Verfahren zur Herstellung der Xanthogensäuren von 21-Oxy-corticosteroiden Es ist bekannt, dass Cortison und verwandte Ver bindungen, die 21-Oxy-pregnene und- pregnadiene, eine günstige Wirkung auf Patienten ausüben, die un ter Schockeinwirkung stehen. Bei der Behandlung dieser Zustände ist es jedoch wesentlich, dass die Wirkung rasch eintritt.
Dies wird jedoch bei diesen Verbindungen dadurch verunmöglicht, dass sie in Wasser nur wenig löslich sind, so dass man sie nicht direkt in Form ihrer wässrigen Lösungen in den Blut- @trom einführen kann. Es war deshalb üblich, das Corticoidin-Äthanol zu lösen und diese Lösungen in ,geeigneter Verdünnung intravenös zu verabreichen. Diese Methode weist jedoch beträchtliche Nachteile auf, insbesondere weil man ziemlich grosse Flüssig keitsmengen einführen muss.
Ausserdem können solche Infusionen nicht gut ausserhalb von Spitälern vorgenommen werden, obschon es oft erwünscht wäre, die Behandlung anderswo, z. B. an einer Unfall stelle, durchzuführen.
Aus diesen und anderen Gründen wäre es er wünscht, Lösungen zur Verfügung zu haben, in denen die erforderliche Dosis in wenigen Millilitern einer Injektionsflüssigkeit enthalten ist und welche die ak tive Substanz in einer Form enthalten, die den An forderungen des Organismus mit genügender Rasch heit zur Verfügung steht.
Verbindungen von Corticoiden, die in wässrigen Flüssigkeiten besser löslich sind als das Corticoid als solches, jedoch nicht notwendigerweise sehr leicht löslich, können auch für andere Zwecke, z. B. zur Herstellung von Salben mit corticoider Wirkung in der Dermatologie, z.
B. gegen Ekzeme, verwendbar sein. Es wurde nun ein Verfahren zur Herstellung .der Xanthogensäuren von 21-Oxy-corticosteroiden und deren Alkalimetallsalzen gefunden, das dadurch gekennzeichnet ist, dass man in der 21-Stellung von 21-Oxy-corticosteroiden unter Mitwirkung der 21- Hydroxylgruppe die -O-CS-SH-Gruppe oder die -O-CS-SMe-Gruppe, wobei Me ein Alkalimetall bedeutet, ausbildet,
indem man eine 21-Oxy-corti- costeroid in einem Lösungsmittel mit einem Alkali metallhydroxyd und Schwefelkohlenstoff umsetzt und aus dem Reaktionsgemisch das Xanthogensäure-De- rivat oder das Alkalimetallsalz davon isoliert.
Die so erfindungsgemäss hergestellten Verbindun gen können als Ester der Dithiokohlensäure mit ihrer Hydroxylgruppe angesehen werden, wobei die saure Sulfhydrylgruppe der Dithiokohlensäure mit Basen in wässrigen Lösungen Xanthogenate bilden kann, deren Löslichkeit durch die Art der Base bestimmt wird.
Diese Xanthogenate hydrolysieren in Gegen wart von Wasser bei pH-Werten in der Nähe des Neutralpunktes sehr rasch, wodurch das Corticoid freigesetzt wird, so dass bei der intravenösen Verab reichung die Corticoidwirkung im menschlichen Or ganismus sehr rasch eintritt.
Bei der biologischen Prüfung des Glucocorticoid- effektes an Ratten zeigten die neuen Verbindungen die gleiche Wirkung wie das entsprechende Steroid- hormon.
Infolge der raschen Hydrolyse der Xanthogenate in ihren Lösungen müssen die neuen Substanzen für den therapeutischen Gebrauch entweder in trockener, wasserlöslicher Form oder in Form der freien Xan- thogensäure, die sich vor Gebrauch leicht mit einer wässrigen Base mischen lässt, oder als Salben, die ein mehr oder weniger lösliches Xanthogenat enthalten und äusserlich verwendet werden sollen, vertrieben werden.
Mit dem erfindungsgemässen Verfahren lassen sich z. B. Xanthogensäuren von d-1-3,20-Diketo-17a, 21-dioxypregnenen und -pregnadienen aus den ent sprechenden Steroiden herstellen, die nicht nur 44-Pregnen-3,11,20-triketo-17a,21-diol-21- xanthogensäure und A4-Pregnen-3,20-diketo-11,17a,21-triol-21- xanthogensäure, (in denen die 11-Hydroxylgruppe in a- oder ss-Stellung sein kann) umfassen, sondern auch 41.4-Pregnadien-3,
11-20-triketo-17a,21-diol-21- xanthogensäure und d1.4-Pregnadien-3,20-diketo-11,17a,21-triol-21- xanthogensäure, in denen die 11-Hydroxylgruppe in a- oder f-Stellung sein kann sowie verwandte Verbindungen. Als Bei spiele für verwandte Verbindungen, seien diejenigen genannt, die den oben erwähnten entsprechen, aber in 6a-Stellung eine Methy bgruppe als Substituenten tragen.
Die bei der Durchführung des erfindungsgemässen Verfahrens verwendeten Lösungsmittel sollen mit Alkali und Schwefelkohlenstoff nicht unter Xantho- genatbildung reagieren. Lösungsmittel mit alkoholi schen Gruppen kommen deshalb im allgemeinen nicht in Frage. Ferner werden vorteilhaft Lösungsmit tel verwendet, die eine gewisse Mischbarkeit mit Was ser besitzen, die vorzugsweise so gross ist, dass sie mindestens 5-10 o/a Wasser auflösen können.
So wird unter sonst gleichen Reaktionsbedingungen die Ausbeute an Xanthogenat bei Verwendung von Me- thylisobutylketon als Reaktionsmedium nur ein Drit tel derjenigen sein, die man erhält, wenn man bei spielsweise in Dioxan arbeitet, was der schlechteren Wassermischbarkeit des ersteren Lösungsmittels ent spricht.
Ausser den bereits genannten Reaktions medien kann man beispielsweise auch noch die fol genden verwenden: Aceton und andere Ketone, wie Methyläthylketon und Dimethylformamid. Aceton und Dimethylformamid ergaben in allen Versuchen ebenso gute Ausbeuten wie Dioxan.
Die erfindungsgemässe Reaktion wird vorzugs- weise so durchgeführt, dass man die Lösung des Steroids im gewählten organischen Lösungsmittel, das mindestens teilweise mit Wasser mischbar ist, mit einem Alkalimetallhydroxyd behandelt und das dabei entstandene Alkoholat mit Schwefelkohlenstoff um setzt, wonach man das entstandene Xanthogenat oder (nach Zusatz einer Säure) die freie Xanthogensäure aus der Lösung isoliert. Die Reaktion wird vorzugs weise unter Kühlung,
nötigenfalls bis nahe an den Gefrierpunkt des Lösungsmittels durchgeführt. Man kann zuerst das Alkalimetallhydroxyd in wässriger Lösung zusetzen und nach kurzem Vermischen der beiden Phasen den Schwefelkohlenstoff als solchen oder im gleichen oder einem ähnlichen Lösungsmittel wie das Steroid gelöst zugeben. Nachdem der Schwe felkohlenstoff einige Zeit eingewirkt hat, so wird das Xanthogenat gebildet sein und die wässrige Phase kann entfernt werden.
Die organische Phase kann nun auf verschiedene Weise behandelt werden, um entweder das Xanthogenat der fraglichen Base oder die freie Xanthogensäure zu isolieren. Im ersteren Falle kann man die Lösung trocknen und zur Trockne eindampfen, oder man kann ein Lösungsmittel, das die Löslichkeit des Xanthogenats herabsetzt, wie Äther, zusetzen, wodurch es ausgeschieden wird.
Im zweiten Falle kann man eine starke Säure, wie Salz- oder Schwefelsäure, und ein Lösungsmittel zusetzen und nach Entfernung der wässrigen Phase die orga nische Phase wiederum zur Trockne verdampfen (wo bei man verbleibendes Wasser durch Trocknen ent fernen kann, um Hydrolyse zu vermeiden), oder man kann ein Lösungsmittel, wie z. B. Aceton, zusetzen, um die freie Säure abzutrennen. Das Xanthogenat oder die Xanthogensäure kann nach der Isolierung umkristallisiert werden und aus der Säure kann man die Salze erhalten, indem man sie in der entspre chenden Base löst.
Die freien Xanthogensäuren sind in Wasser schlecht löslich, doch sind viele Salze leicht löslich und aus diesem Grunde sowie infolge ihrer Zerfliess- lichkeit ist es oft schwer, sie in reinem kristallinem Zustand zu erhalten.
So sind die Natriumsalze zer- fliesslich, während die Kaliumsalze, obschon sie im mer noch sehr leicht löslich sind, leichter im reinen Zustand hergestellt werden können. Selbst die Cal- zium- und Zinksalze lassen sich leicht in fester Form erhalten, doch lassen sich insbesondere gewisse Amin salze speziell leicht in reiner kristalliner Form erhal ten, trotzdem sie gut löslich sind.
Dies ist insbeson dere der Fall bei den Äthanolamin-, Diäthylamin- und N-Äthylpiperidinsalzen. Selbst quaternäre Am moniumsalze lassen sich herstellen.
Als Beispiele für wenig lösliche Aminsalze seien folgende erwähnt: Diisopropylaminsalze und die Salze des Diäthylaminopropylamins, n-Butylamins und Triäthanolamins.
Zur Illustration der Löslichkeit einiger Salze, ins besondere der Aminsalze, wird die Menge einer wäss- rigen Lösung von verschiedenen Basen angegeben, in denen 50 mg Hydrocortison-21-xanthogensäure unter Salzbildung gelöst werden, angegeben.
0,5 ml 1,1 /o ige wässrige Lösung von Äthylamin 1,0 ml 1,2 o/o.ige wässrige Lösung von Triäthylamin 0,5 ml 2,7 o/oige wässrige Lösung von /;

0 %ige wässrige Lösung von 1-Diäthylamin-2-propanol 0,5 ml 0,6 o/oige wässrige Lösung von Ammoniak 1,0 ml 0,5 o/o,ige wässrige Lösung von Natriumhydroxyd. <I>Beispiel 1</I> _J-I-Pregnen-3,11,20-trion-17a, 21-diol-21- xanthoget2säure (Cortison-21-xantlzogensäure). 3,
00 g Cortison werden unter gelindem Erwär men in 90 ml peroxydfreiem Dioxan gelöst und die Lösung in Eiswasser bis zum Gefrierpunkt des Di- oxans gekühlt. Dann gibt man eine vorgekühlte Lö sung von 3,0g KOH in 30 ml Wasser unter kräfti gem Rühren zu. Man rührt und kühlt weitere 5 Mi nuten, wonach man eine Lösung von 3,0 g CS2 in 5 ml Dioxan auf einmal zusetzt. Man rührt weitere 15 Minuten, verdünnt dann mit 200 ml Benzol und säuert mit 9 ml konzentrierter Salzsäure an.
Die wäss- rige Phase wird abgetrennt und die Benzollösung drei mal mit Wasser gewaschen. Nach dem Trocknen über Na2S04 wird die Lösung bei Zimmertemperatur im Vakuum zur Trockne eingedampft und der Rück stand durch Zusatz von Aceton kristallisiert. Die Kri stalle werden auf dem Filter gesammelt, in dünnere Schicht ausgebreitet und bei Zimmertemperatur an der Luft getrocknet. Ausbeute an trockenem Produkt 2,55 g. Die Substanz schmilzt bei 127-130 C, beim Weitererhitzen wird sie wieder fest und schmilzt nochmals bei 230-232 C.
Kaliumsalz <I>der</I> Cortison-21-xanthogensäure. 3,00 g Cortison werden wie oben beschrieben in eine trockene Lösung der Xanthogensäure in Benzol übergeführt und 30 Minuten unter Vakuum gehalten, um überschüssiges CS2 zu entfernen. Hernach gibt man 42 ml 1,6 0/c ige alkoholische KOH zu und lässt einige Zeit stehen. Der Niederschlag wird abfiltriert, mit Benzol gründlich gewaschen und ohne Erhitzen an der Luft getrocknet.
Man erhält so 2,4 g des ge wünschten Produktes vom Smp. 165-167 C (Zers.) mit einem Gehalt an 13,60% S (berechnet 13,51 /o S). <I>Beispiel 2</I> <I>_l</I> I-Pregnen-3, 20-dion-11 ss,17a,21-triol-21- xanthogensäure (Hydrocortison-21-xanthogensäure). 3,
00 g Hydrocortison werden wie im Beispiel 1 beschrieben behandelt, mit einem einzigen Unter schied, dassdieReaktionsmischung statt mit200m1Ben- zol mit 300 ml Äther verdünnt wird. Beim Aus waschen der Lösung mit Wasser fällt die Hydrocorti- son-21-xanthogensäure aus. Sie wird auf dem Filter gesammelt, mit Äther gewaschen und bei Zimmer temperatur an der Luft getrocknet. Man erhält so 3,00 g des gewünschten Produktes.
Es schmilzt bei 110-113 C, wird beim weiteren Erhitzen wieder fest und schmilzt nochmals bei 210-213 C. Kaliumsalz <I>der</I> Hydrocortison-21-xanthogensäure. 1,50 g Hydrocortison-21-xanthogensäure werden mit 10 ml 2 0/c iger alkoholischer Kalilauge vermischt. Die Substanz geht in Lösung. Nach einigem Stehen beginnt sich das Kaliumsalz auszuscheiden und wird nach weiterem Stehen auf dem Filter gesammelt, mit Äther gewaschen und getrocknet.
Ausbeute: 1,30 g; 8,12 "/a K (berechnet 8,2 % K).
Ätlzanolaminsalz <I>der</I> Hydrocortison-21- xanthogensäure. Zu 4,0 g frisch hergestellter Hydrocortison-21- xanthogensäure gibt man 20 ml einer 3,
1%.igen Lö- sung von Äthanolamin in 99 %igem Äthanol. Die Xanthogensäure geht dabei in Lösung und nach we nigen Minuten entsteht ein Niederschlag.
Nach eini gem Stehen wird der Niederschlag abfiltriert, gründ- lich mit 99 % igem Äthanol, dann mit Äther ge- waschen und im Vakuum über P.05 getrocknet.
Man erhält so 2,7 g des gewünschten Salzes. Sein Ultra violettspektrum besitzt Maxima bei 245 m,, (E = 17 500) und 304 mu (E = 14 400) in frisch her gestellter wässriger Lösung. Diäthylaminsalz <I>der</I> Hydrocortison-21- xantlaogensäure.
Zu 4,0 g frisch hergestellter Hydrocortison-21- xanthogensäure gibt man 27 m1 einer 2,5 %igen Lösung von Diäthylamin in 99 % igem Athanol. Die Xantho@gensäure geht in Lösung,
und es entsteht ein Niederschlag. Nach einigem Stehen wird filtriert, gründlich mit 99 % igem Äthanol, dann mit Äther gewaschen und im Vakuum über<B>P205</B> getrocknet. Man erhält 3,5 g des gewünschten Salzes. Sein Ultra violettspektrum besitzt Maxima bei 246 mit (E = 18 200) und 304 my (E = 14 400) in frisch hergestell ter wässriger Lösung.
Tetraäthylammoniumsalz <I>der</I> Hydrocortison-21- xanthogensäure. 5,0 g Tetraäthylammoniumjodid werden in 80 ml Methanol gelöst und zur Lösung 1 ml Wasser und 3,5g Ag20 gegeben. Nach 3 Stunden Rühren wird filtriert und das Filtrat mit Methanol auf 100 ml verdünnt. Zu 4,0 g Hydrocortison-21-xanthogensäure gibt man 48 ml des verdünnten Filtrats. Die Xantho- gensäure geht in Lösung und nach wenigen Minuten wird Äther zugesetzt. Es entsteht ein öliger Nieder schlag.
Die Flüssigkeit wird von diesem abdekantiert und der Niederschlag in Aceton aufgenommen. Man setzt wiederum Äther zu, filtriert vom gebildeten Nie derschlag ab, wäscht gründlich mit Äther und trock net im Vakuum über P205. Man erhält so 2,8 g des gewünschten Salzes. Sein Ultraviolettspektrum zeigt Maxima bei 245 mu (E --_ 17 500) und 303 m,u (E - 14 000) in frisch hergestellter wässriger Lösung.
N-Äthylpiperidinsalz <I>der</I> Hydrocortison-21- xanthogensäure.
Zu 4,0 g frisch hergestellter Hydrocortison-21- xanthogensäure gibt man 4,0g N-Äthylp.iperidin in 20 ml 99 % igem Athanol gelöst. Dabei geht die Xanthogensäure in Lösung, und es entsteht nach we nigen Minuten ein Niederschlag.
Man filtriert, wäscht mit Äther und trocknet im Vakuum über P205. Man erhält so 4,4 g des gewünschten Salzes. Sein Ultra- violettspektrum zeigt Maxima bei 246 mu (E 17 600) und 303 mit (E = 13 800) in frisch her gestellter wässriger Lösung.
Calziumsalz <I>der</I> Hydrocortison-21-xanthogensäure. Zu 85 g Calziumhydroxyd gibt man in einem Mörser 1 g frisch hergestellte Hydrocortison-21- xanthogensäure und 10 ml 99 o/aiges Äthanol. Man verreibt die Mischung während 5 Minuten mit dem Pistill und filtriert.
Die feste Substanz wird mit 99 % igem Alkohol und Äther gewaschen und im Vakuum bei Zimmertemperatur über P205 getrocknet. Man erhält so 1,0 g des gewünschten Salzes. Sein Ultraviolettspektrum besitzt Maxima bei 245 m; c <B>(E=30000)</B> und 304 my <B>(E=22000)</B> in frisch hergestellter wässriger Lösung.
<I>Zinksalz der</I> Hydrocortison-21-xanthogensäure. Zu 452 mg Zinkhydroxyd gibt man im Mörser 40 ml 99o/aiges Äthanol und 4,0 g frisch hergestellte Hydrocortison-21-xanthogensäure, verreibt die Mi schung 5 Minuten mit dem Pistill und filtriert. Der feste Rückstand wird mit 99 o/oigem Äthanol und Äther gewaschen und im Vakuum über<B>P205</B> getrock net.
Man erhält 3,5 g des gewünschten Salzes. Sein Ultraviolettspektrum zeigt Maxima bei 245 m (E = 30 400) und 304 my (E = 28 100) in frisch bereiteter wässriger 0,1n NaOH Lösung.
Wässrige Lösungen der Salze, z. B. der Kalium- salze oder Diäthanolaminsalze werden rasch hydroh- siert. Die Halbwertzeit dieser Salze beträgt beim pH 7,2 und 37 C 15 Minuten. Dabei wird das ent sprechende Steroidhormon, z.
B. Hydrocortison, frei gesetzt. Wenn Lösungen dieser Salze in kleinen Men gen Wasser intravenös eingespritzt werden, wird das Hydrocortison oder allgemein das im Xanthogenat vorhandene Corticoid nach kurzer Zeit im Blut frei- gesetzt. <I>Beispiel 3</I> di.4-Pregnadien-3,11,20-trion-17a,21-diol-21- xanthogensäure (Prednisolon-21-xanthogensäure). 3,
00 g 1-Dehydrocortison werden in 180 ml Dioxan gelöst und die Lösung bis zum Schmelz punkt des Dioxans gekühlt. Dann setzt man unter kräftigem Rühren eine Lösung von 3,0g KOH in 60 ml Wasser zu. Man rührt weitere 5 Minuten, wo nach man eine Lösung von 3,0 g CS2 in 5 ml Dioxan auf einmal zusetzt. Nach weiteren 15 Minuten Rüh ren verdünnt man mit 300 ml Äther und säuert mit 9 ml konzentrierter Salzsäure an.
Die wässrige Phase wird entfernt und die Ätherphase dreimal mit Wasser gewaschen. Nach dem Trocknen über Na2S04 ver dampft man im Vakuum bei Zimmertemperatur zur Trockne und versetzt den Rückstand mit 9 ml Ace ton. Nach einigem Stehen bei 0 C filtriert man von den Xanthogensäurekristallen ab und trocknet sie bei Zimmertemperatur an der Luft. Ausbeute 1,95 g.
Das Produkt schmilzt bei 120-122 C, wird beim Weitererhitzeri wieder fest und schmilzt nochmals bei 222-224 C unter Zersetzung. <I>Beispiel 4</I> dj#4-Pregnadien-3,20-diott-11 /3,17a, 21-triol-21- xanthogensäure. 3,00 g Prednisolon werden unter gelindem Er wärmen in 90 ml peroxydfreiem Dioxan gelöst, und die Lösung in Eiswasser bis zum Gefrierpunkt des Dioxans gekühlt.
Dann setzt man unter kräftigem Rühren eine Lösung von 3,0 g KOH in 30 ml Wasser zu und rührt unter Kühlen fünf Minuten weiter. Hier auf setzt man auf einmal eine Lösung von 3,0 g CS., in 5 ml Dioxan zu. Nach weiteren 15 Minuten Rüh ren verdünnt man das Reaktionsgemisch mit 500 ml Äther und säuert mit 9 ml konzentrierter Salzsäure an. Die wässrige Phase wird entfernt und die Äther phase dreimal mit Wasser gewaschen. Nach kurzem Stehen fällt die Prednisolon-21-xanthogensäure aus.
Der Niederschlag wird auf dem Filter gesammelt, mit Äther gewaschen und bei Zimmertemperatur an der Luft getrocknet. Man erhält 2,1 g des gewünschten Produktes vom Smp. 127-128 C, das beim Weiter erhitzen wieder fest wird und bei 227-228 C noch mals schmilzt.
Kaliumsalz <I>der</I> Pr-ednisolon-21-xanthogettsäure. 1,50 g Prednisolon-21-xanthogensäure werden mit 10 ml 2 % iger alkoholischer Kalilauge vermischt. Die Substanz geht in Lösung. Nach einigem Stehen be ginnt sich das Kaliumsalz auszuscheiden und wird nach weiterem kurzem Stehen auf dem Filter gesam melt, mit Äther gewaschen und getrocknet. Aus beute 1,40 g.
Das Ultraviolettspektrum zeigt Maxima bei<I>244</I> mit<I>(E =</I> 15 200) und 304 m,cc (E = 13 400) in Wasser. <I>Beispiel 5</I> 44-Pregnen-3,20-dion-11 a,17a,21-triol-21 xanthogensäure. 5 g des obengenannten Steroids werden unter ge lindem Erwärmen in 150 ml Dioxan gelöst und die Lösung in Eiswasser bis nahe zum Gefrierpunkt des Dioxans gekühlt. Dann gibt man eine vorgekühlte Lösung von 4 g NaOH in 50 ml Wasser unter kräf tigem Rühren zu.
Man rührt und kühlt weitere 5 Minuten, wonach man eine Lösung von 8 ml CS2 in 10 ml Dioxan auf einmal zusetzt. Nachdem man wei tere 15 Minuten gerührt hat, wird mit 850 ml Äther verdünnt und mit 15 ml konzentrierter Salzsäure aus gesäuert. Die wässrige Phase wird entfernt und die Ätherphase zweimal mit Wasser gewaschen. Die 21- Xanthogensäure fällt dabei aus und wird nach kur zem Stehen bei 0 C auf dem Filter gesammelt, mit Äther gewaschen und bei Zimmertemperatur getrock net. Ausbeute 3,5 g.
Das Produkt schmilzt bei 135 bis 137 C, wird beim Weitererhitzen wieder fest, um bei 216-217 C nochmals zu schmelzen.
<I>Beispiel 6</I> d-I-Pregnett-3,11, 20-trion-17a,21-diol-21- xanthogensäure.
1,0 g Cortison wird unter Erwärmen in 200 ml Methylisobutylketon gelöst und die Lösung auf Zim- mertemperatur abkühlen gelassen. Unter kräftigem Rühren setzt man 1 ml CS., und 1 g KOH in 10 ml Wasser gelöst zu. Man rührt 45 Minuten weiter. Die untere Phase wird entfernt und die Methylisobutyl- ketonphase über Na2S04 getrocknet, filtriert und mit Petroläther versetzt.
Nach einigem Stehen bei 0I IC werden die entstandenen Kristalle durch Filtration entfernt und bei Zimmertemperatur an der Luft ge trocknet. Man erhält 0,3 g der gewünschten Xan- thogensäure.

Die untere Phase- wird entfernt und die verbleibende Di- methylformamidphase mit Äther versetzt.
Nach kur zem Stehen scheidet sich ein Niederschlag aus und der Äther wird abdekantiert. Zum verbleibenden Niederschlag gibt man Äther, filtriert und versetzt die Mutterlauge nochmals mit Äther. Der Niederschlag wird auf dem Filter gesammelt, mit Äther gewaschen und bei Zimmertemperatur im Vakuum über P205 getrocknet. Man erhält 2,7 g des gewünschten ia- liumsalzes.
Das Ultraviolettspektrum zeigt Maxima bei 244 m (F = 15 100) und 304 m,cc (@ <I>=</I> 13 500) in frisch hergestellter wässriger Lösung.
<I>Beispiel 8</I> Kaliumsalz <I>der</I> Hydrocortison <I>- 21 -</I> xanthogensäure. 3 g Hydrocortison werden in 9 ml Dimethyl- formamid gelöst. Unter heftigem Rühren versetzt man bei Zimmertemperatur mit einer Lösung von 3 g KOH in 2,3 ml Wasser und gibt dann 3 ml CS2 zu. Man rührt 45 Minuten. Die untere Phase wird ver worfen und die Dimethylfonnamidphase mit Äraher versetzt.
Es entsteht ein Niederschlag und der Äther wird nach kurzer Zeit von diesem abdekantiert. Man versetzt den Niederschlag mit Aceton, filtriert und versetzt die Mutterlauge wiederum mit Äther. Der Niederschlag wird auf dem Filter gesammelt, mit Äther gewaschen und im Vakuum bei Zimmertempe- ratur über P205 getrocknet. Man erhält 2,7 g des ge wünschten Kaliumsalzes.
<I>Beispiel 9</I> Kaliumsalz <I>der</I> Prednisolon-21-xanthogensäure. Zu einer Mischung von 2 g Prednisolon und 60 ml Aceton gibt man 620 mg KOH und 2 ml CS2. Nach 10 Minuten Rühren wird die Reaktions mischung filtriert und in Äther gegossen. Es entsteht ein Niederschlag, den man abfiltriert, mit Äther wäscht und bei Zimmertemperatur im Vakuum über P205 trocknet. Man erhält 2,1 g des gewünschten Kaliumsalzes.
