Verfahren zur Herstellung organischer Verbindungen Aus Pflanzen der Rauwolfia-Arten, insbesondere wus Rauwolfia canescens, liess sich ein neues Alkaloid mit beruhigender und blutdrucksenkender Wirkung, Deserpidin genannt, in reiner Form gewinnen. Es besitzt als blutdrucksenkendes Mittel grosse therapeu tische Bedeutung. Es lässt sich aus Pflanzenmaterial der Rauwolfia-Arten isolieren, z. B. nach dem am Schluss des Beispiels 1 angegebenen Verfahren. über den chemischen Aufbau des Deserpidins ist bis jetzt nichts bekanntgeworden.
Es hat sich gezeigt, dass man unerwarteterweise durch Behandlung von Deserpidin mit stärkeren, alkalisch verseifenden Mitteln zu einer neuen Car- bonsäure gelangen kann, die den Namen Deserpidin- säure tragen soll. Deserpidinsäure besitzt, wie sich aus unsern Untersuchungen ergeben hat, neben der freien Carboxylgruppe eine freie Hydroxylgruppe und kann durch folgende Formel dargestellt werden:

worin Des den im Deserpidin an die veresterte Hy- droxyl- und Carboxylgruppe gebundenen, zweiwer tigen organischen Rest bedeutet; weiter zeigten unsere Untersuchungen, dass sich Deserpidinsäure durch überführung der Carboxylgruppe in die Carbo- methoxygruppe und der Hydroxylgruppe in die 3,4,5- Trimethoxy-benzoyloxygruppe in Deserpidin zurück verwandeln lässt.
Die Erfindung betrifft ein Verfahren zur Her stellung von Deserpidinsäure-diestern oder deren Salzen, insbesondere derjenigen Ester, in denen die Carboxylgruppe mit Alkanolen, vorzugsweise nie deren, wie Äthanol, Propanol, Butanol, vor allem aber Methanol, verestert und die Hydroxylgruppe durch eine Carbon- oder Sulfonsäure verestert ist.
Bevorzugte Säuren sind Carbonsäuren oder aliphati- schen, alicyclischen, araliphatischen, heterocyclischen oder aromatischen Reihe, besonders solche, die einen ungesättigten Monoring enthalten, vor allem aroma tische und araliphatische Carbonsäuren, deren Phe- nylreste, vorzugsweise in 4-Stellung, durch verätherte Oxy-, insbesondere niedere Alkoxygruppen,
wie Me- thoxy- oder Methylendioxygruppen, substituiert sind, in erster Linie Methoxybenzoesäuren, wie 3,4-Dimethoxy- benzoesäure, 4-Methoxybenzoesäure, besonders aber 3,4,5-Trixnethoxy-benzoesäure. Weitere Säuren dieser Art sind vorab Benzoesäure oder Zimtsäuren, wie Zimt säure, in erster Linie Trialkoxyzimtsäuren, vorzugs weise 3,4,5-Trimethoxyzimtsäure,
O-Carbalkoxy- syringasäuren, wie die O-Carbäthoxy-syringasäure, Phenylessigsäure, Furancarbonsäuren, wie Pyridin-3- carbonsäure, niedere Fettsäuren, z. B. Essigsäure. Be sonders wertvoll sind Ester der Deserpidinsäure der Formel:

worin R einen niederen Alkylrest, vor allem Methyl, darstellt.
Die erfindungsgemäss erhältlichen Diester der Deserpidinsäure weisen wie das Deserpidin wertvolle pharmakologische Eigenschaften auf und können als Heilmittel dienen. So zeigen z. B. folgende Ver bindungen eine sedative und hypotensive Wirkung:
Der 3,4,5-Trimethoxy-benzoesäureester des Deser- pidinsäuremethylesters, der 3,4-Dimethoxy-benzoe- säureester des Deserpidinsäuremethylesters, der 4- Methoxy-benzoesäureester des Deserpidinsäureme- thylesters, der Furan-2-carbonsäureester des Deser- pidinsäuremethylesters, der Nicotinsäureester des Deserpidinsäuremethylesters, der 3,4,
5-Trimethoxy- zimtsäureester des Deserpidinsäuremethylesters, der Essigsäureester des Deserpidinsäuremethylesters, der 3,4,5-Trimethoxy-benzoesäureester des Deserpidin- säureäthylesters, der O-Carbäthoxy-syringasäure- ester des Deserpidinsäuremethylesters.
Das erfindungsgemässe Verfahren zur Herstellung der Diester der Deserpidinsäure oder deren Salzen ist dadurch gekennzeichnet, dass man einen Ester der Deserpidinsäure mit freier Hydroxylgruppe durch Veresterung der Hydroxylgruppe in einen Deserpidin- säureester mit veresterter Hydroxyl- und Carboxyl- gruppe überführt. Zur Veresterung mit der ge
wünschten Säure setzt man die Ester mit freier Hy- droxy baruppe vorteilhaft mit reaktionsfähigen, funktio nellen Säurederivaten, insbesondere mit Halogeniden, wie z. B dem Chlorid, oder Anhydriden um. Die Reaktion wird zweckmässig in Anwesenheit von Ver dünnungsmitteln und/oder Kondensationsmitteln durchgeführt.
Bei Verwendung von Säurehalogeniden arbeitet man vorteilhaft in wasserfreien Lösungsmit teln in Gegenwart säurebindender Mittel, wie Alkali oder Erdalkalicarbonaten, oder starken organischen Basen, wie tertiären Aminen. So verwendet man z. B. für Säurehalogenide Pyridin als Lösungsmittel.
Je nach der Arbeitsweise erhält man die Diester der Deserpidinsäure in freier Form oder als Salze. Da die Deserpidinsäureester eine basische Gruppe auf weisen, können sie Salze mit Säuren bilden.
So lassen sich die erhaltenen Deserpidinsäureester beispielsweise durch Behandeln mit anorganischen oder organischen Säuren, wie Halogenwasserstoffsäuren, Schwefel säure, Phosphorsäuren, Salpetersäure, Oxyäthansul- fonsäure, Toluolsulfonsäure, Essigsäure, Weinsäure, Zitronensäure, in ihre Salze mit Säuren überführen. Aus den Salzen können die Deserpidinsäureester wie der in freier Form gewonnen werden.
Verfahrensgemäss können die Ausgangsstoffe auch in Form der genannten Salze verwendet werden.
Die neuen pharmakologisch wirksamen Ester können als Heilmittel, z. B. in Form pharmazeuti scher Präparate, Verwendung finden, welche sie oder ihre Salze in Mischung mit einem für die enterale oder parenterale Applikation geeigneten pharmazeutischen organischen oder anorganischen Trägermaterial ent halten. Für die Bildung desselben kommen solche Stoffe in Frage, die mit den neuen Verbindungen nicht reagieren, wie z. B.
Wasser, Gelatine, Milch zucker, Stärke, Magnesiumstearat, Talk, pflanzliche Öle, Benzylalkohole, Ascorbinsäure, Gummi, Poly- alkylenglykole, Vaseline, Cholesterin, Tragacanth, Alkohol oder andere bekannte Arzneimittelträger. Die pharmazeutischen Präparate können z. B. als Tabletten, Dragees, Pulver, Salben, Cremen oder in flüssiger Form als Lösungen, Suspensionen oder Emulsionen vorliegen.
Gegebenenfalls sind sie sterili siert und bzw. oder enthalten Hilfsstoffe, wie Kon- servierungs-, Stabilisierungs-, Netz- oder Emulgier- mittel, Salze zur Veränderung des osmotischen Druk- kes oder Puffer. Sie können auch noch andere thera peutisch wertvolle Stoffe, wie 1-Hydrazinophthalazin- hydrochlorid, enthalten. Die Präparate können nach üblichen Methoden gewonnen werden.
In den nachfolgenden Beispielen besteht zwischen Gewichtsteil und Volumteil die gleiche Beziehung wie zwischen Gramm und Kubikzentimeter. Die Temperaturen sind in Celsiusgraden angegeben.
<I>Beispiel 1</I> 0,3 Gewichtsteile Deserpidinsäuremethylester wer den in 2 Volumteilen trockenem, -destilliertem Pyri- din gelöst und die Lösung langsam zu einer ge kühlten Mischung von 0,33 Gewichtsteilen 3,4,5-Tri- methoxy-cinnamoylchlorid in 2 Volumteilen trok- kenem, destilliertem Pyridin zugegeben. Mit 1 Vo- lumteil trockenem Pyridin wird nachgewaschen.
Man lässt dann 4 Tage bei 5 stehen, gibt hierauf die Lö sung in 20 Volumteile Eiswasser, fügt 10 Volumteile einer 10%igen wässrigen Ammoniaklösung zu, arbeitet die Mischung während 5 Minuten gut durch und extra hiert 3mal mit je 15 Volumteilen Methylenchlorid. Die vereinigten Extrakte werden mit 5 Volumteilen einer kalten Natriumchloridlösung gewaschen, über wasserfreiem Kaliumcarbonat getrocknet und im Va kuum zu einem festen Rückstand eingedampft.
0,41 Gewichtsteile davon löst man in 10 Volumteilen Ben zol und 2 Volumteilen Hexan und chromatographiert an 8 Gewichtsteilen aktiviertem Aluminiumoxyd (Woelm; Wirksamkeit I).
Aus der mit Benzol (400 Volumteile) eluierten Fraktion lässt sich nach Ent fernen des Lösungsmittels und Umkristallisieren aus Methanol-Hexan der 3,4,5-Trimethoxy-zimtsäure- ester des Deserpidinsäuremethylesters in Form von kleinen weissen Platten erhalten, die bei 133-143 sintern, bei 182 wieder kristallisieren und endlich bei 216-217 schmelzen. Es besitzt sedative und hypotensive Wirksamkeit.
Die Analyse ergibt eine empirische Formel C34H4oO.N2.
Ultraviolett-Spektrum: Maxima bei: 2, - 226 bis <I>227</I> my (E <I>=</I> 53 900), 291 mu (a = 21600); Minima bei: A = 254-255 m (s = 6700).
Im Infrarot- Spektrum (in Nujol) zeigt er starke Banden bei: 2939-2839, 1729, 1704, 1458, 1313, 1276, 1252, 1182, 1153, 1129 cm-i; mittlere Banden bei: 3402, 1636, 1584, 1507, 1420, 1378, 1044, 995, 831, 728 cm-'; schwache Banden bei: 916, 878 cm-i; und Schultern bei: 3360, 3043, 1330, 1301, 1211, 1102, 1057, 1010, 738 cm-'.
Das als Ausgangsmaterial verwendete 3,4,5-Tri- methoxyzimtsäurechlorid lässt sich wie folgt erhalten: 4 Gewichtsteile 3,4,5-Trimethoxy-zimtsäure wer den 35 Minuten in einem wasserfreien System mit 6 Volumteilen frisch destilliertem Thionylchlorid am Rückfluss gekocht, dann das überschüssige Thionyl- chlorid im Vakuum entfernt und 2mal trockenes Ben- zol zugegeben, das wieder abdestilliert wird.
Der kristalline Rückstand wird 2mal aus Hexan-Äther umkristallisiert. Man erhält so schwach gelbe Pris men vom F. 95-96 .
Der als Ausgangsmaterial verwendete Deserpidin- säuremethylester kann z. B. wie folgt erhalten werden: 500 Gewichtsteile getrocknetes, fein gemahlenes Wurzelmaterial von Rauwolfia canescens werden zuerst während einer Stunde mit 2000 Volumteilen, dann während 45 Minuten mit 1000 Volumteilen und anschliessend 2mal während 30 Minuten mit je 1000 Volumteilen kochendem Methanol extrahiert und die Extrakte jeweils heiss filtriert.
Man engt die vereinigten Extrakte im Vakuum auf 75 Volum- teile einer dicken, sirupähnlichen Lösung ein, gibt dann unter gutem Mischen 75 Volumteile Methanol und 150 Volumteile einer 15%igen Essigsäure zu und extrahiert 2mal mit je 100 Volumteilen Hexan. Die Hexanextrakte zieht man mit 15 Volumteilen 15%iger Essigsäure aus,
vereinigt die Essigsäureauszüge und extrahiert sie 3mal mit je 75 Volumteilen und 1mal mit 50 Volumteilen Äthylenchlorid. Die ersten drei Auszüge werden vereinigt, mit 60 Volumteilen 2-n. Natriumcarbonatlösung und 60 Volumteilen destillier tem Wasser gewaschen und der vierte Äthylenchlorid- extrakt mit den bereits verwendeten Waschlösungen gewaschen.
Man dampft die vereinigten, über Na triumsulfat getrockneten und filtrierten Äthylen chloridextrakte im Vakuum bis zu Gewichtskonstanz ein, löst 1 Gewichtsteil des Rückstandes in 1,5 Vo- lumteilen warmem Methanol und lässt bei 5 18 Stun den stehen. Dann filtriert man die ausgeschiedenen Kristalle, die zu einem grossen Teil aus Reserpin be stehen, ab, wäscht mit kaltem Methanol nach und befreit den Rückstand im Vakuum vom Lösungsmit tel. 2 Gewichtsteile des so erhaltenen rotbraunen, festen Schaums werden 2mal mit je 25 Volumteilen Benzol durchgearbeitet und filtriert.
Die benzol- lösliche Fraktion giesst man auf eine Säule von 40 Ge wichtsteilen aktiviertem Aluminiumoxyd (Woehn, Wirksamkeit I), welche dann zuerst 3mal mit je 50 Volumteilen Benzol und dann 6mal mit je 50 Volumteilen einer Mischung Benzol-Aceton (9:1) eluiert wird. Die erste Benzol-Aceton-Fraktion ver wendet man zur Extraktion des oben erhaltenen ben- zolunlöslichen Teils.
Aus der zweiten Fraktion ge winnt man nach dem Entfernen des Lösungsmittels einen leicht gebräunten, festen Schaum, der nach dem Umkristallisieren aus Methanol farblose, pris matische Nadeln von noch schwach unreinem De- serpidin ergibt. Man adsorbiert 1 Gewichtsteil davon an 20 Volumteile aktiviertem Aluminiumoxyd (Woehn, Wirksamkeit I), eluiert mit Benzol und 0,1% Me thanol enthaltendem Benzol und kristallisiert aus Methanol um.
Man erhält so farblose, prismatische Nadeln von -reinem Deserpidin, das bei 228-232 schmilzt.
Zu 0,5 Gewichtsteilen Deserpidin fügt man eine Lösung von 0,05 Gewichtsteilen Natrium in 25 Vo- lumteilen Methanol und kocht die Mischung unter Stickstoff eine Stunde lang am Rückfluss, wobei sich alles Deserpidin löst. Nach dem Kühlen engt man die erhaltene Lösung im Vakuum auf ein Volumen von ungefähr 10 Volumteilen ein, fügt 30 Volumteile Wasser und langsam konz. Salzsäure zu, bis die Lö sung stark sauer ist.
Man extrahiert erst mit 15 Vo- lumteilen Äther und dann 3mal mit je 10 Volum- teilen Äther. Die wässrige Phase wird mit konz. Am moniak basisch gestellt, mit 15 Volumteilen Methylen- chlorid extrahiert und 3 weitere Male mit je 10 Vo- lumteilen Methylenchlorid ausgezogen.
Die ver einigten Methylenchloridextrakte trocknet man über getrocknetem Kaliumcarbonat und dampft im Va kuum ein, wobei man den Deserpidinsäuremethylester als gelben, festen Schaum, welcher gemäss der Analyse die empirische Formel C22H2s04Nz besitzt, erhält. In der nämlichen Weise erhält man die entsprechenden Deserpidinsäurealkylester, wenn man anstelle von Methanol trockenes Äthanol oder Butanol verwendet.
<I>Beispiel 2</I> 0,5 Gewichtsteile Deserpidinsäuremethylester, der durch 2maliges Abdestillieren von Toluol im Vakuum getrocknet wurde, wird in 5 Volumteilen getrocknetem, frisch destilliertem Pyridin gelöst und 0,5 Volum- teile Essigsäureanhydrid unter Kühlen zugegeben. Die Reaktionsmischung wird 5 Tage bei 5 stehen gelas sen, dann auf 50 Volumteile Eiswasser gegossen und 12 Volumteile 5%iges wässriges Ammoniak zu gegeben.
Man arbeitet die Mischung während 10 Mi nuten gut durch, extrahiert dann ein erstes Mal mit 50 Volumteilen, dann mit 15 Volumteilen und schluss- endlich mit 10 Volumteilen Methylenchlorid, wäscht die vereinigten Extrakte 2mal mit 10 Volumteilen Natriumchloridlösung, trocknet sie über wasserfreiem Kaliumcarbonat und dampft im Vakuum ein.
Der so erhaltene Essigsäureester des Deserpidinsäure- methylesters schmilzt nach dem Umkristallisieren aus Methanol bei 275-278 ; seine Analyse entspricht der empirischen Formel C24H3oO,N2. Er besitzt eine sedative Wirksamkeit. Seine optische Drehung beträgt [a] D = -132 1 (in Chloroform).
Im Infrarot- Spektrum (in Nujol) zeigt es starke Banden bei 2948-2853, 1737, 1709, 1263, 1252, 1092, 732 cm-i; mittlere Banden bei 3386, 1462, 1444, 1379, 1358, 1333, 1314, 1301, 1287, 1214, 1184, 1157, 1116, 1042, 1010, 975, 880, 645 cm-';
schwache Banden bei 954, 928, 916, 908, 850, 829, 804 em-1; Schultern bei 3043, 1490, 1222, 1195, 1145, 1127, 1105, 1056, 1034 Cm-1. Es besitzt im Ultraviolett-Spektrum (in Äthanol) die folgenden Maxima: A, = 226 mu (a <I>=</I> 32200), 282-283 m,u (a = 7340), 289-290 mu (a = 6300) und Minima: A = 247-248 mcc (E <I>=</I> 2070), 288 mu (e = 6240).
<I>Beispiel 3</I> Zu einer Lösung von 0,46 Gewichtsteilen gut getrocknetem Deserpidinsäure-methylester in 5 Vo- lumteilen frisch destilliertem Pyridin werden langsam unter Kühlen 0,46 Gewichtsteile p-Toluolsulfonyl- chlorid in 1 Volumteil trockenem Benzol zugegeben. 1 Volumteil Pyridin wird verwendet, um das Reagens in der Reaktionsmischung zu waschen, worauf das Reaktionsgefäss geschlossen und die Mischung 5 Tage bei 5 stehengelassen wird.
Man gibt dann die röt liche Lösung auf 50 Volumteile Eiswasser, fügt 12 Volumteile 5%ige wässrige Ammoniaklösung zu und rührt das halbfeste Produkt gut während ungefähr 5 Minuten.
Die Reaktionsmischung wird dann mit 50 Volumteilen, 15 Volumteilen und 10 Volumteilen Methylenchlorid extrahiert, die vereinigten Extrakte 3mal mit wenig kalter Natriumchloridlösung ge waschen, über wasserfreiem Kaliumcarbonat getrock net und im Vakuum zu einem halbkristallinen Rück stand eingedampft.
0,63 Gewichtsteile dieses Rück standes löst man in Methylenchlorid, filtriert durch ungefähr 0,02 Gewichtsteile aktivierter Tierkohle über einem Diatomeenerdenfilter, dampft ein und kristallisiert aus 4 Volumteilen Benzol. Durch Auf arbeiten der Mutterlösung wird noch weiteres Ma terial erhalten. Nach dem Umkristallisieren aus Me thanol schmilzt der p-Toluolsulfonsäureester des Deserpidinsäuremethylesters bei 226-228 .
Er zeigt die optische Drehung [a]5, = -85 2 (Chloro form), und seine Analyse ergibt die empirische For mel C29H340"N2S. Sein Ultraviolett-Spektrum in Äthanol zeigt die folgenden Maxima: A, = 225 mu (e = 22250), 282 mu (e = 7860) und Minima: # = 247 ma (e = 2300).
Sein Infrarot-Spektrum (in Nujol) zeigt folgende starke Banden: 2956-2837, 1739, 1464, 1368, 1347, 1334, 1181, 1157, 11<B>1</B>6, 1094, 940, 920, 906, 844, 815, 740 cm-'; mitt lere Banden: 3429, 1600, 1378, 1313, 1303, 1287, 1275, 1266, 1253, 1211, 1142, 1129, 1055, 1041, 1023, 1010, 982, 877, 798, 723, 666 cm-'; schwache Banden bei: 704, 647 cm-1, Schultern bei: 3043, 1582, 1500, 1392, 1325, 1227, 1193, 1101, 830, 807 cm-1.
<I>Beispiel 4</I> 0,5 Gewichtsteile Methyldeserpidat, das durch 2maliges Abdestillieren von Toluol im Vakuum ge trocknet wurde, löst man in 5 Volumteilen trok- kenem destilliertem Pyridin und gibt unter Kühlen 0,5 Volumteile frisch destilliertes 2-Furoylchlorid zu.
Das entstandene Präzipitat wird durch Zufügen von 2 Volumteilen trockenem Benzol wieder gelöst und die Reaktionsmischung nach 5tägigem Stehen bei 5 in 50 Volumteilen Eiswasser gegossen.
Man fügt dann 12 Volumteile einer 5%igen wässrigen Ammo- niaklösung zu, arbeitet die Mischung während un gefähr 10 Minuten gut durch und extrahiert mit 50 Volumteilen,
dann mit 15 Volumteilen und schluss- endlich mit 10 Volumteilen Methylenchlorid. Die kombinierten Extrakte werden 2mal mit je 10 Volum- teilen Natriumchloridlösung gewaschen, über wasser freiem Kaliumcarbonat getrocknet und im Vakuum eingedampft.
0,72 Gewichtsteile des Rückstandes werden in 15 Volumteilen trockenem Benzol gelöst und über 14 Gewichtsteilen aktiviertem Aluminium oxyd (Woelm, Wirksamkeit I) chromatographiert. Aus den mit 200 Volumteilen Benzol und 100 Vo- lumteilen Benzol, das 0,1%Methanol enthält, erhal tenen Fraktionen gewinnt man nach Entfernen der Lösungsmittel und Kristallisieren aus Methanol den Furan-2-carbonsäureester des Deserpidinsäuremethyl- esters in weissen, feinen Nadeln vom F. 244-247 .
Er besitzt sedative und hypotensive Wirksamkeit, zeigt eine optische Rotation [a]5, = -141 0,5 (Chloroform) und entspricht der Formel C27H3406N2. Sein Ultraviolett-Spektrum in Äthanol zeigt die fol genden Maxima: A = 226 mu (e = 37700), = 255 mu (e <I>=</I> 18 000);
Minima: A. = 241 m/t (e = 14800), A, = 278-284 mtt. Sein Infrarot- Spektrum zeigt folgende starke Banden bei: 2941 bis 2816, 1710, 1305, 1187, 1123, 738 cm-1; mittlere Banden bei: 1631, 1575, 1463, 1400, 1378, 1350, 1266, 1230, 1109, 1093, 1061, 1041, 1030, 1015, 979, 969, 933, 766, 755, 746 cm-'; schwache Ban den bei: 3516, 3377, 3283, 917, 902, 885, 853, 822 cm-';
Schultern bei: 3042, 1736, 1441, 1327, 1282, 1223, 1213, 1155, 1145, 1081, 985, 72<B>1</B> cm-1. <I>Beispiel 5</I> Zu einer Lösung von 0,5 Gewichtsteilen Methyl- deserpidat in 4 Volumteilen trockenem, destilliertem Pyridin gibt man 0,5 Gewichtsteile 3,4-Dimethoxy- benzoylchlorid in 2 Volumteilen Benzol langsam unter Kühlen und gutem Rühren zu. Um das Reagens in das Reaktionsgefäss hinein zu waschen, wird 1 Volumteil Pyridin verwendet, dann das Reaktionsgefäss geschlos sen und während 5 Tagen bei 5 gehalten.
Die Reak tionsmischung wird dann in 50 Volumteile Eiswasser gegossen, 2 Volumteile konz. wässrige Ammoniaklösung in 10 Volumteilen Wasser zugegeben und nach 5minutigem gutem Rühren mit 50, dann 15 und 10 Volumteilen Methylenchlorid extrahiert. Die ver einigten Extrakte wäscht man mit 10 Volumteilen gesättigter Natriumchloridlösung, trocknet über was serfreiem Kaliumcarbonat, filtriert und dampft die Lösung im Vakuum zur Trockne ein.
Der Rückstand wird aus 5 Volumteilen Methanol kristallisiert und schmilzt bei 211-215 . Lösen in Methanol-Methy- lenchlorid, Behandeln mit Tierkohle und anschliessen des Umkristallisieren aus Methanol ergibt weisse Pris men des 3,4-Dimethoxy-benzoesäureesters des De- serpidinsäuremethylesters vom F.213-216 und der optischen Drehung [a]25,5 = -1400 20 (Chlo roform). Seine Analyse stimmt auf die empirische For mel C31H3,07N2. Sein Infrarot-Spektrum (Nujol) zeigt folgende starke Banden bei: 2929-2837, 1714, 1467, 1287, 1272, 1230, 1180, 1141,<B>1</B>099 cm-1; mittlere Banden bei 3392, 1605, 1519, 1423, 1381, 1354, 1338, 1324, 1313, 1298, 1251, 1209, 1066, 1028, 980, 953, 925, 880, 826, 762, 741, 727 cm-'; schwache Banden bei: 909, 850, 808, 650 cm-'; Schultern bei: 1596, 1151, 1110, 1038, 1015, 986.
Sein Ultraviolett-Spektrum zeigt die folgenden Ma xima: .? <I>= 224</I> m,tt (e <I>=</I> 52 880), 265 mu (e = 17 900), 284 m,u (e = 13 300), 290 mu (s = 13 360); Minima: 2. --_ 242-243 MA (s = 7350), 281<I>mA (8 =</I> 12980) und 287 mu (s = 12980).
Nimmt man anstelle des 3,4-Dimethoxy-benzoyl- chlorids eine entsprechende Menge 3,4,5-Trimethoxy- benzoylchlorid, so erhält man den 3,4,5-Trimethoxy- benzoesäureester des Deserpidinsäuremethylesters vom F. 228-232 und der optischen Rotation [a] D = 134 (Chloroform).
Aus 3,4,5-Trimethoxy-benzoylchlorid und Äthyl-deserpidat lässt sich der 3,4,5-Trimethoxy- benzoesäureester des Deserpidinsäureäthylesters er halten.
Zu einer Lösung von 0,2 Gewichtsteilen des 3,4,5-Trimethoxy-benzoesäureesters des Deserpidin- säuremethylesters in 3 Volumteilen Methanol und 0,1 Volumteil Methylenchlorid gibt man 0,2 Volum- teile verdünnte Schwefelsäure (1 Volumteil Schwefel säure zu 4 Volumteilen Wasser).
Nach dem Ab dampfen des Methylenchlorids lässt man die Lösung einige Stunden bei 5 stehen und erhält so das schwe felsaure Salz des 3,4,5-Trimethoxy-benzoesäureesters des Deserpidinsäuremethylesters, das mit Wasser in weissen Nadeln kristallisiert und nach dem Abfil- trieren und Waschen mit Methanol bei 266-269 (Zers.) schmilzt.
0,2 Gewichtsteile des oben genannten Esters wer den in 1 Volumteil Methanol aufgeschwemmt und mit mit gasförmiger Salzsäure gesättigtem Methanol so lange versetzt, bis der Ester in Lösung geht. Nach dem Eindampfen zur Trockne erhält man das salz saure Salz des 3,4,5-Trimethoxy-benzoesäureesters des Deserpidinsäuremethylesters, das aus 1 Volum- teil 95%igem Äthanol mit Wasser als Nadeln kristalli siert. Nach dem Filtrieren und Waschen mit Me thanol schmilzt das Produkt bei 253-256 (Zers.).
Zu einer Lösung von 0,2 Gewichtsteilen des beschriebenen Esters mit 3 Volumteilen Methanol und 0,1 Volumteil Methylenchlorid gibt man 0,05 Volumteil verdünnte Salpetersäure (1 Volumteil Sal petersäure zu 4 Volumteilen Wasser). Die Kri stallisation beginnt sofort, wobei nach dem Kühlen auf 5 die erhaltenen Plättchen abfiltriert und mit Me thanol gewaschen werden. Das salpetersaure Salz schmilzt unter Zersetzung bei 254-260 .
Entspre chend erhält man durch Zugabe von 0,3 Volumteil Oxalsäurelösung (1 Volumteil wasserfreie Oxalsäure zu 10 Volumteilen Wasser) das oxalsaure Salz vom F. 239-243 (Zers.).
Auf analoge Weise können weitere Ester von Alkylreserpaten hergestellt werden, so z. B. durch Umsetzung mit geeigneten Säuren, deren Chloriden oder Anhydriden. Als solche können z. B. genannt werden:
4-Methoxy-benzoesäure, Nikotinsäure, Iso- nikotinsäure, Zimtsäure, Phenylessigsäure, Mandel säure, Tropasäure, p-Methoxy-zimtsäure, 3,4,5-Tri- äthoxy-benzoesäure, 3,4-Methylendioxy-benzoesäure, O-Carbäthoxy-syringasäure, Thiophencarbonsäure,\ Picolinsäuren oder Chinolincarbonsäuren.
<I>Beispiel 6</I> Zu 0,5 Gewichtsteilen Äthyldeserpidat, das durch 2maliges Abdestillieren von Toluol getrocknet wurde, in 4 Volumteilen getrocknetem, destilliertem Pyridin werden langsam und unter Rühren 0,5 Gewichtsteile 3,4,5-Trimethoxy-benzoylchlorid in 2 Volumteilen trockenem Benzol zugegeben. Zum Nachwaschen verwendet man 1 Volumteil trockenes Pyridin. Das Reaktionsgefäss wird verschlossen und 3 Tage bei 5 gehalten, dann über Nacht noch bei Raumtemperatur stehengelassen.
Man giesst die Reaktionsmischung auf 50 Volumteile Eiswasser, fügt 2 Volumteile konz. wässrige Ammoniaklösung in 10 Volumteilen Wasser langsam und unter Rühren zu.
Nach 5minutigem Rühren extrahiert man die Mischung zuerst mit 50 Volumteilen, dann mit 15 Volumteilen und end lich mit 10 Volumteilen Methylenchlorid, wäscht die vereinigten Extrakte 2mal mit gesättigter Natrium chloridlösung, trocknet sie über wasserfreiem Kalium- carbonat, filtriert die Lösung und dampft sie im Vakuum zur Trockne ein. 3mal gibt man zum Rück stand Toluol zu und dampft dieses im Vakuum ab.
0,51 Gewichtsteile des erhaltenen Rückstandes löst man in 10 Volumteilen Benzol und chromatographiert über 10 Gewichtsteilen aktiviertem Aluminiumoxyd (Woelm, Wirksamkeit I). Als Waschlösung verwendet man 15 Volumteile Benzol. Die mit Benzol, das 0,1 Methanol und Benzol, das 0,2% Methanol enthält, er haltenen Fraktionen werden eingedampft und in Me thanol gelöst.
Verdünnte Salpetersäure (1 :4) wird zur methanolischen Lösung zugegeben, worauf man das salpetersaure Salz des 3,4,5-Trimethoxy-benzoe- säureesters des Deserpidinsäureäthylesters kristallin erhält, F. 255-260 (Zers.). Es kann aus einer Mi schung Methanol-Methylenchlorid umkristallisiert werden und schmilzt bei 258-260 (Zers.).
<I>Beispiel 7</I> Zu einer Lösung von 0,9 Gewichtsteilen Methyl- deserpidat in 20 Volumteilen trockenem, destilliertem Pyridin gibt man 1,0 Gewichtsteil Nicotinoylchlorid. Man lässt die Reaktion 5 Tage bei 5 stehen, giesst dann in<B>100</B> Volumteile Wasser und fügt 10 Volum- teile wässrige Ammoniaklösung zu.
Die Mischung wird 3mal mit je 30 Volumteilen Methylenchlorid extrahiert, die Extrakte vereinigt, mit 30 Volumteilen einer gesättigten Natriumchloridlösung gewaschen, über Magnesiumsulfat und wasserfreiem Natrium- carbonat getrocknet, filtriert und im Vakuum bei Zimmertemperatur zur Trockne eingedampft.
Der Rückstand wird über 10 Gewichtsteilen Magnesium silikat ( Florex ) chromatographiert, wobei 150 Vo- lumteile Methylenchlorid zum Eluieren verwendet werden. Nach dem Eindampfen des Lösungsmittels und Kristallisation aus Benzol erhält man den Ni kotinsäureester des Deserpidinsäuremethylesters vom F. 167-169 (Zers.).
<I>Beispiel 8</I> Man gibt zu einer Lösung von 0,9 Gewichtsteilen Methyldeserpidat in 20 Volumteilen trockenem, destilliertem Pyridin 1,5 Gewichtsteile Chinolin-6- carbonsäurechlorid, lässt die Reaktionsmischung 5 Tage bei 5 stehen und giesst sie in 100 Volumteile Wasser.
Man fügt 10 Volumteile wässriges Ammo niak zu, extrahiert die Mischung 3mal mit je 30 Vo- lumteilen Methylenchlorid, wäscht die vereinigten Extrakte mit 30 Volumteilen gesättigter, wässriger Natriumchloridlösung, trocknet über Magnesiumsulfat und wasserfreiem Natriumcarbonat, filtriert und dampft zur Trockne bei Raumtemperatur im Vakuum ein. Der Rückstand kristallisiert aus einer Mischung von Methanol und Äther.
Man erhält so das Di- hydrat des Chinolin-6-carbonsäureesters des De- serpidinsäuremethylesters, der bei 172-174 schmilzt (Zers.).
<I>Beispiel 9</I> Zu einer Lösung von 0,9 Gewichtsteilen Deser- pidinsäuremethylester in 20 Volumteilen trockenem, destilliertem Pyridin gibt man 1,2 Gewichtsteile ss Naphthoylchlorid, lässt die Reaktionsmischung 5 Tage bei 5 stehen und giesst in 100 Volumteile Wasser. 10 Volumteile einer wässrigen Ammoniaklösung wer den zugefügt und die Mischung 3mal mit 30 Volum- teilen Methylenchlorid extrahiert.
Die vereinigten Extrakte werden mit 30 Volumteilen einer gesättigten wässrigen Natriumchloridlösung gewaschen, über Ma gnesiumsulfat und wasserfreiem Natriumcarbonat getrocknet, filtriert und im Vakuum bei Raumtem peratur zur Trockne eingedampft. Den Rückstand chromatographiert man über 10 Gewichtsteilen Ma gnesiumsilikat ( Florex ) und verwendet 150 Volum- teile Methylenchlorid zum Auswaschen.
Nach dem Eindampfen des Lösungsmittels und Kristallisation aus Benzol erhält man den fl-Naphthoesäureester des Deserpidinsäuremethylesters vom F. 191-192 .
<I>Beispiel 10</I> Zu einer Lösung von 0,9 Gewichtsteilen Methyl- deserpidat in 20 Volumteilen getrocknetem, destillier tem Pyridin gibt man 1,0 Gewichtsteil 3,4-Methylen- dioxy-benzoylchlorid und lässt die Reaktionsmischung 5 Tage bei 5 stehen.
Man giesst dann in 100 Vo- lumteile Wasser, fügt 10 Volumteile wässrigen Ammo niak zu und extrahiert die Mischung 4mal mit je 30 Volumteilen Methylenchlorid. Die vereinigten Extrakte werden über Magnesiumsulfat und wasser freiem Natriumcarbonat getrocknet, filtriert und zur Trockne eingedampft.
Den Rückstand chromato- graphiert man über 10 Gewichtsteilen Magnesium- silikat ( Florex ) und erhält so nach dem Auslösen mit Methylenchlorid, das 5.% Methanol enthält, Ab dampfen des Lösungsmittels und Kristallisieren aus einer Mischung von Methylenchlorid, Methanol und Ligroin den 3,4-Methylendioxy-benzoesäureester des Deserpidinsäuremethylesters vom F. 195-196 .
<I>Beispiel 11</I> 0,3 Gewichtsteile Deserpidinsäure werden in 20 Volumteilen einer 1 : 1-Mischung von Methylen- chlorid und Dioxan gelöst und eine Lösung von Di- azo-n-butan in Äther langsam unter Kühlung in einem Eisbad so lange zugegeben, bis kein Stickstoff mehr entweicht und die schwach orange Farbe bleibt. Die Mischung wird 24 Stunden bei Raumtemperatur ste hen gelassen, dann im Vakuum vom Lösungsmittel befreit und der Rückstand in Methylenchlorid gelöst.
Die erhaltene Lösung giesst man auf eine kurze Ko lonne von 5 Gewichtsteilen Magnesiumsilikat ( Flo- rex ), eluiert mit Methylenchlorid, das 10% Methanol enthält, und erhält so nach dem Verdampfen des Lö sungsmittels den Deserpidinsäure-n-butylester. Der Rückstand wird in 10 Volumteilen trok- kenem Pyridin gelöst und 5 Volumteile Essigsäure anhydrid zugegeben.
Man lässt 4 Tage bei 5 stehen, giesst die Reaktionsmischung in Wasser und fügt 10 Volumteile einer konz. wässrigen Ammoniaklösung zu. Man extrahiert 4mal mit Methylenchlorid, wäscht die vereinigten Extrakte mit gesättigter, wässriger Natriumchloridlösung und trocknet über Magnesium sulfat und wasserfreiem Natriumcarbonat. Nach dem Eindampfen des Lösungsmittels reinigt man den rohen Rückstand durch Chromatographieren an 5 Ge wichtsteilen Magnesiumsilikat,
wobei als Lösungs mittel Methylenchlorid verwendet wird. Die mit Methylenchlorid, das 10% Methanol enthält, erhal tene Fraktion ergibt -den kristallinen Essigsäureester des Deserpidinsäure-n-butylesters vom F. 226-228 .

<I> Example 3 </I> To a solution of 0.46 parts by weight of well-dried methyl deserpidate in 5 parts by volume of freshly distilled pyridine, 0.46 parts by weight of p-toluenesulfonyl chloride in 1 part by volume of dry benzene are slowly added with cooling. One part by volume of pyridine is used to wash the reagent in the reaction mixture, the reaction vessel is closed and the mixture is allowed to stand at 5 for 5 days.
The reddish solution is then added to 50 parts by volume of ice water, 12 parts by volume of 5% aqueous ammonia solution are added and the semi-solid product is stirred well for about 5 minutes.

Dilute nitric acid (1: 4) is added to the methanolic solution, whereupon the nitric acid salt of the 3,4,5-trimethoxybenzoic acid ester of deserpidic acid ethyl ester is obtained in crystalline form, mp 255-260 (decomp.). It can be recrystallized from a mixture of methanol-methylene chloride and melts at 258-260 (decomp.).
<I> Example 7 </I> 1.0 part by weight of nicotinoyl chloride is added to a solution of 0.9 parts by weight of methyl deserpidate in 20 parts by volume of dry, distilled pyridine. The residue is chromatographed over 10 parts by weight of magnesium silicate (Florex) and 150 parts by volume of methylene chloride are used for washing out.
Evaporation of the solvent and crystallization from benzene gives the fl-naphthoic acid ester of methyl deserpidate with a melting point of 191-192.
