Verfahren zur Herstellung von 1-Benzamido-l-phenyl-3-piperidino-propan l-Benzamido-l-phenyl-3-piperidino-propan ist ein Lokalanästhetikum. In der schweizerischen Patentschrift Nr. 322062 ist ein Verfahren beschrieben, das die Herstellung dieser neuen Verbindung in kleinem Massstab erlaubt, aber für die Industrie weniger geeignet ist.
Das vorliegende Verfahren ist für eine industrielle Synthese gut geeignet. Auch dieses Verfahren ist sehr vorteilhaft, weil es die Herstellung von l-Benzamido- l-phenyl-3-piperidino-propan in ausgezeichneter Reinheit und in einer einzigen Verfahrensstufe aus leicht erhältlichen Ausgangsstoffen ermöglicht.
Nach dem erfindungsgemässen Verfahren wird l-Phenyl-3-piperidino-propanol mit Benzonitril in Gegenwart von konzentrierter Schwefelsäure bei einer Temperatur zwischen 30 und 100"C umgesetzt und das l-Benzamido-l-phenyl-3-piperidino-propan isoliert.
Bei der Berechnung der für die Reaktion erforderlichen Menge an konzentrierter Schwefelsäure ist zu berücksichtigen, dass diese Säure bei der Reaktion nicht nur als Katalysator wirkt, sondern auch als Salz mit l-Phenyl-3-piperidino-propanol und mit 1 Benzamido- 1 -phenyl-3-piperidino-propan gebunden wird. Es ist zweckmässig, die konzentrierte Schwefelsäure in einer Menge von 2,5-3,5 Mol pro Mol 1 Phenyl-3-piperidino-propanol zu verwenden; es können jedoch auch höhere oder niedrigere Molverhältnisse angewendet werden. Das Reaktionsgemisch ist ziemlich viskos, so dass ein wirksames Rühren zur Vermeidung von örtlichen Überhitzungen zweckdienlich ist. Ein inertes Lösungsmittel, wie z. B. Eisessig, kann gewünschtenfalls als Verdünnungsmittel zur Erhöhung der Flüssigkeit des Reaktionsmediums und zur Milderung örtlicher Überhitzungen verwendet werden.
Reaktionstemperaturen über 100" würden sich zwar anwenden lassen, sind jedoch weniger vorteilhaft, da dann Nebenprodukte entstehen. Anderseits würde eine Reaktionstemperatur unterhalb 30 die Reaktionsgeschwindigkeit in unerwünschter Weise verlangsamen. Die bevorzugte Reaktionstemperatur liegt zwischen 50 und 60". Da die mit konzentrierter Schwefelsäure als Katalysator durchgeführte Reaktion zwischen l-Phenyl-3-piperidino-propanol und Benzonitril exotherm verläuft, ist äussere Kühlung zweckdienlich, um die Reaktionstemperatur im bevorzugten Bereich zu halten.
Die Reaktion kann folgendermassen durchgeführt werden: Eine Mischung aus äquimolaren Mengen 1 Phenyl-3-piperidino-propanol und Benzonitril wird in einem Reaktionsgefäss gut gerührt, wobei man etwa drei Mol-Äquivalente konzentrierter Schwefelsäure derart zulaufen lässt, dass eine Überhitzung vermieden wird. Es wird gekühlt, um die Reaktionstemperatur während der Zugabe der Säure zwischen 50 und 60 zu halten. Nach beendeter Zugabe wird das Reaktionsgemisch etwa eine Stunde lang bei Umgebungstemperatur gerührt. Das Reaktionsgemisch wird dann auf zerkleinertes Eis gegossen und das l-Benzamido- l-phenyl-3-piperidino-propan wird als freie Base in nach sich bekannter Weise isoliert, z. B. durch Extraktion mit unmischbaren Lösungsmitteln und anschliessender Neutralisation der im Reaktionsgemisch anwesenden Schwefelsäure.
Das so erhaltene 1-Benzamido-l-phenyl-3-piperidino-propan wird dann entweder über ein kristallisierendes Salz, wie z. B. das Chlorhydrat, oder durch unmittelbares Kristallisieren der freien Base weiter gereinigt. Die freie Base kann auch in kristalliner Form dadurch erhalten werden, dass man sie aus einem Säureanlagerungssalz freisetzt und anschliessend aus einem organischen Lösungsmittel umkristallisiert. Geeignete Lösungsmittel für diesen Zweck sind Methanol, Äthanol, Aceton und Äthylacetat, allein oder in Mischung mit Petroläther.
Beispiel 1
Herstellung von l-Benzamido-l-phenyl-3-piperi- dino-propan und l-Benzamido-l -phenyl-3-piperidino- propan-Chlorhydrat.
Ein aus 21,9 g l-Phenyl-3-piperidino-propanol und 10,3 g Benzonitril bestehendes Gemisch wird in einem Dreihalskolben bereitet, der mit einem Tropftrichter, Thermometer und Rührwerk versehen ist. Der Kolben wird in ein Eisbad gestellt und 18 cm3 konzentrierte Schwefelsäure werden unter Rühren des Gemisches tropfenweise zugesetzt. Hierbei setzt sich das l-Phenyl- 3-piperidino-propanol mit dem Benzonitril unter Bildung von l-Benzamido-l-phenyl-3-piperidino-propan um, das als Sulfat anfällt. Die Reaktionstemperatur wird durch Regulierung der Zugabegeschwindigkeit der Schwefelsäure und der Höhe des Bades um den Kolben zwischen 50 und 60 gehalten.
Nachdem die Zugabe der Schwefelsäure beendet ist, wird das Reaktionsgemisch etwa eine Stunde lang ohne äussere Kühlung gerührt und dann auf 200 cm3 Eis gegossen.
Die so erhaltene saure wässrige Lösung, die l-Benz- amido-l-phenyl-3-piperidino-propan als Sulfat enthält, wird zur Entfernung des nicht umgesetzten Benzonitrils oder von nichtbasischen Nebenprodukten mit 200 cm3 Äther gewaschen. Die wässrige Schicht wird dann mit Natronlauge basisch gestellt. Die in der alkalischen Lösung unlösliche freie Base des l-Benz- amid-l-phenyl-3-piperidino-propans scheidet sich ab und wird mit Äther extrahiert. Es wird zweimal mit je etwa 200cm3 Äther extrahiert, worauf die Ätherextrakte vereinigt und getrocknet werden. Das Hydrochlorid des l-Benzamido-l-phenyl-3-piperidino-pro- pans wird dann durch Einblasen von trockenem HCl-Gas in die Ätherlösung ausgefällt. Es wird abfiltriert und aus einem Gemisch von Methanol und Äthylacetat umkristallisiert.
Es werden so 12, 4 g kristallines l-Benzamido-l-phenyl-3-piperidino-pro- pan-Chlorhydrat mit einem Schmelzpunkt von 186 bis 1880 erhalten.
Die freie Base wird erhalten, indem man das Chlorhydrat in Wasser löst, die erhaltene Lösung mit Natriumhydroxyd basisch macht, die erhaltene unlösliche freie Base in Äther extrahiert, den Äther durch Verdampfen in Vakuum entfernt und die freie Base aus einem Gemisch von Aceton und Petroläther umkristallisiert. Das so erhaltene l-Benzamido-l-phenyl- 3-piperidino-propan schmilzt bei 111-112".
Beispiel 2
Herstellung von l-Benzamido- 1 -phenyl-3-piperi- dino-propan.
Ein Gemisch aus 43,8 kg l-Phenyl-3-piperidinopropanol, 21,2 kg Benzonitril und 100 Litern Eisessig wird in einem mit Glas ausgekleidetem Tank bereitet, der mit einer äussern Kühlvorrichtung, z. B. einem Wassermantel, und einem Rührwerk versehen ist.
36 Liter konzentrierte Schwefelsäure werden diesem Reaktionsgemisch langsam unter den gleichen Bedingungen zugesetzt wie in Beispiel 1. Nach beendeter Schwefelsäurezugabe wird das Reaktionsgemisch eine Stunde lang bei Umgebungstemperatur gerührt. Das Reaktionsgemisch wird dann auf Eis gegossen. Die so erhaltene saure wässrige Lösung enthält das l-Benz- amido-l-phenyl-3-piperidino-propan als Sulfat. Die saure wässrige Schicht wird mit Äther extrahiert, wobei der Ätherextrakt verworfen wird. Die wässrige Schicht wird dann mit Natriumhydroxyd basisch gestellt und das ausfallende l-Benzamido-l-phenyl-3-piperidino- propan zweimal mit jeweils drei Volumen Äther extrahiert.
Die vereinigten Ätherextrakte werden getrocknet und der Äther wird im Vakuum verdampft, wobei das l-Benzamido-l-phenyl-3-piperidino-propan als freie Base zurückbleibt. Weitere Mengen an kristallinem l-Benzamido- l-phenyl-3-piperidino-propan werden durch konzentrieren und Kühlen einer Lösung dieses Rückstands in einem Gemisch von Aceton und Petroläther erhalten. Diese Kristalle schmelzen zwischen 109 und 113". Ihr Gesamtgewicht beträgt 27,5 kg.
Process for the preparation of 1-benzamido-l-phenyl-3-piperidino-propane l-benzamido-l-phenyl-3-piperidino-propane is a local anesthetic. In Swiss patent specification No. 322062 a process is described which allows this new connection to be produced on a small scale, but which is less suitable for industry.
The present process is well suited for industrial synthesis. This process is also very advantageous because it enables the production of l-benzamido-l-phenyl-3-piperidino-propane in excellent purity and in a single process stage from readily available starting materials.
In the process according to the invention, l-phenyl-3-piperidino-propanol is reacted with benzonitrile in the presence of concentrated sulfuric acid at a temperature between 30 and 100 ° C. and the l-benzamido-l-phenyl-3-piperidino-propane is isolated.
When calculating the amount of concentrated sulfuric acid required for the reaction, it must be taken into account that this acid not only acts as a catalyst in the reaction, but also as a salt with 1-phenyl-3-piperidino-propanol and 1-benzamido-1-phenyl -3-piperidino-propane is bound. It is advisable to use the concentrated sulfuric acid in an amount of 2.5-3.5 mol per mol of 1 phenyl-3-piperidino-propanol; however, higher or lower molar ratios can also be used. The reaction mixture is quite viscous so effective stirring is useful to avoid local overheating. An inert solvent such as. B. glacial acetic acid can, if desired, be used as a diluent to increase the fluidity of the reaction medium and to alleviate local overheating.
Reaction temperatures above 100 "could be used, but are less advantageous, since by-products are then formed. On the other hand, a reaction temperature below 30 would slow down the reaction rate in an undesirable manner. The preferred reaction temperature is between 50 and 60". Since the reaction between 1-phenyl-3-piperidino-propanol and benzonitrile, which is carried out with concentrated sulfuric acid as a catalyst, is exothermic, external cooling is useful in order to keep the reaction temperature in the preferred range.
The reaction can be carried out as follows: A mixture of equimolar amounts of 1-phenyl-3-piperidino-propanol and benzonitrile is stirred well in a reaction vessel, about three molar equivalents of concentrated sulfuric acid being added in such a way that overheating is avoided. It is cooled to keep the reaction temperature between 50 and 60 during the addition of the acid. When the addition is complete, the reaction mixture is stirred for about an hour at ambient temperature. The reaction mixture is then poured onto crushed ice and the l-benzamido-l-phenyl-3-piperidino-propane is isolated as the free base in a manner known per se, e.g. B. by extraction with immiscible solvents and subsequent neutralization of the sulfuric acid present in the reaction mixture.
The 1-benzamido-1-phenyl-3-piperidino-propane obtained in this way is then either via a crystallizing salt, such as. B. the chlorohydrate, or further purified by immediate crystallization of the free base. The free base can also be obtained in crystalline form by releasing it from an acid addition salt and then recrystallizing it from an organic solvent. Suitable solvents for this purpose are methanol, ethanol, acetone and ethyl acetate, alone or in admixture with petroleum ether.
example 1
Production of l-benzamido-l-phenyl-3-piperidino-propane and l-benzamido-l-phenyl-3-piperidino-propane chlorohydrate.
A mixture consisting of 21.9 g of 1-phenyl-3-piperidino-propanol and 10.3 g of benzonitrile is prepared in a three-necked flask equipped with a dropping funnel, thermometer and stirrer. The flask is placed in an ice bath and 18 cc of concentrated sulfuric acid is added dropwise while stirring the mixture. Here, the l-phenyl-3-piperidino-propanol reacts with the benzonitrile to form l-benzamido-l-phenyl-3-piperidino-propane, which is obtained as sulfate. The reaction temperature is maintained between 50 and 60 by regulating the rate of addition of the sulfuric acid and the height of the bath around the flask.
After the addition of the sulfuric acid has ended, the reaction mixture is stirred for about an hour without external cooling and then poured onto 200 cm3 of ice.
The acidic aqueous solution thus obtained, which contains 1-benzamido-1-phenyl-3-piperidino-propane as sulfate, is washed with 200 cm3 of ether to remove the unreacted benzonitrile or non-basic by-products. The aqueous layer is then made basic with sodium hydroxide solution. The free base of l-benzamide-l-phenyl-3-piperidino-propane, which is insoluble in the alkaline solution, separates out and is extracted with ether. It is extracted twice with about 200cm3 ether each time, after which the ether extracts are combined and dried. The hydrochloride of the l-benzamido-l-phenyl-3-piperidino-propane is then precipitated by blowing dry HCl gas into the ethereal solution. It is filtered off and recrystallized from a mixture of methanol and ethyl acetate.
This gives 12.4 g of crystalline 1-benzamido-1-phenyl-3-piperidino-propane chlorohydrate with a melting point of 186 to 1880.
The free base is obtained by dissolving the chlorohydrate in water, making the resulting solution basic with sodium hydroxide, extracting the insoluble free base obtained in ether, removing the ether by evaporation in vacuo and recrystallizing the free base from a mixture of acetone and petroleum ether . The l-benzamido-l-phenyl-3-piperidino-propane thus obtained melts at 111-112 ".
Example 2
Production of l-benzamido-1-phenyl-3-piperidino-propane.
A mixture of 43.8 kg of l-phenyl-3-piperidinopropanol, 21.2 kg of benzonitrile and 100 liters of glacial acetic acid is prepared in a glass-lined tank, which is equipped with an external cooling device, e.g. B. a water jacket, and an agitator is provided.
36 liters of concentrated sulfuric acid are slowly added to this reaction mixture under the same conditions as in Example 1. When the addition of sulfuric acid is complete, the reaction mixture is stirred for one hour at ambient temperature. The reaction mixture is then poured onto ice. The acidic aqueous solution thus obtained contains the l-benzamido-l-phenyl-3-piperidino-propane as sulfate. The acidic aqueous layer is extracted with ether, the ether extract being discarded. The aqueous layer is then made basic with sodium hydroxide and the l-benzamido-l-phenyl-3-piperidino-propane which precipitates is extracted twice with three volumes of ether each time.
The combined ether extracts are dried and the ether is evaporated in vacuo, the l-benzamido-l-phenyl-3-piperidino-propane remaining as the free base. Further amounts of crystalline 1-benzamido-1-phenyl-3-piperidino-propane are obtained by concentrating and cooling a solution of this residue in a mixture of acetone and petroleum ether. These crystals melt between 109 and 113 ". Their total weight is 27.5 kg.
