Procédé de préparation de thiocarbanilides
L'invention est relative à des thiocarbanilides (ou
N, N'diaryl-thiourées) à propriétés anti-inflammatoires et antifongiques, renfermant dans leur molécule des groupements halogénés, notamment des groupements fluoro.

Suivant l'invention, de tels thiocarbanilides sont préparés en faisant réagir des isothiocyanates d'aryles appropriés avec des arylamines convenables selon les schémas suivants :

Ce nouveau produit de formule (II) se présente sous forme de fins cristaux incolores, fondant à 1480 C (en tube capillaire). Il est insoluble dans l'eau, peu soluble dans les alcools et les glycols, dans le diméthylformamide et dans l'acide acétique.
Il est soluble dans l'oléate d'éthyle, le myristate d'isopropyle et dans les excipients huileux pour pom- mades et onguents.
Ce produit de formule II peut être préparé par exemple suivant 1'un ou l'autre des procédés décrits aux exemples suivants.
Exemple 1
Le procédé de préparation de cet exemple est basé sur la condensation de l'isothiocyanate de p-fluorophényle avec la 3, 5-dichloroaniline selon l'équation :

On maintient dans une enceinte chaude Ó environ 50-55¯ C, pendant 3 heures, une solution de 50 cm3 d'Úthanol absolu, 16,2g (0,1 mole) de 3,5-dichloroaniline et 15,3 g (0,1 mole) d'isothiocyanate de p-fluorophÚnyle. On laisse reposer durant 10 heures, on essore et on recristallise.
Exemple 2
Le procédé de préparation de cet exemple est basé sur la condensation de l'isothiocyanate de 3, 5-dichlorophényle avec la p-fluoroaniline selon l'équation :

On maintent dans une enceinte chaude à environ 50 550 C, pendant 3 heures, une solution de 50cm3 d'éthanol absolu, 11, 1 g (0, 1 mole) de pfluoroaniline et 20, 4 g (0, 1 mole) d'isothiocyanate de 3, 5-dichlorophényle.
On laisse au repos pendant 6 heures, on essore et on recristallise dans de méthanol.

Activité antifongique in vitro
Les activités mycostatiques des thiocarbanilides halogénés (II), (III), (IV) et (V) ont été étudiées.
Aux tableaux I et II annexés, des résultats sont donnés en ce qui concerne l'activité du produit de formule (II), respectivement après 7 jours et après 15 jours, et ce pour diverses concentrations de ce produit et sur divers organismes.
Pour la bonne compréhension de ces tableaux, on considérera qu'un carré blanc correspond à 100 ouzo d'activité du produit (II) à la concentration correspondante pour l'organisme envisagé, un quart de carré blanc correspond à 25'o/o d'activité, etc. Réci proquement, un quart de carré hachuré correspond à Z5 10/o de croissance de l'organisme, etc.
Certaines activités in vitro des produits (III), (IV) et (V) vis-à-vis de trois organismes (Candida albicans, Microsporum canis et Histoplasma duboisi) sont données ci-dessous.

Etudes expérimentales in vivo
Des études expérimentales in vivo ont été réali- sées pour le produit (II) désigné ci-après par 951 .
A Pouvoir antifongique
Des cobayes étaient infectés expérimentalement avec du Microsporum canis. On prévoyait un traitement journalier des animaux par application de pommade au 951 (1"Vo de produit actif dans du Carbowax). Le traitement débutait le lendemain du jour où l'infection était reconnue et les lésions installées (en moyenne 8 jours après l'inoculation). Des obser vations régulières des sujets étaient faites : contrôle de l'évolution de l'infection (lumière de Wood, examens microscopiques et cultures) et des lésions (examen macroscopique).
Des expériences ont porté sur 7 cobayes traités au 951 et 9 animaux témoins non traités.
Les résultats sont donnés au tableau suivant.

Ces, résultats font apparaître que l'activité antifongique du 951 est très marquée puisque le temps d'infection est fort réduit chez les animaux traités par rapport à celui des témoins (gain moyen de 22 jours), et que le 951 possède une activité intéres- sante sur les lésions mêmes, puisqu'elles sont moins graves chez les sujets traités que chez les témoins et qu'elles guérissent plus rapidement (gain moyen de 17 jours).
Le jour 0 est celui de l'inoculation ; les lésions apparaissent en général au jour 6, l'infection est reconnue au jour 7 et le traitement débute au jour 8 et est maintenu en moyenne pendant 3 semaines.
B Pouvoir anti-inflammatoire
On provoque expérimentalement une dermite chez le cobaye. Dès que les lésions sont installées, on instaure un traitement au 951 à 1 /o dans du Carbowax chez un lot de 5 animaux ; 5 autres animaux sont traités au Carbowax pur et enfin 3 autres ne sont pas traités.
Les résultats font apparaître que le 951 possède une activité anti-inflammatoire nette : dans le groupe traité avec ce produit, la guérison s'obtint en moyenne après 9 jours, alors, alors que chez le groupe traité au Carbowax seul cette guérison s'observe au 14e jour pour 3 animaux et après le 19e jour pour les 2 autres. Enfin, chez les témoins non traités, la guérison s'observe dans un cas au 13e jour et dans les deux autres cas après le 19e jour.
Essais cliniques
Le 951 a été utilisé en applications locales. A titre d'exemples, on peut rapporter les observations suivantes.
-Un cas d'eczématide de la voûte plantaire ; dû
au Ctenomyces inter-digitalis : guérison obtenue
après 15 jours.
-Un cas de mycose inter-digitale compliquée de
dyshidrose, dû an Stenomyces mentagrophyte :
guérison en 3 semaines.
Deux cas de mycose inter-digitale légère, l'un
dû au Ctenomyces mentagrophytes, l'autre au
Trichophyton rubrum : guérison totale dans les
deux cas en 3 semaines.
-De nombreux patients traités au 951 en pommade
ont signalé un soulagement très net dès les
premières applications.
Les thiocarbanilides halogénés, en particulier les thiocarbanilides fluorés sont donc des substances antifongiques intéressantes destinées à combattre les mycoses chez l'homme, chez l'animal et chez les plantes. Ces produits peuvent être utilisés soit par la voie externe (en pulvérisations, en lotions, en pommades etc.), soit par la voie interne (voies orale et parentérale). Aux doses actives, les thiocarbanilides halogénés possèdent une toxicité très faible et sont tolérés facilement à des concentrations dépas- sant 1 g par kilo d'animal. Le spectre d'activité est très vaste et englobe même des organismes, tels que
Histoplasma, qui sont généralement très résistants vis-à-vis de la chimiothérapie.

This new product of formula (II) is in the form of fine colorless crystals, melting at 1480 C (in a capillary tube). It is insoluble in water, sparingly soluble in alcohols and glycols, in dimethylformamide and in acetic acid.
It is soluble in ethyl oleate, isopropyl myristate and in oily excipients for ointments and ointments.
This product of formula II can be prepared, for example, according to either of the methods described in the following examples.

Antifungal activity in vitro
The mycostatic activities of halogenated thiocarbanilides (II), (III), (IV) and (V) were studied.
In tables I and II appended, results are given as regards the activity of the product of formula (II), respectively after 7 days and after 15 days, and this for various concentrations of this product and on various organisms.
For the proper understanding of these tables, we will consider that a white square corresponds to 100 ouzo of activity of the product (II) at the corresponding concentration for the organism envisaged, a quarter of a white square corresponds to 25'o / o d activity, etc. Recently, a hatched quarter of a square corresponds to Z5 10 / o of growth of the organism, etc.

Experimental studies in vivo
Experimental in vivo studies were carried out for the product (II) designated below by 951.
A Antifungal power
Guinea pigs were experimentally infected with Microsporum canis. Daily treatment of the animals was planned by application of 951 ointment (1% of active product in Carbowax). Treatment began the day after the infection was recognized and the lesions installed (on average 8 days after inoculation) Regular observations of the subjects were made: control of the evolution of the infection (Wood's lumen, microscopic examinations and cultures) and lesions (macroscopic examination).
Experiments were carried out on 7 guinea pigs treated with 951 and 9 untreated control animals.
The results are given in the following table.

These results show that the antifungal activity of 951 is very marked since the infection time is greatly reduced in the treated animals compared to that of the controls (mean gain of 22 days), and that the 951 has an interesting activity. - health on the lesions themselves, since they are less serious in the subjects treated than in the controls and they heal more quickly (average gain of 17 days).
Day 0 is that of inoculation; lesions usually appear on day 6, infection is recognized on day 7 and treatment begins on day 8 and is maintained on average for 3 weeks.
B Anti-inflammatory power
Dermatitis is caused experimentally in the guinea pig. As soon as the lesions are installed, a treatment with 951 at 1 / o in Carbowax is initiated in a batch of 5 animals; 5 other animals are treated with pure Carbowax and finally 3 others are not treated.
The results show that 951 has a clear anti-inflammatory activity: in the group treated with this product, healing was obtained on average after 9 days, while in the group treated with Carbowax only this recovery was observed. on the 14th day for 3 animals and after the 19th day for the 2 others. Finally, in untreated controls, healing was observed in one case on the 13th day and in the other two cases after the 19th day.
Clinical tests
The 951 has been used in local applications. As examples, the following observations can be reported.
-A case of eczematid of the arch of the foot; of
with Ctenomyces inter-digitalis: cure obtained
after 15 days.
-A case of inter-digital mycosis complicated by
dyshidrosis, due to Stenomyces mentagrophyte:
healing in 3 weeks.
Two cases of mild inter-digital mycosis, one
due to Ctenomyces mentagrophytes, the other to
Trichophyton rubrum: total cure in
two cases in 3 weeks.
-Many patients treated with 951 ointment
reported very clear relief from the
first applications.
Halogenated thiocarbanilides, in particular fluorinated thiocarbanilides, are therefore valuable antifungal substances intended for combating mycoses in humans, animals and plants. These products can be used either externally (in sprays, lotions, ointments, etc.), or internally (oral and parenteral). At active doses, halogenated thiocarbanilides have very low toxicity and are readily tolerated at concentrations exceeding 1 g per kg of animal. The spectrum of activity is very wide and even includes organisms, such as
Histoplasma, which are generally very resistant to chemotherapy.
