Verfahren zur Herstellung von neuen tricyclischen Hydxoxylaminen Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung neuer tricyclischer Hydroxylamine.

Wie nun gefunden wurde, besitzen solche Verbindungen und, ihre Salze mit anorganischen und organischen Säuren wert volle, insbesondere zentrale sowie auch periphere phar makologische Eigenschaften; sie antagonisieren die Wir kung von Reserpin und Tetrabenazin und potenzieren diejenige von Noradrenalin.
Die neuen Verbindungen .der Formel.' I und ihre pharmazeutisch annehmbaren, verträglichen Salze kön nen beispielsweise zur Behandlung von Geisteskrank heiten, insbesondere von Depressionen, oral und rektal angewendet werden; die wässrigen Lösungen dieser Salze können auch, parenteral verabreicht werden. In den Verbindungen der Formel I kann A z. B. den 1 -Äthylanyl- 2-yliden- (=CH-CH2-), 1-Propanyl-3- yliden- (=CH-CH2-CH2-), 2-Methyl-l-propanyl-3-yli- den -oder den 1-Methyl-1-propanyl-3-yliden-rest bedeu ten.
R kann als niedere Alkylgruppe z. B. die Methyl-, Äthyl-, Propyl-, Isopropyl-, n-Butyl- oder die sek. Butyl- gruppe sein. Bedeutet =AN(OH)R einen N-Hydroxy C- pyrrolildinyladkyliden-, N - Hydroxy - piperidylalkyliden- oder den N-Hydroxy-C-hexahydroazepinylalkyliden-rest, so handelt es sich z.

Die Amme der Formel II werden in Verbindungen der Formel IH übergeführt, indem man eine Lösung eines Acylperoxyds, insbesondere diejenige dies Acetyl- peroxyds, Benzoylperoxyds oder Acetyl-benzoyl-per- oxyds, zu einer solchen eines Amins der Formel;
II fügt - oder umgekehrt, indem man die Lösung eines Acyl- peroxyds vorlegt und diejenige eines Amins zugibt - und eine Reaktionstemperatur von etwa - 50 bis -i- 80 , vorzugsweise eine solche von 0 bis 20 , einhält. Als Lösungsmittel für die Amine und Asylperoxyde eignen sich chlorierte Kohlenwasserstoffe, wie z. B. Chloro form oder Tetrachlorkohlenstoff, ätherartige Stoffe, wie z. B. Diäthyläther oder Tetrahydrofuran, Kohlenwasser stoffe, wie z. B. Benzol oder Toluol, ferner Dimethyl- formamid.
Die erhaltenen Zwischenprodukte, das heisst die N-Acyloxyverbindungen der Formel III, lassen sich z. B. mit einem Alkalihydroxyd, insbesondere mit Kalium hydroxyd, in. einem wässrigen, niederen Alkamol, wie z. B. Methanols oder Äthanol:, zu den Verbindungen der Formel I solvolysieren. Anstelle von wässrig-alkoholi scher Alkalihydroxydlösung kann man zur Solvolyse auch ein Alkalialkoholat, wie z. B. Natriummethylat, in einem niederen Alkanol, wie z. B. Methylalkohol, ver wenden.
Einige Ausgangsstoffe der Formel II sind bekannt. Genannt seien die 5-(2'-Methylamino-äthyliden)-, 5-(2'-Methylamino-propyliden)-, 5-(3'-Methylamino-propyliden)-, 5-(3'-Methylamino-2'- methyl-propyliden)-, 5-[2'-(2"-Pyrrolidinyl)-äthyliden]-, 5-[2'-(2"-Piperidyl)-äthyliden]- und 5-[2'-(2"-Hexahydroazepinyl)-äthyliden]-derivate von 5H-Dibenzo[a,d]cyclohepten und 10,11-Dihydro-5H-dibenzo[a,d]cyclohepten sowie die den genannten Methylaminoverbindungen ent sprechenden Amino-, Äthylamino-, Isopropylamino- und sek. Butylaminoverbindungen.
Die Zwischenprodukte <I>der</I> Formel IH sind neu.
Die neuen Wirkstoffe werden, wie weiter vorn er wähnt, peroral, rektal und parenteral verabreicht. Die täglichen Dosen der freien Basen oder von nichttoxi schen Salzen derselben bewegen sich zwischen 10 und 300 mg für erwachsene Patienten. Geeignete Dosenein heitsformen, wie Dragees, Tabletten, Suppositorien oder Ampullen, enthalten vorzugsweise 5-50 mg eines erfin dungsgemässen Wirkstoffes oder eines nichttoxischen Salzes desselben.
Unter nichttoxischen Sahen der erfindungsgemäss verwendbaren Basen sind Salze mit solchen Säuren zu verstehen, deren Anionen bei den in Frage kommenden Dosierungen pharmakologisch. annehmbar sind, das heisst keine toxischen Wirkungen ausüben. Ferner ist es von Vorteil, wenn,die zu verwendenden Salze gut kri stallisierbar und nicht oder wenig hygroskopisch sind.
Als nichttoxische Salze der Wirkstoffe kommen z. B. die Salze mit der Chlorwasserstoffsäure, Bromwasserstoff säure, Schwefelsäure, Phosphorsäure, Methansulfon- säure, Äthandisulfonsäure und ss-Hydroxyäthansulfon- säure in Betracht.
Doseneinheitsformen für die perorale Anwendung enthaften als Wirkstoff vorzugsweise zwischen 1 und 90 % einer Verbindung .der Formel I oder eines nicht toxischen Salzes einer solchen. Zu ihrer Herstellung kombiniert man: .den Wirkstoff z.
B. mit festen, pulver förmigen, Trägerstoffen, wie Lactose, Saccharose, Sor bit, Mannit; Stärken, wie Kartoffelstärke, Maisstärke oder Amylopektin, ferner Laminariapulver oder Citrus- pulpenpulver; Cellulosederivaten oder Gelatine, gegebe- nenfalls unter Zusatz von Gleitmitteln, wie Magnesium- oder Calciumstearat oder Polyäthylenglykolen (Carbo- waxen) von geeigneten Molekulargewichten zu Tabletten oder zu Drag@e-Kernen.
Letztere überzieht man: bei spielsweise mit konzentrierten Zuckerlösungen, welche z. B. noch arabischen Gummi, Talk und/oder Titan dioxyd enthalten können, oder mit einem in leichtflüch tigen organischen Lösungsmitteln oder Lösungsmittel gemischen gelösten Lack. Diesen Überzügen können Farbstoffe zugefügt werden, z. B. zur Kennzeichnung verschiedener Wirkstoffdosen.
Als Doseneinheitsformen für die rektale Anwendung kommen z. B. Suppositorien, welsche aus einer Kom bination eines Wirkstoffes oder eines geeigneten Salzes desselben mit einer Neutralfettgrundlage bestehen, oder auch Gelatine-Rektalkapseln, welche eine Kombination des Wirkstoffes oder eines geeigneten Salzes, desselben mit Polyäthylenglykolen (Carbowaxen) von geeignetem Molekulargewicht enthalten, in Betracht.
Ampullen zur parenteralen, insbesondere intra muskulären Verabreichung enthalten vorzugsweise ein wasserlösliches Salz eines Wirkstoffes in einer Konzen tration von vorzugsweise <B>0,5-5%,</B> gegebenenfalls zu sammen mit geeigneten Stabilisierungsmitteln und Puf fersubstanzen, in wässriger Lösung.
Die folgenden Vorschriften sollen die Herstellung von Tabletten und Dragees näher erläutern: a) 250g N-[3-(10',11'-Dihydro-5'H-dibenzo[a,d] cyclohepten-5'-yliden)-propyl]-N-methyl- hydroxylamin-hydrochlorid werden mit 175,80 g Lactose und 169,70g Kartoffel stärke vermischt, die Mischung mit einer alkoholischen Lösung von 10 g Stearinsäüre befeuchtet und durch ein Sieb granuliert. Nach dem Trocknen mischt man:
160 g Kartoffelstärke, 200 g Talk, 250 g Magnesiumstearat und 32 g kolloidales Siliciumdioxyd zu und presst die Mischung zu<B>10000</B> - Tabletten vom je 100 mg Gewicht und 25 mg Wirkstoffgehalt (Hydrochlorid), die ge- wünschtenfalls mit Teilkerben zurr feineren Anpassung der Dosierung versehen sein können.
b) Aus 250g N-[3-(10',11'-Dihydro-5'H-dibenzo [a,d]cyclohepten-5'-yliden)-propyl]-N-methyl- hydroxylamin-hydrochlorid, 175,90g Lactose und der alkoholischen Lösung von 10 g Stearinsäure stellt man ein Granulat her, das man nach dem Trocknen mit 56,60 g kolloidalem Silicium dioxyd, 165 g Talk, 20 g Kartoffelstärke und 2,50 g Magnesiumstearat mischt und zu 10 000 Drag@e-Kernen presst. Diese werden anschliessend mit einarm konzen- trierten Sirup aus 502,28g krist.
Saccharose, 6 g Schel lack, 10 g arabischem Gummi, 0,22 g Farbstoff und 1,5 g Titandioxyd überzogen unkt getrocknet. Die er haltenen Dragées wiegen je 120 mg und enthalten je 25 mg Wirkstoff.
Die nachfolgenden Beispiele erläutern die Herstel lung der neuen Verbindungen der Formel I und von bisher nicht beschriebenen Zwischenprodukten näher, stellen jedoch keineswegs die einzigen Ausführungsfor- men derselben dar. Die Temperaturen sind in Celsius graden angegeben.
<I>Beispiel 1</I> a) 13,2 g 5-(3'-Methylamino-propyliden)-10,11-di hydro-5H-dibenzo[a,d]cyclohepten werden in, 300 ml trockenem Diäthyläther gelöst und auf 5 gekühlt. Zu dieser Lösung tropft man innerhalb von 45 Minuten un ter Rühren eine solche von 6,65 g 97%igem Benzoyl- peroxyd in 30 m1' Chloroform und 90 ml trockenem Di- äthyläther. Anschliessend rührt man noch 3 Stunden bei 0 bis 5 . Dann saugt man den entstandenen Nieder schlag, das benzoesaure Salz von 5-(3'-Methylamino-propyliden)-10,11-dihydro- 5H-dibenzo[a,d]cyclohepten, ab und wäscht ihn mit Diäthyläther nach.
Dass klare Filtrat wird mit 2n Natriumcarbonatlösung, 2n Salz säure und Wasser gewaschen, über Natriumsulfat :ge trocknet und im Vakuum eingedampft. Man erhält 8,4 g N-[3-(10',11'-Dihydro-5'H-dibenzo[a,d] cyclohepten-5'-yliden)-propyl]-N-methyl-O- benzoyl-hydroxylamin; dieses kristallisiert aus Methanol, Smp. 90-100 , nach Umkristallisieren aus Diäthyläther-Pentan Smp. 108 bis 110 .
b) 6,3 g des nach a hergestellten Hydroxylamins werden in. 150 ml siedendem Äthanol gelöst und kurz nacheinander mit 10 ml Wasser und' 10 ml 2n Kali lauge versetzt. Man kühlt,die Lösung anschliessend auf 20 ab, lässt sie 15 Minuten bei 20 stehen und dampft das Äthanol im Vakuum ab. Der Rückstand wird mit 20 ml Wasser verdünnt und erschöpfend mit Diäthyl- äther extrahiert.
Man trocknet den ätherischen Ex'trak't über Natriumsulfat und .dampft ihm: in Vakuum ein. Der Rückstand kristallisiert aus Diäthyläther-Pentan. Man erhält 4,0 g N-[3-(10', I 1'-Dihydro-5'H-dibenzo[a,d] cyclohepten-5'-yliden)-propyl]-N-methyl- hydroxylamin, Smp.93-94 .
Aus der freien Base, gelöst in Chloro form, wird mit ä'the'rischer Salzsäure das Hydrochlorid bereitet, Smp. 142-143 .
<I>Beispiel 2</I> a) Analog Beispiel 1 a wird mit Benzoylperoxyd er- halten: a1) aus dem 5-(3'-Amino-propyliden)-10,11- dihydro-5H-dibenzo[a,d]cyclohepten das N-[3-(10',11'-Dihydro-5'H-dibenzo[a,d] cyclohepten-5'-yliden)-propyl]-O-benzoyl- hydroxylamin, a2) aus dem 5-(3'-Methylamino-2'-methyl propyliden)-10,11-dihydro-5H-dibenzo[a,d]- cyclohepten das N-[3-(10',11'-Dihydro-5'H-dibenzo[a,d] cyclohepten-5'-yliden)-2-methyl-propyl]- N-methyl-O-benzoyl-hydroxylamin, a3) aus dem 3-(10',
11'-Dihydro-5'H-dibenzo[a,d]- cyclohepten-5'-ylidenmethyl)-pyrrolidin das 3-(10',11'-Dihydro-5'H-dibenzo[a,d] cyclohepten-5'-ylidenmethyl)-1-benzoyloxy- pyrrolidin, a4) aus dem 3-(10',11'-Dihydro-5'H-dibenzo- [a,d]cyclohepten-5'-ylidenmethyl)-piperidin das 3-(10',11'-Dihydro-5'-H-dibenzo[a,d] cyclohepten-5'-ylidenmethyl)-1-benzoyloxy- piperidin, a5) aus dem 2-[2'-(10",11"-Dihydro-5"H-dibenzo- [a,d]cyclohepten-5 "-yliden)-äthyl]-piperidin das 2-[2'-(10",11"-Dihydro-5"H-dibenzo [a,d]cyclohepten-5"-ylidien)
-äthyl]-1-benzoyl- oxy-piperidin und a5) aus dem 5-(3'-Methylamino-propyliden)- 5H-dibenzo[a,d]cyclohepten das N-[3-(5'H-Dibenzo[a,d]cyclohepten- 5'-yliden)-propyl]-N-methyl-O benzoyl- hydroxylamin.
b) Analog Beispiel 1 b wird mit Kalilauge erhalten: bi) aus dem N-[3-(10',11'-Dihydro-5'H-dibenzo [a,d]cyclohepten-5'-yliden)-propyl]-O-benzoyl- hydroxylamin das N-[3-(10',11'-Dihydro-5'H-dibenzo[a,d]- cyclohepten 5'-yliden)-propyl]-hydroxylamin, b2) aus dem N-[3-(10',11'-Dihydro-5'H- dibenzo [a,d]cyclohepten-5'-yliden)-2-methylpropyl]- N-methyl-O-benzoyl-hydroxylamin dass N-[3-(10',11'-Dihydro-5'H-dibenzo[a,d] cyclohepten-5'-yliden)

As unit dosage forms for rectal use, for. Ampoules for parenteral, in particular intramuscular, administration preferably contain a water-soluble salt of an active ingredient in a concentration of preferably 0.5-5%, optionally together with suitable stabilizers and buffer substances, in an aqueous solution.
The following instructions are intended to explain the production of tablets and dragees in more detail: a) 250g of N- [3- (10 ', 11'-dihydro-5'H-dibenzo [a, d] cyclohepten-5'-ylidene) propyl] -N-methyl-hydroxylamine hydrochloride are mixed with 175.80 g of lactose and 169.70 g of potato starch, the mixture is moistened with an alcoholic solution of 10 g of stearic acid and granulated through a sieve. After drying, mix:
160 g of potato starch, 200 g of talc, 250 g of magnesium stearate and 32 g of colloidal silicon dioxide are added and the mixture is pressed into <B> 10,000 </B> tablets, each weighing 100 mg and containing 25 mg of active ingredient (hydrochloride), if desired Partial notches for a finer adjustment of the dosage can be provided.
b) From 250 g of N- [3- (10 ', 11'-dihydro-5'H-dibenzo [a, d] cyclohepten-5'-ylidene) propyl] -N-methyl-hydroxylamine hydrochloride, 175.90 g Lactose and the alcoholic solution of 10 g of stearic acid are used to produce granules which, after drying, are mixed with 56.60 g of colloidal silicon dioxide, 165 g of talc, 20 g of potato starch and 2.50 g of magnesium stearate and added to 10,000 dragons -Presses cores. These are then mixed with one-arm concentrated syrup made from 502.28 g of crystalline.
Sucrose, 6 g shell lacquer, 10 g arabic gum, 0.22 g dye and 1.5 g titanium dioxide coated and dried. The dragees obtained each weigh 120 mg and each contain 25 mg of active ingredient.
The following examples explain the preparation of the new compounds of the formula I and of intermediates not previously described, but are by no means the only embodiments of the same. The temperatures are given in degrees Celsius.
<I> Example 1 </I> a) 13.2 g of 5- (3'-methylamino-propylidene) -10,11-dihydro-5H-dibenzo [a, d] cycloheptene are dissolved in 300 ml of dry diethyl ether and chilled to 5. A solution of 6.65 g of 97% benzoyl peroxide in 30 ml of chloroform and 90 ml of dry diethyl ether is added dropwise to this solution in the course of 45 minutes, with stirring. The mixture is then stirred for a further 3 hours at 0 to 5. The resulting precipitate, the benzoic acid salt of 5- (3'-methylamino-propylidene) -10,11-dihydro-5H-dibenzo [a, d] cycloheptene, is then suctioned off and it is washed with diethyl ether.
The clear filtrate is washed with 2N sodium carbonate solution, 2N hydrochloric acid and water, dried over sodium sulfate and evaporated in vacuo. 8.4 g of N- [3- (10 ', 11'-dihydro-5'H-dibenzo [a, d] cyclohepten-5'-ylidene) propyl] -N-methyl-O-benzoyl-hydroxylamine are obtained ; this crystallized from methanol, mp 90-100, after recrystallization from diethyl ether-pentane, mp 108-110.
b) 6.3 g of the hydroxylamine prepared according to a are dissolved in. 150 ml of boiling ethanol and 10 ml of water and 10 ml of 2N potassium hydroxide solution are added in quick succession. It is cooled, the solution is then reduced to 20, it is left to stand for 15 minutes at 20 and the ethanol is evaporated off in vacuo. The residue is diluted with 20 ml of water and extracted exhaustively with diethyl ether.
The essential extract is dried over sodium sulphate and evaporated: in a vacuum. The residue crystallizes from diethyl ether-pentane. 4.0 g of N- [3- (10 ', I 1'-dihydro-5'H-dibenzo [a, d] cyclohepten-5'-ylidene) propyl] -N-methyl-hydroxylamine, m.p. 93-94.
The hydrochloride is prepared from the free base, dissolved in chloro form, with ethereal hydrochloric acid, melting point 142-143.
