Verfahren zur Herstellung eines neuen Hexapeptids
Die vorliegende Erfindung bezieht sich auf ein Ver fahren zur Herstellung des neuen Hexapeptids LAlanyl- L-phenylalanyl-L-isoleucycl-glycyl-L-methioninamid, das besonders bei Menschen und Hunden eine hohe gefäss- erweiternde und blutdrucksenkende Wirkung besitzt und daher in der Therapie der Hypertonie Verwendung fin- den kann.
Das erfindungsgemässe Verfahren zur Herstellung des genannten Hexapeptids, das die Formel

aufweist, ist dadurch gekennzeichnet, dass das an der Aminogruppe geschützte Tripeptid L-Alanyl-L-phenyl- alanyl-L-isoleuin der Formel

umgesetzt und hierauf die schützende Gruppe R2 ab gespalten wird.
Die das Amin schützende Gruppe R2 kann z. B. eine toshyl-, Carbobenzoxy-, Carbo-t-butoxy-, Trifuor acetyl-, Tritylgruppe oder eine andere in der Polypeptid- chemie übliche Schutzgruppe sein.
Säurederivate, die mit der Aminogruppe reagieren können, sind das Säurechlorid, Säureazid, p-Nitro-phen ylester usw. ; die Kondensation kann auch in Gegenwart von Dizykolhexylcarbodiimi d ausgefühfrt werden.
Die Abspaltung der-schützenden Gruppe kann z. B. nach bekannten Verfahren ausgeführt werden, je nach
Fall, z. B. durch Behandlung mit Alkalihydroxyden oder mit Natrium in flüssigem Ammoniak oder mit
Chlorwasserstoff oder Bromwasserstoff oder durch kata- lytische Hydrierung.
Die Isolierung und die Reinigung des nach dem er findungsgemässen Verfahren erhaltenen Polypeptids wird vorteilhaft druch Chromatogrpahie an basischer
Tonerde oder an Ionenaustauschern oder druch Gegenstromverteilung ausgeführt.
Das erfindungsgemässe Verfahren kann beispiels- weise wie folgt ausgeführt werden :
Crabobenzoyx-L-alanyl-L-phenylanayl-L-isoleucin (hergestellt durch Kondensation des Carbobenozy- L phenylalanins mit L-Isoleucyl-emthylester und darauf folgende Abspaltung der Carbobenzoxygruppe, Konden- sation des erhaltenen Dipeptids mit Carbobenzoxy-L alanin) und Abspaltung der Methylgruppe wird mit Glycyl-L-leucyl-L-methoin-amid kondensiert (erhalten durch Kondensation des Carbobenzoy-glcycl-L-leuiins mitMethionin-methylester und durch Umsetzung des er haltenen Tripeptids mit gesättigter Ammoniaklösung in Methanol,
wobei das Carbobenzoxy-glycyl-L-leucyl- methioninamid erhalten wird, von dem die Carbobenz- oxygruppe entfernt wurde), wobei das Carbobenzoxy L-alanyl-L-phenylalanyl-L-isoleucy6cl-glyclyl-L-methioinamid entsteht, von dem durch Entfernung der Carbobenzoxygruppe das Hexapeptid gewonnen wird.
Das verfahrensgemäss hergestellte Hexapeptid besitzt eine starke peripher gefässerweiternde Wirkung, die besonders an Menschen und Hunden anchweisebar ist.
Beim Hund sind Dosen von 0, 001-0, 005 y/kg reines Polypeptid, bei intravenöser Verabreichung wirksam ; bei der Injektion in die femorale Arteie beobachtet man Gefässerweiterungen im Gelenk des Hundes bis zu Dosen von 0, 001 y. In der glatten isolierten Darmmuskulatur von Kaninchen, Hunden und.
Meerschweinchen ist das Hexapeptid bis zu Dosen von 0, 001-0, 002 # wirk sam. Das erfindungsgemäss hergestellte Polypeptid oder seine pharmazeutisch verwendbaren Salze können in der Therapie von hypertensiven Anfällen oder jedefalls bei dringender Behandlung von sehr schwerer Hypertension, bei krampfartigein) Gefässyndromen, besonders im Oberflächenmuskelberiech (Bürgersche Krakhiet, Reynaud- sche Krankheit, Ulcera torpida usw.), der Netzhaut gefässe (spastische Blindheit durch zentrale Spasmen der Netzhaut),
der Himhautgefässe (spastische Kopf schmerzen und Migräne) und der Coronargeifässe (Anginaanfälle) angewedet werden.
Das erwähnete Polyppepteid kann subkautan, intramus- kulär, intravenös (Injektion oder INfusion) verabreicht wenden.
Die geeignetsten Lösungsmittel sind Wasser oder physiologische Salzlösung.
Bei subkutaner oder intramuskulöser Verabreichung können sie mit absorpitinsverzögernden Substazen gemischt werden. Der Prozentgehalt an aktiven Bestand- teilen kann sich nach den besonderen pharmazeutischen Formen und nach der gewünschten hypotensiven Wir kung ändern, doch ist er gewöhnlich sehr niedrig,
Beim Menschen kann die tödliche Dosis des wirksamen Bestandteiles von 0, 005-5 mg betragen.
Das Polypeptid besitzt weder akute noch chronische Toxizität.
Ausgangsstoffe können wie folgt hergestellt werden :
1 a) Carbobenzoxy-L-phenylalanyl-L-isoleucin- methylester
29, 9 g Carbobenzoxy-L-phenylalanin (Ber. 91, 1958, S. 462) und 16 g L-Isoleucinmethylester (Helv. Chim.
Acta 38, 1955, S. 1500) werden in 250 ml wasserfreiem Tetrahydrofuran gelöst, Dann werden unter Rühren und Kühlen 24, 7 g Dizyklohexylcarbodiimidy hinzugefügt.
Die Mischung wird 16 Stunden bei Raumtemperatur gerührt. Nach Abfiltrieren des Dizyklohexylharmstoffes wird das Lösungsmittel im Vakuum eingedampft, der Rückstand wird in 500 ml Äthylazetat gelöst und mit
In-Salzsäure, Natriumbicaronat (1 m) und schliesslich mit Wasser extrahiert. Die Lösung wird über Natrium sulfat getrocknet, und dann wicd das Lösungsmittel ab gedampft. Der Rückstand wrid aus Äthylacetat-Petrol äther kristallisiert.
Ausbaute 36 g ; Fp = 106 bis 107 C ; [α]D30 = -10, 5 (c = 2 in Äthanol). b) L-Phenylalnyl-L-isoleucin-methylster chlorhydrat
Zu einer Lösung von 21, 3 g Carbobenzoxy-L-phenyl alanyl-L-isoleucin-methylester in 250 ml Methanol werden 8 g 10%iges Palladium auf Kohle zugefühgt, Die Mischung wird 5 Stunden bei Atmosphärendruck hydriert.
Nach Abfiltrieren des Katalysators und Abdampfen des Lösungsmittels wird der Rückstand in wasserfreiem Äther gelöst, die so erhaltene Lösung mit Chlorwasser- stoff angesäurert und daraufhin zur Trockne eingedampft.
Ausbaute 15 g ; Fp = 74 bis 76 C; [α]D28 = +6,25 (c = 2 in Äthanol). c) Carbobenzoxy-L-alanyl-L-phenylalanyl-L isoleucinmethylester 11, 1 g Carbobenzoyx-L-alainin (J. Biol. Chem. 124, 1938, S. 702) und 15 g L-'Phenylalanyl-'L-isoleucin- mefhylester (freie Base) werden in 150 ml Tetrahydro- furan gelöst. und unter Kühlung und Rühren werden 13, 3 g Dizyklohexylcarbodiimid eingetragen ; nach 16 Stunden Stehen bei Raumtemperatur wird die Mischung filtriert, das Lösungsmittel abdostilliert und der Rückstand in 400 ml Äthylazetat gelöst.
Die Lösung wird mit in Salzsäure, im Natriumbioarbonit. und Wasser gewa schen.
Die Lösung wird über wasserfreiem Natriumsulfat getriodknet ; das Lösungsmittel wird abdestilliert und der Rückstand aus Äthylacetat-Peytroläther umkristallisiert.
Ausbeute 21 g ; Fp = 154 bis 156 C ; [α]D28 = -44, 6 (c = 2, 5 in Äthanol). d) Carbobenzoxy-L-alanyl-L-phenylalanyl-L- isoleucin
Zu einer Lösung von 4,9 g Carbobenzoyx-L-alanyl L-phenyl-L-phenylalanyl-L-isoleucin-methylester in 90 ml Äthanol werden 20 ml in Natronlauge zugegeben.
Nach 90 Min. bei 20 C wird das Äthanol im Vakuum bei Raumtemperatur abdestilliert ; der Rückstand wird wieder mit Wasser aufgenommen, die erhaltene Lösung wird mit Salzsäure angesäuert, und der Niedersclag wird mit Äthylaoetat extrahiert. Nach Trocknen über wasserfreiem natriumsulfat wird das Lösungsmittel ab , destilliert.
Ausbeute 4, 4 g ; Fp = 183 bis 186 C (aus Äthyl- acetat).
2 a) Carbobenzoxy-glycyl-L-leucyl-methionin- methylester
10 g Carbobenzoxy-glycyl-L-leucin (J. Amer. Chem.
Soc. 78, 1956, S. 2130 und 5 g Methioninmethylester (Helv. Chim. Acta 34, 1951, S. 2091) werden in 90 mi Tetrahydrofuran gelöst. Darfaufinh werden 7, 2 g Dizyklophenylcarbodiimid eingetragen, und die Mischung wird 16 Stunden bei Ramtemperatur gerührt. Der Di zyklohexylharnstoff wird abfiltriert, das Läsungsmittel im Vakuum abdestilliert und der Rückstand in 250 ml Äthylazetat gelöst. Die Lösung wird mit In Salzsäure, mit 1 m Natriumbicanbonat und Wasser ausgeschüttelt.
Nach Trocknen über Natriumsulfat wird die Lösung im Vakuum zur Trockne eingeengt. Der ölige Rückstand wird mit Petroläther verrieben, wobei ein kristallines Produkt cntstcht, das aus Äthylacetat-Petroläther umkristallisiert wird.
Ausbeute 12 g ; Fp = 118 bis 119 C ; [α] D30 = -43, 1 (c = 3 in Äthanol). b) Carbobenzoxy-glycyl-L-leucyl-L-methioninamid
10 g Carbobenzoxy-glycyl-L-leucyl-L-methioninmethylester werden in 200 ml mit Ammoniak gesättig- tem wasserfreiem Methanol bei 0 C gelöst. Nach 4 Tagen wird die Lösung bei Raumtemperatur zur Trockne eingeengt, der Rückstand wird mehrmals mit Afhylazetat verrieben und getrocknet.
Ausbaute 8 g ; Fp = 177 bis 179 C (aus Äthylazetat) [α]D30 = -37,5 (c = 5 in Methanol). c) Glycyl-L-leucyl-L-methioninamid
Zu einer Losung von 5, 5 g Carbobenzoxy-glycyl-L- leucyl-L-methioninamid in 250 ml Methanol werden 3 g 10%iges Palladium auf Kohle zugegeben, und es wird d bei Atmosphärendruck hydriert. Nach 3 Stunden werden noch 3 g des Katalysators eingetragen, und die Mischung wird weitere 4 Studne der Hydrierung ausgesetzt. Nach Abfiltrieren des Katalysators wird die Lösung zur Trockene eingeengt, der Rückstand mit Was- ser verrieben und im Vakuum getrocknet.
Ausbeute 2g ; F p = 120 C.
3 a) N-Trityl-glycyl-L-leucin-methylester
In eins Lösung von 31, 7 g N-Trityl-glycin (J. A. C.
S. 78, 1956, S. 1359) und 15, 9 g L-Leuoin-methylester (Helv. Chim. Acta 29, 1946, S. 784) in 300 ml Tetrahydrofuran werden 22, 6 g Dizyklohexylcarbodiimid eingetragen. Das Gemisch wird über Nacht bei Raumtem- peratur gerührt, der Dizyklohexylhamstoff dann abfiltriert und das Lösungsmittel im Vakuum abdestilliert.
Der Rückstand wird mit 300 ml Äthylazetat gelöst, und die Lösung wird mti In Salzsäure (bei 0 C), In Ammoniak und Wasser gewaschen. Nach Trocknen über Natriumsulfat wird das Lösungsmittel im Vakuum abdestilliert.
Ausbeute 35 g eines sirupartigen Produkts. b) N-Trityl-glycyl-L-leucin
35 g N-Trityl-glycyl-L-leuchin-methylester werden in 90 ml In alkoholischer Kalilauge und 50 ml Äthanol erwärmt. Die Lösung wird 11/2 Stunden bei Raumtemperatur stehengelassen, dann mit 300 ml Wasser versetzt und filtriert. Das klare Filtrat wird abgekühlt, mit Essigsäure an, gesäuert, der ausgefallene Niederschlag wird abfiltriert und mit Wasser gewaschen.
Ausbetue 20 g ; Fp = 75 bis 85 C. c) L-Methioninmid
Eine Lösung von 38, 5 g L-Methinin-methylester (Helv. Chim. Acta 34, 1951, S. 2091) in 250 ml was serfreiem Methanol wird bei 5 C mit gasförmigem Ammoniak gesättigt. Nach 4 Tagen Stehenlassen bei +5 C wird, die Lösung im Vakuum zur Trockene eingedampft und der Rückstand einiger Male mit wasserfreiem Athyl- äther verrieben.
Ausbeute 33 g ; Fp = 50 bis 51 C ; [αD17 = 2, 0 (c = 5 in Äthanol). d) N-Trityl-glycyl-L-leucyl-L-methioninamid
In einer Lösung von 17, 2 g N-Trityl-glycyl-L-leucin und 6, 0 g L-Methioninamid in 180 ml Methylenchlorid werden 8, 6 g Dizyklocarbodiimid unter Kühlung eingetragen. Nach Stehenlassen über Nacht bei Raumtem- peratur wird das Reaktionsgemisch filtriert und das Filtrat mit ln Salzsäure (bei 0 C), 1 m Natriumbicarbonat und Wasser gewaschen. Nach Trocknen über Natriumsulfat wird das Lösungsmittel im Vakuum abdestilliert und der Rückstand aus Äthylazetat umkristallisiert.
Ausbeute 15 g ; Fp = 2 ibis 216'C ; [a] D21 = -20 (c = 1, 6 in Äthanol). e) Glycyl-L-leucyl-methioninamid
Eine Lösung von 9, 5 g N-Trityl-glycyl-L-leucylmethionmamid in 50 ml Essigsäure wird mit 50 ml Wasser versetzt und 30 Min. am Wasserbad erhitzt. Nach Zufügen von weiteren 50 ml Wasser wird die Mischung auf 0 C abgekühlt und das Triphenyl-carbinol abfil- triert. Die so erhaltene klare Lösung wird im Vakuum zur Trockene konzentriert.
Der Rückstand wird aus Athanol umkristallisiert ; Fp = 134 bis 136 C. Die wässrige Lösung von Tripeptid-azetat wird an einer Chromatographiekolonne mit Ionenaustauscherharz Dowex 2 (eingetragene Schutzmarke) in basischer Form chromatograpiert, und das Eluat wird im Vakuum zur Trockene konzentriert.
Ausbeute 4, 6 g ; Fp = 156 bis 158 C ; [α]D21 = -50 (c = 2 in Wasser).
Beispiel Carbobenzoxy-L-alanyl-L-phenylalanyl-L-isoleucyl- glycyl-L-leucyl-L-methioninamid
Zu einer Lösung von 2, 7 g Carbobenzoxy-L-alanyl- L-phenylalanyl-L-isoleucin und 1, 8 g Glycyl-L-leucyl- L-methioinamid in 50 ml Dimethylform. amid werden 1, 4 g Dizykolhenxylcarbodiimid eingetragen und die Mischung 20 Stunden bei Raumtemperatur gerührt.
Nach Filtrieren (1, 5 g Dizyklohexylharnstoff) wird die Lö sung im Vakuum zur Trockene eingeengt ; der Rück- stand wird mit wasserfreiem Äther verrieben, der Nie derschlag wird wieder in der geringsten Menge an Di methylformamid gelöst und wiedemun mit Äther ausgefällt.
Ausbeute 3 g ; Fp = 235 bis 238 C (aus Äthanol).
L-Alanyl-L-phenylalanyl-L-isoleucyl-glycyl-L leucyl-L-methioninamid-bromhydrat
Zu 1, 4 ml einer mit Bromwasserstoff gesättigten Essigsäurelösung werden 1, 1 ml Diäthylphosphit und 0, 5 ml Essigsäure hinzugefügt. In dieser Mischung werden 0, 4 g Carbobenzoy-L-alanyl-L-phenylanayl-L-iso leucyl-glycyl-L-leucyl-L-methioninamid gelöst. Nach 75 Min. bei 20 C wird die Lösung mit 50 ml wasserfreiem Äther versetzt. Der. ausgefallene Niederschlag wird mehrmals mit Äther gewaschen und im Vakuum über Kaliumhydroxyd und Phosphoipentoxyd getrocknet.
Ausbeute 0, 36 g ; Fp = 250 bis 255 C ; [a] D20 = -20+4 (c = 1 in Essigsäure).

The cyclohexylurea is filtered off, the solvent is distilled off in vacuo and the residue is dissolved in 250 ml of ethyl acetate. The solution is extracted with 1N hydrochloric acid, 1M sodium bicarbonate and water.
After drying over sodium sulfate, the solution is concentrated to dryness in vacuo. The oily residue is triturated with petroleum ether, resulting in a crystalline product which is recrystallized from ethyl acetate-petroleum ether.
Yield 12 g; M.p. 118 to 119 C; [α] D30 = -43.1 (c = 3 in ethanol). b) carbobenzoxy-glycyl-L-leucyl-L-methionine amide
10 g of carbobenzoxy-glycyl-L-leucyl-L-methionine methyl ester are dissolved in 200 ml of anhydrous methanol saturated with ammonia at 0.degree. After 4 days the solution is evaporated to dryness at room temperature, the residue is triturated several times with ethyl acetate and dried.
Expansion 8 g; Mp = 177 to 179 C (from ethyl acetate) [α] D30 = -37.5 (c = 5 in methanol). c) Glycyl-L-leucyl-L-methionine amide
3 g of 10% palladium on carbon are added to a solution of 5.5 g of carbobenzoxy-glycyl-L-leucyl-L-methionine amide in 250 ml of methanol, and the mixture is hydrogenated at atmospheric pressure. After 3 hours, 3 g of the catalyst are introduced and the mixture is subjected to a further 4 hours of hydrogenation. After the catalyst has been filtered off, the solution is concentrated to dryness, the residue is triturated with water and dried in vacuo.
Yield 2g; F p = 120 C.
3 a) N-trityl-glycyl-L-leucine methyl ester
In a solution of 31.7 g of N-trityl-glycine (J. A. C.
P. 78, 1956, p. 1359) and 15.9 g of L-leuoin methyl ester (Helv. Chim. Acta 29, 1946, p. 784) in 300 ml of tetrahydrofuran are added 22.6 g of dicyclohexylcarbodiimide. The mixture is stirred overnight at room temperature, the dicyclohexylurea is then filtered off and the solvent is distilled off in vacuo.
The residue is dissolved with 300 ml of ethyl acetate, and the solution is washed with in hydrochloric acid (at 0 C), in ammonia and water. After drying over sodium sulfate, the solvent is distilled off in vacuo.
Yield 35 g of a syrupy product. b) N-trityl-glycyl-L-leucine
35 g of N-trityl-glycyl-L-leuchin-methyl ester are heated in 90 ml of alcoholic potassium hydroxide solution and 50 ml of ethanol. The solution is left to stand for 11/2 hours at room temperature, then 300 ml of water are added and the mixture is filtered. The clear filtrate is cooled, acidified with acetic acid, the deposited precipitate is filtered off and washed with water.
Exercise 20 g; Mp = 75 to 85 C. c) L-methionine amide
A solution of 38.5 g of L-methinine methyl ester (Helv. Chim. Acta 34, 1951, p. 2091) in 250 ml of water-free methanol is saturated at 5 C with gaseous ammonia. After standing for 4 days at +5 C, the solution is evaporated to dryness in vacuo and the residue is triturated a few times with anhydrous ethyl ether.
Yield 33 g; M.p. 50 to 51 C; [αD17 = 2.0 (c = 5 in ethanol). d) N-trityl-glycyl-L-leucyl-L-methionine amide
In a solution of 17.2 g of N-trityl-glycyl-L-leucine and 6.0 g of L-methionine amide in 180 ml of methylene chloride, 8.6 g of dicyclocarbodiimide are introduced with cooling. After leaving to stand overnight at room temperature, the reaction mixture is filtered and the filtrate is washed with 1N hydrochloric acid (at 0 C), 1M sodium bicarbonate and water. After drying over sodium sulfate, the solvent is distilled off in vacuo and the residue is recrystallized from ethyl acetate.
Yield 15 g; Mp = 2 to 216'C; [a] D21 = -20 (c = 1.6 in ethanol). e) Glycyl-L-leucyl-methionine amide
A solution of 9.5 g of N-trityl-glycyl-L-leucylmethionmamide in 50 ml of acetic acid is mixed with 50 ml of water and heated for 30 minutes on a water bath. After adding a further 50 ml of water, the mixture is cooled to 0 ° C. and the triphenylcarbinol is filtered off. The clear solution thus obtained is concentrated to dryness in vacuo.
The residue is recrystallized from ethanol; Mp = 134 to 136 C. The aqueous solution of tripeptide acetate is chromatographed in basic form on a chromatography column with ion exchange resin Dowex 2 (registered trademark), and the eluate is concentrated to dryness in vacuo.
Yield 4.6 g; M.p. 156 to 158 C; [α] D21 = -50 (c = 2 in water).
Example carbobenzoxy-L-alanyl-L-phenylalanyl-L-isoleucyl-glycyl-L-leucyl-L-methionine amide
To a solution of 2.7 g of carbobenzoxy-L-alanyl-L-phenylalanyl-L-isoleucine and 1.8 g of glycyl-L-leucyl-L-methioinamide in 50 ml of dimethyl form. amide, 1.4 g of dicykolhenxylcarbodiimide are added and the mixture is stirred at room temperature for 20 hours.
After filtering (1.5 g of dicyclohexylurea) the solution is concentrated to dryness in vacuo; the residue is rubbed with anhydrous ether, the precipitate is again dissolved in the smallest amount of dimethylformamide and again precipitated with ether.
Yield 3 g; Mp = 235-238 ° C (from ethanol).
L-Alanyl-L-phenylalanyl-L-isoleucyl-glycyl-L-leucyl-L-methionine amide bromohydrate
1.1 ml of diethyl phosphite and 0.5 ml of acetic acid are added to 1.4 ml of an acetic acid solution saturated with hydrogen bromide. In this mixture, 0.4 g of carbobenzoy-L-alanyl-L-phenylanayl-L-isoleucyl-glycyl-L-leucyl-L-methionine amide are dissolved. After 75 minutes at 20 ° C., 50 ml of anhydrous ether are added to the solution. Of the. the precipitate is washed several times with ether and dried in vacuo over potassium hydroxide and phosphoipentoxide.
Yield 0.36 g; M.p. 250-255 C; [a] D20 = -20 + 4 (c = 1 in acetic acid).
