Verfahren zur Herstellung eines neuen Cyclobutenderivates
Gegenstand der Erfindung ist ein Verfahren zur Herstellung des 1 -( thylaminomethyl)-5 -methoxy-benzocyclobutens der Formel

Die neue Verbindung besitzt wertvolle pharmakologische Eigenschaften und zeichnet sich gegenüber anderen Verbindungen ähnlicher Struktur, z. B. den in der Schweizer Patentschrift Nr. 436263 beschriebenen Benzocyclobutenverbindungen, aus. So weist sie insbesondere eine antitussive Wirkung an der Katze sowie eine selektiv hemmende Wirkung auf exspiratorische Aktivitäten auf, wie sich im Tierversuch, z. B. an der narkotisierten Taube nach Trachealverschluss, zeigt.
Ferner besitzt sie im Tierversuch, z. B. am Kaninchen, eine Antimorphinwirkung. Die neue Verbindung kann daher als Antitussivum und Analgeticum Verwendung finden. So zeigt sich beispielsweise bei intravenöser Verabreichung der erfindungsgemässen Verbindung in Form ihres Hydrochlorids in Dosen von 3-10 mg/kg an der Katze eine deutliche antitussive Wirkung und bereits in Dosen von 0,3 mg/kg an der Taube eine ausgesprochene hemmende Wirkung auf exspiratorische Aktivitäten, sowie bereits in Dosen von 0,3 mg/kg am Kaninchen eine deutliche morphinantagonistische Wirkung.
Die neue Verbindung ist aber auch ein wertvolles Zwischenprodukt für die Herstellung anderer nützlicher Stoffe, insbesondere von pharmakologisch wirksamen Verbindungen.
Das erfindungsgemässe Verfahren zur Herstellung der neuen Verbindungen ist dadurch gekennzeichnet, dass man eine Verbindung der Formel

mit einer Verbindung der Formel X'-CH=CH3 umsetzt, wobei einer der Reste X und X' für die Aminogruppe und der andere für eine reaktionsfähig veresterte Hydroxylgruppe stehen.
Reaktionsfähig veresterte Hydroxylgruppen sind z. B. Hydroxylgruppen, die mit starken anorganischen oder organischen Säuren, vorzugsweise mit Halogenwasserstoffsäuren, wie Chlor-, Brom- oder Jodwasserstoffsäure, Schwefelsäure, oder mit Arylsulfonsäure, wie Benzol- oder Toluolsulfonsäure, verestert sind.
Die genannte Reaktion kann in an sich bekannter Weise, vorzugsweise in Anwesenheit von Lösungs- und/ oder Kondensationsmitteln, z. B. basischen Kondensationsmitteln, bei tiefer, gewöhnlicher oder erhöhter Temperatur im offenen oder im geschlossenen Gefäss unter Druck durchgeführt werden.
Je nach den Verfahrensbedingungen und Ausgangsstoffen erhält man den Endstoff in freier Form oder in der ebenfalls in der Erfindung inbegriffenen Form seiner Salze. Die Salze des Endstoffs können in an sich bekannter Weise, z.B. mit Alkalien oder Ionenaustauschern in die freie Base übergeführt werden. Von der letzteren lassen sich durch Umsetzung mit organischen oder anorganischen Säuren, insbesondere solchen, die zur Bildung therapeutisch verwendbarer Salze geeignet sind, Salze gewinnen.
Als solche Säuren seien beispielsweise genannt: Halogenwasserstoffsäuren, Schwefelsäuren, Phosphorsäuren, Salpetersäure, Perchlorsäure; aliphatische, alicyclische, aromatische oder heterocyclische Carbon- oder Sulfonsäuren, wie Ameisen-, Essig-, Propion-, Bernstein-, Glykol-, Milch-, Äpfel-, Wein-, Zitronen-, Ascorbin-, Malein-, Hydroxymalein- oder Brenztraubensäure; Phenylessig-, Benzoe-, p-Aminobenzoe-, Anthranil-, p-Hydroxybenzoe-, Salicyl- oder p-Aminosalicylsäure, Embonsäure, Methansulfon-, Athansulfon-, Hydroxyäthansulfon-, Äthylensulfonsäure; Halogenbenzolsulfon-, Toluolsulfon-, Naphthalinsulfonsäuren oder Sulfanilsäure; Methionin, Tryptophan, Lysin oder Arginin.
Diese oder andere Salze der neuen Verbindung, wie z. B. die Pikrate, können auch zur Reinigung der erhaltenen Base dienen, indem man die Base in Salze überführt, diese abtrennt und aus den Salzen wiederum die Base freimacht. Infolge der engen Beziehung zwischen der Base in freier Form und in Form ihrer Salze sind im Vorausgegangenen und nachfolgend unter der freien Base sinn- und zweckgemäss, gegebenenfalls auch die entsprechenden Salze zu verstehen.
Für die erfindungsgemässen Reaktionen werden vornehmlich solche Ausgangsstoffe verwendet, die die oben erwähnten bevorzugten Verbindungen ergeben.
Die Ausgangsstoffe sind bekannt oder können nach an sich bekannten Methoden gewonnen werden.
Die neue Verbindung kann als Heilmittel, z. B. in Form pharmazeutischer Präparate, Verwendung finden, welche sie in freier Form oder in Form ihrer Salze in Mischung mit einem für die enterale oder parenterale Applikation geeigneten pharmazeutischen organischen oder anorganischen, festen oder flüssigen Trägermaterial enthalten. Für die Bildung desselben kommen solche Stoffe in Frage, die mit den neuen Verbindungen nicht reagieren, wie z. B. Wasser, Gelatine, Lactose, Stärke, Stearylalkohol, Magnesiumstearat, Talk, pflanzliche Öle, Benzylalkohole, Gummi, Propylenglykole, Vaseline, Cholesterin oder andere bekannte Arzneimittelträger.
Die pharmazeutischen Präparate können z. B. als Tabletten, Dragees, Kapseln oder in flüssiger Form als Lösungen, Suspensionen oder Emulsionen vorliegen. Gegebenenfalls sind sie sterilisiert und/oder enthalten Hilfsstoffe, wie Konservierungs-, Stabilisierungs-, Netzoder Emulgiermittel, Lösungsvermittler oder Salze zur Veränderung des osmotischen Druckes oder Puffer. Sie können auch andere therapeutisch wertvolle Substanzen enthalten. Die pharmazeutischen Präparate werden nach üblichen Methoden gewonnen.
Die neuen Verbindungen können auch in der Tiermedizin, z. B. in einer der oben genannten Formen, oder in Form von Futtermitteln oder von Zusatzmitteln für Tierfutter verwendet werden. Dabei werden z. B.
die üblichen Streck- und Verdünnungsmittel bzw. Futtermittel angewendet.
Die Erfindung wird in den folgenden Beispielen näher beschrieben. Die Temperaturen sind in Celsiusgraden angegeben.
Beispiel 1
Zu einer Lösung von 1,6 g l-(Aminomethyl)-5methoxy-benzocyclobuten und 4,8 g wasserfreier Pottasche in 10 ml Isopropanol tropft man unter Eiskühlung innert 2 Stunden 1,56 g Ethyljodid in 10 ml Isopropanol. Man rührt 12 Stunden weiter bei Raumtemperatur, filtriert ab und dampft das Filtrat ein. Der teilweise feste Rückstand wird in Äther aufgenommen und das unlösliche quaternäre Salz abfiltriert. Das l-(Aminomethyl)-5-methoxy-benzocyclobuten, 1 -(Äthylaminome- thyl)-5-methoxy-benzocyclobuten und l-(Diäthylamino- methyl)-5-methoxy-benzocyclobuten enthaltende Filtrat wird eingedampft und der Rückstand gaschromatographisch an einer 3 % SE 30-Kolonne (Silicongummi auf Chromport XXX 60/80 Mesh silanisiert) in seine Komponenten aufgetrennt.
Man erhält so das l-(iSXthylamino- methyl)-5-methoxy-benzocyclobuten. Dieses wird in Essigester gelöst und mit in Essigester gelöstem Chlorwasserstoff versetzt, wobei das l-(Sithylaminomethyl)-5- methoxy-benzocyclobuten-hydrochlorid der Formel

als farbloses Salz ausfällt. Durch Umkristallisieren aus Athanol-Essigester erhält man farblose Kristalle vom F. 169-1710
Beispiel 2
10,0 g l-(p-Toluolsulphonyloxymethyl)-5-methoxy- benzocyclobuten und 100 ml absolutes Äthylamin werden bei 0 in ein Bombenrohr eingeschlossen und dann 2 Stunden auf 160-170l erwärmt. Das Reaktionsgemisch wird dann abgekühlt und auf Eis gegossen. Das Reaktionsprodukt wird mit Äther extrahiert.
Der mehrmals mit Wasser gewaschene Atheranteil wird über Natriumsulfat getrocknet, filtriert und eingedampft. Das zurückbleibende braune Öl wird in Essigester gelöst und mit Chlorwasserstoff in Essigester versetzt. Dabei kristallisiert das 1 (Äthylaminomethyl)-5-methoxy-benzo- cyclobuten-hydrochlorid aus, welches mit dem in Beispiel 1 beschriebenen Produkt identisch ist.
Das als Ausgangsmaterial verwendete l-(p-Toluolsulphonyloxymethyl)-5-methoxy-benzocyclobuten kann z. B. wie folgt erhalten werden:
20 g 1 - Cyano-5-methoxy-benzocyclobuten und 300 ml 20 %ige wässrige Natronlauge werden 12 Stunden am Rückfluss erhitzt. Unter Eiskühlung wird konzentrierte Salzsäure zugetropft, wobei die 5-Methoxybenzocyclobuten-l-carbonsäure ausfällt. Sie wird abgenutscht und dann in Methylenchlorid aufgenommen.
Die organische Lösung wird viermal mit Wasser gewaschen, über Natriumsulfat getrocknet, filtriert und eingedampft. Man erhält so rohe, kristalline 5-Methoxybenzocyclobuten-l-carbonsäure, die für die folgende Umsetzung ohne Reinigung verwendet werden kann, F. 93-96 (aus Athanol/Wasser).
38 g rohe 5-Methoxy-benzocyclobuten- 1 -carbon- säure in 230 ml absolutem Tetrahydrofuran werden unter Eiskühlung und Rühren zu 10,3 g Lithiumaluminiumhydrid in 340 ml absolutem Tetrahydrofuran getropft. Das Reaktionsgemisch wird unter Stickstoff 5 Stunden bei Raumtemperatur gerührt. Dann tropft man bei Eiskühlung und kräftigem Rühren 10,3 ml Wasser, 10,3 ml 15 %ige wässrige Natronlauge und schliesslich 30,9 ml Wasser zu. Vom anorganischen Niederschlag wird abgenutscht. Die Tetrahydrofuranlösung wird über Natriumsulfat getrocknet, filtriert und eingedampft. Die Destillation des öligen Rückstandes am Hochvakuum liefert das l-(Hydroxymethyl)-5-me- thoxy-benzocyclobuten als farbloses Ö1 vom Kp. 0,2 100 bis 1010.
31,9 g 1 -(Hydroxymethyl) -5-methoxy-benzocyclo- buten werden in 200 ml absolutem Pyridin gelöst und bei Eiskühlung portionenweise mit 37,2 g fein pulverisiertem p-Toluolsulfonylchlorid versetzt. Dabei soll die Innentemperatur 150 nicht übersteigen. Das Reaktionsgemisch wird dann 24 Stunden bei Raumtemperatur stehengelassen. Dann giesst man das Reaktionsgemisch auf Eis. Den ausgeschiedenen p-Toluolsulphonsäureester extrahiert man mit Äther. Die ätherische Phase wird mit 2n Salzsäure extrahiert, mit 5 %iger Natriumbicarbonatlösung und mit Wasser gewaschen, über Natriumsulfat getrocknet, filtriert und eingedampft. Man erhält so das 1 -(p-Toluolsulphonyloxymethyl)-5 -methoxy- benzocyclobuten als kristallinen Rückstand, das ohne weitere Reinigung weiter umgesetzt werden kann [F. 64 bis 670 (aus Äthanol/Wasser)].
