worin die Substituenten X und Y gleich oder ver schieden sind und Halogenatome, Alkyl- oder Alk- oxylgruppen mit 1-3 Kohlenstoffatomen, A eine gerade oder verzweigte Alkylenkette mit 2-6 Kohlen stoffatomen, R1 und R.,
Wasserstoffatome oder Äl- kylreste mit 1-3 Kohlenstoffatomen bedeuten, welche gegebenenfalls mit dem Stickstoffatom ein hetero- cyclisches Ringsystem, das gegebenenfalls noch ein Heteroatom enthält, bilden.
Diese Verbindungen besitzen bedeutende phar- makodynamische Eigenschaften und können zu thera peutischen Zwecken verwendet werden.
Bisher wurden diese Verbindungen z. B. durch Kondensationen 1,3-disubstituierter Phenothiazine mit Dialkylaminoalkylhalogeniden in Gegenwart von alkalischen Kondensationsmitteln, wie Natrium, Na- triumamid, Natriumhydroxyd und ähnlichen, her gestellt, und zwar entweder ohne Reaktionsmedium oder in Gegenwart von inerten organischen Lösungs mitteln, z. B. Benzol, Toluol und ähnlichen.
Die Isolierung der Reaktionsprodukte wird durch Gegenwart von schmierigen und harzigen Nebenpro- dukten, welche sich während der Reaktion bilden, ziemlich erschwert; infolgedessen ist die Gewinnung der Endprodukte in reiner Form mit grossem Arbeits aufwand verbunden.
Das erfindungsgemässe Verfahren ist dadurch ge kennzeichnet, dass man basische Ester der 1,3-disub- stituierten Phenothiazin-10-karbonsäure der Formel

einer thermischen Dekarboxylierung unterwirft. Vor teilhaft führt man die Dekarboxylierung bei Tempe raturen von 150-250 C durch. Diese Operation kann entweder ohne Reaktionsmedium oder in Gegenwart von inerten Lösungsmitteln, z. B. Diphenyl, Chinolin und ähnlichen, durchgeführt werden.
Das Reaktions produkt kann dann in ein Salz übergeführt werden, welches durch Kristallisation weiter gereinigt wird.
Im Vergleich mit den üblichen Darstellungs methoden kann man die Endprodukte nach dem er findungsgemässen Verfahren leichter isolieren und in hoher Reinheit gewinnen.
Das als Ausgangsmaterial dienende basische Ester der Formel 11 lässt sich durch Reaktion von 1,3-di- substituiertem Phenothiazin -10 - karbonsäurechlorid mit einem Aminoalkohol der Formel

gewinnen; das Phenothiazin-10-karbonsäurechlorid ist durch Reaktion von 1,3-disubstituiertem Pheno- thiazin mit Phosgen, z. B. in Benzol- oder Toluol- lösung und in Gegenwart von einem säurebindenden Mittel, z.
B. Pyridin, zugänglich.
Die erfindungsgemäss hergestellten basischen 1,3- disubstituierten Phenothiazinderivate besitzen an ästhetische, hystaminolytische, ganglio- und neuro- plegische, spasmolytische, analgetische und hypo- thermische Wirksamkeit. Sie können in Form ihrer wasserlöslichen Salze mit organischen oder anorga nischen Säuren oder in Form wasserlöslicher An lagerungsverbindungen verabreicht werden.
<I>Beispiel</I> Ein Gemisch von 6,5 g 1,3-Dichlor-phenothiazin- 10-karbonsäurechlorid, 2,2 g 3-Dimethylaminopro- panol-1 und 20 cm3 wasserfreiem Aceton erhitzt man 2 Stunden zum Sieden unter Rückfluss. Nach Abküh len saugt man das ausgeschiedene 1,3-Dichlor-pheno- thiazin - 10 - karbonsäure - (3 - dimethylaminopropyl)- ester-hydrochlorid (Ausbeute 8,2 g, F.
190-193 C unter Zersetzung) ab, löst es in 5 cm3 Wasser auf, macht die Lösung mit einer 20 % igen wässrigen NaOH-Lösung alkalisch und nimmt die freie Base in 30 cm-" Benzol auf. Man dampft den Benzolauszug ein und erhitzt dann den Rückstand (5,6 g) 40 Minu- ten auf 180-200 C (bis die Coz Entwicklung nach lässt).
Nach Abkühlen löst man die ölartige Base in 5 cm3 Äther auf, und durch Zugabe von einer ätheri schen HCl-Lösung fällt man das Hydrochlorid aus, saugt es ab und kristallisiert es aus 5 cm3 Chlor benzol um. Man erhält das 1,3-Dichlor-10-(3'-di- methylaminopropyl) - phenothiazin - hydrochlorid in einer Ausbeute von 2,9 g (53 no. F. 192-193 C.
Das als Ausgangsmaterial dienende 1,3-Dichlor- phenothiazin-10-karbonsäurechlorid kann man z. B. so herstellen, dass man 56,3 g 1,3-Dichlorphenothia- zin unter Rühren in 300 cm3 einer 17,5 % igen Phos- genlösung in Toluol zur Lösung bringt und hierzu unter Wasserkühlung innerhalb etwa 15 Minuten 25,
6 cm3 wasserfreies Pyridin zutropft. Dann rührt man die Reaktionsmischung 2 Stunden bei Zimmer temperatur und weitere 2 Stunden im siedenden Was serbad. Nach Stehen über Nacht giesst man die Toluol- lösung vom ausgeschiedenen Pyridinhydrochlorid ab und engt sie auf etwa 11., des ursprünglichen Volu mens ein. Nach Abkühlen scheidet sich das 1,3-Di- chlor-phenothiazin-10-karbonsäurechlorid in Form von hellgelben Kristallen aus.
Ausbeute 50,4 g (76,4%), F. 142 C (Benzol-Petroläther 1 : 1).
