Es wurde gefunden, dass die erfindungsgemäss er- hältlichen neuen Verbindungen der Formel I wertvolle Herz- und Kreislaufwirkungen aufweisen.
Das erfindungsgemässe Verfahren zur Herstellung der neuen tricyclischen Aminoalkyl-Derivate ist dadurch gekennzeichnet, dass man Amine der Formel II

Als reaktive Carbonsäure-Derivate kommen vor zugsweise die Säurehalogenide in Betracht. Die Um setzung der Verbindungen der Formeln II und III wird unter Erhitzen in einem inerten Lösungsmittel durch geführt, wobei zweckmässig geeignete Kondensations mittel zugesetzt werden; d. h. man sorgt in üblicher Weise dafür, dass das bei der Reaktion abgespaltene Wasser entfernt wird bzw. man fügt zur Bindung des abgespaltenen Halogenwasserstoffes Basen zu, z. B. einen Überschuss des Amins der Formel II.
Die in der ersten Stufe des Verfahrens erhaltenen Azo- methine bzw. Säureamide werden anschliessend zu den Verbindungen der Formel 1 reduziert, wobei insbeson dere komplexe Metallhydride (z. B. Lithiumaluminium- hydrid, Natriumborhydrid) oder aber die katalytische Hydrierung zur Anwendung kommen. Eine vorteilhafte Verfahrensvariante besteht darin, dass man die Konden sation der Verbindungen der Formeln II und IIIa mit der anschliessenden Reduktion kombiniert, d. h. die Carbonylverbindungen in Gegenwart der Amine redu ziert.
Die nachträgliche N-Alkylierung der Verfahrens produkte der Formel I (falls R5 = Wasserstoff) kann in üblicher Weise erfolgen, z. B. durch katalytische Hydrierung in Gegenwart entsprechender Aldehyde, durch Acylierung und anschliessende Reduktion mittels komplexer Metallhydride oder durch Umsetzung mit den entsprechenden Alkylhalogeniden.
Die basischen Verfahrensprodukte lassen sich mit Hilfe anorganischer oder organischer Säuren in bekann ter Weise in die entsprechenden Salze überführen. Als anorganische Säuren kommen z. B. in Frage Halogen wasserstoffsäuren, Schwefelsäure, Phosphorsäure, und als organische Säuren z. B. Essigsäure, Milchsäure, Maleinsäure, Weinsäure, Zitronensäure, Oxalsäure.
Die oben als Ausgangsprodukte erwähnten Aldehyde der Formel IIIa können z. B. durch Äthinylierung von Ketonen der Formel IV

Die als Ausgangsprodukte benötigten Carbonsäuren der Formel IIIb erhält man z.

lassen sich durch vorsichtige saure Hydrolyse in die ge wünschten Carbonsäuren der Formel IIIb, in der R., eine Hydroxylgruppe bedeutet, umwandeln. Unter kräf tigen Hydrolysebedingungen (z. B. Eisessig, 6n Salz säure) erreicht man eine Wasserabspaltung und Ver seifung unter Bildung von Carbonsäuren der Formel IIIb, in der R., und R.; zusammen eine Kohlenstoffbin dung bilden. Schliesslich kann man auch zunächst unter wasserfreien Bedingungen (z.
B. alkoholische Salzsäure) eine Wasserabspaltung bewirken, dann die C-C-Doppel- bindung reduzieren (z. B. mit amalgamiertem Alumi nium) und anschliessend unter wässrig-sauren Reak tionsbedingungen durch Verseifung die Carbonsäuren der Formel IIIb, in der R2 und R3 Wasserstoff bedeu ten, herstellen.
Im nachstehenden Beispiel ist das erfindungsgemässe Verfahren näher erläutert.
<I>Beispiel</I> N-[2-(6,1 1-Dihydro-dibenzo[b,e]oxepin-11-yliden)- äthyl]-l-phenoxy-2-hydroxy-3-amino-propan 11,8 g 6,11-Dihydro-dibenzo[b,e]oxepin-11-yliden- acetaldehyd (0,05 Mol) werden mit 7,6 g 1-Phenoxy-2- hydroxy-3-aminopropan (0,05 Mol) in 75 ml Benzol eine Stunde im Wasserabscheider erhitzt. Anschliessend dampft man ein, nimmt in 75 ml Methanol auf und ver setzt portionsweise mit 3,8 g Natriumborhydrid (0,1 Mol). Nach einstündigem Rückflusskochen wird dann eingedampft und mit Äther extrahiert.
Aus der getrock neten ätherischen Lösung fällt man durch Zugabe einer Lösung von Oxalsäure in Tetrahydrofuran das Oxalat des N-[2-(6,11-Dihydro-dibenzo[b,e]oxepin-l1-yliden)- äthyl]-1-phenoxy-2-hydroxy-3-amino-propans aus, das aus Isopropanol umkristallisiert werden kann, Die Aus beute des analysenreinen Produktes (Fp. 185 bis 187 C) beträgt 72 % d, Th.
(13,9 g), Der als Ausgangsverbindung verwendete 6,11-Di- hydrö-dibenzo[b,e]-oxepin -11- yliden-acetaldehyd wird durch Umlagerung von 11-Hydroxy-11-äthinyl-6,11-di- hydro-dibenzo[b,e]-oxepin (Fp. 93 bis 94 C; erhalten durch Umsetzung von 6,11-Dihydro-dibenzo[b,e]oxe- pin-11 mit Acetylen und Natrium in flüssigem Ammo niak) in schwefelsaurer, wässrig-alkoholischer Lösung (30 Minuten Reaktionszeit) in 82 % iger Ausbeute dar gestellt:
