form eines Stabes eingearbeitet, welche als Hauptbestandteil einen Kokosöl säureester des Natrium-Isäthionats enthielt. Die verwendete Menge an Produkt des Beispiels 1 betrug 0,2 Gew.,o, bezogen auf das Gesamtgewicht des Detergent-Stabes. Der Stab wurde dann getrennt beim Baden von fünf Individuen verwendet. Danach wurden die fünf Personen zwei Stunden lang der Nachmittags-Sommersonne ausgesetzt. Eine gleichgrosse Gruppe von fünf Personen verwendeten einen ähnlichen Detergents-Stab, der jedoch kein Produkt des Beispiels 1 enthielt. Diese Individuen wurden genau wie die erste Gruppe der Sommersonne ausgesetzt.
Jede Person der zweiten Gruppe zeigte nach der Sonnenbestrahlung Rötungen dr Haut, während keine Person der ersten Gruppe irgnedeinen sichtbaren Beweis für Sonnenbrand zeigte.
Die durchgeführten Versuche zeigen nicht nur die ausgezeichneten Sonnenlicht filternden Eigenschaften der Verbindung des Beispiels 1, sondern auch seine hervorragende Substantivität für menschliche Haut, die beim Baden mit dem Detergent-Stück behandelt wurde.
Beispiel B
Das Verfahren des Beispiels A wurde mit der ab ändernden Massnfhme wiederholt, dass anstelle eines Detergernt-Stücks ein konventionelles Stück Seife (pH ungefähr 10) verwendet wurde, das 0,3 % des Produkts des Beispiels 1 enthielt. Vergleichbare Ergebnisse wie in Beispiel A wurden erhalten.
Beispiel C
Das Produkt des Beispiels 1 wurde als 1 Ogewichts- prozentige Lösung in Wasser hergestellt. In dieser Lö- sung wurde eine Probe Cellulosetextilmaterial eingetaucht, die vorher mit einem dunkelroten Baumwollfarbstoff gefärbt worden war. Nach dem Entfernen und Trocknen der Probe wurde sie zusammen mit einer ähnlich gefärbten Probe, die jedoch nicht mit dem Produkt des Beispiels 1 behandelt war, in einen Fadeometer gegeben. Nach vierstündiger Belichtung zeigte die unbehandelte Probe ein starkes Abfallen der Fär- bung, während die behandelte Ware keine Veränderung der Färbung zeigte.

In general, substances that filter sunlight have been developed which provide quite satisfactory protection against the burning rays, but which allow the tanning rays to pass through. The most deleteriously burning wavelengths of sunlight are in the range of approximately 2900 to 3100 A. The action of these wavelengths on human skin causes the severe burns, redness, blistering and edema that are characteristic of many burns. This is undesirable from an asthetic point of view and at the same time represents a serious danger to human health. In order to overcome the effect of the burning rays of sunlight, various compounds have been used that cancel the burning rays.
The public assumes, and generally accepts, that tanning is a despicable phenomenon and has something to do with good health. The public has been educated to accept the numerous commercially available products which screen the scorching rays and transmit the tanning rays. In recent years it has generally been assumed that while tanning is useful in protecting the skin from continued exposure to the sun, it is associated with a general degradation of the skin from the chemical radiation.
Although the greatest number of people are not severely attacked by tanning rays, there are large numbers that are even sensitive to the tanning rays of the sun, so that irradiation at this wavelength poses great health risks to them. Thus, for those who want protection against both scorching and tanning of the sun's rays and for those who do not want to be tanned, there is a real need for a broad spectrum sunlight filtering agent which gives adequate protection to the skin, sufficiently stable is under the conditions of use and retains its effects for several hours, can easily be applied to the skin and cannot be easily removed, for example by simply washing with water.

Compounds have now been found which have outstanding filter properties for the burning and tanning rays of the sun, and in particular for the rays with wavelengths in the ultraviolet range from 2900 A to about 3500 A. These compounds have excellent K values as well as unexpected light resistance and stability to ultraviolet rays .
The K value is a measure of the effectiveness of the compound in filtering out specific wavelengths of light. A compound with a K value of 1 reduces the transmitted light in a concentration of 0.1 S and a layer thickness of 1 cm to 10% of the incident light. For example, a compound with a K = 10 at a given wavelength in a concentration of 0.1% reduces the radiation passing through to 10% of the incident radiation with a layer thickness of only 1 mm.

Example I.

Example A
The product of Example 1 was incorporated into a neutral detergent composition in the form of a rod, which contained a coconut oil acid ester of sodium isethionate as the main component. The amount of product of Example 1 used was 0.2% by weight, based on the total weight of the detergent stick. The stick was then used separately when bathing five individuals. The five people were then exposed to the afternoon summer sun for two hours. A group of five people of the same size used a similar detergent stick which, however, did not contain the product of Example 1. These individuals, like the first group, were exposed to the summer sun.
Each person in the second group showed reddening of the skin after exposure to the sun, while no person in the first group showed any visible evidence of sunburn.
The tests carried out show not only the excellent sunlight-filtering properties of the compound of Example 1, but also its excellent substantivity for human skin which was treated with the detergent bar while bathing.
Example B.
The procedure of Example A was repeated with the altering measure that a conventional bar of soap (pH approximately 10) containing 0.3% of the product of Example 1 was used instead of a detergent bar. Results comparable to those in Example A were obtained.
Example C
The product of Example 1 was prepared as a 10 percent by weight solution in water. A sample of cellulose textile material which had previously been dyed with a dark red cotton dye was immersed in this solution. After the sample had been removed and dried, it was placed in a fadeometer together with a similarly colored sample which, however, had not been treated with the product of Example 1. After four hours of exposure, the untreated sample showed a sharp drop in color, while the treated goods showed no change in color.
