Verfahren zur Herstellung neuer Antituberkulotica
Es ist bekannt, dass Isonikotinsäurehydrazid, im folgenden INH genannt, antituberkulotische Wirkung besitzt. Seine Nützlichkeit wird aber etwas eingeschränkt durch die Tatsache, dass der Tuberkelbazillus rasch gegen INH widerstandsfähig wird, so dass die Verwendung dieser Substanz in der Therapie beschränkt ist. Auch ist INH in grossen Dosen merklich toxisch.
Es wurde nun gefunden, dass die neuen Stoffe, die man erhält, wenn man INH mit einem Chinolinaldehyd der Formel

worin R ein Wasserstoffatom, eine Alkyl- oder eine Aralkylgruppe bedeutet, umsetzt, antituberkulotische Wirkung zeigen und im besonderen auch gegen INHresistente Stämme des Tuberkelbazillus wirksam sind. Zudem sind die neuen Stoffe bei den anzuwendenden Dosen weniger toxisch als INH.
Gegenstand des vorliegenden Patentes ist nun ein Verfahren zur Herstellung dieser neuen Stoffe der Formel

das dadurch gekennzeichnet ist, dass man einen Chinolinaldehyd der Formel

mit INH umsetzt.
Die Aldehydgruppe kann an jeder beliebigen Stelle des Chinolinkerns sitzen, doch zieht man im allgemeinen 5- oder 2-Stellung vor. Von den neuen Stoffen sind besonders wertvoll das Isonikotinsäurehydrazon des 8-Hydroxy-chinolin-5-aldehyds, welches einen Schmelzpunkt von 239-241"C hat, das Isonikotinsäurehydrazon des 8-Hydroxy-chinolin-2aldehyds, welches einen Schmelzpunkt von 225 bis 2270 C hat, und das Isonikotinsäurehydrazon des 8-Benzyloxychinolin-2-aldehyds, welches sich bei 180-182"C zersetzt.
Die Umsetzung des Chinolinaldehyds mit INH wird zweckmässig in einem Lösungsmittel, z. B.
Alkohol, durchgeführt, vorzugsweise unter Erhitzen am Rückfluss. Die Dauer des Erhitzens hängt vom verwendeten Aldehyd ab, liegt aber im allgemeinen in der Grössenordnung 2-8 Stunden.
Die gebildeten Hydrazone können aus dem Reaktionsgemisch beispielsweise isoliert werden, indem man einen Teil des Lösungsmittels verdampft und das Gemisch auf Oo C abkühlt, wobei die Hydrazone ausfallen; sie können dann leicht entfernt werden, z. B. durch Filtration.
Die neuen Hydrazone bilden sowohl mit Basen, beispielsweise Alkalihydroxyden, als auch mit Mineralsäuren, beispielsweise Salzsäure, Salze. Andere nichttoxische Salze, welche für eine therapeutische Verwendung in Frage kommen, sind u. a. die Salze mit organischen Säuren, z. B. p-Amino-salicylsäure und Glutaminsäure. Es ist bekannt, dass die p-Aminosalicylsäure selbst schon antituberkulotische Wirkung besitzt, so dass die p-Amino-salicylsäure-Salze der Hydrazone bezüglich ihrer antituberkulotischen Wirkung besonders interessant sind.
Die Salze der Hydrazone mit p-Amino-salicylsäure und Glutaminsäure können beispielsweise leicht so hergestellt werden, dass man die freie Säure oder ihr Natriumsalz in wässriger Lösung mit einer Lösung des Hydrazons, z. B. in Salzsäure, umsetzt und das gebildete Salz des Hydrazons durch Zugabe von Alkali ausfällt.
Beispiel I
17,3 Gewichtsteile 8-Hydroxy-chinolin-5-aldehyd und 13,7 Gewichtsteile INH werden zusammen in 800 Volumteilen Alkohol gelöst und die Lösung 4-5 Stunden am Rückfluss gekocht. Dann wird sie auf etwa die Hälfte ihres Volumens eingeengt und auf O"C abgekühlt; dabei fällt das Hydrazon aus und kann am Vakuum abfiltriert und getrocknet werden. Weitere Mengen Hydrazon können durch weitere Konzentrierung der Mutterlauge erhalten werden. Die Gesamtausbeute an Hydrazon beträgt bis ungefähr 700/o.
Das erhaltene Isonikotinsäurehydrazon des 8-Hydroxy-chinolin-5-aldehyd kann aus Alkohol oder einem grossen Volumen Wasser umkristallisiert werden. Es ist löslich in verdünnter Salzsäure oder verdünnter Natronlauge. Das reine Hydrazon schmilzt bei 2392410 C.
Die so erhaltene Verbindung kann wie folgt in Salze übergeführt werden: a) 2,9 Gewichtsteile des Hydrazons werden unter Erhitzen in 50 Volumteilen in Salzsäure gelöst und zu einer Lösung von 2,1 Gewichtsteilen Natrium-pamino-salicylat in 40 Volumteilen Wasser gegeben.
Beim Neutralisieren des Reaktionsgemisches mit 40 Volumteilen in Natriumhydroxyd fällt das p-Aminosalicylat des Hydrazons als braunoranger Niederschlag aus. Dieses Salz zersetzt sich bei 2802850 C und ist in Wasser sehr schlecht löslich. b) 3 Gewichtsteile des Hydrazons werden in 50 Volumteilen in Salzsäure gelöst und ! zu einer Lö- sung von 1,5 Gewichtsteilen l (+)-Glutaminsäure in
10 Volumteilen ln Natriumhydroxyd gegeben. Bei der Zugabe von weiteren 40 Volumteilen in Na triumhydroxyd fällt das l (+)-Glutaminat als gelbbrauner Niederschlag aus. Es ist in Wasser sehr schlecht löslich und zersetzt sich bei 2752800 C.
Beispiel 2
8,7 Gewichtsteile 8-Hydroxy-chinolin-2-aldehyd und 7 Gewichtsteile INH werden in 100 Volumteilen Alkohol gelöst und ungefähr 4 Stunden am Rückfluss gekocht. Bereits beim Erhitzen scheidet sich ein Niederschlag aus, der durch Abkühlen auf 0" C, Abfiltrieren am Vakuum und Trocknen gewonnen wird. Eine weitere Menge des Produktes kann durch Eindampfen der Mutterlauge gewonnen werden. Die Ausbeute beträgt ungefähr 800/0. Das so erhaltene Hydrazon kann aus Alkohol umkristalli- 60 siert werden und schmilzt bei 2252270 C.
Beispiel 3
10 Gewichtsteile 8-Benzyloxy-chinolin-2-aldehyd und 5,2 Gewichtsteile INH werden zusammen in 100 Volumteilen Alkohol gelöst und am Rückfluss 65 4-5 Stunden gekocht. Dann wird ungefähr die Hälfte des Alkohols abdestilliert und der Rückstand auf Oo C abgekühlt. Man erhält ein kristallines Hydrazon mit einer Ausbeute von ungefähr 800/0, das aus Alkohol umkristallisiert werden kann. Es 70 schmilzt bei 180-182" C.

The aldehyde group can be located anywhere on the quinoline nucleus, but the 5- or 2-position is generally preferred. Of the new substances, the isonicotinic acid hydrazone of 8-hydroxy-quinoline-5-aldehyde, which has a melting point of 239-241 "C, and the isonicotinic acid hydrazone of 8-hydroxy-quinoline-2aldehyde, which has a melting point of 225 to 2270 ° C, are particularly valuable and the isonicotinic acid hydrazone of 8-benzyloxyquinoline-2-aldehyde, which decomposes at 180-182 "C.
The reaction of the quinoline aldehyde with INH is conveniently carried out in a solvent, e.g. B.
Alcohol, carried out, preferably with refluxing. The duration of the heating depends on the aldehyde used, but is generally on the order of 2-8 hours.
The hydrazones formed can be isolated from the reaction mixture, for example, by evaporating some of the solvent and cooling the mixture to 0 ° C., the hydrazones precipitating; they can then be easily removed, e.g. B. by filtration.
The new hydrazones form salts both with bases, for example alkali metal hydroxides, and with mineral acids, for example hydrochloric acid. Other non-toxic salts which are suitable for therapeutic use include: a. the salts with organic acids, e.g. B. p-Amino-salicylic acid and glutamic acid. It is known that p-aminosalicylic acid itself already has an antituberculotic effect, so that the p-amino-salicylic acid salts of the hydrazones are of particular interest with regard to their antituberculotic effect.
The salts of the hydrazones with p-aminosalicylic acid and glutamic acid can, for example, easily be prepared by mixing the free acid or its sodium salt in aqueous solution with a solution of the hydrazone, e.g. B. in hydrochloric acid, and the hydrazone salt formed is precipitated by adding alkali.
Example I.
17.3 parts by weight of 8-hydroxyquinoline-5-aldehyde and 13.7 parts by weight of INH are dissolved together in 800 parts by volume of alcohol and the solution is refluxed for 4-5 hours. It is then concentrated to about half its volume and cooled to 0 "C; the hydrazone precipitates out and can be filtered off in vacuo and dried. Further amounts of hydrazone can be obtained by further concentration of the mother liquor. The total yield of hydrazone is up to approximately 700 / o.
The isonicotinic acid hydrazone of 8-hydroxy-quinoline-5-aldehyde can be recrystallized from alcohol or a large volume of water. It is soluble in dilute hydrochloric acid or dilute caustic soda. The pure hydrazone melts at 2392410 C.
The compound obtained in this way can be converted into salts as follows: a) 2.9 parts by weight of the hydrazone are dissolved in 50 parts by volume in hydrochloric acid with heating and added to a solution of 2.1 parts by weight of sodium paminosalicylate in 40 parts by volume of water.

This salt decomposes at 2802850 C and is very poorly soluble in water. b) 3 parts by weight of the hydrazone are dissolved in 50 parts by volume in hydrochloric acid and! to a solution of 1.5 parts by weight of l (+) - glutamic acid in
Put 10 parts by volume in sodium hydroxide. When adding another 40 parts by volume in sodium hydroxide, the l (+) - glutaminate precipitates as a yellow-brown precipitate. It is very poorly soluble in water and decomposes at 2752800 C.
Example 2
8.7 parts by weight of 8-hydroxy-quinoline-2-aldehyde and 7 parts by weight of INH are dissolved in 100 parts by volume of alcohol and refluxed for about 4 hours. A precipitate separates out on heating and is obtained by cooling to 0 "C, filtering off in vacuo and drying. A further amount of the product can be obtained by evaporating the mother liquor. The yield is approximately 800/0. The hydrazone thus obtained can be recrystallized from alcohol and melts at 2252270 C.
Example 3
10 parts by weight of 8-benzyloxy-quinolin-2-aldehyde and 5.2 parts by weight of INH are dissolved together in 100 parts by volume of alcohol and refluxed for 4-5 hours. About half of the alcohol is then distilled off and the residue is cooled to OoC. A crystalline hydrazone is obtained with a yield of approximately 800/0, which can be recrystallized from alcohol. It melts at 180-182 "C.
