Verfahren zur Herstellung von analgetisch wirksamen 1-Phenyi-2 , 3-dimethyl-4morpholinomethyl-pyrazolonen
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung analgetisch wirksamer l-Phenyl-2, 3- dimethyl-4-morpholinomethyl-pyrazolone der Formel

worin R1 ein Wasserstoffatom oder den Phenylrest und R2 ein Wasserstoffatom oder einen niedrigen Alkylrest mit 1 bis 5 C-Atomen in gerader oder verzweigter Kette bedeutet.
Das erfindungsgemässe Verfahren zur Herstellung dieser Verbindungen ist dadurch gekennzeichnet, dass man entsprechend substituiertes Morpholin, Formaldehyd und l-Phenyl-2,3-dimethyl-pyrazolon in wässriger, saurer Lösung miteinander umsetzt. Der Einsatz des Morpholins erfolgt zweckmässig in alkoholischer Lösung.
Die Hydrochloride der Verfahrensprodukte, wie 1-Phenyl-2,3 -dimethyl-4- (2'-phenyl-3'-methyl-mor pholinomethyl)-pyrazolon-(5) -hydrochlorid, fallen hierbei bereits aus der Reaktionslösung aus und können gegebenenfalls aus Alkohol-Aceton umkristallisiert werden. Durch Versetzen der Hydrochloride mit Alkali erhält man nach an sich bekannter Aufarbeitung die entsprechenden Basen, welche in fester Form anfallen.
Die Herstellung von basisch substituierten Verbindungen des l-Phenyl-2,3-dimethyl-pyrazolons-(5) mit Formaldehyd und einer heterocyclischen Base nach dem Prinzip der Mannich-Reaktion ist an sich bekannt. Das Piperidin wurde hierbei zuerst mit dem Formaldehyd mit Hydroxymethylpiperidin umgesetzt, das dann in Gegenwart von wasserfreiem Kaliumcarbonat mit dem 1 l-Phenyl-2, 3-dimethyl-pyrazolon-(5) umgesetzt wurde. Die Ausbeuten bei diesem zweistufigen Verfahren waren ebenso wie bei der einstufigen Durchführung dieser Mannich-Reaktion nicht sehr befriedigend. Über die pharmakologischen Eigenschaften des hierbei erhaltenen Produkts wurde bisher nichts bekannt. Für die Verwendung in der ärztlichen Praxis scheint es nicht geeignet zu sein.
Bei dem erfindungsgemässen Verfahren wird dagegen nicht Piperidin, sondern unsubstituiertes oder in bestimmter Weise, nämlich in 2-Stellung mit einem Phenylrest und in 3-Stellung mit einem niedrigen Alkylrest substituiertes Morpholin in einer Verfahrensstufe in Gegenwart von Formaldehyd mit der Pyrazolonverbindung kondensiert, wobei Ausbeuten von mehr als 80 O/o der Theorie erhalten werden.
Die neuen Verbindungen sind analgetisch besonders wirksam, was sich aus den pharmakologischen Vergleichsversuchen mit bekannten Verbindungen ergibt, deren Ergebnisse in der folgenden Tabelle enthalten sind:

In der vorstehenden Tabelle sind die bei der pharmakologischen Analgesieprüfung gefundenen mittleren therapeutischen Dosen (ADsoII, nach der Methode von Wolff-Hardy mittels dosierten Wärmeeinsatzes durch Punktlichtlampe und Brennlupe bestimmt) von l-Phenyl-2, 3-dimethyl-4-(2'-phenyl-3'- methylmorpholinomethyl)-pyrazolon-(5)
- hydrochlorid im Vergleich mit l-Phenyl-2, 3-dimethyl-4-dimethyl- amino-pyrazolon-(5), Salicylamid und Phenacetin aufgeführt. Hiernach wirkt das neue Morpholinderivat zweimal stärker als das bekannte Pyrazolonderivat und zehnmal stärker als Salicylamid oder Phenacetin.
Berücksichtigt man ferner, dass die analgetische Wirkung von l-Phenyl-2, 3-dimethyl-pyrazolon ein Drittel der Wirksamkeit vom l-Phenyl-2, 3-dimethyl-4-di- methyl-aminopyrazolon beträgt, so wurde gegenüber dem Phenyldimethylpyrazolon durch Einführung des 2-Phenyl-3-methylmorpholino-methylrestes eine Steigerung der Wirksamkeit auf das Sechsfache erzielt.
Die nach dem erfindungsgemässen Verfahren hergestellten l-Phenyl-2, 3 -dimethyl-4-morpholinomethyl- pyrazolone stellen daher insbesondere in Form ihrer Salze in völlig überraschender Weise neue und wertvolle Arzneimittel dar. Die Lösungen der Salze, insbesondere die von organischen Säuren, eignen sich wegen der guten Verträglichkeit der neuen analgetischen Verbindung auch für Injektionen.
Die nach dem erfindungsgemässen Verfahren erhaltenen analgetisch wirksamen Verbindungen können insbesondere in Form der leicht löslichen Salze der Gentisinsäure oder Ascorbinsäure für hochprozentige, injizierbare Lösungen verwendet werden. Die Herstellung dieser Lösungen kann dadurch erfolgen, dass man die in Wasser aufgeschäumte, praktisch unlösliche Base durch Zugabe der organischen Säure in Lösung bringt, wobei man den pH-Wert auf einen neutralen bzw. schwach sauren pH-Bereich von etwa 6-7 einstellt. Das gentisinsaure Salz der neuen Pyrazolonverbindung kann z. B. auch durch Umsetzung äquimolarer Mengen an Gentisinsäure und der Base unter Erwärmen in einem Methanol-Aceton Gemisch hergestellt werden, wobei dann das gewünschte Salz beim Erkalten auskristallisiert, das man isoliert und dann in Aqua destillata entsprechend der gewünschten Konzentration wieder auflöst.
Beispiel I
35,4 g 2-Phenyl-3-methyl-morpholin werden in 50 ml Methanol gelöst und unter Kühlung 20 ml 400/Die Formalinlösung zugegeben. Dann werden 37,6 g 1 -Phenyl-2, 3-dimethylpyrazolon-(5), gelöst in einem Gemisch von 35 ml Wasser und 20 ml konz.
Salzsäure, auf einmal zugefügt und, nachdem der pH-Wert der Reaktionslösung auf pH 2 bis pH 3 eingestellt ist, noch zwei Stunden auf dem Wasserbade bei 25-30OC gerührt. Das ausfallende Hydrochlorid wird aus Methanol-Aceton umkristallisiert.
Schmelzpunkt des l-Phenyl-2,3-dimethy1-4-(2'- phenyl - 3'-methyl - morpholinomethyl) - pyrazolon -(5)- hydrochlorids = 171 bis 172" C unter Zers. Ausbeute: 81,60/0. Die entsprechende Base, welche aus dem Hydrochlorid durch Alkalisieren, Extrahieren und Eindunsten des Extraktionsmittels gewonnen wird, schmilzt bei 149-150 C.
Die freie Base wird mit der äquimolaren Menge Gentisinsäure unter Erwärmen in einem Methanol Aceton-Gemisch umgesetzt und das beim Abkühlen auskristallisierte gentisinsaure Salz, das bei 169 bis 170"C unter Zersetzung schmilzt, isoliert. Das erhaltene Produkt kann in sterilisierter wässriger Lösung zur Injektion verwendet werden.
Beispiel 2
Gemäss Beispiel 1 werden 38,2 g 2-Phenyl3-äthyl-morpholin mit 37,6 g 1-Phenyl-2,3-dimethyl- pyrazolon-(5) umgesetzt. Ausbeute an 1-Phenyl-2,3- dimethyl-4-(2'-pheny1 -3'-äthyl- morpholino- methyl)- pyrazolon-(5)-hydrochlorid : 82,2 2 /o ; Fp. = 174 bis 175"C unter Zers. (aus Aceton-Methanol umkristallisiert).
Beispiel 3
17,4 g Morpholin werden gemäss Beispiel 1 mit 37,6 g 1-Phenyl-2,3-dimethylpyrazolon-(5) und 20 ml 400/obige Formalinlösung umgesetzt. Ausbeute an l-Phenyl-2, 3 -dimethyl-4-morpholinomethylpyrazolon (5)-hydrochlorid: 85,8 O/o. Fp. des Hydrochlorids: 170 bis 171"C unter Zersetzung. Fp. der entsprechend Beispiel 1 anfallenden Base: 130-131"C.

The process according to the invention for the preparation of these compounds is characterized in that appropriately substituted morpholine, formaldehyde and l-phenyl-2,3-dimethylpyrazolone are reacted with one another in an aqueous, acidic solution. The morpholine is expediently used in an alcoholic solution.
The hydrochlorides of the process products, such as 1-phenyl-2,3-dimethyl-4- (2'-phenyl-3'-methyl-mor pholinomethyl) -pyrazolone- (5) -hydrochloride, fall out of the reaction solution and can optionally recrystallized from alcohol-acetone. By adding alkali to the hydrochloride, the corresponding bases, which are obtained in solid form, are obtained after known work-up.
The preparation of basic substituted compounds of l-phenyl-2,3-dimethyl-pyrazolone- (5) with formaldehyde and a heterocyclic base according to the principle of the Mannich reaction is known per se. The piperidine was first reacted with the formaldehyde with hydroxymethylpiperidine, which was then reacted in the presence of anhydrous potassium carbonate with the 11-phenyl-2,3-dimethyl-pyrazolone- (5). The yields in this two-stage process, as in the one-stage implementation of this Mannich reaction, were not very satisfactory. So far nothing has been known about the pharmacological properties of the product obtained in this way. It does not appear to be suitable for use in medical practice.
In the process according to the invention, on the other hand, it is not piperidine but morpholine which is unsubstituted or substituted in a certain way, namely in the 2-position with a phenyl radical and in the 3-position with a lower alkyl radical, condensed in one process stage in the presence of formaldehyde with the pyrazolone compound, yields of more than 80% of theory can be obtained.
The new compounds are particularly effective analgesic, which is evident from the pharmacological comparison tests with known compounds, the results of which are contained in the following table:

The l-phenyl-2,3-dimethyl-4-morpholinomethyl-pyrazolones prepared by the process according to the invention are therefore completely surprisingly new and valuable medicaments, especially in the form of their salts. The solutions of the salts, especially those of organic acids, are suitable because the new analgesic compound is well tolerated, it is also suitable for injections.
The analgesically active compounds obtained by the process according to the invention can be used, in particular, in the form of the easily soluble salts of gentisic acid or ascorbic acid for high-percentage, injectable solutions. These solutions can be prepared by bringing the practically insoluble base, which is foamed in water, into solution by adding the organic acid, the pH being adjusted to a neutral or weakly acidic pH range of about 6-7. The gentisic acid salt of the new pyrazolone compound can, for. B. can also be prepared by reacting equimolar amounts of gentisic acid and the base with heating in a methanol-acetone mixture, the desired salt then crystallizes out on cooling, which is isolated and then redissolved in aqua distillata according to the desired concentration.
Example I.
35.4 g of 2-phenyl-3-methyl-morpholine are dissolved in 50 ml of methanol and 20 ml of 400 / formalin solution are added with cooling. Then 37.6 g of 1-phenyl-2, 3-dimethylpyrazolone (5), dissolved in a mixture of 35 ml of water and 20 ml of conc.
Hydrochloric acid, added all at once and, after the pH of the reaction solution has been adjusted to pH 2 to pH 3, stirred for another two hours on the water bath at 25-30 ° C. The precipitating hydrochloride is recrystallized from methanol-acetone.
