Verfahren zur Herstellung von 3-Acyloxy-46(7)-tropen Gegenstand der vorliegenden Erfindung ist ein Verfahren zur Herstellung von 3-Acyloxy-J6(7)- tropen, das dadurch gekennzeichnet ist, dass man 6-Oxy-3-acyloxy-tropan in 6-Stellung mit einer Mi neralsäure oder einer Sulfonsäure verestert, den er haltenen Ester mit einer wasserfreien organischen Base erwärmt und das so erhaltene 3-Acyloxy-46(7)- tropen abtrennt.
Diese Verbindung kann durch Hy drolyse in das J6(7)-Tropen-3-ol übergeführt werden.
Zur Durchführung dieses Verfahrens kann man das 6 - Oxy - 3 - acyloxy-tropan z. B. mit Phosphor halogenid oder Thionylhalogenid in 3-Acyloxy-6- halogen-tropan überführen und durch Behandlung des letzteren mit wasserfreier organischer Base Ha logenwasserstoff abspalten und so die ungesättigte Bindung erzeugen.
Auf ähnliche Weise kann man das 6-Oxy-3-acyl- oxy-tropan z. B. mit einem Sulfonsäurehalogenid verestern.
Als Sulfonsäurehalogenide können z. B. aroma tische, wie Benzolsulfonsäurechlorid, p-Toluol-sul- fonsäurechlorid, ss-Naphthyl-sulfonsäurechlorid, ver wendet werden. Als aliphatisches Sulfonsäurechlorid kann z. B. Methansulfonchlorid verwendet werden. Zur Abspaltung der Sulfonoxygruppe bzw. des Halogens werden vorteilhaft Pyridin-Basen, z. B. Kollidin, oder tertiäre Alkylamine, wie Triäthylamin, verwendet.

ist ein wertvolles Zwischenprodukt zur Herstellung von zwischen den Kohlenstoffatomen 6 und 7 eine Epoxydsauerstoff - Brücke enthaltenden Tropan- derivaten, so z. B. von. Scopin und dessen Estern, wie Scopolamin und Hyoscin.
<I>Beispiel d</I> 8,3 g dl-3a-Acetoxy-6ss-oxy-tropan (1,0 Mol) und 3,95 g p-Toluol-sulfonylchlorid (0,5 Mol) werden in 8 ml Chloroform 3 Stunden lang unter gelindem Rückfluss erwärmt. Zum Reaktionsgemisch werden dann 60 ml Chloroform hinzugefügt und die Chloro- formlösung viermal mit 10 ml Wasser extrahiert. Die vereinigten wässrigen Extrakte werden sechsmal mit 10 ml Chloroform ausgeschüttelt.
Die vereinigten Chloroformauszüge werden über Natriumsulfat ge trocknet und das Lösungsmittel verdampft. Das zu rückbleibende öl wird kristallisiert und dann aus Azeton-Äthergemisch umkristallisiert. Man erhält auf diese Weise 6,9 g dl-3a-Acetoxy-6f p-toluol- sulfonyloxy-tropan. Ausbeute 93,71/o. Schmelzpunkt: 79-80 C.
Nachdem man die wässrige Schicht bei obiger Trennung mit Kaliumkarbonat alkalisch gemacht hat, kann man die Hälfte des Ausgangsstoffes durch eine weitere Extraktion mit Chloroform fast quanti tativ zurückgewinnen.
3,8 g dl-3a-Acetoxy-6f-p-toluolsulfonyl-oxy- tropan werden nach Hinzufügen von 3 Tropfen Di- äthylanilin in 12 ml abs. Kollid'in gelöst, die Lösung wird in ein Bombenrohr eingeführt und durch die Lösung 5 Minuten lang trockenes sauerstofffreies Stickstoffgas geleitet.
Das Bombenrohr wird in einer Stickstoffatmosphäre zugeschmolzen und 2 Stunden auf 190 C erhitzt, das Reaktionsgemisch aus dem Bombenrohr entfernt und im Vakuum destilliert. Zum Entfernen der Kollidinspuren wird der Rest des Reaktionsgemisches 1 Stunde lang bei 2 mm H- Druck auf 50 C erwärmt. Man erhält auf diese Weise 3,70 g viskoses Öl, welches in 30 ml abs. Chloroform gelöst wird. Diese Lösung wird dreimal mit 50 ml, je 1 g Kaliumkarbonat enthaltendem Wasser ausgeschüttelt.
Die Chloroformlösung wird über Natriumsulfat getrocknet, das Lösungsmittel im Vakuum abgetrieben, wobei man 1,70 g bräunliches, bewegliches Öl erhält, welches man fraktioniert destilliert. Unter 2 mm Hg Druck bei 95 C wird als Hauptfraktion 0,67 g 3a Acetoxy-tropen-6-gewon- nen, dessen Pikrat bei 210 C schmilzt.
<I>Beispiel 11</I> 4 g dl-3a-Acetoxy-6ss-oxy-tropan (1,0 Mol) und 1,14 g Methansulfochlorid (0,5 Mol) werden mit 4 ml Chloroform abgegossen, 1 Stunde lang stehen gelassen, sodann 4 Stunden lang unter Rückfluss er wärmt. Dann wird die Lösung mit 30 ml Chloro form verdünnt, und viermal mit 5 ml Wasser extra hiert. Die Chlorofonnlösung wird über Natrium sulfat getrocknet, das Lösungsmittel verdampft, wo bei man 2,4 g dl-3a-Acetoxy-6ss-methansulfonyloxy- tropan gewinnt.
Ausbeute 86,51/e. Aus dem wässri- gen Extrakt erhält man nach Hinzufügen von Ka- liumkarbonat und mittels Extraktion mit Chloroform 450/D des Ausgangsstoffes zurück.
1,5 g des oben gewonnenen dl-3a Acetoxy-6ss- methansulfonyloxy-tropans werden in ein Bomben rohr eingetragen, mit 3 Tropfen Diäthylanilin ver setzt und in 3 ml Kollidin gelöst. Durch die Lösung wird während 10 Minuten getrocknetes sauerstoff freies Stickstoffgas durchgeführt und das Bomben rohr verschlossen. Das Reaktionsgemisch wird er hitzt und während 2 Stunden auf 190 C gehalten. Danach wird das Kollidin im Wasserstrahl-Vakuum verdampft, und zwecks Verjagen von Spuren dessel ben wird noch 1 Stunde lang bei 50 C in 2 mm Vakuum durch ein Kapillarrohr Stickstoffgas durch geleitet.
Der ölige Rest wird in 15 ml Chloroform ge löst und dreimal mit 2,5 ml 20o/oiger Kalium karbonatlösung ausgeschüttelt. Die Chloroform lösung wird über Natriumsulfat getrocknet, das Lö sungsmittel verdampft, wonach 0,70 g leicht beweg liches Öl gewonnen wird. Dieses Öl wird dann frak tioniert destilliert. Die Hauptfraktion destilliert bei 92-96 C. Auf diese Weise erhält man 0,31 g dl-3a-Acetoxy-trop-6-en, _ dessen Pikrat bei 209 C schmilzt.
<I>Beispiel 111</I> 2,8 g dl-3a-Acetoxy-6ss-oxy-tropan-hydrobromid (aus der Base mittels abs. alkoholischer Bromwasser stoffsäure erzeugt) werden mit 2,7g Phosphortri- bromid und 2 ml abs. Chloroform abgegossen. Das Reaktionsgemisch wird 2 Stunden lang gelinde ge kocht. Das Hydrobromid löst sich langsam, wobei sich milde Bromwasserstoffsäure bildet.
Das Chloro form wird verdampft, der Ölrest in 4 ml Wasser ge löst und der pH-Wert der Lösung mit Kalium- karbonat auf 8 eingestellt. Die wässrige Lösung wird zehnmal mit je 3 ml Chloroform ausgeschüttelt. Die Chloroformlösung wird über Natriumsulfat getrock net, danach das Lösungsmittel verdampft.
Das derart gewonnene Öl wird in abs. Alkohol gelöst und mit abs. alkoholischer Bromwasserstoffsäure in das Hy- drobromid des dl-3a-Acetoxy-6ss-brom-tropans über geführt. Schmelzpunkt 210 C.
1 g 6ss-Brom-3a-acetoxy-tropan wird in 6 ml Kollidin gelöst und durch die Lösung während 10 Minuten getrocknetes sauerstofffreies Stickstoffgas durchgeleitet, sodann das Bombenrohr in Stickstoff atmosphäre verschlossen. Das verschlossene Rohr wird während 4 Stunden bei 180-190 C gehalten. Nach dem Abkühlen wird das ausgeschiedene Kolli- dinbromhydrat abgenutscht und die Mutterlauge zwecks Verjagen der Kollidinspuren 1 Stunde lang auf 50 C unter Durchleiten von Stickstoffgas bei 2 mm Druck erwärmt.
Der zurückgebliebene Rest wird in 10 ml Chloroform gelöst und mit einer Lö sung von 2 g Kaliumkarbonat in 10 ml Wasser aus geschüttelt. Der wässrige Teil wird dreimal mit 5 ml Chloroform ausgeschüttelt und danach die ver einigten Chloroformextrakte im Vakuum abdestil- liert. 0,7 g braunes Öl bleibt zurück, welches unter 2 mm Druck fraktioniert destilliert wird. Die Haupt fraktion wird zwischen 92-96 C gewonnen. Man erhält als Hauptfraktion 0,2 g d1-3 a-Acetoxy-6- tropen, dessen Pikrat bei 209 C schmilzt.
Beim Verwenden von Triäthylamin anstelle von Kollidin kann man durch Arbeiten bei 110 C das selbe Endprodukt erzeugen.
