Verfahren zur Herstellung der Stereoisomeren und des Racemats von
Butan-1,2,3,4-tetrol-1,4-di-(methasulfonat)
Die Erfindung bezieht sich auf ein Verfahren zur Herstellung der Stereoisomeren und des Racemats von Butan-1,2,3,4-tetrol-1 ,4-di-(methansulfonatr mit der folgenden Formel CH3-SO2-O-CH2-CH(OH)-CH(OH)-CH2-O-SO2-CH3.
In Übereinstimmung mit der üblichen Nomenklatur für Butantetrole werden im folgenden die beiden optisch aktiven Isomere mit d-threitol-1,4di-(methansulfonat) bzw. 1-Threitol-1,4-di-(methansulfonat), das Racernat mit dl-Threitol-1,4-di (methansulfonat) und das Mesoisomer mit Erythri tol-1, 4-di-(methansulfonat) bezeichnet. Diese bis jetzt unbekannten Substanzen sind gegen Leukämie wirksam.
Das erfindungsgemässe Verfahren ist dadurch gekennzeichnet, dass ein Isomer oder Racemat von 1,4- Dichiorbutan -2,3 diol, 1 ,4Dibrombutan-2,3- diol oder 1 ,4-Dijodbutan-2,3-diol mit einem Salz der Methansulfonsäure, vorzugsweise dem Silbersalz derselben, oder ein Isomer oder Racemat von 1,2,3,4-Diepoxybutan mit Methansulfonsäure umgesetzt wird.
Die Stereoisomere und die Racemate von 1,4 Dichlorbutan-2,3-diol und 1 ,4-Dibrombutan-2,3diol sind bekannte Substanzen. Die Stereoisomere und das Racemat von 1,4-Dijodbutan-2,3-diol können durch Umsetzung des passenden Isomers oder Racemats von 1,2,3,4-Diepoxybutan mit wässrigem Jodwasserstoff durch ein dem für die Herstellung der Isomere von 1,4-Dibrombutan-2,3-diol von P. W.
Feit in Berichte der Deutschen Chemischen Gesellschaft , Band 93 (1960), Seite 116 beschriebenen Verfahren analoges Verfahren hergestellt werden.
Nach dem Lösen in Aceton und dem Fällen mit Wasser haben die vier so erzeugten 1,4-Dijodbutan- 2,3-diole die folgenden Konstanten: Schmelzpunkt (a) 2D (c = 2, in Methanol)
Meso 131-135 C dl 104-105, 5 C d 105-106,5 C - 8,0"
1 105,5-106,5 C + 7,9"
Beispiel 1 dl-Threitol-1,4-di-(methansulfonat)
Es wurden 12,4 g dl-1,4-Dibrombutan-2,3-diol in 45 cm wasserfreiem Acetonitril gelöst, 30 g des Silbersalzes der Methansulfonsäure zugegeben und die Mischung 1 Stunde lang unter Rücktluss gekocht.
Hierauf wurde das Silberbromid von der heissen Mischung durch Filtrieren getrennt und das Filtrat unter Vakuum zur Trockene eingedampft. Der Rückstand wurde ohne Erhitzen in Aceton zerrieben, welches die erwünschte Substanz löste, jedoch das überschüssige Silbersalz der Methansulfonsäure ungelöst zurückliess. Die letztere Substanz wurde abfiltriert, und die erwünschte Substanz wurde durch Entfernen des Acetons aus dem Filtrat durch Verdampfen unter Vakuum erhalten. Das Rohprodukt wurde aus Äthanol umkristallisiert, in Aceton gelöst und mit Äther gefällt. Der Schmelzpunkt des gereinigten Produktes betrug 100 bis 100, 5 C.
Beispiel 2
Erythritol-1,4-di-(methansulfonat)
Es wurde Meso-1,4-dibrombutan-2,3-diol mit dem Silbersalz der Methansulfonsäure umgesetzt und das Reaktionsprodukt wie im Beispiel 1 isoliert und gereinigt. Hierdurch wurde die erwünschte Substanz mit einem Schmelzpunkt von 120,5 bis 121 C erhalten.
Beispiel 3 d-Threitol-1, ,4-di-(methansulfonat)
Es wurde d-1,4-Dibrombutan-2,3-diol mit dem Silbersalz der Methansulfonsäure umgesetzt und das Reaktionsprodukt wie im Beispiel 1 isoliert und gereinigt. Hierdurch wurde die erwünschte Substanz mit einem Schmelzpunkt von 100 bis 101 C und (a) 2D + 5,50 (c = 2, in Aceton) erhalten.
Beispiel 4 l-Threitol-1,4-di-(methansulfonat)
Es wurde 1-1 ,4-Dibrombutan-2,3-diol mit dem Silbersalz der Methansulfonsäure umgesetzt und das Reaktionsprodukt wie im Beispiel 1 isoliert und gereinigt. Hierdurch wurde die erwünschte Subtsanz mit einem Schmelzpunkt von 100 bis 101 C und (a) 2D0 = -5,3" (c = 2, in Aceton) erhalten.
Beispiel 5
Erythritol-1,4-di-(methansulfonat)
Es wurde zu einer Lösung von 20 g Methansulfonsäure in 50 cm3 Äther eine Lösung von 4,3 g Meso-l ,2,3,4-diepoxybutan in 50 cm3 Ather tropfenweise unter Rühren bei 0 C zugegeben. Die Reaktionsmischung wurde bei Zimmertemperatur 16 Stunden lang stehengelassen, worauf die Ätherlösung von einer öligen Phase dekantiert wurde, aus welcher sich bei Zugabe von Athanol Kristalle ausschieden.
Die Kristalle wurden von der Lösung getrennt und aus Äthanol umkristallisiert. Hierdurch wurde die erwünschte Substanz mit einem Schmelzpunkt von 120,5 bis 121 C erhalten.

In accordance with the usual nomenclature for butanetetrols, the two optically active isomers with d-threitol-1,4-di (methanesulfonate) and 1-threitol-1,4-di- (methanesulfonate), the racernate with dl-threitol 1,4-di (methanesulfonate) and the mesoisomer with erythritol-1,4-di (methanesulfonate). These hitherto unknown substances are effective against leukemia.
The process according to the invention is characterized in that an isomer or racemate of 1,4-dichlorobutane-2,3-diol, 1,4-dibromobutane-2,3-diol or 1,4-diiodobutane-2,3-diol with a salt of methanesulfonic acid , preferably the silver salt thereof, or an isomer or racemate of 1,2,3,4-diepoxybutane is reacted with methanesulfonic acid.
The stereoisomers and the racemates of 1,4 dichlorobutane-2,3-diol and 1,4-dibromobutane-2,3diol are known substances. The stereoisomers and racemate of 1,4-diiodobutane-2,3-diol can be prepared by reacting the appropriate isomer or racemate of 1,2,3,4-diepoxybutane with aqueous hydrogen iodide by a method used to prepare the isomers of 1,4 -Dibromobutane-2,3-diol from PW

Example 1 dl-threitol-1,4-di (methanesulfonate)
12.4 g of dl-1,4-dibromobutane-2,3-diol were dissolved in 45 cm of anhydrous acetonitrile, 30 g of the silver salt of methanesulfonic acid were added and the mixture was refluxed for 1 hour.
The silver bromide was then separated from the hot mixture by filtration and the filtrate was evaporated to dryness in vacuo. The residue was triturated in acetone, without heating, which dissolved the desired substance but left the excess silver salt of methanesulfonic acid undissolved. The latter substance was filtered off, and the desired substance was obtained by removing acetone from the filtrate by evaporation under vacuum. The crude product was recrystallized from ethanol, dissolved in acetone and precipitated with ether. The melting point of the purified product was 100 to 100.5 C.
Example 2
Erythritol 1,4-di- (methanesulfonate)
Meso-1,4-dibromobutane-2,3-diol was reacted with the silver salt of methanesulfonic acid and the reaction product was isolated and purified as in Example 1. As a result, the desired substance having a melting point of 120.5 to 121 ° C was obtained.
Example 3 d-Threitol-1,4-di- (methanesulfonate)
D-1,4-dibromobutane-2,3-diol was reacted with the silver salt of methanesulfonic acid and the reaction product was isolated and purified as in Example 1. This gave the desired substance with a melting point of 100 to 101 C and (a) 2D + 5.50 (c = 2, in acetone).

Example 5
Erythritol 1,4-di- (methanesulfonate)
A solution of 4.3 g of meso-1,2,3,4-diepoxybutane in 50 cm3 of ether was added dropwise with stirring at 0 ° C. to a solution of 20 g of methanesulfonic acid in 50 cm3 of ether. The reaction mixture was left to stand at room temperature for 16 hours, after which the ether solution was decanted from an oily phase from which crystals separated out on addition of ethanol.
The crystals were separated from the solution and recrystallized from ethanol. As a result, the desired substance having a melting point of 120.5 to 121 ° C was obtained.
