Verfahren zur Herstellung von lIarz-Arzneistoffverbilldungen
Die vorliegende Erfindung bezieht sich auf ein Verfahren zur Herstellung neuer Arzneistoffverbindungen und insbesondere neuer therapeutischer Zusammensetzungen mit verbesserten Geruchcharakteristiken. Auf medizinischem Gebiet ist es seit langem bekannt, dass die orale Verabreichung von Arzneistoffen allen anderen Verabreichungsformen im allgemeinen vorzuziehen ist. Es ist ferner auf medizinischem Gebiet bekannt, dass gewisse Arzneimittel einen ausgeprägten Geruch besitzen, der in manchen Fällen so unangenehm sein kann, dass er einen starken Widerwillen des Patienten gegen die Einnahme des betreffenden Arzneimittels hervorruft und dadurch die Anwendungsmöglichkeiten desselben beträchtlich einschränkt.
Es ist daher ein Ziel der vorliegenden Erfindung, ein Verfahren zur Herstellung therapeutischer Präparate mit verbesserten Geruchscharakteristiken zu schaffen, bei welchem ein normalerweise riechender Arzneistoff chemisch derart gebunden ist, dass er im gebundenen Zustand geruchlos ist, ohne dabei seine besonderen therapeutischen Eigenschaften zu verlieren. Gemäss der Erfindung wird ein Verfahren geschaffen zur Verbesserung der Geruchscharakteristiken von normalerweise riechenden Arzneistoffen, welches dadurch gekennzeichnet ist, dass ein normalerweise riechender Arzneistoff mit einem geeigneten Ionenaustauschharz, z. B. einem Harz auf Basis von einem Polymer eines äthylenisch ungesättigten Monomers, unter Bildung einer Harz-Arzneistoffverbindung kombiniert wird.
Die Kombination erfolgt in einer flüssigen Umgebung, z. B. mit dem in Lösung befindlichen Arzneistoff, und die entstandene Verbindung wird anschliessend abgetrennt und getrocknet.
Ein derartiger Arzneistoff von saurer Natur kann mit einem Anionenaustauschharz verbunden werden, um Salze des als Base wirkenden Harzes zu bilden, während der Arzneistoff als Säure wirkt, oder ein Arzneistoff von basischer Natur kann mit einem Kationenaustauschharz verbunden werden, um eine Resinat des Arzneistoffes zu bilden. Der Arzneistoff wird aus seiner Verbindung mit dem Ionenaustauschharz freigesetzt, sobald es mit dem normalen Inhalt des Magens und des Darmtraktes in Berührung kommt.
Die mit dem Harz verbundene Arzneistoffmenge kann geringer sein als zur Sättigung des Harzes erforderlich ist, und der Sättigungsgrad des Harzes mit dem Arzneistoff liegt vorzugsweise zwischen 20 und 95 0/6 Vollsättigung.
Die Teilchengrösse des Harzes beeinflusst den Abgabegrad des Arznei stoffes im Magendarmtrakt, wobei die günstigste Grösse der Teilchen bei Siebfeinheiten zwischen 50 und 400 Maschen liegt.
Die Gestalt der Kurve, die die abgegebene Arzneistoffmenge als Funktion der Zeit darstellt, kann beeinflusst werden durch geeignete Wahl des zu verwendenden Harzes,, durch Veränderung der Gesamtkonzentration des Arzneistoffes in der Resinatverbindung während deren Herstellung und durch Zusatz variierender Mengen reinen Harzes zu der Verbindung, die durch Sättigung des Harzes mit dem Arzneistoff erhalten wurde.
Geeignete Ionenaustauschharze sind im Handel erhältlich (z. B. Zeocarb 225 , Amberlite IR-20 , DeAcidite FF oder Amberlite IRA-400 ) und können entsprechend den funktionellen Gruppen, die der als Harzgrundmasse (resin matrix) bekannten synthetischen, unlöslichen, makromolekularen Verbindung zugeordnet sind, in vier Gruppen eingeteilt werden. Die Harzgrundmasse ist vorzugsweise ein Polymer eines äthylenisch ungesättigten Monomers und ist üblicherweise ein vernetztes Polystyrol, in welchem der Gehalt an Vernetzungen verändert werden kann. Die funktionellen Gruppen der Harzgrundmasse können die folgenden sein: 1. Sulfonsäuregruppen (starkes Kationenaustausch harz), z. B. Zeocarb 225 oder Amberlite IR-20 , oder Dowex 50 ; 2.
Carboxylgruppen (schwaches Kationenaustausch harz), z. B. Zeocarb 226 oder Amberlite IRC-50 ; 3. Quaternäre Ammoniumgruppen (starkes Anionen austauschharz), z. B. DeAcidite FF , Amberlite
IRA-400 oder Dowex 1 und Dowex 2 ; oder 4. Primäre Aminogruppen (schwaches Anionenaus tauschharz), z. B. DeAcidite E oder Amber lite IR-45 .
( Zeocarb 225 ist ein Markenprodukt der Permutit Co. Ltd. und ist ein Polystyrolharz, das mit Divinylbenzol vernetzt ist und Sulfonsäuregruppen enthält, wobei Vernetzungsgrade von 1, 2, 4,5, 8, 12 und 200/al erhältlich sind.
Zeocarb 226 ist ein Markenprodukt der Permutit Co. Ltd. und ist ein Polymethacrylsäureharz, das mit Divinylbenzol vernetzt ist und Carboxylgruppen enthält, wobei der Vernetzungsgrad 2,5 und 4, 5 /e beträgt.
DeAcidite E (Marke) ist ein Polykondensationsharz ohne Vernetzung, das primäre, sekundäre und tertiäre aktive Gruppen enthält.
DeAcidite FF ist ein Markenprodukt der Permutit Co. Ltd. und ist ein Polystyrolharz, das mit aliphatischen Estern vernetzt ist und quaternäre Ammoniumgruppen enthält, wobei die Vernetzungs grade 2 bis 3 30/ei, 3 bis 5 0h und 7 bis 9 9 /o betragen.
Amberlite IR-20 ist ein Markenprodukt der Rohm & Haas Co. und ist ein Harz, das mit Di- vinylbenzol vernetzt ist und Polyaminsulfonsäuregruppen enthält, wobei der Vernetzungsgrad nicht spezifiziert ist.
Amberlite IRA-400 ist ein Markenprodukt der Rohm & Haas Co. und ist ein Polystyrolharz, das mit Divinylbenzol vernetzt ist und quaternäre Ammoniumgruppen enthält, wobei der Vernetzungsgrad nicht spezifiziert ist.
Amberlite IRC-50 ist ein Markenprodukt der Rohm & Haas Co. und ist ein Polymethacrylsäureharz, das mit Divinylbenzol vernetzt ist und Carboxylgruppen enthält, wobei der Vernetzungsgrad nicht spezifiziert ist.

Beispiele von geeigneten Arzneistoffen, die mit einem Ionenaustauschharz gemäss der vorliegenden Erfindung kombiniert werden können, sind Aneurin und Valeriansäure.
Beispielsweise kann Aneurinresinat hergestellt werden, indem man zu 100 ml einer 100/oigen Aneurinhydrochloridlösung 5 g Zeocarb 225 H unter Rühren zusetzt. Der pH-Wert wird überprüft, bis er konstant bleibt. Dann wird das Harz abfiltriert, mit Wasser gewaschen und getrocknet.

Hairzvalerianat kann in ähnlicher Weise hergestellt werden, indem eine 100/oige Valeriansäurelösung und DeAcidite FF (in Chloridform) als Ausgangsstoffe verwendet werden, wobei die Mengen dieselben bleiben.
Bei den nach dem erfindungsgemässen Verfahren hergestellten Produkten kann x , welches den Vernetzungsgrad des Harzes darstellt, im Bereich zwischen 50 und 100 liegen, das heisst, die Vernetzung kann zwischen 20 ovo (eine Moleinheit Divinylbenzol auf 5 Mol Einheiten Styrol) bis 10/a (eine Einheit Divinylbenzol auf 100 Moleinheiten Styrol) variieren.
Das Verhältnis m/n gibt den variablen Sättigungsgrad des Harzes mit dem Arzneistoff an.
Process for the manufacture of larz drug formulations
The present invention relates to a process for the preparation of new drug compounds, and in particular new therapeutic compositions with improved odor characteristics. It has long been known in the medical field that oral administration of drugs is generally preferable to all other forms of administration. It is also known in the medical field that certain medicaments have a distinctive odor which in some cases can be so unpleasant that it causes a strong reluctance in the patient to take the medicament in question and thereby considerably limits the possible uses of the same.
It is therefore an object of the present invention to provide a process for the production of therapeutic preparations with improved odor characteristics, in which a normally smelling medicinal substance is chemically bound in such a way that it is odorless in the bound state without losing its special therapeutic properties. According to the invention there is provided a method for improving the odor characteristics of normally smelling medicinal substances, which is characterized in that a normally smelling medicinal substance is mixed with a suitable ion exchange resin, e.g. B. a resin based on a polymer of an ethylenically unsaturated monomer, is combined to form a resin-drug compound.
The combination takes place in a liquid environment, e.g. B. with the drug in solution, and the resulting compound is then separated and dried.
Such a drug of an acidic nature can be combined with an anion exchange resin to form salts of the base acting resin while the drug acts as an acid, or a drug of basic nature can be combined with a cation exchange resin to form a resinate of the drug . The drug is released from its association with the ion exchange resin as soon as it comes into contact with the normal contents of the stomach and intestinal tract.
The amount of drug associated with the resin can be less than is required to saturate the resin, and the degree of saturation of the resin with the drug is preferably between 20 and 95% saturation.
The particle size of the resin influences the degree of release of the drug in the gastrointestinal tract, with the most favorable size of the particles being between 50 and 400 meshes for sieve fineness.
The shape of the curve, which represents the amount of drug delivered as a function of time, can be influenced by suitable choice of the resin to be used, by changing the total concentration of the drug in the resinate compound during its preparation and by adding varying amounts of pure resin to the compound, obtained by saturating the resin with the drug.
Suitable ion exchange resins are commercially available (e.g. Zeocarb 225, Amberlite IR-20, DeAcidite FF or Amberlite IRA-400) and can be assigned to the synthetic, insoluble, macromolecular compound known as the resin matrix are divided into four groups. The resin base is preferably a polymer of an ethylenically unsaturated monomer and is usually a crosslinked polystyrene in which the content of crosslinks can be changed. Examples of suitable drugs that can be combined with an ion exchange resin according to the present invention are aneurine and valeric acid.
For example, aneurine resinate can be prepared by adding 5 g Zeocarb 225 H to 100 ml of a 100% aneurine hydrochloride solution with stirring. The pH is checked until it remains constant. The resin is then filtered off, washed with water and dried.

Hairzvalerianate can be prepared in a similar manner using a 100% valeric acid solution and DeAcidite FF (in chloride form) as starting materials, the amounts remaining the same.
In the products produced by the process according to the invention, x, which represents the degree of crosslinking of the resin, can be in the range between 50 and 100, i.e. the crosslinking can be between 20 ovo (one mole unit of divinylbenzene per 5 mole units of styrene) to 10 / a ( one unit of divinylbenzene per 100 molar units of styrene).
The ratio m / n indicates the variable degree of saturation of the resin with the drug.
