Process for the preparation of N-aminoalkyl derivatives of azepines The present invention relates to a process for the preparation of new basic alkyl-containing azepines with valuable pharmacological properties.
5-Dibenzo [b, f] azepine, which is referred to below as iminostilbene, and derivatives thereof have not yet become known. It has now been found that N-substituted iminostilbenes of the formula I

wherein X is hydrogen, a halogen atom or the methyl group, Y is an alkylene radical with 2-6 carbon atoms and 2-4 bridge members between N and Am and Am is a low molecular weight dialkylamino radical,
in which the two alkyl radicals can also be connected to one another, mean valuable pharmacological properties, in particular antiallergic and sedative efficacy, and are used, among other things, for the treatment of certain forms of mental illness, in particular mood depression.
Quaternary ammonium salts derived from the tertiary bases defined above act as ganglioplegics.
The process according to the invention for the preparation of compounds of the formula I is characterized in that a reactive ester of a compound of the formula II
EMI0001.0026
in particular a halide, with a secondary amine of the formula III Am-H nI. The reaction can, for example, at a moderately high temperature of, for. B. 80-120 in an inert solvent, such as.
B. a low molecular weight alkanol or alkanone, advantageously an excess of the amine to be reacted is used as an acid-binding agent. Depending on the boiling point of the amine used and the solvent, as well as the required reaction temperature, the reaction may have to be carried out in a closed vessel.
Reactive esters of compounds of the formula II are obtained, for example, by reacting alkali metal compounds of iminostilbenes of the formula IV

with alkylene oxides and reaction of the 5-hydroxyalkyl-iminostilbenes obtained with inorganic acid halides, methanesulfonic acid chloride or arylsulfonic acid chlorides, with 5-haloalkyl-iminostilbenes or
5-Arylsulfonyloxyalkyl-iminostilbene are obtained. These can be reacted, for example, with dimethylamine, methylethylamine, diethylamine, di-n-butylamine, pyrrolidine or piperidine. By addition of halides or Sul faten aliphatic or araliphatic alcohols, z.
B. of methyl iodide, dimethyl sulfate, ethyl bromide, ethyl iodide or benzyl chloride, monoquaternary ammonium compounds can be obtained from the tertiary amines of the formula I in the usual manner, the group Am reacting.
With inorganic or organic acids such as hydrochloric acid, hydrobromic acid, sulfuric acid, phosphoric acid, methanesulfonic acid, ethanedisulfonic acid, acetic acid, citric acid, malic acid, succinic acid, tartaric acid, benzoic acid and phthalic acid, salts can be formed from the tertiary bases, some of which are water-soluble . Fumaric and maleic acids may also be mentioned.
In the following example, the parts mean parts by weight; these relate to parts by volume as to cm3. The temperatures are given in degrees Celsius.
Example 19.3 parts of iminostilbene are in 200 parts by volume of abs. Dissolved benzene and at 45 with stirring in a nitrogen atmosphere a suspension of 4 parts of sodium amide in 30 parts by volume of abs. Benzene was added dropwise. Then the reaction mixture is heated to 75 for 2 hours, then cooled to room temperature,
7 parts by volume of propylene oxide in 7 parts by volume of benzene were added and the whole was stirred for a further 15 hours under a nitrogen atmosphere. The reaction mixture is then mixed with water, the benzene layer is separated off, washed several times with water, dried and evaporated in vacuo.
The residue represents the crude 5- (ss-hydroxypropyl) -iminostilbene, which is mixed with 11.5 parts of methanesulfonic acid chloride at 0 in 40 parts by volume of pyridine.
The reaction mixture is left to stand at room temperature for one hour, then 23 parts of pyridine hydrochloride are added in order to completely convert the reaction product, which consists partly of the methanesulfonic acid ester and partly of the chloride of the starting material, into the latter, and the whole is initially allowed to stand for 15 hours at room temperature and Finally heat it up to 80 for 11, /: 2 hours.
After cooling, the 5- (ss-chloropropyl) -iminostilbene formed is precipitated by diluting it with water and heated to 90 for 3 hours with 100 parts by volume of 33-liter ethanolic dimethylamine solution in a closed vessel. After cooling, water is added to the reaction mixture, it is rendered acidic with dilute hydrochloric acid, it is shaken with ether and the 5- (s-dimethylamino-propyl) -iminostilbene is precipitated with sodium hydroxide solution.
It passes under 0.02 mm pressure at 151-153 and can be converted into hydrochloride using alcoholic hydrochloric acid.
