Neues Verfahren zur Herstellung von 1-Dehyäro-3-oxo-11-hyäroxy-steroiden Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von 1-Dehydro-3-oxo-11-hydroxy- steroiden, insbesondere von 1-Dehydro-3-oxo-lla- hydroxy-steroiden, womit ein neuer Weg zu den in therapeutischer Hinsicht wichtigen 1-Dehydro-corti- coiden, wie Prednison, Prednisolon, 1-Dehydro-Aldo- steron, Dexamethason,
oder 1-Dehydro-testosteron- derivaten, wie dem anabol wirksamen 1-Dehydro- 17a-methyl-testosteron gebahnt ist.
Das Verfahren der vorliegenden Erfindung be steht darin, dass man auf 3-Oxo-1,11-oxido-steroide basische Mittel einwirken lässt.
Die Aufspaltung des Ätherringes und Einfüh- rung der Doppelbindung erfolgt unter basischen Be dingungen, z. B. schon bei der Behandlung mit Al kalimetallsalzen von aliphatischen Carbonsäuren, wie Natriumacetat, Kaliumacetat, Kaliumpropionat usw. Im übrigen können auch Alkalimetallalkoho- late, -bicarbonate, -carbonate und -hydroxyde, z. B.
Natrium-methylat, Kalium-isopropylat, Kalium-ter- tiär-butylat, Natriumbicarbonat, Kaliumcarbonat, Natriumhydroxyd usw., aber auch Erdalkalimetall- hydroxyde, wie Bariumhydroxyd, Calciumhydroxyd, verwendet werden. Ferner können Aluminium-alko- holate, z.
B. Aluminium-isopropylat oder -tertiär- butylat oder tertiäre aliphatische oder cycloaliphati- sche Amine, wie Triäthylamin, N-Methylpiperidin, N-Methyl-morpholin usw. verwendet werden. Die Spaltung gelingt auch beim Chromatographieren an Aluminiumoxyd. Die oben genannten Basen können in geeigneten Lösungsmitteln, in denen sowohl die Oxidoverbindung wie auch die Base löslich sind, verwendet werden, z.
B. in niedrigen aliphatischen Alkoholen, cyclischen Äthern oder aromatischen Kohlenwasserstoffen oder Gemischen derselben.
Als Ausgangsstoffe für das vorliegende Ver fahren eignen sich 3-Oxo-1,11-oxido-, insbesondere 3-Oxo-1 a,11 a-oxido-verbindungen der Cholestan-, Koprostan-, Spirostan-, Cardanolid-, Furostan-, Cholan-, Pregnan- und Androstan-Reihe, die im Ringsystem und in der Seitenkette, insbesondere in einer oder mehreren der Stellungen 2, 4, 5, 6, 7, 8, 9, 10, 12, 14, 15, 16, 17, 18, 19, 20, 21, 22,
wei tere Substituenten aufweisen können, wie veresterte oder verätherte Hydroxylgruppen, freie oder insbe sondere funktionell abgewandelte, z. B. ketalisierte Oxogruppen, funktionell abgewandelte, z.
B. ketali- sierte Oxogruppen, funktionell abgewandelte Carb- oxylgruppen, wie veresterte oder laktonisierte Carb- oxylgruppen, Alkyl- wie Methylgruppen oder Halo genatome. In den Estern sind die Säurereste z. B.
solche von gesättigten oder ungesättigten aliphati- schen oder cycloaliphatischen, von aromatischen oder heterocyclischen Carbonsäuren, beispielsweise solche der Ameisensäure, Essigsäure, Propionsäure, der Buttersäuren, Valeriansäuren, wie n-Valeriansäure oder Trimethylessigsäure, der Capronsäuren, wie /3- Trimethylpropionsäure, der önanth-, Capryl-,
Pelar- gon-, Caprin-, Undecylsäuren, z. B. der Undecylen- säure, der Laurin-, Myristin-, Palmitin- oder Stearin säuren, z.
B. der ölsäure, der Cyclopentyl-, Cyclo- hexyl- oder Phenylessigsäuren oder -propionsäuren, der Benzoesäure, Phenoxy-alkansäuren, wie Phenoxy-essigsäure, p-Chlor-phenoxy-essigsäure, 2,4-Dichlor-phenoxy-essigsäure, 4-tert.-Butyl-phenoxy-essigsäure, 3-P'henoxy-propionsäure, 4-Phenoxy-buttersäure, der Furan-2-carbonsäure,
5-tert.-Butyl-furan-2-carbonsäure, 5-Brom-furan-2-carbonsäure, der Nicotinsäuren, ferner von Dicarbonsäuren, wie Oxal-, Bernstein- oder Glutarsäuren, von substi- tuierten Carbonsäuren, wie ss-Keto-carbonsäuren, z. B. der Acetessig-, Propionylessig-, Butyrylessig- oder Caprinoylessigsäure, von Aminosäuren usw.
Ausserdem können die Ausgangsstoffe auch Doppel bindung aufweisen, z. B. ausgehend vom Kohlen stoffatom 5.
Die zu verwendenden 3-Oxo-1,11-oxido-steroiden lassen sich nach dem in der schweizerischen Patent schrift Nr. 396 889 beschriebenen Verfahren her stellen. Dieses besteht darin, dass man auf lla-Hy- droxy-steroide, welche in 20-Stellung keine weitere freie Hydroxylgruppe aufweisen, oxydierend wir kende Metallacylate, insbesondere Bleitetraacylate, wie Bleitetraacetat,
einwirken lässt und gegebenen falls eine geschützte 3-Oxogruppe in die freie Oxo- gruppe überführt.
Die Verfahrensprodukte sind wertvolle Zwischen produkte zur Herstellung von hochwirksamen Ste- roiden.
Die Temperaturen sind im nachfolgenden Bei spiel in Celsiusgraden angegeben.
<I>Beispiel</I> 2 g 3,17-Dioxo-lla-hydroxy-androstan werden zu einer gerührten, 10! Minuten vorgekochten Suspension von 4 g Calciumcarbonat und 12 g Blei- IV-acetat in 300 ml Methylcyclohexan gegeben. An schliessend kocht man das Reaktionsgemisch eine Stunde am Rückfluss, nutscht die unlöslichen Teile ab und wäscht sie mit Essigester.
Man schüttelt das Filtrat mit einer wässrigen Lösung gleicher Teile Kaliumjodid und Natriumthiosulfat, dann mit Was ser, trocknet und dampft es im Vakuum ein. Die erhaltenen 2,26 g Rückstand werden an 40 g Silicagel (mit 5 % Wasserinhalt) chromatographiert. Aus den eingedampften Methylenchlorid-Eluaten lässt sich das 3,17-Dioxo-l,lla-oxido-androstan ge winnen, das sich aus Methylenchlorid-Äther-Gemisch umkristallisieren lässt.
Es schmilzt bei 175 , kristal- lisiert wieder und schmilzt dann bis 195 . Im IR- Spektrum zeigt es ein 5-Ring- und ein 6-Ring-Keton, aber keine Hydroxyl-Bande. [a] D = -f- 84 (in Chloroform).
Für die weitere Umsetzung wird der ganze rohe Rückstand der Methylenchlorid-Eluate der oben be schriebenen Chromatographie in 50 ml Methanol gelöst und mit einer Lösung von 2 g Kaliumcarbonat in 10 ml Wasser versetzt. Die Lösung kocht man 1 Stunde am Rückfluss, engt sie im Vakuum ein, schüt telt den Rückstand mit Essigester aus, wäscht die Essigester-Lösung mit Wasser, trocknet und dampft sie im Vakuum ein, wobei 1,6g Rückstand erhalten wird.
Diesen chromatographiert man an 27 g Silicagel (mit 5 % Wassergehalt) und eluiert zuerst mit Me- thylenehlorid, dann mit Methylenchlorid, das Aceton in wachsenden Prozenten enthält. Aus den Frak tionen mit 4 und 8 % Aceton erhält man beim Ein dampfen im Vakuum 770 mg Rückstand. Den Rückstand löst man in wenig Benzol und rührt die Lösung mit wenig Wasser, wobei sich Kristalle bil den, die abgenutscht und mit Benzol gewaschen wer den. Diese wasserhaltigen Kristalle werden in Äthanol gelöst.
Die Lösung dampft man im Vakuum ein und kristallisiert den Rückstand aus Methylenchlorid- Äther-Gemisch um, wobei 600 mg des Ai-3,17-Dioxo- 11u-hydroxy-androsten erhalten werden.
Es schmilzt bei 167-169 , kristallisiert dann wieder zu dicken Prismen, die endlich bei 184 schmelzen. Im IR Spektrum, zeigt es ein a,ss@-ungesättägtes Keton, ein 5-Ring-Keton, sowie eine Hydroxyl-Bande [a] D = -I- <B>68,1'</B> (in Chloroform).

The method of the present invention consists in allowing basic agents to act on 3-oxo-1,11-oxido-steroids.

The 3-oxo-1,11-oxido-steroids to be used can be produced according to the method described in Swiss Patent No. 396 889. This consists in that on Ila-hydroxy-steroids, which have no further free hydroxyl group in the 20-position, oxidizing metal acylates, in particular lead tetraacylates, such as lead tetraacetate,

The process products are valuable intermediate products for the manufacture of highly effective steroids.

<I> Example </I> 2 g of 3,17-dioxo-lla-hydroxy-androstane are stirred into a 10! A suspension of 4 g calcium carbonate and 12 g lead IV acetate in 300 ml methylcyclohexane, which has been pre-cooked for minutes, is added. The reaction mixture is then refluxed for one hour, the insoluble parts are filtered off with suction and washed with ethyl acetate.
The filtrate is shaken with an aqueous solution of equal parts of potassium iodide and sodium thiosulphate, then with water, dried and evaporated in vacuo. The 2.26 g residue obtained are chromatographed on 40 g silica gel (with 5% water content). The 3,17-dioxo-1,1a-oxido-androstane can be obtained from the evaporated methylene chloride eluate, which can be recrystallized from a methylene chloride-ether mixture.
It melts at 175, recrystallizes, and then melts until 195. In the IR spectrum it shows a 5-ring and a 6-ring ketone, but no hydroxyl band. [a] D = -f- 84 (in chloroform).
For the further reaction, the entire crude residue of the methylene chloride eluates from the chromatography described above is dissolved in 50 ml of methanol, and a solution of 2 g of potassium carbonate in 10 ml of water is added. The solution is refluxed for 1 hour, concentrated in vacuo, the residue is shaken out with ethyl acetate, the ethyl acetate solution is washed with water, dried and evaporated in vacuo to give 1.6 g of residue.
This is chromatographed on 27 g of silica gel (with 5% water content) and eluted first with methylene chloride, then with methylene chloride, which contains acetone in increasing percentages. From the fractions with 4 and 8% acetone, 770 mg of residue are obtained when evaporating in vacuo. The residue is dissolved in a little benzene and the solution is stirred with a little water, crystals forming which are suction filtered and washed with benzene. These water-containing crystals are dissolved in ethanol.
The solution is evaporated in vacuo and the residue is recrystallized from a methylene chloride-ether mixture, 600 mg of the al-3,17-dioxo-11u-hydroxy-androstene being obtained.
It melts at 167-169, then crystallizes again into thick prisms that finally melt at 184. In the IR spectrum, it shows an a, ss @ -unsaturated ketone, a 5-ring ketone, and a hydroxyl band [a] D = -I- <B> 68.1 '</B> (in chloroform) .
