Verfahren zur Herstellung von lokalanästhetisch wirksamen Toluididen
Die Erfindung betrifft ein neues Verfahren zur Herstellung von lokalan sthetisch wirksamen Tolui diden der Formel

worin R eine Alkylgruppa mit 2 bis 4, vorzugsweise 2 oder 3 Kohlenstoffatommen bedeutet, insbesondere (α-Monoalkylaminopropionyl)-o-toluidinen, in denen die Alkylgruppe Athyl, n-Propyl oder Isopropyl ist und auch Isobutyl-[-CH2-CH-(CH3)2] sein kann, aber vorzugsweise nicht n-Butyl ist, das mehr als 3 C-Atome in einer geraden Kette angejtagert an den Aminostickstoff bestizt.
Die Verbdingune dieser Formel zeigen einen mindestens so guten anästbetischen Effekt beim klinischan Gabmauch wie Lidocain, bei dem es sich um 2, 6-dimethylsubstituiertes Alkylaminoacylanilin, nämlich Diäthylaminoacetyl-2,6-xylidin, handelt, und sie sind völlig frai von Nebeneffekten bei , allure vernünftigen Dosierungen. In Anbetracht ihrer beträchtlich niedtiigeren Toxizität besitzen sie jedoch einen gunstigeren tiherapeutisohan Index als Lidocain. Nach der Erfindung wird daher eine-neue Gruppe von Lokalanästhetika für klinischen Gebrauch verfügbar gemacht.
Ein weiterer Vorteil liegt in der Tatsache, dass o-Toluidin gebraucht werden kann und diese Substanz im Handel in tausreichend Deiner Form für eine Arzneimittelsynthesfe leicht ver fugbar ist.
Der klinische Wert der vorliegenden Verblödun- gen kann auf Grund einiger pharmazeutischer Expe pimente, die durchgeführt worden sind, beuliteilt werden. Die folgende Tabelle enthält die Versuchsergeb- nisse mit verschiedenen Verbindungen gemäss der obigen Formel, in der R, die m d'er Tabelle unter Verbindung angegebene Bedeutung hat, während Lidocain als Standardbezugssubstanz angegeben ist.
Die Verbindungen sind in Form ihrer Hydrochloride geprüft wonden. Die Prüfungen wurden gemäss Standardmethoden der Technik ausgeführt. Toxizitätswerte wurden an weissen Mäuusen durch suboutane und intravenöse Injektionen vorgenommen ; die angegebenen Zahlen sind die Mengen der Verbindung in g/kg und mg/kg Körpergewicht derMäuse, bei der die letale Dosis für 50 % der geprüften Tiere erhalten wurde. Irritation wurde mittels suboutaner Injektion am Kaninchenobr und. an der Innenseite des mensch- lichen Unterarmes beobachtet und bezieht sich auf die Wirkung. an der Injektionsstelle. Der. anästheti- sche Effekt wunde durch subcutane Injektion an der Innenseite des menschlichen Unteranmes geprüft und die Latenzzeit (Lat.) sowie die Duration (Dur.) wurden beobachtet.

Ferner haben klinische Prüfungen mit der Ver tamdunig3,dienachdervorsteheindienTabeUe.an- scheinend besondens güstige Eigenschaften besitzt, gezeigt, dass diese Verbindung dem Lidocain eindeutig als Leitungaanästhetikum m überlegen ist.
Aus s diesen Untersuchungen ist ersichtlich, dass alle fünf f erfindungsgemäss hergestellten Verbindun- gen (2 bis 6) hinsichtlich des anästhetischen Latenzeffektes mindestens so gut wie Lidocain sind und unter Berücksichtigung der Fehlergrenzen alle mit Ausnahme der Isopropylverbindung eine ebenso lange Duration wie dieBezugsverbindung besitzen.
Am eindrucksvollsten ist, dass die Verbindungen 2 bis 4, in denen R eine Alkylgruppe mit 2 oder 3 C-Atomen ist, nicht nur Lidocain gleichwertig, sondern sogar be trächtlich weniger giftig sind als die Bezugssubstanz und auch bezüglich Irritation zumindest beim Menschen, worauf es bei klinischer Anwendung tatsäch- lich ankommt, Lidocain gogenüber nicht nur gleich- wertig sind. Es ist bemerkenswert, dass die n-Propylverbindung einen etwas geringeren irritierenden Effekt als Lidocain hat.
Auch ist es sehr über raschend, dass die Butylverbindungen (5 und 6) bezüglich Irritation am Menschen und bezüglich Giftigkeit dem Lidocain nahekommen. Kaninchenohren sind sehr empfindlich, und die relativ starke Irritation, die man für die Irritation am Kanin cbenohr mit der n-Butylverbindung (6) erhielt, kann bedeutungsvoll sein, da sie. anscheinend andeutet, dass die Methylengruppenkette nicht zu lang sein darf.
Da die Isabutylverbindung eindeutig basser als die n-Butylverbindung ist und dan Athyl-und Pro pylverbindunge. n nahekommt, würde die Schlussfol- gerung möglich erscheinen, dass Verbindungen der obigen, Formel, im denen die Alkylgcuppe R nicht mehr als 3 KohiLenstoffatome in einer geraden Kette angelagert an den Afminfostickstoff ausweist, eme Gruppe wertvollere anästhetischer Verbindungen bildet als solche mit längener gerader Kette.

Für die zur Entfernung aller Doppelbindungen des Restes (III) durchzuführende Hydrierung ist es in einigen Fällen nicht erfonderlich, den Ausgangsstoff der Fonnel (II) aus dem bei seiner Herstellung entstehenden Reaktionsgemisch zu isolieren.
Die bei der Herstellung dieser Lokalanästhetika gemäss dem Verfahren nach der Erfindung erhalte- nen Ausbeuten liegen zwischen 40 und 95 /o und zwar gewöhnlich in der Grössenordnung von 75 bis
90 /o. Abgesehen von der Tatsache, dass das Verfahren zu sehr guten Ausbeuten des betreffenden Endproduktes führt, zeigt sich auch der Vorteil, dass das Produkt in einer sehr reinen Form erhalten wird, da die Hauptverunreinigung meist ein Hydrierungs Katalysator ist, der sich leicht mittels Filtration entfernn lässt.
Beispiel 1
1 Mol Alanyl-o-toluidil und 1 Mol N-Propional dehyd ! werden in 21 Alkohol aufgelöst, worauf ein Brei bestehend aus Alkohol. un) d 10g Katalysator (Palladium auf Kohfle msit 10 /o Pd) zugesetzt wind.
Die Reaktionsmischung wird darauf bei etwa 10 C unter normalem Druck hydriert, bis die theoretische Mené (l Mol) Wasserstofigas ; aufgenommen worden ist. Nach Erhitzen zum Siedepunkt wird die Lösung filtriert und das Filtrat dann im Vakuum einge- dampft. Der Rückstand wind in In-Salzsäure. aufge- löst und darauf durch Schütteln mit Äther. gewa- schen. Die so gereinigte Salzsäurelösung wird durch Zugabe von 2n-Natriumhy, dlroxyd alkalisch gemacht, worauf die freigesetzte Base in Äther aufgenommen wird. Wenn die Ätherlösung mit Na2SO4 getrocknet worden ist, wird. die Lösung abgedampft und der Rückstand bei vermindertem Druck destilliert.
Man erhält ein fasbloses Öl mit K. P. 159 bis 162 /0,1 mm. Die Ausbeute beträgt 50 %.
Beispiel 2
Eine 10"/o-ige Lösung von l Mol Brenztrauben- säure-o-toluiidid m 11 Alkohol, die auch 10g eines aufgeschlämmten Katalysators (Palladium auf Kohle mit 10"/oPd) enthält, wird mit l Mol n-Propylamin versetzt, worauf die Lösung bei Zimmertemperatur unter Rühren hydriert wird. Der Katalysator wird abgesaugt und die Lösung in einem Verdampfer zur Trockne eingedampft. Der Rückstand wird in Ather gelöst und die Base durch Zugabe von ätherischer Salzsäure ausgefällt. Nach einer Umkristallisation aus Alkoholäther erhält man farblose Nadeln. F. P. = 167 bis 168 C. Die Ausbeute beträgt 40 %.
Beispiel 3
1 Mol α-Allylamino-2-methyl-propioanilidhy- drochlorid (F. P. 168 bis 169 C, hergestellt durch Umsetzung von a-Brom-2-methylpropioanilLidí und- AIlylamin durch Erhitzen. auf 80 in Benzol) wird m 1, 51 1 Wesser gelöst und mit 10g Katalysator (Palla- dium auf Kohle mit 10 %Pd) versetzt, woran sich eine Hydrierung bei Zimmertemperatur schliesst, bis 1 Mol (theoretische Menge) Wasserstoff verbraucht worden ist. Nach anschliessender Abfiltrierung des Katalysators wind die Lösung alkalisch gemacht und die so freigesetzte Base in Äther. aufgenommen.
Wenn der laitier über Natriumsulfat getrocknet worden ist, wird das Lösungsmittel abgedampft und der Rückstand unter Vakuum destilliert. K. P. = 159 bis 162 C/0, 1 mm. Die Ausbeute beträgt 93 %.
Beispiel 4
1 Mol Alanyl-o-toluidin und 1 Mol Allyamin werdan in 21 Alkolhol (75 %-iger Alkohol) aufgelöst, worauf 10g Katalysator (Palladium auf Kohle mit 10"/oPd)zugegebenwenden..DieReaktiojnsmischung wird dann bei etwa 10 C'und normalem Druck hydriert, bis die theoretische Menge (2 Mol) Wasser stoffgas verbnaucht worden ist. Darauf wird die Reaktionsmischung, wie schon im Beispiel l beschrieben, behandelt, Die Ausbeute bet1^er¯ägt 55 %.
Die so bergestelltenVerbindungenkönnenals freie Bassen gebraucht werden, jedoch ist es im Hinblick auf ihre Löslichkeit und Retinheit zweckmässig, dass die erhaltenen Basen gemäss an sich bekannten Metbaden in Salze von physiologisch verträglichen organischen oder anorganischen Säuren, z. B. Salzsäure, Schwefelsäure, Phosphorsäure, Essigsäure, Zitronensäure, Weinsäure, Bernsteinsäure, Picrinsäure und natürlichen und halbsynthetischen Penicillinen umgeformt werden. Einige dieser Salze sind hygroskopisch und absorbieren in Berührung mit einer feuchten Atmosphäre Wasser in unterschiedli- chem Ausmass.
