Verfahren zur Herstellung eines neuen Halogenpropans Die Erfindung betrifft ein Verfahren zur Herstellung von 1- Brom -1 - chlor-2,2-difluor-propan der Formel CHBrCl-CF2-CH3. Diese Verbindung ist als nicht ent flammbares Inhalatüons-Anästheticum bei Mensch und Tier verwendbar.
Der Sicherheitsfaktor des 1-Brom- 1-chlor-2,2-difluor- propans bei der Verwendung als, Inhalations-Anästheti- cum ist hoch. Bei pathogenfreien Mäusen ist das LC 50/AC 50-Verhältnis (LC 50 ist die minimale Volu menkonzentration des Dampfes, die benötigt wird, um 50 % der Versuchsmäuse in 30 Minuten zu töten, wäh rend AC 50 die minimale Volumenkonzentration des Dampfes, ist, die benötigt wird, um. bei 50% der Ver suchsmäuse in 30 Minuten volle Anästhesie zu errei chen) 3,0:0,6 das heisst 5.
Unter gleichen Bedingun gen ist das LC50/AC50-Verhältnis von Äther 10,1 zu 3,51 das heisst 2,88, dasjenige von Chloroform 2,63: 0,99 das heisst 2,66 und dasjenige von Halothan 3,75 : 0,8 das heisst 4,69. Bei Vorversuchen mit Mäusen gab es keine verzögerten Todesfälle. Bei Voruntersuchungen an Kaninchen wurde gefunden, dass der Übergang zur vollen chirurgischen Anästhesie und die, Erholung davon mit 1-Brom-1-Chlor-2,2-difluor-propan sanft ist.
Die Herz frequenz unter Anästhesie war bei Kaninchen normal und das Elektrokardiogramm zeigte keine Unregelmä- ssigkeiten. Die Verbindung verursachte bloss eine kleine Depression von geringem Umfang.
Das erfindungsgemässe Verfahren zur Herstellung von 1-Brom-1-chlor-2,2-difluor-propan ist dadurch gekenn zeichnet, dass man 1-Chlor-2,2-diflour-propan bzw. 1- Brom-2,2-dfluor-propan mit Brom bzw. Chlor halo geniert.
Man kann so vorgehen, dass man 1-Chlor-2,2-difluor- propan mit, Brom erhitzt. Nach einer anderen Methode wird die Verbindung durch Erhitzen von 1-Brom-2,2- difluor-propan zusammen mit Chlor gewonnen.
Wird die Reaktion durch Bromierung von 1-Chlor-2,2- difluor-propan in der Gasphase oder durch Chlorierung von 1-Brom-2,2-difluor-propan in der Gasphase durch- geführt, so werden vorzugsweise Temperaturen im Be reich von 250 bis 600 C und insbesondere Tempera turen im Bereich von 400 bis 525 C angewendet.
Die Bromierung und Chlorierung kann auch in Ge genwart von ultraviolettem Licht erfolgen, in welchem Fall niedrigere Temperaturen angewendet werden kön nen als bei der rein thermischen Bromierung bzw. Chlo- rierung. Bei all diesen Halogenierungsverfahren wird vorzugsweise das Chlordifluor-propan bzw. Bromdifluor- propan im molaren Überschuss gegenüber dem Halo gen angewendet.
Inhalationsanästhesie-Mischungen, die 1-Brom-1- chlor-2,2-difluor-propan und Sauerstoff in zur Erzeu- gung von Anästhesie bei Mensch oder Tier geeigneten Mengenverhältnissen enthalten, können auf jede be kannte zweckmässige Art hergestellt und verwendet wer den.
Zum Beispiel kann das Anästheticum in Apparaten oder Maschinen verwendet werden, die die Verdamp fung flüssiger Anästhetica und deren Vermischung mit Sauerstoff oder Luft oder anderen sauerstoffhaltigen Gasgemischen in einem zur Aufrechterhaltung der At mung geeigneten Verhältnis gestatten. Das Inhalations- Anästheticum kann in Behältern gelagert werden, wie sie gewöhnlich für derartige Anästhetica, z.
B. Äther, Chloroform oder Halothan, verwendet werden.
Soweit das 1 - Brom -1-chlor-2,2-difluor-propan für Anästhesiezwecke benutzt wird, sollte es von toxischen Verunreinigungen, die vom Herstellungsverfahren her rühren können, frei sein. Das erfindungsgemäss herge stellte Anästheticum kann mit pharmazeutisch zulässigen Stoffen vermischt werden, z. B. mit Stabilisatoren und dergleichen. Als Stabilisatoren können z.
B. flüchtige Sub stanzen wie Äthanol, das physiologisch unbedenklich ist, oder ein nicht flüchtiges Mittleu, das bei der Verdamp fung nicht mitgenommen wird, z. B. Thymol, verwendet werden.
Das neue Inhalations-Anästheticum kann auch, wenn des erwünscht ist, mit einem oder mehreren bekannten Inhalations-Anästheticas kombiniert werden, z. B. mit Äther, Chloroform, N2O, Äthylen, Cyclopropan, Halo- than, Divinyläther, Trifluoräthyl-vinyl-äther, Methoxy- fluran oder Trichloräthylen.
Das Anästheticum wird vorzugsweise durch Inhala tion verabreicht im Gemisch mit Sauerstoff und <I>gege-</I> benenfalls weiteren Komponenten, z. B. wie folgt: Das Anästheticum wird verdampft und mit Sauerstoff ver mischt, der z.
B. in reiner Form oder in Form von Luft eingesetzt wird, wobei ein gasförmiges, anäsithetisches Gemisch gebildet wird, das einen zur Erzielung der gewünschten Narkosetiefe ausreichenden Anteil an An ästheticum und anderseits genügend Sauerstoff enthält, um die Atmung normal aufrechtzuerhalten. Der Dampf des Anästheticums und der Sauerstoff bzw. das sauer stoffhaltige Gasgemisch können vorgemischt oder auch erst bei der Verabreichung zusammengemischt werden, um die Lunge mit der gewünschten Menge Anästheticum zu versehen.
<I>Beispiel 1</I> 235 g 1-Chlor-2,2-difluor-propan wurden in einen Glaskolben gebracht, auf den ein mit einem Vakuum mantel versener, versilberter Glasfraktionieraufsatz von 90 cm Länge der mit Fenske -Spiralen versehen war, montiert war. Ein Rohr verband den Fraktionier- aufsatz mit einem ringförmigen Gefäss, in dessen Inne rem eine Quecksilberdampflampe angeordnet war. Ein Seitenarm war an das genannte Rohr angeschlossen, um Bromdampf einzulassen. Der Glaskolben wurde, un ter Rückfluss erhitzt und die verdampfte Verbindung ging durch den Fraktionieraufsatz, um mit dien Brom im ringförmigen Gefäss bei einer Temperatur im Bereich von 70 bis 170 C zu reagieren.
Die obersten Arme des ringförmigen Gefässes waren mit Doppelfläche-Rückfluss kühler verbunden und das Kondensat wurde durch eine Siphoneinrichtung direkt in den Glaskolben zurückge führt. Insgesamt wurden 320 g Brom in einer Zeit spanne von 12 Stunden durch den Seitenarm in die Apparatur eingeführt.
Das Rohprodukt im Kolben wurde mit kalter Na- triumhydroxydlösung und dann mit Wasser gewaschen, getrocknet und fraktioniert destilliert. Eine einfache Fraktionierkolonne, die ungefähr 15 theoretische Teller enthielt, wurde zur Destillation verwendet, so dass die erzielte Ausbeute nicht so hoch war wie bei Verwen dung einer wirksameren Fraktionierkolonne.
Es wurden 111 g eines Produktes von 99,5 % Reinheit erhalten, was einer Ausbeute von 27 % des verbrauchten Chlor- difluorpropans entspricht. Das Produkt wurde als 1- Brom-1-chlor-2,2-difluor-propan durch Massenspektro- metrie identifiziert.
