Verfahren zur Herstellung von Dihydrostreptomycin Dihydrostreptomycin ist eine für medizinische Behandlung brauchbare Substanz, weil es ein starkes und breites antibakterielles Spektrum wie Strepto- mycin besitzt und weniger giftig und stabiler als das letztere ist.
Es ist bekannt, dass Dihydrostreptomycin, welches in seinen antibakteriellen Eigenschaften dem Streptomycin ähnlich ist, gegenüber dem letzteren den Vorteil besitzt, dass es weniger giftig auf das Nerven system wirkt. Dihydrostreptomycin wurde bis jetzt durch Hydrierung des Formylrestes von Streptomycin gewonnen und als Sulfat oder Hydrochlorid auf den Markt gebracht. Diese Salze sind farblose oder weisse kristalline oder pulverförmige Substanzen.
Sie sind geruchlos oder fast geruchlos, schmecken schwach bitter, und ihre wässrigen Lösungen sind linksdrehend; eine wässrige Lösung der Sulfatkristalle zeigt eine Drehung von [a]25 = -88 (c = 1). Es ist bekannt, dass Dihydrostreptomycin ein breites antibakterielles Spektrum gegen grampositive, gramne:gative und säurefeste Bakterien besitzt.
Bis anhin wurde Dihydrostreptomycin in der Weise hergestellt, dass man das aus dem Kultur medium von Streptomyces griseus isolierte S.trepto- mycin reduzierte (siehe USA-Patent Nr.2498574). Diese Methode erfordert sehr reines Streptomycin, sonst ist ein beträchtlicher Verlust bei der Reduktion unvermeidlich.
Es wurde bis jetzt kein Bericht über die direkte Herstellung von Dihydrostreptomycin durch Kultur Dihyd@rostreptomycin produzierenderM.ikroorganismen veröffentlicht.
Es wurde nun gefunden, dass man auf einfache Weise unter Verbesserung der Ausbeute und Senkung der Gestehungskosten zu Dihydrostreptomycin ge langt, wenn man einen Stamm eines Dihydrostrepto- mycin erzeugenden Mikroorganismus der Gattung Streptomyces in .einem wässrigen Nährmedium aerob kultiviert.
Das so erzeugte Dihydrostreptomycin kann aus dem Kulturmedium isoliert werden.
Es kann jeder Stamm von jeder zu den Strepto- myceten gehörenden Art, welcher Dihydrostrepto- mycin liefert, verwendet werden. So kann z. B. eine neue Art, welche die Bezeichnung Streptomyces. humi- dus nov. sp. Nr.<B>23572</B> erhalten hat, für diesen Zweck verwendet werden. Diese Art hat die in der folgen den Tabelle aufgeführten charakteristischen Eigen schaften.

Streptomyces humidus erzeugt dagegen Dihydro- streptomycin.
Der oben erwähnte zu Streptomyces humidus ge hörende Stamm ist hygroskopisch, aber einige Stämme, die zu der gleichen Art gehören, haben diese Eigen schaft nicht. Ebenso bilden einige kein Luftmycel, und andere produzieren Pigment.
Dihydrostreptomycin erzeugende Stämme anderer Arten können auch dann für den gleichen Zweck gebraucht werden, wenn sie in ihren Eigenschaften Streptomyces humidus nov. sp. nicht sehr ähnlich sind.
Wie bei Mikroörganismen, insbesondere bei Streptorayceten, beobachtet wird, wechselt ihr Ver halten im Kulturmedium leicht spontan oder sie wer den künstlich geändert. Deshalb ist die Identifizierung einer Art so schwierig, dass jegliche blosse Beschrei bung der Eigenschaften einer Art für die Identifi zierung dieser Art keine definitive Bedeutung hat. Es können auch Mutanten von Dihydrostreptomycin erzeugenden Arten, wie sie z. B. durch Behandlung mit Röntgenstrahlen, ultraviolette Strahlen und Che mikalien entstehen, verwendet werden.

Dihydro- streptomycin als auch Streptomycin resistent. Zum Unterschied von den Antibiotica, die durch Reduk tion von Streptomycin erhalten werden, sind die durch Kultivierung von Dihydrostreptomycin erzeu genden Mikroorganismen erhaltenen Antibiotica im allgemeinen negativ gegenüber der Maltolreaktion. Die Maltolreaktion ist eine Farbreaktion des Strepto- mycins mit Phenolreagens,
die durch Hydrolyse des Streptomycins mittels Alkali erzeugt wird [Schenk et a1., J. Amer. Chem. Soc. 67, 2277 (1945), Boxer, J. Biol. Chem. 169, 153 (1947), Eiseman et a1., Anal. Chem. 21, 1507 (1949)]. Diese Eigenschaften können vorteilhaft zur Trennung verschiedener Stämme zur Anwendung gebracht werden. Es können z.
B. die gewünschten Stämme selektiv durch die Verdün nungsmethode oder durch Kultivierung in einem Dihydrostreptomycin enthaltenden Medium getrennt werden.
Als Kohlenstoffquelle können z. B. Stärke, Lac- tose, Saccharose, Dextrin, Glycerin und Maltose ge braucht werden. Als Stickstoffquelle können orga nische oder anorganische, Stickstoff enthaltende Substanzen verwendet werden, wie z. B. Sojabohnen mehl, Fleischextrakt, Peptone, Erdnusspulver; Kasein, Aminosäuren, Hefe, Kleie, Maismaische, Baumwoll- samenpulver, Nitrate, Harnstoff oder Ammonium verbindungen.
Es können auch kleine Mengen von anorganischen Salzen und das Wachstum fördernde Substanzen der Nährlösung zugesetzt werden. Andere geeignete Wachstumsquellen sind Mycelien eines zu Penicillium gehörenden Stammes oder z. B. dessen Kulturmedium. Jedes für die Kultivierung von Streptomyces griseus geeignete Nährmedium erfüllt den gleichen Zweck. Eine geeignete Muttersubstanz kann, wenn notwendig, beigefügt werden.
Das Kulturmedium kann fest oder flüssig sein; eine submerse und aerobe Kultivierung ist für industrielle Zwecke am vorteilhaftesten.
Wenn Streptomyces humidus als der Dihydro- streptomycin liefernde Mikroorganismus verwendet wird und die Kultivierung unter submersen aeroben Bedingungen durchgeführt wird, so kann dieselbe vorzugsweise bei einer Temperatur von etwa 24 bis 30 C in einer Zeitperiode von 3 bis 8 Tagen vollzogen werden. Aber- die Temperatur und die Zeitperiode muss den anderen Kulturbedingungen angepasst wer den. Bei Verwendung anderer Dihydrostreptomycin erzeugenden Mikroorganismen werden jeweils die für deren Kultivierung geeignetsten Bedingungen gewählt.
Das so erzeugte Dihydrostreptomycin kann mit Hilfe verschieden physikalischer oder chemischer Methoden in optimaler Reinheit aus einem Dihydro- streptomycin enthaltenden Nährmedium isoliert wer den. Hierbei wird ein für den Gebrauchszweck hoher Reinheitsgrad erzielt.
Zur Isolierung des Dihydrostreptomycins aus dem Nährmedium und Abtrennung von den Verunreini gungen kann man z. B. von den Verschiedenheiten zwischen Dihydrostreptomycin oder seinen Salzen und den Verunreinigungen in bezug auf die Adsor- bierbarkeit, die Löslichkeit, den Verteilungskoeffi zienten zwischen zwei Lösungsmitteln und der Stärke, mit der Ionen gebunden werden, Gebrauch machen.
Im Falle der Kultivierung in flüssigem Medium kann das Antibiotikum bis zu einer Konzentration von mehreren tausend Mikrogramm pro em3 im Kultur medium angereichert werden; man kann jedoch das Dihydrostreptomycin schon aus einer nur 10 y/em3 enthaltenden Lösung isolieren.
Die roh aus dem Kulturmedium gewonnene Sub stanz enthält neben Dihydrostreptomycin natürlich verschiedene Verunreinigungen, wie Kohlehydrate, Proteinsubstanzen, Salze, tierische und vegetabilische Substanzen, Mycele von Mikroorganismen und deren Umwandlungsprodukte. Jede dieser Verunreinigun gen enthält z. B. die folgenden Substanzen: Kohlehydrate: Stärke, Zucker, wasserlösliches Kohle hydrat.
Proteinsubstanzen: Aminosäuren, Kasein, Fleisch extrakt, Protein, Peptone.
Tierische und vegetabilische Substanzen: Protein substanzen, Fette, Fettsäuren, Lipoide, Cellulose. Salze: Ammoniumsalze und Amine, Salze von Alkali metallen, von Calcium, Magnesium, Eisen, Mangan, Kupfer, Zink. Umwandlungsprodukte von Mikroorganismen: Enzyme, Proteinsubstanzen, Zucker, mehrwertige Alkohole, Fettsäuren.
Ausser diesen Substanzen können durch Synthese Verbindungen wie Harnstoff erhalten werden.
Die Zusammensetzung der Verunreinigungen ist sehr verwickelt, aber diese variiert in Qualität und Quantität je nach Ausmass und Art der verwendeten Ausgangsstoffe. Im Nährmedium können auch noch andere Verunreinigungen, wie pH-Regulierungsmittel, Fäl#lungsmittel, Adsorbierungsmittel, enthalten sein.
Die Abtrennung des Dihydrostreptomycins aus der Mischung von den genannten Verunreinigungen kann auf verschiedene Weise erfolgen.
Eines dieser Mittel macht von dem Unterschied zwischen Dihydrostreptomycin und den Verunreini gungen in bezug auf Löslichkeit Gebrauch. Bei der flüssigen Kultur wird eine Dihydrostreptomycin- lösung durch Filtration des Gärungsgemisches gewon nen, weil sich dasselbe vorzugsweise im flüssigen Teil befindet. Aber ein kleinerer Anteil der aktiven Ver bindung wird in den Festbestandteilen gefunden und kann durch Extraktion mit heissem Wasser daraus gesammelt werden.
In diesem Falle können Ver unreinigungen der Proteinreihe dadurch eliminiert werden, dass man das pH auf den isoelektrischen Punkt des Proteins einstellt oder indem man vor, nach oder mit gleichzeitiger Entfernung des Mycels Substanzen zusetzt, welche die Verunreinigungen fällen. Wenn das pH des Gärungsgemisches hoch ist, ist der Zusatz einer sauren Substanz angebracht, um das Protein leichter zu fällen. Wenn die Konzentra tion des Dihydrostreptomycins in der Lösung genü gend hoch ist, kann das Antibiotikum durch blosses Zufügen eines geeigneten Fällungsmittels gefällt wer den.
Wenn die Konzentration des Dihydrostrepto- mycins nichtgenügend ist, wird ein Teil des Lösungs mittels verdampft. Als Fällungsmittel werden z. B. Phosphorwolframsäure, Alkyl- oder Aralkylsulfon- säuren, saure Azoverbindungen und Polyhalogen phenolverbindungen verwendet. Alle diese bilden mit Dihydrostreptomycin Salze.
Es können manche andere Verbindungen verwendet werden, und die meisten von ihnen sind saure Verbindungen mit Carboxylresten. In manchen Fällen können die so gebildeten Salze des Dihydrostreptomycins Doppel- oder Komplexsalze sein. Es können auch, wie später beschrieben wird, als eine Art von Fällungsmittel Ionenaustauscher verwendet werden. Um die Fällung der aktiven Substanz oder der Verunreinigungen zu beschleunigen, kann man auch, wenn notwendig, aussalzen.
Wenn feste Kulturen verwendet werden, so wird das Festmaterial mit Wasser, einem wässrigen Lö sungsmittel oder einem anderen geeigneten Lösungs mittel extrahiert, um eine Lösung von Dihydro- streptomycin zu erhalten. Wenn das Gärungsgemisch vorher in eine Festsubstanz übergeführt worden ist, so kann diese, wie oben beschrieben, aufgearbeitet werden; in diesem Falle wird aber die Extraktion bei einem geeigneten pH durchgeführt.
Ein anderes Mittel für die Abtrennung des Di- hydrostreptomycins besteht darin, dass man von der Verschiedenheit zwischen der antibiotisch aktiven Verbindung und den Verunreinigungen in bezug auf die Adsorbierbarkeit Gebrauch macht. Dihydro- streptomycin wird im Kulturfiltrat z. B. durch Zusatz eines geeigneten Adsorbierungsmittels adsorbiert. Wenn notwendig, kann von der Adsorptionschromato- graphie Gebrauch gemacht werden. Als Adsorptions- mittel werden z. B.
Aktivkohle, Alaunerde und Silicagel verwendet. Wenn man das Adsorptions- mittel abtrennt, bleibt der grösste Teil der Verunrei nigungen im flüssigen Anteil. Das Adsorptionsmittel wird dann mit einem geeigneten Lösungsmittel elu fiert, wobei die nicht eluierbaren Verunreinigungen im Ad sorptionsmittel bleiben. Die El:uierung wird im allge meinen mit einem sauren Lösungsmittel durchgeführt. Chromatographie bewirkt eine exaktere Trennung, aber diese Methode erfordert kompliziertere Ver fahren.
Unterschiede zwischen der antibiotisch aktiven Verbindung und den Verunreinigungen in bezug auf den Verteilungskoeffizienten zwischen zwei Lösungs mitteln können ebenfalls für die Trennung verwendet werden. Als Lösungsmittel können Wasser oder ein wässriges Lösungsmittel und ein organisches mit dem letzteren nicht mischbaren Lösungsmittel verwendet werden. In diesem Falle ist die aktive Verbindung geneigt, sich im wässrigen Lösungsmittel zu verteilen, wenn die Lösungsmittel stark sauer sind. Wenn die Lösungsmittel anderseits neutral oder basisch sind und eine höhere Fettsäure, Pikrinsäure und derglei chen mit zugegen sind, so geht die aktive Verbindung in die organische Lösungsmittelschicht über.
Dem gemäss ist es möglich, die aktive Verbindung im Mate rial in ein organisches Lösungsmittel aufzunehmen und dann in Wasser oder ein wässriges Lösungs mittel zu übertragen. Als organische Lösungsmittel werden z. B. Butanole, Pentanole, Phenole, niedere Fettsäureester, Methylisobutylketon, Diäthyläther und halogenierte Kohlenwasserstoffe, wie Chloroform, Tetrachlo:rkohlenstoff, verwendet.
Diese Methode kann in Form einer Verteilungschromatographie oder einer Gegenstromverteilungsmethode zur Anwendung gebracht werden. Die Trennung kann auch so durchgeführt werden, dass von den Unterschieden zwischen der anti biotisch aktiven Verbindung ,und den Verunreinigun gen in bezug auf die Stärke des lonenbindungsver- mögens Gebrauch gemacht wird. Dies wird z. B. da durch erreicht, dass man eine Lösung der aktiven Verbindung mit einem Ionenaustauscher in Kontakt bringt.
In diesem Falle adsorbiert ein Anionen- austauscher anionische Verunreinigungen und belässt die aktive Verbindung in der Lösung, während ein Kationenaustauscher die antibiotisch aktive Verbin dung adsorbiert. Im letzten Falle werden kationische Verunreinigungen ebenfalls adsorbiert, aber letztere Adsorption kann, wenn man einen ,geeigneten Ionen- austauscher verwendet,
möglichst klein gehalten wer den. Die Harze des Sulfonsäuretyps adsorbieren z. B. leicht die Verunreinigungen, aber die Harze des Carboxylsäuretyps haben eine geringe Tendenz hier für. Die Zugabe der Ionenaustauscher kann nach der Mischungsmethode erfolgen, der Gebrauch einer Kolonne für das Harz ist in manchen Fällen eine passende Methode. Diese Methode kann für eine verdünnte Lösung der antibiotisch aktiven Verbin dung wie für das Kulturfiltrat gebraucht werden.
Man kann auch die Dialyse verwenden, um die Verunreinigungen mit hohem Molekulargewicht zu entfernen, aber dieser Prozess ist kompliziert.
Die oben erwähnten Mittel können getrennt für sich oder je nach der Qualität des Materials und dem Verwendungszweck des Produktes, in Kombination zur Anwendung .gelangen. Man kann auch so vor gehen, dass man z. B. das Kulturmedium vorerst filtriert, um die Testbestandteile oder die nach der Kultivierung niedergeschlagenen Substanzen zu ent fernen.
Die antibiotisch aktive Verbindung im Filtrat wird an einen Kationenaustauscher adsorbiert und dann mit einem sauren Lösungsmittel eluiert. Es wird so eine konzentrierte Lösung der aktiven Ver bindung erzielt, welche nur eine kleine Menge von Verunreinigungen enthält. Die Lösung wird, wenn notwendig, weiter konzentriert und einer Adsorptions- Chromatographie unterworfen, um fast alle Verun reinigungen zu entfernen. Aus der konzentrierten Lösung kann das Antibiotikum mit einem geeigne ten Mittel gefällt werden.
Der Niederschlag wird dann in einem geeigneten organischen Lösungsmittel ge löst und eine Säure zugesetzt, worauf das entspre chende saure Salz des Dihydrostreptomycins erhal ten wird. Wenn das Produkt umkristallisiert wird, wird das saure Salz in fast reinem Zustand erhalten.
Alle Arbeitsprozesse, wie Filtrieren, Waschen, Heizen, Kühlen, Mischen, Destillieren, Verdampfen und Trocknen, können nacheinander oder gleich zeitig durchgeführt werden.
Um die Zersetzung der antibiotisch aktiven Ver bindung zu verhindern., wird zweckmässig unter nicht extrem sauren oder .extrem alkalischen Bedingungen gearbeitet. Man kann zwar, wenn notwendig, erhitzen; aber es ist vorzuziehen, nicht allzulange zu erhitzen. Im Vergleich mit anderen Antibiotika ist Dihydro- streptomycin im Kulturmedium verhältnismässig stabil, so dass alle Manipulationen ohne Schwierigkeit durch geführt werden können.
Das so erhaltene Dihydrostreptomycin wurde auf Grund folgender Daten identifiziert: <I>A. Physikalische und chemische Eigenschaften</I> 1. Das Infrarotabsorptionsspektrum wurde von den folgenden Derivaten sowohl des erfindungs gemäss erhältlichen Dihydrostreptomycins (im folgen den als Antibiotikum 21<B>572</B> bezeichnet) als auch des Dihydrostreptomycins des Handels bestimmt. I.
Sulfat, II. Streptidinpicrat, <B>111.</B> a-Methyl-pentaacetyldihydrostreptobio@aminid. Die IR-Spektren der Derivate des erfindungs gemäss erhaltenen Dihydrostreptomycins sind mit den IR-Spektren der entsprechenden Derivate des Di- hydrostreptomycins des Handels identisch.
Alle Spektren sind in Nujolparaffinöl mit einem Natriumchlorid Prisma gemessen.
2. Die ultravioletten Spektren vom Sulfat des Antibiotikums 23 572 und diejenigen eines handeln üblichen Musters von Dihydrostreptomycinsuifat stimmen gut überein, es wurde keine spezifische Ad sorption beobachtet.
3. Die analytischen Werte des Sulfates vom Antibiotikum 23 572 stimmen mit den theoretischen Werten des Dihydrostreptomycinsulfates gut überein. Die Werte für Dihydrostreptomycinsulfat (C21H41012N7)2 - 3 H2S04) sind: Berechnet: C 34,42'%; H 6,0511/G; N 13,3804; S 6,561/0 Gefunden:
C 34,45"/ü; H 6,4211/o; N 12,981/o,; S 6,43% 34,110/0; 6,54 0/0 4. Das Sulfat des Antibiotikums 23 572 zeigt eine Drehung von (a)25 - 87,4 (C = 1, H,0), während das Dihydrostreptomycinsulfat eine Drehung von (a)<B>17</B> - 86,0 zeigt.
Die spezifische Drehung von Dihydrostrepto- mycin wurde von I. A. Solomons et a1. in Science 109, 515 (1949) mit (a)D - 88 und von F. J. Wolf et a1. in J. Am. Chem. Soc. 68, 2163 (1946) mit (a)D - 88,5 angegeben.
5. Das Sulfat des Antibiotikums 23 572 schmilzt unter Zersetzung und Schwarzfärbung bei 250 bis 255 C, das Dihydrostreptomycinsulfat schmilzt eben falls unter Zersetzung und Schwarzfärbung bei 250 bis 255 C.
6. Sowohl das Antibiotikum 23 572 als auch das Dihydrostreptomycin verhalten sich gegenüber der Maltol-Reaktion negativ, Streptomycin spricht da gegen an.
7. Das Antibiotikum<B>23572,</B> das Dihydro- ,streptomycin und Streptomycin verhalten sich gegen über der Sakabguschi-Reaktion positiv.
B. Lösungen des Sulfates vom Antibiotikum <B>23572</B> des Dihyd,rostreptomycinsulfates und des Streptomycinsulfates in 0,1n NaOH in einer Kon zentration von 5 mg/cm3 wurden bei Raumtempe ratur 24 und 48 Stunden lang stehengelassen und die antibakterielle Wirksamkeit jedes Antibiotikums nach der Verdünnungsmethode bestimmt.

Soc. 73, 2290 (1951)] in folgender Weise erhalten: 2 g des Hydrochlorides vom Anti biotikum<B>23572</B> wurden in<B>100</B> cm3 wasserfreiem Methanol, welches 10/9 Chlorwasserstoff enthielt, aufgelöst. Nach 48stündigem Stehen bei Raumtempe ratur wurden zur Lösung unter Rühren 200 cm3 Äther zugesetzt und die entstandenen weissen Kristalle (Streptidinhydrochlorid) durch Zentrifugieren abge trennt. Die Kristalle wurden in 20 cm3 Wasser gelöst und eine gesättigte Picrinsäurelösung zugegeben.
Nach mehrstündigem Stehen wurden die niedergeschlage nen Kristalle abgetrennt und aus Wasser umkristalli- siert. Es wurden als gelbliche Nadeln mit einem Schmelzpunkt von 271-273 C 400 mg Streptidin- pikrat erhalten.
Die Analyse für C8H1$O4N6 - 2 C6HH3N307 zeigte: Berechnet: C 33,340/9; H 3,360/0; N 23,33% Gefunden: C 33,46%; H 3,440;9;
N 23,49% Das Infrarotabsorptionsspektrum des Picrates stimmte mit dem des aus Dihydrostreptomycin her gestellten Picrates wie unter 1 beschrieben überein.
Die über obigem Streptidinhydrochlorid befind liche Flüssigkeit wurde mit einer Lösung von Na- triumhvdroxyd in Methylalkohol neutralisiert, das entstandene Natriumchlorid durch Zentrifugieren ab getrennt und das Lösungsmittel unter vermindertem Druck abdestilliert. Es hinterblieb ein weisses Pulver. Dieser Rückstand wurde in 20 cm3 Pyridin aufgelöst und 7 cm' Es:sigsäureanhydrid zugesetzt.
Nach Stehen über Nacht wurde zur Reaktionsmischung Wasser zugesetzt und das Lösungsmittel abdestilliert. Der Rückstand wurde aus Methanol umkristal isiert. Es wurde in Form von weissen Nadeln mit einem Schmelzpunkt von 194,5 C a-Methyl-pentaacetyldi- hydrostreptobiosaminid erhalten. Die Ausbeute be trug 700 mg.
Die Analyse für C13H19NO8(CH.C0)5(OCH3) ergab: Berechnet: C 51,15%-; H 6,62%; N 2,490/0 Gefunden: C 51,09'%-; H 6,l3'0/9;
N 2,40%. Die Drehung (a) = -120,0 (c = 1%, D in Chloroform).
Das Infrarotabsorptionsspektrum der erhaltenen Verbindung war in guter übereinstimmung mit dem aus Handels-Dihydrostreptomycin erhaltenen a Methyl-pentaacetyldihydrostreptobiosaminid, wie be reits unter 1 beschrieben ist.
Das ss-Methyl-pentaacetyldihydrostreptobiosaminid wurde aus Antibiotikum 23 572 nach dem Verfahren von N. G. Brink [J. Am. Chem. Soc. <B>68,2557</B> (1946)] in folgender Weise erhalten:
10 g Hydrochlorid des Antibiotikums 23 572 wurden in 500 cm3 Methanol, das 1 % Chlorwasserstoff enthielt, aufgelöst. Nach- dem die Lösung bei Raumtemperatur über Nacht gestanden hatte, wurde das Lösungsmittel unter ver mindertem Druck abdestilliert. Der Rückstand wurde in 750 cm3 Methanol aufgelöst und 450 cm3 Äther zugesetzt.
Die Lösung wurde durch eine Kolonne geschickt, die mit 210 g sauer gewaschener Alaunerde beschickt war, die mit einer Methanol-Äther- Mischung im Verhältnis 2: 1 imprägniert war. Die Kolonne wurde mit 1000 cm3 einer Methanol-Äther- Mischung im Verhältnis<B>3:2</B> gewaschen. Die ausge flossene Lösung wurde unter vermindertem Druck zur Trockene eingedampft.
Der Rückstand wurde acety- liert, indem man mit 10 cm3 Essigsäureanhydrid und 10 cm3 Pyrid'in bei Raumtemperatur über Nacht stehen liess. Es wurde dann Wasser dazugegeben und die Lösung im Vakuum zur Trockne verdampft. Der Rückstand wurde in Chloroform aufgelöst und die Chloroformlösung mit Wasser, verdünnter Schwefel säure und schliesslich mit Wasser gewaschen.
Nach dem das Chloroform abdestilliert war, wurde der bräunliche feste Rückstand 2 Minuten lang mit <B>100</B> cm3 Äther gekocht und die Ätherlösung durch Dekantieren abgetrennt. Das gleiche Verfahren wurde nochmals wiederholt. Die in Äther unlösliche Frak tion wurde aus Methanol kristallisiert und liefert a - Methyl - pentaacetyldihydrostreptobiosaminid mit einem Schmelzpunkt von 194,5 C.
Die Ätherlösung wurde auf ungefähr 50 cm3 eingeengt und Petroläther zugesetzt, wobei sich weisse Kristalle ergaben. Um kristallisation aus Methanol ergab ungefähr 100 mg ss - Methyl- pentaacetyldihydrostreptobiosaminid mit einem Schmelzpunkt von 155-156 C. Es wurde keine Schmelzpunktdepression beobachtet, wenn es mit dem ss-Isomeren gemischt wurde, das aus Di- hydrostreptomycin nach dem eben angegebenen Ver fahren hergestellt war.
Die Analyse .ergab für C"H19NOs(CH.C0)5(OCH3): C 50,710/0; H 6,620/a; N 2,6511/o. Die berechneten. Werte sind: C 51,15%; H 6,620/0; N 2,4911/o. (a)= - 36 . (C = 1/Chloroform).
D Das Infrarotspektrum des erhaltenen Produktes war in guter Übereinstimmung mit dem des aus Dihydro- streptomycin erhaltenen ,ss - Methyl - pentaacetyldi- hydrobiosaminid.
Eine Lösung von 260 mg a-Methyl-pentaacetyl- dihydrostreptobiosaminid, das aus dem Antibiotikum <B>23572</B> erhalten war, in 20 cm3 einer 10 n/odgen Salzsäure wurde 3 Stunden lang unter Rückfluss ge kocht. Nach Abkühlen wurde die bräunliche Lösung mit Holzkohle entfärbt und im Vakuum zur Trockne verdampft.
Der Rückstand wurde bei Raumtempe ratur mit 1 cm3 Essigsäureanhydrid und 3 em3 Pyridin acetyliert. Nach Zusatz von Wasser wurde die Lösung zur Trockne eingedampft. Der Rückstand wurde in 50 cm3 Benzol-Petroläther-Mischung im Verhältnis 7 : 3 gelöst.
Die Lösung wurde durch eine Kolonne gelassen, welche mit 4 g einer sauer ge waschenen, mit Petroläther imprägnierten Alaunerde beschickt war, wobei das Acetylierungsprodukt auf der sauer gewaschenen Alaunerde adsorbiert wurde.
Die Kolonne wurde mit 100 cm3 einer Benzol Chloroform-Mischung im Verhältnis 7: 3 behandelt, um die betreffende Substanz zu eluieren. Das Effluans wurde auf ungefähr 10 cm3 eingeengt und 30 cm3 Äther zugesetzt, wobei sich Kristalle ergaben.- Um kristallisation aus Chloroform-Äther ergab 25 mg Nadeln von Pentaacetyl-N-methyl-L-glucosamin mit einem Schmelzpunkt von 158-159 C.
Die Analyse ergab: C 50,71'%; H 6,14 11/a; N 3,25 0/0. Die berechneten Werte für C17H"N010 sind:
C 50,62%; H 6,250/0; N 3,4711/o. (a) D = -102 (c = 0,7'% in Chloroform). Das Infrarotabsorptionsspektrum des erhaltenen Produktes war in völliger übereinstimmung mit dem des Pentaacetyl-N-methyl-L-glucosamins,
das nach der beschriebenen Methode aus Dihydrostreptomycin erhalten war. Es wurde keine Schmelzpunktsdepres- sion beobachtet, wenn es mit Pentaacetyl-N-methyl- L-glucosamin gemischt wurde, das aus Dihydro- streptomycin erhalten war.
13. Antibiotikum 23 572 wurde mit Alkali unter ähnlichen Bedingungen hydrolysiert, wie es für die Hydrolyse von Hydroxystreptomycin verwendet und von F. H. Stodola et a1. [J. Am. Chem. Soc. 73, 2290 (1951)] beschrieben wurde. Demgemäss wurde eine Lösung von 2 g Hydrochlorid des Antibiotikums 23 572 in 40 cm3 l11 NaOH 3 Stunden lang auf 100 C erhitzt. Die Reaktionsmischung wurde mit Salzsäure angesäuert und aufgearbeitet.
Es konnte aber weder eine Substanz mit positiver Reaktion gegenüber Maltot noch eine unter sauren Bedingungen extrahier bare Substanz gefunden werden, das heisst das Anti biotikum 23 572 verhält sich bei dieser Prüfung anders als Streptomycin und Hydroxystreptomycin. Die gleiche Prüfung ergab mit Dihydrostreptomycin das gleiche Resultat.
14. Zu einer wässrigen Lösung von 50000 y/em3 des Antibiotikums 23 572 wurde eine warme Lösung von Methylorange zugesetzt, bis kein Niederschlag mehr gebildet wurde. Der Niederschlag wurde abfil- triert und aus Methanol-Wasser (1 :3) umkristalli siert. Es wurde in Form von Schuppen mit einem Schmelzpunkt von 222-225 C das Helianthat ge wonnen (Wirksamkeit: 340 Streptomycin-Einheiten pro mg).
Die Analyse für Dihydrostreptomycin- helianthat (C03H$00.,1N10s3) ergab: Berechnet: C 50,46%; H 5,7911/o; N 14,95%; S 6,4011/o Gefunden:
C 50,19%; H 5,93%; N 14,93%; S 6,801/0 50,84114; 5,95% 15. Die Papierverteilungschromatographie von Streptomycin und Dihydrostreptomycin wurde von F. H. Stodola et a1. [J. Am. Soc. 73, 2290 (1951)] geprüft.
Es wurde mit Wasser gesättigtes Butanol mit 2 /o p-Toluolsulfonsäure und 2 /o Piperidin als Lö sungsmittel verwendet. Beide Verbindungen wurden mit Hilfe des Bioautogrammes unterschieden.
Nach der gleichen Methode wurde die Papier chromatographie auf die beiden obigen Verbindungen und das Antibiotikum 23 572 angewandt. Die Bio- autogramme wurden mit B. subtilis als Prüfungs- Mikroorganismen geprüft. Es ergab sich, dass das Antibiotikum an der gleichen Stelle eine das Wachs tum verhindernde Zone zeigte, wie das Dihydro- streptomycin.
16. Die kristallographischen Konstanten des Sul fates vom Antibiotikum 23 572 wurden im Vergleich mit denen des von F. J. Wolf et a1. [Science 109, 515 (1949)] berichteten Dihydrostreptomycinsulfates un tersucht.

Die antibakterielle Wirk samkeit des Kulturmediums erreichte 1200 mcg/cm3 gegenüber B. subtilis. <I>Beispiel 3</I> Stamm H-120, welcher zur Streptomyces humi- dus gehört, wurde in verschiedenen Nährlösungen, die aus Siebabfällen von Cerealien bestanden, aerob kultiviert. Die Resultate werden in der folgenden Tabelle gezeigt.
Jede der in den Beispielen 3 bis 8 beschriebenen Kulturen wurde mit 50 cm3 des Kul turmediums in einem Gefäss mit 300 cm3 Fassungs raum unter Schütteln bei 26-28 C durchgeführt.

Zu 700 Liter des, Filtrates des Kultur mediums [pH: 8,5, Wirksamkeit: 35 E/cm3 (Ver dünnungseinheit) gegen E. coli] wurden 7 kg (10/0) aktive Kohle zugesetzt und die Mischung 30 Mi nuten lang gerührt, um die aktive Verbindung zu adsorbieren. Die aktive Kohle wurde mit der 10fachen Menge ihres Volumens an methanolischem Chlor wasserstoff 30 Minuten lang bei einem pH von 2,0 eluiert. Es wurden so 140 Liter Eluat mit einer Wirksamkeit von 100 E/cm3 gewonnen.
Die Eluierung wurde noch einmal' wiederholt, wobei 70 Liter Eluat von einer Wirksamkeit von 35 E/cm3 gewonnen wur den. Die vereinigten Eluate wurden mit n NaOH bis zu einem pH von 6,5 fast neutral gestellt und im Vakuum bei niederer Temperatur bis auf etwa 300 cm3 eingeengt. Während dieser Arbeitsweise wurde das abgeschiedene Natriumchlorid entfernt und dann 3 Liter wasserfreies Aceton zur konzentrierten Lösung zugesetzt.
Die niedergeschlagene aktive Ver bindung wurde im Vakuum getrocknet und ergab ein weisses Pulver. Die Ausbeute betrug 147 g oder 6011/o mit einer Wirksamkeit von 100 E/mg. Das erzielte Produkt ist gegenüber .der Maltolreaktion negativ, gegenüber der Sakaguchireaktion aber positiv.
<I>Beispiel 10</I> Reinigung durch Ionenaustauscher a) 500 Liter des Kulturfiltrates [pH 7,7, Wirk samkeit: 350 E/cm3 (Verdünnungseinheit) gegenüber E. Coli], das wie in Beispiel 9 erhalten wurde, wurden durch einen Turm, der mit 30 Liter Amberlite IRC-50 (H-Typ, eingetragene Marke) gefüllt war, mit einer Geschwindigkeit von 1,8 Liter pro Minute laufen gelassen.
Die Wirksamkeit des Effluens war gegenüber E. coli geringer als 10 E/cm3. Das Harz wurde mit Wasser gewaschen, mit 0,5 n HCl eluiert und 36 Liter des gefärbten Eluates mit einer Wirk samkeit von 3500 E/cms gegenüber E. coli gesam- melt. Das Eluat wurde mit n NaOH auf ein pH von 6,5 eingestellt und bei niederen Temperaturen unter gelegentlicher Entfernung des abgeschiedenen Na triumchlorides auf 1 Liter eingeengt.
Die Wirksam- keit war 160000 E/cm3, die Ausbeute betrug 91,5 %-.
b) Zu 748 Liter des nach Beispiel 9 erhaltenen Filtrates der Züchtung (Gehalt an Dihydrostrepto- mycin ungefähr 2,200 meg/cm3) wurden 4,1 kg Oxal- säure zugesetzt und der erhaltene Niederschlag durch einen De-Laval-Separator abgeschieden. Das pH wurde mit einer Natriumhydroxydlösung auf ungefähr 7,0 ein gestellt und der dabei erhaltene Niederschlag wurde ebenfalls abgetrennt.
Die Lösung wurde durch einen Turm, der mit 6,3 Liter Amberlite IRC-50 (Na-Typ) gefüllt war, mit einer Geschwindigkeit von 40 Liter/ Stunde durchgelassen, worauf das Effluens nur eine Wirksamkeit von 12-6 meg/cm3 zeigte. Das Herz wurde mit Wasser gewaschen und in aktive Verbin dung eluiert, indem man 4 /oige Salzsäure mit einer Geschwindigkeit von 3 Liter/Stunde durchschickte.
20 Liter des so erhaltenen Eluates wurden durch Zusatz von Amberlite IR-45 (OH-Typ) oder Amberlite IRA-400 (OH-Typ) neutralisiert und auf 2,5 Liter unter vermindertem Druck bei niedriger Temperatur eingeengt. Die Wirksamkeit betrug <B>368000</B> meg/cm3.
<I>Beispiel 11</I> Extraktion mit einem organischen Lösungsmittel zu 1 Liter des nach Beispiel<B>10'</B> b) erhaltenen Filtrates der Züchtung mit einer Wirksamkeit von 1001 meg/em3 gegenüber B. subtilis wurden 400 cm3 n-Butylalkohol oder Isoamylalkohol, welcher 3 1/o Laurinsäure enthielt, zugesetzt, die wässerige Schicht wurde auf ein pH von 7,5 eingestellt und die Mi schung 20 Minuten lang geschüttelt.
Nach Abtren nung der Butylal:kohol- oder der Isoamylalkohol- Schicht wurde die gleiche Extraktion noch einmal vorgenommen. Die vereinigten Butylalkohol- oder Isoamylalkohol-Lösungen wurden mit verdünnter Schwefelsäure in Wasser derartig geschüttelt, dass die resultierende wässrige Schicht 120 cm3 betrug und deren pH 2,0 war. Nach Wiederholung der glei chen Extraktion wurden die vereinigten Extrakte mit Äther geschüttelt, um die darin enthaltene Laurin- säure zu entfernen.
Die Wirksamkeit der so erhaltenen Lösung von 25 cm3 war gegenüber B. subtilis 2891 mcg/cm3, die Ausbeute betrug 72,5 %. <I>Beispiel 12</I> Chromatographie auf aktiver Kohle. 1 Liter einer nach Beispiel 10 cz) erhaltenen Lösung mit einer Wirksamkeit von 160000 E/cm3 (Verdünnungsem- heften) gegen E.
coli wurde mit n HCl auf ein. pH von 3,5 eingestellt und durch einen 8 cm breiten und 50 cm hohen Turm geschickt, der mit 50<B>0</B> g aktiver, mit Wasser vom pH 3,5 imprägnierter Kohle beschickt war. Das Effluens wurde in Teile von 300, cm3 geteilt. Der Turm wurde mit destilliertem Wasser behandelt, das mit Salzsäure auf ein pH von 3,5 eingestellt war. Die Fraktionen 1-11 enthielten Natriumchlorid und Verunreinigungen, während die anderen die aktive Verbindung enthielten, wie unten gezeigt wird.
Die Fraktionen 13-21 wurden vereinigt, durch Zusatz von Amberlite IRA-400 (OH-Typ) auf ein pH von 6,5 eingestellt und unter vermindertem Druck bei niederer Temperatur :eingeengt. Zu der konzen trierten Lösung wurde die zehnfache Menge ihres Volumens wasserfreien Acetons zugesetzt, der ent standene Niederschlag durch Dekantieren von der Flüssigkeit getrennt und getrocknet. Es wurden 101 g des Hydrochlorides als eine farblose amorphe Sub stanz gewonnen.
Dies entspricht 783,3 mcg/mg an Dihydrostreptomycin. Zu einer Lösung von 10 g dieses Produktes in 50 cm3 Wasser wurde :eine Lö sung von 6 g M9S04. 7H20 in 7 cm3 Wasser gege ben. Die Mischung wurde mit verdünnter Schwefel säure auf ein pH von 6,0 gestellt, filtriert, tropfen weise unter Rühren mit Methanol versetzt, bis sich die Lösung trübte, und die Temperatur der Mischung bei 50-55 C gehalten.
Zur trüben Lösung wurden einige Kristalle von Dihydrostreptomycinsulfat gege ben und die Lösung bei 50-55 C stehengelassen, worauf die Lösung in kurzer Zeit (etwa 5 Minuten) Kristalle abschied und gleichzeitig klar wurde. Es wurde weiterhin Methanol zugesetzt, bis die Lösung wieder schwach trübe wurde, und die Mischung wie oben stehengelassen. Das Verfahren wurde in, dieser Weise fortgesetzt, bis die Menge des Methanols 100 cm3 erreichte und die Lösung klar wurde.
Dann wurde die Temperatur der Lösung auf Raumtempe ratur herabgesetzt, die Kristalle abfiltriert und diese 5 Minuten lang in 50 %igem Methanol gerührt. Die Kristalle wurden abfiltriert, mit Methanol gewaschen und bei 50 C unter vermindertem Druck getrocknet.
Die Ausbeute betrug 8,9 g (738 mcg/mg als Dihydro- streptomycin). Mehrere Umkristallisationen .erhöhten die Wirksamkeit auf ungefähr 780 mcg/mg als Di- hydrostreptomycin. Es wurde bei dem obigen Prozess gefunden, dass M9S04 :die Wirkung besitzt, im Mate rial befindliche, dem Histamin ähnliche Substanzen zu zerstören.
<I>Beispiel 13</I> 5 cm3 des in Beispiel 10 b) erhaltenen Konzen trates der aktiven Verbindung (Wirksamkeit 368000 mcg/cm3) wurden mit Wasser bis .auf 50 cm3 ver dünnt und eine warme gesättigte Lösung von Methylorange zugesetzt, bis kein Niederschlag mehr gebildet wurde. Nach Kühlen wurde :der Niederschlag abfiltriert und wiederholt aus wässrigem Methanol umkristallisiert, worauf 2,1 g des Helianthates der aktiven Verbindung gewonnen wurden.
Eine Suspen sion des Helianthates in einer kleinen Menge Wasser wurde mit verdünnter Schwefelsäure auf ein pH von 2,0 :eingestellt, worauf sich die aktive Verbindung unter Bildung des Sulfates löste. Die Lösung wurde filtriert, um das ausgeschiedene Methylorange zu ent fernen, und mit einer kleinen Menge Aktivkohle be handelt. Zu der :erhaltenen farblosen Lösung wurde das Zehnfache ihres Volumens an wasserfreiem Aceton zugesetzt, worauf sich 0,6 g des Sulfates der aktiven Verbindung abschieden.
Method for producing dihydrostreptomycin Dihydrostreptomycin is a substance useful for medical treatment because it has a strong and broad antibacterial spectrum like streptomycin and is less toxic and more stable than the latter.
It is known that dihydrostreptomycin, which is similar to streptomycin in its antibacterial properties, has the advantage over the latter that it is less toxic to the nervous system. Up to now, dihydrostreptomycin has been obtained by hydrogenating the formyl radical of streptomycin and has been marketed as a sulfate or hydrochloride. These salts are colorless or white crystalline or powdery substances.
They are odorless or almost odorless, taste slightly bitter, and their aqueous solutions are levorotatory; an aqueous solution of the sulfate crystals shows a rotation of [a] 25 = -88 (c = 1). It is known that dihydrostreptomycin has a broad antibacterial spectrum against gram-positive, gramne: gative and acid-fast bacteria.
Up to now, dihydrostreptomycin has been produced by reducing the S. treptomycin isolated from the culture medium of Streptomyces griseus (see US Pat. No. 2498574). This method requires very pure streptomycin, otherwise a considerable loss in the reduction is inevitable.
No report has yet been published on the direct production of dihydrostreptomycin by culturing dihyd @ rostreptomycin-producing microorganisms.
It has now been found that dihydrostreptomycin is obtained in a simple manner while improving the yield and lowering the production costs if a strain of a dihydrostreptomycin-producing microorganism of the genus Streptomyces is cultivated aerobically in an aqueous nutrient medium.
The dihydrostreptomycin produced in this way can be isolated from the culture medium.
Any strain of any type belonging to the streptomycetes which provides dihydrostreptomycin can be used. So z. B. a new species called Streptomyces. humidus nov. sp. No. <B> 23572 </B> can be used for this purpose. This species has the characteristic properties listed in the table below.

Streptomyces humidus, on the other hand, produces dihydro-streptomycin.
The strain belonging to Streptomyces humidus mentioned above is hygroscopic, but some strains belonging to the same species do not have this property. Likewise, some do not form aerial mycelium and others produce pigment.
Dihydrostreptomycin-producing strains of other species can also be used for the same purpose if their properties are Streptomyces humidus nov. sp. are not very similar.
As is observed in microorganisms, especially in Streptorayceten, their behavior changes slightly spontaneously in the culture medium or they are artificially changed. This is why the identification of a species is so difficult that any mere description of the properties of a species has no definite meaning for the identification of that species. Mutants of dihydrostreptomycin-producing species, such as those described e.g. B. by treatment with X-rays, ultraviolet rays and Che mikalien are used.

The maltol reaction is a color reaction of streptomycin with phenol reagent,
which is generated by hydrolysis of streptomycin by means of alkali [Schenk et al., J. Amer. Chem. Soc. 67, 2277 (1945), Boxer, J. Biol. Chem. 169, 153 (1947), Eiseman et al., Anal. Chem. 21, 1507 (1949)]. These properties can advantageously be used to separate different strains. It can e.g.
B. the desired strains can be selectively separated by the dilution method or by cultivation in a medium containing dihydrostreptomycin.
As a carbon source, for. B. starch, lactose, sucrose, dextrin, glycerine and maltose are needed. As a nitrogen source, organic or inorganic nitrogen-containing substances can be used, such as. B. Soybean flour, meat extract, peptones, peanut powder; Casein, amino acids, yeast, bran, corn mash, cottonseed powder, nitrates, urea or ammonium compounds.
Small amounts of inorganic salts and growth-promoting substances can also be added to the nutrient solution. Other suitable sources of growth are mycelia of a strain belonging to Penicillium or e.g. B. its culture medium. Any nutrient medium suitable for the cultivation of Streptomyces griseus serves the same purpose. A suitable parent substance can be added if necessary.
The culture medium can be solid or liquid; submerged and aerobic cultivation is most advantageous for industrial purposes.
When Streptomyces humidus is used as the dihydro-streptomycin-producing microorganism and the cultivation is carried out under submerged aerobic conditions, it can preferably be carried out at a temperature of about 24 to 30 ° C. for a period of 3 to 8 days. But the temperature and the time period must be adapted to the other culture conditions. When using other dihydrostreptomycin-producing microorganisms, the most suitable conditions for their cultivation are selected in each case.
The dihydrostreptomycin produced in this way can be isolated in optimum purity from a nutrient medium containing dihydrostreptomycin with the aid of various physical or chemical methods. A high degree of purity for the intended use is achieved here.
To isolate the dihydrostreptomycin from the nutrient medium and separate from the impurities, you can z. For example, use can be made of the differences between dihydrostreptomycin or its salts and the impurities in terms of adsorbability, solubility, the distribution coefficient between two solvents and the strength with which ions are bound.
In the case of cultivation in liquid medium, the antibiotic can be concentrated in the culture medium up to a concentration of several thousand micrograms per em3; however, the dihydrostreptomycin can be isolated from a solution containing only 10 μ / cm 3.
In addition to dihydrostreptomycin, the substance obtained raw from the culture medium naturally contains various impurities such as carbohydrates, protein substances, salts, animal and vegetable substances, mycelia from microorganisms and their conversion products. Each of these impurities contains z. B. the following substances: Carbohydrates: starch, sugar, water-soluble carbohydrate.
Protein substances: amino acids, casein, meat extract, protein, peptones.
Animal and vegetable substances: protein substances, fats, fatty acids, lipoids, cellulose. Salts: ammonium salts and amines, salts of alkali metals, of calcium, magnesium, iron, manganese, copper, zinc. Transformation products of microorganisms: enzymes, protein substances, sugars, polyhydric alcohols, fatty acids.
In addition to these substances, compounds such as urea can be obtained by synthesis.
The composition of the impurities is very complex, but this varies in quality and quantity depending on the extent and type of raw materials used. The nutrient medium can also contain other impurities such as pH regulators, precipitants, adsorbents.
The dihydrostreptomycin can be separated from the mixture from the impurities mentioned in various ways.
One of these agents makes use of the difference between dihydrostreptomycin and the impurities in terms of solubility. In the case of liquid culture, a dihydrostreptomycin solution is obtained by filtering the fermentation mixture because it is preferably in the liquid part. But a smaller proportion of the active compound is found in the solid components and can be collected from it by extraction with hot water.
In this case, impurities in the protein series can be eliminated by adjusting the pH to the isoelectric point of the protein or by adding substances which precipitate the impurities before, after or with the simultaneous removal of the mycelium. If the pH of the fermentation mixture is high, the addition of an acidic substance is appropriate to make the protein easier to precipitate. If the concentration of dihydrostreptomycin in the solution is sufficiently high, the antibiotic can be precipitated by simply adding a suitable precipitant.
If the concentration of the dihydrostreptomycin is not sufficient, part of the solvent will be evaporated. As a precipitant z. B. phosphotungstic acid, alkyl or aralkylsulfonic acids, acidic azo compounds and polyhalophenol compounds are used. All of these form salts with dihydrostreptomycin.
Some other compounds can be used and most of them are acidic compounds with carboxyl groups. In some cases the salts of dihydrostreptomycin so formed can be double or complex salts. Also, as will be described later, ion exchangers can be used as a kind of precipitant. In order to accelerate the precipitation of the active substance or the impurities, one can also salt out, if necessary.
If solid cultures are used, the solid material is extracted with water, an aqueous solvent or another suitable solvent to obtain a solution of dihydro-streptomycin. If the fermentation mixture has previously been converted into a solid substance, this can be worked up as described above; in this case, however, the extraction is carried out at a suitable pH.
Another means for separating the dihydrostreptomycin consists in making use of the difference between the antibiotic active compound and the impurities in terms of adsorbability. Dihydro- streptomycin is z in the culture filtrate. B. adsorbed by adding a suitable adsorbent. If necessary, use can be made of adsorption chromatography. As an adsorbent z. B.
Activated charcoal, alum earth and silica gel are used. When the adsorbent is removed, most of the impurities remain in the liquid part. The adsorbent is then eluted with a suitable solvent, the non-elutable impurities remaining in the adsorbent. The elution is generally carried out with an acidic solvent. Chromatography results in a more precise separation, but this method requires more complicated procedures.
Differences between the antibiotic active compound and the impurities in terms of the partition coefficient between two solvents can also be used for the separation. As the solvent, water or an aqueous solvent and an organic solvent immiscible with the latter can be used. In this case the active compound tends to disperse in the aqueous solvent when the solvents are strongly acidic. If, on the other hand, the solvents are neutral or basic and a higher fatty acid, picric acid and the like are also present, the active compound is transferred to the organic solvent layer.
Accordingly, it is possible to take up the active compound in the material in an organic solvent and then transfer it to water or an aqueous solvent. As organic solvents, for. B. butanols, pentanols, phenols, lower fatty acid esters, methyl isobutyl ketone, diethyl ether and halogenated hydrocarbons, such as chloroform, carbon tetrachloride, are used.
This method can be used in the form of a partition chromatography or a countercurrent partition method. The separation can also be carried out in such a way that use is made of the differences between the antibiotically active compound and the impurities with regard to the strength of the ion-binding capacity. This is z. B. achieved by bringing a solution of the active compound with an ion exchanger in contact.
In this case, an anion exchanger adsorbs anionic impurities and leaves the active compound in the solution, while a cation exchanger adsorbs the antibiotic active compound. In the latter case, cationic impurities are also adsorbed, but the latter adsorption can, if a suitable ion exchanger is used,
kept as small as possible. The sulfonic acid type resins adsorb e.g. B. easily the impurities, but the carboxylic acid type resins have little tendency here for. The ion exchangers can be added using the mixing method; the use of a column for the resin is a suitable method in some cases. This method can be used for a dilute solution of the antibiotic active compound as for the culture filtrate.
Dialysis can also be used to remove the high molecular weight contaminants, but the process is complicated.
The above-mentioned agents can be used separately or in combination, depending on the quality of the material and the intended use of the product. One can also proceed in such a way that one z. B. the culture medium is initially filtered to remove the test components or the substances precipitated after cultivation ent.
The antibiotic active compound in the filtrate is adsorbed on a cation exchanger and then eluted with an acidic solvent. In this way, a concentrated solution of the active compound is achieved which contains only a small amount of impurities. If necessary, the solution is further concentrated and subjected to adsorption chromatography in order to remove almost all impurities. The antibiotic can be precipitated from the concentrated solution with a suitable agent.
The precipitate is then dissolved in a suitable organic solvent and an acid added, whereupon the corresponding acidic salt of dihydrostreptomycin is obtained. When the product is recrystallized, the acid salt is obtained in an almost pure state.
All work processes such as filtering, washing, heating, cooling, mixing, distilling, evaporating and drying can be carried out one after the other or at the same time.
In order to prevent the decomposition of the antibiotic active compound. It is advisable to work under conditions that are not extremely acidic or extremely alkaline. You can, of course, heat it if necessary; but it is preferable not to heat too long. In comparison with other antibiotics, dihydro-streptomycin is relatively stable in the culture medium, so that all manipulations can be carried out without difficulty.
The dihydrostreptomycin thus obtained was identified on the basis of the following data: <I> A. Physical and chemical properties 1. The infrared absorption spectrum was determined from the following derivatives of both dihydrostreptomycin available in accordance with the invention (hereinafter referred to as antibiotic 21 572) and commercial dihydrostreptomycin. I.
Sulfate, II. Streptidine picrate, <B> 111. </B> α-methyl-pentaacetyldihydrostreptobio @ aminide. The IR spectra of the derivatives of dihydrostreptomycin obtained according to the invention are identical to the IR spectra of the corresponding derivatives of dihydrostreptomycin commercially available.
All spectra are measured in nujol paraffin oil with a sodium chloride prism.
2. The ultraviolet spectra of the sulfate of antibiotic 23 572 and those of a commercial sample of dihydrostreptomycin sulfate agree well, no specific adsorption was observed.
3. The analytical values of the sulfate of the antibiotic 23 572 agree well with the theoretical values of the dihydrostreptomycin sulfate. The values for dihydrostreptomycin sulfate (C21H41012N7) 2-3 H2S04) are: Calculated: C 34.42%; H 6.0511 / G; N 13.3804; S 6,561 / 0 Found:

The specific rotation of dihydrostreptomycin was determined by I. A. Solomons et al. in Science 109, 515 (1949) with (a) D-88 and by F. J. Wolf et al. in J. Am. Chem. Soc. 68, 2163 (1946) given (a) D - 88.5.
5. The sulfate of antibiotic 23 572 melts with decomposition and blackening at 250 to 255 C, the dihydrostreptomycin sulfate also melts with decomposition and black coloring at 250 to 255 C.
6. Both the antibiotic 23 572 and the dihydrostreptomycin behave negatively towards the maltol reaction, streptomycin reacts against it.

After standing for 48 hours at room temperature, 200 cm3 of ether were added to the solution with stirring and the white crystals (streptidine hydrochloride) formed were separated off by centrifugation. The crystals were dissolved in 20 cm3 of water and a saturated picric acid solution was added.
After standing for several hours, the precipitated crystals were separated off and recrystallized from water. 400 mg of streptidine picrate were obtained as yellowish needles with a melting point of 271-273 C.
Analysis for C8H1 $ O4N6-2 C6HH3N307 showed: Calculated: C, 33,340 / 9; H 3.360 / 0; N 23.33% Found: C 33.46%; H 3.440; 9;
N 23.49% The infrared absorption spectrum of Picrates agreed with that of Picrates made from dihydrostreptomycin as described under 1.
The liquid above the above streptidine hydrochloride was neutralized with a solution of sodium hydroxide in methyl alcohol, the sodium chloride formed was separated off by centrifugation and the solvent was distilled off under reduced pressure. A white powder remained. This residue was dissolved in 20 cm 3 of pyridine and 7 cm 3 of acid anhydride was added.
After standing overnight, water was added to the reaction mixture and the solvent was distilled off. The residue was recrystallized from methanol. It was obtained in the form of white needles with a melting point of 194.5 C α-methyl-pentaacetyldihydrostreptobiosaminide. The yield was 700 mg.
The analysis for C13H19NO8 (CH.C0) 5 (OCH3) resulted in: Calculated: C 51.15% -; H 6.62%; N 2.490 / 0 Found: C 51.09% -; H 6, 13-0 / 9;
N 2.40%. The rotation (a) = -120.0 (c = 1%, D in chloroform).
The infrared absorption spectrum of the compound obtained was in good agreement with the α-methyl-pentaacetyldihydrostreptobiosaminide obtained from commercial dihydrostreptomycin, as already described under 1.
The ss-methyl-pentaacetyldihydrostreptobiosaminide was obtained from antibiotic 23 572 by the method of N. G. Brink [J. At the. Chem. Soc. <B> 68.2557 </B> (1946)] in the following way:
10 g of the hydrochloride of the antibiotic 23 572 were dissolved in 500 cm3 of methanol containing 1% hydrogen chloride. After the solution had stood at room temperature overnight, the solvent was distilled off under reduced pressure. The residue was dissolved in 750 cm3 of methanol and 450 cm3 of ether was added.
The solution was passed through a column which was charged with 210 g of acid-washed alum earth which had been impregnated with a methanol-ether mixture in a ratio of 2: 1. The column was washed with 1000 cm3 of a methanol-ether mixture in the ratio <B> 3: 2 </B>. The flowed out solution was evaporated to dryness under reduced pressure.
The residue was acetylated by allowing it to stand overnight with 10 cm3 of acetic anhydride and 10 cm3 of pyridine at room temperature. Water was then added and the solution was evaporated to dryness in vacuo. The residue was dissolved in chloroform and the chloroform solution was washed with water, dilute sulfuric acid and finally with water.
After the chloroform had distilled off, the brownish solid residue was boiled with 100 cm3 ether for 2 minutes and the ether solution was separated off by decanting. The same procedure was repeated again. The ether-insoluble fraction was crystallized from methanol and yields a-methyl-pentaacetyldihydrostreptobiosaminide with a melting point of 194.5 C.
The ether solution was concentrated to approximately 50 cm3 and petroleum ether was added, giving white crystals. Crystallization from methanol yielded approximately 100 mg of ss-methylpentaacetyldihydrostreptobiosaminide with a melting point of 155-156 C. No melting point depression was observed when it was mixed with the ss-isomer prepared from dihydrostreptomycin by the method just given was.
The analysis gave for C "H19NOs (CH.CO) 5 (OCH3): C 50.710 / 0; H 6.620 / a; N 2.6511 / o. The calculated values are: C 51.15%; H 6.620 / 0; N 2.4911 / o. (A) = -36. (C = 1 / chloroform).
D The infrared spectrum of the product obtained was in good agreement with that of the β-methyl-pentaacetyldihydrobiosaminide obtained from dihydro-streptomycin.
A solution of 260 mg of a-methyl-pentaacetyl-dihydrostreptobiosaminide, which was obtained from the antibiotic <B> 23572 </B>, in 20 cm3 of 10 N / od hydrochloric acid was refluxed for 3 hours. After cooling, the brownish solution was decolorized with charcoal and evaporated to dryness in vacuo.
The residue was acetylated at room temperature with 1 cm 3 of acetic anhydride and 3 cm 3 of pyridine. After adding water, the solution was evaporated to dryness. The residue was dissolved in 50 cm3 of a benzene-petroleum ether mixture in a ratio of 7: 3.
The solution was passed through a column charged with 4 g of an acid washed alum earth impregnated with petroleum ether, the acetylation product being adsorbed on the acid washed alum earth.
The column was treated with 100 cm3 of a 7: 3 benzene / chloroform mixture to elute the substance in question. The effluan was concentrated to approximately 10 cm3 and 30 cm3 of ether were added, which resulted in crystals - Recrystallization from chloroform-ether gave 25 mg of needles of pentaacetyl-N-methyl-L-glucosamine with a melting point of 158-159 C.
Analysis showed: C 50.71%; H 6.14 11 / a; N 3.25 0/0. The calculated values for C17H "N010 are:
C 50.62%; H 6.250 / 0; N 3.4711 / o. (a) D = -102 (c = 0.7% in chloroform). The infrared absorption spectrum of the product obtained was in complete agreement with that of pentaacetyl-N-methyl-L-glucosamine,
which was obtained from dihydrostreptomycin by the method described. No melting point depression was observed when mixed with pentaacetyl-N-methyl-L-glucosamine obtained from dihydro-streptomycin.
13. Antibiotic 23 572 was hydrolyzed with alkali under conditions similar to those used for the hydrolysis of hydroxystreptomycin and described by F. H. Stodola et al. [J. At the. Chem. Soc. 73, 2290 (1951)]. Accordingly, a solution of 2 g of the hydrochloride of the antibiotic 23 572 in 40 cm 3 of 11 NaOH was heated to 100 ° C. for 3 hours. The reaction mixture was acidified with hydrochloric acid and worked up.
However, neither a substance with a positive reaction to maltot nor a substance which can be extracted under acidic conditions could be found, i.e. the antibiotic 23 572 behaves differently than streptomycin and hydroxystreptomycin in this test. The same test gave the same result with dihydrostreptomycin.
14. A warm solution of methyl orange was added to an aqueous solution of 50,000 y / em3 of the antibiotic 23 572 until no more precipitate was formed. The precipitate was filtered off and recrystallized from methanol-water (1: 3). The helianthat was obtained in the form of flakes with a melting point of 222-225 C (potency: 340 streptomycin units per mg).
The analysis for dihydrostreptomycin heliantate (C03H $ 00., 1N10s3) gave: Calculated: C 50.46%; H 5.7911 / o; N 14.95%; S 6.4011 / o Found:
C 50.19%; H 5.93%; N 14.93%; S 6.801 / 0 50.84114; 5.95% 15. The paper distribution chromatography of streptomycin and dihydrostreptomycin was published by F. H. Stodola et al. [J. At the. Soc. 73, 2290 (1951)].
Butanol, saturated with water, with 2 / o p-toluenesulfonic acid and 2 / o piperidine was used as solvent. Both compounds were differentiated with the help of the bioautogram.
Paper chromatography was applied to the above two compounds and the antibiotic 23 572 by the same method. The bio-autographs were tested with B. subtilis as test microorganisms. It was found that the antibiotic showed a growth-preventing zone in the same place as the dihydro-streptomycin.
