Verfahren zur Herstellung eines Acylthio-hydr oxy-steroids Die Erfindung bezieht sich auf ein Verfahren zur Herstellung eines Acylthio-hydroxy-steroids, und zwar im besonderen auf ein neues Verfahren zur Herstellung neuer Acylthio-hydroxy-steroide, welche als Zwischenprodukte für die Synthese wirksamer hormonaler Arzneistoffe verwendbar sind.
Man hat in letzter Zeit die biochemische Akti vität schwefelhaltiger organischer Verbindungen fest gestellt, und es sind auch bereits gewisse Versuche unternommen worden, eine Acylthio-Gruppe in den Steroid-Kern einzuführen. Beispielsweise haben R. M. Dodson et a1. über die Anlagerung von Alkanthiol- säuren an J1.-I-3-oxo- und J1.ti-3-oxo-Steroide be richtet.
Entsprechend der vorliegenden Erfindung wurde nun gefunden, dass eine Epoxy-Bindung im Steroid- Kern durch Alkanthiolsäure gespalten und in eine Acylthio- und eine Hydroxyl-Gruppe übergeführt wird.
Das Epoxy-Steroid lässt sich nämlich durch Be handlung mit Alkanthiolsäure zum entsprechenden Steroid mit einer Acylthio-Gruppe und einer Hy- droxyl-Gruppe am Kern umsetzen, und dieses letz tere kann durch weitere Behandlung in das entspre chende Steroid mit wirksamer therapeutischer Akti vität übergeführt werden.
Die Reaktion zur Anlagerung von Alkanthiol- säure an ein Epoxy-Gruppen aufweisendes Steroid ist bisher in der Literatur nirgends beschrieben worden und ist völlig neu. Zudem sind auch das erfindungs gemäss erhaltene Produkt und seine physiologischen Aktivitäten neu.
Dementsprechend liegt das der Erfindung zu grunde liegende Ziel in der Schaffung einer Methode, mit der eine schwefelhaltige Gruppe an einer be stimmten, gewünschten Stelle in den Steroid-Kern eingeführt werden kann. Ein weiteres Ziel liegt in der Darstellung von Acylthio-hydroxy-steroiden und einem entsprechenden Verfahren zu deren Herstel lung sowie zur Spaltung einer Epoxy-Bindung an einem Steroid-Kern.
Das erfindungsgemässe Verfahren zur Herstellung von Acylthio-hydroxy-steroiden besteht demgemäss darin, dass das entsprechende Epoxy-Steroid mit Alkanthiolsäure behandelt und die Epoxy-Gruppen durch Acylthio- und Hydroxyl-Gruppen ersetzt werden.
Das erfindungsgemässe Verfahren ist allgemein anwendbar auf Epoxy-Steroide von Androstan, Pregnan, Cholan und der Cholestan-Reihe. Die An zahl und Stellung der Epoxy-Bindungen hat dabei keinen wesentlichen Einfluss auf die Wirkung der Reaktion.
Die Reaktion nach der vorliegenden Erfindung kann ausgeführt werden, indem man das Epoxy- Steroid mit einem überschuss von Alkanthiolsäure behandelt. Die Operation kann so durchgeführt wer den, dass man das Epoxy-Steroid in der Alkanthiol- säure auflöst und bei Zimmertemperatur stehenlässt; sie kann nötigenfalls jedoch auch in der Hitze erfol gen.
Obschon die Reaktion im Normalfall keinerlei Lösungsmittel erfordert, kann die Verwendung irgend eines geeigneten Lösungsmittels in gewissen Fällen zu vorteilhaften Resultaten führen.
Das erhaltene Produkt ist ein Steroid, welches in denjenigen Stellungen, welche im Ausgangsmate rial durch die Epoxy-Bindung eingenommen wurden, eine Acylthio-Gruppe und eine Hydroxyl-Gruppe besitzt.
Die erhaltenen Acyl-thio-hydroxy-steroide sind daher als Zwischenprodukte für die Synthese medizi nisch anwendbar, schwefelhaltiger Steroide ver wendbar.
Beispielsweise kann das 5cc-Hydroxy-6P-acetyl- thio-3,20-dion, welches durch Anwendung des erfin- dungsgemässen Verfahrens auf 5a,6a-Epoxy-pregnan- 3,20-dion erhalten wird, durch Dehydrierung in das 6ss-Acetylthio-4-pregnen-3,20-dion übergeführt werden. Die progestationale Wirkung dieser Verbindung ist etwas geringer als diejenige von Progesteron. Die pituitäre gonadotropische Hemmung ist jedoch in der Wirkung etwa zehnmal stärker als diejenige von Progesteron.
Es ist daher zu erwarten, dass die Ver bindung für folgende Zwecke verwendbar ist: 1. als Mittel zur Bekämpfung von Menopause-Er- krankungen, 2. als ovulationshemmendes oder antikonzeptionel les Mittel, 3. als Mittel gegen Hypergonadismus oder vorzei tige Pubertät, 4. als hemmendes Mittel bei gewissen pituitären Tumoren, 5. als tumorstatisches Mittel bei Brustkrebs.
In analoger Weise kann auch jedes andere Zwi schenprodukt der vorliegenden Erfindung in das ent sprechende Endprodukt übergeführt werden.
<I>Beispiel 1</I> 1,56 g 3ss-Acetoxy-5a,6a-epoxy-cholestan wurden in 6 ml Athanthiolsäure gelöst, während 15 Stunden unter Rückfluss gekocht und anschliessend mit Äther extrahiert. Das Extrakt wurde mit Wasser gewaschen, getrocknet und eingedampft. Der Rückstand wurde mit Methanol behandelt, und nach Umkristallisieren der erhaltenen rohen Kristalle aus verdünntem Aceton erhielt man 1,37 g 3ss-Acetoxy-5a-hydroxy- 6ss-acetylthio-cholestan als Blättchen mit einem Schmelzpunkt von 135-l37 C.

C 71,5911/9 H 10,091/0 S 6,091/0 <I>Beispiel 2</I> 6,2 g 5u,6a-Epoxy-pregnan-3,20-dion-3,20-di- äthylenketal wurden in 32 ml Äthanthiolsäure ge löst, während 114 Stunden bei Zimmertemperatur stehengelassen und dann im Vakuum eingedampft. Der Rückstand wurde nach Zugabe von 5 ml Essig säure und 20 ml Wasser auf dem Wasserbad erhitzt. Nach Abdestillieren der Essigsäure im Vakuum wur den die ausgefallenen Kristalle durch Filtrieren ge sammelt, mit Wasser gewaschen und getrocknet.
An schliessend wurden die Kristalle in 20 ml Pyridin und 10 ml Essigsäureanhydrid gelöst und erneut auf dem Wasserbad erhitzt. Das durch Zugabe von Wasser erhaltene rohe Produkt wurde aus Aceton umkristallisiert und ergab 4 g 5a-Hydroxy-6ss-acetyl- thio - pregnan - 3,20 - dion als Prismen mit einem Schmelzpunkt von 227-229 C.
Die Mutterlauge wurde auf Aluminiumoxyd chromatographiert. Die mit PetrolätheriBenzol und Benzol allein eluierte Fraktion lieferte 560 mg 6ss Acetylthio - 4 - pregnen - 3,20 - dion als Nadeln mit Schmelzpunkt 175-185 C.
Die mit Benzol;Äther und Äther(Chloroform eluierte Fraktion ergab dann zusätzliche 370 mg 5a - Hydroxy - 6/3 - acetylthio- pregnan-3,20-dion. Die Gesamtausbeute an 5a-Hy- droxy-6ss-acetylthio-pregnan-3,20-dion betrug 4,37 g. IR (Nujol) ,u: 3,09 (OH); 5,86, 5,91 (3,20-dion); 5,96, 8,91 (SAc).

Durch Behandlung des Rückstandes mit Methanol wurden 132 mg rohe Kristalle von 2a-Acetylthio-3i>- hydroxy-5ss,22ss-spirostan mit Schmelzpunkt 200 bis 209 C erhalten. Durch Umkristallisieren aus Aceton erhöhte sich der Schmelzpunkt auf 213-215 C.

Process for the preparation of an acylthio-hydroxy-steroid The invention relates to a process for the preparation of an acylthio-hydroxy-steroid, and in particular to a new process for the preparation of new acylthio-hydroxy-steroids which are more effective as intermediates for synthesis hormonal drugs are usable.
The biochemical activity of sulfur-containing organic compounds has been noted recently, and certain attempts have been made to introduce an acylthio group into the steroid core. For example, R. M. Dodson et al. reports on the addition of alkanethiolic acids to J1.-I-3-oxo and J1.ti-3-oxo steroids.
According to the present invention, it has now been found that an epoxy bond in the steroid core is cleaved by alkanethiolic acid and converted into an acylthio and a hydroxyl group.
The epoxy steroid can be converted into the corresponding steroid with an acylthio group and a hydroxyl group on the core by treatment with alkanethiolic acid, and this latter can be converted into the corresponding steroid with effective therapeutic activity by further treatment will.
The reaction for the addition of alkanethiolic acid to a steroid containing epoxy groups has not been described anywhere in the literature and is completely new. In addition, the product obtained according to the invention and its physiological activities are also new.
Accordingly, the underlying aim of the invention is to provide a method with which a sulfur-containing group can be introduced into the steroid core at a certain, desired location. Another goal is the preparation of acylthio-hydroxy steroids and a corresponding process for their produc- tion and for cleaving an epoxy bond on a steroid core.
The process according to the invention for the production of acylthio-hydroxy steroids accordingly consists in treating the corresponding epoxy steroid with alkanethiolic acid and replacing the epoxy groups with acylthio and hydroxyl groups.
The method according to the invention is generally applicable to epoxy steroids from Androstane, Pregnan, Cholan and the Cholestan series. The number and position of the epoxy bonds has no significant influence on the effect of the reaction.
The reaction of the present invention can be carried out by treating the epoxy steroid with an excess of alkanethiolic acid. The operation can be carried out by dissolving the epoxy steroid in the alkanethiolic acid and leaving it to stand at room temperature; If necessary, however, it can also be done in the heat.
Although the reaction does not normally require any solvent, the use of any suitable solvent can in certain cases lead to advantageous results.
The product obtained is a steroid which has an acylthio group and a hydroxyl group in those positions which were assumed in the starting material by the epoxy bond.
The acyl-thio-hydroxy-steroids obtained are therefore medically applicable as intermediates for the synthesis, sulfur-containing steroids ver usable.
For example, the 5cc-hydroxy-6P-acetyl-thio-3,20-dione, which is obtained by applying the method according to the invention to 5a, 6a-epoxy-pregnan-3,20-dione, can be converted into the 6ss- Acetylthio-4-pregnen-3,20-dione are converted. The progestational effect of this compound is slightly less than that of progesterone. The pituitary gonadotropic inhibition, however, is about ten times more potent than that of progesterone.
It is therefore to be expected that the compound can be used for the following purposes: 1. as a means of combating menopausal diseases, 2. as an ovulation-inhibiting or contraceptive agent, 3. as an agent against hypergonadism or premature puberty, 4 as an inhibiting agent in certain pituitary tumors, 5. as a tumor static agent in breast cancer.
In an analogous manner, any other intermediate product of the present invention can also be converted into the corresponding end product.
<I> Example 1 </I> 1.56 g of 3ss-acetoxy-5a, 6a-epoxy-cholestane were dissolved in 6 ml of ethanthiolic acid, refluxed for 15 hours and then extracted with ether. The extract was washed with water, dried and evaporated. The residue was treated with methanol, and after recrystallization of the crude crystals obtained from dilute acetone, 1.37 g of 3ss-acetoxy-5a-hydroxy-6ss-acetylthio-cholestane were obtained as flakes with a melting point of 135-137 ° C.

The mother liquor was chromatographed on aluminum oxide. The fraction eluted with petroleum ether benzene and benzene alone yielded 560 mg 6ss acetylthio - 4 - pregnen - 3.20 - dione as needles with a melting point of 175-185 C.
The fraction eluted with benzene, ether and ether (chloroform then gave an additional 370 mg of 5a-hydroxy-6/3 -acetylthio-pregnane-3,20-dione. The total yield of 5a-hydroxy-6ss-acetylthio-pregnane-3 , 20-dione was 4.37 g, IR (Nujol), u: 3.09 (OH); 5.86, 5.91 (3.20-dione); 5.96, 8.91 (SAc).
