Verfahren zur Herstellung neuer Steroide Gegenstand der Erfindung ist ein Verfahren zur Herstellung von Salzen von Steroiden, die in 21-Stel- lung eine veresterte Oxygruppe und in 20-Stellung eine Oxygruppe und einen gegebenenfalls substituier ten Piperidylrest besitzen.
Diese neuartig substituierten Steroide gehören der Allopregnan- oder Pregnanreihe an, wobei es sich auch um Homo- und/oder Nor-Verbindungen und beliebig ungesättigte Verbindungen dieser Reihen handeln kann, wie z. B. Verbindungen, die ausgehend vom Kohlenstoffatom 5 eine Doppelbin dung aufweisen.
Die neuen Verbindungen können beliebig substi tuiert sein, z. B. besonders im Steroidteil, durch freie oder funktionell abgewandelte Oxy- oder Oxo- gruppen oder Halogenatome und/oder gegebenen falls am Piperidylstickstoffatom durch Alkyl-, Acyl- oder Aralkylreste. Eine funktionell abgewandelte Oxygruppe ist insbesondere eine mit einer aliphati- sehen, alicyclischen,
araliphatischen, aromatischen oder heterocyclischen Carbonsäure, Thioncarbon- säure, Thiolcarbonsäure oder Sulfonsäure veresterte Oxygruppe. Als Säuren seien beispielsweise genannt:
Essigsäure, Chloressigsäure, Trifluoressigsäure, Pro- pionsäure, Buttersäuren, Valeriansäuren, Trimethyl- essigsäure, DiäthyIessigsäure, Capronsäuren, Önanth- säuren, Caprinsäuren, Palmitinsäure, Crotonsäure, Undecansäure, Undecylen.saure, Oxalsäure, Bern steinsäure,
Pimelinsäure, Maleinsäure, Milchsäure, Carbaminsäuren, ss-Cyclopentylpropionsäure, Benzoe- säure, Phenylessigsäure, Cyclohexylessigsäure, Fu- ran-2-carbonsäure,
Methansulfonsäure und Toluol- sulfonsäuren. Auch die Oxygruppen in 20- und 21- Stellung können frei oder funktionell abgewandelt, z. B. mit den oben genannten Säuren verestert sein.
N-Substituenten sind besonders niedere Alkyl- reste oder die Acylreste niederer Fettsäuren, Benzoe- Säuren oder einfacher Arylfettsäuren, insbesondere der Methyl- oder Acetylrest.
Die neuen Verbindungen besitzen wertvolle pharmakologische Eigenschaften. So sind sie Herz wirksam und können demgemäss als Arzneimittel Verwendung finden. Gleichzeitig stellen sie wertvolle Zwischenprodukte für die Herstellung von Arznei mitteln dar.
Besonders wertvoll sind Pregnan und Allo- pregnanverbindungen der angegebenen Art, die in 3-Stellung eine freie oder funktionell abgewandelte Oxy- oder Oxogruppe aufweisen und im Ring A ge sättigt sind oder ausgehend vom Kohlenstoffatom 5 eine Doppelbindung aufweisen. Der Piperidylrest ist dabei vorzugsweise in 2-Stellung mit dem Steroid gerüst verknüpft.
Das erfindungsgemässe Verfahren ist dadurch ge kennzeichnet, dass man entsprechende 20-Oxy- steroide mit einer freien oder veresterten Oxygruppe in 21-Stellung, gegebenenfalls unter vorübergehen dem Schutze weiterer in der Ausgangsverbindung vorhandener freier Oxogruppen, mit Pyridyl-metall- Verbindungen zu den entsprechenden 20-Oxy-20- pyridyl-steroiden umsetzt,
eine in 21-Stellung vor handene freie Hydroxylgruppe verestert, die so er haltenen 21-Acyloxy-20-oxo-20-pyridyl-steroide mit Pyridinring reduzierenden Mitteln behandelt und die erhaltenen 21 Acyloxy-20-oxy-20-piperid'yl-steroide in ihre Salze überführt.
Die verfahrensgemässe Umsetzung mit der Pyri- dyl-Metallverbindung, besonders Pyridyl-(2)-lithium kann in üblicher Weise, z. B. in einem inerten Ver dünnungsmittel, vorgenommen, und das Reaktions gemisch wie üblich, z. B. in saurem Medium, auf gearbeitet werden. Sind im Molekül Acyloxygruppen vorhanden, so werden diese dabei zu freien. Oxy- gruppen gespalten.
Sofern weitere freie Oxo-gruppen in der Ausgangsverbindung vorhanden sind, müssen diese von der Umsetzung mit der Pyridyl-Metall- Verbindung durch Überführung in ein Ketal- oder Enol-Derivat geschützt werden.
Als den Pyridinring reduzierendes Mittel ist in erster Linie Wasserstoff in Gegenwart eines Kata- lysators, vorzugsweise eines Edelmetallkatalysators, wie Platin, oder auch Nickel oder Kupferchromit zu nennen, sowie naszierender Wasserstoff, wie er z. B. durch Behandlung von Natrium mit einem Alkohol, z. B. Butanol oder Lithium mit flüssigem Ammo niak, entsteht.
Handelt es sich bei der einer solchen Reduktion unterworfenen Verbindung um ein qua- ternäres Ammonium-, wie Alkylammonium-derivat, so wird eine tertiäre Piperidylverbindung erhalten. In einer durch einen Benzylrest quaternisierten Ver bindung wird dieser jedoch abgespalten. Je nach der Wahl der Reduktionsbedingungen werden im Steroidmolekül vorhandene Doppelbindungen ab gesättigt.
Die gewünschtenfalls auszuführende Verseifung von in den Endstoffen vorhandenen Acyloxygruppen wird in an sich bekannter Weise ausgeführt.
Die freien Basen lassen sich in üblicher Weise in ihre Salze umwandeln, während man aus letzteren in an sich bekannter Weise die Basen freisetzen kann. Als Salze kommen solche anorganischer oder organischer Säuren in Betracht, wie z. B. der Halo genwasserstoffsäuren, Schwefelsäure, Salpetersäure, Phosphorsäure, Rhodanwasserstoffsäure, Essigsäure, Propionsäure, Oxalsäure, Malonsäure, Bernstein säure, Äpfelsäure, Methansulfonsäure, Äthansulfon- säure, Oxyäthansulfonsäure,
Benzol- oder Toluol- sulfonsäure.
Die Ausgangsstoffe sind bekannt oder können nach an sich bekannten Verfahren gewonnen werden. Im folgenden Beispiel sind die Temperaturen in Celsiusgraden angegeben.
<I>Beispiel</I> Zu einer Pyridyl-LithiumLösung aus 2,1g Li- thium, 200 ml Äther, 24 g Brombenzol und 24 g 2-Brompyridin in 100 ml Benzol gibt man innert 30 Minuten bei -30 bis -40 eine Lösung von 10,4 g d5-3ss,21 Diacetoxy-pregnen-20-on und rührt anschliessend 3 Stunden bei -40 weiter. Nach Zugabe von 2n Salzsäure und gegebenenfalls zwecks Lösung Methanol fällt man die Base aus der sauren Lösung mit wässrigem Ammoniak.
Das so erhaltene 45-3ss,20,21- Trihydroxy - 20 - pyridyl - (2') - pregnen wird mittels 50 ml Pyridin und 15 ml Acetanhydrid in üblicher Weise acetyliert, wobei das d5-3ss,21- Diacetoxy-20-hydroxy-20-pyridyl-(2')-pregnen ent- steht. Aus Methanol kristallisiert es in Blättchen vom F. = l59-160 ; [ ]D = -46 - 4 (c = 0,888 in Chloroform).
4,95 g 45-3ss,21-Diacetoxy-20-hydroxy-20-pyri- dyl-(2')-pregnen werden in 80 ml Eisessig in Gegen wart von 300 mg Platinoxyd hydriert. Nach der Aufnahme von 900 ml Wasserstoff filtriert man vom Katalysator ab, dampft das Filtrat im Vakuum ein und isoliert durch Zugabe von wässrigem Ammoniak 3ss,21-Diacetoxy- 20-. hydroxy-20-piperidyl-(2')-allo- pregnan. Die Base wird direkt in das Hydrochlorid übergeführt; F. = über 260 .

Methanesulphonic acid and toluenesulphonic acids. The oxy groups in the 20- and 21-position can be freely or functionally modified, e.g. B. be esterified with the above acids.
N-substituents are particularly lower alkyl radicals or the acyl radicals of lower fatty acids, benzoic acids or simple aryl fatty acids, especially the methyl or acetyl radical.
The new compounds have valuable pharmacological properties. They are heart-active and can therefore be used as medicinal products. At the same time, they represent valuable intermediate products for the manufacture of pharmaceuticals.
Pregnane and allo-pregnane compounds of the specified type which have a free or functionally modified oxy or oxo group in the 3-position and are saturated in ring A or have a double bond starting from carbon atom 5 are particularly valuable. The piperidyl radical is preferably linked to the steroid structure in the 2-position.
The method according to the invention is characterized in that corresponding 20-oxy steroids with a free or esterified oxy group in the 21-position, optionally with temporary protection of further free oxo groups present in the starting compound, with pyridyl metal compounds to give the corresponding 20 -Oxy-20- pyridyl steroids converts,

The process according to the reaction with the pyridyl metal compound, especially pyridyl (2) lithium can be carried out in a conventional manner, for. If further free oxo groups are present in the starting compound, they must be protected from the reaction with the pyridyl metal compound by converting them into a ketal or enol derivative.
The agent that reduces the pyridine ring is primarily hydrogen in the presence of a catalyst, preferably a noble metal catalyst, such as platinum, or else nickel or copper chromite, and nascent hydrogen, such as is used, for example, in US Pat. By treating sodium with an alcohol, e.g. B. butanol or lithium with liquid ammo niac arises.
If the compound subjected to such a reduction is a quaternary ammonium, such as an alkylammonium derivative, a tertiary piperidyl compound is obtained. In a connection quaternized by a benzyl radical, however, this is split off. Depending on the selection of the reduction conditions, existing double bonds in the steroid molecule are saturated.
The saponification, if desired, of acyloxy groups present in the end products is carried out in a manner known per se.
The free bases can be converted into their salts in the usual way, while the bases can be released from the latter in a manner known per se. Suitable salts are those inorganic or organic acids, such as. B. of hydrogen halide acids, sulfuric acid, nitric acid, phosphoric acid, hydrofluoric acid, acetic acid, propionic acid, oxalic acid, malonic acid, succinic acid, malic acid, methanesulfonic acid, ethanesulfonic acid, oxyethanesulfonic acid,
Benzene or toluenesulfonic acid.
The starting materials are known or can be obtained by processes known per se. In the following example the temperatures are given in degrees Celsius.
<I> Example </I> A pyridyl-lithium solution of 2.1 g of lithium, 200 ml of ether, 24 g of bromobenzene and 24 g of 2-bromopyridine in 100 ml of benzene is added within 30 minutes at -30 to -40 Solution of 10.4 g of d5-3ss, 21 diacetoxy-pregnen-20-one and then stirred for a further 3 hours at -40. After adding 2N hydrochloric acid and, if necessary, methanol to dissolve it, the base is precipitated from the acidic solution with aqueous ammonia.
The 45-3ss, 20,21-trihydroxy-20-pyridyl- (2 ') -pregnen thus obtained is acetylated in the usual way using 50 ml of pyridine and 15 ml of acetic anhydride, the d5-3ss, 21-diacetoxy-20-hydroxy -20-pyridyl- (2 ') - pregnen arises. 4.95 g of 45-3ss, 21-diacetoxy-20-hydroxy-20-pyridyl- (2 ') - pregnen are hydrogenated in 80 ml of glacial acetic acid in the presence of 300 mg of platinum oxide. After 900 ml of hydrogen have been taken up, the catalyst is filtered off, the filtrate is evaporated in vacuo and isolated by adding aqueous ammonia 3ss, 21-diacetoxy-20-. hydroxy-20-piperidyl- (2 ') -allo-pregnane. The base is converted directly into the hydrochloride; F. = over 260.
