Verfahren zur Herstellung von 1, 4-Endomethylen 2-cyclohexylmethylamiden
Vorliegende Erfindung bezieht sich auf ein Verfahren zur Herstellung einer von 1, 4-Endomethylen- 2-aminomethylcyclohexan abgeleiteten Gruppe von Amiden.
Die Struktur des 1 ,4-Endomethylen-2-aminome- thylcyclohexans ist folgende:

Nach dem neuen Verfahren werden 1, 4-Endome- thylen-2-cyclohexylmethylamide der Formel

und ihre 5,6-Dehdroanaloge hergestellt, worin n = 0, 1, 2, 3, 4 oder 5 ist, Rt Alkyl, Alkenyl, Cycloalkyl, Aralkyl oder gegebenenfalls durch OH-, Halogen, Alkoxy-. Thioalkyl-, Amino-oder heterocyclische Gruppen teilweise substituiertes Aryl bedeutet,
R2 ein Acylrest ist, und
R3 Wasserstoff, Hydroxyl, Acyloxygruppen, Oxyalkyl, Isopropyl oder Dialkylamino oder eine Morpholino-, Pyrrolidino-, Piperidino- oder Piperazinogruppe bedeutet, wobei die heterocyclischen Gruppen unsubstituiert oder durch Methyl, Äthyl, Isopropyl oder OH substituiert sind.
Das erfindungsgemässe Verfahren ist dadurch gekennzeichnet, dass man ein Acylhalogenid der Formel R2-Hal mit dem entsprechenden Amin umsetzt.
Diese neuen Amide sind bisher nicht beschrieben worden. Sie liegen im allgemeinen in Form von weissen, kristallartigen Substanzen oder leicht gelblichen, zähflüssigen Ölen vor, die nur schwer destillierbar sind. Es wurde gefunden, dass sie neben einer äusserst niedrigen Giftigkeit bedeutende neurotropische Wirksamkeit aufweisen können und deshalb im therapeutischen Gebiet äusserst interessante Anwendungsmöglichkeiten bieten.
Im allgemeinen wurde gefunden, dass die Amide mit Säuren niedrigen Molekulargewicht oder mit aliphatischen Säuren eine erregende und psychostimulierende Wirkung besitzen, während bei aromatischen Säureresten mittleren Molekulargewichts Verbindungen vorliegen, die ausgezeichnet sedative, spasmolytische und anästhetische Wirkungen aufweisen.
Die Verlängerung der Alkylaminkette verursacht in der Regel eine Steigerung der sedativen und anästhetischen Wirksamkeit, während die Gegenwart einer Dialkylaminoalkylgruppe diese Wirksamkeit im allgemeinen zwar erhöht, die Verträglichkeit des Produkts hingegen vermindert.
Die als Ausgangstoffe dienenden Amine können durch Einwirkung von Verbindungen R5(CH2)nX (wo X ein halogen darstellt und R3 die obige Bedeutung besitzt), oder R3(CH2)1COX (wo X ein Halogen ist und R3 die obige Bedeutung besitzt) und darauffolgende Reduktion mit Aluminium-Lithium-Hydrid, oder von Verbindungen R3(CH2)n1CHO und eine anschliessende katalytische Hydrierung hergestellt werden.
Ein Beispiel der Reaktionsfolge wird nachstehend gegeben:

Das erfindungsgemässe Verfahren verläuft nach folgender Reaktion:

Die Reaktion (a) wird in der Regel ausgeführt, indem man beide Reagentien 5-10 Stunden lang auf 1600 C erwärmt, wobei ein Überschuss an Amin im Verhältnis von 3 Mol pro Mol Chlorid als Chlorwasserstoffsäure-Akzeptor benutzt wird.
Das rohe Reaktionsprodukt kann durch Auflösung in Äther, Waschen in alkalihaltigem Wasser und Vakuumdestillation gereinigt werden, und dadurch kann man das überschüssige Amin zurückgewinnen und das reine, N-substituierte Amin mit einer Ausbeute von etwa 90 Oio erhalten.
Die Reaktion (b) wird durchgeführt, indem man vorzugsweise ohne Erhitzung 1 Mol Säurechlorid in Benzol einer Benzollösung zufügt, die in der Regel 1 Mol des n-substituierten Amins sowie 1 Mol Triäthylamin enthält; das letztere dient als Chlorwasserstoffsäure-Akzeptor.
Nach Beendigung der Beimischung wird die Reaktionsmischung vorzugsweise 3-7 Stunden lang am Rückfluss gekocht und das Reaktionsgemisch dann mit 10 0/obigem Natriumhydroxyd, 10 0/oiger Chlorwasserstoffsäure und schliesslich mit Wasser gewaschen.
Nach der Vakuumverdampfung des Lösungsmittels kann man aus der Benzollösung einen leicht gelblichen und schwer destillierbaren öligen Rückstand erhalten, der aus dem gewünschten Amid besteht.
Die Ausbeute beträgt im allgemeinen etwa 90 O/o.
Manchmal kann das Amid in kristalliner Form erhalten werden, wenn man den obigen Rückstand mit einer kleinen Menge Äthylacetat oder mit einem Gemisch von Äthylacetat/Petroläther im Verhältnis 4:1 behandelt.
Beispiel 1 a) Herstellung des Ausgangsmaterials:
11,6 g 1 ,4-Endomethylen-2-aminomethyl-3-methylcyclohexan werden 7 Stunden bei 160 C mit 2,25 g Äthylenchlorhydrin erhitzt.
Durch Alkalisierung mit ln NaOH, Extraktion mit Äthyläther und Destillierung werden 5 g 1, 4-Endome- thylen-2-oxyäthyl-aminomethyl-3 -methylcyclohexan (1) erhalten, während 4 g unreagiertes Amin wiedergewonnen wird. b) Zu 20 g der Verbindung (1) und 11,1 g Tri äthylamin, die in 200 ml wasserfreiem Benzol aufgelöst wurden, werden 19,2 g in 50ml wasserfreiem Benzol aufgelöstes Chlorbenzoylchlorid zugesetzt, während die Mischung gekühlt wird.
Nach Beendung des Zusatzes wird das Ganze unter Umrühren 4 Stunden lang gekocht und nach Waschen mit 10 0/obiger Chlorwasserstoffsäurelösung, 10 0/obiger Natriumhydroxydlösung, und nach Filtrieren durch Norit -Kohle und Verdampfung des Lösungsmittels werden 24 g eines Rückstandes erhalten, der aus dem Chlorbenzamid des 1,4 Endomethylen-2-oxyäthylaminomethyl3 -methylcyclohe- xans besteht.
Dieser Rückstand erscheint wie ein äusserst zähflüssiges leicht gelbliches Öl und nach Behandlung mit einem Gemisch von Äthylacetatl Petroläther im Verhältnis 4:1 kristallisiert er, wobei er weisse Kristalle mit einem Schmelzpunkt von 98 "C bildet. c) Zu 10 g der Verbindung (1) und 5,6 g Triäthylamin in 100 ml einer Lösung von wasserfreiem Benzol werden 7,4 g in 25 ml wasserfreiem Benzol aufgelöstes Lävulinoylchlorid zugesetzt, unter Kühlung der Mischung.
Nach Beendigung der Zufügung wird das Ganze 5 Stunden lang am Rückfluss gekocht, und nach dem Waschen mit 10 0/oiger Chlorwasserstoffsäurelösung, 10 0/einer Natriumhydroxydlösung und Filtrieren durch Norit -Kohle sowie Verdampfung des Lösungsmittels, wird ein leicht gelblich gefärbter öliger Rückstand erhalten, der aus 11,5 g des Lävulinamids des 1 ,4-Endo- methylen-2-oxyäthylaminomethyl-3-methylcyclohexans besteht.
Beispiel 2
Zu 19,6 g 1 ,4-Endomethylen-2-oxyäthylaminomethyl-3-phenylcyclohexan und 10,1 g Triäthylamin in 200 ml einer Lösung von wasserfreiem Benzol werden, unter Kühlung der Mischung 7,85 g Acethylchlorid in 50 ml wasserfreiem Benzol zugesetzt.
Nach Beendung der Zumischung wird das Ganze 3 Stunden lang am Rückfluss gekocht und nach Waschen mit einer 10 öligen Chlorwasserstoffsäurelösung, 10 0/obiger Natriumhydroxydlösung, Filtrieren durch Norit -Kohle und Verdampfung des Lösungsmittels werden 20 g eines sehr zähflüssigen, leicht gelblichen öligen Rückstandes erhalten, welcher aus dem Acetamin des 1, 4-Endomethylen-2-oxyäthylaminomethyl3-phenylcyclohexans besteht.
Beispiel 3 a) Herstellung des Ausgangsmaterials:
Zu 50 g 1 ,4-endomethylen-2-aminomethyl-3- methylcyclohexan und 36,6 g Triäthylamin in 200ml wasserfreiem Benzol werden 43,5 g Isovaleroylchlorid in 50 ml wasserfreiem Benzol langsam zugemischt, unter Kühlung der Mischung.
Nach Beendigung der Zumischung wird das Ganze 5 Stunden lang am Rückfluss gekocht, und der sich bildende Niederschlag, welcher aus Triäthylamin-chlorhydrat besteht, wird durch Filtrieren entfernt.
Aus der klaren Benzollösung wird nach Waschen mit sauren und alkalischen wässrigen Lösungen und nach Verdampfung des Lösungsmittels ein öliger Rückstand gewonnen, welcher nach Vakuumdestillation 60 g Monoisivaleramid des 1, 4-Endomethylen- 2-ami.nomethyl-3-methylcyclohexans ergibt (Verbindungen 11).
27g der Verbindung 11, in 100ml wasserfreiem Äthyläther aufgelöst, werden langsam einer Suspension von 9,2 g Aluminiumlithiumhydrid in 300 ml wasserfreiem Äthyläther zugesetzt.
Nach Beendigung der Beimengung wird das ganze 8 Stunden am Rückfluss gekocht, wonach das überschüssige Aluminiumlithiumhydrid mit Wasser zerstört wird. Die Ätherlösung wird mit 10 5/obiger Natriumhy droxydlösung gewaschen und der Äther verdampft.
Der Rückstand wird im Vakuum destilliert, und man erhält so 21 g 2-Isoamylaminomethyl-3-methyl- 1,4- Endomethylen-cyclohexan, in Form eines sehr beweglichen farblosen Öls (Verbindung III). b) Zu 10 g der Verbindung III und 4,85 g Tri äthylamin in 100 ml wasserfreiem Benzol werden 6,45 g Lävulinoylchlorid in 25 ml wasserfreiem Benzol unter Kühlung des Gemisches beigemengt.
Nach Beendigung der Beimengung wird das Ganze 5 Stunden am Rückfluss gekocht und nach Auswaschen mit 10 0/obiger Natriumhydroxydlösung, 10 0/obiger Chlorwasserstoffsäurelösung, Filtrieren durch Norit -Kohle und Verdampfung des Lösungsmittels werden 11 g eines leicht gelblichen öligen Rückstandes erhalten, der aus dem Lävulinamid des 2-Isoamylami nomethyl-3-methyl-1, ,4-endomethylen-cyclohexans bestehen.
Beispiel 4
Zu 10g 1 ,4-Endomethylen-2-methylaminomethyl3-methylcyclohexan und 6,6 g Triäthylamin in 100 ml wasserfreiem Benzol werden 8,8 g Lävulinoylchlorid in 25 ml wasserfreiem Benzol zugemengt, unter Kühlung der Mischung.
Nach Beendigung der Beimengung wird das ganze 5 Stunden lang am Rückfluss gekocht und nach Waschen mit sauren und alkalischen Bädern, Filtrieren durch Norit -Kohle und Verdampfung des Lösungsmittels werden 11 g eines öligen Rückstandes erhalten, der aus dem Lävulinamid des 1,4-Endomethylen-2- methylaminomethyl-3-methylcyclohexans besteht.
Die chemischen Daten, welche für alle in obigen Beispielen angeführten Verbindungen kennzeichnend sind, werden in der nachstehenden Tabelle 1 dargestellt: Table I

Die pharmakologische Untersuchung der angegebenen Verbindungen wurde an Mäusen, Ratten, Kaninchen,
Katzen und Hunden ausgeführt, sowohl in vivo als auch an isolierten und perfundierten Organen in vitro ausgeführt.
Es wurden durchgeführt: Untersuchungen über die motorische Aktivität (Aktographie), Analgesie (nach D'Amour und Smith), bedingte Reflexe (Pfahlklettertest), Atmungstätigkeit (Spirometrie), Bronchospasmus durch Histamin, Herzblutumlauf (Registrierung des Arteriendrucks und des Elektrokardiogramms), lokalanästhetische Wirkung, experimentelle Hyperämie, Darmdurchgang, in vitro Reaktion des Darms und der Gebärmutter auf ACH, H1, 5HT, BaCl2, morphologisches hämatologisches Blutbild, schützende Wirkung gegenüber des experimentellen Magengeschwürs (hervorgerufen durch Unterbindung des Magenausgangs und durch Immobilisierung).
Besonders sorgfältig wurden die Giftigkeit der Verbindungen sowie ihr Einfluss auf das Betragen untersucht.
Die beruhigende und analgesische Wirkung der Verbindungen (die an Mäusen bzw. an Ratten durchgeführt wurden) wurden quantitativ nach der Methode von Eddy bewertet, unter Schätzung, für jede Dosierhöhe, des Prozentsatzes der Tiere mit abnehmender Bewegungszahl (für 60 Minuten) und Zunahme der Reaktionszeiten gegenüber dem Schmerz (in 60 Minuten), die höher waren als X + 2 S D (X = Durchschnitt S D = Standardabweichung) berechnet in mit Placebo, (Suggestionsmittel) behandelten Kontrolltieren desselben Geschlechts und Gewichts.
Mit diesen Prozentsätzen wurden die Dosis/Effektkurven konstruiert und die entsprechende EDso (Wirkungsdosis 50) berechnet. Die Giftigkeit wurde aus der LD50 der untersuchten Verbindungen nach der Methode von Wilcoxon und Lichtfield abgeleitet. Die Wirkung auf die bedingten Reflexe wurde aus dem Prozentsatz der Hemmungsreaktionen und dem Vergleich der mittleren Latenz-Zeit, gegenüber den Kontrolltieren, zwischen der Hervorrufung des bedingten Reizes und dem Einsetzen der bedingten Reaktion berechnet. Die Unterschiede wurden nach dem Student'schen Test analysiert.
Die Einwirkung auf die Lokalanästhesie wurde durch Oberflächenanästhesie im Vergleich mit Prokain und Lidokain bewertet, und zwar entsprechend dem Frey'schen Test.
Mit den angegebenen Daten wurden die Intensität/ Dauer-Kurven des lokalanästhetischen Effekts aufgetragen.
Die Einwirkung auf den Darmdurchgang (nach Macht) und auf die experimentelle Hyperthermie (nach Burn) wurden jeweils an Mäusen und Kaninchen im Vergleich zu den Kontrolltieren untersucht. Die Unterschiede der beobachteten Werte gegenüber den Kontrolltieren wurden nach dem Student'schen Test untersucht.
Die Untersuchungen über die chronische Toxizität für die verschiedenen Apparate wurden an Ratten durch die Untersuchung des Wachstums, hämatochemische Analysen des Harns, Proben der Wasserdiuresen und histomorphologische Untersuchungen durchgeführt. In den Ratten wurde ausserdem die eventuelle teratogene Wirkung der untersuchten Verbindungen dadurch bewertet, dass man diese gleichzeitig mit der Paarung den Männchen und Weibchen verabreichte, sowie während der nächsten 21 Tage. Untersuchungen über das Betragen und Versuche über subakute Giftigkeit wurden 30 Tage lang in erwachsenen Hunden bei- derlei Geschlechts vorgenommen. In diesen Tieren wurde ausserdem der Verlauf der Glykämie, der Azot ämie, der Blutgerinnung, der Sedimentierung von Erythrozyten und das Elektrokardiogramm untersucht.
Versuche über das Betragen wurden ausserdem an Katzen und Mäusen durchgeführt (kämpferisches Betragen).
Die geschwürverhütende Wirkung nach Unterbindung des Magenausganges und Immobilisierung wurde in Ratten für zwei Höhen von täglichen Dosen untersucht. Sie wurde auf Grund des Prozentsatzes von magengeschwürfreien Tieren gegenüber der Inzidenz in den Kontrolltieren, aus den Messungen des pH des Magensaftes und qualitativ mit Bezug auf die Anzahl und Grösse der vorgefundenen Geschwüre bewertet.
Zusammenfassend weisen die in der Tabelle I mit einem Kreuz bezeichneten Verbindungen folgende pharmakologische Eigenschaften auf:
1. Beruhigende und analgesische Wirkung ohne Beeinträchtigung der Muskelleistung und der bedingten Verteidigungsreflexe.
2. Durch anticholinergische, antihistaminische und antiserotoninische Elemente gekennzeichnete spasmolytische Wirkung.
3. Schutzwirkung auf durch Unterbindung des Magenausganges und Immobilisierung hervorgerufene Experimentalgeschwüre.
4. Lokalanästhetische, vernarbende und antimikrobische Wirkung.
5. Antipyretische Wirkung.
Die obigen Verbindungen scheinen keine schädlichen Nebenwirkungen auf den Herzblutumlauf und den Atmungsapparat, das Blut, die Fortpflanzung und die Nachkommenschaft auszuüben. Der berechnete Toleranzbereich gegenüber der sedativen Wirkung schwankt zwischen 1:30 und 1:20, je nach dem betrachteten Termin. Die Toleranz gegenüber der chronischen Verabreichung ist sehr hoch. Das Acetamid des 1 ,4-Endomethylen-2-oxyäthyl-aminomethyl-3 -phenyl- cyclohexans erzeugt eine psychostimulierende Wirkung in den verschiedenen Versuchstieren. In Tafel 2 wird ein Teilbild der therapeutisch verwendbaren pharmakologischen Wirkungen der in Tafel 1 mit einem Kreuz bezeichneten Produkte wiedergegeben.
Auf Grund der pharmakologischen Tests ergibt sich, dass die Reihe von neuen Amiden interessante therapeutische Verwendungsmöglichkeiten im Bereich der Psychoneurosen, der gastroenterischen und kardiozirkulatorischen Pathologie aus psychosomatischer Äthiopathogenes e verspricht.

Process for the preparation of 1, 4-endomethylene 2-cyclohexylmethylamides
The present invention relates to a process for the preparation of a group of amides derived from 1,4-endomethylene-2-aminomethylcyclohexane.
The structure of 1,4-endomethylene-2-aminomethylcyclohexane is as follows:

The new process gives 1,4-endomethylene-2-cyclohexylmethylamides of the formula

and their 5,6-dehydro analogs, where n = 0, 1, 2, 3, 4 or 5, Rt is alkyl, alkenyl, cycloalkyl, aralkyl or optionally by OH-, halogen, alkoxy-. Thioalkyl, amino or heterocyclic groups denotes partially substituted aryl,
R2 is an acyl radical, and
R3 denotes hydrogen, hydroxyl, acyloxy groups, oxyalkyl, isopropyl or dialkylamino or a morpholino, pyrrolidino, piperidino or piperazino group, the heterocyclic groups being unsubstituted or substituted by methyl, ethyl, isopropyl or OH.
The process according to the invention is characterized in that an acyl halide of the formula R2-Hal is reacted with the corresponding amine.
These new amides have not yet been described. They are generally in the form of white, crystal-like substances or slightly yellowish, viscous oils which are difficult to distill. It has been found that, in addition to extremely low toxicity, they can have significant neurotropic effectiveness and therefore offer extremely interesting application possibilities in the therapeutic field.
In general, it has been found that the amides with acids of low molecular weight or with aliphatic acids have an excitatory and psychostimulating effect, while in the case of aromatic acid residues of medium molecular weight compounds are present which have excellent sedative, spasmolytic and anesthetic effects.
The lengthening of the alkylamine chain generally causes an increase in the sedative and anesthetic effectiveness, while the presence of a dialkylaminoalkyl group generally increases this effectiveness, but reduces the tolerability of the product.
The amines used as starting materials can be obtained by the action of compounds R5 (CH2) nX (where X is a halogen and R3 has the above meaning), or R3 (CH2) 1COX (where X is a halogen and R3 has the meaning above) and the following Reduction with aluminum-lithium hydride, or of compounds R3 (CH2) n1CHO and a subsequent catalytic hydrogenation can be produced.

Reaction (a) is usually carried out by heating both reagents at 1600 ° C. for 5-10 hours using an excess of amine in the ratio of 3 moles per mole of chloride as the hydrochloric acid acceptor.
The crude reaction product can be purified by dissolving in ether, washing in alkaline water and vacuum distillation, and thereby one can recover the excess amine and obtain the pure, N-substituted amine in a yield of about 90%.
Reaction (b) is carried out by adding, preferably without heating, 1 mol of acid chloride in benzene to a benzene solution which generally contains 1 mol of the n-substituted amine and 1 mol of triethylamine; the latter serves as a hydrochloric acid acceptor.
After the end of the admixture, the reaction mixture is preferably refluxed for 3-7 hours and the reaction mixture is then washed with 100% sodium hydroxide, 100% hydrochloric acid and finally with water.
After evaporation of the solvent in vacuo, a slightly yellowish oily residue which is difficult to distill and which consists of the desired amide can be obtained from the benzene solution.
The yield is generally about 90%.
Sometimes the amide can be obtained in crystalline form by treating the above residue with a small amount of ethyl acetate or with a 4: 1 mixture of ethyl acetate / petroleum ether.
Example 1 a) Production of the starting material:
11.6 g of 1,4-endomethylene-2-aminomethyl-3-methylcyclohexane are heated for 7 hours at 160 ° C. with 2.25 g of ethylene chlorohydrin.
By alkalization with 1N NaOH, extraction with ethyl ether and distillation, 5 g of 1,4-endomethyl-2-oxyethyl-aminomethyl-3-methylcyclohexane (1) are obtained, while 4 g of unreacted amine are recovered. b) To 20 g of the compound (1) and 11.1 g of triethylamine dissolved in 200 ml of anhydrous benzene, 19.2 g of chlorobenzoyl chloride dissolved in 50 ml of anhydrous benzene is added while the mixture is cooled.
After completion of the addition, the whole is boiled with stirring for 4 hours and after washing with 10 0 / above hydrochloric acid solution, 10 0 / above sodium hydroxide solution, and after filtering through Norit carbon and evaporation of the solvent, 24 g of a residue are obtained, which is from the Chlorobenzamide of 1,4 endomethylene-2-oxyäthylaminomethyl3 -methylcyclohexans consists.
This residue appears like an extremely viscous, slightly yellowish oil and after treatment with a mixture of ethyl acetate / petroleum ether in a ratio of 4: 1 it crystallizes, forming white crystals with a melting point of 98 "C. c) To 10 g of compound (1) and 5.6 g of triethylamine in 100 ml of a solution of anhydrous benzene, 7.4 g of levulinoyl chloride dissolved in 25 ml of anhydrous benzene are added while cooling the mixture.

Example 2
7.85 g of ethyl chloride in 50 ml of anhydrous benzene are added to 19.6 g of 1,4-endomethylene-2-oxyethylaminomethyl-3-phenylcyclohexane and 10.1 g of triethylamine in 200 ml of a solution of anhydrous benzene, with the mixture being cooled.
After the addition is complete, the whole is refluxed for 3 hours and after washing with an oily hydrochloric acid solution, 10% sodium hydroxide solution, filtering through Norit carbon and evaporation of the solvent, 20 g of a very viscous, slightly yellowish oily residue are obtained, which consists of the acetamine of 1, 4-endomethylene-2-oxyäthylaminomethyl3-phenylcyclohexane.
Example 3 a) Preparation of the starting material:
43.5 g of isovaleroyl chloride in 50 ml of anhydrous benzene are slowly added to 50 g of 1,4-endomethylene-2-aminomethyl-3-methylcyclohexane and 36.6 g of triethylamine in 200 ml of anhydrous benzene, while the mixture is cooled.
When the admixture is complete, the whole is refluxed for 5 hours and the precipitate which forms, which consists of triethylamine chlorohydrate, is removed by filtration.
After washing with acidic and alkaline aqueous solutions and after evaporation of the solvent, an oily residue is obtained from the clear benzene solution, which after vacuum distillation gives 60 g of monoisivaleramide of 1,4-endomethylene-2-aminomethyl-3-methylcyclohexane (compounds 11) .
27 g of compound 11, dissolved in 100 ml of anhydrous ethyl ether, are slowly added to a suspension of 9.2 g of aluminum lithium hydride in 300 ml of anhydrous ethyl ether.
When the addition is complete, the whole is refluxed for 8 hours, after which the excess aluminum lithium hydride is destroyed with water. The ether solution is washed with 10 5 / above sodium hydroxide solution and the ether is evaporated.
The residue is distilled in vacuo, and 21 g of 2-isoamylaminomethyl-3-methyl-1,4-endomethylene-cyclohexane are obtained in this way in the form of a very mobile, colorless oil (compound III). b) 6.45 g of levulinoyl chloride in 25 ml of anhydrous benzene are added to 10 g of compound III and 4.85 g of triethylamine in 100 ml of anhydrous benzene while cooling the mixture.
After the addition is complete, the whole is refluxed for 5 hours and after washing with 10 0 / above sodium hydroxide solution, 10 0 / above hydrochloric acid solution, filtering through Norit carbon and evaporation of the solvent, 11 g of a slightly yellowish oily residue are obtained, which is obtained from the Levulinamide of 2-Isoamylami nomethyl-3-methyl-1,, 4-endomethylene-cyclohexane exist.
Example 4
8.8 g of levulinoyl chloride in 25 ml of anhydrous benzene are added to 10 g of 1,4-endomethylene-2-methylaminomethyl3-methylcyclohexane and 6.6 g of triethylamine in 100 ml of anhydrous benzene, with the mixture being cooled.
After the addition is complete, it is refluxed for 5 hours and after washing with acidic and alkaline baths, filtering through Norit carbon and evaporation of the solvent, 11 g of an oily residue are obtained, which is derived from the levulinamide of 1,4-endomethylene 2-methylaminomethyl-3-methylcyclohexane.

The pharmacological investigation of the compounds mentioned was carried out on mice, rats, rabbits,
Cats and dogs carried out both in vivo and carried out on isolated and perfused organs in vitro.
The following were carried out: examinations of motor activity (actography), analgesia (according to D'Amour and Smith), conditioned reflexes (pole climbing test), breathing activity (spirometry), bronchospasm due to histamine, cardiac circulation (recording of arterial pressure and the electrocardiogram), local anesthetic effects , experimental hyperemia, intestinal passage, in vitro reaction of the intestine and the uterus to ACH, H1, 5HT, BaCl2, morphological hematological blood count, protective effect against the experimental gastric ulcer (caused by tying off the gastric outlet and by immobilization).
The toxicity of the compounds and their influence on behavior were examined with particular care.
The calming and analgesic effects of the compounds (which were carried out on mice and on rats, respectively) were assessed quantitatively according to Eddy's method, with an estimate of the percentage of animals with decreasing number of movements (for 60 minutes) and increasing reaction times for each dose level versus pain (in 60 minutes) that was greater than X + 2 SD (X = mean SD = standard deviation) calculated in placebo, (placebo) treated control animals of the same sex and weight.
The dose / effect curves were constructed with these percentages and the corresponding ED 50 (effective dose 50) was calculated. The toxicity was derived from the LD50 of the compounds examined using the Wilcoxon and Lichtfield method. The effect on the conditioned reflexes was calculated from the percentage of the inhibitory responses and the comparison of the mean latency time, compared with the control animals, between the elicitation of the conditioned stimulus and the onset of the conditioned response. The differences were analyzed according to Student's test.
The effect on local anesthesia was assessed by surface anesthesia in comparison with procaine and lidocaine according to Frey's test.
The intensity / duration curves of the local anesthetic effect were plotted with the data given.
The effects on the intestinal passage (according to Macht) and on the experimental hyperthermia (according to Burn) were examined in each case on mice and rabbits in comparison with the control animals. The differences between the observed values and the control animals were investigated using Student's test.
The studies on the chronic toxicity for the various apparatuses were carried out on rats by studying the growth, hematochemical analyzes of the urine, samples of the water diuresis and histomorphological studies. In the rats, the possible teratogenic effect of the investigated compounds was also assessed by the fact that they were administered to the males and females at the same time as the mating and during the next 21 days. Behavioral studies and subacute toxicity tests were carried out in adult dogs of both sexes for 30 days. The course of glycaemia, azotemia, blood coagulation, sedimentation of erythrocytes and the electrocardiogram were also examined in these animals.
Experiments on behavior were also carried out on cats and mice (combative behavior).
The anti-ulcer effect after ligation of the gastric outlet and immobilization was examined in rats for two levels of daily doses. It was assessed on the basis of the percentage of gastric ulcer-free animals versus the incidence in the control animals, from measurements of the pH of the gastric juice and qualitatively with reference to the number and size of the ulcers found.
In summary, the compounds marked with a cross in Table I have the following pharmacological properties:
1. Soothing and analgesic effect without impairing muscle performance and the related defensive reflexes.
2. Spasmolytic effect characterized by anticholinergic, antihistaminic and antiserotoninic elements.
3. Protective effect on experimental ulcers caused by ligation of the gastric outlet and immobilization.
4. Local anesthetic, scarring and antimicrobial effect.
5. Antipyretic effect.
The above compounds do not appear to have any deleterious side effects on the cardiovascular and respiratory system, blood, reproduction and progeny. The calculated tolerance range for the sedative effect varies between 1:30 and 1:20, depending on the date considered. The tolerance to chronic administration is very high. The acetamide of 1,4-endomethylene-2-oxyethyl-aminomethyl-3-phenyl-cyclohexane produces a psychostimulating effect in the various test animals. Table 2 shows a partial picture of the therapeutically usable pharmacological effects of the products marked with a cross in Table 1.
The pharmacological tests show that the range of new amides promises interesting therapeutic uses in the field of psychoneuroses, gastroenteric and cardio-circulatory pathology from psychosomatic ethiopathogenes.
