Verfahren zur Herstellung von Penicillinderivaten
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von Penicillinderivaten. Die darnach hergestellten neuen Verbindungen sind wertvolle antibakterielle Agenzien. Sie dienen als Zusätze zu tierischem Futter, als Agenzien f r die Behandlung von Mas, titis bei Hornvieh und als therapeutische Präpa- rate zur Behandlung insbesondere von infekti¯sen Erkrankungen, welche durch Gram-positive Bakterien verursacht sind, bei Federvieh, bei Säugetieren sowie beim Menschen.
Antibakterielle Agenzien, wie das Benzylpenicil- Hn, haben sich in der Vergangenheit als hochwirksam in der Therapie infektiöser Erkrankungen, welche durch Gram-positive Bakterien verursacht sind, erwiesen. Jedoch weisen solche Agenzien den schwer- wiegenden Nachteil auf, dass sie gegenüber wässrigen Säuren instabil sind, was sich z. B. bei oraler Ver abreichung auswirkt. Ferner sind diese Agenzien unwirksam gegenüber manchen Bakterienstämmen, welche Penicillinase produzieren.
Viele der erfin- dungsgemäss hergestellten neuen) Verbindungen zeigen au¯er ihrer starken antibakteriellen Wirksamkeit Wi derstandsfähigkeit gegenüber der Zerstörung durch SÏure oder durch Penicillinase auf und sind ferner wirksam gegenüber benzylpenicillinresistenten Bak terienstämmen.
Die erfindungsgemäss bergestellten neuen Verbindungen weisen die allgemeine Formel

auf. In der Formel bedeutet Ri einen Cyclopentyl-, Cyclohexyl-, Benzyl-, Phenyl-oder substituierten Phenylrest oder eine aliphatische Gruppe mit 1 bis 20 Kohlenstoffatomen. R2 ist ein Cyclopentyl-, Cyclo- hexyl-oder niedrigerer Alkylrest, wobei unter letzterem gerade oder verzweigte, gesättigte, aliphatische Kohlenwasserstoffgruppen mit 1 bis 6 Kohlenstoff- atomen verstanden werden.

Das erfindungsgemässe Verfahren richtet sich auch auf die Herstellung nicht toxischer Salze von Säuren der allgemeinen Formel (I). Darunter werden ver standen die Salze des Natriums, Kaliums, Kaliums und Aluminiums, die Ammoniumsalze und substituierten Ammoniumsalze, z. B. Salze solcher nicht toxischer Amine, wie der Trialkylamine, einschliesslich des TriÏthylamins, Procains, Dibenzylamins, N-Ben- zyl-¯-phenethylamins, 1-Ephenamins, N,N'-Dibenzyl Ïthylendiamins, Dehydroabietylamins, N, N'-bis-De hydroabietyläthylendiamins sowie anderer Amine, welche sich in der Vergangenheit zur Bildung von Salzen des Benzylpenicillins bewährt haben.
Der im vorstehenden verwendete Ausdruck ali- phatischo bezieht sich auf gerade und verzweigte, ge sättigte und ungesÏttigte, äliphatiische Kohlenwasser- stoffgruppen mit 1 bis 20 Kohlenstoffatomen, z. B.
Methyl, Athyl, Propyl, Allyl, Butyl, Isobutyl, a-und ss-Betenyl, Amyl, Hexyl, Lauryl, Octadecyl, Tetradecyl, Tetradecenyl, Hexadecyl, Hexadecenyl, Octa- decyl, Octadecenyl, Eicosenyl usw. Hierunter sind die niedrigeren aliphatischen Gruppen, das heisst diejeni- gen mit 1 bis 6 Kohlenstoffatomen, bevorzugt.
Das erfindungsgemässe Herstellungsverfahren rich tet sich vorzugsweise auf solche Penicillinderivate, in welchen R1 eine niedrigere Alfkylgruppe oder Phenyl- gruppe und R9 eine niedrigere Alkylgruppe, insbeson- dere Dimethylgruppe, darstellt.
Das Herstellungsverfahren ; nach der Erfindung ist dadurch gekennzeichnet, da¯ 6-Aminopenicillansäure oder vorzugsweise eines ihrer Neutralsalze, wie das Natrium-oder Triäthylaminsalz, mit einem Acylie rungsmittel der allgemeinen Formel

worin Z ein bei der Acylierung primärer Aminogruppen austretender funktioneller Rest ist, umgesetzt wird. Als Acylierungsmittel dieser Art haben sich insbesondere die Säurehalogenide, wie das Säurechlorid und das Säurebromid, ferner die Säureanhydride und gemischten Anhydride mit anderen Carbonsäuren einschliesslich Monoester solcher Carbonsäuren und insbesondere niedrigere aliphatische Ester der Kohlensäure, als geeignet erwiesen.
Die vorgenannten, als Acylierungsmittel dienenden Säurechloride, SÏurebromide oder Säureanhydride können aus den entsprechenden a- (aliphatisch)-thio- a-substituierten Essigsäuren oder a-arylthio-a-substi- tuierten Essigsäuren, worin der a-Substituent die Bedeutung von R2 in den vorgenannten Formelle aufweist, nach den in der Literatur bekannten Arbeitsweisen zur Herstellung analoger Säuren, wie der Phenylessigsäure und der Phenoxyessigsäure, her- gestellt werden.
In denjenigen Fällen, wo derartige substituierte Essigsäuren noch nicht beschrieben sind, können sie aus dem geeigneten aliphatischen Mercaptan oder substituierten Phenylthiol und der geeigneten a-chlor-oder a-bromsubstituierten Essigsäure nach bekannben Methoden hergestellt werden.
Im Falle der Hersbellung von Säuren der all gemeinen Formel (II), worin eine der Gruppen R3, R4 und R. eine Amino-oder Alkylaminogruppe ist, das heisst, worin der Substituent am Phenylring ein reaktionsfähiges Wasserstoffatom trägt, welches mit einem Acylierungsmittel für primäre Aminogruppen acyliert werden k¯nnte, werden derartige Aminooder Alkylaminogruppen mit Vorteil vor der Acylie- rungsreaktion geschützt. Die nachfolgende Entfer- nung der schützenden Gruppe zur Bildung der freien aminosubstituierten oder alkylaminosubstituierten.
Penicillinverbindung kann vermittels katalytischer Hydrierung, z. B. mit Palladium oder Platin auf Bariumkarbonat oder Kohle als Träger, durchgeführt werden. Geeignete schützende Gruppen hierzu sind solche der allgemeinen Formel R"-O-CO-, worin R"-eine Alkyl-, Benzyl-, substituierte Benzyl, Phe- nyl-, substituierte Phenyl-oder Tritylgruppe ist. Eine andere Art zur Herstellung geeigneter Acylierungs- mittel, in welchen die Gruppen Rs, R4 oder Ro Aminogruppen sind, besbeht in der Darstellung ent sprechender Nitroverbindungen, welche dann in be- kannter Weise zu den Aminoverbindungen hydriert werden können.
Eine besondere Arbeitsweise des erfindungsgemÏ Ben Herstellungsverfahrens geht aus von Mischanhy driden. Dieselben k¯nnen hergestellt werden aus SÏuren der allgemeinen Formel

und einem Alkylester der Chlorkohlensäure. Hierbei arbeitet man mit Vorteil in Gegenwart eines tertiären Amins, wie des Triäthylamins und in einem wasserfreien inerten und vorzugsweise mit Wasser misch- baren Lösungsmittel, wie in p-Dioxan, dem gegebe nenfalls eine geringe Menge reinen trockenem Acetons zugef gt werden kann. Man arbeitet dabei am besten in der Kälte bei ungefähr 4 C und benötigt Reaktionsdauern von ungefähr 30 Minuten.
Zur L¯sung derart hergestellter Mischanhydride wird danach eine abgekühlte L¯sung der 6-Aminopenicillansäure und ein tertiäres Kohlenwasserstoffamin, z. B. Triäthyl- amin, in einem Lösungsmittel, wie Wasser, hinzugegeben. Anschliessend wird das Reaktionsgemisch während ungefähr einer Stunde gerührt, wobei sich das substituierte Ammoniumsalz des gewünschten Produktes bildet. Hiernach kann das Gemenge gegebenenfalls bei alkalischem pH-Wert mi't einem mit Wasser nicht mischbaren Lösungsmittel, wie mit Ather, extrahiert werden, um nicht umgesetztes Ausgangsmaterial zu entfernen. Anschliessend kann das Produkt in der wässrigen Phase in die freie SÏure übergeführt werden, was vorzugsweise in der Kälte unter einer Schicht von ¯ther durch Zugabe von verdünnter Mineralsäure, z.
B. 5n Schwefelsäure bis zum pH-Wert 2, geschieht. Die freie Säure wird hiernach mit einem mit Wasser nicht mischbaren, neutralenl, organischen Lösungsmittel, wie Ather, ex trahiert und der Extrakt gegebenenifalls mit Wasser in der KÏlte rasch gewaschen und dann getrocknet.
Das im ätherischen Extrakt in freier Säureform enthaltene Produkt kann hiernach in jedes gewünschte Salz eines Meta1'Is oder Amins durch Behandlung mit der geeigneten Base übergeführt werden. Hierzu kann man eine freie Aminobase, wie das Procain oder eine L¯sung von Kalium-2-äUhylhexanoat in trockenem no Butanol, verwenden. Diese Satze sind üblicherweise in Lösungsmitteln, wie Ather, unlöslich und können in reiner Form durch blosses Filtrieren abgetrennt werden.
Eine andere Arbeitsweise zur Herstellung einer ätherischen L¯sung der Säureform der neuen Verbindungen geht aus von einer wässrigen ! Lösung von 6-Aminopenicillansäure und Natriumbikarbonat, zu welcher das Säurechlorid hinzugefügt wird und welche dann bei Zimmertemperatur wÏhrend ungefähr 20 bis 60 Minuten kräftig geschüttelt wird. Hiernach kann das Gemenge mit ¯ther zur Entfernung nicht umgesetzten oder hydrolysierten Ausgangsmaterials extrahiert werden. Anschliessend wird die L¯sun, auf den pH-Wert 2 gebracht und die freie Säureform des Produktes mit ¯ther extrahiert.
Der ätherische Extrakt kann getrocknet werden, was mit Vorteil, mit wasserfreiem Natriumsu'lfat geschieht, wonach das Trocknungsmittel entfernt wird und worauf eine trockene ätherische L¯sung des gewünschten Produktes verbleibt. Letzteres kann aus der L¯sung leicht abgeschieden werden, vorzugsweise in Form eines ätherunlöslichen Salzes, wie des Kaliumsalzes. Diese Arbeitsweise wird mit Vorteil dann angewandt, wenn das Säurechlorid mit einem primären Amin schneller reagiert als es dies mit Wasser tut, was sich durch einen Tes't leicht feststellen lässt. Bei dieser Arbeits- weise kann das Säurechlorid durch eine äquimole- kulare Menge des entsprechenden Säurebromids oder Säureanhydrids ersetzt werden.
Da einige der erfindungsgemäss hergestellten an) d- biotischen Substanzen verhältnismässig instabil sind und leicht chemischen Anderungen unterliegen, welche einen Verlust an antibiotischer Aktivität mit sich bringen, werden mit Vorteil derartig milde Reaktionsbedingungen eingehalten, dass die Zersetzung der Substanzen unterbleibt. Die auszuwählenden Reaktionsbedingungen hängen naturgemäss weitgehend von der Reaktionsbereitschaft der verwendeten chemischen Substanzen ab. In den meisten Fällen ist ein Kompromiss zu schliessen zwischen der Anwendung sehr milder Bedingungen, während längerer Reaktionszeiben und der Verwendung kräftigerer Bedinr gungen. wÏhrend kürzerer Zeiten mit der Möglichkeit einer teilweisen Zersetzung der antibiotischen Substanz.
Die bei den Herstellungsverfahren von Derivaten der 6-Aminopenicillansäure einzuhaltenden Temperaturen sollten im allgemeinen 30¯C nicht berschreiten. In manchen Fällen ist Raumtemperatur geeignet.
Weiterhin sollte die e EinhaDtung stark saurer oder al kalischer Bedingungen beim erfindungsgemässen Verfahren vermieden werden. Es ist vorteilhaft, im pH Bereich zwischen 6 und 9 zu arbeiten, was in üblicher Weise durch Anwendung eines Puffers, z. B. einer L¯sung von Natriumbikarbonat oder eines Natriumt- phosphatpuffers, erreicht werden kann. Ausser der Verwendung wässriger Reaktionsmedien unter Einschlu¯ filtrierter Fermentationsbrühen oder wässriger Lösungen roher 6-Aminopenicillansäure können auch organische Lösungsmittel verwendet werden, z. B.
Dimethylformamid, Dimethylacetamid, Chloroform, Aceton, Methyl-isobutyl-keton und Dioxan. Häufig hat es sich als besonders vorteilhaft erwiesen, eine wässrige L¯sung eines Salzes der 6-Aminopenicillansäure zu einer L¯sung des Acylierungsmittels in einem inerten, und vorzugsweise in einem inerten L¯sungsmittel, welches mit Wasser mischbar ist, sowie Aceton oderDimethylformamid hinzuzugebenl. Kräftiges Rüh ren ist naturgemäss dann von Vorteil, wenn mehr als eine Phase zugegen ist, z. B. feste und flüssige Phase oder zwei flüssige Phasen.
Nach Abschluss der eigentlichen Herstellungsreak- tion können die Endprodukte gegebenenfalls mit den jenigen Techniken isoliert und aufgearbeitet werden, wie sie bei der Herstellung von Benzylpenicillin und Phenoxymethylpenicillin Anwendung gefunden haben.
So kann das Endprodukt mit Diäthyläther oder n Butanol bei saurem pH-Wert extrahiert und anschlie- ssend durch Lyophilisation gewonnen werden, oder es kann in ein lösungsmittelunlösliches Salz, z. B. durch Neutralisation mit einer n-butanolischen Lö- sung von Kalium-2-äthylhexanoat, übergeführt werden. Man kann das Produkt auch aus wässriger Lösung als wasserunlösliches Salz eines Amins ausfäl- len oder direkt daraus durch Lyophilisation gewinnen.
Letzteres geschieht vorzugsweise in Form eines Natrium-oder Kaliumsalizes. Wenn das Produkt als Tri äthylaminsalz erhalten wurde, kann es in die Form der freien Säure und hernach in ein anderes Salz übergeführt werden, in der Art und Weise, wie dies f r Benzylpenicillin und andere Penicillino bekannt ist. So ergibt die Behandlung einer solchen, TriÏthyl aminverbindung in Wasser mit Natriumhydroxyd das Natriumsalz, wonach das Triäthylamin durch Extraktion, z. B. mit Toluol, abgeschieden werden kann.
Behandlung des Natriumsalzes mit starken wässrigen Säuren führt zur Bildung der Säureform, welche ihrerseits in andere Aminsalze, z. B. das Salz des Procains, durch Umsetzung mit der entsprechenden Aminobase übergeführt werden kann. Derart hergestellte Salze können durch Lyophilisation oder, sofern sie unlöslich sind, durch blosses Filtrieren abgetrennt werden.
Eine Arbeitsweise zur Isolierung des Produktes als kristallines Kaliumsalz besteht in der Extraktion aus einer sauren wϯrigen L¯sung z. B. vom pH-Wert 2 mit Diäthyläther, Trocknen des Ätherextrakts und Zugabe von wenigstens einem Aquivalent einer Lösung von Kalium-2-äthylhexanoat in trockenem n Butanol. Das Kaliumsalz fällt normalerweise in kristalliner Form aus und kann durch Filtrieren, oder Dekantieren ; gewonnen werden.
Beispiel I
0, 15 Mol = 20, 9 g a-Allylthiopropionsäure werden in einer Mischung von 75 ml trockenem Aceton und 330 ml trockenem Dioxan gelöst. Zur L¯sung werden unter Rühren'und Kühlung 21, 1 ml Triäthyl- amin = 0, 15 Mol hinzugefügt. Die erhaltene kalte L¯sung (etwa-3 C) wird langsam tropfenweise mit 0, 15 Mol = 14, 2 ml Isobutylchlorformiat versetzt, wobei die Temperatur des Reaktionsgemisches unter 5 C während der ganzen Dauer der Zugabe gehalten wird.
Die erhaltene L¯sung wird bei ungefähr 0 C während 1 Stunde gerührt. Zum Gemisch wird eine L¯sung von 0, 15 Mol = 32, 4 g 6-AminopenicillansÏure in 45 ml Wasser und 22 ml TriÅathylamin rasch hinzugegeben, wobei die Temperatur bei ungefähr 5 C gehalten wird. Die erhaltene L¯sung wird in der KÏlte bei ungefähr 0 bis 8 C ungefähr 2 Stunden lang gerührt. Hiernach, wird das Reaktionsgemenge mit 600 ml Eiswasser, welches 12, 6 g Natriumbikarbonat enthält, verdünnt und das Gemenge zweimal mit 450ml-Portionen Äther zur Entfernung nicht umgesetztes Ausgangsmaterial extrahiert. Die Atherextrakte werden verworfen.
Die wässrige Schicht wird gekühlt und mit 65 ml Schwefelsäure auf einen pH-Wert knapp unter 2 angesäuert. Die wässrige Lösung wird hiernach zweimal mit 450-ml-Portionen ¯ther extrahiert und die vereinigten Atherextrakte mit 100 ml kaltem Wasser gewaschen, über wasserfreiem Natriumsulfat getrocknet, filtriert und mit 75 ml einer 40% igen L¯sung von Kalium-2-äthylhexanoat in n-Butanol behandelt, worauf das Kaliumsalz der 6-(α-Allylthio- propionamino)-penicillansäure sich als ein Íl abschei- det. Das das Reaktionsprodukt enthaltende Gemenge wird mit 2 ml trockenem Äther verd nnt. Der ¯ther wird vom Íl dekantiert und das 01 hiernach mit einem Liter trockenem Ather gewaschen und der Ather dekantiert.
Das Verfahrensprodukt wird dann mit 250 ml trockenem Aceton gemischt und mit trokkenem Ather auf 2 Liter verdünnt. Nach dem Kratzen fÏllt das Produkt aus, wird abfittriert, mit Äther gewaschen, bei Zimmertemperatur im Vakuum ber Phosphorpentoxyd getrocknet und als eine wasserlös- liche, wei¯e, kristalline, feste Substanz im Gewicht von 49, 8 g erhalten. Die Substanz schmilzt bei 180 bis 190 C unter einiger Zersetzung bei ungefähr 160 C.
Sie enthält die ¯-Lactamstruktur, nachweisbar durch Infrarotanalyse und inhibierte Staph. aureus Smith bei einer Konzentration von 0, 5 mcg/ml. Die 50 % ige Heilungsdosis (CDbo) bei intramuskulärer Injektion in Mäusen gegenüber Staph. aureus Smith liegt bei 0, 35 mcg/kg.
Beispiel 2
Das Verfahren des Beispiels 1 wird nachgearbeitet unter Verwendung von a-Butylthiopropionsäure (0, 15 Mol = 24, 3 g) anstelle der α-AllylthiopropionsÏure.
Das Kaliumsalz der 6-(α-Butylthiopropionamido)- penicillansäure wird als eine wasserlösliche, kristalline, feste Substanz enthalten, welche eine ss-Lactamstruk- tur enthÏlt, nachweisbar durch Infrarotanalyse und Staph. aureus Smith bei einer Konzentration von 0, 001 Gew. % inhibiert.
Beispiel 3
Das Verfahren des Beispiels 1 wird nachgearbeitet unter Verwendung von a-Dodecylthio-propion- sÏure (0, 15 Mol = 41, 1 g) anstelle von a-Allylthiopropionsäure. Das Kaliumsalz der 6-(α-Dodecylthio- propionamido)-penicillansaure scheidet sich als was serlösliche kristalline Substanz ab ; es enthält die durch Infrarotanalyse nachweisbare ss-Lactamstruktur und inhibiert Staph. aureus Smith bei einer Konzentration von 0, 001 Gew. %.
Beispiel 4
Das Verfahren gemäss Beispiel 1 wird nachgear- beitet unter Verwendung von a-Methylthiobuttersäure (0, 15 Mol = 20, 1 g) anstelle von a-Allylthiopropion- säure. Das Kaliumsalz der 6- (a-Methylthiobutyr- amido)-peniciHansäure wird erhalten als wasserlös- lidhe kristalline Substanz, welche eine ¯-Lactamstruktur enthält, wie durch Infrarotanalyse nachgewiesen werden kann. Es inhibiert Staph. aureus Smith bei einer Konzentration von 0, 001 Gew. %.
Beispiel 5
Das Verfahren. gemϯ Beispiel 1 wird nachgear- beitet unter Verwendung von a-Isopropylthioisovale- riansäure (0, 15 Mol = 26, 6 g) anstelle von a-Allyl- thiopropionsäure. Das Kaliumsalz der 6- (a-Isopropyl- thioisovaleramido)-penicillansäure wird erhalten als wasserlösliche kristalline Substanz, welche eine ss- Lactamstruktur enfflält, wie durch Infrarotanalyse nachgewiesen werden, kann. Es inhibiert Staph. aureus Smith bei einer Konzentration von 0, 001 Gew. %.
Beispiel 6
0, 01 Mol α-¯thylthiopropionsÏure, 0, 011 Mol Triäthylamin und 0, 01 Mol Isobutylchlorformiat werden in 20 ml reinem, trockenem Dioxan und 2 ml trockenem Aceton während ungefähr 30 Minuten bei etwa 4 C gerührt. Zu dieser L¯sung wird eine abgekü'hlte L¯sung von 0, 01 Mol 6-Aminopenicillan- sÏure und 0, 01 Mol Triäthylamin in 20 ml Wasser hinzugegeben und das Gemisch während 1 Stunde in der Kälte gerührb. Nach Zugabe von 1, 0 g Natrum- bikarbonat in 30 ml kaltem Wasser wird die L¯sung zweimal mit 25 ml Äther extrahiert, und die ätheri- schen Extrakte werden verworfen.
Die wässrige Lösung wird in einem Eisbad abgek hlt und gerührt, überschichtet mit 75 ml ¯ther und ihr pH-Wert mit 5n Schwefel'säure auf 2 eingestellt. Der Ather wird abgetrennt und die wässrige Lösung erneut mit 75 ml ¯ther extrahiert. Die vereinigten ätherischen Extrakte enthalten die 6- (a-Athylthiopropionamido)-penicillan- säure. Sie werden rasch ber wasserfreiem Natrium sulfat getrocknet und filtriert. Zugabe von 6 ml trokkenem n-Butanol, enthaltend 0, 375 g/ml Kalium-2 äthyihexaooat, gefolgt von zusätzlichem trockenem Äther, führt zur Ausfällung des Kaliumsalzes.
Nach dem Verreiben, mit Ather wird das Kaliumsalz im Vakuum ber Phosphorpentoxyd getrocknet und abgeschieden als wassedösliches Pulver, welches das Wachstum von Staph. aureus Smith bei einer Konzentration von 0, 001 Gew. % hemmt.
Beispiel 7
Wird in der Arbeitsweise gemäss Beispiel 1 die u-Allylthiopropionsäure ersetzt durch 0, 15 Mol a-Cyclohexylthiopropionsäure, a-Cyclopentylthiopropionsäure, a-Benzylthiopropionsäure, α-¯thylthio-α-cyclohexylessigsÏure, a-Methylthio-a-cyclopentylessigsaure, a-Hexadecenylbhiobuttersäure, a-Athylthiocapronsäure, α-OctadecylthiopropionsÏure, α-(2-¯thylhexyl)-thiopropionsÏure, α-¯thylthiobuttersÏure, a-Äthylthioisovaleriansäure, a-Benzylthiobuttersäure, a-Benzylthioisovaleriansäure, α-MethylthiobuttersÏure, a-Butylthioisovaleriansäure, a-Benzylthiocapronsäure und α

Beispiel 8
Zu 25 ml = 35, 7 g = 0, 3 Mol mässig am R ckfluss siedenden Thionylchlorid werden 25 g = 0, 143 Mol α-PhenylthiopropionsÏure im Verlauf von 20 Minuten zugesetzt. Das Reaktionsgemisch, wird auf einem Wasserbad 2 Stunden lang erwärmt und wird hiernach beim Vakuum der Wasserstrahlpumpe de stilliert, wobei 21, 1 g von a-Phenylthiopropionylchlo- rid erhalten werden, das einen Siedebereidh von 140 bis 145 C im Vakuum der Wasserstrahlpumpe aufweist.
22, 7 g = 0, 105 Mol 6-AminopenicillansÏure werden in einer L¯sung von 18 g = 0, 215 Mol Natrium- bikarbonat in 225 ml Wasser gelöst und die L¯sung auf ungefähr 5 C abgekühlt. 21, 1 g = 0, 105 Mol des α-Phenylthiopropionylchlorids, dessen Herstellung vorstehend beschrieben wurde, werden in 225 cm3 Aceton gel¯st und die erhaltene L¯sung langsam im Verlauf von 30 Minuten zur L¯sung der 6-Aminopenicillansäure zugesetzt. Die Acylierungsmischung wird zunächst während 30 Minuten in einem Eisbad und anschliessend während 90 Minuten nach Ent fernung der äusserlichen Kühlung gerührt.
Nach dieser Behandlung wird das Reaktions- gemenge mit 300 ml eiskaltem Wasser verdünnt und zweimal mit Ather extrahiert, um nicht umgesetzte Ausgangsprodukte zu entfernen. Die Ätberextrakte werden verworfen. Die wϯrige Schicht wird abgetrennt, mit 400 ml Sither überschichtet, gekühlt und mit 100 ml 6n SchwefelsÏure angesÏuert.
Die Ïtherische Schicht, in welcher das Verfahrensprodukt enthalten ist, wird abgetrennt, zweimal mit kaltem Was- ser gewaschen, über wasserfreiem Natriumsulfat getrocknet, filtriert und mit 50 ml einer 40% igen) Lö- sung von n Kalium-2-Ïthylhexanoat in n-Butanol behandelt, worauf das Kaliumsalz der 6-(a-Phenylthio- propionsäureamido)-penicillansäure ausfällt und abfiltriert wird. Nach dem Verreiben mit Ather wird das Kaliumsalz bei Zimmertemperatur im Vakuum über Phosphorpentoxyd getrocknet und als wasserlösliches Pulver im m Gewicht von 36, 75 g gewonnen.
Es zersetzt sich bei 195 bis 197 C, enthält die ss-Lactam- struktur, wie durch Infrarotanalyse nachgewiesen werden kann, und inhibiert Staph. aureus Smith bei einer Konzentration von 0, 5 mcg/m1. CD50 gegen ber Staph. aureus Smith bei intramuskulärer Injektion in Mäusen = 0, 35 mcg/kg.
Beispiel 9
Das Beispiel 6 wird nachgearbeiteb unter Verwen- dung von a-Phenylt'hiopropionsäure (0, 01 Mol) anstelle von a-Athylthiopropionsäure. Das Kaliumsalz der 6- (α-Phenylthiopropionamido)-penicillansÏure wird erhalten als wasserlösliches Produkt, welches eine ss-Lactamstruktur enthält, wie durch Infrarot- analyse nachgewiesen werden kaan. Es inhibiert das Wachstum von Staph. aureus Smith bei einer Konh zenbration von 0, 5 mcg/ml. CD., gegenüber Staph. aureus Smith bei intramuskulärer Injektion in MÏusen = 0, 35 mcg/kg.
Beispiel 10
Das Verfahren, des Beispiels 6 wird nachgearbeitet unter Verwendung von α-Paranitrophenylthiopropion- sÏure (0, 01 Mol) anstelle von a-Äthylthiopropion- säure. Das Kaliumsalz der 6-(a-Paranitrophenylliio- propionamido)-penicillansäure wird erhalten, als wasserlösliches Pulver, welches das Wachstum von Staph. aureus Smith bei einer Konzentration von 0, 001 Gew. % hemmt.
Beispiel 11
Das Verfahren wird gemäss Beispiel 6 nachgear- beitet unter Verwendung von 0, 01 Mol a-Phenyl- thiobuttersäure anstelle von a-Äthylthiopropionsäure.
Das Kaliumsalz der 6-(α-Phenylthiobutyramido)-peni- cillansäure wird erhalten als wasserlösliches Pulver, welches das Wachstum von Staph. aureus, Smith bei einer Konzentration von 0, 001 Gew. % hemmt.
Beispiel 12
Das Verfahren gemϯ Beispiel 6 wird nachgeafr- beitet unter Verwendung von a-Phenylfhiocapron- sÏure anstelle von a-Athylthiopropionsäure. Das Kaliumsalz der 6-(a-Phenylthiobutyramid'o3-peniicillan- sÏure wird erhalten als wasserlösliches Pulver, welches das Wachstum von Staph. aureus Smith bei einer Konzentration von 0,001 Gew. % hemmt.

Das Verfahren gemäss Beispiel 6 wird nachgear beitet unter Verwendung von 0, 001 Mol a-Phenylthio- isovaleriansäure anstelle von a-Athylthliopropionsäure.
Das Kaliumsalz der 6-(a-Phenylithioisovaleramido) penicillansäure wird erhalten als wasserlösliches Pulver, welches das Wachstum von Staph. aureus Smith bei einer Konzentration von 0, 001 Gew. % hemmt.
Beispiel 14
Zur Herstellung des 6-Aminopenicillansäurederi- vats der a-Parachlorphenylthiopropionsäure werden 22, 7 g = 0, 105 Moli 6-Aminopenicillansäure in einer L¯sung von 18 g = 0, 125 Mol Natriumbikarbonat in.
225 ml Wasser gelöst und die L¯sung auf ungefähr 5 C abgekühlt. Die α-Parachlorphenylthiopropion- sÏure wird mit Thionylchlorid zum a-Paraehlorphe- nylthiopropionylchlorid umgesetzt, wovon 21, 0 g= 0, 105 Mol in 225 cm3 Aceton aufgelöst werden Die erhaltene Mischung wird langsam im Verlauf von ungefähr 30 Minuten der L¯sung der 6-AminopenicillansÏure hinzugegeben. Das Acylierungsgemisoh wird zunächst 30 Minuten lang in einem Eisbad und anschliessend 90 Minuten lang nach Entfernung der äusserlichen Kühlung gerührt.
Nach der vorbeschriebenen Behandlung wird das Reaktionsgemenge mit 300 ml eiskaltem Wasser verd nnt und zweimal mit Ather extrahiert, um nicht umgesetzte Ausgangsprodukte zu entfernen. Die ätherischen, Extrakte werden verworfen. Die wϯrige Schicht wird abgetrennt, mit 400 ml Äther uberschichtet, gekühlt und mit 100 ml 6n Schwefelsäure angesäuert. Die ätherische Lösung, in welcher das Reaktionsprodukt enthalten ist, wird abgetrennt, zweimal mit kaltem Wasser gewaschen, mit wasserfreiem Natriumsulfat getrocknet, filtriert und mit 50 ml einer 40% igen L¯sung von Kalium-2-äthylhexanoat in n-Butanol behandelt, worauf das Kaliumsalz der 6- (a-Parachlorphenylthiopropionamido)-penicillan- säure ausfällt und abfiltriert wird.
Nach dem Verreiben mit Äther wird das Kaliumsalz bei Zimmertemperatur im Vakuum ülber Phosphorpentoxyd getrocknet und als wasserlösliches Pulver gewonnen. Es enthÏlt die ¯-Lactamstruktur, wie durch Infrarotanalyse nachgewiesen wird,. Das Salz inhibiert Staph. aureus Smith bei einer Konzentration von 0, 001 Gew. %.

Furthermore, adherence to strongly acidic or alkaline conditions should be avoided in the process according to the invention. The end product can be extracted with diethyl ether or n-butanol at acidic pH and then obtained by lyophilization, or it can be converted into a solvent-insoluble salt, e.g. B. by neutralization with a n-butanol solution of potassium 2-ethylhexanoate, can be converted. The product can also be precipitated from aqueous solution as the water-insoluble salt of an amine or obtained directly from it by lyophilization.
The latter is preferably done in the form of a sodium or potassium salt. If the product is obtained as the triethylamine salt, it can be converted to the free acid form and then to another salt in the manner known for benzylpenicillin and other penicillinos. Thus, the treatment of such, TriÏthyl aminverbindungen in water with sodium hydroxide gives the sodium salt, after which the triethylamine by extraction, for. B. with toluene, can be deposited.
Treatment of the sodium salt with strong aqueous acids leads to the formation of the acid form, which in turn is converted into other amine salts, e.g. B. the salt of procaine, can be converted by reaction with the appropriate amino base. Salts produced in this way can be separated off by lyophilization or, if they are insoluble, by simple filtration.
One way of working to isolate the product as a crystalline potassium salt is to extract it from an acidic aqueous solution, e.g. B. from pH 2 with diethyl ether, drying the ether extract and adding at least one equivalent of a solution of potassium 2-ethylhexanoate in dry n-butanol. The potassium salt usually precipitates in crystalline form and can be filtered or decanted; be won.
Example I.
