Verfahren zur Herstellung von N-substituierten Phenothiazinen
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung neuer 10-(3 -N-Formyl-N-methylaminopropyl)-phenothiazine, welche wertvolle pharmakologische Eigenschaften besitzen.

in welcher X Wasserstoff oder Chlor bedeutet, sind bis jetzt noch nicht bekannt geworden.
Wie überraschenderweise gefunden wurde, besitzen die erfindungsgemässen Verbindungen wertvolle pharmakologische Eigenschaften: Sie zeigen bei gewissen Nagern bei geringer Toxizität eine dämpfende Wirkung auf das Zentralnervensystem, und sie verringern das Schmerzempfinden. Diese Eigenschaften kennzeichnen die Verbindungen als geeignet zur Behandlung von gewissen Formen von Geisteskrankheiten.
Verbindungen der Formel I werden hergestellt durch Formylierung der entsprechenden l0-(3-Methylamino- propyl)-phenothiazine. Folgende Verfahren seien genannt: (1) Umsetzung des 10-(3-Methylaminopropyl)-phe- nothiazins oder dessen 3-Chlorderivats, zunächst mit einem niederen Alkylmagnesiumhalogenid, wie z.B. Methylmagnesiumjodid, und anschliessende Umsetzung des entstandenen komplexen Magnesiumsalzes mit einem niederen Alkylester der Ameisensäure, z.B. mit Ameisensäuremethylester.

Die Umsetzung des Phenothiazinderivats mit dem Grignard Reagens wird in einem nicht-polaren wasserfreien organischen Medium, wie beispielsweise in einem ätherischen Lösungsmittel, vorzugsweise in Diäthyläther, durchgeführt. Das gleiche Lösungsmittel ist für die Umsetzung des entstandenen komplexen Magnesiumsalzes mit dem niederen Ameisensäurealkylester geeignet: vorteilhaft verwendet man Ameisensäure-methylester. Beide Reaktionsstufen werden vorzugsweise bei der Rückflusstemperatur des Lösungsmittels ausgeführt.
(2) Formylierung von 10-(3-Methylaminopropyl)- -phenothiazin oder dessen 3-Chlorderivat mittels Ameisensäure und eines geeigneten niederen Fettsäureanhydrids, beispielsweise Essigsäureanhydrid. Diese Umset- zung wird gemäss W.J. Gensler, J. Org. Chem. 21, 336 bis 339 (1956) durchgeführt, indem das Phenothiazinderivat in Ameisensäure gelöst und diese Lösung mit z.B. Essigsäureanhydrid bei einer Temperatur von vorzugsweise 55 - 600 umgesetzt wird.
In Abänderung dieses Verfahrens kann eine geeignete niedere Fettsäure, beispielsweise Essigsäure, anstelle eines niederen Fettsäureanhydrids verwendet werden.
(3) Formylierung von 10-(3 -Methylaminopropyl)- -phenothiazin oder dessen 3-Chlorderivat mit Chloral oder Chloralhydrat gemäss F.F. Blicke und Chi-jung Lu, J. Am. Chem. Soc. 74, 3933 - 3934 (1952). Bei der Durchführung dieser Umsetzung wird beispielsweise Chloral langsam zu einer gekühlten Lösung einer der oben angegebenen Ausgangsverbindungen in Chloroform zugegeben. Das sich ergebende Gemisch wird einige Stunden bei Raumtemperatur gerührt und dann kurze Zeit erhitzt.
(4) Formylierung von 10-(3-Methylaminopropyl)- -phenothiazin oder dessen 3-Chlorderivat mit Ameisensäure in Gegenwart eines Tri(nieder)alkylamins, vorzugsweise des Trimethylamins. Dabei wird die Ameisensäure beispielsweise einer Lösung des Ausgangsphenothiazins im tert. Amin unter Kühlung zugefügt und dies sich ergebende Gemisch mehrere Stunden am Rückfluss gekocht.
Die jeweils erfindungsgemäss gewonnenen Reaktionsgemische werden nach bekannten Verfahren aufgearbeitet.
Die verwendeten Ausgangsmaterialien sind entweder im Handel erhältlich oder können nach an sich bekannten Methoden leicht hergestellt werden.
Der hierin verwendete Ausdruck niederes Alkyl bezeichnet geradkettige gesättigte aliphatische Gruppen mit 1 bis 4 Kohlenstoffatomen. Beispiele solcher Alkylgruppen sind die Methyl-, Äthyl-, n-Propyl- und n-Butylgruppe. Der hierin verwendete Ausdruck niedere Fettsäure bedeutet z.B. Essigsäure, Propionsäure oder Buttersäure.
In den nachstehenden Beispielen sind die Temperaturen in Celsiusgraden angegeben.
Beispiel I 10-(3-N-formyl-N-methylanlin.opropyl)-phenothiazin
In einen 1 Liter fassenden 3-Halskolben, welcher mit Rührer, Rückflusskühler, Trocknungsrohr und zusätzlichem Trichter versehen ist, werden 1,3 g Mg-Späne eingebracht und letztere mit 25 ml trockenem Äther überschichtet. Zu diesem Gemisch werden dann 7,8 g frisch destilliertes Methyljodid in 150 ml trockenem Äther tropfenweise während einem Zeitraum von einer halben Stunde zugegeben. Das Gemisch wird während 30 Minuten am Rückfluss erhitzt, dann auf Zimmertemperatur abgekühlt, hierauf eine Lösung von 13,7 g 10-(3-N-Methylaminopropyl)-phenothiazin in 200 ml trockenem Äther während 40 Minuten zugetropft und das Reak tionsgemisch während 4 Stunden am Rückfluss erhitzt.
Nach dem Abkühlen werden 6,05 g Methylformiat wäh rend 10 Minuten zugetropft. Dann wird das Reaktionsgemisch während 12 Stunden gerührt und hierauf in eine Ammoniumchloridlösung gegossen. Die Ätherschicht wird abgetrennt und wässrige Phase mit Äthylacetat extrahiert. Die vereinigten organischen Phasen werden über Na > SO4 getrocknet, filtriert und die Lösungsmittel im Vakuum entfernt. Die Destillation des Rückstandes ergibt ein gelbes öl vom Kp. 2260/0,15 mm Hg.
Beispiel 2 1O-(3-N-Foml-N-rnethyiaminop'opyl)-phenothicizin
Zu der Lösung von 3,5 g (0,0125 Mol) 10-(3-N-Methylaminopropyl)-phenothiazin in 62,5 ml 98 - 100%iger
Ameisensäure werden unter Rühren und Kühlen 20,5 ml
Essigsäure anhydrid gegeben, dann wird während 15 Mi nuten auf 55 - 600 erwärmt. Das Reaktionsgemisch wird im Vakuum eingedampft. Der Rückstand wird gekühlt und mit 5 ml Wasser und 5 ml 3n NaOH versetzt. Das
Gemisch wird mit Benzol extrahiert, die Benzolphase über Na2SOG getrocknet und im Vakuum eingeengt. So erhält man 2 g eines Öls, das nach I.R.-Spektrum das gewünsch te Produkt darstellt.
Beispiel 3 10-(3-N-Formyl-N-methylaminopropyl)-phenothîazin
Zu 6,85 g (0,025 Mol) 10-(3-N-Methylaminopropyl) -phenothiazin in 125 ml 98 - 100 ,1Oiger Ameisensäure wer den unter Rühren und Kühlen 41 ml Essigsäure gegeben, anschliessend wird während 15 Minuten auf 55 - 600 erwärmt. Das Reaktionsgemisch wird im Vakuum eingedampft. Der Rückstand wird gekühlt und mit 10 ml Wasser und anschliessend mit 10ml NaOH versetzt. Das Gemisch wird mit Benzol extrahiert, die Benzollösung wird über Na2SO4 getrocknet und im Vakuum eingedampft, wobei das gesuchte Produkt als öl erhalten wird, dessen I.R.-Spektrum mit demjenigen einer authentischen Probe übereinstimmt.
Beispiel 4 10-(3-N-Formyl-N-methylaminopropyl) -phenothiazin
Zu 3,4 g (0,0125 Mol) 10-(3-N-Methylaminopropyl) -phenothiazin in 6 ml Trimethylamin werden unter Kühlung 8 ml 98%iger Ameisensäure zugetropft. Anschliessend kocht man das Reaktionsgemisch während 9 Stunden am Rückfluss, engt es im Vakuum ein, kühlt ab, fügt 10 ml Wasser bei und extrahiert mit Äther. Der Ätherextrakt wird über NaSO4 getrocknet und eingedampft.
Der ölige Rückstand wird als das gesuchte Produkt identifiziert (I.R.-Spektrum).
Beispiel 5
Verwendet man die in obenstehenden Beispielen äquivalenten Mengen von 3-Chlor- 10-(3-N-methylaminopro- pyl)-phenothiazin statt 10-(3-N-Methylaminopropyl)-phe- nothiazin, so erhält man in analoger Weise das 3-Chlor - 10- (3- N - formyl - N - methylaminopropyl)-phenothiazin, Smp. 93 - 940C.
Process for the preparation of N-substituted phenothiazines
The present invention relates to a process for the preparation of new 10- (3-N-formyl-N-methylaminopropyl) -phenothiazines which have valuable pharmacological properties.

As has surprisingly been found, the compounds according to the invention have valuable pharmacological properties: in certain rodents they show a dampening effect on the central nervous system with low toxicity, and they reduce the sensation of pain. These properties characterize the compounds as useful in the treatment of certain forms of mental illness.
Compounds of the formula I are prepared by formylating the corresponding 10- (3-methylaminopropyl) phenothiazines. The following processes may be mentioned: (1) Reaction of 10- (3-methylaminopropyl) -phenothiazine or its 3-chloro derivative, initially with a lower alkyl magnesium halide, e.g. Methyl magnesium iodide, and subsequent reaction of the complex magnesium salt formed with a lower alkyl ester of formic acid, e.g. with methyl formate.

The reaction of the phenothiazine derivative with the Grignard reagent is carried out in a non-polar anhydrous organic medium, such as, for example, in an ethereal solvent, preferably in diethyl ether. The same solvent is suitable for the reaction of the complex magnesium salt formed with the lower alkyl formate: it is advantageous to use methyl formate. Both reaction steps are preferably carried out at the reflux temperature of the solvent.
(2) Formylation of 10- (3-methylaminopropyl) -phenothiazine or its 3-chloro derivative using formic acid and a suitable lower fatty acid anhydride, for example acetic anhydride. According to W.J. Gensler, J. Org. Chem. 21, 336 to 339 (1956) by dissolving the phenothiazine derivative in formic acid and this solution with e.g. Acetic anhydride is reacted at a temperature of preferably 55-600.
As a modification of this procedure, a suitable lower fatty acid, for example acetic acid, can be used in place of a lower fatty acid anhydride.
(3) Formylation of 10- (3-methylaminopropyl) - -phenothiazine or its 3-chloro derivative with chloral or chloral hydrate according to F.F. Glances and Chi-jung Lu, J. Am. Chem. Soc. 74, 3933-3934 (1952). In carrying out this reaction, chloral, for example, is slowly added to a cooled solution of one of the above starting compounds in chloroform. The resulting mixture is stirred for a few hours at room temperature and then heated for a short time.
(4) Formylation of 10- (3-methylaminopropyl) - phenothiazine or its 3-chloro derivative with formic acid in the presence of a tri (lower) alkylamine, preferably trimethylamine. The formic acid is, for example, a solution of the starting phenothiazine in tert. Amine was added with cooling and the resulting mixture was refluxed for several hours.
The reaction mixtures obtained in each case according to the invention are worked up by known processes.
The starting materials used are either commercially available or can easily be prepared by methods known per se.
As used herein, lower alkyl denotes straight chain saturated aliphatic groups having 1 to 4 carbon atoms. Examples of such alkyl groups are the methyl, ethyl, n-propyl and n-butyl groups. The term lower fatty acid used herein means e.g. Acetic acid, propionic acid or butyric acid.
In the examples below, the temperatures are given in degrees Celsius.
Example I 10- (3-N-formyl-N-methylanlin.opropyl) -phenothiazine
1.3 g of Mg shavings are placed in a 1 liter 3-necked flask equipped with a stirrer, reflux condenser, drying tube and additional funnel, and the latter is covered with 25 ml of dry ether. 7.8 g of freshly distilled methyl iodide in 150 ml of dry ether are then added dropwise to this mixture over a period of half an hour. The mixture is refluxed for 30 minutes, then cooled to room temperature, then a solution of 13.7 g of 10- (3-N-methylaminopropyl) phenothiazine in 200 ml of dry ether is added dropwise over 40 minutes and the reaction mixture over 4 hours heated to reflux.
After cooling, 6.05 g of methyl formate are added dropwise during 10 minutes. The reaction mixture is then stirred for 12 hours and then poured into an ammonium chloride solution. The ether layer is separated off and the aqueous phase is extracted with ethyl acetate. The combined organic phases are dried over Na> SO4, filtered and the solvents are removed in vacuo. Distillation of the residue gives a yellow oil with a b.p. 2260 / 0.15 mm Hg.
Example 2 10- (3-N-Forml-N-methyiaminop'opyl) -phenothicizine
To the solution of 3.5 g (0.0125 mol) of 10- (3-N-methylaminopropyl) -phenothiazine in 62.5 ml of 98-100%

Acetic anhydride is added, then the mixture is heated to 55-600 for 15 minutes. The reaction mixture is evaporated in vacuo. The residue is cooled and 5 ml of water and 5 ml of 3N NaOH are added. The
The mixture is extracted with benzene, the benzene phase is dried over Na2SOG and concentrated in vacuo. This gives 2 g of an oil which, according to the I.R. spectrum, represents the desired product.
Example 3 10- (3-N-Formyl-N-methylaminopropyl) -phenothîazine
To 6.85 g (0.025 mol) of 10- (3-N-methylaminopropyl) -phenothiazine in 125 ml of 98-100.10 formic acid, 41 ml of acetic acid are added with stirring and cooling, then heated to 55-600 for 15 minutes . The reaction mixture is evaporated in vacuo. The residue is cooled and 10 ml of water and then 10 ml of NaOH are added. The mixture is extracted with benzene, the benzene solution is dried over Na2SO4 and evaporated in vacuo, the sought product being obtained as an oil whose I.R. spectrum corresponds to that of an authentic sample.
Example 4 10- (3-N-Formyl-N-methylaminopropyl) -phenothiazine
To 3.4 g (0.0125 mol) of 10- (3-N-methylaminopropyl) -phenothiazine in 6 ml of trimethylamine, 8 ml of 98% formic acid are added dropwise with cooling. The reaction mixture is then refluxed for 9 hours, concentrated in vacuo, cooled, 10 ml of water added and extracted with ether. The ether extract is dried over NaSO4 and evaporated.
The oily residue is identified as the product sought (I.R. spectrum).
Example 5
If the equivalent amounts of 3-chloro-10- (3-N-methylaminopropyl) -phenothiazine in the above examples are used instead of 10- (3-N-methylaminopropyl) -phenothiazine, the 3- Chlorine-10- (3-N-formyl-N-methylaminopropyl) -phenothiazine, m.p. 93-940C.
