The present invention relates to a process for the preparation of new glycols of the formula
EMI1.1
where Rt is an alkyl, alkenyl or cycloalkyl radical or an aryl radical optionally substituted by halogen atoms or low molecular weight alkyl or alkoxy groups, R2 and R3 are low molecular weight alkyl radicals, where R2 can also be connected to an alkyl radical Rj, and R4 is an unsaturated aliphatic hydrocarbon radical with 2- 3 mean carbon atoms. As has now been found, these compounds have sedative, hypnotic to narcotic activity. They also have an anticonvulsant effect and are particularly useful therapeutically as tranquilizers.
They also potentiate the effect of other narcotics and can also be used as intermediate products for the manufacture of other pharmacologically valuable substances.

with an organometallic compound of magnesium or zinc and a halide of the formula Hal-CH2-R4 III in which Hal is chlorine, bromine or iodine, and if desired, if the radical R4 of the reaction product contains a triple bond, this is hydrogenated to a double bond. Generally good yields can be expected if, according to the invention, one mole of an α-hydroxy-ketone of the formula II is mixed with approximately two moles of an organometallic compound of magnesium and a halide of the formula III in an organic solvent which has one or more oxygen atoms bonded between hydrocarbon radicals contains, e.g.
B. an ether or ethereal solvent, such as diethyl ether or tetrahydrofuran, or an acetal, such as dimethyl formal.
The a-hydroxy-ketones of the formula II, where R2 and R3 are methyl groups, are obtained, for example, by condensation of methyl ketones of the formula R-CO-CH3 with alkali metal acetylides and subsequent addition of water, e.g. B. in the presence of mercury oxide and sulfuric acid, to the ethynyl-methyl-carbinole thus obtained. Furthermore, that of Billimoria et al. (J.
Chem. Soc. 1951, 3067) mentioned reaction of α-hydroxycarboxylic acids with methyllithium; α-Hydroxy-ketones of the general formula II can also be obtained from ketones of the general formula Rt-CO-R2. The O-acyl derivatives, O-tetrahydropyranyl derivatives or O- (a-alkoxy-ethyl) - derivatives of nitriles of branched a-hydroxy-alkanecarboxylic acids, of a-hydroxy-a-cycloalkyl-aceto-nitriles or a-hydroxy-a Aryl-acetonitriles can be reacted with alkylmagnesium halides of the formula R3-Mg-Hal to give imines, from which the α-hydroxy-ketones of the general formula II can be obtained by hydrolysis.
Examples of suitable a-hydroxy-ketones are 3-methyl-3-hydroxy-butanone- (2),

α-Ketols of the general formula II, in which R1 and R2 together represent an alkylene radical and R3 methyl, are obtained, for example, by condensation of cycloaliphatic ketones with alkali metal acetylidene and addition of water to the ethynylcycloalkanols thus obtained, or by the above-mentioned reaction of α-hydroxycarboxylic acids with Methyllithium; or, if R3 represents an alkyl radical with 2-4 carbon atoms, by reacting cyanohydrins of suitable cycloaliphatic ketones (after closing the hydroxyl group as described above) with low molecular weight alkylmagnesium halides to form imines, from which the α-hydroxy-ketones of the general formula II can be obtained by hydrolysis are.
Examples of such compounds are l-acetyl, l-propionyl, l-butyryl and l-isobutyryl-cyclo-pentanol,

Example 1
To an allyl magnesium bromide solution prepared according to Organic Synthesis 36, 61 (1956) (from 24.3 parts of Mg, 1 crystal of iodine and 60.5 parts of allyl bromide, using a total of 350 parts by volume of absolute ether), 28.8 Parts of 3,5-dimethyl -3-hydroxyhexanone - (2), boiling point 6: 650, dissolved in 50 parts by volume of abs. Ether, added dropwise at a reaction temperature below 201.
After stirring for 4 hours at room temperature, 100 parts of water are first added dropwise to the reaction mixture, while cooling with ice, whereupon it is made acidic to Congo by carefully adding 50% sulfuric acid. After stirring for a further hour at room temperature, a further 100 parts of water are added. The precipitate is filtered off with suction and washed out with water, the organic layer of the filtrate is separated off and the aqueous layer is extracted 3 times with 100 parts by volume of ether. The combined organic solution is washed neutral, dried over sodium sulfate, and after removing the solvent, the residue is purified by repeated distillation under reduced pressure.
