Verfahren zur Herstellung von bisher unbekannten Nortropinon-Derivaten
Es wurde gefunden, dass man zu bisher unbekann- ten Nortropinon-Derivaten der Formel I,

worin Ri eine Acyloxyalkylgruppe und R2 einen niederen Alkylrest bedeuten, gelangen kann, indem man die nach dem Hauptpatent erhaltenen 1-Oxyalkyl- nortropinon-Derivate acyliert.
Gegenstand des Hauptpatentes ist ein Verfahren zur Herstellung von l-Oxyalkyl-nortropinon-Deriva- ten. Dieses ist dadurch gekennzeichnet, dass y-Keto- aldehyde mit Acetondicarbonsäure und primären Aminen, kondensiert werden. Die vorliegende Erfindung betrifft eine Weiterbildung des im Hauptpatent beschriebenen Verfahrens. Bei der Weiterbearbeitung des Erfindungsgedankens wurde nämlich gefunden, dass es gelingt, die Hydroxylgruppe in 1-Oxyalkyl nortropinon-Derivaten durch Behandeln mit reak tionsfähigen Derivaten organischer Säuren zu acylieren. Die Acylierung kann nach bekannten Metho- den ausgeführt werden, z.
B. durch Behandlung mit einem Säurehalogenid in Gegenwart oder bei Abwesenheit eines säurebindenden Mittels, durch Behandlung mit einem Säureanhydrid oder auch durch Alkoholyse, das heisst indem man auf alkoholartige Nortropinon-Derivate einen Säureester unter dem katalytischen Einfluss eines Alkalimetalls einwirken lässt.
Die so erhaltenen basischen Ester sind wertvolle Zwischenprodukte für die Herstellung von Medi kamenten.
Beispiel I
40, 5 g 2, 5-Dimethoxy-2-oxymethyl-tetrahydro- furan werden in 390 cm3 0, 1n-Schwefelsäure eine halbe Stunde in eine Mischung von 75 g Acetondicarbonsäure (neutralisiert in 440 cm3 Wasser mit 52, 2 g Kalk) und 240 cm3 35 /siger Methylamin- lösung unter Rühren eingetropft. Die Reaktionsmischung wird drei Stunden weitergerührt und dann zwei Tage stehengelassen, wonach sie bei etwa 40 im Vakuum bis zur Trockne eingedampft wird. Der Rückstand wird in so viel n-Schwefelsäure aufgenom- men, dass die trübe Lösung kongosauer reagiert.
Die zunächst starke CO2-Entwicklung lϯt sich durch ErwÏrmen auf dem Wasserbad zu Ende führer, worauf man die Flüssigkeit auf Zimmertemperatur abkühlt und vom ausge-schiedenen Caiciumsulfat abfiltriert. Das hellbraune Filtrat wird mit Kalilauge stark alkalisch gemacht, mit Natriumchlorid gesättigt, erneut filtriert und während drei Tagen mit Ather extrahiert. Beim Abdampfen des Athers bleiben 25 g rohe Base zur ck, welche nach der Destillation im Hochvakuum in 100 cm3 Alkohol aufgenommen und mit der berechneten Menge alkoholischer Pikrinsäure heil3 versetzt wird, Beim Abkühlen kristallisieren 46, 5 g Pikrat.
Nach mehrmaligem Umkristallisieren schmilzt das so erhaltene Pikrat des 1-Oxymethyl-tropinons bei 191 (Zers.). Man stellt aus 1-Oxymethyl-tropinonpikrat in übMcher Weise die freie Base dar. Hierauf werden 3 g 1-Oxymethyl-tropinon mit 4, 1 g Diphenylessig- säurechlorid in Pyridinlösung versetzt. Nach sechs Stunden wird das Pyridin im Vakuum entfernt, der Rückstand mit Wasser verrührt und mit der entsprechenden Natronlauge alkalisch gemacht. Die alkalische Lösung wird mit Chloroform ausgeschüttelt.
Nach dem Trocknen des Chloroformextraktes und Entfernen des Lösungsmittels wird das zurüokgeblie- bene 01 im Hochvakuum destilliert. Kp. 160 bis 170 / 0, 06 mm. Aus dem Ester wird in Alkohollösung mit der berechneten Menge Pikrinsäure das Pikrat des 1-Oxymethyl-tropinon-diphenylessigsäureesters hergestellt. F. = 187 (Zers.).
Beispiel 2
Durch Behandlung mit Acetanhydrid in Pyridin analog Beispiel 1 wird 1-Oxymethyl-tropinon acetyliert. Das so erhaltene 1-Acetoxymethyl-tropinon der Formel

siedet bei 105 bis 110 (0, 04 mm). Das Pikrat schmilzt, aus Alkohol umkristallisiert, bei 165 .

The subject of the main patent is a process for the production of l-oxyalkylnortropinone derivatives. This is characterized in that γ-ketoaldehydes are condensed with acetone dicarboxylic acid and primary amines. The present invention relates to a further development of the method described in the main patent. In the further processing of the inventive idea it was found that it is possible to acylate the hydroxyl group in 1-oxyalkylnortropinone derivatives by treatment with reactive derivatives of organic acids. The acylation can be carried out by known methods, eg.
B. by treatment with an acid halide in the presence or absence of an acid-binding agent, by treatment with an acid anhydride or by alcoholysis, that is, by allowing an acid ester to act on alcohol-like nortropinone derivatives under the catalytic influence of an alkali metal.
The basic esters obtained in this way are valuable intermediates for the production of drugs.
Example I.
40.5 g of 2,5-dimethoxy-2-oxymethyl-tetrahydrofuran are in 390 cm3 of 0.1N sulfuric acid for half an hour in a mixture of 75 g of acetone dicarboxylic acid (neutralized in 440 cm3 of water with 52.2 g of lime) and 240 cm3 of 35 / s methylamine solution was added dropwise with stirring. The reaction mixture is stirred for a further three hours and then allowed to stand for two days, after which it is evaporated to dryness at about 40 in vacuo. The residue is taken up in so much n-sulfuric acid that the cloudy solution reacts acidic to the Congo.
The initially strong development of CO2 can be brought to an end by warming up on a water bath, whereupon the liquid is cooled to room temperature and the calcium sulphate which has separated out is filtered off. The light brown filtrate is made strongly alkaline with potassium hydroxide solution, saturated with sodium chloride, filtered again and extracted with ether for three days. When the ether is evaporated, 25 g of crude base remain, which is taken up in 100 cm3 of alcohol after distillation in a high vacuum and mixed with the calculated amount of alcoholic picric acid. 46.5 g of picrate crystallize on cooling.
After repeated recrystallization, the picrate of 1-oxymethyl-tropinone thus obtained melts at 191 (dec.). The free base is prepared from 1-oxymethyl-tropinone picrate in the usual way. 3 g of 1-oxymethyl-tropinone are then mixed with 4.1 g of diphenyl acetic acid chloride in pyridine solution. After six hours, the pyridine is removed in vacuo, the residue is stirred with water and made alkaline with the appropriate sodium hydroxide solution. The alkaline solution is extracted with chloroform.
After the chloroform extract has been dried and the solvent has been removed, the oil that remains is distilled in a high vacuum. 160 to 170 / 0.06 mm. The picrate of the 1-oxymethyl-tropinone-diphenylacetic acid ester is prepared from the ester in alcohol solution with the calculated amount of picric acid. F. = 187 (dec.).
Example 2
1-Oxymethyl-tropinone is acetylated by treatment with acetic anhydride in pyridine as in Example 1. The 1-acetoxymethyl-tropinone of the formula obtained in this way
