Gegenstand der Erfindung ist ein Verfahren zur Herstellung des 1-Isopropylamino-2-hydroxy-3-(o-pro pargyloxy-phenoxy)-propans der Formel

Die neue Verbindung besitzt wertvolle phamiakolügi- sche Eigenschassssen. Insbesondere bewirkt sie eine Hem mung adrenergischer ¯-Rezeptoren. Sie kann dementsprechend bei Herz- und kreislauferkrankungen als Me dikamente angewendet werdisn.
Das erfindungsgemϯe Verfahren zur Henstetluag der neuen Verbindung ist dadurch gekennzeichnet, da¯ man o-Propargyloxy-phleíol mit emer Verbindung der aügememen Formel

worin X2 eine Hydroxylgruppe und X1 eine reaktionsfähig veresterte Hydroxylgmippe bedeuten oder Xi und XO gemeinsam eine Sauersboffbrücke bilden, umsetzt.
Ein reakttonsfäbigefr Ester ist dabei z. B. derjenige einer starken organischen oder anorganischen SÏure, wie vor allem einsr Halogenwafssorstoffsäure, z. B. der Chlor-, Brom- oder JodwasserstoffsÏure, oder einer Sul fonsäure, wie einer ArylsulfonsÏure, z. B. der p-Toluol- sulfonsÏure. Dabei arbeitet man in blicher Weise. Bei Verwendung der Ester verfährt man zweckmässig in Gegenwart eines sÏurebindenden Kondensationsmittels, insbesondere eines zur Salzbildung mit dem Phenol geeignten Kondensationsmittels, oder man verwendet ein Metall-, wie Alkalisalz des Phenols. So kann man in Gegenwart von Alkalialkoholaten arbeiten.
Die AusgamsOoffe sind bekannt oder können nach an sich bekannten Methoden gewonnen werden.
Je nach den Verfahrensbedingungen und Aus, gangsstoffen erhÏlt man den Endstoff in freier Form oder in der ebenfalls in der Erfindung inbegriffenen Form seiner Salze. Die Salze des Endstoffes k¯nnen in an sich be- kannter Weise, z. B. mit Alkalien oder Ionenaustau schern in die freie Base bergef hrt werden. Von der letzteren lassen sich durch Umsetzung milt organischen oder anorganischen Säuren, insbesondere solchen, die zur Bildung von therapeutisch verwendbaren Salzen geeignet sind, Salze gewinnen.
Als solche Säuren seien beispielsweise genannt : HalogenwasserstoffsÏuren, SchwefelsÏuren, PhosphorsÏuren, Salpetersäure, PerchlorsÏure, aliphatische, alicyclische, aromatische oder heterocyoliache Carbon-oder Sulfonsäuren, wie Ameisen-, Essig-, Propion-, Bernstein-, Glykol-, Milch-, apte-, Wein-, Zitronen-, Ascorbin-, Malein-, Hydroxy malein-oder Brenztraubensäure ; Phenylesstg-, Benzoe-, p-Aminobenzoe-, Anthranil-, p-Hydroxy-benzoe-, Salicyl-oder p-AminosalicylsÏure, EmbonsÏure, Methan sulfon-, Abhansulfon-, Hydroxyäthansulfon-, Athylen- suffonsäure ;
Halogenbenzolsulfon-, Toluolsulfon-, NaphthalinsulfonsÏure oder SulfanilsÏure ; Methionin, Trypto phan, Lysin odeur Argmia.

B. die Pikrate, können auch zur Reinigung der er- haltenen freien Base dienen, indem man die freie Base in Salze überführt, diese abtrennt und aus den Salzen wiederum die Base freimacht. Infolge der engen Beziehungen zwischen der neuen Verbindung in. freier Form und in Form ihrer Salze sind im vorausgegange- nen und nachfolgend unter der freien Base sinn-und zweckmϯig, gegebenenfalls auch dfie entsprechenden Salze zu verstehen.
Die noue Verbindung kann als Racemat oder in Form der Antipoden vorliegen. Das Racemat lässt sich in üblicher Weise in die Antipoden zerlegen.
Die neue Verbindung kann z. B. in Form pharmazeutischer Präparate Verwendung findien, welche sie in freier Form oder gegebenenfalls in Form ihrer Salze in Mischung mit einem für die enterale oder parenterale Applikation geeigneten pharmazeutischen, organischen oder anorganischen, festen oder flüssigen Trägermaterial enthalten.
Im folgenden Beispiel werden die Temperaturen in Celsiusgraden angegeben.
Beispiel
10 g o-Propargyloxy-phenol, 8 g 3-Isopropylamino2-hydroxy-l-chlorpropan und 10 g Pottasche werden in 150 ml Aceton während 5 Stunden bei 50 gerührt.
Hierauf filtriert man die Pottasche ab und dampft im Vakuum zur Trockne ein. Den Rückstand l¯st man in Ather und extrahert mit In Natronlauge. Die Atherschicht wird abgetrennt und eingedampft. Es verbleibt das 1-Isopropylamino-2-hydroxy-
3-(o-propargyloxy-phenoxy)-propan der Formel

das bei 48-52 schmilzt.
Das Hydrochlorid der Verbindung schmilzt bei 96-97 .
Das als Ausgangsmatenal 1 verwendete A-Propargyloxy-phenol kann auf folgende Weise hergestellt werden.
110 g Brenzkatechin werden in l Liter Aoeton ge- l¯st und unter gutem Rühren mit 150 g Pottasche und 120 g Propargylbromid wÏhrend 4 Stunden auf 80 erwärmt. Hierauf filtriert man den ungelösten Anteil ab und dampft im Vakuum das Lösuingsmitttel ab. Den Rückstand destilliert man im Wasserstrahivakuum. Man erhält das o-Propargyloxy-phenol, das bei 120 bis 130¯/14 mm siedet.
