<tb> Progesterone <SEP> (+ TP) <SEP> 87.6 <SEP> 94.0 <SEP> 39.5 The above-mentioned method according to the invention can, for example, be carried out in such a way that a starting compound <B> 11 </ B> condensed with a compound of the formula III in the presence of a basic catalyst.
As a catalyst, for example, an inorganic base such as an alkali metal hydroxide, such. Sodium hydroxide, potassium hydroxide, or the like, and alkaline earth metal hydroxide, e.g. Calcium hydroxide, an alkali metal carbonate e.g. Potassium carbonate, an alkali metal alkoxide, e.g. Sodium methoxide, potassium t-butoxide, sodium isopropoxide or the like, an alkali metal amide or hydride, e.g. Sodium amide, sodium hydride; an organic base such as an amine, amine salt or ammonium compound, e.g.
B. triethylamine, dimethylaniline, pyridine, pyridine hydrochloride, picoline, collidine, trimethylbenzylammonium hydroxide or the like and triphenylmethyl sodium or the like. The reaction with III, e.g.
Acrylonitrile or the like., In an inert solvent such as methanol, ethanol, isopropanol, t-butanol, benzene, ether, tetrahydrofuran, dioxane, or the like., Or mixtures thereof ben, or in some cases without a solvent, in one Temperature range from the melting point to the boiling point of the solvent, within a reaction time not more than two days, are carried out; this creates an intermediate product of the formula:
EMI0003.0011
which with an alkali, e.g. Sodium hydroxide or the like, hydrolyzed and then subjected to a Grignard reaction with an alkyl magnesium halide (B-CH2MgX, where B is as defined above and X is a halogen atom) and ring closure.
In the above-mentioned process according to the invention, the reaction intermediates do not always need to be isolated and purified, since the best yields are not obtained by the isolation in each stage. For the isolation or purification of the products, for example, distillation processes, in particular distillation under reduced pressure, chromatography, or chemical conversion by means of a carbonyl derivative with a carbonyl reactant such as semicarbazide, Girard T or the like, can be used.

After removing the dioxane under reduced pressure, the residue is extracted with dichloromethane.
The extract is washed with water and then with a sodium chloride solution, dried over anhydrous sodium sulphate and then evaporated under reduced pressure; a reddish-orange colored oil product is obtained which gives 51.4 g of 1-ethyl-2-oxo-6-methoxy-1,2,3,4-tetrahydronaphthalene-1-proprionitrile with a yield of 79.9% . <B> 250 </B> ml of an aqueous 20% potassium hydroxide solution are added to a solution of <B> 30.5 g </B> of the compound obtained above in <B> 250 </B> ml of ethanol, and the resulting mixture is refluxed for 6 hours under a nitrogen atmosphere, during which ammonia gas is evolved. The reaction mixture is concentrated under reduced pressure in order to remove the ethanol and the residue is diluted with water. The alkaline solution thus obtained is washed with dichloromethane, acidified with concentrated hydrochloric acid and extracted with ether.
After that, the extract is washed with sodium chloride solution, dried over anhydrous sodium sulfate, and evaporated under reduced pressure; 31.6 g of crude keto acid are obtained, which is 1-ethyl-2-oxo-6-methoxy-1, 2, 3, 4-tetrahydronaphthalene-1-propionic acid (IR: CCl 4 3200-2500 broad, 1710, 1610, 1575, max <B> 1501, 1265, </B> 1045 cm- ').
1.38 g of the keto acid obtained is dissolved in a mixture of 40 ml of acetic anhydride and 0.02 g of anhydrous sodium acetate. The solution is refluxed for three hours under a nitrogen atmosphere and then distilled under reduced pressure to remove the acetic anhydride. The concentrate obtained in this way is diluted with water and extracted with ether.
The extract is washed with dilute aqueous sodium carbonate solution and then with saturated aqueous sodium chloride solution, dried over anhydrous sodium sulfate, and evaporated under reduced pressure; 1.14 g of the corresponding enol lactone are obtained, d. 1. 4a-ethyl-7-methoxy-3, 4, 4a, 9-tetrahydro-2H-1-oxaphenanthene-2-one in the crude state. The distillation gives a pure substance of Kp0.2-0.3 at 200-220 ° C.
