worin Res die oben genannte Bedeutung besitzt, R für einen Alkylrest, z. B. einen Methyl-, Äthyl-, Propyl- oder Butylrest, und R' für einen Methyl-, Propyl- oder Butylrest steht, oder von Salzen solcher Ester.
Die neuen Ester besitzen ähnlich der im Haupt patent beschriebenen Verbindung eine dem Reserpin vergleichbare ausgeprägte blutdrucksenkende Wir kung, jedoch sind sie nicht oder nur wenig sedativ hypnotisch wirksam. Ganz besonders ist das Methyl- reserpat-carbopropoxysyringat durch eine hohe hypo- tensive Wirksamkeit ohne sedative Komponente aus gezeichnet. Die neuen Ester sollen als Heilmittel Ver wendung finden.
Das erfindungsgemässe Verfahren ist dadurch ge kennzeichnet, dass man einen O-Syringoyl-reserp- säurealkylester der Formel

abgebenden Verbindung umsetzt, in welchen For meln R und R' die oben gegebene Bedeutung be sitzen.
So kann man beispielsweise auf den Reserp- säureester einen Kohlensäuremethyl-, -propyl- oder -butylester oder ein reaktionsfähiges funktionelles Derivat eines solchen Esters, wie ein Halogenformiat, besonders ein Alkylchlorformiat, einwirken lassen.
Die Reaktion wird zweckmässig in Anwesenheit von Verdünnungsmitteln und bzw. oder Konden sationsmitteln durchgeführt. Bei Verwendung von Säurehalogeniden arbeitet man vorteilhaft in wasser freien Lösungsmitteln in Gegenwart säurebindender Mittel, wie Alkali- oder Erdalkalicarbonaten, oder starken organischen Basen, wie tertiären Aminen. So verwendet man z. B. für Säurehalogenide Pyridin als Lösungsmittel.
Je nach der Arbeitsweise erhält man die neuen Diester in freier Form oder als Salze. Letztere lassen sich auch aus den freien Estern durch Umsetzung mit anorganischen oder organischen Säuren, wie Ha logenwasserstoffsäuren, Schwefelsäure, Phosphor säure, Salpetersäure, Essigsäure, Propionsäure, Zi- tronensäure, Milchsäure, Oxalsäure, Bernsteinsäure, Äpfelsäure, Weinsäure, Ascorbinsäure, Methansul- fonsäure, Äthansulfonsäure, Oxyäthansulfonsäure, Benzoesäure,
Salicylsäure, p-Amino-benzoesäure oder Toluolsulfonsäure, gewinnen. Aus den Salzen kön nen die neuen Ester in freier Form, z. B. durch Be handeln ihrer Lösung mit Silbercarbonat, erhalten werden.
In den nachfolgenden Beispielen besteht zwischen Gewichtsteil und Volumteil die gleiche Beziehung wie zwischen Gramm und Kubikzentimeter. Die Temperaturen sind in Celsiusgraden angegeben.
<I>Beispiel 1</I> Zu einer Lösung von 0,2 Gewichtsteilen Methyl- O-syringoyl-reserpat in 20 Volumteilen Benzol und 2 Volumteilen Pyridin gibt man 2 Volumteile Methyl- chlorformiat. Die Mischung wird während 5 Minuten am Rückfluss gekocht und im Vakuum eingedampft. Man behandelt den Rückstand mit 25 Volumteilen Wasser, bis er körnig geworden ist, filtriert das Was ser, wäscht den Rückstand mit Wasser, nimmt in 15 Volumteile Methanol auf.
Die erhaltene Lösung behandelt man während 5 Minuten mit 0,3 Gewichts teilen Silbercarbonat, filtriert und dampft im Vakuum zur Trockne ein. Man erhält so das Methyl-O-(O'- carboxymethyl-syringoyl)-reserpat, das nach dem Umkristallisieren aus 5 Volumteilen Äthanol bei 231 bis 232 schmilzt.
Das im Beispiel als Ausgangsstoff verwendete Methyl-O-syringoyl-reserpat kann auf die folgende Weise erhalten werden: 0,34 Gewichtsteile Methyl-O-(O'-carbäthoxy- syringoyl)-reserpat werden in 0,1 Gewichtsteil 3-n. alkoholischem Ammoniak gelöst und 3 Tage lang bei 5 stehengelassen, dann im Vakuum zur Trockne eingedampft und der feste Rückstand mit wenig wasserfreiem Äthanol umkristallisiert. Man erhält 0,12 Gewichtsteile Methyl-O-syringoyl-reserpat vom F. 190-192 .
Zur Bestimmung der Identität der erhaltenen Verbindungen löst man 0,03 Gewichtsteile davon in Methanol und lässt mit einem überschuss Diazomethan bei Raumtemperatur über Nacht stehen. Man dampft das Lösungsmittel ein und kristallisiert das erhaltene Reserpin aus Aceton um.
Es besitzt den gleichen Schmelzpunkt (264-265 ) und die gleiche Infrarot-Absorptionskurve wie auf andere Weise erhaltenes Reserpin. <I>Beispiel 2</I> Zu einer Lösung von 0,2 Volumteilen Methyl- O-syringoyl-reserpat in 20 Volumteilen Benzol und 2 Volumteilen Pyridin gibt man 2 Volumteile Propyl- chlorformiat und kocht die Mischung 5 Minuten am Rückfluss. Nach 30 Minuten Stehen bei Raumtempe ratur dampft man im Vakuum zur Trockne ein,
wäscht das körnige Produkt mit Wasser, nimmt es in Methanol auf und rührt mit 0,3 Gewichtsteilen Silbercarbonat während 5 Minuten. Nach dem Ab- filtrieren dampft man im Vakuum ein und kristalli siert das so erhaltene Methyl-O-(O'-carbopropoxy- syringoyl)-reserpat aus Aceton-Wasser um. F. 180 bis 184 .
Verwendet man anstelle des Propylchlorformiats eine entsprechende Menge Butyl- oder Isobutylchlor- formiat und verfährt im übrigen wie oben beschrie ben, so erhält man das Methyl-O-(O'-carbobutoxy- syringoyl)-reserpat vom F. 187-189 bzw. das Me- thyl-O-(O'-carboisobutoxy-syringoyl)-reserpat, das ein Hydrochlorid-monohydrat vom F. 224-225 bildet.
Dieses Hydrochlorid lässt sich durch Schütteln seiner methanolischen Lösung mit Silbercarbonat in die freie Base überführen, die nach Umkristallisieren aus Äthanol bei 192-194 schmilzt.
Geht man anstelle vom Methyl-O-syringoyl-reser- pat, das in den Beispielen 1 und 2 als Ausgangsstoff verwendet wurde, vom entsprechenden Äthyl-O- syringoyl-reserpat aus, so erhält man die folgenden Verbindungen: Äthyl-O-(O'-carbomethoxy-syringoyl)-reserpat, Äthyl-O-(O'-carbopropoxy-syringoyl)-reserpat, Äthyl-O-(O'-carbobutoxy-syringoyl)-reserpat, Äthyl-O-(O'-carboisobutoxy-syringoyl)-reserpat.
