<Desc/Clms Page number 3>

<Desc/Clms Page number 5>

<Desc/Clms Page number 6>
<Desc/Clms Page number 7>
<Desc/Clms Page number 8>
Schema 2).

Process for the preparation of salts of triterpenic acids insoluble in water It is known that triterpenic acids occur in numerous other plants apart from hawthorn, in particular plants of the family of the labiate, rosaceae and ericaceae.
In contrast to Crataegus oxyyacantha and monogyna, which in leaves and flowers also contain crataegolic acid in addition to oleanolic and ursolic acid, in other plants, as far as could be determined, crataegolic acid occurs only rarely or in very small amounts, so that it usually occurs only contain other triterpenic acid, especially oleanolic and ursolic acid.
The main patent describes a process for the production of salts of the water-insoluble triterpenic acids from hawthorn. The subject of the present patent is a process for the production of salts of water-insoluble triterpene acids, which is characterized in that other raw materials containing such triterpene acids than Crataegus are extracted with solvents which contain halogenated liquid hydrocarbons that boil above 40 and below 200 ,
that the extracts are freed of fat and fat-soluble colorants after evaporation of the solvents and the triterpenic acids are converted into salts after further purification by treatment with bases.
Mixtures of the above-mentioned halogenated hydrocarbons with low-boiling, oxygen-containing solvents, in particular the initial members of the aliphatic alcohol series, which have 1 to 5 carbon atoms, are particularly suitable for extracting the triterpene acids, the alcohol content in such a mixture being 50% by volume, but especially 30% by volume. / o should not exceed.
Mixtures of this type consistently give by far the highest yields of triterpenic acids. In particular, the yields are consistently better than when the halogenated hydrocarbons are used without the addition of alcohol. Instead of mixtures with alcohols, mixtures can be used which, instead of the alcohols, contain other oxygen-containing compounds such as lower aliphatic ketones or esters, albeit with somewhat less success.
By far the cheapest halogenated hydrocarbon is trichlorethylene, as it gives very good yields of triterpenic acids even with little or no addition of alcohol. However, the addition of alcohol is generally advantageous even with trichlorethylene. As a result, trichlorethylene or mixtures which, in addition to trichlorethylene, contain up to 50% by volume of alcohol, preferably up to 30% by volume, can be used.
In the case of the other halogenated hydrocarbons, the addition of oxygen-containing, low-boiling solvents, in particular alcohols, should be at least 5, but preferably 10 vol%, so that a good yield is obtained with a low solvent consumption.
In the same way, extracts from such drugs, which contain water-soluble components in addition to water-insoluble components, can also be treated for the purpose of separating off the triterpenic acid.
From the triterpenic acid-containing extracts obtained with the aid of the solvent, fatty substances and fat-soluble dyes are separated off, preferably in accordance with the process of the main patent, by treatment with aliphatic or cycloaliphatic hydrocarbons such as ligroin, gasoline, cyclohexane and the like.
Even if the starting drug has already been defatted, such treatment of the extracts is always carried out according to the invention, since small residues of fats and, above all, larger amounts of fat-soluble colorants are still present in the extracts.

Yields of crude triterpenic acids obtained by the process from various plants with the aid of trichlorethylene containing 5% by volume of methanol are shown in the table below.

If other halogenated hydrocarbons are used, a higher alcohol content is always necessary to achieve the same yields.
The raw triterpenic acid mixture freed from fat and fat-soluble dyes is still colored throughout by dyes that are not fat-soluble and is therefore further purified according to the invention, e.g.
B. by adsorption. It is best to use solutions of the triterpenic acid mixture in a mixture of halogenated hydrocarbons, for example trichlorethylene, and a lower aliphatic alcohol, preferably with 1 to 3 carbon atoms. A solution in a mixture of equal parts of halogenated hydrocarbon and alcohol is particularly favorable. Activated carbon, active aluminum oxide, bentonite and the like can be used as the adsorbent.
Depending on the amount of adsorbent used and the starting material from which the triterpenic acid mixture was isolated, the purified product is practically colorless to pale yellow or light green after the solvent has been distilled off.
<Desc / Clms Page number 3>
Further purification of the crude triterpenic acid mixture by selective adsorption is mostly not necessary if the individual triterpenic acids are to be isolated; however, it turned out to be advantageous because the separation is easier.
The further purification of the triterpenic acid mixture is also possible by treatment with an ester with a maximum of 3 carbon atoms, ie methyl formate, methyl acetate and ethyl formate, but especially methyl formate, in the cold. In this way, the colorants are largely removed, together with any sitosterols and small amounts of low-melting triterpenic acids of a still unknown constitution.
However, it is also possible to use both methods for further purification of the triterpenic acid mixture one after the other.
A separation into the individual triterpenic acids is possible. As has been established, however, fractional concentration or fractional precipitation does not result in very pure individuals, but the individual acids are still contaminated with other acids.
As has been found, better separation can be achieved by treatment with esters which have at most 3 carbon atoms, in particular methyl formate, in the heat. The ursolic acid remains practically undissolved, while oleanolic acid and lower-melting triterpenic acids, together with any remaining amounts of dye, go into solution.
Any crataegolic acid that may be present only partially dissolves; the main amount mostly remains with the undissolved urolic acid. If there is no crataegolic acid in the plant in question, the residue from treatment with hot methyl formate consists of almost pure ursolic acid.
According to the invention, the triterpenic acids obtained are converted into salts by treatment with bases. The hydroxides of the alkali metals, ammonium hydroxide and water-soluble organic bases can be used as bases. Example 1 Erica flowers were completely dried in a drying cabinet at 95 ° C. To achieve the maximum yield of triterpene acids, the dry drug was ground in a mill to form drug meal.
3500 g of this comminuted Erica flower drug were extracted in three stages, each with five times the amount (17.5) of a solvent mixture consisting of 80 parts of trichlorethylene and 20 parts of methanol at the boiling point under reflux. For each stage, the heating time was two hours, in each case from the start of boiling, followed by hot filtration.
The extract filtrates were combined regardless of the partially flocculated triterpenic acid components and the solvent was completely distilled off in vacuo, initially at 10 mm Hg and finally at 15 mm Hg.
The thick, fatty, yellow-brown (green in most other plants) residue was taken up in two liters of gasoline with a boiling point of 60 to 70 ° C. while stirring and the undissolved triterpenic acid residue was separated off by filtration. The residue on the filter was washed with gasoline until the filtrate appeared almost colorless.
The crude triterpenic acid mixture obtained in this way has a yellow color after drying, weighs 189 g and melts at 225 to 227 ° C.
The sodium salts of the acids were prepared by treating a solution of the triterpenic acids obtained in this way and further purified in accordance with the later explanations, for example a solution in trichlorethylene or ether with sodium hydroxide solution.
In the same way, i. H. Using the same solvent mixture, the same temperature, proportions and other conditions, a further number of plants were treated, which are listed in detail in Table I below with the results obtained.

The yields obtained were practically the same within the error limit, and consistently somewhat lower, at 2 to 6%.
The color and melting point of the triterpenic acid products obtained with trichlorethylene corresponded to those in the table.
Example 2 In the treatment of the finely ground crude triterpenic acid mixtures obtained according to Example 1, each with 15 times the amount of methyl formate at a temperature of 15 ° C., the sitosterols, most of the green and yellow fat-insoluble dyes and some of the lowest melting triterpenic acid (175 ° C. ) and other impurities.
The procedure is as follows: The triterpenic acid powder is stirred with 10 times the volume of the weight of the triterpenic acid methyl formate for 60 minutes and then the cooled mixture (15 ° C.) is filtered through suction filters under low vacuum. The residue on the filter is largely washed with the remaining 5-fold amount of methyl formate until the filtrate appears almost colorless.
The dried triterpenic acid residues are only weakly colored or even almost colorless. The yields listed in Table I decrease by almost 15 to 25% after treatment with methyl formate. The amount of separated accompanying substances differs from plant to plant.

It was heated for two hours from the start of boiling and then filtered while hot. Despite partial crystallization of the triterpenic acids, the filtrates were combined and completely separated from the solvent in vacuo, initially at 100 mm Hg and then at 15 mm Hg.
The fatty, viscous residue was taken up in two liters of gasoline with a boiling point of 60 to 70 ° C. while stirring, and the undissolved triterpenic acid residue was separated off by filtration. The residue on the filter was washed with gasoline until the filtrate appeared colorless. After drying, a weight of 150 g was found. This residue melted at 240 to 243 ° C and was green in color.
The potassium salts of the triterpenic acids were produced by treating an ethereal solution of the further purified triterpenic acids with an in potassium hydroxide solution with stirring. Other plants were treated in the same way.
The results obtained are summarized in Table 1I.

For comparison, the first 10 drugs were extracted with chloroform, the yields being 10 to 15% lower than indicated in Table II. Chloroform without the addition of an alcohol therefore gives significantly lower yields and is consequently inferior to the mixtures and trichlorethylene.
Example 4 The crude triterpenic acids of plants 1, 5, 7, 11, 12, 14, 18, 19 and 27 of Table TI are treated with 10 times the volume of methyl formate per part by weight of crude triterpenic acids, in such a way that, similar to example 2 going on, but at
<Desc / Clms Page number 6>
a temperature of 10 ° C works. In the methyl formate, the sitosterols, the green and yellow coloring matter and part of the lowest melting triterpenic acid dissolve. The triterpenic acids lighten, most of them become colorless.
The weight of the yields is reduced by an average of 20%. In the case of the Uva Ursi example, the weight of the yield z. B. by 18%, so that 113g with a melting point of 249 to 254 'C remain with a light green color.
In each case 1 g of the triterpenic acids purified in this way was dissolved in 10 g of alcohol and diethanolamine was added until a pH of 7.7 was reached, and 10 g of propylene glycol were added to the heated mixture, followed by further diethanolamine until a pn value of 8.5 was reached added. Durable solutions of other organic amines can be prepared in the same way.
Example S The crude triterpenic acid mixtures of the drugs according to Tables I and II or the mixtures aftertreated with methyl formate were further purified by treatment with adsorbents as follows: 100 g of each of the somewhat colored triterpenic acid mixtures are mixed with 20 times the amount of a solvent mixture of trichlorethylene and methanol 1: 1 dissolved under reflux in the warmth and filtered hot. With the addition of one to three teaspoons of decolorizing charcoal, the solution is refluxed for three to six hours and filtered while hot.
The filtrate solution is brought to dryness under a vacuum of initially 100 mm Hg and finally 15 mm Hg and the white triterpenic acid masses are collected. The melting points of the white products increased only slightly compared to the products purified with methyl formate.
The triterpenic acids purified in this way were dissolved in trichlorethylene and converted into the difficult-to-melt potassium salts by shaking with a 1/2 potassium hydroxide solution. Lithium, sodium, calcium and ammonium salts can also be produced in the same way.
It has been observed that most triterpenic acid mixtures contain 3 to 5 individual compounds. The following examples serve to illustrate the complete separation of the triterpenic acid mixtures into individual compounds.
This separation method can be carried out both with the triterpenic acids that are still colored and with the decolored products.
EXAMPLE 6 182 g of triterpenic acids were obtained from 3500 g of Flores ericae according to Example 1 by extraction with trichlorethylene and degreasing with gasoline. The dried greenish yellow powder melted at 2240 ° C. The yield of 182 g, based on the weight of the drug, was 5.2%.
For further purification, this crude mixture was finely pulverized and treated with 1800 cm3 of methyl formate at 10 ° C. for 60 minutes with stirring and filtered through a suction filter under a low vacuum. The residue on the suction filter was washed with 1000 cm3 of cooled methyl formate until the filtrate was only slightly colored.
After drying of the undissolved part B, 151 g of triterpenic acids remained, which were almost colorless and melted at 236 to 240.degree.
The separated part B 1 is in the methyl formate solution, from which 20 g of solid substance with a melting point of 154 to 157 ° C. could be separated by treatment with the lowest-boiling ligroin. This solid substance contains the sitosterols, together with part of the lowest melting point triterpenic acid, which when purified melts at about 175 ° C.
The main group of triterpenic acids with the designation B was refluxed in four stages with 1000 cm3 of methyl formate each and filtered while hot.
18 g dissolve in the hot methyl formate, these are deposited as group B2 and further separated separately. Group A of the triterpenic acids remained undissolved; it weighed 113 g and melted at 241 to 244 ° C.
Separation of the triterpenic acid group A: To separate the immediately crystallizable ursolic acid from this group, proceed as follows: The triterpenic acid group A, which has been ground to a fine powder, was treated with methylene chloride in four stages with 1000 cm3 each at boiling point for 60 minutes and was hot filtered. 102 g, which was almost pure ursolic acid and melted at 254 ° C., remained insoluble.
In order to obtain the crystallized ursolic acid from it, this 102 g was dissolved in 5000 cm of ethyl alcohol and the solution was placed in a refrigerator at 2 ° C. for several days for crystallization. The solution in ethyl alcohol must be made at the boiling point. 89 g of ursolic acid crystallized with a melting point of 279 to 282 C.
Another 8 g of crystallized ursolic acid could be obtained from the mother liquor as follows: The dry residue of the mother liquor was boiled twice more with 100 cubic meters of methylene chloride at boiling point for 30 minutes each time and the residue was dissolved in ethyl alcohol after drying. 8 g of further ursolic acid then crystallized from this solution.
In order to keep the products of the triterpenic acid group A completely white, it is advisable to dissolve them in a solvent mixture of 1 part trichlorethylene and 1 part methanol before treatment with methylene chloride. One needed for
<Desc / Clms Page number 7>
100 g triterpenic acids 2000 cm9 of the mixture. The discoloration is done with charcoal while the solution is boiled for five hours.
Separation of the triterpenic acid group B2: If the methyl formate solution obtained by hot extraction from the triterpenic acid group B is brought to dryness by distilling off the solvent, 18 g of residue with a melting point of 224 to 226 ° C. are obtained.
To separate the compounds contained in this residue, extraction is also carried out with methylene chloride while hot.
Group B2 was thoroughly boiled for 60 minutes with 250 cm3 of methylene chloride each time. The residue B G 1, which had a melting point of 262 to 264 ° C. and weighed 5.7 g, remained undissolved.
Pure oleanolic acid could be obtained from this residue B G 1 by direct crystallization with alcohol.
The residue of 5.7 g was dissolved in 200 cm3 of alcohol and filtered in the heat, the last traces of dye were decolorized with a teaspoon of charcoal, the volume was reduced to 50 cm3 after the charcoal had been separated off and allowed to crystallize.
3.8 g of the purest oleanolic acid with a melting point of 300 ° C. crystallize in the refrigerator. After a single dissolution from ethyl alcohol, oleanolic acid is obtained, which melts at 304 ° C. The further separation of the triterpenic acids remaining in the methylene chloride solution took place again as follows.
The methylene chloride solution of group B 2 was concentrated to a tenth volume and placed in ice. 5.2 g of a triterpenic acid separated out, which appears quite pure in the chromatogram and melted at 205 ° C. (B G 2).
The clear filtrate was freed from solvent by distilling off and the dry residue was collected. The melting point of the not yet completely pure, lowest melting point triterpenic acid was 160 to 156 ° C. (B G 3).
The group A methylene chloride solution was concentrated to one tenth volume and placed in ice. 17 g of a triterpenic acid separated out, which melted at 222 to 224 ° C. and appeared fairly pure (AG2).
The pale filtrate of the remaining methylene chloride solution was evaporated to dryness. Since the remaining mass is sticky, no melting point could be determined. The amount was 3.8 g (AG 3).
The pure oleanolic acid and ursolic acid produced in this way were each dissolved in trichlorethylene and stirred with sodium hydroxide solution, and the sodium salts formed were separated off.
For a better overview, reference is made to the scheme -l.
EXAMPLE 7 3200 g of apple pomace were very finely ground after drying and extracted using the procedure of Example 1 with a solvent mixture consisting of 80 parts of trichlorethylene and 20 parts of methanol.
The crude triterpenic acid mixture obtained in this way weighed 63.5 g (1.98% yield, based on the dry starting drug). The color of the triterpenic acids was light yellow; Melting point 223 to 225 ° C.
This finely powdered triterpenic acid mixture was first treated with 600 cm3 of methyl formate at 10 ° C. with stirring for 60 minutes and then filtered through a suction filter. The pale yellow residue was washed with 300 cm3 of cooled methyl formate and then dried.
Part B of the triterpenic acids remained undissolved (43 g). This residue is almost colorless and has a melting point of 246 to 249 ° C.
Part B 1 of the triterpenic acid mixture is in the methyl formate filtrate. The methyl formate was distilled off and the residue was boiled with gasoline. 19 g with a melting point of 136 to 139 ° C. remained undissolved.
The gasoline solution left 8.5 g of a sticky residue on evaporation.
The main group B of the triterpenic acids was vigorously refluxed in four stages with 500 em3 methyl formate each and filtered hot.
17 g, with a melting point of 234 to 236 ° C., dissolve in the hot methyl formate; these are deposited as group B 2 and further separated separately.
Group A of the triterpenic acids remained undissolved and weighed 21.9 g, it melted at 255 to 258 ° C. Separation of the triterpenic acid group A: To separate the ursolic acid that is immediately crystallizable from this group, the following procedure is used: The one ground to a fine powder Triterpenic acid group A was treated with methylene chloride in four stages with 300 cm3 each at the boiling point for 30 minutes and filtered while hot.
15 g remained undissolved, which are almost pure ursolic acid and had a melting point of 268 to 269 ° C. (AG 1).
In order to obtain crystallized ursolic acid from it, this amount is dissolved in 600 cm3 of ethyl alcohol at the boiling point and this solution is prepared for crystallization after filtration.
After a few days, 13 g of ursolic acid with a melting point of 279 to 282 ° C. crystallize from this solution.
The methylene chloride filtrates were concentrated to 200 cm3 and the concentrated methylene chloride solution was placed in ice water for 60 minutes. 3 g of a triterpenic acid (AG2) precipitated out, which melted at 247 to 248 ° C. without purification.
After the solvent has been distilled off, the mother liquor of the methylene chloride solution leaves a residue (AG3) of 3.5 g, which melted at 205 to 208 ° C.
<Desc / Clms Page number 8>
Separation of the triterpenic acid group B2: The methyl formate solution obtained by hot extraction was brought to dryness, a residue of 17 g remained, which melted at 234 to 236 ° C.
Group B2 was thoroughly boiled four times for 30 minutes each time with 250 cm3 of methylene chloride each time.
A residue B G 1, which weighed 6 g and had a melting point of 250 to 258 ° C., remained undissolved.
This time it was not possible to obtain oleanolic acid from this residue by direct crystallization with alcohol, but again 1.5 g of ursolic acid with a melting point of 279 to 281 ° C.
The triterpenic acids remaining in the methylene chloride solution were separated as follows: The methylene chloride solution was concentrated to a tenth volume and placed in ice.
2.8 g of triterpenic acid (BG2), which had a melting point of 210 to 215 ° C., separated out.
The melting point of the residue of the methylene chloride solution after the methylene chloride had been distilled off was 163 to 165 ° C. and weighed 4.5 g (B G 3).
In this experimental example, too, it was advisable to remove the last remains of the dye with decolorizing charcoal in order to obtain snow-white crystals.
The pure ursolic acid obtained by the above method was dissolved in chloroform and converted into the potassium salt of ursolic acid by shaking with a 1/2 potassium hydroxide solution (cf. Scheme 2).
