Verfahren zur Herstellung eines Impfstoffes zur gleichzeitigen
Immunisierung von Hunden oder Füchsen gegen Staupe, Hepatitis contagiosa canis und Leptospirose (Stuttgarter Hundeseuche und Weil'sche Krankheit)
Die belgische Patentschrift Nr. 619 228 beschreibt ein Verfahren zur Herstellung eines Impfstoffes zur gleichzeitigen Immunisierung von Hunden oder Füchsen gegen Staupe und Hepatitis contagiosa canis, bei welchem man ein aus Hundelebern oder Hundemilzen durch Extraktion oder aus Gewebekultur gewonnenes Hepatitis contagiosa canis-Virus-haltiges Material mittels Formaldehyd inaktiviert, hierauf den freien Formaldehydanteil durch Zusatz einer Lösung, die die 1,5-2-fach stöchiometrische Menge an sauren Salzen der schwefeligen Säure, insbesondere Natrumbisulfit, enthält,
abbindet und das so gewonnene Produkt mit einer aus einer Gewebekultur nach dem in der belgischen Patentschrift Nr. 594 749 beschriebenen Verfahren gewonnenen, durch wenigstens 50 aufeinanderfolgende Passagen modifiziertes Staupevirus enthaltenden Suspension mischt und die Mischung gefriertrocknet.
Nach dem genannten Verfahren wird somit eine Vaccine gewonnen, die zur gleichzeitigen Immunisierung von Hunden und Füchsen gegen Hepatitis contagiosa canis und Staupe geeignet ist.
In weiterer Ausgestaltung dieses Gegenstandes wurde nun gefunden, dass man auch eine Vaccine zur gleichzeitigen Immunisierung von Hunden oder Füchsen gegen Hepatitis contagiosa canis, Staupe und Leptospirose herstellen kann. Dabei wird zu der genannten Vaccine zur gleichzeitigen Immunisierung von Hunden oder Füchsen gegen Staupe und Hepatitis contagiosa canis eine durch Tieffrieren abgetötete Leptospira canicola-Kultursuspension und eine durch Tieffrieren abgetötete Leptospira icterohaemorrhagiae Kultur-suspension zugegeben und gegebenenfalls lyophilisiert.
Die Erfindung betrifft daher ein Verfahren zur Herstellung eines Impfstoffes zur gleichzeitigen Immunisierung von Hunden oder Füchsen gegen Staupe, Hepatitis contagiosa canis und Leptospirose (Stuttgarter Hundeseuche und Weil'sche Krankheit), das dadurch gekennzeichnet ist, dass man ein aus Hundelebern oder Hundemilzen durch Extraktion oder aus Gewebekultur gewonnenes Hepatitis contagiosa canis Virus-haltiges Material mittels Formaldehyd inaktiviert, hierauf den freien Formaldehydanteil durch Zusatz einer Lösung, die die 1,5-2-fach stöchiome- trische Menge an sauren Salzen der schwefligen Säure, insbesondere Natriumbisulfit, enthält, abbindet und das erhaltene Produkt mit einer aus einer Gewebekultur gewonnenen,
durch wenigstens 50 aufeinanderfolgende Passagen modifiziertes Staupevirus enthaltenden Suspension sowie mit einer durch Tieffrieren abgetöteten Leptospira canicola-Kultursuspension und einer Leptospira icterohaemorrhagiae-Kultursuspension mischt und die Mischung gefriertrocknet.
Die genannte, den Staupevirus enthaltende, Suspension kann gemäss belgischer Patentschrift Nr. 594 749 gewonnen werden.
Zweckmässig geht man so vor, dass man ein Mischungsverhältnis von einem gegen Staupe wirksamen Anteil, der aus etwa 25% eines modifizierten staupevirushaltigen Gewebekulturmaterials, 60% Rinderbouillon vom pH-Wert 7,6 und 15% einer 50% igen Glukoselösung besteht, zu einem annähernd gleich grossen, gegen Hepatitis contagiosa canis wirksamen Anteil, der aus etwa 90% eines inaktivierten Hepatitis contagiosa canis-Virus-haltigen Extraktes und 10% einer 2% igen Aluminiumhydroxydlösung besteht,
einhält und zu 20 Volumenanteilen dieser Mischung 1 Volumenanteil einer im Dunkelfeld mikroskopisch auf die Dichte von 100.106 Organismen pro ml eingestellten und durch Tieffrieren abgetöteten Leptospira canicola-Kultursuspension und 1 Volumenanteil auf die Dichte von 100.106 Organismen pro ml eingestellten und durch Tieffrieren abgetöteten Leptospira icterohaemorrhagiae-Kultursuspension zusetzt.
Beispiel I
1.000 ml einer aus Hundenierenepithelkultur gewonnenen, modifiziertes Staupevirus enthaltenden Suspension werden mit 1.000 ml einer mit Formaldehyd inaktivierten und Natriumbisulfit behandelten 0,2% Aluminiumhydroxyd enthaltenden Hepatitis contagiosa canis-Virussuspension unter Eiskühlung gemischt.
Dazu werden 100 ml einer im Dunkelfeld mikroskopisch auf eine Dichte von 100.106 Organismen pro ml eingestellten und durch Leptospira canicola-Kultursuspension und 100 ml in gleicher Weise auf eine Dichte von 100.105 Organismen pro ml eingestellten und durch Tieffrieren abgetöteten Leptospira icterohaemorrhagiae-Kultursuspension zugesetzt. Je 2,2 ml dieser so erhaltenen Mischung werden in Fläschchen abgefüllt, im Stehen bei einer Temperatur von-40 C eingefroren und in einer Gefriertrocknungsanlage lyophilisiert.
Die getrocknete Substanz, die sich in evakuierten Fläschchen befindet, stellt einen Impfstoff gegen Staupe, Hepatitis contagiosa canis und Leptospirose (Stuttgarter Hundeseuche und Weil'sche Krankheit) in haltbarer und lagerfähiger Form dar.
Bestimmung der Vertraglichkeit und Wirksamkeit der gemäss der Erfindung hergestellten Vaccine A. Prütung am Hund Immunisierung ss/'Hng.'
8 für das Staupe-und das Hepatitis contagiosa canis-Virus und für Leptospirose empfängliche 8 Wochen alte Welpen eines Wurfes werden in einem Isolierzwinger untergebracht, 4 der Hunde werden mit je 1 Dosis = 2 ml der erfindungsgemäss hergestellten Vaccine subkutan vacciniert. Die Vaccine besitzt einen Staupevirusgehalt von 104, 375 TCID50. Die Abkürzung TCID50 bedeutet 50% der Tissue Culture Infectivity Dosis.
Sie errechnet sich nach der Methode von REED und MUENCH (Am. J. Hyg. 27,493,1938). 4 Hunde verblieben als unbehandelte Kontrollen. Davon werden 2 Hunde für die Testinfektion mit pathogenem Staupevirus und 2 für die Testinfektion mit pathogenem Hepatitis contagiosa canis-Virus verwendet.
Verträglichekit:
Alle 8 Hunde bleiben während einer Beobachtungszeit von 10 Wochen gesund. Lediglich bei den 4 vac cinierten Hunden tritt am 4. oder 5. Tag ein ephemerer Temperaturanstieg um 0,4 bis 0,8 C auf, ohne dass sonstige klinische Symptome erkennbar wären. Durch das in der Vaccine enthaltene Aluminiumhydroxyd bildet sich am Applikationsort ein Vaccinedepot aus, das Bohnengrösse erreichen kann und innerhalb von 1 bis 3 Wochen resorbiert ist.
Wirksamkeit : a) Bestimmung der Antikorper gegen Staupe, Hepatitis contagiosa canis und Leptospirose
Von sämtlichen Versuchshunden werden vor Versuchsbeginn und 4 Wocheri nach der Impfung Blutproben entnommen und auf Antikörper gegen Staupe und Hepatitis contagiosa canis im Virusneutralisationstest in der Gewebekultur und auf Antikörper (Agglutinine) gegen Leptospirose (L. canicola und L. icterohaemorrhagiae) in der Agglutinations-Lysis-Reaktion untersucht.
Der Impfeffekt ist in der beigefügten Tabelle schematisch dargestellt (Tabelle 1). Alle Hunde besassen vor der Impfung keine Antikörper gegen Staupe und Hepatitis contagiosa canis und keine Agglutinine gegen Leptospira canicola und Leptospira icterohaemorrhagiae. 4 Wochen nach der Impfung weisen alle vaccinierten Hunde einen gleichmässigen und hohen Gehalt an Staupe-und Hepatitis contagiosa canis Virus-neutralisierenden Antikörpern und an Agglutininen gegen L. canicola und L. icterohaemorrhagiae auf. Die Virus-neutralisierenden Dosen (VND) werden nach der Methode von REED und MUENCH (umseitig zitiert) errechnet.
Sie ergeben sich aus der Anzahl der im Virus-Neutralisationstest verwendeten Vi rus-ID50 multipliziert mit dem reziproken 50% igen neutralisierenden Endwert des geprüften Serums. Die VDN werden auf 1 ml des unverdünnten Serums bezogen (zitiert : DRÄGER, ACKERMANN, BARTH : Medizin und Chemie VI, 532,1958). b) Belastungsinfektion mit pathogenem Staupe-und Hepatitis contagiosa canis-Virus (H. c. c.-Virus)
6 Wochen post vaccinationem (p. v.) werden sowohl die vaccinierten als auch die beiden Staupekontrollhunde mit einem hundepathogenen Staupevirusstamm subkutan infiziert. Der verwendete Virusstamm führt bei staupeempfänglichen Hunden zum sogenannten Virusstadium der Staupe, das charakterisiert wird durch eine zweigipfelige Fieberkurve.
Auf der Höhe der zweiten Fieberzacke lässt sich Staupevirus in den Organen der erkrankten Hunde durch die Komplementbindungsreaktion (KBR) nachweisen. Nach der Testinfektion bleiben alle vaccinierten Hunde vollkommen gesund und fieberfrei. Die beiden unbehandelten Staupekontrollhunde zeigen dagegen die typischen Symptome einer Staupeinfektion. Das Staupevirus wird durch die KBR nachgewiesen.
2 Wochen nach der Testinfektion mit Staupevirus werden alle vaccinierten Hunde und die beiden unbehandelten Hepatitis contagiosa canis-Kontrollen mit einem hundepathogenen H. c. c.-Virus infiziert. Alle vaccinierten Hunde bleiben danach gesund und zeigen keinerlei Symptome, die für die Hepatitis contagiosa canis sprechen. Die unbehandelten Kontrolltiere dagegen erkranken mit Fieber an Hepatitis contagiosa canis und verenden zum Teil. H. c. c. wird bei ihnen durch den Sektionsbefund und den Nachweis des Virus in der KBR festgestellt.
Die erfindungsgemäss hergestellte Vaccine wurde an Hunden in weiteren Versuchen und unter Praxisbedingungen geprüft, wobei die im vorangegangenen Beispiel erhaltenen Ergebnisse ihre Bestätigung fanden.
Die Vaccine erwies sich als gut verträglich und führte zu keinen bemerkenswert grösseren Nebenerscheinungen.
B. Prütung an Laboratoriumstieren
Die erfindungsgemäss hergestellte Vaccine wird auf Verträglichkeit und Unschädlichkeit ausser an Hunden an weissen Mäusen, Meerschweinchen und Goldhamstern geprüft. Meerschweinchen und Goldhamster werden auch zur Wirksamkeitsprüfung gegen Leptospirose (Stuttgarter Hundeseuche und Weil'sche Krankheit) herangezogen. Zur Verträglichkeits-und Unschädlichkeitsprüfung werden weisse Mäuse mit der wiedergelösten gebrauchsfertigen Vaccine intracerebral und Meerschweinchen und Goldhamster subkutan vacciniert. Die Tiere werden 10 Tage beobachtet und dürfen nicht erkranken.
Zur Wirksamkeitsprüfung gegen L. canicola und L. icterohaemorrhagiae werden Meerschweinchen mit je 0,5 ml und Goldhamster mit je 0,3 ml der wiedergelösten gebrauchsfertigen Vaccine subkutan in einwöchigem Abstand vacciniert. 14 Tage nach der Impfung werden die Tiere entblutet und das aus den Blutproben gewonnene Serum auf Agglutinine (Antikörper) gegen L. canicola und L. icterohaemorrhagiae untersucht. Es besteht auch die Möglichkeit, bei Meerschweinchen die Ausbildung von H. c. c.-Antikörpern durch die Untersuchung der Seren im Virusneutralisationstest in der Gewebekultur zu prüfen. In der nachstehenden Tabelle ist der Prüfungsablauf einer Herstellungscharge der erfindungsgemäss hergestellten Vaccine wiedergegeben (Tabelle 2).
Anstelle der serologischen Untersuchung auf An tikörper gegen Leptospiren kann die Wirksamkeit durch eine Testinfektion mit pathogenen Leptospirenkulturen geprüft werden. Der in Tabelle 3 dargestellte Versuch wird analog wie in der Tabelle 2 wiedergegeben durchgeführt. Anstelle des Entblutens werden die Goldhamster mit pathogenen L. canicola-Kulturen i. p. infiziert. Dabei bleiben die vaccinierten Tiere gesund, die nicht vaccinierten Kontrolltiere erkranken oder verenden an Leptospirose. Die Diagnose wird durch den Sektionsbefund gefestigt.
Wie aus den dargelegten Beispielen zu ersehen ist, führt die Anwendung der erfindungsgemäss hergestellten Vaccine zu einem gleichzeitigen Schutz gegen Staupe, Hepatitis contagiosa canis und Leptospirose (L. canicola und L. icterohaemorrhagiae), der sich durch seine Güte und Gleichmässigkeit auszeichnet.

sets and the product thus obtained is mixed with a suspension containing distemper virus obtained from a tissue culture by the method described in Belgian patent specification No. 594 749 and containing modified by at least 50 successive passages, and the mixture is freeze-dried.
According to the method mentioned, a vaccine is thus obtained which is suitable for the simultaneous immunization of dogs and foxes against hepatitis contagiosa canis and distemper.
In a further embodiment of this subject it has now been found that a vaccine for the simultaneous immunization of dogs or foxes against hepatitis contagiosa canis, distemper and leptospirosis can also be produced. In this case, a Leptospira canicola culture suspension killed by deep freezing and a Leptospira icterohaemorrhagiae culture suspension killed by deep freezing are added to the vaccine mentioned for the simultaneous immunization of dogs or foxes against distemper and hepatitis contagiosa canis and, if necessary, lyophilized.
The invention therefore relates to a method for the production of a vaccine for the simultaneous immunization of dogs or foxes against distemper, hepatitis contagiosa canis and leptospirosis (Stuttgart dog disease and Weil's disease), which is characterized in that one from dog livers or dog spleen by extraction or Material containing hepatitis contagiosa canis virus obtained from tissue culture is inactivated by means of formaldehyde, and the free formaldehyde content is then inactivated by adding a solution which contains 1.5-2 times the stoichiometric amount of acidic salts of sulphurous acid, in particular sodium bisulfite, and binds the product obtained with one obtained from a tissue culture,

It is advisable to proceed in such a way that a mixing ratio of a proportion effective against distemper, which consists of about 25% of a modified tissue culture material containing distemper virus, 60% beef broth with a pH of 7.6 and 15% of a 50% glucose solution, is approximately the same proportion effective against contagious hepatitis canis, which consists of around 90% of an extract containing inactivated contagious hepatitis canis virus and 10% of a 2% aluminum hydroxide solution,
and 20 parts by volume of this mixture 1 part by volume of a Leptospira canicola culture suspension adjusted microscopically in the dark field to the density of 100,106 organisms per ml and killed by freezing and 1 part by volume to the density of 100,106 organisms per ml and killed by freezing culture suspension Leptospira icterohaemorrhagiae clogs.
Example I.
1,000 ml of a modified canine distemper virus suspension obtained from dog kidney epithelial culture are mixed with 1,000 ml of a formaldehyde-inactivated and sodium bisulfite-treated 0.2% aluminum hydroxide containing hepatitis contagiosa canis virus suspension with ice cooling.
To this end, 100 ml of a Leptospira icterohaemorrhagiae culture suspension which has been adjusted microscopically in the dark field to a density of 100,106 organisms per ml and which has been adjusted to a density of 100,105 organisms per ml and killed by freezing by Leptospira canicola culture suspension and 100 ml in the same way. 2.2 ml each of this mixture obtained in this way are filled into vials, frozen while standing at a temperature of −40 ° C. and lyophilized in a freeze-drying system.
The dried substance, which is in evacuated vials, is a vaccine against distemper, hepatitis contagiosa canis and leptospirosis (Stuttgart dog disease and Weil's disease) in a durable and storable form.
Determination of the compatibility and effectiveness of the vaccines A. Testing on dogs, immunization ss / 'Hng.'
8 8-week-old puppies of a litter susceptible to the canine distemper virus and hepatitis contagiosa virus and to leptospirosis are housed in an isolation kennel, 4 of the dogs are vaccinated subcutaneously with 1 dose = 2 ml of the vaccine produced according to the invention. The vaccine has a distemper virus content of 104,375 TCID50. The abbreviation TCID50 means 50% of the tissue culture infectivity dose.
It is calculated using the method of REED and MUENCH (Am. J. Hyg. 27, 493, 1938). 4 dogs remained as untreated controls. Of these, 2 dogs are used for the test infection with pathogenic distemper virus and 2 for the test infection with pathogenic hepatitis contagiosa canis virus.
Compatibility kit:
All 8 dogs remain healthy during an observation period of 10 weeks. Only in the 4 vacated dogs did an ephemeral rise in temperature of 0.4 to 0.8 C occur on the 4th or 5th day without any other clinical symptoms being discernible. The aluminum hydroxide contained in the vaccine forms a vaccine depot at the application site, which can reach the size of a bean and is absorbed within 1 to 3 weeks.
Efficacy: a) Determination of antibodies against distemper, hepatitis contagiosa canis and leptospirosis
Blood samples are taken from all test dogs before the start of the experiment and 4 weeks after the vaccination and for antibodies against distemper and contagious hepatitis canis in the virus neutralization test in the tissue culture and for antibodies (agglutinins) against leptospirosis (L. canicola and L. icterohaemorrhagiae) in the agglutination lysis Reaction examined.
The vaccination effect is shown schematically in the attached table (Table 1). Before vaccination, none of the dogs had antibodies against distemper or hepatitis contagiosa canis and no agglutinins against Leptospira canicola and Leptospira icterohaemorrhagiae. 4 weeks after the vaccination, all vaccinated dogs show an even and high content of antibodies that neutralize distemper and hepatitis contagiosa canis virus and of agglutinins against L. canicola and L. icterohaemorrhagiae. The virus-neutralizing doses (VND) are calculated using the method of REED and MUENCH (quoted overleaf).
They result from the number of virus ID50 used in the virus neutralization test multiplied by the reciprocal 50% neutralizing end value of the tested serum. The VDN are based on 1 ml of the undiluted serum (cited: DRÄGER, ACKERMANN, BARTH: Medizin und Chemie VI, 532, 1958). b) Stress infection with pathogenic distemper and hepatitis contagiosa canis virus (H. c. c. virus)
6 weeks after vaccinationem (p. V.), Both the vaccinated and the two congestion control dogs are infected subcutaneously with a canine pathogenic distemper virus strain. The virus strain used leads to the so-called virus stage of distemper in dogs susceptible to distemper, which is characterized by a two-peaked fever curve.
At the level of the second fever spike, distemper virus can be detected in the organs of the diseased dog through the complement fixation reaction (KBR). After the test infection, all vaccinated dogs remain perfectly healthy and free of fever. The two untreated congestion control dogs, on the other hand, show the typical symptoms of a congestion infection. The distemper virus is detected by the KBR.
2 weeks after the test infection with distemper virus, all vaccinated dogs and the two untreated hepatitis contagiosa canis controls are treated with a dog-pathogenic H. c. c. virus infected. All vaccinated dogs remain healthy afterwards and show no symptoms that speak for hepatitis contagiosa canis. The untreated control animals, on the other hand, contract hepatitis contagiosa canis with fever and some of them die. H. c. c. is determined by the autopsy findings and the detection of the virus in the KBR.
The vaccine produced according to the invention was tested on dogs in further experiments and under practical conditions, the results obtained in the previous example being confirmed.
The vaccine proved to be well tolerated and did not lead to any noticeably major side effects.
B. Testing on laboratory animals
The vaccine produced according to the invention is tested for compatibility and harmlessness, except in dogs, on white mice, guinea pigs and golden hamsters. Guinea pigs and golden hamsters are also used to test the effectiveness against leptospirosis (Stuttgart dog disease and Weil's disease). To test compatibility and harmlessness, white mice are vaccinated intracerebrally with the redissolved ready-to-use vaccine and guinea pigs and golden hamsters are vaccinated subcutaneously. The animals are observed for 10 days and must not become ill.
To test the effectiveness against L. canicola and L. icterohaemorrhagiae, guinea pigs are vaccinated subcutaneously at one-week intervals with 0.5 ml each and golden hamsters with 0.3 ml each of the redissolved ready-to-use vaccines. 14 days after vaccination, the animals are bled and the serum obtained from the blood samples is examined for agglutinins (antibodies) against L. canicola and L. icterohaemorrhagiae. It is also possible to develop H. c. In guinea pigs. c. antibodies by examining the sera in the virus neutralization test in the tissue culture. The following table shows the test sequence for a production batch of the vaccines produced according to the invention (Table 2).
Instead of a serological test for antibodies against leptospira, the effectiveness can be checked by a test infection with pathogenic leptospira cultures. The experiment shown in Table 3 is carried out analogously to that shown in Table 2. Instead of bleeding the golden hamsters with pathogenic L. canicola cultures i. p. infected. The vaccinated animals remain healthy, the non-vaccinated control animals become ill or die of leptospirosis. The diagnosis is confirmed by the autopsy findings.
As can be seen from the examples given, the use of the vaccines prepared according to the invention leads to simultaneous protection against distemper, contagious hepatitis and leptospirosis (L. canicola and L. icterohaemorrhagiae), which is distinguished by its quality and uniformity.
