Verfahren zur Herstellung von Phenyläthanolaminderivaten
Es ist bekannt, dass 3,5-Dioxy-phenyl-alkanolamine eine mit dem Adrenalin vergleichbare physiologische Wirkung zeigen, dass sie aber gegenüber diesem den Vorteil besitzen, dass sie wesentlich beständiger und daher, im Gegensatz zu Adrenalin, peroral anwendbar sind.
Die Herstellung dieser Verbindungen gelang bisher durch tÇberführung der entsprechenden, am l-C-Atom gegebenenfalls durch Alkylreste substituierten 3,5-Dioxy-co-halogen-acetophenone, deren OH-Gruppen gegebenenfalls geschützt sind, in Aminoketone nach an sich bekannten Methoden und anschliessende Reduktion der Aminoketone zu den entsprechenden Aminoalkoholen. Unter den bekannten Methoden fanden vor allem die Umsetzung der Halogenketone mit Ammoniak bzw. Aminen und, im Falle der Herstellung von Phenyläthanolaminen mit freier Aminogruppe, die Umsetzung der Halogenketone mit Natriumazid und anschliessende, gleichzeitige Hydrienmg der Azidogruppe und der Ketogruppe praktische Anwendung.
Die Herstellung der 3,5-Dioxy-phenylalkanolamine, ausgehend von den o-Halogenketonen, bringt jedoch einige schwerwiegende Nachteile mit sich.
Allein die oj-Halogenketone selbst sind äusserst unangenehm zu handhaben, da sie die Schleimhäute, besonders die der Augen, sehr stark reizen. Ferner sind die Ausbeuten bei der Umsetzung mit Ammoniak bzw. Aminen sehr schlecht. Wählt man aber das Verfahren über die Azidoacetophenone, so sind die Ausbeuten zwar wesentlich besser, doch besitzen die Azidoacetophenone wiederum die unangenehme Eigenschaft, dass sie sehr explosionsfreudig sind. So detonieren Dioxyazidoacetophenone bereits bei Temperaturen unter 150 C, wenn sie im Röhrchen erhitzt werden, während beispielsweise Natriumazid unter den gleichen Bedingungen bis über 400 C erhitzt werden kann, ohne zu detonieren.
Es ist klar, dass die Handhabung einer solchen Substanz, besonders in grösseren Mengen, eine erhebliche Gefahrenquelle darstellt.
Gegenstand der vorliegenden Erfindung ist ein neues Verfahren zur Herstellung von Phenyläthanolaminen mit unsubstituierter Aminogruppe, das wesentlich höhere Ausbeuten liefert, und bei dem das Arbeiten mit Substanzen, die die Augen oder andere Schleimhäute reizen, oder die explosions freudig sind, vermieden wird.
Es konnte nämlich gefunden werden, dass sich Phenyläthanolamine der Formel I :

kondensiert und diese Nitroalkanole anschliessend, vorzugsweise katalytisch, reduziert, wobei gleichzeitig oder anschliessend gegebenenfalls vorhandene Aralk oxygruppen in die freien Oxygruppen übergeführt werden.
Die Herstellung von Nitroalkanolen durch Kondensation von Aldehyden mit Nitroalkanen ist im Prinzip bekannt. Es ist dabei aber einem Fachmann geläufig, dass diese Kondensation in geeigneten Lösungsmitteln, vor allem in niederen Alkoholen, durchgeführt wird, und dass dabei nur dann gute Ausbeuten erzielt werden können, wenn der als Ausgangsprodukt gewählte Aldehyd bei 0 C in diesen, Alkoholen gut löslich ist. Ist dagegen der Aldehyd bei Temperaturen um 0 C nur schwer oder gar nicht in niederen Alkoholen löslich, werden nur sehr schlechte Ausbeuten erzielt, weil die Reaktion dann unter ungünstigen physikalischen Bedingungen durchgeführt werden muss. Der Grund dafür liegt hauptschlich darin, dal3 bei der Kondensation von Aldehyden mit Nitroalkanen in Gegenwart von Alkali, z. B.
Natriummethylat, primär die Alkalisalze der entsprechenden Pseudosäuren gebildet werden und aus dem Reaktionsgemisch ausfallen. Ist nun der Aldehyd nicht leichter im verwendeten Lösungsmittel löslich als das Alkalisalz der gebildeten Pseudosäure, wird er von diesem eingeschlossen und dadurch der weiteren Umsetzung entzogen. Dadurch ist eine nur teilweise Reaktion des Aldehyds möglich. Eine weitere Ausbeuteverminderung muss dann noch in Kauf genommen werden, da die Aufarbeitung eines solchen, nur teilweise ausreagierten Reaktionsgemisches auf Grund der Instabilität der gebildeten Nitroalkanole schwierig und verIustreich ist.
Da die Aldehyde der Formel II in niederen Alkoholen nur äusserst schwer löslich sind, erschien die Anwendung der Nitroalkan-Kondensation zur Herstellung der Phenyl-alkanolamine der Formel I aus den oben geschilderten Gründen von vornherein nicht aussichtsreich. Auch Versuche, andere Lösungsmittel, in denen die Aldehyde der Formel II bei etwa 0 C gut löslich sind, wie beispielsweise Toluol, Benzol oder Essigester, als Reaktionsmedium zu verwenden, waren zum Scheitern verurteilt, da Essigester unter den Reaktionsbedingungen selbst Reaktionen eingeht und bei Reaktion in Gegenwart von festem Alkalialkoholat, das ja in Benzol und Toluol nicht löslich ist, keine guten Ausbeuten erzielt werden konnten. Daher erschien diese Reaktionsweise für den erfindungsgemässen Zweck zunächst nicht anwendbar.
Oberraschenderweise konnte nun gefunden werden, dass die Kondensation der Aldehyde der Formel II mit Nitromethan trotzdem, und zwar sogar in sehr guter Ausbeute möglich ist, wenn man als Reaktionsmedium ein Lösungsmittelgemisch, bestehend aus niederen aliphatischen Alkoholen, aromatischen Kohlenwasserstoffen und Aminen verwendet.
Gegenstand der vorliegenden Erfindung ist demnach ein Verfahren zur Herstellung der Phenyl- äthanolamine der Formel I, das dadurch gekenn- zeichnet ist, dass Aldehyde der Formel II in einem Lösungsmittelgemisch, bestehend aus niederen aliphatischen Alkoholen, aromatischen Kohlenwasserstoffen und Aminen bei Temperaturen von-10 bis +30 C und in Gegenwart von Alkalialkoholaten als Kondensationsmittel mit Nitromethan zu Nitroalkanolen der Formel III kondensiert und diese dann anschliessend reduziert werden, wobei gegebenenfalls vorhandene Aralkoxygruppen gleichzeitig oder anschliessend in die freien Oxygruppen übergeführt werden.
Die so gebildeten Phenyläthanolamine der Formel I können dann aus dem Reaktionsgemisch entweder als freie Basen oder in Form ihrer Salze mit anorganischen oder organischen Säuren gewonnen werden.
Die Durchführung der Nitromethankondensation erfolgt besonders zweckmässig bei Temperaturen zwischen-5 und-F 10 C. Als Komponenten des Lösungsmittelgemisches haben sich alle niederen aliphatischen Alkohole, z. B. Methanol, Athanol, Isopropanol, und die aromatischen Kohlenwasserstoffe Benzol, Toluol und Xylol besonders bewährt.
Als Amine sind sowohl primäre, sekundäre als auch tertiäre Amine einschliesslich stickstoffhaltiger Heterocyclen geeignet. Als Beispiele hierfür können Pyridin, Pyrrol, Anilin, Dimethylanilin, Diäthylamin, Triäthylamin, ss-Dimethylam, inoäthanol, Diäthanol- amin und Triäthanolamin genannt werden. Besonders zweckmässig ist die Verwendung eines Gemisches von Athanol, Benzol und Pyridin, und zwar vorzugsweise in einem Mischungsverhältnis von 5-12 Volumteilen absolutem Benzol und 0,5-1,2 Volumteilen Pyridin auf je 10 Volumteile absolutes Äthanol.
Die anschliessende Reduktion der Nitroalkanole der Formel III zu den Aminoalkanolen der Formel I kann in an sich bekannter Weise erfolgen. Besonders bevorzugt ist eine katalytische Reduktion bei Normaldruck und Raumtemperatur, beispielsweise mit Palladium als Katalysator. Handelt es sich um eine Anwendung des erfindungsgemässen Verfahrens zur Herstellung von Verbindungen der Formel I, die im aromatischen Kern freie Hydroxylgruppen tragen, so wird von Aldehyden ausgegangen, in denen diese Hydroxylgruppen durch Aralkylreste, vorzugsweise Benzylreste, geschützt sind. In einem solchen Falle kann die Abspaltung der schützenden Reste gleich- zeitig mit der Reduktion der Nitrogruppe durch ka talytische Hydrierung erfolgen.
Wird die Nitrogruppe unter Bedingungen reduziert, bei denen die Aralkoxy- gruppen erhalten bleiben, so muss an die Reduktion z. B. eine katalytische Abspaltung der Aralkylgruppen mittels Wasserstoff angeschlossen werden.
Nach Beendigung der katalytischen Reduktion kann das Reaktionsgemisch auf einfache Weise, z. B. durch Abtrennen des Katalysators mittels Filtration und Eindampfen des Filtrates zum Aminoalkohol erfolgen. In manchen Fällen empfiehlt es sich auch, den Aminoalkohol der Formel I gleich in Form seines Salzes mit anorganischen oder organischen Säuren zu isolieren.
Im Fall der Isolierung als Hydrohalogenide hat sich dabei folgende Variante des erfindungsgemässen Verfahrens besonders bewahrt. Die zur Salzbildung nötige Säure wird nicht dem Reaktionsgemisch als solche zugesetzt, sondern durch katalytische Spal- tung von Verbindungen, die unter diesen Bedingungen Halogenwasserstoff tiefem, im Reaktionsgemisch erzeugt. Als solche Substanzen können beispielsweise Benzylchlorid oder Benzylbromid genannt werden. Zur Durchführung wird nach beendeter katalytischer Reduktion der Nitrogruppe dem Reaktionsgemisch die äquivalente Menge Benzylchlorid bzw. Benzylbromid zugesetzt und das Gemisch bis zur beendeten Wasserstoffaufnahme weiterhydriert. Das auf diese Weise gebildete Salz kann dann auf übliche Weise isoliert werden.
Beispiel 1
189,3 g 3,5-Diacetoxy-benzaldehyd, 342,7 g Kaliumbicarbonat, 325,6 g Benzylchlorid und 2570 ml Athanol werden 9 Stunden unter Stickstoff bei Siedetemperatur gerührt. Nach dem Erkalten auf Raumtemperatur lässt man das Reaktionsgemisch über Nacht bei-20 C stehen. Hierauf saugt man ab und wäscht den Niederschlag mit kaltem Athanol. Der Niederschlag wird dann mit 1000 ml Benzol ausgerührt, die Lösung von dem ausgeschiedenen Kaliumchlorid und Kaliumacetat abfiltriert und die Salze mit etwas Benzol ausgewaschen. Das Filtrat wird nun im Vakuum eingeengt und der Rückstand mit Methanol versetzt, wobei sofort ein dicker Kristall- brei entsteht. Nach Abkühlen auf-20 C saugt man die Kristalle ab, wäscht mit Methanol und trocknet das Kristallisat.
Man erhält als erstes Kristallisat 152,9 g (56,4% der Theorie) 3,5-Dibenzyloxy-benzaldehyd vom Schmelzpunkt 80, 0-80,5 C.
Durch Eindampfen der Mutterlauge im Vakuum werden weitere 37,8 g (13,9% der Theorie) Rohprodukt erhalten.
47,5 g 3,5-Dibenzyloxy-benzaldehyd, 212 ml absolutes Athanol, 108 ml absolutes Benzol, 18,5 g absolutes Pyridin und 14,4 g Nitromethan werden bei etwa 0 C gerührt, wobei eine Lösung entsteht. Hierauf wird im Verlauf von rund 1/2 Stunde eine Lösung von 7,1 g Natrium in 108 ml Methanol zugetropft. Nach beendeter Zugabe wird das Gemisch etwa 4 Stunden bei 0 C gehalten. Bereits während der Zugabe beginnt das Natriumsalz der gebildeten Pseudosäure auszufallen. Nach beendeter Reaktionszeit wird das Reaktionsgemisch in einem Gemisch von 54 ml Eisessig und 90 ml absolutem Athanol gelöst. Dabei entsteht eine klare Lösung, welche im Vakuum bei 30-40 C Badtemperatur weitgehend d eingedampft wird.
Der kristallisierte Rückstand, der aus dem gebildeten Nitroalkanol und Natriumacetat' besteht, wird nun in Essigester und Wasser gelöst.
Die Essigesterlösung wird abgetrennt, mit Wasser, Natriumbicarbonatlösung und wieder mehrmals mit Wasser gewaschen und mit Natriumsulfat getrocknet.
Nach Abdampfen des Lösungsmittels erhält man 51, 46 g rohes 1- (3', 5'-Dibenzyloxy-phenyl)-2-nitro- äthanol- (l) (91, 0% der theoretisch möglichen Menge) vom Schmelzpunkt 52-54 C. Zur Reinigung löst man dieses Rohprodukt bei 30 C in absolutem Xylol und kühlt auf-20 C. Nach Absaugen erhält man 45,6 g reines Nitroalkanol, das sind 88,6 % der eingesetzten Rohproduktmenge. Schmelzpunkt 62 bis 65,5 C.
23,0 g 1- (3', 5'-Dibenzyloxy-phenyl)-2-nitro- äthanol- (l) werden mit 230 ml Eisessig und 2,3 g Palladiumkohle (10% Pd) versetzt und mit Wasserstoff reduziert. Nach Stillstand der Wasserstoffaufnahme wird das Reaktionsgemisch mit 7,0 g Benzylchlorid versetzt. Die Hydrierung wird nun fortgesetzt, das Benzylchlorid dabei in Toluol und HC1 gespalten und dadurch das Hydrochlorid des Aminoalkanols gebildet. Nach Verbrauch der für die Spaltung des Benzylchlorids notwendigen Wasser- stoffmenge werden das ausgefallene 1-(3', 5'-Di- hydroxy-phenyl)-2-aminoäthanol- hydrochlorid und der Katalysator durch Filtration isoliert.
Der Niederschlag wird nun mit 100 ml Methanol behandelt, die entstandene Lösung vom Katalysator abfiltriert und letzterer mit etwas Methanol ausgewaschen. Das erhaltene Filtrat wird nun im Vakuum unter Wasserstoff eingedampft, der kristalline Rückstand wird mit 30 ml Isopropylalkohol digeriert und gekühlt und isoliert. Es werden 9,56 g Hydrochlorid (83,9 % der theoretisch möglichen Menge) vom Schmelzpunkt 212-214 C erhalten.
Der als Ausgangsprodukt benötigte 3,5-Diacet oxy-benzaldehyd wird aus 3,5-Diacetoxy-benzoylchlorid durch Reduktion nach Rosenmund erhalten.
Beispiel 2
36,96 g 3,4,5-Triacetoxy-benzaldehyd, 98,70 g Benzylchlorid, 91,2 g Kaliumcarbonat und 658 ml Athanol werden 12 Stunden unter Stickstoff bei Siedetemperatur gerührt. Nach dem Erkalten wird abgesaugt und der Niederschlag mit 300 ml Athanol gewaschen. Dann wird der Niederschlag mit 750 ml Wasser digeriert, wieder abgesaugt, mit Wasser gewaschen und getrocknet. Nach Trocknung im Vakuum werden 49,1 g 3,4,5-Tribenzyloxy-benzaldehyd erhalten, das sind 87,5 der theoretisch möglichen Menge. Zur Reinigung kristallisiert man das Pro- dukt aus 1400 ml Methanol um, wobei 36,41 g Reinprodukt (74,1 % der eingesetzten Menge) vom Schmelzpunkt 101, 5-102,5 C erhalten werden.
15,0 g 3,4,5-Tribenzyloxy-benzaldehyd, 52 mI absolutes Benzol, 52 ml absolutes Athanol, 5,2 g absolutes Pyridin und 3,2 g Nitromethan werden bei 0 C gerührt, bis sich der Aldehyd fast vollständig aufgelöst hat. Dann wird eine Lösung von 1,7 g Natrium in 26 ml Methanol zugetropft und nach der Zugabe die Temperatur 5 Stunden bei etwa 0 C gehalten. Nach dieser Zeit wird eine Lösung von 14,6 ml Eisessig in 20 ml absolutem Alkohol bei 0 C zufliessen gelassen und die erhaltene Lösung im Vakuum eingedampft. Der Abdampfrückstand wird analog Beispiel 1 aufgearbeitet. Es werden 16, 7 g rohes 1- (3', 4', 5'-Tribenzyloxy-phenyl)-2-nitro äthanol- (l) erhalten, das sind 97,3 % der Theorie.
Zur Reinigung wird dieses Rohprodukt in Ather gelöst und diese Lösung langsam unter Rühren in Petroläther einfliessen gelassen. Nach Absaugen, Waschen mit Petroläther und Trocknung werden
12,9 g reines Nitroalkanol vom Schmelzpunkt 93 bis 94,5 C erhalten.
9,2 g 1- (3', 4', 5'-Tribenzyloxy-phenyl)-2-nitro- äthanol- (l), 1,0 g Palladiumkohle (10% Pd), 50 ml Eisessig und 50 ml Methanol werden vermischt und wie üblich bei Normaldruck und Raumtemperatur reduziert. Dabei werden bei 738 Torr und 23 C 2790 ml Wasserstoff verbraucht, ein Wert, der weitgehend mit dem theoretisch zu erwartenden Verbrauch übereinstimmt. Das Reaktionsgemisch wird nun mit 2,1 g Benzylchlorid versetzt und die Hydrierung fortgesetzt.
Nach Aufnahme der theoretisch berechneten Menge Wasserstoff wird vom Kontakt durch Filtration abgetrennt und letzterer mit 150 ml Eisessig/Methanoll 1 : 1 gewaschen. Das Filtrat wird im Vakuum unter Wasserstoff bei 40 C zur Trockne eingedampft. Der kristallisierte Rückstand wird mit 35 ml Isopropylalkohol digeriert, abgesaugt, das Hydrochlorid mit 50 ml gekühltem Isopropylalkohol gewaschen und im Vakuum bei Raumtemperatur über Atznatron getrocknet. Es werden 3,31 g
1- (3', 4', 5'-Trihydroxyphenyl)-2-aminoäthanol- (l)-hydrochlorid erhalten, das sind 90, 0% der theoretisch möglichen Menge. Die Substanz kann zur Reinigung aus Methanol/Isopropylalkohol umkristallisiert werden.
Schmelzpunkt : 164-165,5 C Zersetzung.
Der als Ausgangsmaterial benötigte 3,4,5-Triacetoxybenzaldehyd wird aus 3,4,5-Triacetoxybenzoylchlorid durch Reduktion nach Rosenmund dargestellt.
Beispiel 3
113,16 g 3,4,5-Tribenzyloxybenzaldehyd werden in 2260 ml Eisessig'gelöst, die Lösung mit 33,9 ml 66 S igem Bromwasserstoff versetzt und 21/2 Stunden bei 50 C gehalten. Hierauf wird im Vakuum bei 50 C weitgehend eingeengt. Der Rückstand wird mit 4n NaOH alkalisch gemacht. Das Natriumsalz des 3,5-Dibenzyloxy-4-hydroxy-benzaldehyds kristallisiert aus und wird durch Absaugen isoliert.
Es wird zweimal mit je 100 ml Essigester digeriert, wieder abgesaugt, mit Essigester gewaschen und getrocknet. Es werden 73,51 g Natriumsalz erhalten.
Das erhaltene Natriumsalz wird in 700 ml heissem Methanol gelöst, die Lösung mit 4n HCI I angesäuert, 1 Stunde bei Raumtemperatur und 5 Stunden bei-20 C stehengelassen. Das kristallisierte Produkt wird durch Absaugen isoliert, zweimal mit je 100 ml Wasser digeriert, abgesaugt, mit Wasser gewaschen und getrocknet. Es werden so 44,41 g roher 3,5-Dibenzyloxy-4-hydroxy-benzaldehyd erhalten. Schmelzpunkt 118-120 C. Das Produkt wird aus 750 ml Isopropylalkohol umkristallisiert. Ausbeute 40,23 g vom Schmelzpunkt 120-122 C.
2,0 g Kaliumhydroxyd werden in 110 ml Athanol gelöst. In dieser Lösung werden 8,9 g 3,5-Dibenzyloxy-4-hydroxybenzaldehyd aufgelöst, die Lösung mit 5,2 g Methyljodid versetzt und das Reaktionsgemisch 3 Stunden bei Siedetemperatur gerührt. Nach dem Erkalten saugt man ab, wäscht den Niederschlag mit etwas gekühltem Athanol und verreibt ihn zweimal mit Wasser. Nach Absaugen und Waschen mit Wasser wird das Rohprodukt getrocknet. Es werden 6, 9 g 3,5-Dibenzyloxy-4-methoxybenzaldehyd erhalten, das sind 74,4% der theoretisch möglichen Menge.
Zur Reinigung wird das Rohprodukt aus Isopropyl- alkohol umkristallisiert. Schmelzpunkt 86-87,5 C.
10,0 g 3,5-Dibenzyloxy-4-methoxy-benzaldehyd, 40 ml absolutes Athanol, 30 ml absolutes Benzol, 3,5 g Pyridin und 2,7 g Nitromethan werden bei etwa 0 C gerührt und langsam eine Lösung von 1,35 g Natrium in 25 ml Methanol zugetropft. Nach der Zugabe wird 7 Stunden bei 0 C gehalten. Dann wird das Reaktionsgemisch mit einer gekühlten Lösung von 10,5 ml Eisessig in 20 ml absolutem Athanol angesäuert und im Vakuum bei 30-40 C eingeengt. Die weitere Aufarbeitung erfolgt wie in Beispiel 1. Es werden 10,84 g (92SE der theoretisch möglichen Menge) kristallisiertes rohes 1- (3', 5'-Di- benzyloxy-4'-methoxyphenyl)-2-nitroäthanol- (l) erhalten.
Schmelzpunkt 132-135 C.
3,97 g des oben beschriebenen Nitroalkanols werden in 50 ml Eisessig in Gegenwart von 0,4 g Palladiumkohle (10 Sig) reduziert. Es werden bei 735 Torr und 25 C 1230 ml Wasserstoff verbraucht (theoretischer H2-Verbrauch 1229 ml). Dann werden 1,1 g Benzylchlorid zugesetzt und wieder aushydriert. Der Kontakt wird durch Filtration abgetrennt und mit Methanol ausgewaschen. Das Filtrat wird im Vakuum bei 40 C unter Wasserstoff eingedampft. Der kristallisierte Rückstand (2,35 g) wird mit 10 ml Isopropylalkohol digeriert.
Nach Absaugen, Waschen mit 10 ml kaltem Isopropylalkohol und Trocknen im Vakuum werden 1,68 g des
1- (3', 5'^Dihydroxy-4'-methoxy-phenyl)
2-amino-äthanol- (l)-hydrochlorids (82,1 % der Theorie) erhalten. Das Produkt kann aus Methanol/Aceton oder Methanol/Ather umkristalli- siert werden. Schmelzpunkt 214-215 C.
Beispiel 4
Zur Herstellung des 3-Methoxy-4,5-dibenzyloxy benzaldehyds wird der 3-Methoxy-4, 5-diacetoxybenzaldehyd analog Beispiel 2 oder 3 in alkoholischer Lösung in Gegenwart von Kaliumcarbonat mit Benzylchlorid umgesetzt. Man erhält in 55 % iger Ausbeute den 3-Methoxy-4,5-dibenzyloxy-benzaldehyd vom Schmelzpunkt o, 2 : 217-220 C.
12,4 g des Aldehyds, 50 ml absolutes Athanol,
37,5 ml absolutes Benzol, 4,4 g Pyridin und 3,4 g Nitromethan werden bei 0 C gerührt und eine Lö- sung von 1,6 g Natrium in 31 ml Methanol zuge- tropft. Nach insgesamt 5stündigem Rühren bei 0 C wird mit einer gekühlten Lösung von 13 ml Eisessig in 25 ml absolutem Athanol angesäuert und die erhaltene Lösung bei 30-40 C im Vakuum einge dampft. Die Aufarbeitung erfolgt analog Beispiel l.
Es werden so 14,1 g rohes 1-(3'-Methoxy-4', 5'-di- benzyloxy-phenyl)-2-nitro-äthanol- (l) (97 % der Theorie) erhalten, das nach Umkristallisieren aus, Aceton-Xylol-Petroläther 1 : 2 : 2 bei 104-107 C schmilzt.
8,15 g obigen Nitroalkanols werden in 100 ml Eisessig in Gegenwart von 0, 8 g Palladiumkohle (10% Pd) bei Normaldruck und Raumtemperatur reduziert. Wenn die entsprechende Menge Wasserstoff verbraucht worden und die Wasserstoffaufnahme beendet ist, wird das Reaktionsgemisch mit 2,11 g Benzylchlorid versetzt und die Hydrierung so lange fortgesetzt, bis kein Wasserstoff mehr verbraucht wird. Dann wird vom Katalysator abgesaugt und dieser mit 20 ml Eisessig gewaschen. Das Filtrat wird im Vakuum unter Wasserstoff bei 30-40 C' eingedampft.
Der zunächst ölige Abdampfrückstand kristallisiert beim Digerieren mit Isopropylalkohol, und es können 60% der theoretisch möglichen Menge an reinem
1'- (3'-Methoxy-4', 5'dihydóxy-phenyl)
2-aminoäthanol- hydrochlbrid isoliert werden. Schmelzpunkt 157-157,5 C Zersetzung. Der 3-Methoxy-4,5-diacetoxy-benzaldehyd wird aus dem 3-Methoxy-4, 5-diacetoxy-benzoyl- chlorid durch Reduktion nach Rosenmund gewonnen.
Beispiel 5
3-Athoxy-4, 5-diacetoxy-benzaldehyd wird analog zu den Beispielen 1-3 in äthylalkoholischer Lösung in Gegenwart von Kaliumbicarbonat oder Kaliumcarbonat mit Benzylchlorid umgesetzt, und man erhält analog zu Beispiel 4 den 3-Athoxy-4,5-dibenzyl oxy-benzaldehyd vom Schmelzpunkt 0, 2 220-223 C.
Analog zu den andern Beispielen werden 18,8 g dieses Aldehyds, 72 ml absoluter Alkohol, 54 ml absolutes Benzol, 6,38 g Pyridin und 4,9 g Nitromethan gerührt und bei einer Temperatur von 0 bis 5 C eine Lösung von 2,3 g Natrium in 45 ml Methanol zufliessen gelassen. Anschliessend wird 7 Stunden bei dieser Temperatur gerührt, das Reaktions- gemisch hierauf mit einer gekühlten Lösung von 25 ml Eisessig in 36 ml absolutem Alkohol angesäuert und wie bei den andern Beispiel aufgearbeitet. Man erhält in 93 % iger Ausbeute rohes
1- (3'-Athoxy-4', 5'-dibenzyloxy-phenyl)-
2-nitroäthanol- (1), welches nach Umkristallisieren aus 50 ml Xylol bei 109-112 C schmilzt.
Bei katalytischer Reduktion dieses 1-(3'-Athoxy- 4', 5'-dibenzyloxy-phenyl)-2-nitroäthanol- (l) analog den andern Beispielen und Reduktion in Gegenwart von Benzylchl'orid erhält man 1- (3'-Athoxy-4', 5'-di- hydroxy-phenyl)-2-aminoäthanol- (l)-hydrochlorid vom Schmelzpunkt 174 C Zersetzung. Ausbeute : 61,8 %.
Beispiel 6
7,0 g 3,4,5-Tribenzyloxy-benzaldehyd, 26 ml Toluol, 26 ml Methanol, 2,0 g Triäthylamin und 1,6 g Nitromethan werden bei 0 C gerührt und analog Beispiel 2 mit einer Lösung von 0, 85 g Natrium in 13 ml Methanol umgesetzt. Nach 5stündigem Rühren bei etwa 0 C wird das Reaktions- gemisch mit einer Lösung von 7,5 ml Eisessig in 10 ml Methanol angesäuert und wie in Beispiel 2 aufgearbeitet. Es werden 8,0 g kristallisiertes, rohes l- (3', 4', 5'-Trlbenzyloxy-pheny13-2-nitroäthanol-(1) (100% der Theorie) erhalten. Nach Umkristallisieren aus 70 ml absolutem Xylol erhält man 6,02 g Reinprodukt mit einem Schmelzpunkt von 92-94 C.
Die Reduktion zum Aminoalkanol erfolgt wie in Beispiel 2.
Beispiel 7
7,0 g 3,4,5-Tribenzyloxy-benzaldehyd, 26 ml Benzol, 26 ml Methanol, 2,9 g Triäthanolamin und 1,6 g Nitromethan werden analog Beispiel 2 mit einer Lösung von 0, 85 g Natrium in 13 ml Methanol umgesetzt. Nach 5stündigem Rühren bei etwa 0 C wird das Reaktionsgemisch mit einer Lösung von 7,5 ml Eisessig und 10 ml Methanol angesäuert und wie in Beispiel 2 beschrieben aufgearbeitet. Es werden 7,85 g kristallisiertes, rohes l- (3', 4', 5'-Tri- benzyloxy-phenyl)-2-nitroäthanol- (1) (98,1% der Theorie) erhalten. Nach Umkristallisieren aus 70 mi Xylol werden 5,51 g Reinprodukt vom Schmelz- punkt 93-95 C erhalten.
Die weitere Verarbeitung zum 1- (3', 4', 5'-Trihydroxy-phenyl)-2-aminoäthanol- hydrochlorid erfolgt wie in Beispiel 2.
Beispiel 8
5,7 g 3,4,5-Tribenzyloxy-benzaldehyd, 22 ml absolutes Xylol, 22 ml absolutes Athanol, 1,63 g Di äthanolamin und 1,3 g Nitromethan werden analog Beispiel 2 mit einer Lösung von 0,85 g Natrium in 13 ml Methanol umgesetzt. Das Reaktionsgemisch wird weiter nach Beispiel 2 bearbeitet. Es werden 6,55 g öliges, rohes 1- (3', 4', 5'-Tribenzyloxy-phenyl)- 2-nitroäthanol (1) erhalten. Durch Umkristallisieren desselben aus 56 ml Xylol werden 4,66 g (84,4% der Theorie) Reinprodukt erhalten. Die weitere Verarbeitung zum Aminoalkanol erfolgt wie in Beispiel 2.
Beispiel 9
7,0 g 3,4,5-Tribenzyloxy-benzaldehyd, 26 ml Toluol, 26 ml absoluter Alkohol, 1, 6 g Nitromethan und 2,1 g Dimethylanilin werden analog Beispiel 2 mit einer Lösung von 0,85 g Natrium in 13 ml Methanol umgesetzt. Weitere Aufarbeitung des Reaktionsgemisches wie in Beispiel 2. Es werden 9,2 g öliges, rohes 1- (3', 4', 5'-Tribenzyloxy-phenyl)-2-nitro- äthanol- (l) erhalten, aus dem nach Umkristallisieren aus 80 ml Xylol 5,75 g Reinprodukt mit einem Schmelzpunkt von 93-95 C erhalten werden. Die weitere Verabeitung zum Aminoalkanol erfolgt wie in Beispiel 2.
Beispiel 10
7,0 g 3,4,5-Tribenzyloxy-benzaldehyd, 26 ml absolutes Xylol, 26 ml Isopropylalkohol, 1,6 g Nitromethan und 1,8 g Anilin werden wie in Beispiel 2 mit einer Lösung von 0,85 g Natrium in 13 ml Methanol umgesetzt. Weitere Aufarbeitung wie in Beispiel 2. Es werden 11,2 g teilweise kristallisiertes 1- (3', 4', 5'-Tribenzyloxy-phenyl)-2-nitroäthanol- (l) (xylolhaltig) isoliert. Nach Umkristallisieren aus 70 ml Xylol werden 6,25 g Reinprodukt (78,1 % der Theorie) mit dem Schmelzpunkt 93-95 C erhalten.
Beispiel 11
99,5 g 3,4-Diacetoxy-5-isopropoxy-benzaldehyd, 199,4 g Benzylchlorid, 162,8 g Kaliumcarbonat und 1660 ml Athanol werden 12 Stunden unter Stickstoff- atmosphäre bei Siedetemperatur gerührt. Nach beendeter Reaktion lässt man abkühlen, trennt die ausgeschiedenen Salze durch Filtration ab und dampft das Filtrat im Vakuum ein. Der Eindampfrückstand wird in Benzol aufgenommen und die Benzollosung nacheinander mit Wasser, 2% iger Natronlauge und 5 % iger Essigsäure gewaschen und getrocknet. Nach Eindampfen der Lösung im Vakuum erhält man in n praktisch quantitativer Ausbeute den 3,4-Dibenzyl oxy-5-isopropoxybenzaldehyd in Form eines viskosen Ols.
19,5 g 3,4-Dibenzyloxy-5-isopropoxybenzaldehyd, 72 ml absoluter Alkohol, 54 ml absolutes Benzol, 6, 38 g Pyridin und 4,90 g Nitromethan werden gemischt und auf 0 C abgekühlt. Darauf trägt man unter Rühren innerhalb von 15 Minuten eine Lösung von 2,3 g Natrium in 45 ml Methanol ein und rührt die Mischung einige Stunden bei 0 C. Zur Aufarbeitung wird das dabei gebildete Natriumsalz der Pseudosäure mit einer Lösung von 25 ml Eisessig in 36 ml Athanol zersetzt, die erhaltene Lösung wird im Vakuum eingedampft und der Eindampfrückstand wie in Beispiel beschrieben aufgearbeitet.
Es werden 18,9 g kristallisiertes 1- (3', 4'-Dibenzyloxy-5'-iso- propoxy-phenyl)-2-nitroäthanol- (1) erhalten, das entspricht einer Ausbeute von 83 % der Theorie. Nach Digerieren in einer Mischung von Xylol und Petrol- äther schmilzt das Produkt bei 94-96, 5 C.
5,0 g 1-(3', 4'-Dibenzyloxy-5'-isopropoxy-phenyl)- 2-nitroäthanol- (1) werden in 70 ml Eisessig gelöst und bei Raumtemperatur und Normaldruck in Ge genwart von 3,0 g Pal'ladiumkohle (10% ig) als Katalysator hydriert. Nach Aufnahme der berechneten Menge Wasserstoff kommt die Hydrierung zum Stillstand, worauf 1,2 g Benzylchlorid zugesetzt werden und das Gemisch bis zur Beendigung der Wasserstoffaufnahme hydriert wird. Anschliessend wird der Katalysator abfiltriert, das Filtrat im Vakuum unter Wasserstoff eingedampft und der Eindampfrückstand durch Digerieren mit Essigester-Ather gereinigt.
Man erhält so 1,8 g dl-1-(3', 4'-Dihydroxy-5'-isopropoxy- phenyl)-2-aminoäthanol-hydrochlorid in öliger Form.
Der als Ausgangsmaterial verwendete 3, 4-Di- acetoxy-5-isopropoxy-benzaldehyd kann wie folgt hergestellt werden : Der durch Kondensation von Gallussäuremethylester mit Benzalchlorid in Pyridin erhältliche 3,4-Dioxybenzyliden-5-hydroxy-benzoesäuremethylester wird in absolutem Athanol in Ge genwart von Kalium mit Isopropyljodid unter dreistündigem Erhitzen zum Sieden umgesetzt, wobei man den 3,4-Dioxybenzyliden-5-isopropoxy-benzoesäuremethylester erhält, der sich alkalisch verseifen lässt.
Die dabei, erhaltene 3,4-Dioxybenzyliden-5-iso propoxy-benzoesäure vom Schmelzpunkt 153-154 C wird in Eisessig mit Palladiumkohle als Katalysator bei 50 C und Normaldruck hydriert, wobei die 3,4 Dihydroxy-5-isopropoxy-benzoesäure vom Schmelz- punkt 178-182 C erhalten wird. Durch Acetylieren kann daraus die 3,4-Diacetoxy-5-isopropoxy-benzoesäure vom Schmelzpunkt 154-157 C gewonnen werden, aus der man durch Umsatz mit Thionyl- chlorid das 3,4-Diacetoxy-5-isopropoxy-benzoylchlorid arhält. Dieses kann durch Reduktion nach Rosenmund in Xylol bei 140 C in den 3,4-Diacet oxy-5-isopropoxy-benzaldehyd übergeführt werden.

It is known that 3,5-dioxy-phenyl-alkanolamines show a physiological effect comparable to that of adrenaline, but that they have the advantage over this that they are much more stable and therefore, in contrast to adrenaline, can be used orally.
These compounds have hitherto been prepared by converting the corresponding 3,5-dioxy-co-halogenoacetophenones, optionally substituted on the IC atom by alkyl radicals, the OH groups of which may be protected, into amino ketones by methods known per se and then reducing the Aminoketones to the corresponding amino alcohols. Among the known methods, the reaction of the haloketones with ammonia or amines and, in the case of the preparation of phenylethanolamines with a free amino group, the reaction of the haloketones with sodium azide and subsequent simultaneous hydrogenation of the azido group and the keto group have found practical application.
The preparation of the 3,5-dioxy-phenylalkanolamines, starting from the o-halo ketones, has some serious disadvantages, however.
Only the oj-halogen ketones themselves are extremely uncomfortable to handle because they are very irritating to the mucous membranes, especially those of the eyes. Furthermore, the yields in the reaction with ammonia or amines are very poor. If one chooses the process via the azidoacetophenones, the yields are much better, but the azidoacetophenones again have the unpleasant property that they are very explosive. Dioxyazidoacetophenones detonate at temperatures below 150 C if they are heated in a tube, while sodium azide, for example, can be heated to over 400 C under the same conditions without detonating.
It is clear that the handling of such a substance, especially in larger quantities, represents a significant source of danger.
The present invention is a new process for the preparation of phenylethanolamines with unsubstituted amino groups, which gives significantly higher yields, and in which working with substances that irritate the eyes or other mucous membranes, or that are explosive, is avoided.

The production of nitroalkanols by condensation of aldehydes with nitroalkanes is known in principle. However, a person skilled in the art is familiar with the fact that this condensation is carried out in suitable solvents, especially in lower alcohols, and that good yields can only be achieved if the aldehyde chosen as the starting material is readily soluble in these alcohols at 0 ° C. is. If, on the other hand, the aldehyde is only sparingly soluble or not at all soluble in lower alcohols at temperatures around 0 C, only very poor yields are achieved because the reaction must then be carried out under unfavorable physical conditions. The main reason for this is that in the condensation of aldehydes with nitroalkanes in the presence of alkali, e.g. B.
Sodium methylate, primarily the alkali metal salts of the corresponding pseudo-acids, are formed and precipitate from the reaction mixture. If the aldehyde is not more soluble in the solvent used than the alkali salt of the pseudo-acid formed, it will be enclosed by this and thus withdrawn from further reaction. As a result, the aldehyde can only partially react. A further reduction in yield must then be accepted, since the work-up of such an only partially fully reacted reaction mixture is difficult and wasteful due to the instability of the nitroalkanols formed.
Since the aldehydes of the formula II are only extremely sparingly soluble in lower alcohols, the use of the nitroalkane condensation for the preparation of the phenylalkanolamines of the formula I did not appear to be promising from the outset, for the reasons outlined above. Attempts to use other solvents in which the aldehydes of the formula II are readily soluble at about 0 C, such as toluene, benzene or ethyl acetate, as the reaction medium, were doomed to failure, since ethyl acetate itself reacts under the reaction conditions and during reaction in the presence of solid alkali alcoholate, which is not soluble in benzene and toluene, no good yields could be achieved. Therefore, this mode of reaction initially did not appear to be applicable for the purpose according to the invention.
Surprisingly, it has now been found that the condensation of the aldehydes of the formula II with nitromethane is still possible, and indeed even in very good yield, if a solvent mixture consisting of lower aliphatic alcohols, aromatic hydrocarbons and amines is used as the reaction medium.
The present invention accordingly provides a process for the preparation of the phenylethanolamines of the formula I, which is characterized in that aldehydes of the formula II in a solvent mixture consisting of lower aliphatic alcohols, aromatic hydrocarbons and amines at temperatures from -10 to +30 C and in the presence of alkali alcoholates as condensation agents with nitromethane to form nitroalkanols of the formula III and these are then reduced, with any aralkoxy groups present being converted simultaneously or subsequently into the free oxy groups.
The phenylethanolamines of the formula I thus formed can then be obtained from the reaction mixture either as free bases or in the form of their salts with inorganic or organic acids.
The nitromethane condensation is particularly conveniently carried out at temperatures between -5 and -F 10 C. All lower aliphatic alcohols, e.g. B. methanol, ethanol, isopropanol, and the aromatic hydrocarbons benzene, toluene and xylene are particularly effective.
Primary, secondary and tertiary amines, including nitrogen-containing heterocycles, are suitable as amines. Examples of this are pyridine, pyrrole, aniline, dimethylaniline, diethylamine, triethylamine, β-dimethylamine, inoethanol, diethanolamine and triethanolamine. It is particularly useful to use a mixture of ethanol, benzene and pyridine, preferably in a mixing ratio of 5-12 parts by volume of absolute benzene and 0.5-1.2 parts by volume of pyridine per 10 parts by volume of absolute ethanol.
The subsequent reduction of the nitroalkanols of the formula III to the aminoalkanols of the formula I can be carried out in a manner known per se. Catalytic reduction at normal pressure and room temperature, for example with palladium as the catalyst, is particularly preferred. If the process according to the invention is used for the preparation of compounds of the formula I which have free hydroxyl groups in the aromatic nucleus, aldehydes in which these hydroxyl groups are protected by aralkyl groups, preferably benzyl groups, are used. In such a case, the protective radicals can be split off at the same time as the reduction of the nitro group by catalytic hydrogenation.
If the nitro group is reduced under conditions in which the aralkoxy groups are retained, the reduction must be carried out e.g. B. a catalytic cleavage of the aralkyl groups can be connected by means of hydrogen.
After the catalytic reduction has ended, the reaction mixture can be prepared in a simple manner, e.g. B. by separating the catalyst by filtration and evaporation of the filtrate to the amino alcohol. In some cases it is also advisable to isolate the amino alcohol of the formula I directly in the form of its salt with inorganic or organic acids.
In the case of isolation as hydrohalides, the following variant of the process according to the invention has proven particularly useful. The acid required for salt formation is not added to the reaction mixture as such, but rather through the catalytic cleavage of compounds which, under these conditions, are generated in the reaction mixture which are lower in hydrogen halide. Benzyl chloride or benzyl bromide, for example, can be mentioned as such substances. To carry this out, after the catalytic reduction of the nitro group has ended, the equivalent amount of benzyl chloride or benzyl bromide is added to the reaction mixture and the mixture is further hydrogenated until the uptake of hydrogen has ceased. 189.3 g of 3,5-diacetoxy-benzaldehyde, 342.7 g of potassium bicarbonate, 325.6 g of benzyl chloride and 2570 ml of ethanol are stirred for 9 hours under nitrogen at boiling temperature. 47.5 g of 3,5-dibenzyloxy-benzaldehyde, 212 ml of absolute ethanol, 108 ml of absolute benzene, 18.5 g of absolute pyridine and 14.4 g of nitromethane are stirred at about 0 ° C., a solution being formed. The filtrate obtained is then evaporated in vacuo under hydrogen, the crystalline residue is digested with 30 ml of isopropyl alcohol and cooled and isolated. 9.56 g of hydrochloride (83.9% of the theoretically possible amount) with a melting point of 212-214 ° C. are obtained.
The 3,5-diacetoxy-benzaldehyde required as the starting product is obtained from 3,5-diacetoxy-benzoyl chloride by reduction according to Rosenmund.
Example 2
36.96 g of 3,4,5-triacetoxy-benzaldehyde, 98.70 g of benzyl chloride, 91.2 g of potassium carbonate and 658 ml of ethanol are stirred under nitrogen at boiling temperature for 12 hours. After cooling, it is filtered off with suction and the precipitate is washed with 300 ml of ethanol. The precipitate is then digested with 750 ml of water, again filtered off with suction, washed with water and dried. Example 3
113.16 g of 3,4,5-tribenzyloxybenzaldehyde are dissolved in 2260 ml of glacial acetic acid, 33.9 ml of 66% hydrogen bromide are added to the solution and kept at 50 ° C. for 21/2 hours. It is then largely concentrated in vacuo at 50.degree. The residue is made alkaline with 4N NaOH. The sodium salt of 3,5-dibenzyloxy-4-hydroxy-benzaldehyde crystallizes out and is isolated by suction.
It is digested twice with 100 ml of ethyl acetate each time, filtered off with suction again, washed with ethyl acetate and dried. 73.51 g of the sodium salt are obtained.
The sodium salt obtained is dissolved in 700 ml of hot methanol, the solution is acidified with 4N HCl I and left to stand at room temperature for 1 hour and at -20.degree. C. for 5 hours. The crystallized product is isolated by suction, digested twice with 100 ml of water each time, suction filtered, washed with water and dried. 44.41 g of crude 3,5-dibenzyloxy-4-hydroxy-benzaldehyde are obtained in this way. Melting point 118-120 ° C. The product is recrystallized from 750 ml of isopropyl alcohol. Yield 40.23 g with a melting point of 120-122 C.
2.0 g of potassium hydroxide are dissolved in 110 ml of ethanol. 8.9 g of 3,5-dibenzyloxy-4-hydroxybenzaldehyde are dissolved in this solution, 5.2 g of methyl iodide are added to the solution and the reaction mixture is stirred at boiling temperature for 3 hours. After cooling, it is suctioned off, the precipitate is washed with a little chilled ethanol and rubbed twice with water. After filtering off with suction and washing with water, the crude product is dried. 6.9 g of 3,5-dibenzyloxy-4-methoxybenzaldehyde are obtained, that is 74.4% of the theoretically possible amount.
For purification, the crude product is recrystallized from isopropyl alcohol. Melting point 86-87.5 C.
10.0 g of 3,5-dibenzyloxy-4-methoxy-benzaldehyde, 40 ml of absolute ethanol, 30 ml of absolute benzene, 3.5 g of pyridine and 2.7 g of nitromethane are stirred at about 0 C and slowly a solution of 1, 35 g of sodium in 25 ml of methanol were added dropwise. Example 4
To produce 3-methoxy-4,5-dibenzyloxy benzaldehyde, 3-methoxy-4,5-diacetoxybenzaldehyde is reacted with benzyl chloride in an alcoholic solution in the presence of potassium carbonate as in Example 2 or 3. 3-methoxy-4,5-dibenzyloxy-benzaldehyde with a melting point of 0.2: 217-220 ° C. is obtained in a 55% yield.
12.4 g of the aldehyde, 50 ml of absolute ethanol,
37.5 ml of absolute benzene, 4.4 g of pyridine and 3.4 g of nitromethane are stirred at 0 ° C. and a solution of 1.6 g of sodium in 31 ml of methanol is added dropwise. After stirring for a total of 5 hours at 0 C, it is acidified with a cooled solution of 13 ml of glacial acetic acid in 25 ml of absolute ethanol and the resulting solution is evaporated at 30-40 C in a vacuum. The work-up takes place analogously to Example 1.
This gives 14.1 g of crude 1- (3'-methoxy-4 ', 5'-di-benzyloxyphenyl) -2-nitro-ethanol- (l) (97% of theory), which after recrystallization , Acetone-xylene-petroleum ether 1: 2: 2 melts at 104-107 C.
8.15 g of the above nitroalkanol are reduced in 100 ml of glacial acetic acid in the presence of 0.8 g of palladium carbon (10% Pd) at normal pressure and room temperature. 2-aminoethanol hydrochloride can be isolated. Melting point 157-157.5 C decomposition. The 3-methoxy-4,5-diacetoxy-benzaldehyde is obtained from the 3-methoxy-4,5-diacetoxy-benzoyl chloride by reduction according to Rosenmund.
Example 5
3-Ethoxy-4, 5-diacetoxy-benzaldehyde is reacted with benzyl chloride analogously to Examples 1-3 in ethyl alcoholic solution in the presence of potassium bicarbonate or potassium carbonate, and 3-ethoxy-4,5-dibenzyl oxy is obtained analogously to Example 4 -benzaldehyde with a melting point of 0.220-223 C.
Analogously to the other examples, 18.8 g of this aldehyde, 72 ml of absolute alcohol, 54 ml of absolute benzene, 6.38 g of pyridine and 4.9 g of nitromethane are stirred and a solution of 2.3 is stirred at a temperature of 0 to 5 ° C g of sodium in 45 ml of methanol are allowed to flow in. 7.0 g of 3,4,5-tribenzyloxy-benzaldehyde, 26 ml of benzene, 26 ml of methanol, 2.9 g of triethanolamine and 1.6 g of nitromethane are treated as in Example 2 with a solution of 0.85 g of sodium in 13 ml of methanol implemented. 5.7 g of 3,4,5-tribenzyloxy-benzaldehyde, 22 ml of absolute xylene, 22 ml of absolute ethanol, 1.63 g of diethanolamine and 1.3 g of nitromethane are analogous to Example 2 with a solution of 0.85 g of sodium in 13 ml of methanol reacted. Example 11
99.5 g of 3,4-diacetoxy-5-isopropoxy-benzaldehyde, 199.4 g of benzyl chloride, 162.8 g of potassium carbonate and 1660 ml of ethanol are stirred for 12 hours under a nitrogen atmosphere at the boiling point. After the reaction has ended, the mixture is allowed to cool, the precipitated salts are separated off by filtration and the filtrate is evaporated in vacuo. The evaporation residue is taken up in benzene and the benzene solution is washed in succession with water, 2% strength sodium hydroxide solution and 5% strength acetic acid and dried. 19.5 g of 3,4-dibenzyloxy-5-isopropoxybenzaldehyde, 72 ml of absolute alcohol, 54 ml of absolute benzene, 6.38 g of pyridine and 4.90 g of nitromethane are mixed and cooled to 0.degree.
