Verfahren zur künstlichen Kultivierung von Pilzmaterial Es wurde ein Verfahren gefunden, halluzinogen wirksame Pilzarten der Gattungen Stropharia, Cono- cybe, Panaeolus, Amanita, Russula sowie folgender Pilzarten der Gattung Psilocybe:
Psiloeybe semper- viva Heim et Cailleux, Psilocybe caerulescens Murrill var. Mazatecorum Heim, Psilocybe Zapote- corum Heim und Psiloeybe Aztecorum Heim im La boratorium unter solchen Kulturbedingungen zu züchten, dass dabei aktives Ausgangsmaterial in Mengen gebildet wird,
die für die Gewinnung der Wirkstoffe Psilocybin und Psilocin in präparativem Massstab ausreichen.
Aus künstlich gezüchtetem Pilzmaterial, erhal ten aus Reinkulturen oder aus biologischen Varianten oder Mutanten von Pilzen der Gattungen Stropharia, Conocybe, Panaeolus, Amanita, Russula sowie der folgenden Pilzarten der Gattung Psilocybe:
Psi'locybe semperviva Heim et Cailleux, Psilocybe caerulescens Murrill var. Mazatecarum Heim, Psilocybe Zapote- corum Heim, Psilocybe Aztecorum Heim wurde durch Impfung derselben auf natürlichem oder künst lichem Nährboden und Bebrütung dieser Kulturen am Tageslicht oder im Dunkeln bei einer konstanten Temperatur zwischen 18 und 27 ,
die für die Gewin nung der Wirkstoffe Psilocybin und Psilocin in prä- parativem Massstab technisch brauchbare Menge an aktivem Ausgangsmaterial erhalten.
Das erfindungsgemässe Verfahren wird vorzugs weise folgendermassen ausgeführt: Zur Züchtung auf natürlichem Substrat wird ein Kompost aus gego renem Weizenstroh, einem Gemisch von Maislaub und -Stengeln oder -Halmen von wilden Gräsern hergestellt, mit fliessendem Wasser reichlich gewa schen, in Tonschalen verteilt und im Autoklagen sterilisiert.
Der Kompost wird mit Mycel aus Pri märkulturen beimpft, während etwa 2 Wochen bei 24 bis 27 bebrütet, die Kulturen mit sterilem Sand gedeckt und im Glaskasten im Tageslicht bei einer Temperatur von 18 bis 27 bei Konstanthaltung des Feuchtigkeitsgehaltes sich selbst über'l'assen. Nach 4 bis 5 Wochen erscheinen die Fruchtkörper; die Ernte erfolgt ab und zu während eines bis zwei Monaten bei beginnender Sporenbildung.
Da die Gewinnung grösserer Mengen Ausgangs material auf diesem Wege jedoch etwas umständ lich ist, wurden andere Züchtungsmethoden ausge arbeitet. Dabei wurde unerwarteterweise gefunden, dass die Pilze in vitro auf nährstoffreichen Substraten anstelle von Mycel mit Fruchtkörpern fast ausschliess lich aktives Mycel und teilweise Sklerotien in grosser Menge bilden, auf nährstoffarmen dagegen die be kannten Fruchtkörper.
Zum Beispiel auf einem Agar- Nährboden (mit 1,5 11/o Agar) liegt das Optimum für die Fruchtkörperbildung bei einer Konzentration an Malzextrakttrockensubstanz von 0,2 bis 0,7"/o,
das Optimum für die Mycel- und Sklerotienbildung da- gegen bei Konzentrationen von 4 bis 10% je nach Pilzart.
Während das Tageslicht für die Fruchtkörper bildung unentbehrlich ist, wurde hingegen gefunden, dass bei Bebrütung der Kulturen im Dunkeln die Sklerotien- bzw. Mycelbildung wesentlich üppiger ist.
Die besten Ausbeuten an aktivem Pilzmaterial (Mycel und Sklerotien) werden erhalten, indem man Oberflächenkulturen mit Malzextrakt-Nährlösungen (Bierwürze oder käufliche Malzextraktpräparate) von 4 bis 101/o Gehalt an Trockensubstanz anlegt und diese Kulturen im Dunkeln bei einer konstanten Temperatur zwischen 22 und 2f bebrüten lässt.
Für ein gutes Wachstum muss der Nährlösung 0,2 % Agar zugesetzt werden: In diesem noch beinahe flüs sigen Medium findet der Pilz gerade genügend Halt, um rasch eine gut geschlossene Mycel'decke zu bil den.
Der Zusatz von Eisen-II-Salzen ist notwendig, ferner erweisen sich Zusätze von Zink-, Kalium-, Calcium- und Magnesium-Salzen von Nitrat-, Phos phat- und Sulfat-Ionen sowie von Hefeextrakt oder sogenannten Cornsteep Solids (Konzentrat von Maisquellwasser) als sehr vorteilhaft.
Dieses Züch tungsverfahren bedeutet einen grossen technischen Fortschritt, da es ermöglicht, in kürzerer Zeit und unter kleinerem Arbeitsaufwand eine - im Vergleich mit der Züchtung auf natürlichem Substrat - mehr als zehnfache Ausbeute an aktivem Ausgangsmaterial zu erhalten, das für die Isolierung des psychotrop wirksamen Psilocins und Psilocybins verwendet wer den soll.
<I>Beispiel 1</I> Reinkulturen von Psilocybe semperviva Heim et Cailleux a) Herstellung des Impfstoffes: Man fegt von Basidiosporen des mexikanischen Hutpilzes Psilocybe semperviva Heim et Cailleux Reinkulturen auf Bierwürze-Agar an. Zu diesem Zweck werden die von den Lamellen eines reifen Fruchtkörpers abfallenden Sporen auf einer sterilen Unterlage aufgefangen und auf Bierwürze-Agar ge bracht und bebrütet.
Aus den so entstandenen Pri märkulturen wird das Impfungsmaterial in genügen der Menge folgendermassen hergestellt: Das Mycel wird unter sterilem Leitungswasser mit einem rauhen Spatel abgeschabt, so dass eine Suspension feiner Mycel-Flocken entsteht. Mit dieser Suspension werden Erlenmeyer-Kolben beimpft, welche eine Nährlösung und sattelförmige Porzellan- fül'Ikörper, wie sie zur Füllung von Destillations- kolonnen gebräuchlich sind, enthalten.
Beste Er fahrungen machten wir mit 300-cm3-Erlenmeyern, enthaltend 50 g Sattelfüllkörper (Grösse 1 cm, Ge wicht etwa 0,9 g) und 80 cm3 Nährlösung, be- stehend aus Bierwürze von etwa 4% Trockensubstanz und 0,2%. Agar.
Die Kultur wird bei einer Temperatur von 24 bebrütet; nach zwei Wochen hat sich auf der Ober fläche eine kompakte Myceldecke gebildet. Die ganze Kultur wird 30 Minuten lang auf der rotierenden Schüttelmaschine geschüttelt, wobei die Sattelfüll- körper durch ihre scharfen Kanten das Mycel zer mahlen: Es entsteht dabei eine feine Suspension von Mycelflocken. Der so erhaltene Impfstoff genügt zur Beimpfung von 50 Liter Nährlösung.
b) Herstellung der Kultur: Frische, helle Bierwürze ohne Hopfenzusatz wird mit Leitungswasser auf einen Gehalt von 8 % an Trockensubstanz verdünnt.
Zu jedem Liter dieser Lösung fügt man:
