Verfahren zur Herstellung neuer 3,6-Diogo-A-nor-B-homo-steroide Gegenstand der vorliegenden Erfindung ist ein besonderes Verfahren zur Herstellung von neuen 3,6- Dioxo-A-nor-B-homo-steroiden, ausgehend von 3- Oxo-4,5-oxido-steroiden.
Die genannten Verbindungen bilden eine neue Klasse von biologisch wirksamen Steroiden oder Zwi schenprodukten zu deren Darstellung. So weisen die 3,6 - Dioxo - A - nor - B - homo - androstane und<B>-AL</B> androstene, die in 17ss-Stellung eine freie oder ver- esterte Hydroxylgruppe besitzen, bei sehr geringer androgener Aktivität eine hohe anabole Wirksamkeit auf.
Die 3,6-Dioxo A-nor-B-homo-pregnan Verbinn dungen, insbesondere solche, die in 20- und ge gebenenfalls in 11-Stellung eine Oxogruppe, in 17- bzw. oder in 21- und 11j3-Stellung Hydroxylgruppen aufweisen, sind besonders als neuartige Verbindungen mit entzündungshemmender bzw. antiöstro@gener und progestativer Wirkung von grossem Interesse.
Die bis heute bekannten Steroide mit A-nor--B- homo-Gerüst, das 6-Oxo-A-nor-B-homo-cholestan und das 6-Oxo-17ss-hydroxy-5a- und -5ss-androstan und deren 17-Benzoate wurden durch basische Be handlung der entsprechenden 4a-Tosyloxy-5a-hy- droxy-Verbindungen hergestellt.
Es wurde nun gefunden, dass man zu 3,6-Dioxo- A-nor-Bzhomo-steroiden auf einem neuartigen und einfacheren Weg gelangen kann, wenn man 3-Oxo- 4,5-oxido-steroide mit ultraviolettem Licht bestrahlt.
Die gebildeten 3,6-Dioxo-A-nor-B homo-steroide können, wenn erwünscht, durch Behandeln mit acylie- renden Mitteln in ihre 6 Enolacylate übergeführt werden oder durch Umsetzen mit Hydrazinen in die cyclischen Hydrazonderivate umgewandelt werden.
Das vorliegende Verfahren wird durch das fol gende Partialformelschema, z. B. unter Verwendung von Ausgangsstoffen der Gruppierung I, veranschau licht.

R1 = H oder Acylrest; R = H oder CH3 Die als Ausgangsstoffe verwendeten in Stellung 1 und 2 gesättigten 3-Oxo-4a,5a- sowie -4ss,5ss-oxida- steroide können aus den entsprechenden @4-3-Oxo- Verbindungen in an sich bekannter Weise, z. B. durch Behandlung mit alkalischem Wasserstoffsuperoxyd oder Persäuren, oder aber durch Hydrierung der /\4-3-Oxo-4,5-oxido-steroide, hergestellt werden.
Die letztgenannten Ausgangsstoffe sind neu; sie können überraschenderweise direkt aus den L\.1,'-3-Oxo- steroiddienen durch Behandlung mit Persäuren, z. B. mit organischen Persäuren, wie niederaliphatischen und aromatischen Persäuren, z.
B. mit %,essigsäure, Benzopersäure und Phthalmonopersäure oder aber durch Dehydrierung der oben genannten 1,2-gesättig- ten 3-Oxo-4,5-oxido-.steroide, z. B. mit Selendioxyd oder Dicyano-dichlor-chinon erhalten werden.
Die verfahrensmässige Bestrahlung der 3-Oxo- 4,5-oxido-steroide wird vorteilhaft in organischen Lösungsmitteln, z. B. in aliphatischen und cycloali- phatischen Kohlenwasserstoffen wie Pentan, Hexan., Cyclohexan und Methylcyclohexan ausgeführt.
Be sonders geeignet sind aliphatische und cyclische Äther wie z. B. Diäthyläther und Dioxan.
Als Lichtquelle eignet sich künstliches oder star kes natürliches Licht; vorzugsweise verwendet man Ultraviolettlicht, wie es von Quecksilber-Niederdruck- und Hochdruckbrennern erzeugt wird, oder starkes Sonnenlicht. Die Bestrahlung erfolgt vorzugsweise bei Temperaturen zwischen 0 und +80 .
Die als Reaktionsprodukt anfallenden 3,6-Dioxo- A-nor-B-homo-steroide, die als ss-Diketoverbindun- gen zum überwiegenden Teile in Enolform vorliegen, können in an .sich bekannter Weise in ihre funktio nellen, von der Enolform sich ableitbaren Keton- Derivate übergeführt werden. So erhält man z. B. durch Acylierung, so z.
B. durch Behandlung mit Car- bonsäureanhydriden, wie Acetanhydrid oder Propion- säureanhydrid, die entsprechenden Enolester. Durch Umsetzen der Verfahrensprodukte z. B. mit Hydrazi- nen entstehen cyclische Hydrazone, die auch als Pyrazolderivate angesehen werden können..
Die als Ausgangsstoffe für das vorliegende Ver fahren verwendeten 3-Oxo-4a,5a- und -4ss,5ss-oxido- steroide gehören vorzugsweise den Reihen der Androstane, rPegnane, Cholane, Cholestane, Spiro- stane und Cardanolide,
sowie den entsprechenden 19-Nor-Reihen an und können zusätzlich zu den ge nannten Gruppierungen in einer oder in mehreren der Stellungen 7, 7a, 8, 9, 11, 12, 14, 15, 16, 17, 18, 19, 20 und 21 weitere Substituenten aufweisen wie Alkyl- (z.
B. Methyl)-Gruppen und/oder Halogen atome, freie oder funktionell abgewandelte Oxogrup- pen und freie, veresterte oder verätherte Hydroxyl- gruppen. Ausserdem können die Ausgangsstoffe Dop pelbindungen enthalten, insbesondere wie oben er wähnt, zwischen den Kohlenstoffatomen 1 und 2. Es können auch Gemische der beiden in 4,5-Stellung isomeren Epoxyde verwendet werden.
Besonders wichtige Ausgangsstoffe sind z. B. die folgenden Verbindungen: 3-Oxo-4a,5a-oxido- und -4ss,5ss-oxido-17-hydroxy-androstan und seine Ester, 3,17-Dioxo-4a,5,a-oxido- und -4ss,5ss-oxido-androstan und dessen 17-Äthylen-Ketal, 3-Oxo-4a,5a-oxido- und 4ss,5ss-oxido-17a-alkyl-, -17a-alkenyl- und -17a- alkinyl-17ss-hydroxy-androstane und ihre Ester, wie z.
B. das 3-Oxo-4a,5a-oxido- und -4ss,5ss-oxido-17ss- hydroxy-17a-methyl-, -17a-äthyl-, -17a-vinyl-, -17a- äthinyl- und -17u-allyl-androstan, 3,20-Dioxo-4a,5a- oxido- und -4ss,5ss-oxido-pregnan, 3,20 Dioxo-4a,5a- oxido- und -4ss,5ss-oxido-17a.-hydroxy-pregnan und ihre Ester, 3,20-Dioxo-4a,
5a-oxido- und -4ss,5ss- oxido-21-hydroxy-pregnan und deren Ester, 3,20- Dioxo-4a,5a-oxido- und -4ss,5ss-oxido-17a,21-di- hydroxy-pregnan, ihre Ester und. 17,20;
20,21-Bis- methylendioxy-Verbindungen, 3,20-Dioxo-4a, 5a- oxido- und -4ss,5ss-oxido-l lss,17a.,21-trihydroxy- pregnan, ihre Ester und 17,20,20,2l Bis-methylen- dioxy Verbindungen, ferner die in Stellung 1 und 2 ungesättigten Derivate der oben genannten Verbin dungen.
Die 17- oder 20-Oxo-Verbindungen werden vorzugsweise in Form ihrer 17- bzw. 20-Monoketale verwendet.
Die nach dem erfindungsgemässen Verfahren er hältlichen Produkte können sowohl in Form der 3,6- Dioxo-A-nor-B-homo-steroide als auch ihren tauto- meren Enolverbindungen anfallen. Man kann auch die entsprechenden, daraus ableitbaren Enolester organischer, insbesondere niederaliphatischer und aromatischer Carbon- und. Sulfosäuren, wie z.
B. der Essigsäure, Propionsäure, Buttersäure, Benzoesäure, p-Toluolsulfonsäure herstellen.
Besonders zu nennen sind die folgenden 3,6- Dioxo-A-nor-B-homo-androstane und -pregnane: 3,6, 17-Trioxo-A-nor-B-homo-androstan, 3,6 Dioxo-17ss- hydroxy-A-nor-B-homo-androstan und seine 17 Ester, 3,6-Dioxo-17ss-hydroxy-17a-alkyl-, -17a-alkenyl- und -17a-alkinyl-A-nor-B-homo-androstane, wie z.
B. das 3,6-Dioxo-17r3-hydroxy-17a-methyl-, -17a-äthyl-, -17a vinyl-, -17a-äthinyl-, -17a-allyl- und -17a-tri- fluorpropinyl-A-nor-B-homo-androstan und ihre 17- Ester, ferner 3,6,20-Trioxo-A-nor-B-homo-pregnan, 3,6,20-Trioxo-17a-acetoxy-pregnane, 3,6,11,20-Te- traoxo-17a,21-dihydroxy-A-nor-B-homo-pregnan und seine Ester, 3,6,20-Trioxo-11ss,17a,
21-Trihydroxy- A-nor-B homo-pregnan, und die entsprechenden in Stellung 1,2 ungesättigten Derivate der genannten Verbindungen, insbesondere das C#,1-3,6-Dioxo-17ss- hydroxy-A-nor-B-homo-androsten und seine Ester und die in 17a-Stellung durch gesättigte und unge sättigte Niederalkylreste substituierten Verbindungen wie z.B. das 01-3,6-Dioxo-17ss-hydroxy-17a-methyl-, -17a-vinyl und -17cs-äthinyl-A-nor-B-homo-androsten und ihre Ester, wie auch das A1-3,6,
20-Trioxo-A- nor-B-homo-pregnen, L,.1-3,6,20-Trioxo-17a-acyloxy- pregnen, G1-3,6,20-Trioxo-11ss,17a,21-trihydroxy-A- nor-B-homo-pregnen und seine Ester; ferner die ent sprechenden Enolester.
In den oben genannten Estern sind die Säure reste, insbesondere solche von aliphatischen, cyclo- aliphatischen, araliphatischen, aromatischen und heterocyclischen Carbonsäuren mit 1-15 Kohlenstoff atomen, z.
B. Formiate, Acetate, Propionate, Buty- ratei, Trimethylacetate, Capronate, Valerianate, Ön- anthate, Decanoate, Cyclopentylpropionate, Hexa- hydrobenzoate, Phenylpropionate, Benzoate, Furoate,, Trifluoracetate, Äthyl- und Methylcarbonate usw.
In den folgenden Beispielen sind die Temperatu ren in Celsiusgraden angegeben. Die UV.-Absorp- tionsspektren sind in Äthanol-Lösung aufgenommen.
Die UV.-Bestrahlungen werden unter Stickstoff atmosphäre in einem Quarzgefäss ausgeführt, in wel chem die Lichtquelle zentral angeordnet und mit Wasser gekühlt ist. Als Lichtquellen dienen Nieder- druckrbrenner NK 6/20 (monochromatisches Licht der Wellenlänge 254 m,y) und Hochdruckbrenner Q 81 (beideQuarzlampen GmbH. Hanau).

Die abgekühlte Lösung wird darauf im Vakuum zur Trockene eingedampft und die erhaltenen Kri stalle (300 mg), welche gemäss Dünnschichtchromato- gramm [Fliessmittel:
Benzol-Methanol-(10:1)] ein- heitlich sind, zweimal aus Methanol umkristallisiert. Man erhält das Pyrazolderivat von 3,6-Dioxo-17ss- hydroxy-A-nor-B-homo-androstan vom konstanten Smp. 152-l55 . [a]D = +57 (c = 0,52). IR.-Spek- trum (CHCIs): = 3630, 3640, 1635 cm-' (schwach).
UV-Spektrum AB,", = 227<I>mg,</I> log e = 3,74.
<I>Beispiel 9</I> 700 mg eines Gemisches der isomeren 3-Oxo-4ss, 5ss- und 4a,5a-oxido-17ss-hydroxy-17a-methyl-Al- androstene werden in 200 ml Dioxan wie in Beispiel 3 beschrieben während 46 Stunden bestrahlt. Die Lö sung wird darauf im Vakuum eingedampft. Zwei- maliges Umfällen des kristallinen Rohproduktes er gibt 458 mg A1-3,6-Dioxo-17a-methyl-17ss-hydroxy- A-nor-B-homo-androsten vom F. 126-128 . [ab = +79,8 (c = 0,49).
UV-Spektrum: lm", = 239 m,u (e <I>=</I> 6700). IR.-Spektrum: v <I>=</I> 3610, 1761, 1708 cm-1.
Das in diesem Beispiel verwendete Ausgangs material Iässt sich z. B. wie folgt erhalten: 9 g A1.4-3-Oxo-17a-methyl-17ss-hydroxy-andro- stadien werden in einer Lösung von Benzopersäure in 140 ml Chloroform (35 mg/ml) 4 Tage im Dunkeln bei Zimmertemperatur stehen gelassen. Die Re aktionslösung wird darauf mit Äther verdünnt und nacheinander mit wässriger Kaliumjodid- und Na- triumthiosulfatlösung geschüttelt.
Man erhält 9,1 g Rohprodukt, das an neutralem Aluminiumoxyd (Akt. 11I; 30fache Menge) chromatographiert wird.
Benzol und Benzol-Äther-(9:1)-Gemisch eluiert 1830 mg amorphes Material, das nochmals an neu tralem Aluminiumoxyd (Akt. III; 60fache Menge) chromatographiert wird.
Dabei erhält man mit Ben- zol-Äther-(9:1)-Gemisch 1015 mg kristallines Mate rial, das nach zweimaliger Kristallisation aus Aceton- Petroläther bei l65-167 schmilzt (800 mg). [a]D = 105,1 (c = 0,56). UV.-Spektrum: ),o" = 232 m,y (e = 9000). IR.-Spektrum: v = 3600, 1674, 1617 cm-1.
Es liegt ein Gemisch von 85 0/0 3-Oxo-4ss,5ss- oxido-17ss-hydroxy-17a-methyl-Al-androsten und 15 0!o 3-Oxo-4a-5a-oxido-17ss-hydroxy-17a-methyl- Al-androsten vor. Die Äther-Fraktionen aus beiden Chromatogrammen ergeben nach einmaliger Kristal lisation aus Aceton-Petroläther 5,3 g unverändertes Ausgangsmaterial zurück.
<I>Beispiel 10</I> Unter den in Beispiel 1 angegebenen Bedingungen wird aus der Bis-methylendioxyverbindung von Pred- nisolon: über die entsprechende, ebenfalls nach den Angaben des Beispiels 1 hergestellte 4a,5a- oder 4ss, 5ss-Oxidoverbindung die 17,20;20,21-Bis-methylen- dioxyverbindung von a1-3,6,20-Trioxo-llss,17a,21- trihyäroxy-A nor-B-bomo-pregnen erhalten.
Diese liefert nach Hydrolyse das freie L.1-3,6,20-Trioxo- l lss,17a,21-trihydroxy-A-nor-B-homo@pregnen.
In analoger Weise liefert die Bestrahlung des A'-3-Oxo-4a,5a- und 4ss,5ss-Oxido-17ss-acetoxy-19- nor-androstens das L1-3,6-Dioxo-17ss-acetoxy-A-nor@ B-homo-19-nor-androsten. <I>Beispiel 11</I> 1,5 g pl-3,20-Dioxo-4,5-oxido-17a-acetoxy- pregnen in 500 ml Dioxan werden 72 Stunden mit einem Niederdruckbrenner bestrahlt (Temperatur: 26 bis 29 ) und die Lösung darauf im Vakuum einge dampft.
Der Rückstand wird in 50 ml Alkohol ge löst, mit 150 ml Äther verdünnt und im Scheide trichter mit 10 ml eiskalter l0o/oiger Natronlauge, in 50 ml Wasser geschüttelt.
Nach erschöpfender Ex traktion mit Natronlauge werden die alkalischen Aus züge vereinigt und mit verd. Salzsäure angesäuert, wobei 850 mg rohes O1-3,6,20-Trioxo-17a-acetoxy- A-nor-B-homo-pregnen erhalten werden. Nach Kri stallisation aus Methylenchlorid-Äther schmilzt die reine Verbindung bei 198-199 , [a]D = +76 (c = 0,945). Im Dünnschichtchromatogramm (System: Benzol/Methanol 9:1) ist ein einziger Fleck ,sichtbar.
Der mit Natronlauge nicht reagierende Anteil be trägt 0,5 g und stellt unverändertes Ausgangsmaterial dar.
Der Ausgangsstoff lässt sich wie folgt herstellen: 10 g p4-3,20-Dioxo-17a-acetoxy-pregnen werden in 100 ml Methylenchlorid und 300 ml Methanol ge löst und bei 0 tropfenweise mit 60 ml 30o/oigem Wasserstoffperoxyd und 20 ml l0o/oiger Natronlauge versetzt und 27 Stunden bei 0 gerührt. Die Re aktionsmischung wird dann auf Eis-Wasser gegossen und mit Methylenchlorid und Äther extrahiert. Nach dem Neutralwaschen,
Trocknen und Eindampfen im Vakuum erhält man 9 g rohes 3,20-Dioxo-17a-ace- toxy-4,5-oxido-pregnan, das ohne weitere Reinigung für den nächsten Schritt verwendet wird.
5,16 g rohes 3,20-Dioxo-17a-acetoxy-4,5-oxido- pregnan werden in 75 ml tert. Amylalkohol gelöst. Nach Zugabe von 2,5 ml Eisessig und 0,5g Queck silber wird. zum Sieden erhitzt und unter gutem Rüh ren eine Lösung von 2,5 g Selendioxyd in 32 ml tert. Amylalkohol zugetropft. Nach 14 Stunden wird ab gekühlt, über eine Schicht Norit abgenutscht und das Filtrat im Vakuum eingedampft.
Der Rückstand wird in Essigester aufgenommen und nacheinander mit l0o/oiger Kaliumjodlösung, 20o/oiger Natrium- thiosulfatlösung, 10o,'oiger Sololösung und Wasser ge waschen.
Nach dem Eindampfen des Lösungsmittels erhält man 5,78 g rohes A1-3,20-Dioxo-4,5-oxido- 17a-acetoxy-pregnen, das nach Filtration über Alu- miniumoxyd (Aktivität II) und Kristallisation aus Methylenchlorid-Äther bei 203-212 schmilzt. <I>Beispiel 12</I> 4,15 g A1-3-Oxo-4,
5-oxido-17a-methyl-17ss- acetoxy-androsten in 500 ml Dioxan werden 72 Stun den mit einem Niederdruckbrenner bestrahlt (Tempe ratur: 26-30 ) und nach den Angaben in Beispiel 11 aufgearbeitet.
Der natronlaugelösliche Anteil beträgt 3,63 g und stellt das /\1-3,6-Dioxo-17a-methyl-17ss- acetoxy A-nor-B-homo-androsten dar, das nach Kri stallisation aus Methylenchlorid-,Äther bei 206-210 schmilzt.
Der für die Bestrahlung benützte Ausgangsstoff lässt sich wie folgt herstellen: p4-3-Oxo-17a-methyl-17ss-acetoxy- androsten wird nach den Angaben in Beispiel 11 mit alkalischem Wasserstoffperoxydzum3-Oxo-4.,5-oxido-17a-methyl- 17ss-acetoxy-androstan (F.
= 119-121', aus Aceton- Hexan) oxydiert und anschliessend mittels Selen dioxyd zum @1-3-Oxo-4,5-oxido-17a-methyl-17ss- acetoxy-androsten dehydriert. <I>Beispiel 13</I> 3 g Al-3-Oxo-4,5-oxido-20ss-acetoxy-pregnen in 500 ml Dioxan werden 70 Stunden mit einem Niederdxuckbrenner bestrahlt (Temperatur: 27-30 ) und nach den Angaben von Beispiel 11 aufgearbeitet.
Der natronlaugelösliche Anteil liefert 2,16 g p1-3,6 Dioxo-20ss-acetoxy A-nor-B-homo-pregnen, das nach Kristallisation aus Methylenchlorid-Äther bei 161 bis <B>163'</B> schmilzt. [ab = +134' in Chloroform (c = 0,511). Daneben werden noch 550 mg Ausgangs- material zurückgewonnen.
Der obige Ausgangsstoff lässt sich wie folgt ge winnen: Das A'-3-Oxo-20ss-hydroxy-pregnen vom F. 167 bis 170 wird nach den Angaben in Beispiel 11 mit alkalischem Wasserstoffperoxyd oxydiert, das er- haltene Rohprodukt mit Acetanhydrid-Pyridin behan delt und das 3-Oxo-4,5-oxido-20ss-acetoxy-pregnan mittels Selendioxyd zum A1-3-Oxo-4,5-oxido-20ss- acetoxy-pregnen dehydriert. <I>Beispiel 14</I> 1,
5g Lakton des A1-3-Oxo-4,5-oxido-17ss- hydroxy-17a-(ss-carboxy-äthyl)-androstens in 500 ml Dioxan werden 72 Stunden mit einem Niederdruck- brenner (Temperatur: 26-29 ) bestrahlt, die Lösung darauf im Vakuum eingedampft und wie in Bei spiel 11 aufgearbeitet.
Der Natronlauge-Auszug stellt das Lakton des O1-3,6-Dioxo-17ss-hydroxy-17a-(ss- carboxy-äthyl)-A-nor-B-homo-androstens dar; es schmilzt nach Kristallisation aus Methylenchlorid- Äther bei 185-186 , [a]D = +85 .
Der Ausgangsstoff lässt sich aus dem Lakton des A4-3-Oxo-17ss-hydroxy-17a-(ss-carboxyäthyl)- androsten durch Oxydation mit Wasserstoffperoxyd und anschliessend Dehydrierung mittels Selendioxyd gemäss den Angaben in; Beispiel 11 gewinnen.

Die Ausgangsstoffe gewinnt man ausgehend von A4 3,20-Dioxo-17a-capronyloxy-pregnen bzw. A4- 3,20-Dioxo-17a-methyl-pregnen durch Oxydation mittels Wasserstoffperoxyd, gefolgt von Selendioxyd- Dehydrierung gemäss den Angaben in Beispiel 11.
<I>Beispiel 17</I> 1,5 g pl-3-Oxo-4,5-oxido-7a-methyl-17ss-acet- oxy androsten in 500 nal Dioxan werden wie in Bei spiel 11 beschrieben bestrahlt, wobei das A1-3,6- Dioxo-7a-methyl-17,pracetoxy A-nor-B-homo-andro- sten erhalten wird.
In analoger Weise wird das Al-3-Oxo-4,5-oxido- 7a,17a dimethyl-17ss-hydroxy-androsten in das Al- 3,6-Dioxo-7a,17a-dimethyl-17,ss-hydroxy A-nor-B- homo-androsten überführt.
Das al-3-Oxo-4,5-oxido-7a-tnethyl-17ss-acetoxy- androsten und das Al-3-Oxo-4,5-oxido-7a,17a- dimethyl-17ss-hydroxy-androsten gewinnt man bei spielsweise durch Umsetzung des A1,4-3-Oxo-7a- methyl-17f-acetoxy-androstadiens bzw. al>4-3-Oxo- 7a,17a -dimethyl-17ss -hydroxy- androstadiens mit Benzopersäure gemäss den Angaben in Beispiel 1.
<I>Beispiel 18</I> 1,5g A1-3,20-Dioxo-4,5-oxido-llss,17a-di- hydroxy-21-acetoxy-pregnen werden nach den An gaben von Beispiel 11 bestrahlt und aufgearbeitet, wo bei das A1-3,6,20-Trioxo-Ilss,17a-dihydroxy-21- acetoxy A-nor B homo-pregnen und das durch teil- weise Hydrolyse während der Natronlauge-Extraktion daraus entstandene A1-3,6,20-Trioxo-Ilss,17a,21-tri- hydroxy-A-nor-B-homo-pregnen erhalten werden.
Den oben verwendeten Ausgangsstoff gewinnt man aus A1,4-3,20-Dioxo-11ss,17a-idihydroxy-21- acetoxy-pregnadien durch Umsetzung mit Benzoper säure gemäss den Angaben in Beispiel<B>1</B> .
<I>Beispiel 19</I> 1,8g des 17,20;20,21-Bis-methylendioxyderivats von Prednisolon werden in einer Lösung vonBenzoper- säure in 100 ml Chloroform (45 mg/ml) 7 Tage im Dunkeln bei Zimmertemperatur stehen gelassen. Die Reaktionslösung wird darauf mit Äther verdünnt und nacheinander mit wässriger Kaliumjodid- und Natriumthiosulfatlösung, Wasser, Natriumhydrogen carbonatlösung und nochmals mit Wasser gewaschen.
Nach dem Eindampfen werden 1,4 g eines Gemisches enthaltend das 17,20;20,21-Bismethylendioxyderivat von 4a,5a- und 4ss,5ss-Oxido-prednisolon erhalten.
1,4 g des obigen Gemisches werden in 170 ml Dioxan während 14 Stunden mit einer Niederdruck- Quecksilberlampe belichtet. Nach Aufarbeiten ge- mäss den Angaben in Beispiel 11 erhält man 420 mg des 17,20;
21,21-Bismethylendioxyderivats von A, 3,6,20-Trioxo-11ss,17a,21-trihydroxy-A-nor-B-homo- pregnen,das nach Filtration über Silicagel, Entfär- bung mit Tierkohle und Kristallisation aus Aceton- Petroläther bei 210-230 unter Zersetzung schmilzt; [a]D = -7 (c = 0,7). UV.-Spektrum: Am", = 236 bzw. 310 m,y (E <I>=</I> 8000 bzw. 6080).
Process for the production of new 3,6-diogo-A-nor-B-homosteroids The present invention relates to a special process for the production of new 3,6-dioxo-A-nor-B-homosteroids, starting from FIG - Oxo-4,5-oxido steroids.
The compounds mentioned form a new class of biologically active steroids or inter mediate products for their preparation. Thus, the 3,6-dioxo-A-nor-B-homo-androstane and <B> -AL </B> androstene, which have a free or esterified hydroxyl group in the 17ss position, have a very low androgenic activity high anabolic efficacy.
The 3,6-dioxo A-nor-B-homo-pregnane compounds, in particular those which have an oxo group in the 20- and possibly in the 11-position, and hydroxyl groups in the 17- or or 21- and 11j3-positions , are of particular interest as novel compounds with anti-inflammatory or anti-estrogen and progestative effects.
The steroids known to date with A-nor-B-homo-skeleton, 6-oxo-A-nor-B-homo-cholestane and 6-oxo-17ss-hydroxy-5a- and -5ss-androstane and their 17-Benzoates were prepared by basic treatment of the corresponding 4a-tosyloxy-5a-hydroxy compounds.
It has now been found that 3,6-dioxo-A-nor-Bzhomo-steroids can be obtained in a novel and simpler way if 3-oxo-4,5-oxido-steroids are irradiated with ultraviolet light.
The 3,6-dioxo-A-nor-B homosteroids formed can, if desired, be converted into their 6 enol acylates by treatment with acylating agents or converted into the cyclic hydrazone derivatives by reaction with hydrazines.
