Procédé de préparation de bases de Mannich de 1'a-6-deoxy-5-oxy-tétracycline
Cette invention concerne la préparation de nouveaux dérivés des a-6-déoxy-5-oxytétracyclines qui sont utiles pour l'administration orale et pour l'administration parentérale, savoir les bases de Mannich des a-6-déoxy-5oxytétracyclines et leurs sels d'addition d'acides correspondants pharmaceutiquement acceptables.
L'a-6-déoxy-5-oxytétracycline est un agent thérapeutique intéressant (voir brevet des Etats-Unis d'Amérique No 3200149) qui est extrêmement efficace contre les micro-organismes Gram positifs et Gram négatifs.
L'un des problèmes associés aux tétracyclines est la difficulté rencontrée quand on les administre par voie intraveineuse, intramusculaire, ainsi que par voie orale.
On préfère en général, pour l'administration intraveineuse ou intramusculaire, employer des solutions aqueuses des agents thérapeutiques. Malheureusement, l'a- 6- déoxy-5-oxytétracycline et les autres bases tétracyclines ne sont que peu solubles dans l'eau et ne sont par conséquent pas adaptables à une utilisation sous forme de doses aqueuses. On a dans le passé surmonté ce problême en employant des dérivés des tétracyclines solubles dans l'eau. Des dérivés préférés à cet égard ont été les bases de Mannich, qui sont préparées par condensation de la tétracycline avec du formaldéhyde et une amine primaire ou secondaire. Les bases de Mannich des tétracyclines ont été également aussi bien préparées à partir d'amino-acides.
Un autre avantage des bases de Mannich est le fait que dans de nombreux cas on constate qu'elles sont moins toxiques par voie parentérale que les tétracyclines parents correspondantes.
Les bases de Mannich des a-6-déoxy-5-oxytétracycli- nes ont une solubilité dans l'eau plus grande que le composé parent et elles présentent encore l'avantage que leurs sels acides ne s'hydrolysent pas rapidement en précipitant dans les milieux aqueux quand on les laisse au repos ou quand on ajuste le pH d'environ 3 à 8. De plus, les nouveaux composés obtenus selon la présente invention apparaissent comme étant absorbés rapidement et complètement dans le tractus gastrointestinal. Ils fournissent aux animaux la protection contre divers types d'infections contractées soit expérimentalement soit naturellement.
Les nouveaux composés sont particulièrement bien adaptés pour une administration parentérale, donnant des teneurs de sérum plus prolongées que l'antibiotique parent correspondant, quand on effectue une administration par voie intramusculaire. Leur solubilité améliorée dans l'eau dans un large domaine de pH les rend supérieurs à leurs antibiotiques parents pour l'administration intraveineuse de l'antibiotique. Les sels d'addition d'acides des nouveaux composés sont plus solubles dans l'eau que les formes bases libres, à la fois dans l'eau et dans les solvants organiques hydroxyliques tels que les alcanols inférieurs, le propylène-glycol, le glycérol et les solvants semblables. Ceci les adapte à une utilisation dans diverses préparations pharmaceutiques stérilisées.
Les seSs d'addition d'acides sont particulièrement bien absorbés dans le tractus gastrointestinal et ils fournissent des teneurs de l'antibiotique dans le sang satisfaisantes par administration orale ou parentérale.
Bien que tous les nouveaux composés obtenus soient intéressants pour des applications orales ou vétérinaires, dans le cas de l'utilisation parentérale pour des humains on préfère particulièrement l'amido-N-(pyrrolidinomé thyl)a-6-déoxy-5-oxytétracycline et l'amido-N-(morpho linométhyl)s-6-déoxy-5-oxytétracycline à cause du fait qu'elles sont bien exemptes de caractéristiques d'irritation à l'endroit infecté.
La présente invention a pour objet un procédé de préparation des nouveaux composés ayant la formule:

Le procédé selon la présente invention est caractérisé par la réaction de l'a-6-déoxy-5.oxytétracycline, ou bien d'un de ses sels d'addition d'acides, avec un composé ayant la formule:

R est tel qu'on l'a défini ci-dessus et Z est H, en présence de formaldéhyde ou d'un composé engendrant du formaldéhyde ou bien, autrement, par réaction de l'a-6 déoxy-5-oxytétracycline, ou d'un de ses sels d'addition avec ledit composé II ou un de ses sels d'addition d'acides, où Z est Y - CH2 - où Y est le chlore, le brome, le groupement hydroxyle, un groupement alcoxyle inférieur, le groupement S03 -. hydrosulfure, phtalimide, succinimide,
un groupement ester malonique d'alcoyle ou un groupement ester malonique d'acyle, en l'absence de formaldéhyde.
Le terme alcoyle inférieur tel qu'il est utilisé ici se rapporte aux groupements alcoyle ayant jusqu'à 6 atomes de carbone et pouvant être à chaîne droite ou ramifiée.
Le terme alcoyle comprend également les groupements alcoyle à chaîne droite et à chaîne ramifiée.
Sont donc compris parmi les composés de formule I les produits dimères dérivés de 2 moles d' -6-déoxy-5- oxytétracycline, 2 moles de formaldéhyde et 1 mole de pipérazine. La formule est convenablement représentée par:

où T est la fraction a-6-déoxy-5-oxytétracycline, ladite
fraction étant liée à travers le groupement 2-carboxamide
au groupement 1,4-bis-méthylènepipérazine. Les nou
veaux composés obtenus sont amphotères et ils forment
donc des sels à la fois avec les acides et les bases.
Les composés obtenus selon la présente invention ont
une solubilité dans l'eau supérieure à celle de l'antibioti
que parent correspondant et ils ont encore l'avantage que
leurs sels d'acides ne s'hydrolysent pas rapidement en
précipitant dans les milieux aqueux au repos ou bien
quand on ajuste le pH dans un domaine de 3 à 8.
Dans la première possibilité d'exécution du procédé,
on fait réagir l'antibiotique parent avec le formaldéhyde
ou un équivalent, par exemple, le paraformaldéhyde, et
un composé ayant au moins un groupement amine pri
maire ou secondaire. La réaction est effectuée par simple
contact de l'a-6-déoxy-S-oxytétracycline avec le formal
déhyde et l'amine désirée dans un milieu aqueux ou
alcoolique pouvant être modifié comme on le désire par
addition de solvants organiques solubles dans l'eau pour
améliorer la solubilité des divers corps réagissants.
Par
exemple, I'a- 6-déoxy-5 -oxytétracyd i ne employée sous for
me de la base libre anhydre est de préférence dissoute dans un solvant miscible dans l'eau, par exemple, le
méthanol, 1'méthanol, le dioxane, le tétrahydrofuranne, des
éthers d'alcoyle inférieur de l'éthylène-glycol ou du
diéthylène-glycol tels que le diméthyléther, le diméthyl
formamide, etc. On préfère employer suffisamment de
solvant pour maintenir l'antibiotique en solution quand
on l'ajoute au milieu réactionnel aqueux. Les corps réa
gissants peuvent être ajoutés simultanément ou indivi
duellement, dans un ordre quelconque.
On préfère en
général ajouter l'a- 6-déoxy- 5-oxytétracycline au mélange de formaldéhyde et du composé aminé puisque l'on obtient ainsi les meilleurs résultats. Autrement, on peut traiter d'abord l'antibiotique par le formaldéhyde, puis ajouter le composé aminé au mélange réactionnel. Avec ce dernier procédé il est dans quelque cas possible d'isoler le produit intermédiaire de la réaction de l'antibiotique et du formaldéhyde avant réaction avec le composé aminé, bien qu'il y ait assez peu d'avantages à suivre un tel procédé.
On a constaté qu'une agitation efficace du milieu réactionnel est particulièrement utile, en particulier au cours de l'addition de l'antibiotique parent, bien qu'elle ne soit pas essentiellc. On peut mener la réaction dans un grand domaine de température, par exemple d'environ -150 C à environ 800 C. L'intervalle de température préféré est d'environ flo à à environ 300 C.
Dans l'autre possibilité, on utilise des composés aminométhylés du type approprié pour la transaminométhylation, c'est-à-dire ceux ayant la formule Y - CH.2 - R dans laquelle R est tel qu'on l'a défini ci-dessus et Y est choisi dans le groupe composé du chlore, du brome, du groupement hydroxyle, des groupements alcoxyle inférieur, du groupement SO-, hydrosulfure, phtalimide, succinimide, des esters maloniques d'alcoyle et des esters maloniques d'acyle.
Les corps réactifs sont de préférence employés dans un rapport sensiblement équimoléculaire. Bien que l'on puisse employer des excès de formaldéhyde et du composé aminé choisi, aucun avantage appréciable n'apparait en faisant ceci. La formation du produit est presque instantanée lors du mélange des corps réagissants. On obtient fréquemment les produits sous forme d'huiles pouvant être cristallisées par des procédés classiques. Un tel procédé met en jeu l'addition de l'huile à un corps non solvant agité mécaniquement, après quoi on obtient le produit solide par des procédés classiques, par exemple, filtration. Des non-solvants convenables sont l'acétone, I'éther, les hydrocarbures aromatiques, le toluène, les hydrocarbures aliphatiques liquides tels que l'hexane, les alcanols, tels que l'isopropanol, etc.
D'autres procédés de cristallisation des produits huileux se suggèrent d'euxmêmes à ceux qui sont spécialistes de la technique.
Les produits obtenus selon la présente invention sont capables de former des sels soit avec les acides organiques soit avec les acides minéraux à cause de la présence de groupements basiques et acides. Représentatifs de tels acides sont l'acide chlorhydrique, sulfurique, phosphorique, acétique, butyrique, palmitique, oléique, citrique, gluconique, lactique, tartrique, ascorbique, pantothénique, les pénicilîlines, le demi-ester succinique du D-(-) thréo - 2 - dichloroacètamino - 1 -p-nitrophényl - 1,3-propane- diol, l'acide phytique, et l'acide maléique. Certains composés peuvent former des sels polyacides. Par exemple. l'amido-N-(lysinométhyl)-a- 6-déoxy- 5-oxytétracycline est capable de former un mono- un di- ou un trichlorhydrate à cause de la présence de 3 groupements amine.
De plus, à cause de la présence du groupement carboxyle libre, les composés dérivant des amino-acides peuvent former une variété de sels avec les bases soit minérales soit organiques. Des exemples de ces sels sont les sels de métaux alcalins, tels que les sels de sodium ou de potassium, le sel d'ammonium et les sels de métaux alcalino-terreux, tels que le sel de calcium, ainsi que les sels avec les amines organiques. De tels sels sont naturellement préparés par des procédés classiques connus de ceux qui sont spscialistes de la technique.
Les diverses formes de 1'a-6-déoxy-5-oxytétracycline peuvent être employées dans le présent procédé, par exemple, les sels d'addition d'acides ainsi que la base libre sous forme hydratée ou anhydre. Les composés aminés employés dans le présent procédé peuvent être utilisés sous forme de leurs sels d'addition d'acides ou bien sous forme du composé aminé libre. Si on utilise un sel de la tétracycline ou du composé aminé, on préfère en général employer les sels acides minéraux, dont un exemple est constitué par les sels de l'acide chlorhydrique, puisque ceux-ci sont aisément disponibles et donnent d'excellents résultats.
Ceux-ci comprennent naturellement les sels d'addition de l'acide phosphorique, sulfurique, nitrique, bromhydrique. et des acides semblables, en plus des sels d'addition de l'acide chlorhydrique. Comme cela est connu de ceux qui sont spécialistes de la technique, le produit obtenu correspondra aux formes choisies de l'a-6-déoxy-5-oxytétracycline et du composé aminé. Par exemple, quand on fait réagir la ss-alanine avec l'a-6- déoxy-5-oxytétracycline, le produit obtenu est la base libre. I'amidon-N-(ss-alaninométhyl) -6-déoxy-5-oxyté- tracycline.
Quand elle est employée dans le présent procédé, la lysine peut être utilisée sous l'une de ses formes optiquement actives, par exemple la lysine dextrogyre ou la lysine lévogyre, ainsi que le composé racémique, la lysine DL, ou bien sous forme d'un mélange des formes optiquement actives. On préfère en général employer la lysine L puisque l'acide amino L est la forme active biologiquement. On peut trouver le produit en résultant plus avantageux pour une utilisation dans le corps des animaux. Naturellement, le produit obtenu avec les autres formes de la lysine possède les propriétés intéressantes décrites précédemment. Le spectre de l'antibiotique in vitro des nouveaux composés obtenus correspond à celui de l'antibiotique parent à partir duquel il dérive.
Tel qu'il est utilisé dans le mémoire descriptif le symbole < ( w signifie oméga , et désigne l'atome de carbone terminal d'une chaîne alcoyle.
Pour l'administration parentérale par voie intramusculaire ou intraveineuse, la dose journalière est habituellement égale à environ 0,05 à 1,0 g. Pour les formulations intramusculaires, on utilise des concentrations dans l'intervalle de 50 à 100 mg/ml. Pour les administrations intraveineuses la concentration est environ 10 mg/ml.
Si on le désire, les nouveaux composés peuvent également être administrés oralement, bien que, comme cela est indiqué, on préfère l'administration parentérale.
Les exemples suivants illustrent l'invention.
Exemple I
Chlorhydrate d'amido-N-(Lysinométhyl)-a-6-déoxy-
5 -oxytétracycline
A 18.3 g (0,10 mole) de monochlorhydrate de lysine L dissous dans 100 ml d'eau, on ajoute 10 ml d'une solution aqueuse de formaldéhyde à 37 o/o On ajoute au mélange résultant 44,4 g (0,10 mole) d'a-6-déoxy-5-oxytétracycline anhydre dissous dans 500 ml de tétrahydrofuranne. Après mélange intime, le produit se forme en environ 15 minutes sous forme d'une couche huileuse qui, après séparation de la phase aqueuse, est ajoutée goutte à goutte à 3 litres d'alcool isopropylique agité. On récupère le produit par filtration, on le remet en suspension dans l'acétone, on le filtre et on le sèche à 650 C sous pression réduite.
Exemple ll:
On répète le processus de l'exemple I en employant, à la place de la lysine, des quantités équivalentes des amino acides suivants:

Exemple III:
A mid o-N-(ss-Alaninométhyl)-a-6-déoxy-5-oxytétracycline
A un ballon agité, on ajoute les corps réagissants suivants dans l'ordre:
eau 100 ml
ss-alanine 4,5 g
formaldéhyde à 37 oxo 10 ml
dimèthyléther d'éthylène-glycol 25 ml
eau . . ml
s < -6-déoxy-5-oxytétracycline dissoute
dans 250 ml de diméthyl éther
d'éthylène-glycol 22,2 g
Presque immédiatement après l'agitation, il se forme un solide gommeux. On sépare la gomme du mélange, on la met dans 500 ml de diméthyléther d'éthylène-glycol et on agite jusqu'à ce qu'il se produise un produit filtrable.
On obtient ensuite le produit par filtration suivie d'un lavage à l'éther.
Exemple IV:
Amido-N-(Glycinamidométhyl)-α-6-déoxy-
5-oxytétracycline
On répète le processus de l'exemple III en employant du glycinamide (3,7 g) à la place de la ss-alanine, on obtient le composé du titre de l'exemple.
Quand on remplace le glycinamide par les amides de la -alanine, de la lysine et de la sérine, on obtient les produits correspondants.
Exemple V:
Amido-N-(4'-(ss-hydroxyéthyl)pipérazinométhyl]- a-6-d éoxy-5-oxytétracycline
A une solution contenant 13 g de N-(ss-hydroxyéthyl)- pipérazine dans 500 ml d'isopropanol, on ajoute 3 g de paraformaldéhyde, et on chauffe le mélange à 600 C pendant 30 minutes jusqu'à dissolution complète. Après refroidissement de la solution à 400 C, on ajoute 22,2 g d'a-6-déoxy-5-oxytétracycline anhydre, et on laisse la réaction se poursuivre pendant trois heures tout en agitant et en faisant barboter un courant d'azote sec dans cette solution. On filtre ensuite la solution et on lave deux fois le cake de filtration par 200 ml d'isopropanol.
On met le cake en suspension dans 100 ml d'éther anhydre, on le filtre, on le lave trois fois par 500 ml d'éther anhydre et on le sèche sous vide.
Exemple VI..
Acétate d'amido-N-(pipéridinométhyl)-α-6-déoxy-
5-oxytétracycline
A de l'a-6-déoxy-5-oxytétracycline (22,2 g) dissoute dans 500 ml de tétrahydrofuranne, on ajoute, tout en agitant. de l'acétate de pipéridine (7,3 g) et 7 ml d'une solution de formaline à 37 /o, à une température de 200 C. On agite le mélange pendant deux heures, on recueille le précipité par filtration, on le lave par du tétrahydrofuranne et on le sèche sous vide à environ 400 C.
Exemple Vit.
Chlorhyd rate d'amido-N-(pyrrolidinométhyl)- a-6-déoxy-5-oxytétracycline
A 450 ml d'éthanol on ajoute successivement 3,60 ml d'une solution aqueuse à 37 % de formaldéhyde, 2,52 ml de pyrrolidine, et 13,2 g d'a-6-déoxy-5-oxytétracycline.
On agite le mélange résultant pendant quelques minutes à température ambiante et on recueille le solide qui se sépare de la solution (1,32 g) par filtration. On amorce le filtrat par une très petite quantité de ce corps solide et on agite à température ambiante pendant environ 2 heures 1/2.
L'amido-N-(pyrrolidinométhyl)-a-6-déoxy-5-oxytétracycline qui se sépare de la solution est filtrée et lavée successivement par du méthanol et de l'acétone. Le rendement est 12.7 g.
Analyse.
Calculé pour C2,H3lN308, 1/2 H2O: C = 60,43: H = 6,38: N = 7,83
Trouvé:
C = 60,49; H = 6,21: N = 7,82 L'amido-N-(pyrrolidinométhyl)-α-6-déoxy-5-oxytétra- cycline (12 g) est ajoutée à 260 ml d'eau. Le pH du mélange est ajusté à 1,9 par de l'acide chlorhydrique concentré et filtré. La lyophilisation du filtrat donne 13g d'un mélange du mono- et du dichlorhydrate d'amino-N (pyrrolidinométhyl)-α-6-déoxy-5-oxytétracycline.
Analyse.

Exemple Viii.
Chlorhydrate d' amido-N-(morpholinométhyl)- a-6-d éoxy-5-oxytétracycline
A 512 ml d'une solution 0,01 N d'acide chlorhydrique dans du méthanol, on ajoute 4,09 mi d'une solution aqueuse à 37 /o de formaldéhyde, 2,96 ml de morpholine, et 15 g d'a-6-déoxy-5-oxytétracycline. On ajuste le pH du milieu résultant à environ 4,7 avec de l'acide chlorhydrique concentré et la petite quantité (cad. 0,4 g) du corps cristallin qui se sépare de la solution est recueilli par filtration. Le filtrat est amorcé par une petite quantité de ces cristaux et agité pendant environ 3 heures à température ambiante.
Les cristaux d'amido-N-(morpholinomé- thyl)-a-6-déoxy-5-oxytétracycline qui se séparent de la solution à la fin de cette période sont filtrés et lavés successivement par du méthanol et de l'acétone.
Analyse.

Exemple IX:
Diacérate d'amido-N-(4'-méthylpipérazinométhyl) a-6-d éoxy-5-oxytétracycline
Dans 2 litres de méthanol on dissout, à température ambiante, de l'α-6-déoxy-5-oxytétracycline (22,2 g), 16,9 ml d'une solution formaline (à 37 /o) et du diacétate de N-méthylpipérazine (11,0 g). On laisse reposer le mélange réactionnel 8 heures au réfrigérateur et on le concentre ensuite sous pression réduite jusqu'à ce que le produit se sépare.
Le précipité obtenu est isolé, lavé par du méthanol et séché sous pression réduite à 400 C.
Exemple X.
Chlorhydrate d'amido-N-(2,6-diméthylmorpholino- ,n éthyl)-a-6-déoxy-5-oxytétracycline
De l'éther de chlorométhyl-méthyle (8,0 g) est ajouté goutte à goutte, à 1 5o C tout en agitant, à une solution de 2,6-diméthylmorpholine (8,7 g) et d'a-6-déoxy-5-oxytétracycline (44,4 g) dans 800 ml de tétrahydrofuranne. Après 2 heures, on isole le précipité, on le lave par du tétrahydrofuranne et de l'éther froids et on le sèche sous pression réduite.
Exemple Xl: Amido-N-(dibenzylaminométhyl)-α-6-déoxy-
5-oxytétracycline
De la dibenzylamine fraîchement distillée (9,9g) et de la formaline (à 37 O/o, 6,0ml) sont ajoutées à une suspension d'a-6-déoxy-5-oxytétracycline (11,1 g) dans 500 ml d'alcool de tert.-butyle et le mélange est agité à température ambiante pendant 30 minutes, puis mis à l'ébullition pendant 15 minutes. La solution chaude est ensuite filtrée, refroidie à température ambiante et le solide est séparé, lavé par de l'alcool de tert.-butyle et séché sous pression réduite.

Exemple Xlil:
A scorbate d'amido-N-(g-hydroxyéthylaminométhyl)- a-6-déoxy-5-oxytétracycline
Du formaldéhyde gazeux (6,0 g) est ajouté à un mélange de monoéthanolamine (12,2 g), d'acide ascorbique (35,2 g) et d'a-6-déoxy-5-oxytétracycline (88,8 g) dans
1500 ml de méthanol à 100 C.
On laisse le mélange reposer pendant I heure et on le concentre ensuite sous pression réduite à température ambiante. Le concentrat est ajouté à de l'éther sec (4000 ml), le produit est séparé, lavé par de l'éther et séché. A un mélange agité de 20 ml de méthanol, de ss-di- éthylaminoéthylamino-éthyle (5,8 g), d'acide acétique glacial (6,0 mi) et de formaldéhyde méthanolique (8,8 mi d'une solution à 18 /o), on ajoute de 1' < -6-déoxy-5-oxy- tétracycline (22,2 g) à 200 C en 90 minutes. On agite le mélange résultant pendant 30 autres minutes et on le concentre ensuite sous pression réduite. On ajoute le concentrat à I litre d'éther, on sépare le solide, on le lave par de l'éther et on le sèche.

On transforme le produit de l'exemple I en la base libre en le traitant par une solution aqueuse contenant une quantité équivalente de bicarbonate de sodium. La base libre ainsi obtenue est transformée en le dichlorhydrate par dissolution dans du méthanol contenant 2 équivalents de chlorure d'hydrogène. On obtient le produit par évaporation de la solution résultante sous pression réduite.
De manière similaire, les produits de l'exemple II sont transformés en le sel chlorhydrate désiré. D'autres sels d'addition d'acides sont préparés en remplaçant

This problem has in the past been overcome by employing water soluble tetracycline derivatives. Preferred derivatives in this regard have been the Mannich bases, which are prepared by condensation of tetracycline with formaldehyde and a primary or secondary amine. Mannich bases of tetracyclines have also been prepared from amino acids as well.
Another advantage of Mannich bases is the fact that in many cases they are found to be less toxic by the parenteral route than the corresponding parent tetracyclines.
The Mannich bases of α-6-deoxy-5-oxytetracyclines have greater solubility in water than the parent compound and they still have the advantage that their acid salts do not hydrolyze rapidly by precipitating in them. aqueous media when left to stand or when the pH is adjusted to about 3 to 8. Amido-N- (4 '- (ss-hydroxyethyl) piperazinomethyl] - a-6-deoxy-5-oxytetracycline

Amido-N- (morpholinomethyl) - a-6-deoxy-5-oxytetracycline hydrochloride
To 512 ml of a 0.01 N solution of hydrochloric acid in methanol is added 4.09 ml of a 37% aqueous solution of formaldehyde, 2.96 ml of morpholine, and 15 g of a -6-deoxy-5-oxytetracycline. Amido-N- (g-hydroxyethylaminomethyl) - a-6-deoxy-5-oxytetracycline scorbate
Formaldehyde gas (6.0 g) is added to a mixture of monoethanolamine (12.2 g), ascorbic acid (35.2 g) and α-6-deoxy-5-oxytetracycline (88.8 g ) in
1500 ml of methanol at 100 C.
The mixture is left to stand for 1 hour and then concentrated under reduced pressure at room temperature. The concentrate is added to dry ether (4000 ml), the product is separated, washed with ether and dried. A mid o-N- (aminor1lethyl) -a-6-deoxy-5-oxytetracycline
Methanolic formaldehyde (1.7 ml of an 18% solution) is added to a solution of α-6-deoxy-5-oxytetracycline (ammonium salt) (4.4 g) in 100 ml of methanol. The mixture is allowed to stand for 3 hours, the precipitate is separated. it is washed with ether and dried.

Has a stirred mixture of 20 ml of methanol, ss-di-ethylaminoethylamino-ethyl (5.8 g), glacial acetic acid (6.0 ml) and methanolic formaldehyde (8.8 ml of a solution to 18 / o), 1 '<-6-deoxy-5-oxy-tetracycline (22.2 g) is added at 200 ° C. over 90 minutes. Example XVI:
I, 4-bis (α-6-deoxy-5-oxytetracycline-2-carboxamido-
methyl) -piperazine
Piperazine (4.3g) and formalin (14ml of a 36.7 0 / o solution) are dissolved in 200ml of water at 150 C.

Example XVII:
The product of Example I is converted to the free base by treating it with an aqueous solution containing an equivalent amount of sodium bicarbonate. The free base thus obtained is converted into the dihydrochloride by dissolution in methanol containing 2 equivalents of hydrogen chloride. The product is obtained by evaporation of the resulting solution under reduced pressure.
Similarly, the products of Example II are converted to the desired hydrochloride salt. Other acid addition salts are prepared by replacing

It is dissolved in an aqueous solution containing one equivalent of sodium hydroxide. The resulting solution is frozen and dried under reduced pressure. the sodium salt is obtained in the form of a powder.

Salts of the novel compounds obtained with pharmaceutically acceptable acids and bases are particularly useful for the preparation of various dosage forms. The salts of the pharmaceutically unacceptable acids and bases are useful for the purification of the present compounds and also for the preparation of the pharmaceutically acceptable salts.
Example XIX:
The products of Examples VI-X, XIII and XV are converted into their respective free bases by treatment with an equivalent amount of sodium bicarbonate in aqueous solution. The free bases thus obtained are transformed into other acid addition salts by dissolution in methanol containing an equivalent of one of the following acids: hydrobromic acid, tartaric acid, gluconic, citric, palmitic, stearic, pantothenic, phytic , benzyl-penicillin, butyric acid and the succinic half-ester of A - (-) - threo-2-dichloroacetamino-1 -para-nitrophenyl-1, 3-propanediol.
Similarly, the free bases of examples V,
XI, XII, XIV and XVI are converted into the corresponding acid addition salts.
Example XX:
Following the procedure of Example XVIII, the free bases of the products of Examples V-XV are treated with sodium, potassium, calcium, or magnesium hydroxide to give the corresponding salts.
