<B>Verfahren</B> zur <B>Herstellung neuer Derivate der</B> 7-Amino-cephalosporansäure Gegenstand der Erfindung ist ein Verfahren zur Herstellung neuer therapeutisch wirksamer Derivate der 7-Aminocephalosporansäure der Formel I:

worin A, und A_ gegebenenfalls durch Niederalkoxy- gruppen oder Halogenatome substituierte gerade oder verzweigte Alkylreste mit höchstens 6 Kohlenstoffato- men bedeuten, mit der Massgabe, dass höchstens einer der Reste A, und A, durch Halogenatome substituiert ist, und ihrer Salze.
Als Alkylreste sind insbesondere zu nennen Methyl, Äthyl, Propyl, und iso-Propyl.
A, und A_ können gleich oder verschieden sein.
Als Halogenatome kommen Brom, Fluor, Jod, und vor allem Chlor in Betracht, als Niederalkoxygruppen vor allem N4ethoxy- oder Äthoxygruppen. Die Substitu- enten befinden sich vorzuesweise in ,3-Stellung zur Carbamylgruppe:
A, oder A_ ist beispielsweise eine 2-Halogenätlivl-, 2-Halogenpropyl- oder 1-Methyl- 2-halogenpropyleruppe. Es können auch mehrere Sub- stituenten in A, und A_ vorhanden oder nur der eine von A, und A_ substituiert sein.
Die Salze der neuen Verbindungen sind Metall salze, vor allem solche von therapeutisch anwendbaren Alkali- oder Erdalkalimetallen, wie Natrium, Kalium oder Calcium, oder Salze mit organischen Basen, z. B. Triäthylamin, N-Äthylpiperidin.
Die neuen Verbindungen besitzen antibakterielle Aktivität gegenüber grampositiven Bakterien, beispiels weise Bacillus subtilis, Bacterium megatherium und Staphylococcus aureus, insbesondere auch gegenüber penicillinresistenten Stämmen, vor allem aber auch ge- genüber grammnegativen Bakterien, z.
B. Escherichia coli, Klebsiella pneumoniae, Salmonella typhi murium. Sie können daher als Heilmittel in der Human- und Veterinärmedizin verwendet werden, ferner als Futter mittelzusätze und zur Konservierung von Nahrungsmit tel.
Die neuen Verbindungen werden nach an sich bekann ten Methoden hergestellt. So werden sie erhalten, wenn man Verbindungen der Formel 1I:

worin R für Wasserstoff oder die Gruppe CO-NH-A, steht, mit desacetylierenden Mitteln behandelt und die erhaltene O-Desacetylverbindung mit Isocyanaten der Formel 11I: A_--N#C==0 (III) umsetzt. Die Desacetylierung wird beispielsweise mit Acetylesterase durchgeführt.
Diese an sich bekannte Methode zur Herstelluna von Desacetyl-7-amino- cephalosporansäure kann wesentlich dadurch verbes sert werden, dass man nach Beendigung der enzymati schen Reaktion die Lösung auf pH 4,5 ansäuert, wobei die Säure aus der Lösung ausfällt und abgetrennt wer den kann.
Die Umsetzung der Desacetylverbindung mit dem Isocyanat wird vorzugsweise in Gegenwart einer star ken organischen Stickstoffbase, wie Triäthylamin, in einem inerten Lösungsmittel, beispielsweise Dimethyl- formamid, Methylenchlorid, Tetrahydrofuran, vorge nommen. Falls die Ausgangsverbindung bereits einen Substituenten A, enthält, kann die Gruppe A= des zur Umsetzung verwendeten Isocyanats verschieden von A, oder gleich A, sein.
Auf beliebiger Stufe des Verfahrens kann auch die Carboxylgruppe mit sauer leicht hydrolysierbaren Alkoholen, vorzugsweise Benzhydrylalkohol, verestert und auf einer späteren Stufe wieder abgespalten wer den. Die vorübergehende Überführung der Säure in den Ester eignet sich sehr gut zur Reinigung der Produkte.
Die neuen Verbindungen können als Heilmittel, z. B. in Form pharmazeutischer Präparate, Verwen dung finden. Diese enthalten die Verbindungen in Mischung mit einem für die enterale, topicale oder parenterale Applikation geeigneten pharmazeutischen organischen oder anorganischen, festen oder flüssigen Trägermaterial. Für die Bildung desselben kommen solche Stoffe in Frage, die mit den neuen Verbindun gen nicht reagieren, wie z. B.
Wasser, Gelatine, Lac- tose, Stärke, Stearylalkohol, Magnesiumstearat, Talk, pflanzliche Öle, Benzvlalkohole, Gummi, Propylengly- kol, Polyalkylenglykole. Vaseline, Cholesterin oder an dere bekannte Arzneimittelträger. Die pharmazeuti scher. Präparate können z. B. als Tabletten, Dragees, Salben, Creams, Kapseln oder in flüssiger Form als Lösungen, Suspensionen oder Emulsionen vorliegen.
Gegebenenfalls sind sie sterilisiert und./oder enthalten Hilfsstoffe, wie Konservierungs-, Stabilisierungs-. Netz- oder Emulgiermittel. Lösungsvermittler oder Salze zur Veränderung des osmotischen Drucks oder Puffer. Sie können auch andere therapeutisch wertvolle Substan zen enthalten. Die Präparate werden nach üblichen Methoden erhalten.
Die folgenden Beispiele erläutern die Erfindung ohne sie jedoch einzuschränken. Die Temperaturen sind in Celsiusgraden angegeben.
Bei der Dünnschichtchromatographie werden fol gende Systeme verwendet: System 1: n-Buatnol-Essigsäure (10:1), gesättigt mit Wasser.
System 1I: Essigester-Pyridin-Essigsäure-Wasser (60:20:6:11).
System III: n-Butanol-Pyridin-Essigsäure-Wasser (30:20:6:24).
Farblose Flecke auf violettem Unterrund nach Be sprühen mit Jodstärke-Essigsäure-Rea;ens.
<I>Beispiel 1</I> 112 m±! Desacetvl-7-amino-ceplialosporansäure werden in 9 nil abs. Dimethvlformamid mit 100 mg abs. Triä tlivlamin und 100 ms Äthyli-ocvanat versetzt und während 2 Std. unter Feuchtigkeitsausschluss bei 50' gehalten. Bei Beginn der Reaktion wird 2elcgent- lich geschüttelt bis das ungelöste Material in Lösung gegangen ist.
Nach beendigter Reaktion wird die Lösung bei 30\ im Hochvakuum am Rotationsver dampfer zur Trockene eingeengt. Der Rückstand wird mit 0,5-n. wässeriger Kaliumliydroaenearbonatlesr-.nR versetzt und mit Essigester mehrmals ausgschüttelt. \.lach dem Ans'iuren der extrahierter, wä;
seri_7en Phaw mit Salzsäure auf pH 3 extrahiert man erschöpfend mit Essigester, wäscht die vereinigten Essigesterextrakte mit Wasser und gesättigter Kochsalzlösung, trocknet mit Natriumsulfat und dampft zur Trockene ein. Der fast farblose Rückstand enthält O-Desacetyl-O-ätliyl- carbamoyl-7-(N'-äthylureido)-cephalosporansäur(#. Diese weist im in Nujol aufgenommenen IR-Spektrum typische Banden bei 5,6; 5,9 und 6,1 u auf.
Das Mate rial zeigt in vitro hohe antibakterielle Wirksamkeit ge genüber gramnegativen Bakterien. Minimale Hemm konzentration in @igiml: Escherichia coli 30 Salmonella typhosa 15 Klebsiella pneumoniae 15 Die als Ausgangsmaterial verwendete 0-Desacetyl- 7-amirio-ceplialosporansäure kann wie folgt hergestellt werden:
<I>273 mg</I> (1 mLIol) 7-Amino-ceplialosporansäure werden in @9 ml dest. Wasser aufgeschlämmt und unter energischem Rühren so lange tropfenweise mit 0,2-n. Kalilauge versetzt, bis ein pH von 6,7 erreicht ist, wobei alles Material in Lösung Weht. Hierauf gibt man 4 ml wäss. Lösung von Acetylesterase, welche mit Natriumoxalat stabilisiert ist, zu. Die frei werdende Essigsäure wird mit Hilfe eines automatischen Titrators mit 0.2-n.
Kalilauge neutralisiert (Einstellung auf pH 6,7). Nach 9 Std. sind 18 0;o d. Theorie an Kali lauge verbraucht. Man gibt jetzt weitere 2 ml Acetyl- esteraselösung zu; es werden in den folgenden 14 Std. aber lediglich weitere 4 0 o an Kalilauge verbraucht. Die gelbe, leicht trübe Reaktionslösung wird durch Celite filtriert. Die Desacetvl-7-amino-cephalosporansäure wird durch Ansäuern des klaren Filtrates mit 2-n.
Es sigsäure auf pI-I 4.5 ausgefällt. Ausbeute 161 mg IR.- Spektrum in Nujol: Banden u. a. bei 2,96 c(, 3,15 . 5,56y. Eine Esterbande ist eindeutig nicht mehr vor handen.
Das Produkt zeigt im Dünnschichtchromato- gramm auf Silicagel imv System n-Butanol-Pyridin- Essigsäure-Wasser (30:20:6:24) einen Rf-W ert von 0,33 (7-Amino-cephalosporansäure im gleichen System:
Rf 0,-10). Fleischfarbene Flecke beim Besprii- hen mit Ninhydrin-Collidin; farblose Flecke auf violet tem Untergrund nach Besprühen mit Jodstärke-Essig- säure-Reagens (Ausführung nach R. Thomas, Nature 191, 1161, (1961<B>1</B>).
In der Elektrophorese zeigt Desacetyl-7-amino- cephalosporansäure (1) im Vergleich zu 7-Amino- cephalosporansäure (1I) folgendes Verhalten (je 90 Min. bei 46 V/cm auf Whatman Nr. 1-Papier elek- trophoresiert; die Zahlen bedeuten die Anzahl cm Wanderungen gegen die Anode):

Von 10 kg niögliclist dicksclialisen- Oranoen wird das Flavedo (äusserste orange Rindenschicht) mit @inec Bircher-Raffel abgeraspclt. Die so erhaltenen 1600 Flavedo werden in 8 Portionen ä 200 g mit je 200 ml eiskalter 3-proz. Natriunichioridlösung homogenisiert. Der entstehende Brei jeder Portion wird sofort mit 20 g Celite vermischt und durch einen
Nylonfilter ab- genutscht. Zur Stabilisierung der Esterase siittigt man die eisgekühlten vereinigten Filtrate mit 52<B><U>g</U></B> Natrium oxalat,
filtriert sie durch eine dünne Schicht Hyflo- Supercel und versetzt das Filtrat mit 7-l2 <B><U>2</U></B> Ammoni- umsulfat (70 % der zur Sättigung benötigten Menge). Man lässt über Nacht bei 0<B>'</B> stehen und saugt den ge bildeten Niederschlag ab.
Die das Enzym enthaltende Fällung wird in 200 ml 0,1-m. Natriuinoxalatlösung aufgenommen und und 10 Tage bei 00 gegen 0,1-m. Natriumoxalat dialysiert, wobei die Aussenlö sung jeden Tag erneuert wird. Die Enzymlösung wird schliesslich filtriert und bei 0 aufbewahrt.
<I>Beispiel 2</I> 300 mg (0,65 mN4ol) Triäthylammoniumsalz von roher Desacet5-l-7-(clilor-tert.-butylureido)-cephalospo- ransäure (ca. 60-proz.) werden in 3 ml frisch entgastem Dimethylformamid, 2,4m1 einer 10-proz. Lösung von absolutem Tributvlamin (1,3 mMol)
in Dimethylform- amid und 0.95 ml einer 10-proz. Lösung von Athyliso- cyanat (1.3 mMol) in Dimethylformamid aufgenommen und 16 Std. bei 20' rea,-,ieren gelassen. Während der Tanzen Reaktionszeit lässt man Ultraschall der Fre quenz -15 kHz einwirken.
Anschliessend verdampft man im N'a@Utim zur Trockne, löst in einem Gemisch Essigester-0,1-m. Phosphatpuffer pH 7 (1:1) und säu ert mit konz. Phosphorsäure auf pH 2,0 an. Nach '!_ Std. Rühren bei 20' wird das Gemisch mit 50-proz. wässriger Trikaliumphosphatlösung auf pH 8,0 ge stellt und mit Essigester gewaschen. Die wässrige Phase stellt man mit konz. Salzsäure auf pH 2,0 und extrahiert das Produkt mit Essigester.
Der über Natri- uinstiffat getrocknete Extrakt gibt beim Eindampfen im Vakuum 99 mg Desacetyl-0-äthylcarbomyl-7-(chlor- tert.-butylureido)-cephalosporansäure. Minimale Hemmkonzentraionen in vitro und Rf-Werte im Dünn- schichtchromatoL,ramm, vgl. Tabelle.
Die als Aus gangsmaterial verwendete 0-Desacetyl-7-(cWor-tert.- butylureido)-cephalosporansätire kann wie folgt herge stellt werden: 544 mg (2 mbtol) 7-Amino-cephalosporansäure werden in 10 ml Metliylenchlorid und 0,56 ml Triäthyl- amin mit 279 mg (2,1 mMol) Chlor-tert.-butyl-isocya- nat 3 Std. unter "elindem Rückfluss gekocht.
Nach dem Eindampfen wird der Rückstand in<B>50</B> ml Chloro form und 50 ml Essigester aufgenommen und mit 3 ml 2-n. Salzsäure und 20 ml Wasser ausgeschüttelt. Die Chloroform-Essigesterlösung wird hierauf nochmals mit 10 ml Wasser ausLeschüttelt, über Natriumsulfat getrocknet und schonend eingedampft. Der Rückstand enthält die 7-(Chlor-tert.-butvlureido)-cephalosporan- sätire, die mittels Natriumätlivlhexanoat in Aceton in das Natriumsalz überueführt wird.
4,3 mg (0,01 niMol) Natriumsalz der 7-(Chlor-tert.- butylureido)-ceplialosporansäure werden in 0,8 ml Was ser gelöst und das pH mit 0.02-n. Natronlauge auf 6,7 gestellt. Man gibt 0,2 ml Acetylesteraselösuna zu und neutralisiert anschliessend die bei der eintretenden Des- acetvlierung frei werdende Essigsäure mittels eines auto matischen Titrators mit 0,02-n.
Natronlauge auf einen pH-Wert von 6, 7. Nach 135 Nlin. sind 80<B>','0</B> d. -h. an Natronlauge vebraucht. Die entstandene Desacetyl-7- (chlor-tert. butyl-ureido)-cephalosporansäure zeigt fol ;ende Rf-Werte im Diinnschichtchromato2ranim an Sili- ca#-,cl (entwickelt mit Jodst;irkc-Essi;siiure-Rea2ens): System 1: 0,42; System<B>11:</B> 0,34: System 111: 0,56.
<I>Beispiel 3</I> Aus 300 mg (0.65 mN-Iol) rohem Triäthylammoni- unisalz der 1)esacetv1-7-(clilor-tert.-btitvlureido)-cepha- losporansäure erhält man durch Umsetzen mit 1,3 mMol Methyhsocyanat analog Beispiel 2 89 g Des acetyl-0-methylcarbamoyl-7-(chlor-tert.-butylureido)-ce- phalosporansäure. Charakterisierung siehe Tabelle.
<I>Beispiel 4</I> 300 mg (0,69 mMol) Triäthylammoniumsalz aus ca. 60 0,"oigem Desacetyl-7-(3-chloräthylureido)-cepha- losporansäure werden entsprechend Beispiel ? in 3 ml Dimethylformanüd und 2,55 ml 1011:
oigem Tributyl- amin in Dimethylformamid (1,37 mMol) mit 0,97 nil 10 !oigem Ätliylisocyanat in Dimethvlformamid (1,37 mMol) während 16 Std. zur Reaktion eebracht und aufgearbeitet. Man erhält 81 mg Desacetyl 0-äthylcarbamoyl-7(ü-chloräthylureido)-cephalosporan- säure. Eigenschaften siehe Tabelle.
Die als Ausgangsmaterial verwendete 0-Desacetyl- 7-(#i-chloräthylureido)-cephalosporansäure kann wie folgt hergestellt werden: 544 mg (2 mMol) 7-Amino-cephalosporansäure werden in 10 ml NIethylenchlorid und 0,56 ml Tri- äthylamin mit 222 mg (2.1 mNiol) ";-Chloräthyl-isocva- nat 8 Std. unter gelindem Rückfluss gekocht.
Nach dem Eindampfen wird der Rückstand in 50 ml Chloro form und 50 ml Essigester aufgenommen und mit 3 ml 2-n. Salzsäure und 20 ml Wasser aus,-,eschüttelt. Die Chloroform-Essigesterlösuno wird hierauf nochmals mit 10 ml Wasser ausgeschüttelt. über Natriumsulfat getrocknet und schonend eingedampft. Aus dem Rück stand erhält man nach Umkristallisieren aus Aceton- Essigsäure-Petroläther die 7-[N'-#3-Chloräthyl)-ureido]- cephalosporansäure als nahezu farblose Kristalle vom F. 165 (Zers.).
Das mittels Natriumätliylhexanoat in Aceton herge stellte Natriumsalz verfärbt sich bei 170= und zersetzt sich bei Temperaturen über 200-.
4,0 mg (0,01 mMol) Natriumsaltz der 7- [N'-(3-chloräthvl)-ureido]-cephalosporansäure werden in 0,8 ml destilliertem Wasser gelöst und das pH mit 0,02-n. Natronlauge auf 6.7 gestellt. Man gibt 0,2 ml Acetylesteraselösung zu und neutralisiert anschliessend die bei der eintretenden Desacetylierung frei werdende Essigsäure mittels eines automatischen Titrators mit 0,02-n. Natronlauge auf einen pH-Wert von 6,7. Nach 135 Miii. sind 80 0 0 d. Th. an Natronlauge verbraucht.

in which A and A are straight or branched alkyl radicals optionally substituted by lower alkoxy groups or halogen atoms and have a maximum of 6 carbon atoms, with the proviso that at most one of the radicals A and A is substituted by halogen atoms, and their salts.
Particularly suitable alkyl radicals are methyl, ethyl, propyl and iso-propyl.
A and A_ can be the same or different.
The halogen atoms are bromine, fluorine, iodine and, above all, chlorine, and the lower alkoxy groups are primarily N, ethoxy or ethoxy groups. The substituents are preferably in the 3-position to the carbamyl group:
A, or A_ is, for example, a 2-halogenoethyl, 2-halogenopropyl or 1-methyl-2-halogenopropyl group. Several substituents can also be present in A 1 and A_ or only one of A 1 and A_ can be substituted.
