Verfahren zur Herstellung neuer Zuckerderivate
Gegenstand der Erfindung ist die Herstellung von 2 -O-(, B-Carboxy-propionyl)-glucofuranosiden der Formel

worin Rl einen niederen Alkylrest und R3, R5 und R6 gegebenenfalls substituierte Benzylreste bedeuten.
Niedere Alkylreste sind vor allem solche mit höch- stens 5 Kohlenstoffatomen, wie Propyl-, Isopropyl-, sek. -Butyl-oder besonders Athylreste. Die aromatischen Rin ge der Benzylreste können Substituenten tragen, z. B. insbesondere niedere Alkoxy-, wie Methoxy-, Äthoxy-oder Propoxy-oder Methylendioxygruppen, Halogenatome, wie Chlor oder Brom, Alkylreste, wie Methyl-, Athyl-, Propyl-oder Butyl-und/oder Trifluormethylreste,
Die neuen Verbindungen besitzen wertvolle pharma kologische Eigenschaften. So weisen sie insbesondere eine antiinflammatorische Wirkung auf, wie sich im Tierversuch, z. B. an der Ratte, zeigt. Ferner besitzen sie, z.
B. im Tierversuch, wie z. B. an Meerschweinchen, eine antiallergische Wirkung. Die neuen Verbindungen können daher als Antiphlogistika Verwendung finden. Die neuen Verbindungen sind aber auch wertvolle Zwischenpro- dukte für die Herstellung anderer nützlicher Stoffe, insbesondere von pharmakologisch wirksamen Verbindungen.
Besonders hervorzuheben sind die Niederalkyl-2-0- (ss- -carboxy-propiionyl)-3, 5, 6-tri-O-benzyl- D-glucofuranosi- de und insbesondere das Athyl-2-0- (f3-carboxy-propio- nyl)-3, 5, 6-tri-O-benzyl-D-glucofuranosid, das beispielsweise in Form seines Natriumsalzes an der Ratte bei intraperitonealer Gabe in Dosen von 100 bis 300 mg/ kg eine ausgesprochen antiinflammatorische Wirkung aufweist.
Die Salze, besonders die Alkalimetallsalze der neuen Verbindungen zeichnen sich, im Gegensatz zu ähnlichen 2-0-unsubstituierten Verbindungen, durch eine gute Was serlöslichkeit aus.
Die neuen Verbindungen können in freier Form oder in Form ihrer Salze, wie Metallsalze oder Salze von organischen Basen insbesondere therapeutisch verwendbarer Salze, vorzugsweise der Alkalimetallsalze, besonders der Natriumsalze vorliegen. Freie Säuren und ihre Salze lassen sich in üblicher Weise ineinander überführen. Die Salze können auch zur Reinigung der freien Säuren dienen. Infolge der engen Beziehungen zwischen den neuen Verbindungen in freier Form und in Form ihrer Salze sind im Vorausgegangenen und nachfolgend unter den freien Verbindungen sinn-und zweckmässig, gegebenenfalls auch die entsprechenden Salze zu verstehen.
Die neuen Verbindungen können als reine Anomere oder als Anomerengemische vorliegen. Letztere können aufgrund der physikalisch-chemischen Unterschiede der Bestandteile in bekannter Weise in die beiden reinen Anomeren aufgetrennt werden, beispielsweise durch Chromatographie und/oder fraktionierte Kristallisation.
Vorzugsweise isoliert man den wirksameren der beiden Anomeren.
Die neuen Verbindungen werden erhalten, wenn man Glucofuranoside der Formel

mit den 3-Carboxy-propionylrest einführenden, acylierenden Mitteln umsetzt.
Als acylierende Mitte] kommen dabei insbesondere Bernsteinsäureanhydrid oder auch reaktionsfähige funktionelle Monosäurederivate der Bernsteinsäure, wie die Halogenide, besonders das Bernsteinsäuremonochlorid, in Frage. Vorzugsweise geht man so vor, dass man die genannten Glucofuranoside in Gegenwart von sauren oder basischen Katalysatoren, z. B. in Gegenwart von Pyridin, mit Bernsteinsäureanhydrid umsetzt.
Man kann aber auch die Glucofuranoside mit Bernsteinsäuremonohalogeniden, z. B. mit Bernsteinsäuremo- nochlorid in Gegenwart von säurebindenden Kondensationsmitteln, wie tertiären Basen, Natriumacetat und dergleichen, umsetzen.
Die Erfindung betrifft auch diejenigen Ausführungs- formen des Verfahrens, bei welchem man einen Aus gangsstoff in Form eines unter den Reaktionsbedingun- en erhältlichen rohen Reaktionsgemisches einsetzt.
Die Ausgangsstoffe sind bekannt oder können nach an sich bekannten Methoden gewonnen werden.
Die neuen Verbindungen oder ihre Salze können als Heilmittel, z. B. in Form pharmazeutischer Präparate, Verwendung finden.
Die Erfindung wird in den nachfolgenden Beispielen näher beschrieben. Die Temperaturen sind in Celsiusgraden angegeben.
Beispiel I
Zu einer Lösung von 28, 8 g Äthyl-3, 5, 6-tri-0-benzyl-D-gluco-furanosid in 75 mi absolutem Pyridin gibt man 6, 6 g fein pulverisiertes Bernsteinsäureanhydrid hinzu und erhitzt unter Rühren und Feuchtigkeitsausschluss bei 70 während 24 Stunden. Der durch Eindampfen im Vakuum bei 55 erhaltene Rückstand wird mit 30 ml Eiswasser versetzt und während 45 Minuten geschüttelt. Man extrahiert dann mit Ather, wäscht die Ätherschicht fünfmal mit 50 ml eisgekühlter 5n Salzsäure und 50 ml Eiswasser, trocknet sie über Natriumsulfat. engt auf 70 ml ein und schüttelt mit 50 ml gesättigter. wässeriger Natriumbicarbonatlösung und 50 ml Wasser.
Nachdem man am Rotationsverdampfer den Äther im Vakuum entfernt hat, verdünnt man mit 700 ml Wasser und filtriert durch Diatomeerde. Die klare Lösung wird mit eiskalter 2n Salzsäure auf pH 2, 5 gestellt, worauf das ausgeschiedene öl mit Ather extrahiert wird. Die über Natriumsulfat getrockneten und am Wasserstrahl und anschliessend am Hochvakuum eingedampften Atherextrakte liefern das Äthyl-2-0- (ss-carboxy-propionyl)- -3. 5. 6-tri-O-benzyl-D-glucofuranosid der Formel dampfen des Äthers unter vermindertem Druck wird die wässrige Lösung filtriert und lyophilisiert, wobei das Natriumsalz als dickflüssiges Öl anfällt.
Im Dünnschichtchromatogramm auf Silicagel mit dem System n-Butanol/Essigsäure/Wasser (150 Vol. Teile ; 15 Vol. Teile ; 42 Vol. Teile), angefärbt mit 50% iger ätha- nolischer Schwefelsäure, zeigt die Substanz den Rf-Wert von 0, 68.
Beim kurzen Stehenlassen einer Lösung der freien Verbindung in einem Überschuss l-n. wässrig-äthanolischem Natriumhydroxyd wird der Ester quantitativ verseift. Das erhaltene Äthyl-3, 5, 6-tri-O-benzyl-D-glucofuranosid wird im Dünnschichtchromatogramm, System Chloroform-Essigsäureäthylester (85 : 15), Silicagel (PF 254 der Firma Merck, Darmstadt), Indikator : Schwefelsäure, aufgetrennt ; das x-Anomere zeigt einen Rf-Wert von 0, 56; [α]20D = +21 (c = 1 in Chloroform) ; und das 3-Anomere einen Rf-Wert von 0, 27, fa] 20D =-560 (c-1 in Chloroform).

Eine Lösung von 10, 12 g Isobutyl-3, 5, 6-tri-O-ben- zyl-D-glucofuranosid in 40 ml absolutem Pyridin wird mit 2, 8 g fein pulverisiertem Bernsteinsäureanhydrid versetzt, und unter Rühren und Feuchtigkeitsausschluss bei 70 C während 38 Stunden erhitzt. Anschliessend destilliert man die Hauptmenge Pyridin unter vermindertem Druck bei 60 ab und schüttelt den Rückstand mit 40 ml Eiswasser während 10 Minuten.
Nach dem Abdekantie- ren des Wassers nimmt man das 51 in 150 ml Ather auf. und extrahiert die ätherische Lösung fünfmal mit 50 ml 2-n. eisgekühlter Salzsäure und fünfmal mit 50 ml Wasser. Die über Natriumsulfat getrocknete und unter vermindertem Druck eingedampfte Ätherlösung liefert das Isobutyl-2-O- (, 8-carboxy-propionyl)-3, 5, 6-tri-O-ben zyl-D-glucofuranosid der Formel

Zur Herstellung des Natriumsalzes wird die Säure in Äther gelöst, mit der berechneten Menge wässriger Natriumhydrogencarbonatlösung geschüttelt, der Äther unter vermindertem Druck abgedampft und anschliessend lyophilisiert, wobei das Natriumsalz als dickflüssiges Öl anfällt. Aus dem Natriumsalz lässt sich eine klare 3% ige wässrige Lösung herstellen.
Beim kurzen Stehenlassen einer Lösung der freien Verbindung in einem Überschuss ln. wässrig-äthanoli- schem Natriumhydroxyd wird der Ester quantitativ verseift. Das erhaltene Isobutyl-3, 5, 6-tri-O-benzyl-D-glucofuranosid zeigt im Dünnschichtchromatogramm auf Silicagel im System Chloroform-Essigsäureäthylester (85 : 15) einen Rf-Wert von 0, 64 für das a-Anomere und einen Rf-Wert von 0, 30 für das p-Anomere.
Das Ausgangsmaterial kann wie folgt hergestellt werden :
Eine Lösung von 250 g 1, 2-O-Isopropyliden-3, 5, 6 -tri-O-benzy]-a-D-glucofuranose in 5000 ml einer 1-n.
Lösung trockener Salzsäure in Isobutanol wird während 24 Stunden bei Zimmertemperatur stehen gelassen. Nach dem Abkühlen auf 0-5 neutralisiert man mit 10-n. wässeriger Natronlauge, destilliert die Hauptmenge Isobutanol unter vermindertem Druck im Rotationsverdampfer bei 50-60 ab und extrahiert den Rückstand mit Chloroform. Die mit Wasser gewaschene, über Natriumsulfat getrocknete Chloroformlösung wird unter vermindertem Druck eingedampft und im Hochvakuum vom restlichen Lösungsmittel befreit.
Der so erhaltene Rückstand wird einer Kurzweg-Destillation im Hochvakuum unterworfen, wobei man das Isobutyl-3, 5, 6-tri-O-benzyl-D-glucofuran- osid als dickflüssiges 61 bei 240-245 X 0, 01 mm Hg erhält : M"n = -34 ¯ 1 (c = 1 in Chloroform).

Beim Umsetzen von 3, 8 g Athyl-3, 5, 6-tri-O-benzyl- -x-D-glucofuranosid in 25 ml Pyridin mit 0, 87 g Bernsteinsäureanhydrid erhält man nach 40stündigem Erhit- zen auf 70 und nach der im Beispiel 1 beschriebenen Aufarbeitungsmethode das Äthyl-2-0- (ss-carboxy-propio- nyl)-3, 5, 6-tri-O-benzyl-α-D-glucofuranosid der Formel

Beispiel 5
Eine Lösung von 10 g n-Propyl-3, 5, 6-tri-O-benzyl-D -glucofuranosid in 40 ml absolutem Pyridin wird mit 2, 24 g fein gepulvertem Bernsteinsäureanhydrid versetzt und unter Rühren während 28 Stunden erhitzt. Anschliessend wird die Hauptmenge Pyridin unter vermindertem Druck bei 55 abdestilliert und der Rückstand während 45 Minuten mit 40 ml Eiswasser geschüttelt.
Nach dem Abdekantieren des Wassers, nimmt man das 61 in Ather auf und schüttelt fünfmal mit eiskalter 2-n.
Salzsäure und Wasser aus. Die Ätherlösung wird über Natriumsulfat getrocknet, filtriert und unter vermindertem Druck bei 40 eingedampft. Der Rückstand wird noch zweimal mit je 50 ml Petroläther ausgerührt, das Lösungsmittel abdekantiert und das 61 im Hochvakuum entgast. Man erhält auf diese Weise das n-Propyl-2-O -(, B-carboxy-propionyl)-3, 5, 6-tri-O-benzyl-D-glucofuranosid der Formel

Beispiel 6
Eine Lösung von 10 g n-Butyl-3, 5, 6-tri-O-benzyl-D- -glucofuranosid in 40 ml absolutem Pyridin wird mit 2, 2 g fein gepulvertem Bernsteinsäureanhydrid versetzt und unter Rühren und Feuchtigkeitsausschluss bei 70 während 75 Stunden erhitzt. Anschliessend destilliert man die Hauptmenge Pyridin bei 50 unter vermindertem Druck ab, schüttelt den erhaltenen Rückstand während 10 Minuten mit 45 ml Eiswasser dekantiert die wässerige Schicht ab und nimmt das öl in 150 ml Ather auf. Die ätherische Lösung wird fünfmal mit 50 ml eisgekühlter 5-n. Salzsäure und 50 ml Eiswasser gewaschen, über Natriumsulfat getrocknet und unter vermindertem Druck engedampft.
Der Rückstand ist das n-Butyl-2-O-(ss-carboxypropionyl)-3,5,6-tri-O-benzyl-D-glucofuraosid der Formel

Zur Herstellung des Natriumsalzes des n-Butyl-2-O- (-carboxy-propionyl)-3, 5, 6-tri-O-benzyl-D-glucofuran- osids wird die Säure in Äther gelöst und mit der berechneten Menge wässeriger Natriumhydrogencarbonatlösung geschüttelt. Nach Abdampfen des Äthers wird die wässrige Lösung lyophylisiert, wobei das Natriumsalz als dickflüssiges öl anfällt. Aus dem Lyophylisat lässt sich eine klare 3, 2%ige wässrige Lösung herstellen.
Das im obigen Beispiel verwendete Ausgangsmaterial kann folgendermassen hergestellt werden :
Eine Lösung von 79 g 1, 2-O-Isopropyliden-3, 5, 6-tri -O-benzyl-x-D-glucofuranose in 1800 ml einer 1-n. Lö- sung von Salzsäure in n-Butanol wird während 22 Stunden bei Zimmertemperatur stehen gelassen. Nach dem Abkühlen auf 0-5 wird mit 10-n. wässriger Natronlauge neutralisiert. Nach dem Abdestillieren der Hauptmenge n-Butanol unter vermindertem Druck wird mit Chloroform extrahiert ; die Chloroformlösung wird mit wässri ger Natriumbisulfitlösung und Wasser gewaschen, über Natriumsulfat getrocknet und unter vermindertem Druck eingedampft.
Der im Hochvakuum bei 40 vom restlichen Lösungsmittel befreite Rückstand stellt das n-Butyl -3. 5. 6-tri-O-benzyl-D-glucofuranosid dar, welches sich im Hochvakuum in einer Kurzweg Destillationsappara- tur bei 240-250 /0, 01 mm Hg destillieren lässt, [] 20D = -36 + 1 (c = 1 in Chloroform).

After most of the n-butanol has been distilled off under reduced pressure, the mixture is extracted with chloroform; the chloroform solution is washed with aqueous sodium bisulfite solution and water, dried over sodium sulfate and evaporated under reduced pressure.
The residue freed from the remaining solvent in a high vacuum at 40 is n-butyl -3. 5. 6-tri-O-benzyl-D-glucofuranoside, which can be distilled in a high vacuum in a short-path distillation apparatus at 240-250/0.01 mm Hg, [] 20D = -36 + 1 (c = 1 in chloroform).
