Verfahren zur Herstellung von Penicillinen
Die vorliegende Erfindung betrifft ein syntheti- sches Verfahren zur Herstellung von neuen Penicillinen mit wertvollen antibakteriellen Eigenschaften.
Sie dienen als Zusätze zu tierischem Futter, als Agentien zur Behandlung von Mastitis bei Hornvieh und als therapeutische Präparate für Federvieh und Säugetiere, sowie zur Behandlung des Menschen, insbesondere bei infektiösen Erkrankungen, welche durch gram-positive Bakterien verursacht worden sind.
Antibakterielle Agentien, wie das Benzylpenicil- lin, haben sich als hervorragend wirksam in der The- rapie von Infektionen durch gram-positive Bakterien erwiesen. Jedoch zeigen solche Agentien den schwer wiegenden Nachteil, dass sie in wässrigen Säuren, z. B. bei Verabreichung durch den Mund, unstabil sind und sich ferner unwirksam erweisen gegen zahlreiche sogenannte resistente Bakterienstämme, z. B.
Penicillin-resistente Stämme von Staphylococcus aureus (Micrococcus pyogenes var. aureus). Ausserdem ist Benzylpenicillin ungenügend wirksam gegen manche Bakterien, welche Penicillinase produzieren.
Viele der erfindungsgomäss hergestellten Verbindungen zeigen ausser kräftiger antibakterieller Aktivität Widerstandsvermögen gegen die Zerstörung durch Säure oder durch Penicillinase und sind deswegen gegen Benzylpenicillin-resistente Bakterienstämme wirksam oder inhibieren die Benzylpenicillinase und verstärken auf diese Weise die Wirkung des Benzylpeni- cillins,, wenn sie mit letzterem vermischt werden.

Die nach dem erfindungsgemässen Verfahren her stellbaren nicht-toxischen Salze von Säuren der For mel I sind z. B. Salze des Natrums, Kaliums, Calciums und Aluminiums, Ammoniumsalze und sub- stituierte Ammoniumsalze, ferner Salze nicht-toxischer Amine, wie der niederen Trialkylamine ein- schliesslich Triäthylamin, Procain, Dibenzylamin, N Benzyl--phenäthylandn, l-Ephenamin, N, N'-Diben- zyl-äthylendiamin, Dehydroabiethylamin, N, N'-bis Dehydroabietyl-äthylendiamin, niedere Alkylpiperidine, wie sie bisher zur Bildung von Salzen des Ben zylpenicillins schon Verwendung gefunden haben.
Die Alkylreste können gerad-und verzweigtkettig sein und enthalten vorzugsweise 1 bis 12 Kohlenstoffatome. Der Ausdruck niedere Alkylgruppen bezieht sich, wie üblich, auf Alkylgruppen mit 1 bis 6 Kohlenstoffatomen.
Von besonderem Interesse sind solche Penicilline der Formel I, worin R, ein Allylradikal oder eine niedere Alkylgruppe, insbesondere N-Propyl, Isopropyl, n-Amyl, n-Butyl und tertiäres Butyl, ist. Aus den erfindungsgemäss erhaltenen Penicillinen k¯nnen entsprechende Ester hergestellt werden, welche sich durch chemische oder enzymatische Hydrolyse wiederum in die freie Säure überführen lassen.
Mit Vorteil kann man beim erfindungsgemässen Verfahren von einem Neutralsalz der 6-Amino-penicillansäure ausgehen.
Als Acylierungsmittel verwendet man vorteilhafterweise ein Säurehalogenid, insbesondere ein Säure- chlorid der Formel

Eine besonders vorteilhafte Herstellungsweise benutzt als Acylierungsmittel ein Mischanhydrid, wie es hergestellt werden kann durch Umsetzung der Säure

mit einem Alkylester, insbesondere einem niederen Alkylester der Cblorkohlensäure, in Gegenwart eines tertiären Amins und eines wasserfreien inerten Lö sungsmittels.
Die erfindungsgemäss mit der 6-Amino-penicillansäure umzusetzenden Acylierungsmittel leiten sich ab von N-substituierterPhthalaminsäure. DiesePhthal- aminsäurederivate lassen sich leicht nach in der technischen Literatur beschriebenen Methoden herstellen, wovon eine im nachfolgenden Beispiel 1 angegeben wird. Das für diese Herstellung benötigte Phthalsäure- anhydrid und manche der dazu verwendeten Amine sind handelsüblich erhältlich, bzw. können leicht nach bekannten präparativen Methoden hergestellt werden.
Eine vorteilhafte Arbeitsweise zur erfindungsgemässen Herstellung von Verbindungen der Formel I unter Verwendung eines Mischanhydrids mit Äthoxy-oder Isobutoxycarbonsäure besteht darin, dass eine Phthalaminsäure der oben angegebenen Formel mit z. B. Isobutylchlorformiat und einem ter tiären Amin, wie Triäthylamin, in einem wasserfreien inerten und vorzugsweise mit Wasser mischbaren Lo sungsmittel, wie p-Dioxan und gegebenenfalls mit einer geringen Menge trockenen Acetons, während ungefähr 30 Minuten bei etwa 4 gemischt wird. Zur L¯sung des dabei entstandenen Mischanhydrids wird eine abgekühlte L¯sung von 6-Amino-penicillansäure und einem tertiären Amin, z. B. Triäthylamin, in einem Lösungsmittel, wie Wasser, hinzugegeben.
Das Reaktionsgemenge wird während etwa 1 Stunde ger hrt, wobei das substituierte Ammoniumsalz des gewünschten Endproduktes entsteht. Die Mischung wird hierauf günstigerweise bei alkalischem pH-Wert mit einem mit Wasser nicht mischbaren Lösungsmittel, wie z. B. Ather, extrahiert, um nicht umgesetzte Aus gangsprodukte zu entfernen. Das in der wässrigen Phase enthaltene Produkt wird hierauf in die freie Säure übergeführt, was vorzugsweise in der Kälte unter einer Schicht von Ather durch Zugabe von ver dünnter Mineralsäure, z. B. 5n H. SO, bis zum pH Wert 2 geschieht. Die freie Säure wird hiernach mit einem mit Wasser nicht mischbaren neutralen organischen Lösungs. mittel, wie z.
B. Ather, extrahiert und der Extrakt mit Wasser in der Kälte rasch gewaschen und hiernach gegebenenfalls getrocknet. Die im äthe- ris. chen Extrakt enthaltene freie Säure kann hierauf in jedes gewünschte Metallsalz oder Aminsalz über- geführt werden, indem man sie mit der geeigneten Base behandelt. Solche Basen sind z. B. freie Amine, wie Procainbase oder eine L¯sung von Kalium-2 äthylhexanoat in trockenem n-Butanol. Die dabei entstehenden Salze sind normalerweise unlöslich in Lö sungsmitteln, z. B. Äther, und können daraus durch einfaches Filtrieren abgeschieden werden.
Einige der erfindungsgemäss hergestellten Substanzen sind verhältnismässig unstabile Verbindungen und unterliegen chemischen Anderungen, die zu einer r Verminderung ihrer antibakteriellen Aktivität führen.
Aus diesem Gninde werden beim Herstellungsverfahren mit Vorteil derart milde Reaktionsbedingungen angewandt, dass eine Zersetzung der Substanzen unterbleibt. Die auszuwählenden Reaktionsbedingungen hängen natürlich in weitem Mass von der Reaktionsbereitschaft der umzusetzenden Verbindungen ab. In den meisten Fällen ist es erforderlich, einen Kompro- miss zu schliessen zwischen der Anwend. ung sehr milder Bedingungen während längerer Zeitdauer oder der Anwendung kräftigerer Reaktionsbedingungen wäh- rend kürzerer Zeit mit der Möglichkeit einer Zersetzung eines Teils der antibiotischen Substanz.
Die zur Herstellung von Derivaten der 6-Aminopenicillansäure einzuhaltende Temperatur sollte im allgemeinen 30 nicht überschreiten. In manchen Fäl- len erweist sich Zimmertemperatur als geeignet. In der Regel sollten beim erfindungsgemässen Verfahren stark saure oder stark alkalische Reaktionsbedingun- gen womöglich vermieden werden. Vorzugsweise wird die Umsetzung daher im pH-Bereich zwischen 6 und 9 durchgeführt, was in üblicher Weise durch Verwen- dung eines Puffers, z. B. einer L¯sung von Natriumbicarbonat oder eines Natriumphosphatpuffers, erreicht werden kann.
Ausser der Anwendung wässriger Reaktionsmedien, einschliesslich filtrierter Fermenta tionsbrühen, oder wässriger Lösungen, roher 6-Ami no-penicillansäure, können auch organische Lösungsmittel angewendet werden, wie z. B. Dimethylformamid, Dimethylacetamid, Chloroform, Aceton, Methyl isobutyl-keton und Dioxan. Häufig erweist es sich als besonders günstig, eine wässrige e L¯sung eines Salzes der 6-Amino-penicillansäure zu einer L¯sung des Acylierungsmittels in einem inerten und vorzugsweise mit Wasser mischbaren Lösungsmittel, wie Aceton oder Dimethylformamid, hinzuzugeben. Kräftiges Rühren ist naturgemäss dann von Vorteil, wenn mehr als eine Phase, z. B. feste und flüssige oder zwei flüsige Phasen, zugegen sind.
Die erfindungsgemäss hergestellten Produkte kön- nen nach denjenigen Techniken isoliert werden, wie sie für Benzylpenicillin und Phenoxymethylpenicillin bekannt sind. So kann das Produkt mit Diäthyläther oder n-Butanol bei saurem pH-Wert extrahiert und hierauf durch Lyophilisation abgeschieden werden, oder es kann in ein im Lösungsmittel nicht lösliches Salz übergeführt werden, z. B. vermittels Neutralisation mit einer L¯sung von Kalium-2-äthylhexanoat in n-Butanol. Man kann das Produkt auch aus wässriger L¯sung, als wasserunlösliches Salz eines Amins ausfällen oder direkt vermittels Lyophilisation gewinnen, was vorzugsweise in Form eines Natrium-oder Ka liumsalzes geschieht.
Falls das Triäthylaminsalz erhalten wurde, kann das Produkt in die freie Säure übergeführt und hiernach in ein anderes Salz umgesetzt werden, wie dies bei den Herstellungsverfahren von Benzylpenicillin und anderen Penicillinen bekannt ist. So ergibt die Behandlung eines solchen Triäthyl- aminsalzes in Wasser mit Natriumhydroxyd das Natriumsalz, wonach das Triäthylamin vermittels Extraktion, z. B. mit Toluol, entfernt werden kann. An schliessend ergibt die Behandlung des Natriumsalzes mit starker wässriger Säure die freie Säure der Verbindung, welche ihrerseits wieder in andere Aminsalze, z. B. in das Procainsalz durch Umsetzung mit der betreffenden Base, übergeführt werden kann.
Derart erhaltene Salze werden mit Vorteil durch Lyophilisation oder, sofern sie unlöslich sind, durch Filtration abgetrennt. Eine Arbeitsweise zur Isolierung des er findungs. gemäss hergestellten Produktes als kristallines Kaliumsalz besteht darin, dass das Produkt aus einer sauren wässrigen Lösung (z. B. vom pH-Wert 2) mit Diäthyläther extrahiert wird, worauf der Ätherextrakt getrocknet und mindestens ein Äquivalent einer Lösung von Kalium-2-äthylhexanoat in trockenem n Butanol hinzugegeben wird. Das Kaliumsalz fällt aus, meistens in kristalliner Form, und kann abfiltriert oder durch Dekantation gewonnen werden.
Beispiel I a) 74, 0 g (0, 5 Mol) Phthalsäureanhydrid und 300 ml Benzol wurden in einem 1-Liter-Dreihals Rundkolben gemischt. Das Gemisch wurde 20 Minuten lang gerührt und hiernach eine Mischung von 0, 5 Mol (28, 5 g) Allylamin in 100 ml Benzol tropfenweise im Verlauf von einer halben Stunde hinzugegeben.
Die erhaltene Mischung wurde weitere 20 Minuten bei Zimmertemperatur gerührt. Das Reaktionsgemisch wurde hiernach auf ein Wasserbad gebracht und am Rückfluss eine Stunde lang gekocht. Das Gemenge wurde auf Zimmertemperatur abkühlen gelassen und hiernach während ungefähr 20 Minuten auf ein Eisbad gesetzt, worauf das Reaktionsprodukt ausfiel. Die N-Allyl-phthalaminsäure wurde abfiltriert, über Nacht in einem Exsikkator getrocknet und wog 92, 4 g. Sie hatte einen Smp. von 115 bis 117 . Das nach dem Umkristallisieren aus Aceton erhaltene Produkt wog 60, 2 g und hatte einen Smp. von 115 bis 117 .
Analyse, berechnet C = 64, 38 /o, H= 5, 40"/o.
Gefunden C = 64, 40 /o, H = 5, 50 I/o.
Nach ähnlichen Arbeitsweisen mit Aceton als Lö sungsmittel wurden N-Benzyl-phthalaminsäure, N-n Propyl-phthalaminsäure und andere Säuren hergestellt, wobei die Reaktionsbedingungen in Abhängigkeit von den Eigenschaften der verschiedenen verwendeten Amine leicht variiert wurden. b) 10 ml Athylchlorformiat wurden tropfenweise unter Rühren bei ungefähr-5 C zu einer L¯sung von 20, 52g (0, 1 Mol) N-Allyl-phthalaminsäure, 14ml Tri Ïthylamin, 70 ml p-Dioxan und 30 ml trockenen Ace- ton hinzugegeben.
Nachdem das Gemenge bei-5 15 Minuten lang gerührt worden war, wurde eine Lösung von 21, 6 g (0, 1 Mol) 6-Arnino-penicillansäure in 50 ml Wasser, die zuvor auf 0 C abgekühlt worden war, und 15 ml Triäthylamin in einem Guss zugefügt. Das Reaktionsgemisch wurde 2 Stunden lang kräftig bei 0 gerührt. Hiernach wurde es mit einem gleichen Volumen Wasser verdünnt und dreimal mit Methyl isobutyl-keton extrahiert, wobei die Extrakte verwor- fen wurden. Die kalte wässrige L¯sung wurde mit Me thyl-isobutyl-keton überschichtet und mit 42 %iger Phosphorsäure auf den pH-Wert 2 angesäuert.
Die angesäuerte L¯sung wurde mit 700 ml Methyl-isoW butyl-keton in 3 Portionen extrahiert. Die Methyl-iso butyl-keton-Extrakte, worin die 6- (N-allyl-N'-phthal- amido)-penicillansäure enthalten war, wurden einmal mit Wasser gewaschen, mit Natriumsulfat getrocknet, filtriert und mit einer L¯sung von Kalium-2-äthyl- hexanoat in Ather behandelt. Das Lösungsmittel wurde abdekantiert und das zurückbleibende Produkt mit 400 ml Aceton iiberschichtet. Das erhaltene Kaliumsalz der 6- (N-allyl-N'-phthalamido)-penicillansäure wurde abfiltriert, im Vakuum über Phosphor- pentoxyd getrocknet und wog hiernach 20, 0 g.
Es enthielt die, B-Lactamstruktür, wie durch Infrarot- Analyse nachgewiesen werden konnte, und inhibierte Staph. aureus Smith bei einer Konzentration von 0, 8 mcg/ml. Bei intramuskulärer Injektion in Mäuse lag die Heilungsdosis CD-,, bei 6, 8 mcg/kg.
Beispiel 2
13, 9 g = 0, 1 Mol Triäthylamin wurden in einer Portion zu einer Suspension von 25, 5 g (0, 1 Mol) N Benzylphthalaminsäure in 150 ml Tetrahydrofuran hinzugegeben, wobei sich eine klare Lösung bildete.
Die L¯sung wurde auf ungefÏhr -5¯ abgek hlt und 13, 7g (0, 1 Mol) Isobutylchlorformiat tropfenweise hinzugefügt, während die Temperatur des Gemisches bei ungefähr-5 gehalten wurde. Die erhaltene Reak tionsmischung wurde eine halbe Stunde lang gerührt und hiernach in einer Portion eine abgekühlte Lösung von 21, 6g (0, 1 Mol) 6-Amino-penicillansäure in 40 ml Wasser und 15 ml Triäthylamin hinzugegeben. Das Reaktionsgemenge wurde eine halbe Stunde lang bei-5 in einem Kältebad gerührt und hiernach weitere 2 Stunden bei Zimmertemperatur. Anschlies- send wurde das Gemenge mit einem gleichen Volumen Wasser verdünnt und zweimal mit Methyl-isow butyl-keton extrahiert, wobei die Extrakte verworfen wurden.
Die wässrige L¯sung wurde mit Methyl-iso butyl-keton überschichtet, abgekühlt und mit 42, 5 %iger Phosphorsäure zum pH-Wert 2 angesäuert. Die angesäuerte wässrige Lösung, welche die 6- (N-benzyl N'-phthalamido)-penicillansäure enthielt, wurde zwei- mal mit Methyl-isobutyl-keton extrahiert, wonach die vereinigten Extrakte mit abgekühltem Wasser gewaschen, durch Natriumsulfat filtriert und über Natriumsulfat getrocknet wurden. Das Natriumsulfat wurde aus den Extrakten abgeschieden und zu letzteren 33, 2 ml einer 50 %igen L¯sung von Natrium-2 äthylhexanoat zugegeben, worauf sich eine Fällung bildete.
Das ausgefallene Natriumsalz der 6- (N-ben- zyl-N'-phthalamido)-penicillansäure wurde abfiltriert, in Aceton aufgeschlämmt, erneut filtriert, an Luft und schliesslich im Vakuum über Phosphorpentoxyd getrocknet. Das erhaltene Produkt wog 11, 8 g. Es enthielt die jS-Lactamstruktur, nachgewiesen mittels Infrarot-Analyse und inhibierte Staph. aureus Smith bei einer Konzentration von 0, 5 mcg/ml. Bei intramuskulärer Injektion in Mäusen betrug die : Heilungs- dosis CD50 17 mcg/kg.
Analyse, berechnet für C25H22N3OsSNa : C = 58, 1 /o, H = 4, 66%. Gefunden : C = 54, 65 /o, H = 4, 75 /o.
Beispiel 3
13, 9 g = 0, 1 Mol Triäthylamin wurden in einer Portion zu einer Suspension von 20, 7 g (0, 1 Mol) N- n-Propyl-phthalaminsäure in 100 ml Tetrahydrofuran hinzugegeben, wobei eine klare L¯sung entstund. Die L¯sung wurde in einem Bad aus Eis, Salz und Aceton abgekühlt und 13, 7 (0, 1 Mol) Isobutylchlorformiat tropfenweise hinzugegeben, wobei die Temperatur der L¯sung auf der Temperatur des Kältebades gehalten wurde. Das erhaltene Reaktionsgemisch wurde eine halbe Stunde lang gerührt und hiernach in einer Portion zu einer abgekühlten L¯sung von 21, 6 g (0, 1 Mol) 6-Amino-penicillansäure in 40 ml Wasser und 15 ml Triäthylamin hinzugefügt.
Das erhaltene Reak tionsgemenge wurde vom Kältebad weggenommen, 2t/2 Stunden lang bei Zimmertemperatur gerührt und hiernach mit einem gleichen Volumen Wasser verd nnt. Es wurde nunmehr zweimal mit Methyl-isobutyl-keton extrahiert, wobei die Extrakte verworfen wurden. Die wässrige Lösung wurde mit Methyl-isobutyl-keton überschichtet, abgekühlt und mit 42, 5 o,'o- iger Phosphorsäure zum pH-Wert 2 angesäuert. Die angesäuerte wässrige L¯sung, welche die 6- (N-n-pro pyl-N'-phthalamido)-penicillansäure enthielt, wurde zweimal mit Methyl-isobutyl-keton extrahiert, worauf die vereinigten Extrakte mit abgekühltem Wasser gewaschen, durch Natriumsulfat filtriert und über Natriumsulfat getrocknet wurden.
Das Natriumsulfat wurde vom Extrakt abgetrennt und 40 ml einer 50"tu igen L¯sun von Kalium-2-äthylenhexanoat in n-Butanol zum letzteren hinzugefügt, worauf eine Fällung entstund. Das ausgefallene Kaliumsalz der 6- (N-n- propyl-N'-phthalamido)-penicillansäure wurde abfil- triert, in Aceton aufgeschlämmt, erneut filtriert, an Luft und schliesslich im Vakuum über Phosphorpent- oxyd getrocknet. Das erhaltene Produkt wog 22, 5 g.
Es zeigte bei derInfrarot-Analyse einess-Lactamstruk- tur. Es inhibierte Staph. aureus Smith bei einer Kon zentration von 1, 6 mcg/ml.
Analyse, berechnet für CtaHNgO. SK : C51, 5 /o, H = 5, 00 /o. Gefunden : C = 48, 9 solo, H = 5, 28 %.
Beispiel 4
13, 9 g = 0, 1 Mol Triäthylamin wurden in einer Portion zu einer Suspension von 23, 5 g (0, 1 Mol) N n-Amylphthalaminsäure in 150 ml Tetrahydrofuran hinzugegeben, wobei eine klare L¯sung entstund. Die L¯sung wurde auf ungefähr-5 abgekühlt, worauf 13, 6 g (0, 1 Mol) Isobutylchlorformiat in 20 ml Tetrahydrofuran tropfenweise hinzugefügt wurden, wobei die Temperatur der L¯sung bei ungefähr-5 gehalten wurde. Das erhaltene Reaktionsgemisch wurde eine halbe Stunde lang bei ungefähr 0 gerührt und hierauf in einer Portion zu einer abgekühlten Lösung von 21, 6 g (0, 1 Mol) 6-Amino-peniciIlansäure in 60ml Wasser und 15 ml Triäthylamin hinzugefügt.
Das erhaltene Reaktionsgemisch wurde eine halbe Stunde lang auf dem Kältebad gerührt und hiernach weitere 2 Stunden bei Zimmertemperatur. Das Reaktions- gemenge wurde mit einem gleichen Volumen Wasser verdünnt und zweimal mit Methyl-isobutyl-keton extrahiert, wobei die Extrakte verworfen wurden. Die wässrige L¯sung wurde mit Methyl-isobutyl-keton überschichtet, abgekühlt und mit 42, 5 %iger Phosphorsäure zum pH-Wert 2 anges. äuert. Die angesäuerte wässrige Lösung, welche die 6- (N-n-methyl N'-phthalamido)-penicillansäure enthielt, wurde zweimal mit Methyl-isobutyl-keton extrahiert, wonach die vereinigten Extrakte mit abgekühltem Wasser gewaschen, durch Natriumsulfat filtriert und über Natriumsulfat getrocknet wurden.
Das Natriumsulfat wurde vom Extrakt abgetrennt und zu letzterem 40 ml einer 50%igen L¯sung von Kalium-2-äthylenhexanoat in n-Butanol hinzugefügt, wobei ein Niederschlag entstund. Das ausgefallene Kaliumsalz der 6- (N-n-amyl N'-phthalamido)-penicillansäure wurde abfiltriert, mit Aceton aufgeschlämmt, erneut filtriert und schliesslich an Luft und im Vakuum über Phosphorpentoxyd getrocknet. Das erhaltene Produkt wog 28, 0 g und zersetzte sich beim Erwärmen bei 140 bis 145 . Es enthielt eine durch Infrarot-Analyse nachgewiesene p-Lactamstruktur. Es inhibierte Staph. aureus Smith bei einer Konzentration von 0, 8 mcg/ml.
Analyse, berechnet für C21H26N3O5SK: C = 53, 5 /o, H = 5, 52 /o. Gefunden : C = 49, 65 %, H = 5, 18 /o.

Es wurde nach der Arbeitsweise des Beispiels. 4 verfahren, wobei jedoch die N-n-Amyl-phthalamin- säure ersetzt wurde durch 24, 5 g (0, 1 Mol) N-Fur furyl-phthalaminsäure. Erhalten wurde dabei das Kaliumsalz der 6- (N-Furfuryl-N-phthalamido)-penicil- lansäure, welche 26, 5 g wog und sich beim Erhitzen bei 120 zersetzte. Es enthielt dio ss-Lactamstruktur, nachweisbar durch Infrarot-Analyse und inhibierte Staph. aureus Smith bei einer Konzentration von 0, 8 mcg/ml.

Beispiel 6
Die Arbeitsweise des Beispiels 4 wurde nachgearbeitet, wobei die N-n-Amyl-phthalaminsäure ersetzt wurde durch 23, 1 g (0, 1 Mol) N- (1, 2, 5, 6-Tetra hydro-pyridyl)-N'-phthalaminsäure. Erhalten wurde dabei das Kaliumsalz der 6- [N- (1, 2, 5, 6-Tetrahydro pyridyl)-N'-phthaIamido]-penicilIansäure im Gewicht von 37, 5 g. Es zersetzte sich oberhalb 140 beim Erwärmen. Es enthielt die ss-Lactamstruktur, nachgewie- sen vermittels Infrarot-Analyse. Die Substanz inhibierte Staph. aureus Smith bei einer Konzentration von 0, 4 mcg/ml.
Analyse, berechnet f r C21H22N3O5SK : C = 53, 96 %, H = 4, 71 %. Gefunden : C = 49, 90 /o, H = 5, 56 /o.
Beispiel 7
10 ml Athylehlorformiat wurden tropfenweise unter Rühren bei ungefähr-5 zu einer L¯sung von 21, 9 g (0, 1 Mol) N, N-Tetramethylen-N'-phthalamin- säure, 14 ml TriÏthylamin, 70 ml p-Dioxan und 30 ml trockenem Aceton hingefiigt. Nachdem 15 Mi- nuten lang bei5 gerührt worden war, wurde eine L¯sung von 21, 6 g (0, 1 Mol) 6-Amino-penicillansäure in 50 ml vorher auf 0 abgekühltes Wasser und 14 ml Triäthylamin in einem Guss hinzugegeben. Das Reak tionsgemenge wurde kräftig 2 Stunden lang bei 0 gerührt. Es wurde hiernach dreimal mit Methyl-isobutyl-keton extrahiert, wobei die Extrakte verworfen wurden.
Die kalte wässrige L¯sung wurde überschichtet mit Methyl-isobutyl-keton und mit 42 %iger Phosphorsäure zum pH-Wert 2 angesäuert. Die angesäuerte Losung wurde mit 700 ml Methyl-isobutylketon in 3 Portionen extrahiert. Die Methyl-isobutyl keton-Extrakte, welche die 6- (N, N-Tetramethylen- N'-phthalamido)-penicillansäure enthielten, wurden mit Natriumsulfat getrocknet, filtriert und mit einer Lösung von Kalium-2-äthylhexanoat in n-Butanol behandelt. Die L¯sun wurde im Vakuum eingeengt und mit trockenem Ather verdünnt, wonach das. erhaltene Produkt abgetrennt und mit Aceton gewaschen wurde.
Das feste Kalium-6- (N, N-totramethylen-N'-phthal- amido)-penicillinat wog nach dem Trocknen im Vakuum über Phosphorpentoxyd 21, 0 g. Es schmolz unter Zersetzung bei 150 und enthielt die ss-Lactam- struktur, wie durch Infrarot-Analyse nachgewiesen werden konnte. Es inhibierte Staph. aureus Smith bei einer Konzentration von 1, 6 mcg/ml und zeigte bei intramuskulärer Injektion in Mäusen eine Heilungs- dosis CD50 von 9 mcg/kg gegenüber Staph. aureus Smith.
Beispiel 8
10 ml Athylchlorformiat wurden tropfenweise unter Rühren zu einer auf ungefähr-5 abgekühlten L¯sung von 26, 9 g (0, 1 Mol) N- (2-Phenyläthyl)- phthalaminsäure, 14 ml Triäthylamin, 70 ml p-Dio- xan und 30 ml trockenem Aceton hinzugefügt. Nachdem 15 Minuten lang bei-5 gerührt worden war, wurde eine vorgängig auf 0 abgekühlte Lösung von 21, 6 g (0, 1 Mol) 6-AminoWpenicillansäure in 50 ml Wasser und 15 ml Triäthylamin in einem Guss hinzugegeben. Das Reaktionsgemenge wurde kräftig 2 Stunden lang bei 0¯ ger hrt. Hiernach wurde es dreimal mit Methyl-isobutyl-keton extrahiert, wobei die Extrakte verworfen wurden.
Die kalte wässrige Lösung wurde mit Methyl-isobutyl-keton überschichtet und mit 42 /Oiger Phosphorsäure zum pH-Wert 2 angesäuert. Die angesäuerte Lösung wurde mit 700 ml Methyl-isobutyl-keton in 3 Portionen extrahiert. Die Methyl-isobutyl-keton-Extrakte, welche die 6- [N- (2- Phenyläthyl)-N'-phthalamido]-penicillansäure enthiel- ten, wurden mit Natriumsulfat getrocknet, filtriert und mit einer L¯sung von Kalium-2-äthylhexanoat in n Butanol behandelt. Die L¯sung wurde vom ausgefallenen Produkt abdekantiert, welches hiernach mit 400 ml Aceton überschichtet wurde.
Das feste Kaliumsalz der 6-[N-(2-Phenyläthyl)-N'-phthalamido]- penicillansäure wurde hierauf abfiltriert, im Vakuum über Phosphorpentoxyd getrocknet und wog 21, 6 g.
Es schmolz unter Zersetzung bei 130 und enthielt die ss-Lactamstruktur, wie durch Infrarot-Analyse nachgewiesen werden konnte. Es inhibierte Staph. aureus Smith bei einer Konzentration von 0, 8 mcg/ ml. Bei intramuskulärer Injektion in Mäusen erwies sich seine Heilungsdosis CD50 gegeniiber Staph. aureus Smith als 23 mcg/kg.
Beispiel 9

Die kalte wässrige Schicht wurde mit Methyl isobutyl-keton überschichtet und mit 42"/piger Phosphorsäure auf den pH-Wert 2 angesäuert. Die angesäuerte L¯sung wurde mit 700ml Methyl-isobutylketon in 3 Portionen extrahiert. Die Methyl-isobutylketon-Extrakte, welche die 6- (N, N-Pentamethylen N'-phthalamido)-penicillansäure enthielten, wurden mit Natriumsulfat getrocknet, filtriert und mit einer L¯sung von Kalium-2-äthylhexanoat in n-Butanol behandelt. Die L¯sung wurde im Vakuum eingeengt und mit trockenem ¯ther verdünnt, wobei eine Fällung entstund.
Das ausgefallene Kaliumsalz der 6- (N, N Pentamethylen-N'-phthalamido)-penicillansäure wurde abfiltriert, im Vakuum über Phosphorpentoxyd getrocknet und wog hiernach 40, 0 g. Es schmolz bei 150 unter Zersetzung. Es enthielt die ¯-Lactamstruktur, nachweisbar durch Infrarot-Analyse. Es inhibierte Staph. aureus Smith bei einer Konzentration von 6, 35 mcg/ml. Gegenüber Staph. aureus Smith erwies sich die Heilungsdosis bei intramuskulärer Injektion in Mäusen CI ? o zu 28 mcg/kg.
Beispiel 10
13, 9 g = 0, 1 Mol Triäthylamin wurden in einer Portion zu einer Suspension von 20, 5 g (0, 1 Mol) N Isopropyl-phthalaminsäure in 100 mlTetrahydrofuran hinzugegeben, wobei eine klare L¯sung entstund. Die L¯sung wurde auf ungefähr-12 C abgekühlt und 13, 7 g = 0, 1 Mol Isobutylchlorformiat in 20 ml Te- trahydrofuran tropfenweise im Verlauf von 20 Minuten hinzugefügt, wobei die Temperatur der L¯sung bei ungefähr-10 gehalten wurde. Das entstandene Reaktionsgemisch wurde'/, Stunde lang bei ungefähr 0¯ ger hrt, worauf dazu in einem Guss eine abgek hlte L¯sung von 21,6 g (0,1 Mol) 6-Amino-penicillansäure in 40 ml Wasser und 15 ml Triäthylamin hinzugegeben wurde.
Das erhaltene Reaktionsgemisch wurde 3 Stunden lang bei-10 gerührt, wonach das Rühren bei Zimmertemperatur eine weitere Stunde fortgesetzt wurde. Das Gemenge wurde mit einem gleichen Anteil Wasser gemischt und zweimal mit Methyl-isobutyl-keton extrahiert, wobei die Extrakte verworfen wurden. Die wässrige L¯sung wurde mit Methyl-isobutyl-keton überschichtet, abgekühlt und mit 42, 5 /oiger Phosphorsäure zum pH-Wert 2 angesäuert. Die angesäuerte wässrige L¯sung, welche die 6- (N-Isopropyl-N'-phthalamido)-penicillansäure enthielt, wurde zweimal mit Methyl-isobutyl-keton extrahiert, wonach die vereinigten Extrakte mit abgekühltem Wasser gewaschen, durch Natriumsulfat filtriert und über Natriumsulfat getrocknet wurden.
Das Natriumsulfat wurde von den Extrakten abgetrennt und zu letzteren 40 ml einer 50 loigen Lösung von Kalium-2-äthylhexanoat in n-Butanol hinzugege- ben. Das Gemisch wurde nunmehr im Vakuum zur Trockne konzentriert und n-Pentan zum Rückstand hinzugef gt. Das entstandene Kaliumsalz der 6-(N Isopropyl-N'-phthalamido)-penicillansäure wurde abfiltriert, an Luft und hiernach im Vakuum über Phos phorpentoxyd getrocknet. Das noch etwas feuchte Produkt wog 15, 3 g. Es enthielt die p-Lactamstruk- tur, wie durch Infrarot-Analyse nachgewiesen werden konnte. Es inhibierte Staph. aureus Smith bei einer Konzentration von 3, 12 mcg/ml.
Seine Heilungsdosis Go gegenüber Staph. aureus Smith bei intramuskulärer Injektion in Mäusen lag bei 19 mcg/kg.
Beispiel 11
Das Verfahren des Beispiels 2 wurde nachgearbeitet, wobei dieN-Benzyl-phthalaminsäure ersetzt wurde durch 0, 1 Mol N- (a-Methylbenzyl)-phthalaminsäure.
Erhalten wurde dabei das Natriumsalz der 6- (N-a- Methylbenzyl-N'-phthalamido)-penicillansäure im Gewicht von 11, 0 g. Sie enthielt die -Lactamstruktur, wie durch Infrarot-Analyse nachgewiesen werden konnte. Die Substanz inhibierte Staph. aureus Smith bei 1, 6 mcg/ml. Gegenüber Staph. aureus, Smith erwies sich die Heilungsdosis CDso bei intramuskulärer Injoktion in Mäusen bei 25 mcg ! kg.
Analyse, berechnet für C24H24N3O5SNa: C = 58, 9 /o, H = 4, 94 /o. Gefunden : C = 56, 5 /o, H = 5, 14 /o.
Beispiel 12
Gemäss Beispiel 5 wurde unter Ersatz der N-Furfuryl-phthalaminsäure durch 0, 1 Mol N, N-Hexamethylen-phthalaminsäure das Kaliumsalz der 6- (N- Hexamethylen-N'-phthalamido)-penicillansäure im Gewicht von 40, 5 g erhalten. Die Substanz enthielt die ss-Lactamstruktur, wie durch Infrarot-Analyse nachgewiesen werden konnte. Sie inhibierte Staph. aureus Smith bei einer Konzentration von 1, 6 mcg/ml.
Die Heilungsdosis. CDg gegenüber Staph. aureus Smith bei intramuskulärer Injektion in Mäusen lag bei 9 mcg/kg.
Beispiel 13
Gemäss Beispiel 5 wurde unter Ersatz der N Furfuryl-phthalaminsäure durch 0, 1 Mol N-Tetra- hydrofurfuryl-phthalaminsäure das Kaliumsalz der 6 (N-Tetrahydrofurfuryl-N'-phthalamido)-penicillan- säure erhalten. Es wog 33, 0 g und enthielt die ss-Lac- tamstruktur, wie durch Infrarot-Analyse nachgewiesen werden konnte. Die Substanz inhibierte Staph. aureus Smith bei 1, 6 mcg/ml.
Beispiel 14
Gemäss Beispiel 1 wurde unter Ersatz der N-Furfuryl-phthalaminsäure durch 0, 1 Mol N-Morpholino- phthalaminsäure das Kaliumsalz der 6- (N-Morpho- lino-N'-phthalamido)-penicillansäure im Gewicht von 25, 0 g erhalten. Die Subs. tanz enthielt die ss-Lactam- struktur, wie durch Infrarot-Analyse nachgewiesen werden konnte. Sie zersetzte sich bei 165¯ beim Er wärmen und inhibierte Staph. aureus Smith bei 3, 12 mcg/ml.
Beispiel 15
Gemäss Beis. piel 5 wurde anstelle von N-FurfurylphthalaminsÏure 0, 1 Mol N- (1, 2, 3, 4-Tetrahydro-chi- naldino)-phthalaminsäure verwendet. Erhalten wurde dabei das Kaliumsalz der 6- [o- (2'-Methyl-l', 2', 3', 4' tetrahydro-chinolylcarbonyl)-benzamido]-penicillan- säure im Gewicht von 43, 5 g.
Die Substanz enthielt den ss-Lactamring, wie durch Infrarot-Analyse nachgewiesen werden konnte und zersetzte sich beim Erwärmen bei 150 . Sie inhibierte Staph. aureus Smith bei 1, 6 mcg/ml. Bei intramuskulärer Injektion in Mäusen lag die Heilungsdosis CDgo gegenüber Staph. aureus Smith bei 45 mcg/kg.
Analyse, berechnet für CAN, O, SK : C = 58, 75"/o, H = 4, 89 /o. Gefunden : C = 58, 15 /o, H = 5, 52 %.
Beispiel 16
13, 9g (0, 1 Mol) Triäthylamin wurden in einer Portion zu einer Suspension von 24, 5 (0, 1 Mol) N, N-Diallyl-phthalaminsÏure in 100 ml Tetrahydrofuran zugegeben, wobei eine L¯sung entstund. Die L¯sung wurde auf un. gefähr-5 abgekühlt und 13, 6 (0, 1 Mol) Isobutylchlorformiat in 20 ml Tetrahydro- furan tropfenweise hinzugefügt, wobei die Temperatur der L¯sung auf ungefähr-5 gehalten wurde.
Die erhaltene Reaktionsmischung wurde 1/2 Stunde lang bei ungefÏhr 0 gerührt, wonach in einer Portion eine abgekühlte L¯sung von 21, 6 g (0, 1 Mol) 6 Amino-penicillansäure in 60 ml Wasser und 15ml Triäthylamin hinzugegeben wurde. Das erhaltene Gemenge wurde eine halbe Stunde lang im Kältebad und hiernach weitere 2 Stunden lang bei Zimmertemperatur gerührt. Das Reaktionsgemisch wurde mit einem gleichen Volumen Wasser verdünnt und zweimal mit Methyl-isobutyl-keton extrahiert, wobei die Extrakte verworfen wurden. Die wässrige L¯sung wurde mit Methyl-isobutyl-keton überschichtet, ab- gekühlt und mit 42, 5 /oiger Phosphorsäure zum pH Wert 2 angesäuert.
Die angesäuerte wässrige Lösung, welche die 6- (N, N-Diallyl-N'-phthalamido)-penicil- lansäure enthielt, wurde zweimal mit Methyl-isobutylketon extrahiert, wonach die vereinigten Extrakte mit abgekühltem Wasser gewaschen, durch Natriumsulfat filtriert und über Natriumsulfat getrocknet wurden.
Das Natriumsulfat wurde abgetrennt und der Extrakt mit 40 ml einer 50 /oigen Lösung von Kalium-2 äthylhexanoat in n-Butanol versetzt, wonach die Lösung auf ein niedriges Volumen eingeengt und mit Ather verdünnt wurde. Hierbei entstund eine Fällung.
Das ausgefallene Kaliumsalz der 6-(N, N-Diallyl-N' phthalamido)-penicillansäure wurde abfiltriert, in Aceton aufgeschlämmt, erneut filtriert, an Luft und schliesslich im Vakuum über Phosphorpentoxyd getrocknet. Das derart erhaltene Produkt wog 40, 5 g.
Es enthielt die ss-Lactamstruktur, wie mit Infrarot- Analyse nachgewiesen werden konnte. Es inhibierte Staph. aureus Smith bei einer Konzentration von 0, 8 mcg/ml. Die Heilungsdosis CDvo gegenüber Staph. aureus Smith bei intramuskulärer Injektion in Mäusen lag bei 4 mcg/kg.
Beispiel 17
Es wurde gemäss Beispiel 16 gearbeitet, wobei die N, N-Diallylphthalaminsäure ersetzt wurde durch 0, 1 Mol N, N-Diisopropyl-phthalaminsÏure bzw. N, N-Di äthyl-phthalaminsäure bzw. N, N-Dimethyl-phthal aminsäure. Erhalten wurden dabei die Kaliumsalze der 6- (N, N-Diisopropyl-N'-phthalamido)-penicillan- säure, 6- (N, N-Diäthyl-N'-phthalamido)-penicillan- säure und 6-(N, N-Dimethyl-N'-phthalamido3-penicil- lansäure. Alle diese Salze wurden in fester Form iso- liert. Sie wiesen nach der Infrarot-Analyse alle einen ss-Lactamring auf.
Sie inhibierten Staph. aureus Smith bei Konzentrationen von 0, 001 Gew.- /o.
Beispiel 18
Gemäss Beispiel 4 wurde unter Ersatz der N- (n Amyl)-phthalaminsäure durch 0, 1 Mol N-Dodecylphthalaminsäure das wasserlösliche Kaliumsalz der 6-(N-Dodecyl-N'-phthalamido)-penicillansäure im Gewicht von 10, 5 g erhalten. Die Substanz enthielt den P-Lactamring (nachweisbar durch Infrarot-Analyse) und inhibierte Staph. aureus Smith bei 0, 4 mcg/ml.
Beispiel 19
13, 9 g (0, 1 Mol) Triäthylamin wurden in einer Portion zu einer Suspension von 22, 1 g (0, 1 Mol) N-t-Butyl-phthalaminsäure in 100 ml Tetrahydro- furan hinzugegeben, wobei sich eine homogene Lösung bildete.
Die L¯sung wurde auf ungefähr-5 abgekühlt und 13, 7 g (0, 1 Mol) Chlorformiat in 20 ml Tetrahydrofuran tropfenweise im Verlauf von 20 Mi- nuten zu derselben hinzugefügt, wobei die Temperatur auf ungefähr-10 gehalten wurde. Das entstan- dene Reaktionsgemisch wurde 1/2 Stunde lang bei ungefähr 0 C gerührt und hiernach in einer Portion eine abgekühlte L¯sung von 21, 6 g (0, 1 Mol) 6 Amino-penicillansäure in 50 ml Wasser und 15 ml Triäthylamin hinzugefügt. Das erhaltene Gemenge wurde 11/2 Stunden lang bei einer Temperatur von -5 bis +5 und hierauf bei Zimmertemperatur gerührt, bis die Entwicklung von CO2 aufgehört hatte.
Das Reaktionsgemisch wurde mit einem gleichen Gewicht an Wasser verdünnt und zweimal mit Methyl isobutyl-keton extrahiert, wobei die Extrakte verwor- fen wurden. Die wässrige Lösung wurde mit Methyl isobutyl-keton überschichtet, abgekühlt und auf den pH-Wert 2 mit 42, 5 %iger PhosphorsÏure angesÏuert.
Die angesäuerte wässrige L¯sung, welche die 6- (N-t- Butyl-N'-phthalamido)-penicillansäure enthielt, wurde zweimal mit Methyl-isobutyl-keton extrahiert, worauf die vereinigten Extrakte mit abgekühltem Wasser gewaschen, durch Natriumsulfat filtriert und über Natriumsulfat getrocknet wurden. Das Natriumsulfat wurde von den Extrakten abgetrennt und letztere mit 45 ml einer 40 /oigen L¯sung von Kalium-2-äthyl- hexanoat in n-Butanol versetzt, wobei sich eine Fällung abschied. Das Gemenge wurde im Vakuum zur Trockne eingeengt.
Der Rückstand des Kaliumsal- zes der 6- (N-t-Butyl-N'-phthalamido)-penicillansäure wurde abfiltriert, an der Luft und schliesslich im Vakuum über Phosphorpentoxyd getrocknet. Die Substanz, welche noch etwas Feuchtigkeit enthielt, wog 24, 0 g. Sie zeigte einen Smp. von 136 bis 137 unter Zersetzung. Sie enthielt die ss-Lactamstruktur (nach- weisbar durch Infrarot-Analyse) und inhibierte Staph. aureus Smith bei einer Konzentration von 3, 12 mcg/ ml. Die Heilungsdosis CD50 gegenüber Staph. aureus Smith sowie einem Penicillin-G-resistenten Stamm von Staph. aureus bei intramuskulärer Injektion in Mäusen lag bei 27 mcg/kg bzw. 20 mcg/kg.

The non-toxic salts of acids of the formula I which can be produced by the process according to the invention are z. B. salts of sodium, potassium, calcium and aluminum, ammonium salts and substituted ammonium salts, also salts of non-toxic amines, such as the lower trialkylamines including triethylamine, procaine, dibenzylamine, N benzyl-phenethylandn, l-ephenamine, N , N'-Dibenzyl-ethylenediamine, dehydroabiethylamine, N, N'-bis dehydroabietyl-ethylenediamine, lower alkylpiperidines, as they have been used for the formation of salts of Ben zylpenicillins.
The alkyl radicals can be straight-chain or branched and preferably contain 1 to 12 carbon atoms. The term lower alkyl groups refers, as usual, to alkyl groups having 1 to 6 carbon atoms.
Of particular interest are those penicillins of the formula I in which R 1 is an allyl radical or a lower alkyl group, in particular n-propyl, isopropyl, n-amyl, n-butyl and tertiary butyl. Corresponding esters can be prepared from the penicillins obtained according to the invention, which esters can be converted into the free acid by chemical or enzymatic hydrolysis.
In the process according to the invention, it is advantageous to start from a neutral salt of 6-amino-penicillanic acid.
The acylating agent used is advantageously an acid halide, in particular an acid chloride of the formula

The acylating agents to be reacted with 6-amino-penicillanic acid according to the invention are derived from N-substituted phthalamic acid. These phthalamic acid derivatives can easily be prepared by methods described in the technical literature, one of which is given in Example 1 below. The phthalic anhydride required for this preparation and some of the amines used for this are commercially available or can easily be prepared by known preparative methods.
An advantageous procedure for the inventive preparation of compounds of the formula I using a mixed anhydride with ethoxy- or isobutoxycarboxylic acid is that a phthalamic acid of the formula given above with z. B. isobutyl chloroformate and a tertiary amine, such as triethylamine, in an anhydrous, inert and preferably water-miscible solvent such as p-dioxane and optionally with a small amount of dry acetone, for about 30 minutes at about 4 is mixed. To dissolve the resulting mixed anhydride, a cooled solution of 6-amino-penicillanic acid and a tertiary amine, e.g. B. triethylamine, added in a solvent such as water.
The reaction mixture is stirred for about 1 hour, the substituted ammonium salt of the desired end product being formed. The mixture is then favorably at an alkaline pH with a water-immiscible solvent, such as. B. Ather, extracted to remove unreacted from output products. Some of the substances prepared according to the invention are relatively unstable compounds and are subject to chemical changes which lead to a reduction in their antibacterial activity.
For this reason, such mild reaction conditions are advantageously used in the production process that decomposition of the substances does not occur. The reaction conditions to be selected naturally depend to a large extent on the reactivity of the compounds to be converted. In most cases it is necessary to make a compromise between the users. The use of very mild conditions for an extended period of time or the use of more vigorous reaction conditions for a shorter period of time with the possibility of some of the antibiotic substance decomposing.
The temperature to be maintained for the preparation of derivatives of 6-aminopenicillanic acid should generally not exceed 30. In some cases room temperature proves to be suitable. As a rule, strongly acidic or strongly alkaline reaction conditions should, if possible, be avoided in the process according to the invention. The reaction is therefore preferably carried out in the pH range between 6 and 9, which can be achieved in the usual way by using a buffer, e.g. B. a solution of sodium bicarbonate or a sodium phosphate buffer can be achieved.
In addition to the use of aqueous reaction media, including filtered fermentation broths, or aqueous solutions, crude 6-amino no-penicillanic acid, organic solvents can also be used, such as. B. dimethylformamide, dimethylacetamide, chloroform, acetone, methyl isobutyl ketone and dioxane. It often proves to be particularly advantageous to add an aqueous solution of a salt of 6-aminopenicillanic acid to a solution of the acylating agent in an inert and preferably water-miscible solvent, such as acetone or dimethylformamide. Vigorous stirring is naturally advantageous when more than one phase, e.g. B. solid and liquid or two liquid phases are present.
The products prepared according to the invention can be isolated using those techniques as are known for benzylpenicillin and phenoxymethylpenicillin. For example, the product can be extracted with diethyl ether or n-butanol at acidic pH and then deposited by lyophilization, or it can be converted into a salt that is not soluble in the solvent, e.g. B. by means of neutralization with a solution of potassium 2-ethylhexanoate in n-butanol. The product can also be precipitated from aqueous solution as a water-insoluble salt of an amine or obtained directly by means of lyophilization, which is preferably done in the form of a sodium or potassium salt.
If the triethylamine salt has been obtained, the product can be converted into the free acid and then converted into another salt, as is known in the production process of benzylpenicillin and other penicillins. Thus, the treatment of such a triethylamine salt in water with sodium hydroxide gives the sodium salt, after which the triethylamine by means of extraction, eg. B. with toluene can be removed. The treatment of the sodium salt with strong aqueous acid then gives the free acid of the compound, which in turn is converted into other amine salts, e.g. B. can be converted into the procaine salt by reaction with the base in question.
Salts obtained in this way are advantageously separated off by lyophilization or, if they are insoluble, by filtration. A way of working to isolate the he invention. according to the product prepared as a crystalline potassium salt consists in that the product is extracted from an acidic aqueous solution (e.g. pH 2) with diethyl ether, whereupon the ether extract is dried and at least one equivalent of a solution of potassium 2-ethylhexanoate in dry n butanol is added. The potassium salt precipitates, mostly in crystalline form, and can be filtered off or obtained by decantation.
Example I a) 74.0 g (0.5 mol) of phthalic anhydride and 300 ml of benzene were mixed in a 1 liter three-necked round bottom flask. The mixture was stirred for 20 minutes and then a mixture of 0.5 moles (28.5 g) of allylamine in 100 ml of benzene was added dropwise over half an hour.
The resulting mixture was stirred for an additional 20 minutes at room temperature. The reaction mixture was then placed on a water bath and refluxed for one hour. The mixture was allowed to cool to room temperature and then placed on an ice bath for about 20 minutes, whereupon the reaction product precipitated. The N-allyl-phthalamic acid was filtered off, dried overnight in a desiccator and weighed 92.4 g. She had a m.p. from 115 to 117. The product obtained after recrystallization from acetone weighed 60.2 g and had a melting point of 115 to 117.

Following similar procedures with acetone as the solvent, N-benzyl-phthalamic acid, N-n-propyl-phthalamic acid and other acids were prepared, the reaction conditions being slightly varied depending on the properties of the various amines used. b) 10 ml of ethyl chloroformate were added dropwise with stirring at approximately -5 ° C. to a solution of 20.52 g (0.1 mol) of N-allyl phthalamic acid, 14 ml of triethylamine, 70 ml of p-dioxane and 30 ml of dry acetic acid tone added.
After the mixture had been stirred at -5 for 15 minutes, a solution of 21.6 g (0.1 mol) of 6-amino-penicillanic acid in 50 ml of water, which had previously been cooled to 0 C, and 15 ml of triethylamine added in one pour. The reaction mixture was stirred vigorously at 0 for 2 hours. It was then diluted with an equal volume of water and extracted three times with methyl isobutyl ketone, the extracts being discarded. The cold aqueous solution was covered with a layer of methyl isobutyl ketone and acidified to pH 2 with 42% phosphoric acid.
The acidified solution was extracted in 3 portions with 700 ml of methyl isoW butyl ketone. The methyl isobutyl ketone extracts, which contained 6- (N-allyl-N'-phthalamido) -penicillanic acid, were washed once with water, dried with sodium sulfate, filtered and with a solution of potassium Treated -2-ethyl hexanoate in ether. The solvent was decanted off and the remaining product was covered with 400 ml of acetone. The resulting potassium salt of 6- (N-allyl-N'-phthalamido) -penicillanic acid was filtered off, dried in vacuo over phosphorus pentoxide and then weighed 20.0 g.
It contained the, B-lactam structure as shown by infrared analysis and inhibited Staph. aureus Smith at a concentration of 0.8 mcg / ml. When injected intramuscularly into mice, the healing dose CD- ,, was 6.8 mcg / kg.
Example 2
13.9 g = 0.1 mol of triethylamine were added in one portion to a suspension of 25.5 g (0.1 mol) of N benzylphthalamic acid in 150 ml of tetrahydrofuran, a clear solution being formed.
The solution was cooled to about -5¯ and 13.7g (0.1 mole) of isobutyl chloroformate was added dropwise while maintaining the temperature of the mixture at about -5. The resulting reaction mixture was stirred for half an hour and then a cooled solution of 21.6g (0.1 mol) of 6-aminopenicillanic acid in 40 ml of water and 15 ml of triethylamine was added in one portion. The reaction mixture was stirred for half an hour at -5 in a cold bath and then for a further 2 hours at room temperature. The mixture was then diluted with an equal volume of water and extracted twice with methyl isow butyl ketone, the extracts being discarded.
The aqueous solution was covered with a layer of methyl isobutyl ketone, cooled and acidified to pH 2 with 42.5% phosphoric acid. The acidified aqueous solution containing the 6- (N-benzyl N'-phthalamido) -penicillanic acid was extracted twice with methyl isobutyl ketone, after which the combined extracts were washed with cooled water, filtered through sodium sulfate and dried over sodium sulfate were. The sodium sulfate was separated from the extracts and 33.2 ml of a 50% solution of sodium 2-ethylhexanoate were added to the latter, whereupon a precipitate formed.
The precipitated sodium salt of 6- (N-benzyl-N'-phthalamido) -penicillanic acid was filtered off, suspended in acetone, filtered again, dried in air and finally in vacuo over phosphorus pentoxide. The product obtained weighed 11.8 g. It contained the jS-lactam structure, detected by infrared analysis and inhibited Staph. aureus Smith at a concentration of 0.5 mcg / ml. When injected intramuscularly into mice, the: healing dose CD50 was 17 mcg / kg.

Example 3
13.9 g = 0.1 mol of triethylamine were added in one portion to a suspension of 20.7 g (0.1 mol) of N-n-propyl-phthalamic acid in 100 ml of tetrahydrofuran, a clear solution being formed. The solution was cooled in an ice, salt and acetone bath and 13.7 (0.1 mol) isobutyl chloroformate were added dropwise, the temperature of the solution being kept at the temperature of the cooling bath. The reaction mixture obtained was stirred for half an hour and then added in one portion to a cooled solution of 21.6 g (0.1 mol) of 6-aminopenicillanic acid in 40 ml of water and 15 ml of triethylamine.
The resulting reaction mixture was removed from the cooling bath, stirred for 2 t / 2 hours at room temperature and then diluted with an equal volume of water. It was now extracted twice with methyl isobutyl ketone, the extracts being discarded. The aqueous solution was covered with a layer of methyl isobutyl ketone, cooled and acidified to pH 2 with 42.5% phosphoric acid. The acidified aqueous solution, which contained the 6- (Nn-propyl-N'-phthalamido) -penicillanic acid, was extracted twice with methyl isobutyl ketone, whereupon the combined extracts were washed with cooled water, filtered through sodium sulfate and over Sodium sulfate were dried.
The sodium sulfate was separated from the extract and 40 ml of a 50 igen solution of potassium 2-ethylene hexanoate in n-butanol was added to the latter, whereupon a precipitation occurred. The precipitated potassium salt of the 6- (Nn-propyl-N'- phthalamido) penicillanic acid was filtered off, suspended in acetone, filtered again, in air and finally dried over phosphorus pentoxide in vacuo. The product obtained weighed 22.5 g.
It showed a ß-lactam structure by infrared analysis. It inhibited staph. aureus Smith at a concentration of 1.6 mcg / ml.

Example 4
13.9 g = 0.1 mol of triethylamine were added in one portion to a suspension of 23.5 g (0.1 mol) of N n-amylphthalamic acid in 150 ml of tetrahydrofuran, a clear solution being formed. The solution was cooled to about -5 and 13.6 g (0.1 mol) of isobutyl chloroformate in 20 ml of tetrahydrofuran was added dropwise while maintaining the temperature of the solution at about -5. The resulting reaction mixture was stirred for half an hour at about 0 and then added in one portion to a cooled solution of 21.6 g (0.1 mol) of 6-amino penicilanic acid in 60 ml of water and 15 ml of triethylamine.

The product obtained weighed 28.0 g and decomposed at 140-145 on heating. It contained a p-lactam structure detected by infrared analysis. It inhibited staph. aureus Smith at a concentration of 0.8 mcg / ml.

The potassium salt of 6- [N- (1, 2, 5, 6-tetrahydropyridyl) -N'-phthaIamido] -penicillanic acid with a weight of 37.5 g was obtained. It decomposed above 140 when heated. The solid potassium 6- (N, N-totramethylene-N'-phthalamido) penicillinate weighed 21.0 g after drying in vacuo over phosphorus pentoxide. It melted with decomposition at 150 and contained the β-lactam structure, as could be demonstrated by infrared analysis. The methyl isobutyl ketone extracts, which contained 6- [N- (2-phenylethyl) -N'-phthalamido] -penicillanic acid, were dried with sodium sulfate, filtered and treated with a solution of potassium-2- Treated ethyl hexanoate in n butanol. The acidified aqueous solution, which contained 6- (N-isopropyl-N'-phthalamido) -penicillanic acid, was extracted twice with methyl isobutyl ketone, after which the combined extracts were washed with cooled water, filtered through sodium sulfate and over sodium sulfate were dried.
The sodium sulfate was separated from the extracts and 40 ml of a 50 ml solution of potassium 2-ethylhexanoate in n-butanol were added to the latter. The mixture was then concentrated to dryness in vacuo and n-pentane was added to the residue. The resulting potassium salt of 6- (N isopropyl-N'-phthalamido) -penicillanic acid was filtered off, in air and then dried in vacuo over phosphorus pentoxide. The still slightly moist product weighed 15.3 g. It contained the p-lactam structure, as could be demonstrated by infrared analysis. It inhibited staph. aureus Smith at a concentration of 3.12 mcg / ml.

Example 11
The procedure of Example 2 was followed, the N-benzyl-phthalamic acid being replaced by 0.1 mol of N- (a-methylbenzyl) -phthalamic acid.
The sodium salt of 6- (N-α-methylbenzyl-N'-phthalamido) penicillanic acid with a weight of 11.0 g was obtained. It contained the lactam structure as could be demonstrated by infrared analysis. The substance inhibited Staph. aureus Smith at 1.6 mcg / ml. Opposite Staph. aureus, Smith, the healing dose CDo for intramuscular injection in mice was found to be 25 mcg! kg.

Example 12
According to Example 5, replacing N-furfuryl-phthalamic acid with 0.1 mol of N, N-hexamethylene-phthalamic acid, the potassium salt of 6- (N-hexamethylene-N'-phthalamido) -penicillanic acid weighing 40.5 g was obtained. The substance contained the ß-lactam structure as could be demonstrated by infrared analysis. She inhibited Staph. aureus Smith at a concentration of 1.6 mcg / ml.

According to Example 5, the potassium salt of 6 (N-tetrahydrofurfuryl-N'-phthalamido) penicillanic acid was obtained by replacing the N-furfuryl-phthalamic acid with 0.1 mol of N-tetrahydrofurfuryl-phthalamic acid. It weighed 33.0 g and contained the ss-lactam structure, as could be demonstrated by infrared analysis. The substance inhibited Staph. aureus Smith at 1.6 mcg / ml.
Example 14
According to Example 1, the potassium salt of 6- (N-morpholino-N'-phthalamido) penicillanic acid weighing 25.0 g was obtained by replacing the N-furfuryl-phthalamic acid with 0.1 mol of N-morpholinophthalamic acid. The subs. Tanz contained the SS-lactam structure, as could be demonstrated by infrared analysis. It decomposed at 165¯ on heating and inhibited staph. aureus Smith at 3.12 mcg / ml.
Example 15
According to Example 5, 0.1 mol of N- (1, 2, 3, 4-tetrahydroquinaldino) phthalamic acid was used instead of N-furfurylphthalamic acid. The potassium salt of 6- [o- (2'-methyl-1 ', 2', 3 ', 4' tetrahydroquinolylcarbonyl) -benzamido] -penicillanic acid with a weight of 43.5 g was obtained.
The substance contained the ß-lactam ring as could be detected by infrared analysis and decomposed when heated at 150. She inhibited Staph. aureus Smith at 1.6 mcg / ml. When injected intramuscularly into mice, the healing dose was CDgo versus Staph. aureus Smith at 45 mcg / kg.
Analysis calculated for CAN, O, SK: C = 58.75 "/ o, H = 4.89 / o. Found: C = 58.15 / o, H = 5.52%.
Example 16
13.9 g (0.1 mol) of triethylamine were added in one portion to a suspension of 24.5 (0.1 mol) of N, N-diallyl-phthalamic acid in 100 ml of tetrahydrofuran, a solution being formed. The solution was on un. Cooled at about -5 and 13.6 (0.1 mol) isobutyl chloroformate in 20 ml tetrahydrofuran added dropwise, the temperature of the solution being kept at about -5.
The resulting reaction mixture was stirred at about 0 for 1/2 hour, after which a cooled solution of 21.6 g (0.1 mol) of 6 aminopenicillanic acid in 60 ml of water and 15 ml of triethylamine was added in one portion. The mixture obtained was stirred for half an hour in the cold bath and then for a further 2 hours at room temperature. The reaction mixture was diluted with an equal volume of water and extracted twice with methyl isobutyl ketone, the extracts being discarded. The aqueous solution was covered with a layer of methyl isobutyl ketone, cooled and acidified to pH 2 with 42.5% phosphoric acid.
The acidified aqueous solution, which contained the 6- (N, N-diallyl-N'-phthalamido) -penicillanic acid, was extracted twice with methyl isobutyl ketone, after which the combined extracts were washed with cooled water, filtered through sodium sulfate and over sodium sulfate were dried.
The sodium sulfate was separated off and the extract was treated with 40 ml of a 50% solution of potassium 2-ethylhexanoate in n-butanol, after which the solution was concentrated to a low volume and diluted with ether. This produced a precipitate.
The precipitated potassium salt of 6- (N, N-diallyl-N 'phthalamido) -penicillanic acid was filtered off, suspended in acetone, filtered again, dried in air and finally in vacuo over phosphorus pentoxide. The product thus obtained weighed 40.5 g.
It contained the ß-lactam structure as could be demonstrated by infrared analysis. It inhibited staph. aureus Smith at a concentration of 0.8 mcg / ml. The cure dose CDvo to Staph. aureus Smith when injected intramuscularly in mice was 4 mcg / kg.
Example 17
The procedure was as in Example 16, the N, N-diallylphthalamic acid being replaced by 0.1 mol of N, N-diisopropyl-phthalamic acid or N, N-diethyl-phthalamic acid or N, N-dimethyl-phthalamic acid. The potassium salts of 6- (N, N-diisopropyl-N'-phthalamido) -penicillanic acid, 6- (N, N-diethyl-N'-phthalamido) -penicillanic acid and 6- (N, N -Dimethyl-N'-phthalamido3-penicillanic acid. All these salts were isolated in solid form. According to infrared analysis, they all had an ß-lactam ring.
They inhibited Staph. aureus Smith at concentrations of 0.001 wt / o.
Example 18
According to Example 4, replacing the N- (n-amyl) -phthalamic acid with 0.1 mol of N-dodecylphthalamic acid, the water-soluble potassium salt of 6- (N-dodecyl-N'-phthalamido) -penicillanic acid weighing 10.5 g was obtained. The substance contained the P-lactam ring (detectable by infrared analysis) and inhibited Staph. aureus Smith at 0.4 mcg / ml.
Example 19
13.9 g (0.1 mol) of triethylamine were added in one portion to a suspension of 22.1 g (0.1 mol) of N-t-butyl phthalamic acid in 100 ml of tetrahydrofuran, a homogeneous solution being formed.
The solution was cooled to about -5 and 13.7 g (0.1 mol) of chloroformate in 20 ml of tetrahydrofuran was added thereto dropwise over 20 minutes while maintaining the temperature at about -10. The resulting reaction mixture was stirred for 1/2 hour at about 0 C and then a cooled solution of 21.6 g (0.1 mol) of 6 aminopenicillanic acid in 50 ml of water and 15 ml of triethylamine was added in one portion . The mixture obtained was stirred for 11/2 hours at a temperature of -5 to +5 and then at room temperature until the evolution of CO2 had ceased.
The reaction mixture was diluted with an equal weight of water and extracted twice with methyl isobutyl ketone, the extracts being discarded. The aqueous solution was covered with a layer of methyl isobutyl ketone, cooled and acidified to pH 2 with 42.5% phosphoric acid.
The acidified aqueous solution, which contained 6- (Nt-butyl-N'-phthalamido) -penicillanic acid, was extracted twice with methyl isobutyl ketone, whereupon the combined extracts were washed with cooled water, filtered through sodium sulfate and over sodium sulfate were dried. The sodium sulphate was separated from the extracts and 45 ml of a 40% solution of potassium 2-ethylhexanoate in n-butanol were added to the latter, a precipitate separating out. The mixture was concentrated to dryness in vacuo.
The residue of the potassium salt of 6- (N-t-butyl-N'-phthalamido) -penicillanic acid was filtered off, dried in air and finally in vacuo over phosphorus pentoxide. The substance, which still contained some moisture, weighed 24.0 g. It showed a m.p. from 136 to 137 with decomposition. It contained the ß-lactam structure (detectable by infrared analysis) and inhibited Staph. aureus Smith at a concentration of 3.12 mcg / ml. The CD50 healing dose versus Staph. aureus Smith and a penicillin G resistant strain of Staph. aureus when injected intramuscularly into mice was 27 mcg / kg and 20 mcg / kg, respectively.

Using methods analogous to those described in the preceding examples, new penicillins were prepared by the process according to the invention, where R2 in formula I was hydrogen and R3 represented the following radicals:
