und deren Salze blutzuckersenkende Eigenschaften be sitzen.
EMI0001.0050
vorzugsweise folgende Bedeutung haben: Piperidin, a-Methyl-piperidin, ss-Methyl-piperidin, y-Methylpiperidin, a,y-Dimethyl-piperid'in, a,a'-Dimethyl- piperidin.

Process for the preparation of benzenesulfonylsemicarbazides It is known that certain benzenesulfonylurea derivatives have blood sugar-lowering properties and are suitable as antidiabetics which can be administered orally (cf., for example, Arzneimittelforschung, Volume 8 [1958], pages 444-454).
It has now been found that benzenesulfonyl semicarbazide of the formula
EMI0001.0015
wherein X is an alkyl radical containing 2 to 6 carbon atoms and -Z-Z'- is an alkylene chain containing 5 carbon atoms, which may optionally be substituted by low molecular weight alkyl radicals,
and their salts have blood sugar lowering properties.
The present invention relates to a process for the production of such benzenesulfonyl-semicarbazide, which is characterized in that in the corresponding benzenesulfonyl-thiosemicarbazides, for example with the help of oxides or salts of heavy metals or by using oxidizing agents such as hydrogen peroxide replaced the sulfur with oxygen.
The substituent X can, for example, have the following meaning: p-ethyl, p- (n) -propyl, p-isopropyl, p-tert-butyl, p- (n) -butyl, p-isobutyl, p-pentyl (3) , p-tert-amyl, p- (n) -hexyl.
The group can counter this
EMI0001.0050
preferably have the following meanings: piperidine, α-methyl-piperidine, β-methyl-piperidine, γ-methylpiperidine, α, γ-dimethyl-piperidin, α, α-dimethylpiperidine.
The benzenesulfonyl semicarbazides obtainable by the process according to the invention are valuable medicaments which are characterized in particular by a good blood sugar-lowering effectiveness with low toxicity.
For example, 4- (4-ethylbenzenesulfonyl) -1,1-pentamethylene-semicarbazide, as can be seen from the table below, lowers the blood sugar level in test animals for much longer than N- (4-methylbenzenesulfonyl, which is known as an oral antidiabetic agent ) - N'-n-butylurea. In the test, the compounds are administered to the rabbits in a dose of 400 mg / kg in the form of the sodium salts and the dogs in a dose of 5 mg / kg in the form of the free compound.

serve to treat diabetes mellitus, the sulfonyl semicarbazides can be used either as such or in the form of their salts with bases or acids or in the presence of substances which lead to salt formation. For example, the following can be used for salt formation:
Alkaline agents, such as alkali or alkaline earth hydroxides, carbonates, bicarbonates and physiologically compatible organic bases; also acids, such as hydrochloric acid, hydrobromic acid, sulfuric acid and amidosulfonic acid.
The medical preparations are preferably tablets which, in addition to the process products, contain the usual auxiliaries and carriers, such as talc, starch, lactose, tragacanth, magnesium stearate.
Example 4- (4-Ethyl-benzenesulfonyl) -1,1-pentamethylene-semicarbaide 4.9 g of 4- (4-ethyl-benzenesulfonyl) -1,1-pentamethylene-thiosemicarbazide [melting point 177- 178 C, made from N- (4-ethyl-benzenesulfonyl) -N1-phenyl-thiourea and N-amino-piperidine] are dissolved in 20 ml of 2N sodium hydroxide solution and 100 ml of water while warming.
Cool slightly, add 10 ml of hydrogen peroxide solution (35%) and heat for 30 minutes on the steam bath. The solution is cooled, clarified with charcoal and acidified with dilute acetic acid. It is filtered off with suction, reprecipitated from dilute ammonia and recrystallized from ethyl acetate. The 4- (4-ethyl-benzenesulfonyl) -1,1-pentamethylene-semicarbazide melts at 174-176 C.
