Die neuen Verbindungen, nämlich das 3-Cyclo- pentylmethyl- und das 3-Cyclohexylmethyl-6-chlor-7- sulfamyl- 3,4 - dihydro -1,2,4 - benzothiadiazin -1,1- di- oxyd, und deren Salze zeigen eine hohe diuretische und natriuretische Wirksamkeit und sollen als Heil mittel Verwendung finden, besonders in Form von pharmazeutischen Präparaten.
Die neuen Verbindungen werden erhalten, wenn man das Nitrobenzol der Formel

mit einem Aldehyd der Formel R-CH.-CHO, worin R die oben genannte Bedeutung besitzt, um setzt und reduziert, wobei intermediär die Nitro- gruppe zu einer Aminogruppe reduziert wird. Die Reaktion wird vorzugsweise in Gegenwart eines Lö sungsmittels, besonders eines mit Wasser mischbaren Lösungsmittels, wie eines Alkanols, z. B. Methanol oder Athanol, eines Äthers, z.
B. Athylenglykol-di- methyläther, Tetrahydrofuran, eines Formamids, wie Formamid selbst oder N,N-Diäthylformamid, eines Alkanons, wie Aceton, Wasser oder Mischungen, be sonders wässrige Mischungen, mit den genannten Lö sungsmitteln durchführt.
Der Aldehyd kann als solcher oder in Form eines diesen Aldehyd abgebenden Mittels, z. B. als Poly meres oder als reaktionsfähiges funktionelles Deri vat, wie eines Acetals mit einem Alkanol, z. B. 1,1- Dimethoxy-2-cyclopentyl- oder -cyclohexyl-äthan, oder anderer funktioneller Derivate der diesen zu grunde liegenden 1,1-Dioxy-2-cycloalkyl-äthane ver wendet werden. Solche Derivate werden vorzugsweise verwendet, wenn die Reduktion chemisch in Gegen wart einer Säure, wie einer starken anorganischen Säure, z. B. einer Mineralsäure, wie Salzsäure oder Schwefelsäure, durchgeführt wird.
Man kann aber auch den Aldehyd in Lösung zur Reaktionsmischung zugeben, wobei als Lösungsmittel eines der vorge nannten organischen Lösungsmittel oder Wasser ver wendet werden kann. Man verwendet die Reaktions teilnehmer, das heisst das Sulfamyl-nitrobenzolderi- vat und den Aldehyd oder eines seiner Derivate, vor zugsweise in ungefähr äquivalenten Mengen. Man kann zur Vervollständigung der Reaktion auch einen überschuss an Aldehyd anwenden.
Die Reduktion lässt sich vorzugsweise durch Be handeln mit katalytisch aktiviertem Wasserstoff vor nehmen. Als Katalysator kommt dabei ein Metall der B. Gruppe des periodischen Systems, wie Nickel, z. B. Raney-Nickel, oder ein anderer äquivalenter Metallkatalysator zur Anwendung. Die Reaktion wird vorzugsweise unter atmosphärischem Druck, sofern notwendig, auch unter höheren Drucken, durch geführt. Auch kann man kleine Mengen alkalische Reagenzien, wie Alkalimetallhydroxyde, z. B. Na triumhydroxyd, Kaliumhydroxyd, beigeben, wobei allerdings der Aldehyd vorzugsweise als solcher zu gegeben wird. Die Reaktion lässt sich bei Raumtem peratur oder bei erhöhter Temperatur durchführen.
Die Reduktion kann auch mittels eines Metalls in Gegenwart einer Säure, wie z. B. Zink, Zinn oder Eisen, in Gegenwart einer Mineralsäure, z. B. Salz säure, Bromwasserstoffsäure oder Schwefelsäure, durchgeführt werden. Je nach den gewählten Bedin gungen und den gewählten Metallen kann dabei die Reaktion unter Kühlen oder Erhitzen durchgeführt werden.
Der zu diesem Verfahren verwendete Ausgangs stoff lässt sich erhalten, wenn man ein 2,4-Dihalogen- 5-chlor-nitrobenzol mit einem Mercaptan der For mel Ri-SH, worin R1 einen aliphatischen oder arali- phatischen Kohlenwasserstoffrest, z.
B. Methyl, Äthyl oder besonders Benzyl, darstellt, oder einem Halo genid der Formel Rl Hal in Gegenwart von Thio- harnstoff umsetzt, die erhaltenen 2,4-Di-Ri mercapto- 5-chlor-nitrobenzole zu den 2,4-Sulfonsäuren oxy diert, z. B. mit Wasserstoffperoxyd, und mit einem Halogenierungsmittel, z. B. Phosphorpentachlorid, in die 5 - Chlor - nitrobenzol - 2,4 - disulfonylhalogenide überführt.
Oxydiert man die 2,4-R,- Mercapto-5- chlor-nitrobenzole mit einem Halogen oder in Gegen wart eines Halogens, gelangt man direkt zu den Di- sulfohalogeniden. Letztere können dann durch Be handeln mit Ammoniak in die Di-sulfamylverbindung übergeführt werden.
Je nach den Reaktionsbedingungen erhält man die neuen Verbindungen in freier Form oder in Form ihrer Salze. Erhaltene Metallsalze können zum Bei spiel durch Reaktion mit wässrigen sauren Mitteln, wie Mineralsäure, z. B. Halogenwasserstoffsäure, bei spielsweise Salzsäure oder Schwefelsäure, in die freien Verbindungen übergeführt werden. Diese wiederum lassen sich in die Metallsalze, wie Alkalimetallsalze, überführen durch Behandeln zum Beispiel mit einem Metallhydroxyd, wie Natrium- oder Kaliumhydroxyd, in einem Lösungsmittel, wie einem Alkanol, z. B.
Methanol oder Äthanol, oder in Wasser und an schliessendem Abdampfen des Lösungsmittels. Dabei können Mono- oder Polysalze gebildet werden.
Im nachfolgenden Beispiel sind die Temperaturen in Celsiusgraden angegeben.
<I>Beispiel</I> Zu einer Suspension von 4,0g 5-Chlor-2,4-disulf- amyl-nitrobenzol in 10 cm3 95 /o igem Äthanol und 1,5 cm3 Wasser, welches 0,55g Natriumhydroxyd gelöst enthält, gibt man 2,75g Cyclopentyl-acetalde- hyd-natriumbisulfitverbindung und rührt die Mi- schung mehrere Stunden.
Man fügt 1 g Eisenpulver und 2,3 cm3 konzentrierte Salzsäure zu, erhitzt die Mischung 2 Stunden am Rückflusskühler, wobei man noch weitere 2 g Eisenpulver zugibt. Nach dem Zu fügen von weiterem wässrigem Äthanol filtriert man die warme Mischung. Nach dem Zugeben von Was ser kristallisiert das 3-Cyclopentylmethyl-6-chlor-7- sulfamyl- 3,4- dihydro -1,2,4- benzothiadiazin-1,1- di- oxyd aus.
Es schmilzt nach dem Umkristallisieren aus wässrigem Äthanol bei 225 .
In analoger Weise, ausgehend von einer entspre chenden Menge Cyclohexyl - acetaldehyd - natrium- bisulfitverbindung, erhält man das 3-Cyclohexyl- methyl- 6 - chlor- 7-sulfamyl-3,4-dihydro-1,2,4-benzo- thiadiazin-1,1-dioxyd, welches nach dem Umkristal- lisieren aus Äthanol, das wenig Wasser enthält, bei 224-226 schmilzt.

The reaction is preferably carried out in the presence of a solvent, especially a water-miscible solvent such as an alkanol, e.g. B. methanol or ethanol, an ether, e.g.
B. ethylene glycol dimethyl ether, tetrahydrofuran, a formamide such as formamide itself or N, N-diethylformamide, an alkanone such as acetone, water or mixtures, especially aqueous mixtures, with the solvents mentioned.
The aldehyde can be used as such or in the form of an aldehyde releasing agent, e.g. B. as poly meres or as a reactive functional Deri vat, such as an acetal with an alkanol, z. B. 1,1-dimethoxy-2-cyclopentyl or -cyclohexyl-ethane, or other functional derivatives of these underlying 1,1-dioxy-2-cycloalkyl-ethane can be used ver. Such derivatives are preferably used when the reduction is performed chemically in the presence of an acid such as a strong inorganic acid, e.g. B. a mineral acid such as hydrochloric acid or sulfuric acid is carried out.

If the 2,4-R, - mercapto-5-chloro-nitrobenzenes are oxidized with a halogen or in the presence of a halogen, the disulfohalides are obtained directly. The latter can then be converted into the disulfamyl compound by treating with ammonia.
Depending on the reaction conditions, the new compounds are obtained in free form or in the form of their salts. Metal salts obtained can, for example, by reaction with aqueous acidic agents, such as mineral acid, e.g. B. hydrohalic acid, for example hydrochloric acid or sulfuric acid, are converted into the free compounds. <I> Example </I> To a suspension of 4.0 g of 5-chloro-2,4-disulphamyl-nitrobenzene in 10 cm3 of 95% ethanol and 1.5 cm3 of water which contains 0.55 g of dissolved sodium hydroxide , 2.75 g of cyclopentyl acetaldehyde sodium bisulfite compound are added and the mixture is stirred for several hours.
1 g of iron powder and 2.3 cm3 of concentrated hydrochloric acid are added, the mixture is heated for 2 hours on the reflux condenser, a further 2 g of iron powder being added. After adding more aqueous ethanol, the warm mixture is filtered. After adding water, the 3-cyclopentylmethyl-6-chloro-7-sulfamyl-3,4-dihydro-1,2,4-benzothiadiazine-1,1-dioxide crystallizes out.
It melts at 225 after recrystallization from aqueous ethanol.
In an analogous manner, starting from a corresponding amount of cyclohexyl-acetaldehyde-sodium bisulfite compound, 3-cyclohexylmethyl-6-chloro-7-sulfamyl-3,4-dihydro-1,2,4-benzothiadiazine is obtained -1,1-dioxide, which after recrystallizing from ethanol, which contains little water, melts at 224-226.
