Einrichtung zum Beimpfen von festen Nährböden mit Keimen oder Sera In der mikrobiologischen Methodik ist die Beimp- fung von Nährböden mit Keimsuspensionen eine viel verwendete Technik. Als Nährböden verwendet man unter anderem feste Nährböden, die, in sogenannte Petri-Schalen gegossen, zur Aufnahme des Impfgutes dienen. Zur übertragung einer Keimsuspension auf den festen Nährboden dient häufig eine Impfnadel, mit der man die Keime aus einer geeigneten Keimsuspension auf den festen Nährboden überträgt.
Aus praktischen Grün den ist man bestrebt, mehrere Organismen auf ein und demselben Nährboden anznimpfen. Bei der bisher üb lichen Technik muss jeder zu verwendende Impfstamm einzeln unter sterilen Kautelen auf den festen Nährbo den übertragen werden. Die kunstgerechte Arbeitsweise verlangt, dass die Impfnadel abgeflammt, in die Keim suspension eingetaucht, anschliessend die Impfstelle des festen Nährbodens bestrichen und der beimpfte Nähr boden wieder bedeckt wird. Derselbe Vorgang wiedjr- holt sich für jeden der zu verwendenden Impfstämme.
Um stets vollständige Sterilität zu gewährleisten und auch jede Infektion aus der Luft oder die übertragung von Rückständen von dem vorher berührten Nährbo den zu vermeiden, ist der Vorgang des Abflammens auch dann nötig, wenn nur mit einer einzigen Keim suspension gearbeitet wird. Dabei ist bei diesem Abflam men mit Vorteil darauf zu achten, dass vor jedem Ein tauchen der Nadel in die Keimsuspension die Nadel hinreichend abgekühlt wird, um zu verhindern, dass durch eine zu heisse Nadel die Keime in der Keimsus pension teilweise abgetötet werden, was besonders bei kleinen Volumina der Keimsuspension die Keimdichte beeinflussen würde.
Dieselben Schwierigkeiten bestehen bei der Beimpfung von festen Nährböden mit Typisie- rungssera.
Um zwischen den einzelnen Impfstellen auf ein und demselben Nährboden einen einigermassen gleichmässi- gen Abstand einzuhalten, wurde schon die Verwendung von Impfnadelträgern versucht, in denen mehrere, bei spielsweise vier, fünf oder sogar neun Impfnadeln an- geordnet sind, die gleichzeitig je in einen Impfgutbe- hälter, d. h. einen Keimsuspensionsbehälter bzw. Se rumbehälter getaucht und nachher gleichzeitig mit dem Nährboden in Berührung gebracht und dann gemein sam abgeflammt werden. Die Arbeit mit einem solchen Impfnadelträger ist jedoch umständlich.
Mit der vorliegenden Erfindung wird nun eine Ein richtung zur Verfügung gestellt, die zum Beimpfen von festen Nährböden mit Keimen oder Sera dient und die eine wesentliche Arbeitsersparnis und damit verbunden eine entsprechende Beschleunigung der Arbeit er möglicht und zudem gestattet, Nadelträger mit verhält- nismässig vielen Impfnadeln zu verwenden, wobei gleich zeitig erreicht wird, das Eintauchen von zu heissen Impf nadeln in das Impfgut zu verhindern.
Die erfindungsgemässe Einrichtung ist dadurch ge kennzeichnet, dass sie mindestens vier je mit mehreren Impfnadeln versehene Nadelträger, eine Station, an der die Impfnadeln das Impfgut aufnehmen, eine Impf station und mindestens eine Abflammstation sowie vor teilhafterweise vor der Impfgutaufnahmestation eine Kühlzone aufweist sowie eine Transportvorrichtung, die bei einem Arbeitsschritt die Nadeln des ersten Nadel trägers zu einem Nährboden und von dort zur nächsten Station,
die Nadeln des zweiten Nadelträgers in die Flammen der Abflammstation und von dort zur näch sten Station eventuell die Nadeln des dritten Nadelträ gers in die Kühlzone und von dort zur nächsten Station, und die Nadeln des vierten Nadelträgers in Impfgutbe- hälter und von dort zur nächsten Station fortbewegt.
Nachfolgend wird anhand der beiliegenden Zeich nung ein Ausführungsbeispiel der Erfindung beschrie ben.
In der Zeichnung zeigen: Fig. 1 eine Darstellung der gesamten Einrichtung, Fig. 2 einen Horizontalschnitt durch den Hauptteil der Einrichtung nach der Linie 11-II, Fig. 3 die ihrerseits einen Vertikalschnitt nach der Linie III-III der Fig. 2 darstellt, während Fig.4 Details der die Drehbewegung steuernden Steuerplatten in grösserem Massstab, Fig. 5 ein Schaltschema der Steuerung, und Fig. d eine schematische Darstellung des Arbeits laufes darstellen.
Wie aus den Fig. 2 und 3 ersichtlich ist, ist auf ei ner durch die Verschalung 1 abgedeckten Bodenplatte 2 mit Schrauben 3 der zentrale Ständer 4 befestigt. In diesen Ständer, der eine durchgehende, mit einzelnen Absätzen versehene Bohrung aufweist, ist von unten der mit einem Deckel 6 versehene Druckzylinder 5 einge schraubt. Der Einfachheit halber sind in der Fig. 3 der Kolben 7 und die mit ihm verbundenen Teile in der rechten und der linken Figurenhälfte in zwei verschie denen Stellungen dargestellt. Die linke Hälfte zeigt den Ruhezustand der eingeschalteten Apparatur, während die rechte Hälfte einen nachfolgend noch beschriebenen Zustand eines Bewegungsablaufes darstellt.
Auf der Kolbenstange 8 ist eine Lagerkappe 9 aufgeschraubt, auf welcher frei drehbar die Hubstange 10 aufsteht, an deren oberem Ende die Halteplatte 11 mittels eines Stiftes 11 a befestigt ist. In der Halteplatte 11 sind acht Rohrabschnitte 12 eingesetzt, in denen je eine Füh rungshülse 13 sitzt, die durch einen Stift 14 gesichert ist. Aussen auf jeder dieser Führungshülsen 13, die unten durch einen Schraubdeckel 17 abgeschlossen sind, sitzt eine durch einen Stift 18 gesicherte Hülse 19 mit einem Zentrierbolzen 20 und eine durch einen Stift 21 ge sicherte Hülse 22 mit einem Steuerbolzen 23. In jedem Schraubdeckel 17 ist eine Leitstange 24 vertikal ver schiebbar gelagert.
Am oberen Ende jeder dieser Stan gen 24 ist durch einen Stift 25 ein Führungszapfen 26 befestigt, der durch einen Keil 27 gegen Drehung ge sichert ist. Eine Feder 28, die zwischen dem Schraub deckel 17 und dem Führungszapfen 26 eingespannt ist, drückt die Leitstange 24 nach oben, so dass, falls sich die unten angebrachte Rolle 29 im Bereich der Steuer scheibe 30 befindet, sie an diese angepresst wird. In den übrigen Bereichen wird die Leitstange so weit nach oben gedrückt, dass die unten an ihr angebrachte Trag vorrichtung 31 am Schraubdeckel 17 ansteht, wie das aus der rechten Hälfte der Fig. 3 ersichtlich ist. An je der Tragvorrichtung 31 ist ein je mit neun Impfnadeln 33 versehener Nadelträger 32 befestigt.
Die einzelnen Nadeln sind im Nadelträger derart vertikal verschieb bar gelagert, dass, wenn sie auf einer Unterlage aufste hen und der Nadelträger nach unten bewegt wird, sie sich im Nadelträger 32 nach oben verschieben können, wodurch eine Beschädigung vermieden werden kann.
Aussen am zentralen Ständer 4 sind an zwei einan der gegenüberliegenden Stellen Steuerelemente ange bracht, die die Drehbewegung der Halteplatte<B>11</B> und damit der mit ihm verbundenen Teile, die zusammen ein Karussell bilden. steuern. Diese Steuerelemente sind in der Fig. 4 in grösserem Massstab dargestellt. Sie wer den je durch drei Platten 34, 35 und 36 sowie zwei durch Federn 39 bzw. 40 belastete Weichen 37 und 38 gebildet, die zusammen Führungen für die Zentrier bolzen 20 und die Steuerbolzen 23 bilden. Das ganze Karussell ist durch eine Haube 41 abgedeckt, aus wel cher nur die Nadelträger 32 herausragen.
Auf der Verschalung 1 ist ein Platz 45 für die Auf nahme einer soggenannten Petri-Schale 42, also einer mit einem Agar-Nährboden also einem festen Nährboden 43 versehenen Glasschale vorgesehen. Zweckmässiger- weise kann an dieser Impfstation der elektrische Steuer knopf 44 angebracht sein, der sich jedoch auch anders wo anbringen oder durch einen Fussschalter ersetzen liesse. Neben dieser Impfstation 45 ist ein Platz 46 für die Aufnahme von neun Impfgutbehältern 47, also Keimsuspensionsbehältern oder Serabehältern ausgebil det.
Diese beiden Stationen weisen einen gegenseitigen Abstand von 45 , bezogen auf die Achse der Hubstange 10, auf. Zwei weitere Stationen, von denen die eine einen Abstand von 90 und die andere einen solchen von 135 zur Impfstation 45 besitzt, sind je mit neun Brennstellen 48 versehen, die alle an eine gemeinsame Gaszuleitung 49 angeschlossen sind.
Der vorstehend beschriebene Apparateteil ist, wie man aus der Fig. 1 ersehen kann, mit einem als Ganzes mit 50 bezeichneten Steuerteil durch Leitungen für Gas, Wasser und Elektrizität verbunden. Die im Steuerteil 50 angeordneten Schalt- und Steuerelemente sind im rech ten Teil der Fig. 5 schematisch dargestellt, während der linke Teil dieser Fig. 5 eine schematische Darstellung des vorstehend beschriebenen Hauptteiles ist. Wie man sieht, ist der Druckzylinder 5 durch eine Leitung 51 über ein Dreiwegventil 52 mit einer Druckwasserleitung 53 verbunden. Die dritte am Dreiwegventil 52 ange schlossene Leitung ist die Entleerungsleitung 54.
An stelle einer ans Wasserversorgungsnetz angeschlossenen Druckwasserleitung 53 liesse sich natürlich auch eine an eine Druckluftanlage angeschlossene Leitung ver wenden.
Ein Schalter 55 wird beim Herunterkommen des gan zen Karussells durch eine der Rollen 29 geschlossen und schliesst dadurch den durch das Relais 56 gehenden Stromkreis, so dass dessen Anker anzieht und die bei den Kontakte 56a und 56b schliesst, wodurch einer seits das Relais 56 selbst hält, auch wenn der Kontakt 55 wieder geöffnet ist, und andererseits das das Drei wegventil 52 steuernde Relais 57 angezogen wird, so dass Wasser in den Druckzylinder 5 strömen kann, bis der Kolben 7 seine oberste Stellung einnimmt.
Diesen Betriebszustand zeigt sowohl das in der Fig. 5 darge stellte Schaltschema wie auch die linke Hälfte der Fig. 3, wobei natürlich der Hauptschalter 58 geschlossen und dadurch das elektromagnetische Absperrventil 61 der Gaszuleitung 49 geöffnet ist, die elektrischen Anschlüsse mit dem Stromnetz verbunden und auch die Gas- und die Wasserleitungen an entsprechende Leitungen ange schlossen sind und die Brennstellen 48 bei geöffnetem Hahn 62 brennen.
Wenn nun ein Nährboden geimpft werden soll, so wird der Steuerknopf 44 betätigt, wodurch das Relais 56 stromlos wird und abfällt, so dass durch das öff nen des Kontaktes 56b auch das Relais 57 stromlos wird und sich der Zylinder 5 unter dem Gewicht des Karussells über das nun geöffnete Dreiwegventil 52 und die Leitung 54 entleeren kann. Das Karussell bewegt sich also nach unten. Die über der Petri-Schale 42 be findlichen Impfnadeln impfen den Nährboden 43, die über dem Rand der Steuerscheibe 30 befindliche Rolle 29 gleitet unter diese Steuerscheibe und die über den Suspensionsbehältern 47 befindlichen Nadeln werden in diese Behälter eingetaucht.
Sobald nun eine der Rollen 29 den Schalter 55 schliesst, wie das in der rechten Hälfte der Fig. 3 dargestellt ist, hebt sich das Karussell wieder. Dabei wird dem im mittleren Schlitz der Steuer scheibe 35 geführten Steuerbolzen 23 durch die Weiche 37 eine Drehbewegung erteilt, und er verlässt die Steuer führung auf der rechten Seite in der Fig. 4, so dass der Steuerbolzen 23 der benachbarten Führungshülse 13 von links in die Steuerführung eintritt. Auf diese Art und Weise wird das Karussell bei jedem Hub um 45 gedreht, während das Absenken ohne Drehbewegung er folgt.
Die Geschwindigkeit des Absenkens und die Ge schwindigkeit der Aufwärtsbewegung lassen sich unab hängig voneinander durch Regulierventile 59 und 60 verändern, die in der Zuleitung 53 bzw. der Ableitung 54 angeordnet sind. Die Fig. 6 zeigt schematisch den Bewegungsablauf der Nadelträger.
Selbstverständlich kann je nach Bedarf eine der bei den Abflammstationen ausgeschaltet werden, oder es könnte auch noch eine weitere Abflammstation vorge sehen sein. Dadurch, dass zwischen der letzten Ab flammstation und der Station, an welcher die Impfna deln die Suspension aufnehmen, mehrere leere Stationen vorhanden sind, ist hinreichend Gewähr dafür geboten, dass die Nadeln sich soweit abkühlen, dass sie die Sus pension nicht zerstören können. Je nach Bedarf lassen sich allerdings auch noch Flüssigkeitskühlstationen vor sehen.
Auch könnte die Zahl von acht Nadelträgern und ebenso vielen Stationen vermehrt oder vermin dert werden, jedoch sollten mindestens vier vorhanden sein, damit gleichzeitig eine in die Suspension eintau chen, die andere die Impfung vornehmen, die dritte abgeflammt und die vierte abgekühlt werden kann.
Natürlich könnte anstelle der vorstehend beschriebe nen Schalt- und Steuereinrichtung auch eine einfachere Einrichtung verwendet werden, bei welcher beispiels weise das Dreiwegventil 52 von Hand in die Entlee rungsstellung gebracht und direkt durch das sich sen kende Karussell umgeschaltet wird.

The same difficulties exist when inoculating solid culture media with typing sera.
In order to maintain a reasonably even distance between the individual inoculation sites on the same nutrient medium, attempts have already been made to use inoculation needle holders in which several, for example four, five or even nine inoculation needles are arranged, each of which is simultaneously in a vaccine - container, d. With the present invention, a device is now made available which is used to inoculate solid nutrient media with germs or sera and which enables substantial labor savings and, associated with this, a corresponding acceleration of work and also allows needle carriers with a relatively large number of inoculation needles to use, at the same time achieved to prevent the immersion of hot needles in the vaccine.

Two further stations, one of which is 90 from the vaccination station 45 and the other 135 from the vaccination station 45, are each provided with nine burners 48 which are all connected to a common gas supply line 49.
The apparatus part described above is, as can be seen from FIG. 1, connected to a control part designated as a whole with 50 by lines for gas, water and electricity. The switching and control elements arranged in the control part 50 are shown schematically in the right part of FIG. 5, while the left part of this FIG. 5 is a schematic representation of the main part described above. As can be seen, the pressure cylinder 5 is connected to a pressurized water line 53 by a line 51 via a three-way valve 52. The third line connected to the three-way valve 52 is the drain line 54.
Instead of a pressurized water line 53 connected to the water supply network, a line connected to a compressed air system could of course also be used.
A switch 55 is closed by one of the rollers 29 when the whole carousel comes down and thereby closes the circuit going through the relay 56, so that its armature picks up and the one at the contacts 56a and 56b closes, which on the one hand holds the relay 56 itself , even if the contact 55 is opened again, and on the other hand the relay 57 controlling the three-way valve 52 is attracted so that water can flow into the pressure cylinder 5 until the piston 7 assumes its uppermost position.

As soon as one of the rollers 29 closes the switch 55, as shown in the right half of FIG. 3, the carousel rises again. The control pin 23 guided in the middle slot of the control disk 35 is given a rotary movement by the switch 37, and it leaves the control guide on the right side in FIG. 4, so that the control pin 23 of the adjacent guide sleeve 13 from the left into the Tax guidance occurs. The speed of lowering and the speed of the upward movement can be changed independently of one another by regulating valves 59 and 60 which are arranged in the supply line 53 and the discharge line 54, respectively. 6 shows schematically the sequence of movements of the needle carriers.
Of course, one of the flaming stations can be switched off as required, or another flaming station could also be provided. The fact that there are several empty stations between the last flame station and the station at which the vaccination needles pick up the suspension is sufficient to ensure that the needles cool down to such an extent that they cannot destroy the suspension. However, liquid cooling stations can also be provided as required.
The number of eight needle carriers and the same number of stations could also be increased or decreased, but there should be at least four so that one can be immersed in the suspension, the other can perform the vaccination, the third can be flamed and the fourth can be cooled down.
Of course, instead of the switching and control device described above, a simpler device could also be used, in which, for example, the three-way valve 52 is brought into the emptying position by hand and switched directly by the carousel which is kende itself.
