Verfahren zur Herstellung von 16a-Alkyl-20-keto-steroiden Es ist bekannt, dass man 416-20-Ketone der Pregnanreihe durch Umsetzung mit Alkylmagnesium- halogeniden in Gegenwart von Kupfer-(1)-halogeni- den in 16a-Alkyl-20-keto-pregnane überführen kann. Nach den in der Literatur beschriebenen Angaben für diese Reaktion erhält man aber nur in einzelnen Fällen befriedigende Ausbeuten.
Bei der Übertragung dieser Reaktion auf das A16-3 Hydroxy-20-keto- pregnen bzw. seine Ester beträgt beispielsweise die Ausbeute an der 16a-Alkylverbindung nicht mehr wie etwa 2004.
Es wurde nun gefunden, dass man 416-20-Keto- pregnene und d16-20-Keto-allopregnene sowie ihre Derivate, die im Ringsystem eine bzw. mehrere wei tere Doppelbindungen enthalten, durch Behandlung mit Alkylmagnesiumhalogeniden in Gegenwart von Kupfer-(1)-halogeniden in hoher Ausbeute in die ent sprechenden 16a-Alkyl-20-keto-steroide überführen kann, wenn man die Umsetzung mit dem Alkyl- magnesiumhalogenid und dem Kupfer-(1)
-halogenid in Gegenwart von Tetrahydrofuran und in Abwesen heit von Alkylhalogenid durchführt.
Nach diesem Verfahren gelingt es, in beliebigen Z116-20-Keto-pregnenen und -allopregnenen sowie in ihren obengenannten Derivaten in 16a-Stellung einen Alkyl-, insbesondere einen niederen Alkyl-, z. B. den Methylrest, in hoher Ausbeute einzuführen. Spezi fische Ausgangsstoffe sind z. B.
j16-3fl- und -3a-Hydroxy-20-keto-pregnen und -allopregnen, j 16-3f'-Hydroxy-11,20-diketo-allopregnen, J 15-3ss,11 a-Dihydroxy-20-keto-allopregnen, J 16-3a-Hydroxy-11,20-diketo-pregnen, J 16_3fl-Hydroxy-12,20-diketo-allopregnen, J 16-3 a-Hydroxy-12,20-diketo-pregnen, 45,
16-3f Hydroxy-20-keto-pregnadien, A5,16_3ss_Hydroxy-11,20-diketo-pregnadien, A5,16-3ss, 11 a-Dihydroxy-20-keto-pregnadien, 45,16-3ss-Hydroxy-12,20-diketo-pregnadien, 46 (11),16-3ss-Hydroxy-20-keto-allopregnadien, A9(11),16-3a-Hydroxy-20-keto-pregnadien, ihre 3-Ester und 3-Äther, und entsprechende Ver bindungen, die in 21-Stellung eine freie, veresterte oder verätherte Hydroxygruppe aufweisen.
Von den in 21-Stellung substituierten Verbindungen sind ins besondere zu nennen das 45,16-3ss,21-Dihydroxy-20- keto-pregnadien und das 45#16-3-Athylen-dioxy-21- hydroxy-11,20-diketo-pregnadien.
Als Alkylinagnesiumverbindungen verwendet man in erster Linie niedere Alkylmagnesiumhalogenide, z. B. Methylmagnesiumjodid. Bei der Durchführung der verfahrensgemässen Reaktion ist darauf zu achten, dass in der verwendeten Alkylmagnesiumhalogenid- Lösung kein Alkylhalogenid mehr vorhanden ist, was leicht durch Ausdestillieren, eventuell unter Verwen dung eines zusätzlichen Lösungsmittels, z. B. Benzol, oder durch Anwendung eines überschusses an Ma gnesium erreicht werden kann.
Als Kupfer-(1)-halo- genide können die für diese Reaktion üblichen Ver bindungen, insbesondere Kupfer-(1)-chlorid, verwen det werden.
Die Umsetzung wird normalerweise bei Raum temperatur durchgeführt. Die Aufarbeitung der Re aktionsmischung kann in an sich bekannter Weise erfolgen, z. B. durch Zersetzung mit verdünnten Säuren, wie Salzsäure und Schwefelsäure, oder mit Ammoniumchlorid-Lösung. Die erhaltenen 16a-A1- kyl-20-keto-steroide mit freien Hydroxygruppen in 3- bzw. 3- und 21-Stellung lassen sich, wenn er wünscht, in bekannter Weise in ihre Ester undloder Äther überführen.
Die nach dem vorliegenden Verfahren erhaltenen 16a-Alkyl-20-keto-steroide dienen als Zwischen produkte, in erster Linie zur Herstellung von Cortico- steroiden mit einem 16ra-Alkylrest, wie z. B. 16a-Me- thyl-9a-fluor-prednisolon und -prednison.
Die Temperaturen sind in den nachfolgenden Bei spielen in Celsiusgraden angegeben.
<I>Beispiel 1</I> Eine Grignard-Lösung aus 2 g Magnesium und 7 em3 Methyljodid in 100 cm3 abs. Äther versetzt man mit 150 cm3 reinem Tetrahydrofuran. Unter Calciumchlorid-Verschluss destilliert man den Äther und den Methyljodid-Überschuss vollständig ab. Die erhaltene Methyhnagnesiumjodid-Suspension in Tetra- hydrofuran kühlt man auf 20 ab und füllt das Re aktionsgefäss mit sauerstofffreiem, trockenem Stick stoff.
Nun werden 400 mg Kupfer-(1)-chlorid zuge geben. Unter Rühren und unter Stickstoff lässt man dann bei der gleichen Temperatur eine Lösung von 5 g d 16 - 3ss - Acetoxy -1l,20 - diketo - allopregnen in 50 cms reinem Tetrahydrofuran zutropfen. Die Lö sung rührt man noch 30 Minuten, kühlt sie dann von aussen mit Eis ab und zersetzt vorsichtig mit einer wässrigen Ammoniumchlorid-Lösung. Das erhaltene Gemisch verdünnt man mit Äther,
wäscht die äthe rische Lösung mit einer Ammoniumchlorid-Lösung, Wasser, einer Natriumthiosulfat-Lösung und Wasser, trocknet und dampft sie ein. Der amorphe Rückstand stellt das rohe 3f'-Hydroxy-11,20-diketo-16a-methyl- allopregnan dar. Es wird in 10 cms trockenem Pyri- din und 20 cm?, Acetanhydrid gelöst und 15 Stunden bei 20 stehengelassen. Die Lösung versetzt man dann mit Wasser, engt die Lösung im Vakuum ein und nimmt den Rückstand in Äther auf.
Die ätherische Lösung wäscht man mit verdünnter Salz säure und Wasser, trocknet und dampft sie ein. Der Rückstand wird aus Äther allein, Äther-Pentan- oder Äther-Isopropyläther-Gemisch umkristallisiert und gibt 4,58 g des 3fl'-Acetoxy-16a-methyl-11,20-diketo- allopregnans. Es schmilzt bei 142-143 , kristallisiert dann wieder zu breiten Prismen, die endgültig bei 153 schmelzen; [a]27 = + 80 (in Chloroform);
IR-Spektrum in Methylenchlorid: Banden u. a. bei 5,79 1t, 5,86,u, <I>7,20</I> ,u (Schulter),<I>7,26</I> ,u (Schulter), 7,35,u, 8,13 /t und 9,74 p.
<I>Beispiel 2</I> Eine Grignard-Lösung aus 7 g Magnesium und 30 cm3 Methyljodid in 200 cm3 abs. Äther versetzt man mit 300 cm3 reinem Tetrahydrofuran. Unter Calciumehlorid-Verschluss destilliert man den Äther und den Methyljodid-überschuss vollständig ab.
Die erhaltene Methyhnagnesiumjodid-Suspension in Tetra- hydrofuran kühlt man auf 20 ab und füllt das Reak tionsgefäss mit sauerstofüreiem, trockenem Stick stoff. Nun wird 1 g Kupfer-(1)-chlorid zugegeben.
Unter Rühren und unter Stickstoff lässt man dann bei der gleichen Temperatur eine Lösung von 10 g 416-3ss,lla-Diacetoxy-20-keto-allopregnen in 150 cm3 Tetrahydrofuran zutropfen. Die Lösung rührt man dann noch 30 Minuten, kühlt sie dann von aussen mit Eis ab und zersetzt vorsichtig mit einer wässrigen Ammoniumchlorid-Lösung. Das erhalt--ne Gemisch verdünnt man mit Äther, wäscht die ätherische Lö sung mit einer Ammoniumchlorid-Lösung, Wasser, einer Natriumthiosulfat-Lösung und Wasser,
trocknet und dampft sie ein. Der amorphe Rückstand stellt das rohe 3fl,11 a - Dihydroxy - 16a - methyl-20-keto- allopregnan dar. Es wird in 40 cm3 Pyridin und 20 cm3 Acetanhydrid gelöst und 15 Stunden bei 20 stehengelassen. Die Lösung versetzt man dann mit Wasser, engt sie im Vakuum ein und nimmt den Rückstand in Äther auf. Die ätherische Lösung wäscht man mit verdünnter Salzsäure und Wasser, trocknet und dampft sie ein.
Der kristalline Rück stand (10,3 g) wird aus Methanol umkristallisiert, wobei 8,6 g des 3/3,11 a-Diacetoxy-16a-methyl-20- keto-allopregnans vom F. 172-174 erhalten werden. IR-Spektrum in Methylenchlorid: Banden u. a. bei 5,79 ,u, <I>5,86</I> ,et, 7,20 @t (Schulter),<I>7,26</I> ,c1 (Schulter), 7,34 ,u., 8,13 , < t, 8,64<I>,et</I> und 9,76 P.. Aus den Mutter laugen lassen sich weitere 0,74 g der gleichen Ver bindung gewinnen.
<I>Beispiel 3</I> Eine Grignard-Lösung aus 6,9g Magnesium und 25 em3 Methyljodid in 200 cm- abs. Äther versetzt man mit 200 cm-' reinem Tetrahydrofuran. Unter Calciumchlorid-Verschluss destilliert man den Äther und den Methyljodid-überschuss vollständig ab. Die erhaltene Methylmagnesiumjodid-Suspension in Te- trahydrofuran kühlt man auf 20 ab und füllt das Reaktionsgefäss mit sauerstofffreiem, trockenem Stickstoff.
Nun werden 600 mg Kupfer-(1)-chlorid zugegeben. Unter Rühren und unter Stickstoff lässt man dann bei der gleichen Temperatur eine Lösung von 10 g d5#16-3fl-Acetoxy-20-keto-pregnadien in 100 cm3 reinem Tetrahydrofuran zutropfen. Die Lö sung rührt man noch 30 Minuten, kühlt sie dann von aussen mit Eis ab und zersetzt vorsichtig mit einer wässrigen Ammoniumchlorid-Lösung. Das erhaltene Gemisch verdünnt man mit Äther, wäscht die äthe rische Lösung mit einer Anunoniumchlorid-Lösung, Wasser,
einer Natriumthiosulfat-Lösung und Wasser, trocknet und dampft sie ein. Der Rückstand wird aus Aceton umkristallisiert, wobei 8,48 g des J5-3fl- Hydroxy-16a-methyl-20-keto-pregnens erhalten wer den; es schmilzt ab 182 , kristallisiert zum Teil wie der und schmilzt definitiv bei 188 .
2 g dieser Verbindung werden in 10 cm3 Pyridin und 20 cm3 Acetanhydrid in der Wärme gelöst. Die Lösung lässt man 15 Stunden bei 20 stehen, zersetzt die Lösung mit Wasser und dampft sie im Vakuum ein. Den Rückstand löst man in Äther, wäscht die ätherische Lösung mit verdünnter Salzsäure und Wasser, trocknet und dampft sie ein.
Der Rückstand wird aus Aceton-Isopropyläther-Gemisch umkristalli- siert, wobei 1,9 g des J5-3fl-Acetoxy-16-methyl-20- keto-pregnen vom F. 182-184 erhalten werden; [a120 = 0 (Chloroform); IR-Spektrum in Methylen- chlorid: Banden u. a. bei 5,76 ,u, 5,86 ,u, <I>7,20</I> ,u (Schulter), 7,27,u, 7,34,u und 9,70 ,1c.
<I>Beispiel 4</I> Ausgehend von 5 g dis - 3a - Acetoxy -11,20- diketo-pregnan und unter Verwendung des im Bei spiel 1 beschriebenen Verfahrens erhält man 4,6 g 3a - Acetoxy -16a - methyl-11,20-diketo-pregnan, das nach Kristallisation aus einem Äther-Pentan-Gemisch bei 157-158 schmilzt; [a]D = + 118 (Chloroform).
<I>Beispiel 5</I> Unter Verwendung des im Beispiel 3 beschrie benen Verfahrens erhält man, ausgehend von 10 g 416-3ss-Acetoxy-20-keto-pregnen, 8,3 g 3ss-Hydroxy- 16a - methyl - 20 - keto - pregnan vom F. 203-205 . Durch Acetylierung gewinnt man daraus das 3ss- Acetoxy-16a-methyl-20-keto-pregnan vom F. 139 bis 142 , [a]D = + 93" (Chloroform).
In analoger Weise erhält man, ausgehend vom 45,1s - 3ss21 - Diacetoxy - 20 - keto -pregnadien, das A5-3ss,21 -Dihydroxy -16a - methyl-20-keto-pregnen, das bei der Acetylierung das 45-3ss,21-Diacetoxy- 16a-methyl-20-keto-pregnen vom F. 160-170 (aus Hexan) liefert.
<I>Beispiel 6</I> Je 30 cms trockenes, peroxydfreies Tetrahydro- furan und trockenes Toluol werden mit 10 cm3 einer 2,14-molaren Methylmagnesiumjodidlösung (mit grossem Magnesiumüberschuss bereitet aus 4 g Ma gnesiumspänen, 3,2 cm3 Methyljodid und 53 cm3 Äther und Abdestillieren des grösseren Teils Äther)
in einer Stickstoffatmosphäre zusammengegeben und erwärmt, bis die Temperatur im Gemisch 78 be trägt, wobei 17 cm3 Lösungsmittel abdestillieren. Nach Abkühlen auf 25 wird 100 mg trockenes Kupfer-(1)-chlorid zugesetzt und während 5 Minuten mit dem Grignard-Gemisch gerührt.
Durch das Ab destillieren von Lösungsmittel sowie durch drei ent nommene Proben des Reaktionsgemisches zu je 4 cm3 vermindert sich dessen Volumen auf 41 cm3. Die am Ende der Vorbehandlung entnommene Probe entwickelt mit 256loiger Ammoniumchloridlösung 20,7 cm3 Methan, was einem Gehalt des Reaktions gemisches an Grignard-Komplex von 9,47 Millimol entspricht.
Nun lässt man bei -10 bis 0 eine Lösung von 1,6 g 16-Dehydro-progesteron in 30 cm3 Toluol auf einmal zufliessen und nach Entfernen der äussern Kühlung während 30 Minuten bei -10 bis + 12 reagieren. Das Aufarbeiten erfolgt durch Vermischen mit überschüssiger Ammoniumchloridlösung unter Zusatz von wenig Natriumthiosulfat, Aufnehmen in Benzol, Trocknen und Abdestillieren der Lösungs mittel im Vakuum. Beim Vermischen des Roh produktes mit wenig Äther wird 1,2 g kristallines 16a-Methyl-progesteron gewonnen.
Die chromato- graphische Reinigung der Mutterlauge liefert weitere 0,25 g. Das aus n-Hexan umkristallisierte Produkt schmilzt bei 137-138 ; E 241 M = 14 400. IR-Spek- trum: 5,87,u (20-Keton) und 5,98 y <I>+</I> 6,17 ,u (44-3-Keton).
Process for the preparation of 16a-alkyl-20-keto-steroids It is known that 416-20-ketones of the pregnane series can be converted into 16a-alkyl-20 by reaction with alkylmagnesium halides in the presence of copper (1) halides -keto-pregnane. According to the information for this reaction described in the literature, however, satisfactory yields are only obtained in individual cases.
When this reaction is transferred to the A16-3 hydroxy-20-keto-pregnen or its esters, for example, the yield of the 16a-alkyl compound is no longer as in 2004.
It has now been found that 416-20-keto-pregnene and d16-20-keto-allopregnene and their derivatives, which contain one or more additional double bonds in the ring system, by treatment with alkyl magnesium halides in the presence of copper (1) Halides can be converted into the corresponding 16a-alkyl-20-keto-steroids in high yield if the reaction with the alkyl magnesium halide and the copper (1)
-halide in the presence of tetrahydrofuran and in the absence of alkyl halide unit.
According to this method, it is possible in any Z116-20-keto-pregnenen and -allopregnenen and in their above-mentioned derivatives in the 16a-position an alkyl, in particular a lower alkyl, z. B. to introduce the methyl radical in high yield. Specific starting materials are z. B.
j16-3fl- and -3a-hydroxy-20-keto-pregnen and -allopregnen, j 16-3f'-hydroxy-11,20-diketo-allopregnen, J 15-3ss, 11 a-dihydroxy-20-keto-allopregnen , J 16-3a-Hydroxy-11,20-diketo-pregnen, J 16_3fl-Hydroxy-12,20-diketo-allopregnen, J 16-3 a-Hydroxy-12,20-diketo-pregnen, 45,
16-3f hydroxy-20-keto-pregnadiene, A5,16_3ss_Hydroxy-11,20-diketo-pregnadien, A5,16-3ss, 11 a-dihydroxy-20-keto-pregnadiene, 45,16-3ss-hydroxy-12, 20-diketo-pregnadiene, 46 (11), 16-3ss-hydroxy-20-keto-allopregnadiene, A9 (11), 16-3a-hydroxy-20-keto-pregnadiene, its 3-esters and 3-ethers, and corresponding Ver compounds which have a free, esterified or etherified hydroxy group in the 21-position.
Of the compounds substituted in the 21-position, particular mention should be made of 45,16-3ss, 21-dihydroxy-20-keto-pregnadiene and 45 # 16-3-ethylene-dioxy-21-hydroxy-11,20-diketo -pregnadia.
The alkylinagnesium compounds used are primarily lower alkylmagnesium halides, e.g. B. methyl magnesium iodide. When carrying out the process according to the reaction, care must be taken that no more alkyl halide is present in the alkylmagnesium halide solution used, which can easily be achieved by distillation, possibly using an additional solvent, e.g. B. benzene, or by using an excess of magnesium can be achieved.
The compounds customary for this reaction, in particular copper (1) chloride, can be used as copper (1) halides.
The reaction is usually carried out at room temperature. The reaction mixture can be worked up in a manner known per se, for. B. by decomposition with dilute acids such as hydrochloric acid and sulfuric acid, or with ammonium chloride solution. The 16a-alkyl-20-keto-steroids obtained with free hydroxyl groups in the 3- or 3- and 21-positions can, if desired, be converted into their esters and / or ethers in a known manner.
The 16a-alkyl-20-keto-steroids obtained by the present process are used as intermediate products, primarily for the production of corticosteroids with a 16ra-alkyl radical, such as. B. 16a-methyl-9a-fluoro-prednisolone and -prednisone.
The temperatures are given in degrees Celsius in the following examples.
<I> Example 1 </I> A Grignard solution of 2 g magnesium and 7 em3 methyl iodide in 100 cm3 abs. 150 cm3 of pure tetrahydrofuran are added to ether. The ether and excess methyl iodide are completely distilled off under a calcium chloride seal. The methyhnagnesium iodide suspension obtained in tetrahydrofuran is cooled to 20 and the reaction vessel is filled with oxygen-free, dry nitrogen.
Now 400 mg of copper (1) chloride are added. A solution of 5 g d 16-3ss-acetoxy-1l, 20-diketo-allopregnene in 50 cms of pure tetrahydrofuran is then added dropwise at the same temperature with stirring and under nitrogen. The solution is stirred for a further 30 minutes, then cooled from the outside with ice and carefully decomposed with an aqueous ammonium chloride solution. The mixture obtained is diluted with ether,
the ethereal solution washes with an ammonium chloride solution, water, a sodium thiosulphate solution and water, dried and evaporated. The amorphous residue is the crude 3f'-hydroxy-11,20-diketo-16a-methylallopregnane. It is dissolved in 10 cms of dry pyridine and 20 cm? Of acetic anhydride and left to stand at 20 for 15 hours. The solution is then mixed with water, the solution is concentrated in vacuo and the residue is taken up in ether.
The ethereal solution is washed with dilute hydrochloric acid and water, dried and evaporated. The residue is recrystallized from ether alone, ether-pentane or ether-isopropyl ether mixture and gives 4.58 g of 3fl'-acetoxy-16a-methyl-11,20-diketo- allopregnans. It melts at 142-143, then crystallizes again into broad prisms that finally melt at 153; [a] 27 = + 80 (in chloroform);
IR spectrum in methylene chloride: bands u. a. at 5.79 1t, 5.86, u, <I> 7.20 </I>, u (shoulder), <I> 7.26 </I>, u (shoulder), 7.35, u, 8.13 / t and 9.74 p.
<I> Example 2 </I> A Grignard solution of 7 g of magnesium and 30 cm3 of methyl iodide in 200 cm3 of abs. 300 cm3 of pure tetrahydrofuran are added to ether. The ether and excess methyl iodide are completely distilled off with a calcium chloride seal.
The methyhnagnesium iodide suspension obtained in tetrahydrofuran is cooled to 20 and the reaction vessel is filled with oxygen-free, dry nitrogen. 1 g of copper (1) chloride is now added.
A solution of 10 g of 416-3ss, IIIa-diacetoxy-20-keto-allopregnene in 150 cm3 of tetrahydrofuran is then added dropwise at the same temperature with stirring and under nitrogen. The solution is then stirred for a further 30 minutes, then cooled from the outside with ice and carefully decomposed with an aqueous ammonium chloride solution. The mixture obtained is diluted with ether, the ethereal solution is washed with an ammonium chloride solution, water, a sodium thiosulphate solution and water,
dries and steams them. The amorphous residue is the crude 3fl, 11a - dihydroxy - 16a - methyl-20-ketoallopregnan. It is dissolved in 40 cm3 of pyridine and 20 cm3 of acetic anhydride and left to stand at 20 for 15 hours. The solution is then mixed with water, concentrated in vacuo and the residue is taken up in ether. The ethereal solution is washed with dilute hydrochloric acid and water, dried and evaporated.
The crystalline residue (10.3 g) is recrystallized from methanol, 8.6 g of the 3 / 3.11 a-diacetoxy-16a-methyl-20-keto-allopregnans of F. 172-174 being obtained. IR spectrum in methylene chloride: bands u. a. at 5.79, u, <I> 5.86 </I>, et, 7.20 @t (shoulder), <I> 7.26 </I>, c1 (shoulder), 7.34, u ., 8.13, <t, 8.64 <I>, et </I> and 9.76 P .. A further 0.74 g of the same compound can be obtained from the mother liquors.
<I> Example 3 </I> A Grignard solution of 6.9 g magnesium and 25 em3 methyl iodide in 200 cm abs. Ether is mixed with 200 cm- 'pure tetrahydrofuran. The ether and excess methyl iodide are completely distilled off under a calcium chloride seal. The methyl magnesium iodide suspension obtained in tetrahydrofuran is cooled to 20 and the reaction vessel is filled with oxygen-free, dry nitrogen.
600 mg of copper (1) chloride are now added. A solution of 10 g of d5 # 16-3fl-acetoxy-20-keto-pregnadiene in 100 cm3 of pure tetrahydrofuran is then added dropwise at the same temperature with stirring and under nitrogen. The solution is stirred for a further 30 minutes, then cooled from the outside with ice and carefully decomposed with an aqueous ammonium chloride solution. The mixture obtained is diluted with ether, the ethereal solution is washed with an ammonium chloride solution, water,
a sodium thiosulfate solution and water, dry and evaporate them. The residue is recrystallized from acetone, 8.48 g of the J5-3fl-hydroxy-16a-methyl-20-keto-pregnens being obtained; it melts from 182, partially crystallizes again and definitely melts at 188.
2 g of this compound are dissolved in 10 cm3 of pyridine and 20 cm3 of acetic anhydride in the heat. The solution is left to stand at 20 for 15 hours, the solution is decomposed with water and evaporated in vacuo. The residue is dissolved in ether, the ethereal solution is washed with dilute hydrochloric acid and water, dried and evaporated.
The residue is recrystallized from an acetone / isopropyl ether mixture, 1.9 g of J5-3fl-acetoxy-16-methyl-20-keto-pregnen having a melting point of 182-184 being obtained; [a120 = 0 (chloroform); IR spectrum in methylene chloride: bands u. a. at 5.76, u, 5.86, u, <I> 7.20 </I>, u (shoulder), 7.27, u, 7.34, u and 9.70, 1c.
<I> Example 4 </I> Starting from 5 g of dis - 3a - acetoxy -11,20- diketo-pregnane and using the method described in Example 1, 4.6 g of 3a - acetoxy -16a - methyl- 11,20-diketo-pregnane, which after crystallization from an ether-pentane mixture melts at 157-158; [a] D = + 118 (chloroform).
<I> Example 5 </I> Using the method described in Example 3, starting from 10 g of 416-3ss-acetoxy-20-keto-pregnene, 8.3 g of 3ss-hydroxy-16a-methyl-20 are obtained - keto - pregnan from F. 203-205. Acetylation gives 3ss-acetoxy-16a-methyl-20-keto-pregnane with a melting point of 139 to 142, [a] D = + 93 "(chloroform).
In an analogous manner, starting from 45.1s - 3ss21 - diacetoxy - 20 - keto -pregnadiene, the A5-3ss, 21 -dihydroxy -16a - methyl-20-keto-pregnen, which in the acetylation produces the 45-3ss, 21-Diacetoxy-16a-methyl-20-keto-pregnen of F. 160-170 (from hexane) provides.
<I> Example 6 </I> 30 cms each of dry, peroxide-free tetrahydrofuran and dry toluene are mixed with 10 cm3 of a 2.14 molar methylmagnesium iodide solution (with a large excess of magnesium prepared from 4 g of magnesium shavings, 3.2 cm3 of methyl iodide and 53 cm3 ether and distilling off the greater part of the ether)
combined in a nitrogen atmosphere and heated until the temperature in the mixture is 78, with 17 cm3 of solvent distilling off. After cooling to 25, 100 mg of dry copper (1) chloride is added and the mixture is stirred with the Grignard mixture for 5 minutes.
By distilling off the solvent as well as by taking three samples of the reaction mixture of 4 cm3 each, its volume is reduced to 41 cm3. The sample taken at the end of the pretreatment develops 20.7 cm3 of methane with 256,000 ammonium chloride solution, which corresponds to a Grignard complex content of 9.47 millimoles in the reaction mixture.
A solution of 1.6 g of 16-dehydro-progesterone in 30 cm3 of toluene is now allowed to flow in all at once at -10 to 0 and, after removing the external cooling, react for 30 minutes at -10 to +12. Working up is carried out by mixing with excess ammonium chloride solution with the addition of a little sodium thiosulfate, taking up in benzene, drying and distilling off the solvent in vacuo. When the raw product is mixed with a little ether, 1.2 g of crystalline 16a-methyl-progesterone is obtained.
Chromatographic purification of the mother liquor yields a further 0.25 g. The product recrystallized from n-hexane melts at 137-138; E 241 M = 14,400. IR spectrum: 5.87, u (20-ketone) and 5.98 y <I> + </I> 6.17, u (44-3-ketone).
