Verfahren zur Einführung von Resten wenig reaktionsfähiger Carbonsäuren, wie des ss-Cyklopentyl-propionylrestes, in die 17a-Oxygruppe solcher Steroide, die am Kohlenstoffatom 17 neben der Oxygruppe eine Acetylgruppe tragen Bekanntlich ist die 17a-Oxygruppe solcher Steroide, die am Kohlenstoffatom 17 gleichzeitig eine CH.C0-Gruppe tragen,
nur unter Anwendung ver schärfter Reaktionsbedingungen zu verestern. Immer hin gelingt diese Veresterung mit den im allgemeinen gut reaktionsfähigen aliphatischen Carbonsäuren recht glatt, wenn man deren Anhydride bzw. Säure chloride, letztere vorteilhaft als Lösung in der zu gehörigen Säure, in Gegenwart stark wirksamer Ver- esterungskatalysatoren wie p-Toluolsulfosäure auf die obengenannten 17a-Oxysteroide einwirken lässt.
Auch diese Methode versagt indessen beim Ver such der Veresterung genannter Steroide mit reak tionsträgeren Säuren wie ss-Cyklopentyl-propion- säure. Denn selbst bei Ausdehnung der Einwirkungs zeit von Cyklopentylpropionsäurechlorid in Gegen wart von Cyklopentylpropionsäure als Lösungsmittel auf 8 Tage lässt sich unter Verwendung von bis zu 1,
1 Moläquivalenten p-Toluolsulfosäure als Ver- esterungskatalysator weder bei 20 noch bei 37 C eine auch nur andeutungsweise Veresterung des 17a Oxypregn-5-en-3ss-ol-20-on-3-acetats in 17-Stellung erzielen.
Es wurde nun gefunden, dass man die angestrebte Veresterung der 17a-Oxygruppe mit reaktionsträ geren Säuren, wie der 6-Cyklopentylpropionsäure, auch bei Steroiden, die am 17-Kohlenstoffatom gleichzeitig eine Acetylgruppe tragen, gleichwohl er zwingen kann, wenn man die reaktionsträgere Säure wasserfei in Gegenwart von Trifluoressigsäure- anhydrid bei erhöhten Temperaturen, vorzugsweise bei 80-90 C, auf die genannten Steroide einwir ken lässt.
Die Einführung des ss-Cyklopentylpropionsäure- restes in die 17a-Oxygruppe erfolgt hierbei in über raschend kurzer Zeit und mit ausgezeichneter Aus beute. Sterische Umlagerungen des Steroids im Be reich des Cyklopentanringes treten nicht in Erschei nung (vgl. Beispiel 2).
Sogar gegebenenfalls zusätz lich irn Steroidmolekül vorhandene Ketogruppen, die bekanntlich bei Verwendung von p-Toluolsulfo- säure als Veresterungsmittel in Enolester übergehen, bleiben bei der erfindungsgemässen Veresterungs- methode unangegriffen.
Die neuen Steroidester sollen als Heilmittel oder als Zwischenprodukte zur Herstellung solcher ver wendet werden. Sie zeichnen sich vor den schon länger bekannten Cyklopentylpropionaten anderer Steroide durch eine vergleichsweise überraschend gute Löslichkeit in öligen Lösungsmitteln aus.
Natürlich ist die erfindungsgemässe Veresterungs- methode auch auf die reaktionsfähigeren aliphati- schen Carbonsäuren übertragbar, allerdings ohne hierbei im allgemeinen besondere Vorteile zu bieten. <I>Beispiel 1</I> 5 g 17a-Oxypregn-5-en-3-ol-20-on-3-acetat wer den in eine 80 C heisse Mischung aus 10 cm3 Cy- klopentylpropionsäure und 5 cm3 Trifluoressigsäure- anhydrid eingetragen.
Das homogene Reaktions gemisch wird 45 Minuten bei dieser Temperatur ge halten und nach Erkalten in Wasser eingegossen. Das hierbei ausgefallene Öl wird in Äther aufgenom men, zur Entfernung der überschüssigen Säure mit verdünnter Natronlauge ausgewaschen, über Na triumsulfat getrocknet und durch Eindampfen im Vakuum vom Äther befreit. Der ölige Rückstand wird mit Pentan zur Kristallisation gebracht und aus Isopropyläther umkristallisiert.
Ausbeute 5,53 g = 83,71/o der Theorie 17a- Oxypregnenolen - 3 - acetat-17 -cyklopentylpropionat, F. = 137-138,5 .
<I>Beispiel 2</I> 1 g 17a-Oxyprogesteron, 4 cm3 Capronsäure und 1 cm3 Trifluoressigsäureanhydrid werden analog Beispiel 1 umgesetzt und aufgearbeitet. Ausbeute 860 mg =<B>66,501o</B> der Theorie 17a-Oxyprogesteron- capronat, F. = 120-121 C.
Es ist identisch mit authentischem Material, das nach bekanntem Verfahren mit Capronsäureanhydrid in Gegenwart von p-Toluolsulfosäure dargestellt wurde. Das beweist, dass trotz der sehr energischen Reaktionsbedingungen keine sterischen Umlagerun gen im Bereich des Cyklopentanringes eingetreten. sind.
<I>Beispiel 3</I> 1 g 17a-Oxyprogesteron wird der 80 C heissen Mischung aus 4 cm3 Cyklopentylpropionsäure und 1 cm3 Trifluoressigsäureanhydrid zugesetzt und ana- log Beispiel 1 umgesetzt und aufgearbeitet.

Even keto groups possibly additionally present in the steroid molecule, which are known to be converted into enol esters when p-toluenesulphonic acid is used as esterification agent, remain unaffected by the esterification method according to the invention.
The new steroid esters are to be used as medicinal products or as intermediates for the manufacture of such ver. They are distinguished from the cyclopentylpropionates of other steroids, which have long been known, by a comparatively surprisingly good solubility in oily solvents.
